1. Effects of growth hormone (GH) were examined on short-term aspects of seawater adaptation in coho salmon smolts. 2. Injection of somatostatin (SRIF) immediately prior to seawater entry suppressed plasma GH levels, but did not have any significant effects at 6 or 12 hr on hematocrits, plasma glucose or plasma Na+ levels. 3. Plasma GH levels increased 250% within 36 hr after seawater exposure. 4. Plasma glucose levels, in contrast, were significantly lower in the seawater fish after 36 hr post-exposure. 5. Plasma Na+ levels increased to 190 mEq/1 by 24 hr but subsequently returned to freshwater levels while hematocrits showed no significant changes over the 72 hr of exposure. 6. The significance of these results is discussed in terms of successful seawater adaptation in coho salmon.  相似文献

Increased growth during winter increased the incidence of age 1+ Salmo salar smolts in spring. High condition factor in spring and good growth in summer was associated with a high incidence of sexually mature males in autumn. In two experiments, groups (n=160–237 per group) of individually identified parr, either ungraded (lower and upper modal groups: LMG, UMG) or size-graded (LMG only), were reared at either 10, 6 or 3 °C overwinter (Nov to May). At 10 °C, up to 51% of parr originally in the LMG became smolts in spring at age 1+. In contrast, at either 6 or 3 °C (control), < 6% of LMG parr became smolts. The probability of being recruited into the UMG overwinter was positively related to initial body size, and was increased by size-grading. Smolt recruitment was two-fold higher among females compared to males; a proportion of males by age 0+ had already opted to mature at age 1+ rather than smolt at age 1+. In contrast, smolting at age 1+ was not inhibited in males previously mature at age 0+. During summer (May to Nov), all experimental groups were reared at ambient temperature, each subdivided between fresh water (max 21 °C) or seawater (max 15 °C). Good growth in seawater of winter recruits into the UMG confirmed they had completed smolting. Mortality in seawater among parr was 41–83%, and among smolts was 10–22%. Specific growth rate during summer was inversely related to winter rearing temperature. The incidence of sexual maturity in autumn at age 1+ among male parr was positively related to winter rearing temperature, fork length and condition factor in May, but there was large variation among individuals with respect to body size and maturity. Summer rearing in seawater reduced growth and the incidence of maturation. Parr and post-smolt maturity was 84–99% and 100% in fresh water respectively, 21–58% and 0% in seawater.  相似文献

Serum protein, glucose and fat levels were determined for Coho salmon parrs, freshwater smolts, seawater smolts, seawater stunts and freshwater desmolts. Liver and muscle glycogen, fat, protein and water concentrations were also calculated. Serum protein, glucose and fat levels were significantly lower in the freshwater smolts than in the parrs. Furthermore, both liver and muscle total fat levels were markedly decreased in the smolts, suggesting that smoltification is associated with increased catabolism. Smolts that failed to reach seawater by late summer reverted to a parr-like appearance and also regained the biochemical characteristics of parrs (high body fat and glycogen). Premature transfer of Coho presmolts into seawater caused impaired growth (stunting). Stunted Coho had higher muscle protein levels than normal smolts and parrs. Tissue water levels were not significantly different between stunts and normal seawater smolts, suggesting that the stunting phenomenon may not be caused primarily by osmoregulatory failure but may be due to shifts in metabolic patterns. In contrast to the pronounced changes seen in Coho salmon, transfer of King salmon to sea water did not result in increased catabolism of body reserves or in the production of stunts.  相似文献

To examine the changes in secretion of growth hormone (GH) and prolactin (PRL) with reference to their osmoregulatory roles, changes in pituitary mRNA levels and plasma concentrations of these hormones were examined during seawater adaptation in silvery juveniles (smolts) and precociously mature males (dark parr) of amago salmon (Oncorhynchus rhodurus). Transfer to seawater increased plasma sodium levels in both smolts and dark parr. Smolts adjusted their plasma sodium to the level associated with seawater-adaptation (165 mEq/liter) within 3 days, whereas no adjustment was seen in dark parr; the latter failed to survive in seawater for more than 3 days. In smolts, plasma GH levels increased significantly 1 day after transfer, whereas there was no significant change in dark parr. An increase in GH mRNA levels was observed in smolts in association with increased plasma GH, whereas there was no change in dark parr. In contrast, a reduction in plasma PRL levels was consistently observed in both smolts and dark parr after transfer to seawater. However, there was no significant change in PRL mRNA levels in either smolts or dark parr. These results suggest that both gene expression and release of GH are activated by seawater transfer only in smolts with adequate seawater adaptability, whereas PRL gene expression is decreased after seawater transfer regardless of seawater adaptability.  相似文献

During three consequtive years, 1975–1977, Individually tagged Baltic salmon Salmo salar smolts of sexually immature male and female fish (n = 35027, mean size: 15.2 cm) and precocious males (n = 6518, mean size: 14.2 cm) were released into Umeälven (Ume river), northern Sweden. Rate of survival (% captured adults) based on 3714 recoveries was significantly higher (p < 0.01) for smolts from immature fish (10.2%) than those from smolts of early maturing males, i.e. precocious males (2.2%). corresponding to an average yield of 474 and 85 kg per KHX) smolts released, respectively. Gain in survival was on average 2.5% and 1.4% per cm increase in smolt size for immature smolts and smolts from precocious males, respectively. The poor survival among smolts of precocious males is suggested to he related to an interaction between sexual maturation and smolting linked to incompletely resorbed gonads leading to a non migratory behaviour. These non migratory males are then suggested to suffer heavily by predation in the river.
The two smolt categories had a similar growth pattern in sea. Smolts from precocious males did not mature early in sea indicating no relation to grisling, i.e. sexually maturing fish returning after first winter in sea. Adult weight of fish returning the fourth summer after release was related to smolt size (P < 0.05). Our Response Surface Model (RSA) predicted that large smolts (19.0 cm) had a higher specific growth rate over their life-span compared to small smolts (<15.0 cm), 0.86% d−1 and 0.46% d−1, respectively. Large smolts (19.0 cm) attained a size of 3.0 kg during their second winter in sea about six months earlier than small smolts (13.0 cm). The paper discusses alternative release strategies that can be employed if the ultimate goal of salmon stocking is maximizing yield.  相似文献

Synopsis The hypothesis that an increase in the mortality rate of Atlantic salmon would be produced by the synergistic effect of osmotic stress and of stress due to the presence of predators was tested by putting two groups of smolts (one acclimated to seawater and the other not acclimated) into a tank containing predators. These smolts were tested afterwards in a seawater-challenge test, together with smolts of the control groups for both experimental groups. The mortality rate of the non-acclimated and predator-exposed smolts was 90%, and that of the seawater acclimated and predator-exposed smolts 43%, compared to no mortality among the control groups. Lack of acclimation to seawater did not in itself induce any lethal stress, but in conjunction with predator-stress there was a synergistic effect, leading to an increase in the mortality rate. The hypothesis is therefore considered to be valid.  相似文献

To elucidate the ultrastructural modifications of the gill epithelium during smoltification, gills of the Atlantic salmon (Salmo salar) were examined by electron microscopy at three stages of this process, which were defined as follows: "parrs" were freshwater fish that had not yet started their transformation; "freshwater smolts" were freshwater fish that were ready to enter seawater; and "seawater smolts" were smolts that had been transferred from fresh water and maintained for 4 days in seawater (35%). In the gill epithelium of parrs, there were two types of chloride cells. The large chloride cells contained deeply stained mitochondria and numerous apical, irregular, dense, membrane-bound bodies that formed 77% of the chloride cell population and were distinguished easily from small chloride cells that have distinctly paler mitochondria and no dense bodies in their apical cytoplasm. In freshwater smolts, the large chloride cells formed 95% of the chloride-cell population. In contrast to the small chloride cells that were not modified, they almost doubled in size. Their tubular system developed extensively to form a tight network with regular meshes significantly smaller than those observed in parr chloride cells. Forty percent of the large chloride cells were associated with a new type of cell, the accessory cell, to which they were bound by shallow apical junctions. Half of these accessory cells were not seen to be in contact with the external medium. In seawater smolts, 80% of the large chloride cells were associated with accessory cells. Most accessory cells reached the external medium and sent numerous cytoplasmic interdigitations within the apical portion of the adjacent chloride cells. As a result, a section through the apical portion of the chloride cells and their associated accessory cells revealed a mosaic of interlocked cell processes bound together by an extended, shallow apical junction. It was concluded that the Atlantic salmon develops in fresh water most of the ultrastructural modifications of the gill epithelium which in most euryhaline fish are triggered by exposure to seawater. The effective transfer into seawater would act only as a final stimulus to achieve some adequacy between the freshwater smolt and its new environment.  相似文献

Seasonal, diel and tidal rhythmic activity of hatchery-reared Atlantic salmon ( Salmo salar ) smolts migrating through a large estuary was studied by ultrasonic tracking of 46 individuals during two seasons. Prior to 10 May each year most smolts were inactive and remained near shore in shallow water. After 10 May nearly all smolts moved away from the release point into swift water and made rapid seaward progress. Initiation of migration each year occurred when river and hatchery water temperatures rose above 9°C. Migration in the estuary was largely passive drift, and as a result there were distinct tidal rhythms of ground ('swimming') speed and net seaward travel. There were no diel rhythms in ground speed or net seaward travel; smolts drifted seaward on the tides during both day and night. Smolts may be slightly deeper during day than night.  相似文献

The intestinal role in osmoregulatory preadaptation of freshwater-adapted (FW) yearling coho salmon during smoltification was examined. Measurements of intestinal net fluid absorption (JV) using an in vitro sac preparation showed that JV of FW parr was significantly less than that of either FW or seawater-adapted (SW) smolts. From two to four weeks following the springtime thyroxin surge, JV increases in FW coho to a level comparable with that observed for SW smolts. JV remained elevated in FW smolts throughout most of the summer and then decreased in the autumn. Two months after the thyroxin peak, intestinal wet weight per unit serosal surface area increases. Coho stunts resulting from premature transfer into sea water had significantly higher JV than that seen in SW smolts. The elevated JV of stunts is only partially explicable on the basis of differences in body weight between stunts and smolts.
Our observations indicate that an increase in intestinal fluid absorption is a preadaptive change associated with smoltification occurring in concert with alterations in renal and branchial osmoregulatory mechanisms. The timing of the increase in JV suggests a phase relationship to the thyroxin surge. This temporal relationship may help to explain the efficacy of using changes in plasma thyroxin levels to predict hatchery-fish release dates that may optimize eventual seawater survival.  相似文献

Carbonic anhydrase (CA) activity was measured in blood and in gill tissue of coho salmon smolts during chronic exercise and subsequent transfer into seawater. The mean level of CA activity was higher in blood than gill tissue in both freshwater and seawater. CA activity in gill tissue increased significantly after the smolts had adapted to seawater. CA activity in blood decreased significantly in the group of fish given the highest exercise level after they had adapted to seawater. There were no significant differences in CA activity in gill tissue between control and exercise groups in either freshwater or seawater.  相似文献

This study was designed to elucidate the mechanism by which somatostatin administration ameliorates or abolishes diarrhea in pancreatic cholera syndrome (PCS). Absorption (or secretion) of water and electrolytes was measured in 30-cm segments of jejunum of 18 healthy volunteers in whom PCS was mimicked by intravenous infusion of VIP. Using the triple-lumen tube technique, the intestine was perfused with a plasma-like electrolyte solution while administering intravenous saline (control), VIP (400 pmol/kg/hr), somatostatin (5000 pmol/kg/hr), or VIP plus somatostatin. VIP infusion abolished water and electrolyte absorption and somatostatin had no effect on these VIP-induced transport changes regardless of whether somatostatin infusion was started before or after VIP infusion. Somatostatin infusion had no effect on VIP plasma concentration when elevated by intravenous VIP infusion (control: 10 +/- 1 pmol/l; during VIP infusion: 108 +/- 6). In a patient with pancreatic cholera syndrome identical perfusion experiments showed jejunal water secretion (93 ml/30 cm/hr) which changed to absorption (65 ml/30 cm/hr) when somatostatin was infused (5000 pmol/kg/hr). Plasma VIP concentration fell from 145 to 74 pmol/l (normal less than 50) during somatostatin infusion. Stool weight fell from 3722 g to 819 g per 24 hours when somatostatin was given at a dose of 2500 pmol/kg/hr for two days. Our observations in healthy subjects show that somatostatin has no effect on intestinal transport at the mucosal level when circulating VIP concentration is elevated.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Twenty-two salmon smolts, Salmo salar L., carrying miniature sonic tags were tracked individually for periods of up to 175 h in Loch Voil, Scotland, during May 1979 and 1980. Activity was predominantly nocturnal, 80% occurring between 21.00 and 06.00 hours, and was apparently undirected. Average velocities during this active interval were 0.6 body lengths per second (bl s−1), with 98 and 93% of the time spent moving at less than 2 and less than 1 bl s−1, respectively. The rates of downstream displacement were 0.04 bl s−1 in 1979 and 0.01 bl s−1 in 1980. The direction of displacement of smolts and of movement of water at a depth of 1 m was positively correlated ( P <0.001) and smolt displacement was biassed slightly ahead of water movement. Mean step lengths were 141 and 200 m in 1979 and 1980, respectively. Rates of downstream passage of 327 ICES plate-tagged smolts released 16.8 km upstream of the fish trap at Clunie dam, Loch Tummel, during the spring migrations of 1975 and 1976 averaged 0.13 bl s−1 in each year: net surface water movement was about 3.7 times this rate during the same intervals. These data are consistent with the model of passive smolt migration postulated by Tytler et al. (1978) and suggest that the active component required to ensure passage through a loch (Thorpe & Morgan, 1978) is very small.  相似文献

The distal rabbit colon was used as a model to investigate the influence of the cationophore Salinomycin in vivo with a single-pass perfusion, and in vitro with a modified Ussing chamber technique. For in vivo experiments with labelled 14C-PEG as a volume marker in the perfusate, a dose of 10E-4 mol/l Salinomycin was used. Net water (53 microliters/h/cm), net chloride (3 mumol/h/cm) and net sodium (3.6 mumol/h/cm) absorption was not significantly influenced, but net potassium secretion (-3 mumol/h/cm) was decreased to zero and transepithelial potential (PD) reduced from -45 mV to -33 mV. 10E-4 mol/l Salinomycin, applied in vitro on the muscosal side, decreased PD in 80 min and 10E-3 mol/l in 30 min from 18 mV to zero. Both concentrations decreased the short-circuit current (Isc = 77 microA/cm2) in 60 min, respectively 30 min to 40 microA/cm2. After 60 min mucosal 10E-4 mol/l Salinomycin the Isc increased, resulting from a transepithelial conductance (Gt) increase from 3 to 40 mS/cm2. A dose-related effect was present on PD, Isc and Gt at concentrations between 10E-7 and 10E-6 mol/l. The unidirectional 22Na fluxes were increased to 20 times the control values and net Na transport disappeared. We conclude that Salinomycin when applied in usual doses (10E-4 mol/l) as a coccidiostatic feed additive, profoundly affects colonic electrolyte transport.  相似文献

Functions of the anatomically obvious, yet peculiar, pyloric ceca of the fish gut have been a source of conjecture for over two millennia since Aristotle hypothesized on digestive utilities. Here, we demonstrate regulated and adaptive changes in osmoregulatory physiology of ceca from chinook salmon (Onchorhynchus tshawytscha). Transfer of salmon from freshwater to seawater (both short- and long-term) significantly stimulated both fluid uptake from 5.1 to 8.3-9.3 microl/cm2/hr and also Na+/K+ -ATPase from 6.5 to 8.3-9.6 micromol/ADP/mg protein/hr. Similar changes were induced with implants of cortisol, which resulted in high physiological cortisol levels in plasma. Ceca, which can number about 200 in chinook salmon, were estimated to account for the majority of fluid uptake capacity of the intestine and, after long-term seawater adaptation, the proportion of uptake capacity was sixfold higher. Transport physiology of ceca is thus under environmental and endocrine control indicative of an important role in salt and water homeostasis.  相似文献

The early migration and habitat use of brown trout Salmo trutta post‐smolts tagged with acoustic transmitters (n = 50) were investigated in a fjord system in central Norway from 30 April to 26 November 2014. The main aims were to investigate return rate, marine residence time and spatial use of the fjord system. Median seaward migration and return to fresh water dates were 22 May and 4 July, respectively. Of the 40 seaward migrating smolts, 26 returned to fresh water, giving a minimum return rate to fresh water of 65%. Entrance to the fjord from the river occurred mainly at night (80% of the S. trutta), however, no such diurnal pattern was observed during the return migration. Mean marine residence time was 38 days, but with large individual variation (22–99 days). The innermost parts of the study area were more utilized than the outer part of the fjord system during the sea residency, and with more use of the near shore habitat than the open, pelagic areas. Many post‐smolts also utilized the outer part of the fjord system, however, and 94% of the post‐smolts were recorded at least 14 km from the home river mouth. Marine survival and distribution in the fjord were size dependent with the largest individuals utilizing outer fjord areas and having higher return rates to fresh water. As far as is known, this is the first published study on temporal and spatial behaviour in the marine environment of first‐time S. trutta migrants during the full course of their first trip to sea.  相似文献

Legionella pneumophila in sterile distilled water was not detected after ultraviolet irradiation by FLONLIZER, a new-type sterilizer, at a flow rate of 82.5 to 364.8 liters/hr. When irradiated by FLONLIZER at a flow rate of under 324.0 liters/hr, no viable cells of legionellae, other heterotrophic bacteria and bacterivorous protozoa were detected in the cooling tower water, which was found to contain L. pneumophila. No viable cells of L. pneumophila and L. bozemanii suspended in sterile distilled water were detected after the irradiation with UV-doses of over 6.16 X 10(3) micro W.sec/cm2. At the irradiation of low UV-doses under 1.06 X 10(4) micro W.sec/cm2, the viable count of legionellae recuperated by photoreactivation from UV-damage increased with the exposure time under a white fluorescent lamp. However, in the samples irradiated with UV-doses of over 3.52 X 10(4) micro W.sec/cm2, equal to the FLONLIZER, legionellae did not recuperate even after 18 hr illumination with a white fluorescent lamp. FLONLIZER is thus expected to act as a sterilizer which can control the legionellae inhabiting cooling tower systems placed in outdoor space.  相似文献

Atlantic salmon Salmo salar smolts were exposed to one of the four different aerobic exercise regimens for 10 weeks followed by a 1 week final smoltification period in fresh water and a subsequent eight‐day seawater transfer period. Samples of gill and intestinal tissue were taken at each time point and gene expression was used to assess the effects of exercise training on both branchial and intestinal osmoregulatory pathways. Real‐time polymerase chain reaction (PCR) analysis revealed that exercise training up‐regulated the expression of seawater relevant genes in the gills of S. salar smolts, including Na+, K+ ATPase (nka) subunit α1b, the Na+, K+, 2 Cl? co‐transporter (nkcc1) and cftr channel. These findings suggest that aerobic exercise stimulates expression of seawater ion transport pathways that may act to shift the seawater transfer window for S. salar smolts. Aerobic exercise also appeared to stimulate freshwater ion uptake mechanisms probably associated with an osmorespiratory compromise related to increased exercise. No differences were observed in plasma Na+ and Cl? concentrations as a consequence of exercise treatment, but plasma Na+ was lower during the final smoltification period in all treatments. No effects of exercise were observed for intestinal nkcc2, nor the Mg2+ transporters slc41a2 and transient receptor protein M7 (trpm7); however, expression of both Mg2+ transporters was affected by salinity transfer suggesting a dynamic role in Mg2+ homeostasis in fishes.  相似文献

The impacts of polychlorinated biphenyl (PCB) exposure on smoltification and subsequent seawater performance were investigated in hatchery-reared, anadromous Arctic charr (Salvelinus alpinus). The fish were subjected to a 2-month summer seawater residence, after which they were orally dosed with 0 (Control, C), 1 (Low Dose, LD) or 100 mg Aroclor 1254 kg(-1) body mass (High Dose, HD) in November. They were then held in fresh water, without being fed (to mimic their natural overwintering in freshwater), until they had smolted in June the next year. The smolts were then transferred to seawater and fed to mimic their summer feeding residence in seawater, followed by a period without food in freshwater from August until maturation in October. Compared with C and LD charr, the HD charr had either a transient or a permanent reduction in plasma growth hormone, insulin-like growth factor-1, and thyroxin and triiodothyronine titers during the period of smoltification. These hormonal alterations in the HD charr corresponded with impaired hyposmoregulatory ability in May and June, as well as reduced growth rate and survival after transference to seawater. Consequently, fewer fish in the HD group matured in October compared to the other two treatments. The HD fish had a liver PCB concentration ranging between 14 and 42 mg kg(-1) wet mass, whereas there were similar, and very low, liver PCB concentrations in LD and C fish throughout the smolting period. Our findings suggest that PCB might compromise mechanisms important for fitness in a fish species living in an extreme environment.  相似文献

Energy contents of immature parr and smolts, and mature resident and anadromous brown trout Salmo trutta sampled from a small stream in southern Norway were estimated from lipid, protein and carbohydrate concentrations. In immatures the lipid concentrations were highest in parr in the autumn. Mean lipid concentrations increased significantly with age in parr sampled in autumn (1·3% in age 0+ to 3·4% in age 3+), whereas they did not in smolts. The lipid concentrations of parr in spring were not significantly different from those of similarly aged smolts. By contrast, the relative water content (%) decreased with age in parr in the autumn and increased with age in smolts, mean values being slightly higher in smolts (78%) than in parr (77%). Protein and carbohydrate concentrations did not vary with age in the immature fish, mean protein concentrations being 18·0, 17·5 and 16·8% in parr in the autumn and spring, and in smolts, respectively. In residents, the concentrations of lipids were lower and of water higher, in age group 1 than in older fish, whereas there was no significant variation with age amongst anadromous trout. The energy concentration of 2+ smolts (349 kJ 100 g-1) was similar to that of 0+ parr in the autumn. Mean somatic energy density in autumn was 1·1 times higher in freshwater residents than in parr at age 1+ (407 and 387 kJ 100 g-1) and marginally different at age 2+ (462 and 426 kJ 100 g-1, respectively). The energy contents per unit mass of residents were 1·3–1·6 times that of similar aged smolts. Mean somatic energy density of anadromous trout (504 kJ 100 g-1) was higher than that of residents (455 kJ 100 g-1). Somatic energy, lipid and protein concentrations were correlated highly with water contents of all life stages, age and sex groups.  相似文献

We determined net fluid secretion rate across the pigmented rabbit conjunctiva in the presence and absence of pharmacological agents known to affect active Cl- secretion and Na+ absorption. Fluid flow across a freshly excised pigmented rabbit conjunctiva mounted between two Lucite half chambers was measured by a pair of capacitance probes in an enclosed cabinet maintained at 37 degrees C and a relative humidity of 70%. Fluid transport was also measured in the presence of compounds known to affect active Cl- secretion (cAMP, UTP, and ouabain), Na+ absorption (D-glucose), or under the Cl--free condition on both sides of the tissue. Net fluid secretion rate across the pigmented rabbit conjunctiva in the serosal-to-mucosal direction at baseline was 4.3+/-0.2 microl/hr/cm2 (mean +/- s.e.m.). Net fluid secretion rate was increased approximately two-fold by mucosally applied 1 mM 8-Br cAMP (8.4+/-0.4 microl/hr/cm2) and 10 microM UTP (9.8+/-0.6 microl/hr/cm2), but was abolished by either serosally applied 0.5 mM ouabain (0.3+/-0.1 microl/hr/cm2) or under the Cl--free conditions (0.06+/-0.04 microl/hr/cm2). Mucosal addition of 20 mM D-glucose decreased net fluid secretion rate to 1.0+/-0.5 microl/hr/cm2. In conclusion, the pigmented rabbit conjunctiva appears to secrete fluid secondary to active Cl- secretion. This net fluid secretion is subject to modulation by changes in active Cl- secretion rate and in mucosal fluid composition such as glucose concentration.  相似文献
