Bt cotton (Cry1Ac) has been commercially grown in China since 1997, saving China's cotton production from attack by Bt‐target pests and also tremendously reducing pesticide usage. In recent years, however, Bt cotton, with 4.2 million ha of cultivation, has suffered from a secondary target pest, Spodoptera exigua (Hübner) (Lepidoptera: Noctuidae). In China, growers have even had to re‐adopt conventional pesticides to control the pest, and this practice has already caused serious pesticide residue. In order to clarify the sublethal effects of chemical pesticide, the responses of a Bt‐susceptible and a Bt‐tolerant (Bt10) S. exigua strain to three treatment combinations were examined, including Bt toxin, sublethal chlorpyrifos, and Bt + sublethal chlorpyrifos. The susceptible and the Bt10 strain responded differently to dual pressure. Bt toxin + sublethal chlorpyrifos treatment lowered larval mortality and stimulated population increase of the susceptible S. exigua, whereas it delayed growth and development of the Bt10 strain. Under dual pressure, although larvae of the Bt10 strain developed faster than larvae of the susceptible strain, the Bt10 population experienced higher larval mortality, prolonged pupal duration, decreased pupal weight, decreased emergence rate, and shortened adult longevity. Compared with the susceptible strain, the Bt10 strain was deleteriously affected by sublethal chlorpyrifos. The Bt‐tolerant/resistant S. exigua population was more vulnerable to chemical pesticides like chlorpyrifos regardless of whether it was exposed to Bt toxin or not. Our study provides a reference for increasing the efficacy of control of S. exigua in Bt‐cotton planting areas.  相似文献

Mythimna unipuncta Haworth (Lepidoptera: Noctuidae) is a well‐known moth species whose larvae can cause devastating damage to some Poaceae crops, including maize (Zea mays L.). The low susceptibility to the Bacillus thuringiensis Berliner (Bt) toxin observed in L6 larvae of this species has been the object of several studies. This study aimed to clarify whether the toxin eliminated from the content of the peritrophic membrane is degraded or excreted and whether the effects of the Bt toxin depend on the doses ingested. To this end, L6 larvae were fed on diets with different amounts of lyophilized Bt or non‐Bt maize leaves. The effect of the Bt concentrations on larval development was measured and the fate of the toxin in the larval tissues was tracked. Results indicated that the larvae of M. unipuncta fed on the various diets showed few differences in weight gain, duration of development, or pupal weight between sublethal Bt concentrations. The larvae rapidly excreted a large part of the toxin ingested, whereas inside the peritrophic membrane the toxin was eliminated, degraded, or sequestered at a rate that increased with the dose and the duration of feeding. As a consequence, little toxin reached the midgut epithelium and therefore the binding sites of the toxin. Moreover, larvae fed on the Bt toxin recovered quickly when they were transferred to a non‐Bt diet.  相似文献

外源蛋白在环境中的残留与积累是转Bt基因作物环境安全评价的重要内容之一。我国已育成多个具有商业化前景的Bt稻品系,但目前多年种植Bt稻后Bt外源蛋白是否会在土壤中积累还不清楚。本研究在同一试验田连续9年种植了转cry1Ab/1Ac基因明恢63(华恢1号)和转cry2A基因明恢63水稻,采用酶联免疫吸附法(ELISA)跟踪监测了分蘖期和收获后60 d根际土中外源蛋白含量变化,试验第1年(2012年)和最后1年(2020年)还测定了苗期、开花期和成熟期根际土中外源蛋白含量。结果表明: 2012年,转cry1Ab/1Ac基因明恢63在苗期、分蘖期、开花期、成熟期和收获后60 d根际土中外源蛋白含量分别为1.25、1.77、1.97、1.71和0.30 ng·g-1,2020年分别为1.30、1.69、2.03、1.77和0.43 ng·g-1;2012年,转cry2A基因明恢63在苗期、分蘖期、开花期、成熟期和收获后60 d根际土中外源蛋白含量分别为0.91、1.52、1.53、1.37和0.12 ng·g-1,2020年分别为0.95、1.43、1.61、1.40和0.15 ng·g-1。多因素方差分析显示,时间效应对Bt外源蛋白积累不显著,而品种和生育期效应显著。Bt稻生长过程中根际土中可以检测出微量的Bt外源蛋白,但收获后60 d已经基本降解完毕,根际土中Bt外源蛋白含量不会随着种植时间的增加而累积。  相似文献

转Bt基因玉米Bt11花粉对玉米螟赤眼蜂繁殖和存活的影响   总被引:7,自引:0,他引:7  
在室内评价了表达Cry1Ab杀虫蛋白的转基因抗虫玉米Bt11花粉作为食物对玉米螟赤眼蜂Trichogramma ostriniae雌蜂寿命、繁殖力、子代羽化数和性比的影响。结果显示,Bt11玉米花粉中含有115.13±7.29 ng/g FW Cry1Ab杀虫蛋白。饲喂水+Bt玉米花粉和水+对照玉米花粉的雌蜂寿命、繁殖力和子代羽化数明显高于只提供水的处理,但性比没有显著差异。饲喂10%蜂蜜水的雌蜂寿命、繁殖力和子代羽化数与饲喂10%蜂蜜水+Bt玉米或10%蜂蜜水+对照玉米花粉的处理雌蜂相似,没有显著差异。饲喂水+玉米花粉和只提供水的各处理的寿命、繁殖力和子代羽化数均显著低于蜂蜜水+玉米花粉和只提供蜂蜜水的各个处理,但性比显著高。饲喂水+Bt玉米花粉与水+对照玉米花粉之间以及蜂蜜水+Bt玉米花粉与蜂蜜水+对照玉米花粉之间的寿命、繁殖力和子代羽化数没有显著差异。由此得出结论,同只提供水相比,玉米花粉可以提高玉米螟赤眼蜂的寿命和繁殖力。表达Cry1Ab杀虫蛋白的Bt11玉米花粉对玉米螟赤眼蜂的寿命、繁殖力、子代羽化数和性比无不利影响。  相似文献

There is no conclusive evidence that Helicoverpa spp. (Lepidoptera: Noctuidae) in Australia have evolved significant levels of resistance to Bollgard II® cotton (which expresses two Bt toxin genes, cry1Ac and cry2Ab). However, there is evidence of surviving larvae on Bollgard II cotton in the field. The distribution and survival of early‐instar Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae) larvae were examined on whole Bollgard II and non‐Bt cotton plants in greenhouse bioassays. The expression of Cry toxins in various parts of Bollgard II plants was compared to the survival of larvae in those locations. Only 1% of larvae survived after 6 days on greenhouse‐grown Bollgard II plants compared to 31% on non‐Bt cotton plants. Overall, and across all time intervals, more larvae survived on reproductive parts (squares, flowers, and bolls) than on vegetative parts (leaves, stems, and petioles) on Bollgard II plants. The concentration of Cry1Ac toxin did not differ between plant structures, whereas Cry2Ab toxin differed significantly, but there was no relationship between the level of expression and the location of larvae. This study provides no evidence that lower expression of Cry toxins in the reproductive parts of plants explains the survival of H. armigera larvae on Bollgard II cotton.  相似文献

在室内对转基因水稻KMD1中的Cry1Ab毒蛋白经食物链在几种主要害虫及其捕食性天敌体内的积累进行了研究。结果表明: 无论是水稻孕穗期还是成熟期,二化螟Chilo suppressalis连续取食KMD1或取食KMD1.36 h后移至对照品种秀水11上取食不同时间后,幼虫体内的Cry1Ab含量均随取食时间延长逐渐下降。稻眼蝶 Mycalesis gotama幼虫连续取食KMD1或在KMD1上取食两天后移至秀水11上继续取食不同时间,体内的Cry1Ab含量也都随取食时间延长而下降,但下降速度较二化螟更快。取食KMD1的二化螟和稻眼蝶幼虫的粪便中均检测到较高浓度的Cry1Ab,对照组中均无Cry1Ab。取食KMD1的二化螟幼虫血淋巴中检测到Cry1Ab,含量为3.5 ng/g。取食KMD1的褐飞虱 Nilaparvata lugens、稻蚜Sitobion avenae以及饲喂取食过KMD1的二化螟或稻眼蝶幼虫的拟水狼蛛Pirata subpiraticus体内都含有一定浓度的Cry1Ab,其中,拟水狼蛛体内的CrylAb含量以饲喂取食KMD1稻眼蝶幼虫的含量最高,约为饲喂取食KMD1二化螟幼虫的60倍。这些结果表明Cry1Ab可以沿水稻害虫天敌食物链传递。  相似文献

Steroid alkaloids have been shown to elicit a wide range of pharmacological effects that include anticancer and antifungal activities. Understanding the biosynthesis of these molecules is essential to bioengineering for sustainable production. Herein, we investigate the biosynthetic pathway to cyclopamine, a steroid alkaloid that shows promising antineoplastic activities. Supply of cyclopamine is limited, as the current source is solely derived from wild collection of the plant Veratrum californicum. To elucidate the early stages of the pathway to cyclopamine, we interrogated a V. californicum RNA‐seq dataset using the cyclopamine accumulation profile as a predefined model for gene expression with the pattern‐matching algorithm Haystack. Refactoring candidate genes in Sf9 insect cells led to discovery of four enzymes that catalyze the first six steps in steroid alkaloid biosynthesis to produce verazine, a predicted precursor to cyclopamine. Three of the enzymes are cytochromes P450 while the fourth is a γ‐aminobutyrate transaminase; together they produce verazine from cholesterol.  相似文献

Mortality of western corn rootworm (Diabrotica virgifera virgifera LeConte) due to feeding on MON863 transgenic maize (Zea mays L.) expressing the Cry3Bb1 protein was evaluated at three Missouri sites in both 2003 and 2004 and at one site each in South Dakota, Nebraska and Iowa in 2004. To do this, survivorship relative to survivorship on isoline maize (i.e. the same genetic background, but without Cry3Bb1) was evaluated. Comparisons were made using low (1650–2500 eggs/m) and high (3300–3500 eggs/m) western corn rootworm egg densities. Significantly fewer beetles were recovered from MON863 than from isoline maize. Emergence from MON863 as a percentage of viable eggs ranged from 0.02% to 0.10%, whereas percentage emergence from isoline maize ranged from 1.09% to 7.14%. Survivorship on MON863 relative to survivorship on isoline averaged 1.51% when averaged across all environments and both years, so mortality because of the Cry3Bb1 protein averaged 98.49%. The average time delay to 50% cumulative beetle emergence from MON863 was 18.3 days later than from isoline maize. Females comprised 56% and 71% of total beetles recovered from MON863 in 2003 and 2004, respectively. Results are discussed in relation to insect resistance management (IRM) of western corn rootworm.  相似文献

To verify current thresholds for Bollgard II® cotton in Australia, the impact of Helicoverpa spp. (Lepidoptera: Noctuidae) larvae on yield, development, and quality under various infestation intensities and durations, and stages of growth, was tested using small plot field experiments over two seasons. Infestation with up to 80 eggs m?1 of Helicoverpa armigera (Hübner) and Helicoverpa punctigera Wallengren showed that species, infestation level, and stage of growth had no significant effect on yields of seed‐cotton or lint and on maturity and fibre quality. The duration of infestation of white flowers with H. punctigera neonates (maximum of every day for up to 4 weeks) had no impact on the yield of seed‐cotton or lint, maturity, and fibre quality, but when 100% of flowers were infested (compared with 0 or 50%), seed‐cotton and lint yields were significantly reduced and maturity was delayed. Infestation with up to 18 medium H. armigera larvae m?1 at several plant stages did not significantly affect yields of seed‐cotton and lint, maturity, and fibre quality. A heliocide spray applied on a commercial farm at the current threshold resulted in a significantly higher lint yield, compared with a farm where no spray was applied. In conclusion, Bollgard II® cotton is highly resistant to Helicoverpa spp. infestation.  相似文献

Nitrogen is a critical factor for plant development and nitrogen input is one of the important tactics to enhance the development and yield of crops. Nevertheless, nitrogen input could influence the occurrence of insects positively or negatively. Nitrogen is also one of the main elements composing the insecticidal crystal (Cry) protein. Cry protein production could affect nitrogen partitioning in Bt plants and as such nitrogen input may influence insect pest management in transgenic Bt rice, Oryza sativa L. (Poaceae). To test this possibility, we evaluated the impacts of nitrogen regimes on the main insect pests and their predators on two Bt rice lines, T2A‐1 and T1C‐19, expressing Cry2A and Cry1C, respectively, and their non‐transgenic parental counterpart MH63. The results showed that Cry proteins with different nitrogen regimes have enough insecticidal activity on rice leaffolder, Cnaphalocrocis medinalis Guenée (Lepidoptera: Crambidae), in both laboratory and field experiments. Laboratory studies indicated that relevant parameters of ecological fitness in brown planthopper, Nilaparvata lugens (Stål) (Hemiptera: Delphacidae), a non‐target insect pest, were significantly affected by nitrogen input both on Bt and MH63 rice lines. Nymphal survival, female adult longevity, and egg hatchability in N. lugens differed significantly among rice varieties. The experiments conducted in rice fields also demonstrated that nitrogen was positively correlated with the abundance of N. lugens on Bt rice, similar to that on MH63 rice. The abundances of two predators – the wolf spider Pirata subpiraticus (Boesenberg & Strand) (Araneae: Lycosidae) and the bug Cyrthorhinus lividipennis Reuter (Hemiptera: Miridae) – were significantly affected by rice growth stages but not by nitrogen input and rice varieties. In conclusion, the above results indicate that high nitrogen regimes for Bt rice (T2A‐1 and T1C‐19) and non‐Bt rice (MH63) cannot facilitate the management of insect pests.  相似文献

【目的】通过研究中华稻蝗Oxya chinensis几丁质脱乙酰基酶1 (chitin deacetylase 1, CDA1)基因的分子特性和生物学功能, 为新型农药靶标筛选提供理论基础。【方法】在中华稻蝗转录组数据库搜索获得几丁质脱乙酰基酶基因片段, 用生物信息学方法对其进行同源比对分析并作聚类关系图; 利用cDNA末端快速扩增(rapid-amplification of cDNA ends, RACE)获得该基因cDNA全长序列; 通过Real-time quantitative PCR(qPCR)方法检测OcCDA1在5龄若虫不同组织部位和不同发育时期的表达情况, 并采用RNAi技术研究OcCDA1在飞蝗生长发育中的功能。【结果】获得中华稻蝗几丁质脱乙酰基酶1基因全长cDNA,命名为OcCDA1(GenBank登录号:KP271171)。OcCDA1在前肠、体壁、后肠和脂肪体组织中高表达, 在5龄第1天表达量显著高于以后几天。RNAi结果显示, 注射该基因的dsRNA后, OcCDA1基因的表达量降低了93.3%。OcCDA1基因沉默后虫体出现进食减少、发育迟缓现象, 总死亡率达到95.2%,其中38.1%的试虫在蜕皮前死亡, 57.1%出现因蜕皮困难而死亡的表型。【结论】OcCDA1在中华稻蝗的发育中起着重要作用, 该基因沉默引起虫体进食减少和蜕皮异常从而导致死亡。  相似文献

The fall armyworm, Spodoptera frugiperda (J.E. Smith), is a major target pest of Bt crops (e.g., corn, cotton, and soybean) in North and South America. This pest has recently invaded Africa and Asia including China and the invasion has placed a great threat to the food security in many countries of these two continents. Due to the extensive use of Bt crops, practical resistance of S. frugiperda to Cry1F corn (TC 1507) with field control problems has widely occurred in Puerto Rico, Brazil, Argentina, and the mainland United States. Analyzing data generated from decade-long studies showed that several factors might have contributed to the wide development of the resistance. These factors include (1) limited modes of action of Bt proteins used in Bt crops; (2) cross-resistance among Cry1 proteins; (3) use of nonhigh dose Bt crop traits; (4) that the resistance is complete on Bt corn plants; (5) abundant in initial Cry1F resistance alleles; and (6) lack of fitness costs/recessive fitness costs of the resistance. The long-term use of Bt crop technology in the Americas suggests that Bt corn can be an effective tool for controlling S. frugiperda in China. IRM programs for Bt corn in China should be as simple as possible to be easily adopted by small-scale growers. The following aspects may be considered in its Bt corn IRM programs: (1) use of only “high dose” traits for both S. frugiperda and stalk borers; (2) developing and implementing a combined resistance monitoring program; (3) use “gene pyramiding” as a primary IRM strategy; and (4) if possible, Bt corn may not be planted in the areas where S. frugiperda overwinters. Lessons and experience gained from the global long-term use of Bt crops should have values in improving IRM programs in the Americas, as well as for a sustainable use of Bt corn technology in China.  相似文献

采用ELISA法研究了田间种植条件下转Bt基因玉米MON810生育期根际土壤及还田秸秆中Cry1Ab蛋白的田间残留降解动态,并分别用移动对数模型、指数模型和双指数模型对秸秆分解释放Cry1Ab蛋白的田间降解动态进行拟合,估算了DT5o和DT90值.结果表明:Bt玉米不同生育期根际土壤中Cry1 Ab蛋白含量差异较大,但总体随生育期的延长而显著降低.收获后地表覆盖和埋入土壤两种秸秆还田方式下,秸秆中Cry1Ab蛋白在土壤中的降解规律基本一致,均呈现前期大量快速降解,中后期极少量稳定降解两个阶段.秸秆还田7d内,地表覆盖处理的Cry1Ab蛋白降解率均极显著高于埋入土壤处理;10 d时两处理的降解率基本一致,分别为88.8%和88.6%;20 d后,两处理秸秆中Cry1Ab蛋白的降解日趋缓慢;至180 d时仍能检测到少量的Cry1Ab蛋白.3种模型均能较好地反映秸秆中Cry1Ab蛋白的田间降解规律,从相关系数(R)及DTgo值与实测值的吻合程度来看,双指数模型最优.  相似文献

Cotton fibers are single‐celled trichomes that initiate from the epidermal cells of the ovules at or before anthesis. Here, we identified that the histone deacetylase (HDAC ) activity is essential for proper cotton fiber initiation. We further identified 15 HDAC s homoeologs in each of the A‐ and D‐subgenomes of Gossypium hirsutum . Few of these HDAC homoeologs expressed preferentially during the early stages of fiber development [?1, 0 and 6 days post‐anthesis (DPA )]. Among them, GhHDA 5 expressed significantly at the time of fiber initiation (?1 and 0 DPA). The in vitro assay for HDAC activity indicated that GhHDA 5 primarily deacetylates H3K9 acetylation marks. Moreover, the reduced expression of GhHDA 5 also suppresses fiber initiation and lint yield in the RNA interference (RNA i) lines. The 0 DPA ovules of GhHDA 5 RNA i lines also showed alterations in reactive oxygen species homeostasis and elevated autophagic cell death in the developing fibers. The differentially expressed genes (DEG s) identified through RNA ‐seq of RNA i line (DEP 12) and their pathway analysis showed that GhHDA 5 modulates expression of many stress and development‐related genes involved in fiber development. The reduced expression of GhHDA 5 in the RNA i lines also resulted in H3K9 hyper‐acetylation on the promoter region of few DEG s assessed by chromatin immunoprecipitation assay. The positively co‐expressed genes with GhHDA 5 showed cumulative higher expression during fiber initiation, and gene ontology annotation suggests their involvement in fiber development. Furthermore, the predicted protein interaction network in the positively co‐expressed genes indicates HDA 5 modulates fiber initiation‐specific gene expression through a complex involving reported repressors.  相似文献

Proteins belonging to the enhancer of RNA interference‐1 subfamily of 3′–5′ exoribonucleases participate in divergent RNA pathways. They degrade small interfering RNAs (siRNAs), thus suppressing RNA interference, and are involved in the maturation of ribosomal RNAs and the degradation of histone messenger RNAs (mRNAs). Here, we report evidence for the role of the plant homologue of these proteins, which we termed ENHANCED RNA INTERFERENCE‐1‐LIKE‐1 (ERIL1), in chloroplast function. In vitro assays with AtERIL1 proved that the conserved 3′–5′ exonuclease activity is shared among all homologues studied. Confocal microscopy revealed that ERL1, a nucleus‐encoded protein, is targeted to the chloroplast. To gain insight into its role in plants, we used Nicotiana benthamiana and Arabidopsis thaliana plants that constitutively overexpress or suppress ERIL1. In the mutant lines of both species we observed malfunctions in photosynthetic ability. Molecular analysis showed that ERIL1 participates in the processing of chloroplastic ribosomal RNAs (rRNAs). Lastly, our results suggest that the missexpression of ERIL1 may have an indirect effect on the microRNA (miRNA) pathway. Altogether our data point to an additional piece of the puzzle in the complex RNA metabolism of chloroplasts.  相似文献

The Microrchidia (MORC) family proteins are important nuclear regulators in both animals and plants with critical roles in epigenetic gene silencing and genome stabilization. In the crop plant barley (Hordeum vulgare), seven MORC gene family members have been described. While barley HvMORC1 has been functionally characterized, very little information is available about other HvMORC paralogs. In this study, we elucidate the role of HvMORC6a and its potential interactors in regulating plant immunity via analysis of CRISPR/SpCas9-mediated single and double knockout (dKO) mutants, hvmorc1 (previously generated and characterized by our group), hvmorc6a, and hvmorc1/6a. For generation of hvmorc1/6a, we utilized two different strategies: (i) successive Agrobacterium-mediated transformation of homozygous single mutants, hvmorc1 and hvmorc6a, with the respective second construct, and (ii) simultaneous transformation with both hvmorc1 and hvmorc6a CRISPR/SpCas9 constructs. Total mutation efficiency in transformed homozygous single mutants ranged from 80 to 90%, while upon simultaneous transformation, SpCas9-induced mutation in both HvMORC1 and HvMORC6a genes was observed in 58% of T0 plants. Subsequent infection assays showed that HvMORC6a covers a key role in resistance to biotrophic (Blumeria graminis) and necrotrophic (Fusarium graminearum) plant pathogenic fungi, where the dKO hvmorc1/6a showed the strongest resistant phenotype. Consistent with this, the dKO showed highest levels of basal PR gene expression and derepression of TEs. Finally, we demonstrate that HvMORC1 and HvMORC6a form distinct nucleocytoplasmic homo-/heteromers with other HvMORCs and interact with components of the RNA-directed DNA methylation (RdDM) pathway, further substantiating that MORC proteins are involved in the regulation of TEs in barley.  相似文献

Experiments were conducted in small arenas and on whole plants to explore the effect of cotton aphids, Aphis gossypii Glover (Hemiptera: Aphididae), as alternative prey on the predation of Helicoverpa armigera Hübner (Lepidoptera: Noctuidae) larvae by green lacewing larvae, Mallada signatus Schneider (Neuroptera: Chrysopidae). Transgenic Bt (Bollgard II®) and conventional cotton plants were included to explore potential differences in the predator's performance on these cotton types. In small arenas, the presence of 20 aphids reduced predation on H. armigera larvae by 22% (from 5.5 to 3.3 of 10) by a single lacewing larva over a 24‐h period. The presence of H. armigera reduced predation on aphids by ca. 29% (from 16.8 to 11.0 of 20) over 24 h. On whole plants, the presence of alternative prey had no effect on the number of H. armigera larvae or aphids remaining after 3 days. The presence of H. armigera larvae alone, without the predator, caused a 24% reduction in the numbers of aphids on conventional, but not on Bt cotton plants. The combination of Bt cotton and lacewing larvae caused a 96.6% removal of early‐stage H. armigera larvae, a statistically significant increase over the addition of the proportions (91.6%) removed by each factor measured separately, providing evidence of synergism. These studies suggest that the presence of aphids as alternative prey would not necessarily disrupt the predation by green lacewing on larvae of H. armigera, especially on Bt cotton.  相似文献
