A cluster of three pyritized, naturally arranged, conoidal sclerites has been found on a bedding surface in the lower Cambrian Chapel Island Formation in southeastern Newfoundland. The elements are about 4 mm in length and are morphologically similar to protoconodonts but neither their original chemical composition nor internal structure is preserved. Arrangement of the elements suggests that they belonged to an apparatus that performed a grasping function. The specimen is described here as ProtohertzinaEfficacynadia n.sp. This is the oldest hitherto described protoconodont apparatus, apart from clusters of Mongolodus described from the lower Cambrian of the Lesser Himalaya. This paper adds to the scant knowledge of early protoconodonts that we consider to be the earliest biomineralized predators.  相似文献

Following the advent of digital subtraction angiography (DSA), carbon dioxide gas has become a useful contrast agent for arterial angiography. Former manual injection methods had precluded accurate dosing and reproducibility. An original gas injector was therefore developed and tested in a circulatory system model. This permitted an accurate evaluation of vascular gas-filling. it also proved possible to measure the injection parameters taking different influencing factors into account. It was shown that vessels up to 10 mm in diameter are virtually completely filled with gas. In vessels larger than 10 mm in diameter, a residual fluid level remains, which in turn reduces the possibility for complete vessel imaging. The injection flow is dependent primarily on the vascular circulation rate, vessel diameter and the inclination of the vessel. With respect to other factors examined, neither catheter size, number of holes nor the direction of the injection have any relevant influence.  相似文献

The ability to detect viable contaminants in cultures propagated from the original host-expression system ensures that the integrity and purity of seed banks, fermentation broths, and ultimately the final product are continually controlled and maintained. The method developed to detect such agents must be selective for a broad spectrum of microbes, which may be present at very low levels, while discriminating from the host organisms. Although Pichia pastoris strains are frequently used as cell lines for the expression of heterologous proteins, a method that is specific for monitoring culture purity has yet to be reported for this type of organism. An assay that is capable of recovering contaminating bacteria, fungi, and closely related yeast from cultures of P. pastoris at parts per million detection limits is described here.  相似文献

Consistent asymmetric folding of the commissure is a characteristic feature of a small but significant number of brachiopod species. The feature may be obligate or facultative and is almost entirely confined to rhynchonellids, most of which are Mesozoic. The detailed nature of the asymmetry is very variable, but does not extend to internal hard parts such as crura. Taken as a whole, asymmetric brachiopod species show no preference for any particular environment or geographic region, and in no circumstances seem to have been markedly more or less successful than symmetric species. We are thus led to suggest that asymmetry was a genetically based condition which cropped up periodically in brachiopod evolution, and which possibly was selected neither particularly for nor against.  相似文献

A very simple and rapid GC-MS procedure for the separation and quantitation of - and -serine has been developed utilizing a conventional bonded-phase capillary column. The procedure involves initial esterification with isobutanol followed by acylation with the chiral derivatizing reagent S-(−)-N-(heptafluorobutyryl)prolyl chloride (HPC). This procedure requires neither extraction nor clean-up steps and is sensitive to 50 pg on-column. Total time of the procedure is under 3 h and derivatives are stable at room temperature for at least 5 days, making this procedure ideal for automated injections. A simple, one-day synthesis of HPC is described which yields >99.9% optical purity.  相似文献

Imaging can be thought of as the most direct of experiments. You see something; you report what you see. If only things were truly this simple. Modern imaging technology has brought about a revolution in the kinds of questions we can approach, but this comes at the price of increasingly complex equipment. Moreover, in an attempt to market competing systems, the microscopes have often been inappropriately described as easy to use and suitable for near-beginners. Insufficient understanding of the experimental manipulations and equipment set-up leads to the introduction of errors during image acquisition. In this feature, I review some of the most common practical pitfalls faced by researchers during image acquisition, and how they can affect the interpretation of the experimental data.This article is targeted neither to the microscopy gurus who push forward the frontiers of imaging technology nor to my imaging specialist colleagues who may wince at the overly simplistic comments and lack of detail. Instead, this is for beginners who gulp with alarm when they hear the word "confocal pinhole" or sigh as they watch their cells fade and die in front of their very eyes time and time again at the microscope. Take heart, beginners, if microscopes were actually so simple then many people (including myself) would suddenly be out of a job!  相似文献

Summary The previous simple model for treating concerted evolution of multigene families has been revised to be compatible with various new observations on the immunoglobulin variable region family and other families. In the previous model, gene conversion and unequal crossing-over were considered, and it was assumed that genes are randomly arranged on the chromosome; neither subdivision nor correlation of gene identity and chromosomal distance were considered. Although this model satisfactorily explains the observed amino acid diversity within and between species, it fails to predict the very ancient branching of the mouse immunoglobulin heavy chain V-gene family. By incorporating subdivided structure and genetic correlation with chromosomal distance into the simple model, the data of divergence may be satisfactorily explained, as well as the rate of nucleotide substitution and the amino acid diversity. The rate at which a V-gene is duplicated or deleted by conversion or by unequal crossing-over is estimated by the new model to be on the order of 10–6 per year. We have expanded previous observations on olfactory metabolic enzymes by examining the content of various metabolic enzymes in the olfactory mucosa of the male Long-Evans rat at different ages. Age-related changes in metabolic enzyme content may be related to changes in susceptibility to toxicants with age and may also contribute to altered odorant perception in the elderly. While some enzymes did not vary over the age range examined, decreases in the microsomal content of other enzymes were observed. While mRNA for acetyltransferase enzymes has previously been described in olfactory mucosa, the markedly higher activity of olfactory acetyltransferases compared to liver had not previously been described. Acetyltransferases are important in the metabolism of drugs and toxicants that are aromatic amine derivatives and may contribute to the bioactivation of rodent olfactory mucosal carcinogens such as 2,6-dimethylaniline and alachlor. These studies show that the olfactory mucosa varies in its metabolic capacity with age, and characterize another class of metabolic enzymes in the olfactory mucosa, both of which may impact significantly on responses to toxicants and therapeutic agents in the nasal cavity.  相似文献

Triazine herbicides are among the most heavily used agricultural pesticides. Although they possess a very low acute toxicity in animals, a mammary tumor response has been consistently observed in Sprague-Dawley (SD) female rats following chronic oral dosing of atrazine and simazine at and above maximum tolerated doses. However, a substantial collection of detailed research has clearly shown that triazines are not genotoxic or mutagenic, nor do they possess estrogenic agonist activity that might promote mammary tumor growth. Examination of estrous cycling records of atrazine-treated SD rats revealed a premature appearance of persistent estrous episodes, beyond the prevalent occurrence normally seen in untreated, aging SD rats. A significant correlation has been found between early or severe estrous cycle disruption of atrazine-treated rats and the early appearance of mammary tumors. In studies using SD female rats fed atrazine for 6 months, then ovariectomized and administered an estrogen-containing silastic s.c. implant, a deficient luteinizing hormone surge was observed at a 400 parts per million (ppm) dose, but not at 25 or 50 ppm. Because SD rats exhibiting persistent estrus also have a prolonged elevation of estrogen secretion, it is proposed that the triazine-associated mammary tumor response is promoted by the test animal's own estrogen from ovarian follicles that fail to ovulate because gonadotropin surge sufficiency is blocked by the high dose of herbicide. It is further proposed that, because reproductive senescence in SD rats is fundamentally different from menopause in women, the animal response to dosing, as well as the enormous requisite dosing level, establishes a safety margin of very low risk to human health from this mode of action.  相似文献

The fine structure of mitochondria from the ventricular myocardium of canaries, sparrows, zebra finches, quail, and geese has been studied. The first three of these birds have very fast heart rates, the quail being intermediate, and the goose has a relatively slow rate. The canary heart has a unique form of mitochondrion containing large, parallel arrays of zigzag or angled cristae. Other cristae, continuous with the zigzag ones and also occupying large parts of the mitochondrial volume, are named retiform because of the hexagonal network which they form, sometimes in a single plane and sometimes three dimensional. These two types of cristae appear to be interconnectible. It is possible that there is a direct functional significance in these peculiar forms, but, in any case, the relative constancy of dimensions in these arrays is probably related to specific properties of the molecules of which the cristal membrane is composed. It is also demonstrated that this membrane is composed in part of approximately 30-A particles which are believed to be protein molecules. This unusual mitochondrial morphology is not seen either in the other fast bird hearts or in the slower ones, so that there is neither a simple correlation with heart rate nor probably with the separate parts of the cardiac cycle. Although none of the other four hearts shows more than an occasional angled crista, there does seem to be a rather gross correlation between heart rate and mitochondrial size and complexity of crista structure, but no correlation with presence or absence of zigzag forms. The cristae of quail heart mitochondria are disposed in unusually large close-packed whorls.  相似文献

Laboratory toxicity tests generally expose organisms to a constant concentration of a toxicant in a uniform environment. The two techniques outlined in this paper assess the impact of a model toxicant, copper, on marine fouling assemblages in the field. These techniques allow natural assemblages to develop under a regime of toxicant dosing. The experiments were conducted using both techniques over time frames ranging from 2 weeks to 6 months. Short term experiments using the techniques assess the effects of toxicant exposure on recruitment. The longer term experiments enabled detection of certain indirect impacts of pollution. These include effects due to competition for space and variations in the susceptibility of organisms with age. Over the course of the experiments, a range of responses to copper was observed for different taxa. Both techniques have the potential to be used for testing the impact of other toxicants. Approaches such as those described are necessary in order to understand how communities react to toxicants in real world situations.  相似文献

Although stressing polymers have been widely and successfully used to determine the osmotic properties of solutes in aqueous media, the osmotic stress method presents some limitations. To overcome these drawbacks, an alternative and more direct method, which has been named the osmomanometer, is described in this letter. The osmotic pressure accessible by this method ranges typically from 1 to 30 kPa using a simple hydrostatic effect and can be extended to higher pressures by using pressurized gas. This method needs neither a pressure sensor nor calibration.  相似文献

There is a rich literature on automatic species identification of a specific target taxon as regards various vocalizing animals. Research usually is restricted to specific species – in most cases a single one. It is only very recently that the number of monitored species has started to increase for certain habitats involving birds. Automatic acoustic monitoring has not yet been proven to be generic enough to scale to other taxa and habitats than the ones described in the original research. Although attracting much attention, the acoustic monitoring procedure is neither well established yet nor universally adopted as a biodiversity monitoring tool. Recently, the multi-instance multi-label framework on bird vocalizations has been introduced to face the obstacle of simultaneously vocalizing birds of different species. We build on this framework to integrate novel, image-based heterogeneous features designed to capture different aspects of the spectrum. We applied our approach to a taxon-rich habitat that included 78 birds, 8 insect species and 1 amphibian. This dataset constituted the Multi-label Bird Species Classification Challenge-NIPS 2013 where the proposed approach achieved an average accuracy of 91.25% on unseen data.  相似文献

The tumor-inherent vasculature plays a major role with respect to tumor sensitivity to ionizing radiation. Ultrastructural studies of the tumor vasculature are necessary in order to obtain more detailed information on the architecture and structure and on the sites affected by tumor therapy. The vasculature of xenotransplanted human tumors on nude mice was investigated in this study by means of scanning and transmission electron microscopy. Structurally complete, real arteries or veins are neither to be seen in the periphery nor in the center. Large-caliber vessels basically have a capillary wall structure. The endothelial cells show a very alternating height and electron density. Frequently, different cell types and tumor cells themselves are apparently involved in the formation of the vessel wall. The endothelium is characterized by very simple, immature cell contacts. In some tumors, the amount of vessels without or with incomplete endothelium seems to be higher than the number of structurally real capillaries. This has consequences as well for radiotherapy as for hyperthermia and chemotherapy.  相似文献

Oil bodies are lipid storage organelles which have been analyzed biochemically due to the economic importance of oil seeds. Although oil bodies are structurally simple, the mechanisms involved in their formation and degradation remain controversial. At present, only two proteins associated with oil bodies have been described, oleosin and caleosin. Oleosin is thought to be important for oil body stabilization in the cytosol, although neither the structure nor the function of oleosin has been fully elucidated. Even less is known about caleosin, which has only recently been described [Chen et al. (1999) Plant Cell Physiol 40: 1079–1086; Næsted et al. (2000) Plant Mol Biol 44: 463–476]. Caleosin and caleosin-like proteins are not unique to oil bodies and are associated with an endoplasmatic reticulum subdomain in some cell types. Here we review the synthesis and degradation of oil bodies as they relate to structural and functional aspects of oleosin and caleosin.  相似文献

Summary Pigeon muscles lacking muscle spindles were grafted into sites which normally have a muscle containing spindles. The reciprocal transplantations were also made. After two to eight months, the graft of the donor muscle without spindles had regenerated into a muscle containing muscle spindles. The reciprocal grafts, muscles containing spindles transplanted to a site lacking spindle innervation, had neither muscle spindles nor remnants of the spindles. These experiments demonstrate that 1) the innervation is required for formation of the spindle; 2) the original spindles do not survive transplantation; and 3) parts of the original spindle are not required for spindle regeneration.This work was supported in part by NSF grants PCM 77-15960 and PCM 79-16540  相似文献

A simple method for assaying the mitochondrial carnitine/acylcarnitine translocase of muscles that needs only few milligrams of fresh tissue is described. The procedure involves monitoring of the sulphobetaine (an inhibitor of the translocase)-sensitive acetylation of sub-saturating concentrations of carnitine in the medium, linked to the oxidation of [2-14C]pyruvate in the presence of malonate. Conditions affecting the reliability of the outlined procedure and the ancillary information to be collected, namely the activities of pyruvate oxidase system and carnitine acetyltransferase, for detecting possible deficiency of the translocase are described, together with data on the translocase activity in human skeletal muscle, in rat red and white skeletal muscles and in rat heart. The concepts outlined should allow development of assays of other mitochondrial transporters that also would require neither isolation of mitochondria nor availability of a large quantity of tissue, both of which are otherwise needed at present.  相似文献
