Comparative efficacy of the determination of the sensitivity of bacterial cells to barium ions was evaluated on a synthetic nutrient medium, FMH agar, Mueller-Hinton agar, and AGV agar. The synthetic nutrient medium developed for this study contained L-proline and L-glutamine as the sole nitrogen and carbon source, which promoted growth of all Pseudomonas strains and ensured the minimal level of barium binding. The sensitivity of 80 strains belonging to 11 Pseudomonas species, including the type strains, as well as of 80 strains of 22 other bacterial species, was studied. The sensitivity of bacteria to barium ions was determined by using serial dilutions of barium chloride in the nutrient medium. The highest level of analytical sensitivity of pseudomonads to barium ions was determined on the synthetic nutrient medium: the minimal inhibitory concentration (MIC) values of barium chloride ranged from 0.5 to 6 g/L, the MIC90 value was 2 g/L. At the same time, 86.1% of all strains of fluorescent Pseudomonas species produced fluorescein on the control BaCl2-free synthetic nutrient medium. For representatives of other genera grown on all the studied nutrient media, the MIC values of barium chloride ranged from 20 to 50 g/L. The proposed method for determination of the sensitivity of bacteria to barium ions using the synthetic nutrient medium with 6 g/L of barium chloride as a criterion for the classification of barium-sensitive strains to the genus Pseudomonas is suitable for standardization.  相似文献

Early blight (Alternaria solani) is an important disease causing severe damage in tomato. The eleven isolates of A. solani designated as So, Dh, Sh, Va-5, Ka, Ma, Hy, Ba-1, My, Va-3 and Mi were collected from different agroclimatic conditions and these isolates were characterized for cultural, morphological, pathogenic and molecular variations. The pigmentation varied from yellow, brown, black, brownish to greenish black in isolates of A. solani on potato dextrose agar medium. In general, radial growth of all isolates ranged between 14.9 mm and 32.2 mm on PDA and 24.3 mm to 53.7 mm on three selective media i.e., ASM, V-8 juice agar and V-8 juice agar (synthetic) on the fourth day. The fastest radial growth was recorded in the So isolate and slowest in the Ka isolate on PDA, while isolates Dh, Ba-1 and Va-3 were recorded to be faster in growth on ASM, V-8 juice agar and V-8 juice agar (synthetic) medium. The thickness of conidiogenous hyphae varied between 1.17 μ and 9.56 μ, with maximum in the Va-5 and Ma isolates. Most of the isolates showed smooth mycelial growth with circular and irregular margin and without concentric zonation. Sporulation was not found in any of the isolates on four different nutrient media, whereas conidiogenous hyphal length was observed in V-8 juice agar medium only. Based on the pathogenicity, isolates of A. solani were rated as virulent or less virulent based on percentage disease incidence data. Molecular variability studies were also done to find out the best annealing temperature and eighty-six primers were screened to select for maximum polymorphism of DNA. The best annealing temperature was recorded between 32.5 °C and 34.0 °C for the pathogen, and most efficient amplification and polymorphism of DNA was found with random primer 5′-CGCGTTCCTG-3′.  相似文献

A total of 28 species representing 15 fungal genera were found to be associated with the all different stages of the Egyptian cotton leafworm,Spodoptera littoralis Boisd., its faeces and the air of the containers where it was reared on Czapek Dox agar medium at 28°C.Aspergillus was the most common genus and was frequently isolated from all substrates.A. flavus was the only fungus associated with all insect stages. At least two well known facultative pathogens were obtained:Beauveria alba andCephaliophora tropica. Nearly all fungi recovered from insect faeces or its environment were also encountered from different insect stages. Forty-two isolates were tested for their abilities for growth on a synthetic medium (nitrogen- and carbon-free) containing partially purified chitin. Most (76.2%) had moderate growth rates.  相似文献

In order to assess the suitability of the Starch glutamate ampicillin penicillin-10C agar for the isolation ofAeromonas spp. from waters it was necessary to compare the properties of this medium with those of three others, Starch ampicillin agar, Ampicillin dextrin agar and m-Aeromonas medium, and to monitor different kinds of waters. A selection of forty eight samples were taken from moderately polluted river water, highly polluted river water, polluted sea water (littoral) and treatment & distribution water and monitored using these media. The results were similar with Ampicillin dextrin agar, m-Aeromonas medium and Starch glutamate ampicillin penicillin-10C, but the simplicity of composition and use and its selectivity recommends the last medium as the most adequate for the isolation ofAeromonas spp.Abbreviations ADA ampicillin dextrin agar - mA m-aeromonas medium - SA starch ampicillin agar - SGAP-10C starch glutamate ampicillin penicillin-10C  相似文献

Summary Alginase-secreting bacteria associated with actively growing tissues of the marine Phaeophyta speciesSargassum fluitans andS. natans have been isolated and evaluated for their ability to degrade alginate (ALG), carboxymethylcellulose, and agar. Of seven isolates selected for their ability to grow on 2% agar containing 1% sodium alginate, none were able to grow on either 2% agar or 2% agar supplemented with 0.1% carboxymethylcellulose. Two of these with fermentative potential, i.e., ALG-A and ALG-G, showed selective activities with respect to their ability to degrade native alginate and/or take up the products resulting from alginate degradation. The ALG-A isolate was able to rapidly degrade native alginate with the generation of a stable polymer fraction and small oligouronides, most of which were dissimilated for growth. The ALG-G isolate was able to completely degrade native alginate with the accumulation of significant quantities of unsaturated dimeric and trimeric oligouronides. A limit polymer was generated from the action of a polymannuronan-specific extracellular alginate lyase purified from exponential cultures of the ALG-A organism. This product proved to be an effective substrate for the alginate lyase activity obtained from the medium of exponential phase cultures of the ALG-G isolate, and upon incubation with concentrated and dialyzed ALG-G medium was converted to the products that were observed to accumulate in the medium of the ALG-G isolate grown on native alginate. These organisms represent examples of the microflora associated with actively growingSargassum tissues, each with a selective ability to degrade and dissimilate the biomass of the marine brown algae.  相似文献

Antibiotic susceptibility testing of Helicobacter pylori isolates was performed by broth microdilution method with MegaCellTM RPMI-1640 Medium (SIGMA). Fifty five clinical isolates of H. pylori were tested against metronidazole, tinidazole, amoxicillin, and clarithromycin. The results were compared to those obtained by standard agar dilution method. The microdilution method performed with new medium, showed excellent correlation with agar dilution results, with 100% agreement for metronidazole, 96.3% for amoxicillin, 90.7% for clarithromycin, and 92.8% for tinidazole. MICs determined by proposed method were highly reproducible: replicate results were variable within one-two-fold dilution by using different inocula and different batches of medium.  相似文献

Tubercle bacilli were preincubated with lecithin-cholesterol liposomes to be subsequently exposed to phospholipase A2. After further incubation in the environment of acidic buffer, viable units in the final mixture were enumerated by inoculating the serial dilutions of an aliquot onto Kirchner agar medium containing horse serum in 5%. Another aliquot was used for lipid analyses to confirm hydrolysis of lecithin. In addition to this bactericidal type of experiments, bacteriostatic tests were also conducted with Kirchner semi-solid agar medium, into which liposome-treated bacilli were inoculated with the enzyme at a time. Various natural and synthetic lecithins different in constituent fatty acids were employed. The results indicated that toxic fatty acids released from lecithin acted to kill the bacilli or to inhibit their growth.  相似文献

Guar gum, a galactomannan, has been reported to be an inexpensive substitute of agar for microbial culture media. However, its use is restricted probably because of (1) its highly viscous nature even at high temperatures, making dispensing of the media to Petri plates difficult and (2) lesser clarity of the guar gum gelled media than agar media due to impurities present in guar gum. To overcome these problems, three guar gum derivatives, carboxymethyl guar, carboxymethyl hydroxypropyl guar and hydroxypropyl guar, were tested as gelling agents for microbial growth and differentiation. These were also evaluated for their suitability for other routine microbiological methods, such as, enumeration, use of selective and differential media, and antibiotic sensitivity test. For evaluation purpose, growth and differentiation of eight fungi and eight bacteria grown on the media gelled with agar (1.5%), guar gum (4%) or one of the guar gum derivatives (4%), were compared. All fungi and bacteria exhibited normal growth and differentiation on all these media. Generally, growth of most of the fungi was better on guar gum derivatives gelled medium than on agar medium. The enumeration carried out for Serratia sp. and Pseudomonas aeruginosa by serial dilution and pour plate method yielded similar counts in all the treatments. Likewise, the selective succinate medium, specific for P. aeruginosa, did not allow growth of co-inoculated Bacillus sp. even if gelled with guar gum derivatives. The differential medium, Congo red mannitol agar could not differentiate between Agrobacterium tumefaciens and Rhizobium meliloti on color basis, if gelled with guar gum or any of its derivatives However, for antibiotic sensitivity tests for both Gram-positive and -negative bacteria, guar gum and its derivatives were as effective as agar.  相似文献

Aims: The aim of this study was to develop a modified selective medium to improve the recovery rate of Brachyspira hyodysenteriae and other clinically significant intestinal spirochaetes from porcine faeces. Methods and Results: The susceptibility of five Brachyspira spp. type strains and five Thai field isolates of B. hyodysenteriae to the antimicrobials halquinol and flavomycin was determined by in vitro susceptibility tests in the agar dilution method, and optimal incorporation rates were confirmed by broth dilution. All the spirochaetes were susceptible to halquinol at ≤1 μg ml?1, while 16 μg ml?1 of flavomycin (F) allowed their growth, and therefore, only the latter was selected for further use. F and different combinations of colistin (C), spectinomycin (S) and rifampacin (R) were incorporated into pre‐enrichment broths and/or agar plates, and growth of the spirochaetes from seeded faeces was determined. Two solid media were selected for further testing using faeces from 90 finishing pigs on 10 farms. A previously recommended method of pre‐enrichment did not increase the recovery rate. The use of blood agar modified medium (BAM) containing F (16 μg ml?1), S (400 μg ml?1), R (30 μg ml?1) and colistin (C, 100 U ml?1) (assigning as BAM‐CSRF) reduced the growth of contaminating intestinal microbiota and resulted in a significantly higher rate of spirochaete recovery than the previous recommended medium. Conclusion: BAM‐CSRF is a useful new selective medium for the isolation of B. hyodysenteriae and other intestinal spirochaetes from pig faeces. Significance and Impact of the Study: The new selective medium for isolating B. hyodysenteriae and other Brachyspira spp. from pig faeces will improve their recovery and subsequent disease diagnosis.  相似文献

Summary 1) A simple technique for the preparation of dissolved autoclaved antigens and of sonic vibrated antigens, the immunisation of rabbits, and testing methods by precipitation in agar medium and in capillary tubes are described.2) Five different strains ofSporotrichum belonging to three species,S. Schenckii, S. Beurmanni, andS. asteroides are studied as to their precipitin lines formation in agar medium.3) The similarity in precipitin lines formation is interpreted to demonstrate the antigenic identity of these differentSporotrichum species.4) Comparable results with minor differentiation but in major sensitivity are obtained by precipitin tests in capillary tubes.5) Temperatures of 4° C are shown to stimulate intensity of precipitin lines.6) Dissolved sonic vibrated antigens alter the precipitin formation by changing and intensifying the precipitin lines.7) Absorption experiments with dissolved antigens give unsatisfactory results if precipitin tests in capillary tubes are used.This work was done during a Fulbright Fellowship spent with Dr.Norman F. To develop a rapid reporter system for the screening of stationary-phase promoters in Escherichia coli, the expression pattern of the green fluorescent protein (GFP) during bacterial cultivation was compared with that of the commonly used β-galactosidase. Using GFP with enhanced fluorescence, the expression pattern of both reporter systems GFP and β-galactosidase were similar and showed a typical induction of gene activity of the reporter genes, i.e. increase of expression at the transition from exponential to stationary phase. The expression was affected by the culture medium, i.e. in contrast to the complex medium (LB medium), the stationary-phase specific induction was only observed in synthetic medium (M9) when amino acids were added, whereas there was generally no induction in MOPS medium. To develop a rapid screening method on agar plates for stationary-phase promoters, a photographic approach was used, continued with computational image treatment. A screening method is presented which enables an on-line monitoring of gene activity.  相似文献

The emergence of methicillin-resistant Staphylococcus aureus (MRSA) is responsible for nosocomial and community-acquired infections. Hence, rapid and accurate laboratory diagnosis of MRSA is a vital constituent of control measures. The present study evaluated five different methods for the identification of MRSA. A total of 207 S. aureus clinical isolates that consisted of 89 MRSA and 118 methicillin-susceptible S. aureus (MSSA) strains confirmed by PCR were tested. MRSA strains were evaluated by five different methods: chromogenic MRSA agar (CMRSA), oxacillin resistance screening agar base (ORSAB), mannitol salt oxacillin agar (MSO), mannitol salt cefoxitin agar with two different concentrations of cefoxitin [4 μg/ml (MSC-4) and 6 μg/ml (MSC-6)]. The results of the different methods were compared to mecA PCR as the gold standard. MSC-6 showed only six false-positive MRSA in comparison with PCR. The sensitivities and specificities of MSC-6, MSC-4, MSO-4, ORSAB, and CMRSA were as follows: 98.9/94.9%, 100/83.1%, 89.9/87.3%, 97.8/96.6%, and 95.5/94.9%, respectively. In comparison with PCR, it was found that both MSC-6 and ORSAB were relatively the least expensive screening tests (0.70 and0.70 and 1.00, respectively). In conclusion, all methods were comparable, but MSC-6 was the least expensive medium for MRSA screening.  相似文献

Summary Studies were carried out to evaluate sugarcane bagasse as an alternative to agar for micropropagation of apple clones to reduce the cost of micropropagation and improve the quality of the propagules. Significant improvement in the in vitro rooting process, coupled with cost reduction, were obtained by the use of sugarcane bagasse as a substitute for the traditionally used agar-gelled medium. The tests were undertaken with micro-cuttings of the apple rootstock Marubakaido (Malus prunifolia Borkh.) using a rooting medium composed of half-strength Murashige and Skoog salts and vitamins, 3% (w/v) sucrose, and 0.49 μM indole-3-butyric acid. The plants grown on sugarcane bagasse yielded a 22% increase in root length, 20% increase in plant length, and 63% increase in the number of roots, compared with agar-grown micro-cuttings. Particle size of the sugarcane bagasse had a significant impact on all those parameters, and the best results were obtained with bagasse comprising particles smaller than 0.18 mm. The results demonstrated that the sugarcane bagasse could be used effectively as a substitute for agar during rooting of apple shoots.  相似文献

为确定峨眉山金蝉花孢梗束培养的最佳工艺条件,对金蝉花孢梗束生长的常用培养基配方进行筛选试验,选用大米、小米、黄豆等原料进行配方试验,采用均匀设计试验法优化培养基配方。结果表明,金蝉花在以无机盐为主要营养成分的Richard琼脂培养基和Czapek琼脂培养基上均能产生孢梗束。在大米等的配方试验中,以大米为营养物质的金蝉花孢梗束生长最快,金蝉花氨基酸含量为23.4%,粗多糖含量为26.5%,蛋白质含量为29.3%,均高于野生金蝉花中该物质的含量。在均匀设计的配方试验中,10号培养基中的金蝉花孢梗束以珊瑚状为主,菌体形态最好,孢梗束产率可达57.4%。10号配方孢梗束中蛋白质的含量高达42.5%,氨基酸含量高达33.3%,均远高于野生金蝉花。  相似文献

S ummary . Two methods of viable counting have been compared for unirradiated and γ irradiated Escherichia coli NCTC 5933. The spread plate method has been shown to be more reproducible than a serial dilution method in a liquid medium, but the survivor-dose curves of γ irradiated bacteria obtained by the two methods were identical. The spread plate method was used to compare the survival of γ irradiated E. coli on peptone agar and on a synthetic glucose-salts agar over a range of survivor levels of c. 9 log cycles. The log % survivor-dose curve was linear for the synthetic medium but had a double exponential shape for the peptone agar. Recovery was much higher on peptone than on a synthetic medium. Using a tube dilution method in a liquid glucose-salts medium, 19 l -amino acids were tested for their effect on recovery. The slight increases in number of survivors attributable to some amino acids were associated with a change in the first part (100–1% survivors) of the curve rather than an alteration in final slope. The phosphate concentration of the solid synthetic medium markedly affected its ability to recover γ irradiated cells.  相似文献

Using synthetic substrates, an uncomplicated and sensitive procedure for the determination of extracellular aminopeptidase was developed. The studied enzyme produced by the tested plant material (calli, cell suspension culture and roots of Amsonia tabernaemontana Walt. seedlings) hydrolyzed the substrates β‐naphthylamides (βNA) and 4‐(phenylazo) phenylamides (PAP‐amide) of the amino acids to β‐naphthylamine and 4‐(phenylazo) aniline, respectively, and amino acid. The β‐naphthylamides of the amino acids were applied for the identification of extracellular aminopeptidase, whereas the 4‐(phenylazo) phenylamides of the amino acids were used for the determination of intra‐ and extracellular aminopeptidase activity. By simultaneous azocoupling of β‐naphthol with Fast Garnet GBC salt on agar plates a corresponding brown‐red hardly water‐soluble azo‐dye was produced. The evaluation of dyed zones allowed the extracellular aminopeptidase activity to be assessed. No coloration of the agar medium was observed without inoculum, with heat‐inactivated cells (10 min at 100 °C) or in medium inoculated without substrate. On the agar plates with substrate and sterile Amsonia seedlings, changes in coloration were observed indicating a release of aminopeptidase from the roots during germination. The results show a 91.0 % intracellular and 9.0 % extracellular distribution of aminopeptidase activity, when a cell suspension culture of A. tabernaemontana Walt. as the plant material was used. The agar plate method described permits the rapid, uncomplicated and specific detection of plant producers of extracellular aminopeptidase, which could be particularly useful in future inhibitory and/or biotechnological studies.  相似文献

Results of minimal inhibitory concentration tests with a diversity of bacterial strains showed that 9-chloro-9-(4-diethylaminophenyl)-10-phenylacridan (C-390) inhibited the growth of all microorganisms tested (other than Pseudomonas aeruginosa) at 25 microgram/ml or less, whereas MICs obtained for P. aeruginosa ranged from to to greater than 100 microgram/ml. Therefore, C-390 was evaluated as a potential selective agent for P. aeruginosa in pseudomonas agar F. Recovery tests were conducted on this medium with 53 strains o P. aeruginosa, and the results were compared to those obtained in similar tests on commercially available selective media, i.e., pseudomonas isolation agar and Pseudosel agar. The results of these comparisons indicated that pseudomonas agar F with C-390 was significantly less inhibitory than Pseudosel agar and pseudomonas isolation agar and more selective than pseudomonas isolation agar. The incorporation of C-390 in pseudomonas agar F also provided a medium that was both selective and differential. Preliminary evidence also suggested that C-390 may be added to other basal media with comparable results.  相似文献

A simple, rapid and reliable outline for identification of clostridia isolates from human infections was developed. It consists of a combination of API ZYM and API LRA Oxidase tests. The enzymatic activities were performed with strains sub-cultured onto carbohydrate-free medium (Columbia blood agar). Fifty-five strains of Clostridium difficile, C. bifermentans, C. sordellii, and C. perfringens from clinical specimens and eight reference standard strains representing different species of the same genus were analyzed. The accuracy of the new method was evaluated by comparison with the results obtained by DNA/DNA analysis.  相似文献

Summary Agitated layers of liquid medium were created on platform shakers in jars with 25–30 ml of medium (similar to conventional agar culture) rotating at 90 rpm. Thin films were scaled up in larger rectangular vessels on tilted shelves that periodically rock. In jars of liquid medium with a density of 180 explants per liter, multiplication rates of Hota tokudama var. ‘Newberry Gold’ were optimal with a media sucrose concentration of 5% [both with and without 1 μM benzyladenine (BA)]. Endogenous levels of soluble sugars were directly related to the concentration of sucrose in the medium. Three Hosta cultivars (‘Striptease’, ‘Minuteman’, and ‘Stiletto’) with plant densities of 40–200 explants per liter of medium were tested in larger, agitated, thin-film vessels in media with 5% sucrose and directly compared to agar medium. Higher rates of multiplication were observed in liquid than agar with the magnitude of the difference dependent on explant density. Pooled results for the three varieties with 200 explants per liter showed multiplication rates of 1.7x and 2.3x for agar and thin-film liquid, respectively. At 40 explants per liter, the multiplication rate was increased to 2.1x for agar and 3.4x for thin-film liquid. Sugar uptake was greater in liquid than agar and was greater in the higher densities, with the magnitude of the effect dependent on plant variety. Increased vessel size in the liquid, thin-film system and greater sugar uptake allowed more, larger plants to be harvested. Alocasia macrorrhizos was cultured in growth medium containing 1μM BA and 5% sucrose with plant densities in the range of 33–330 explants per liter. Dry weight and multiplication rate were greater in the liquid system than agar with the magnitude of the difference dependent on plant density. With approximately 165 explants per liter, and greater at the initiation of culture, plant density limited growth in both agar and liquid thin-film systems. In a multiplication medium (3 μM BA and 3 μM ancymidol) plant size was reduced by 50% and 60% (fresh weight) in liquid and agar, respectively. Initial density in the range of 165–330 explants per liter did not limit growth with the smaller plants in liquid or semisolid multiplication medium. Sugar uptake was greater in liquid than agar. While ample sugar was present in media for growth at any density on agar, sugar depletion was limiting growth at highest densities with the larger plants in liquid growth medium. In semisolid agar medium, sugar uptake by plants was more rapid than diffusion across the agar medium, resulting in non-equilibrium conditions following the culture cycle. In agitated, liquid medium, a greater transfer of sugars to plant tissue was related to accelerated growth.  相似文献
