目的:研究共轭亚油酸甘油酯对高脂饮食大鼠血清脂肪酸组成变化的影响;方法:60只SD大鼠随机分为5组:正常对照组、高脂对照组,共轭亚油酸甘油酯低(2 g/kg·bw)、中(4 g/kg·bw)、高(6 g/kg·bw)剂量组,除基础对照组外其余各组均喂饲高脂饲料,建立高脂模型,以灌胃方式给予受试物,6周后取血清测定其脂肪酸组成。采用一步法直接对血清中脂肪酸进行甲酯化,气相色谱毛细管柱分离,氢火焰离子化检测器(FID)定性定量分析;结果:共轭亚油酸甘油酯低、中、高剂量组大鼠血清中单不饱和脂肪酸含量为25.66%,18.74%,17.72%,与高脂对照组相比显著性下降(P0.01),各剂量组饱和脂肪酸和多不饱和脂肪酸含量分别为48.08%,48.52%,51.15%和27.03%,29.28%,31.13%,与高脂对照组相比显著性升高(P0.01)。几种代表性脂肪酸如各剂量组中的油酸、亚油酸与高脂对照组相比分别增加了26.48%,41.56%,51.26%和9.18%,8.61%,8.73%,各剂量组中棕榈酸与高脂对照组相比降低了5.28%,8.80%,10.92%。结论:共轭亚油酸甘油酯能够增加大鼠血清中饱和脂肪酸和多不饱和脂肪酸的含量,减少单不饱和脂肪酸含量,改变血清脂肪酸组成。  相似文献

摘要:目的:了解不同哺乳期母乳中各种脂肪酸水平的差异及其变化趋势,为母乳喂养及婴幼儿配方奶粉的配制提供科学依据。方法:收集2015年7月—2016年3月,在北京大学人民医院足月顺产的25~40岁产妇产后1~5d的初乳、6~10d的过渡乳和30d的成熟乳123份,采用气相色谱质谱联用法(GC-MS)检测母乳中的脂肪酸成分,分析母乳中各脂肪酸水平的差异及随时间的变化趋势。结果:随着泌乳时间的延长,C18∶0、C14∶1ω5、C18∶3ω6水平和ω-6/ω-3比例呈增高趋势;C24∶0、C16∶1ω9、C20∶1ω9、C20∶2ω6、C20∶3ω6、C20∶4ω6(AA)、C22∶2ω6、C22∶4ω6、C22∶5ω6、C22∶5ω3和C22∶6ω3(DHA)水平呈下降趋势,总体SFA含量逐渐增加,但无显著性意义;而PUFA 尤其是ω-3 PUFA水平显著减少(P<0.05)。 结论:母乳中的各种脂肪酸随着泌乳时间的延长而不断变化,新生儿喂养及配方奶粉的配制时应考虑母乳中各种脂肪酸含量的微观变化。  相似文献

共轭亚油酸(Conjugated linoleic acid,CLA)具有抗癌、抗动脉粥样硬化、减肥和免疫调节等生理活性。共轭亚油酸可以通过酶法异构化获得,将底物亚油酸异构形成具有生物活性物质-共轭亚油酸的异构酶称为亚油酸异构酶。因此,通过介绍亚油酸异构酶的来源、作用机制、酶学性质和基因工程菌生产等方面的研究进展,结合不断发展的基因工程技术,旨在提高亚油酸异构酶的活性、产量和异构化效率,以扩大反应底物范围,降低生产成本,从而推进共轭亚油酸的规模化、可持续性的工业生产。  相似文献

目的:分析西安市冠心病(CHD)患者膳食脂肪酸供能比与血脂水平的相关关系。方法:选取西安市三级甲等医院的CHD患者;采用膳食频率调查表,根据每日食物摄入种类及数量计算各种膳食脂肪酸供能比;测定体格指标和空腹血脂水平;采用协方差分析探讨CHD患者膳食脂肪酸供能比与血脂水平的相关关系。结果:本次调查共纳入符合要求的研究对象152名,平均年龄60.91±10.94岁,饱和脂肪酸(SFA)、单不饱和脂肪酸(MUFA)和多不饱和脂肪酸(PUFA)供能比分别为7.9%±2.4%、16.6%±7.8%、11.4%±5.6%。在校正混杂因素后,膳食MUFA供能比与血清ApoA呈正相关(P<0.05);低密度脂蛋白胆固醇(LDL C)和低密度脂蛋白胆固醇/高密度脂蛋白胆固醇(LDL C/HDL C)比值随膳食MUFA供能比上升而下降(P<0.05)。膳食SFA和PUFA供能比分别与LDL C、HDL C等间无显著相关性(P>0.05)。结论:CHD患者膳食MUFA供能比与血脂水平相关,提示提高膳食MUFA供能比,对改善CHD患者血脂水平具有重要作用。  相似文献

妊娠期糖尿病(GDM)是妊娠期常见的一种并发症。由于妊娠孕妇所处阶段特殊性,强调防治措施首先以调整饮食为主。GDM孕妇在发病机理方面存在着胰岛素抵抗和胰腺β细胞凋亡现象,脂肪酸诱导胰岛素抵抗在GDM发生发展中作用目前仍存在一定争议。本文综述膳食饱和脂肪酸、多不饱和脂肪酸、单不饱和脂肪酸及反式脂肪酸与GDM关系的研究进展,为GDM孕妇摄入合理膳食脂肪酸提供科学依据。  相似文献

观察侧脑室注射共轭亚油酸(Conjugated linoleic acid,CLA)对SD大鼠糖脂代谢的影响及其可能的机制。方法:向正常SD大鼠侧脑室内注射共轭亚油酸(CLA),分别于注射后2、4、8、12、24小时采血,试剂盒法测血糖、胰岛素、瘦素、血甘油三脂、血胆固醇、血高密度脂蛋白。48小时后处死,分离动物的脂肪(皮下、内脏、肾周、睾周)进行称重,计算体脂比。结果:与对照组相比,侧脑室注射CLA48小时后,大鼠脂体比、皮下脂肪、睾周脂肪均下降。术后2-12h血糖降低,血清胰岛素浓度也降低,而且持续的时间较长(48h)。侧脑室注射CLA对脂代谢有影响,2h时血清甘油三酯升高、胆固醇降低、4h时高密度脂蛋白升高。结论:共轭亚油酸能够通过中枢神经系统调节外周糖脂代谢,这可能与其能减轻体重的机制有关。  相似文献

以胶束电动色谱法对奶样中共轭亚油酸主要的两种异构体进行了分析。在优化条件下(80 mM pH9.0的磷酸盐缓冲液,54 mMSDS,4%(w/v)β-CD,8 M尿素,4%(v/v)乙醇作为运行缓冲液,分离电压25 kV,柱温20℃),胶束电动色谱可在15 min内对奶样中两种主要CLA,即9c,11t-CLA和10t,12c-CLA进行分离测定,最低检出限为0.081 ng/mL。分析结果显示,不同处理奶样中的CLA含量差异显著(P<0.001),但CLA的组成相近,其中的10t,12c-CLA含量差异不显著P=0.999,约为3%;不同品种奶样,如牛奶、水牛奶和羊奶中的CLA含量差异显著(P<0.001),其中CLA含量次序为牛奶>羊奶>水牛奶,并且不同品种奶的9c,11t-CLA与10t,12c-CLA比例差异显著(P<0.05)。  相似文献

目的建立高脂血症大鼠模型,分析共轭亚油酸(CLA)对其脂质代谢和visfatin基因表达水平的影响,为进一步研究CLA对脂肪代谢的调控机制奠定基础。方法用高脂饲料饲喂雄性Wistar大鼠,4周后断尾采血测定血清甘油三酯(TG)、总胆固醇(CHO)和血糖(GLU)含量,将构建成功的高脂血症大鼠分为实验组和对照组,实验组每天定时灌胃CLA(0.8 mL/0.1 kg),每周定时称重,记录采食量;4周后眼球采血,测定血清中GLU、CHO、TG、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)水平;断颈处死大鼠,分离体脂并提取肝脏总RNA,半定量RT-PCR分析visfatin基因表达水平。结果高脂饲料饲喂4周后,模型组大鼠血清TG、CHO、GLU明显高于对照组大鼠(P〈0.05),表明高脂大鼠模型构建成功。灌胃CLA 4周后,实验组大鼠体重、体脂和采食量低于对照组大鼠(P〈0.05),血清中GLU、CHO、TG、LDL含量明显降低,但实验组HDL浓度升高。RT-PCR实验结果表明,实验组大鼠visfatin基因表达水平明显低于对照组大鼠(P〈0.05)。结论CLA能降低高脂血症大鼠摄食量,改善高血脂大鼠脂质代谢,并能降低visfatin基因的表达水平。  相似文献

In two experiments with growing-finishing pigs six different dietary fats were added to a conventional diet (control - C) to study the effects of dietary monounsaturated (MUFA) and polyunsaturated fatty acids (PUFA) on the fatty acid composition of backfat and kidney fat at similar amounts of double bonds in feed (Exp. 1:7% pork fat - PF, 4.95% olive oil - OO, 3.17% soybean oil - SO) or a constant amount of 5% of processed fats (Exp. 2: partially hydrogenated fat - SAT, fractionated pork fats: olein - OLE, stearin - STE). Compared with the control, PUFA were only slightly increased in backfat of pigs fed PF, OLE, STE or OO, although dietary PUFA intake was up to 70% higher. With SO PUFA were significantly increased in adipose tissues, predominantly at the expense of MUFA. Consequently, a non-linear relationship was found between PUFA intake and proportion in backfat. MUFA were incorporated at the expense of SFA, therefore, adipose tissues of OO fed animals were lowest in SFA. Despite comparable amounts of double bonds in feed (Exp. 1), the degree of unsaturation measured as fat score (sum of double bonds) was in the order SO > OO > PF > C. In contrast, the proportion of SFA was C > PF = SO > OO. Regarding the decisive role of SFA for fat consistency it may be concluded that MUFA should also be considered in feeding recommendations for pigs. Furthermore, in case of a high dietary supply of MUFA, a simple index of double bonds might not be sufficiently conclusive to judge pig fat quality.  相似文献

Dramatic shifts in the Western diet have led to a marked increase in the dietary intake of the n-6 polyunsaturated fatty acid (PUFA), linoleic acid (LA). Dietary LA can then be converted to arachidonic acid (ARA) utilizing three enzymatic steps. Two of these steps are encoded for by the fatty acid desaturase (FADS) cluster (chromosome 11, 11q12.2-q13) and certain genetic variants within the cluster are highly associated with ARA levels. However, no study to date has examined whether these variants further influence pro-inflammatory, cyclooxygenase and lipoxygenase eicosanoid products. This study examined the impact of a highly influential FADS SNP, rs174537 on leukotriene, HETE, prostaglandin, and thromboxane biosynthesis in stimulated whole blood. Thirty subjects were genotyped at rs174537 (GG, n = 11; GT, n = 13; TT, n = 6), a panel of fatty acids from whole serum was analyzed, and precursor-to-product PUFA ratios were calculated as a marker of the capacity of tissues (particularly the liver) to synthesize long chain PUFAs. Eicosanoids produced by stimulated human blood were measured by LC-MS/MS. We observed an association between rs174537 and the ratio of ARA/LA, leukotriene B4, and 5-HETE but no effect on levels of cyclooxygenase products. Our results suggest that variation at rs174537 not only impacts the synthesis of ARA but the overall capacity of whole blood to synthesize 5-lipoxygenase products; these genotype-related changes in eicosanoid levels could have important implications in a variety of inflammatory diseases.  相似文献

通过皂化法和尿素包合法对热榨、冷榨栝楼(Trichosanthes kirilowii)籽油进行提取,采用气相色谱-质谱联用(GC-MS)技术检测分析栝楼籽油中亚麻酸、亚油酸成分、含量。结果表明,热榨栝楼籽油中亚麻酸相对含量为4.16%~11.58%,亚油酸相对含量为68.62%~95.84%。冷榨栝楼籽油中不含亚麻酸成分。此方法适用于栝楼籽油的成分分析。  相似文献

Forty Large White pigs were fed from 30kg to 103kg body mass on diets supplemented with 6% of pure high-oleic sunflower oil (HO) or HO plus increasing amounts of partially hydrogenated rape seed oil (HR; 1.85%, 3.70%, 5.55%), containing high levels of j 6 to j 11 C 18:1 trans fatty acid isomers. Increasing dietary C 18: trans fatty acids resulted in a linear increase in C 18:1 trans fatty acids and conjugated linoleic acid (cis-9, trans-11 CLA) in backfat (BF) as well as in neutral lipids (NL) and phospholipids (PL) of M. long. dorsi. Thus, the rate of bioconversion of trans vaccenic acid (TVA) into CLA and incorporation of C 18:1 trans and CLA into pig adipose tissue was not limited up to 25g total C 18:1 trans fatty acids including 3.3g of TVA perkg feed. BF was higher in C 18:1 trans fatty acids and CLA than M. long. dorsi NL and PL. In BF and NL the sum of saturated fatty acids (SFA) increased with increasing dietary amounts of HR, while in PL SFA were reduced. Thus, according to their physical properties, C 18:1 trans fatty acids partly replaced SFA in PL. Firmness of backfat was also significantly increased (P<0.05) with increasing amounts of HR in feed.  相似文献

We showed previously in cultures of primary human adipocytes and preadipocytes that lipopolysaccharide and trans-10,cis-12-conjugated linoleic acid (10,12-CLA) activate the inflammatory signaling that promotes insulin resistance. Because our published data demonstrated that preadipocytes are the primary instigators of inflammatory signaling in lipopolysaccharide-treated cultures, we hypothesized that they played the same role in 10,12-CLA-mediated inflammation. To test this hypothesis, we employed four distinct models. In model 1, a differentiation model, CLA activation of MAPK and induction of interleukin-8 (IL-8), IL-6, IL-1β, and cyclo-oxygenase-2 (COX-2) were greatest in differentiated compared with undifferentiated cultures. In model 2, a cell separation model, the mRNA levels of these inflammatory proteins were increased by 10,12-CLA compared with bovine serum albumin vehicle in the adipocyte fraction and the preadipocyte fraction. In model 3, a co-culture insert model, inserts containing ∼50% adipocytes (AD50) or ∼100% preadipocytes (AD0) were suspended over wells containing AD50 or AD0 cultures. 10,12-CLA-induced IL-8, IL-6, IL-1β, and COX-2 mRNA levels were highest in AD50 cultures when co-cultured with AD0 inserts. In model 4, a conditioned medium (CM) model, CM collected from CLA-treated AD50 but not AD0 cultures induced IL-8 and IL-6 mRNA levels and activated phosphorylation of MAPK in naive AD0 and AD50 cultures. Consistent with these data, 10,12-CLA-mediated secretions of IL-8 and IL-6 from AD50 cultures were higher than from AD0 cultures. Notably, blocking adipocytokine secretion prevented the inflammatory capacity of CM from 10,12-CLA-treated cultures. These data suggest that CLA instigates the release of inflammatory signals from adipocytes that subsequently activate adjacent preadipocytes.  相似文献

Changing the dietary ratio of the essential fatty acids (EFA), 18:2n6 and 18:3n3, while keeping the amounts of other fatty acids in the diet constant can rapidly and specifically alter the proportions of n6 and n3 22-carbon fatty acids in the brain of the weanling rat. A dietary 18:2n6/18:3n3 ratio of 165 versus 1.8 caused higher n6 and lower n3 22-carbon fatty acid levels, without changing total 22-carbon fatty acid levels, in phosphatidylethanolamine and phosphatidylcholine from several neural membrane fractions. This was apparent after only 2 weeks and showed no sign of plateauing after 12 weeks. Other neural fatty acids were essentially unaffected. The three most abundant 22-carbon fatty acids responded somewhat differently to increments in the dietary 18:2n6/18:3n3 ratio (1.8, 9, 36, and 165). Levels of 22:4n6 increased by similar absolute amounts for each four-fold increase in dietary 18:2n6/18:3n3 ratio; in contrast, the largest absolute changes in 22:5n6 and 22:6n3 levels occurred as the 18:2n6/18:3n3 ratio increased from 36 to 165. This study shows that the 18:2n6/18:3n3 ratio of diets high in fat (40% of energy) and adequate in EFA, both typical of diets in developed countries, can substantially and relatively quickly affect the 22-carbon fatty acids in the brain, even after the rapid accumulation of these fatty acids during neural growth has ceased.  相似文献
