A study was conducted to characterize patterns of mobilizationand translocation of seedpiece nitrogen (N) from single-eyeseedcores cut from 5 and 17-month-old potato seed-tubers. Differencesin mobilization efficiency were related to age-induced, morphologicaldifferences in plant development. Seedcores from older seed-tuberssprouted earlier and produced an average of 6.6 shoots per eyecompared to a single shoot from younger seed-tubers. Shoot vigour(d. wt per shoot) from 17-month-old was four-fold lower thanthat from 5-month-old seed-tubers following 25 d of growth.However, total shoot dry matter from older seedcores was two-foldgreater than that from younger seedcores. Differences in vigourper shoot were not explained entirely by differences in shootnumber. Rates and absolute amounts of free amino, soluble protein andtotal-N mobilized from 17-month-old seedcores were greater thanfrom younger seedcores. However, a higher degree of intersproutcompetition from older seedcores translated into a lower amountof mobilized N available to support growth of individual shoots.Furthermore, before seedcore N became limiting, concentrationof foliar N (mg g d. wt–1) from older seedcores was lowercompared to that from younger seedcores, indicating a lowersink strength for N per unit increase in dry weight of the multipleshoots. Seedpiece N did not appear to limit shoot growth fromyounger seedcores. Age-induced loss of vigour on an individualshoot basis may thus be related to decreased sink strength andincreased competition among multiple shoots for seedcore N. During the early stages of growth, developing sprouts rely heavilyupon tuber carbohydrate reserves. Strong sprouts are producedfrom young potato (Solaman tuberosum L.) seed-tubers, whereastubers of advanced physiological age produce sprouts with reducedvigour. Single-eye seedcores from 7- and 19-month-old seed-tuberswere sprouted in order to study the effect of tuber age on carbohydratereserve mobilization. Following 24 d of growth, the same amountof total shoot dry matter was produced from both ages of seedcores.However, d. wt of individual shoots from older seedcores was93 % lower than that from younger cores, reflecting a significantloss in apical dominance with age. Furthermore, core d. wt lossper unit gain in plant d. wt was greater from 19-month-old seedcores,indicating an age-related decrease in the efficiency of reservemobilization over the growth interval. During sprouting, ratesof starch hydrolysis and subsequent increases in total solublecarbohydrates were similar for both tuber ages. Reducing sugars(mg g d. wt–1) accumulated faster in older than in youngercores; non-reducing sugars (mainly sucrose) decreased 53% inolder seedcores over the 24 d growth interval. Non-reducingsugar content (mg g d. wt–1, mg core–1) of youngercores remained constant. Growth potential of potato (Solanum tuberosum L.) plants is influenced by seed-tuber age. After 24 days of growth, single-eye seedcores from 7-month-old seed-tubers produced 64% more foliar dry matter than those from 19-month-old seed-tubers, reflecting a higher growth rate. This study was initiated to determine if differences in polyamine (PA) metabolism are associated with aging and age-reduced vigor of potato seed-tubers. As tubers aged in storage, putrescine (Put) increased 2.2-fold, while spermidine (Spd) and spermine (Spm) decreased 33% and 38%, respectively. Ethylene content of the tuber tissue also increased with advancing age, suggesting that during the aging process S-adenosylmethionine was directed toward ethylene biosynthesis at the expense of the PAs. Single-eye cores from 7- and 19-month-old tubers were sown and PA levels in core and shoot tissues were monitored during plant development. Put titer of younger cores increased 8.8-fold by 12 days. In contrast, the increase in Put over the initial titer in older cores was 2.9-fold. The reduced ability of older cores to synthesize Put during plant establishment is probably due to a 45% decline in ornithine decarboxylase activity between 12 and 16 days after planting. Lack of available Put substrate limited the biosynthesis of Spd and Spm, and thus their concentrations remained lower in older cores than in younger cores. Lower PA titer in older cores during plant establishment is thus coincident with reduced growth potential. Concentrations of Put and Spd were higher in shoots developing from older cores throughout the study, but there was no age-related difference in Spm content. In contrast, activities of arginine and S-adenosylmethionine decarboxylases were higher in shoots from younger cores during establishment. The results indicate that aging affects PA metabolism in both tuber and developing plant tissues, and this may relate to loss of growth potential with advancing seed-tuber age.  相似文献

In vitro treatment of microsomal membranes from the axes ofsoybean (Glycine max (L.) Merr.) seeds with free radicals simulatesthe type of membrane injury observed following a lethal desiccationstress—the accumulation of free fatty acids in the membranebilayer, the loss of lipid-P, and the formation of gel phasedomains. The major phospholipids in the microsomal fractionwere phosphatidylcholine, phosphatidylethanolamine and phosphatidylinositol.Although these treatments induced an extensive loss of totalphospholipid from the microsomal fraction following desiccation,the ratio of the major phospholipids remained unchanged. Neitherlysophosphatides nor phosphatidic acid accumulated in the fraction,but free fatty acid levels increased. Therefore, cleavage ofboth acyl chains from the phospholipid molecule occurred followingdesiccation of the axes and in vitro free radical treatmentof the membrane. Both treatments also promoted formation of gel phase domainsas shown by wide angle x-ray diffraction and increased microviscosityas determined by the fluorescent probe, DPH (1,6-diphenyl-1,3,5-hexatriene).This could be simulated in liposomes prepared from the totalmicrosomal lipid fraction by the addition of saturated freefatty acids (16:0 and 18:0) at the levels observed followingstress. In contrast, the addition of unsaturated fatty acidsperturbed the bilayer and reduced microviscosity. The inclusionof both saturated and unsaturated free fatty acids as observedin vivo promoted a response similar to that observed with onlythe addition of the saturated free fatty acids. Desiccation of the axes also promoted a loss of microsomal protein,which was recovered in the 165 000 x g supernatant, and an apparentloss of thiol groups from the membrane as determined by a thiolspecific fluorescence probe, dansylaziridine. This loss of thiolgroups could also be simulated by exposure of the membranesto gamma irradiation, which was used as a non-enzymatic sourceof free radicals. Collectively, these data support the hypothesisthat membrane disassembly following desiccation stress is mediatedby a free radical mechanism, and that the consequent de-esterificationof membrane phospholipid and accumulation of saturated freefatty acids alter the physical properties of the membrane. Small amounts of a mixture of fatty acid short‐chain‐alkyl esters (FASCAEs) were obtained from the fruits of twelve plant species of Celastraceae family, and in five of them the FASCAEs were present not only in the arils but also in the seeds. These mixtures contained 32 individual FASCAE species, which formed four separate fractions, viz. FA methyl, ethyl, isopropyl, and butyl esters (FAMEs, FAEEs, FAIPEs, and FABEs, resp.). The FASCAE acyl components included the residues of 16 individual C14–C24 saturated, mono‐, di‐, and trienoic FAs. Linoleic, oleic, and palmitic acids, and, in some cases, also α‐linolenic acid predominated in FAMEs and FAEEs, while myristic acid was predominant in FAIPEs. It can be suggested that, in the fruit arils of some plant species, FAMEs and FAEEs were formed at the expense of a same FA pool characteristic of a given species and were strongly different from FAIPEs and FABEs esters regarding the mechanism of their biosynthesis. However, as a whole, the qualitative and quantitative composition of various FASCAE fractions, as well as their FA composition, varied considerably depending on various factors. Therefore, separate FASCAE fractions seem to be synthesized from different FA pools other than those used for triacylglycerol formation.  相似文献

H+-ATPase was solubilized from the tonoplast of mung bean (Vignaradiata L.) hypocotyls and purified by fast protein liquid chromatographyon a Mono Q ion-exchange column. The purified ATPase hardlycontained any phospholipid, but it did contain 10 to 15 moleculesof sterol and 25 to 30 molecules of glycolipid per ATPase molecule,and it had little activity without exogenously added phospholipids.Each individual polar head group, acylglyceride and fatty acidthat constituted a phospholipid was incapable by itself of activatingthe ATPase. Sterols and cerebroside had little activating effect.Maximal activation of ATPase was noted with asolectin or variousmolecular species of phosphatidylcholine (PC) at 0.005% to 0.01%(w/v). The activation by the various molecular species of PCwas dependent on the length and degree of unsaturation of fattyacyl chains. PC with two saturated and long fatty acyl chainsof more than 18 carbon atoms failed entirely to activate theATPase. PC, PS and PG with 1-palmitoyl (16:0)-2-oleoyl(18:1)fatty acyl chains all activated ATPase to nearly the same extentas asolectin, but the activation by PE and PA with the samefatty acyl composition was 52% and 15% of that by asolectin,respectively. The molecular species of PC with phase-transitiontemperatures below 50C activated ATPase, as determined at 38C.The dependence on temperature of the activation by the molecularspecies of PC indicated that the activation of the ATPase beganclose to the temperature of the phase transition of the PC added.These data indicate that phospholipids in the liquid-crystallinephase are essential for the catalytic activity of the ATPase. The effect of advanced meristem age on growth and accumulationof plant nitrogen (N) in potato (Solanum tuberosum L.) was studied.Etiolated plantlets, excised from sprouted, single-eye-containingcores from 7 and 19-month-old seed-tubers, were transplantedinto aerated nutrient culture. Rates of shoot and root dry matterand shoot soluble-N (which included nitrate-N) accumulationwere similar for plants from both meristem ages over a 30 dinterval of log-linear growth. The rate at which nitrate-N accumulatedwas consistently 17 per cent higher in shoots from 19-month-oldcompared to those from 7-month-old meristems. However, accumulationof free amino-N and soluble protein-N were 21 and 15 per centlower, respectively in shoots from 19-month-old meristems. Abuild-up of shoot nitrate, along with lower rates of accumulationof amino-N and soluble protein-N, suggests a lower capacityfor nitrate reduction during early growth of plants from oldermeristems. Furthermore, these effects can be attributed to age-inducedchanges in the meristem or bud tissue as the plants were separatedfrom the tuber tissue initially in the study. Long-term ageingof seed-potatoes apparently affects changes within meristemsthat translate into a lower capacity to accumulate reduced formsof nitrogen during early plant growth. The effect of seed-tuber age on the ability of tuber tissue to synthesize protein during sprouting was examined. As seed-tuber age advanced from 4 to 32 months (at 4°C, 95% relative humidity), soluble protein concentration of tubers decreased linearly, with a concomitant increase in free amino acid concentration. The age-induced loss of tuber protein may thus be due to increased proteolysis, decreased protein synthesis, or both. Five- and 17-month-old seed-tubers were compared for their ability to incorporate radiolabeled amino acids into soluble protein at equivalent stages of sprout development. Tuber respiration was profiled through each sprouting stage to characterize the physiological status of the seed-tubers prior to incorporation studies. Five-month-old seed-tubers maintained a constant rate of respiration during sprouting. In contrast, respiration of 17-month-old tubers increased as sprout dry matter increased, resulting in a 2- to 3-fold greater respiratory rate from the older tubers, relative to the younger tubers, at similar stages of sprout development. Prior to sprouting, the rate of incorporation of amino acids into trichloroacetic acid-precipitable protein of tissue from 5-month-old tubers was 2. 9-fold higher than that from 17-month-old tubers. More importantly, protein-synthetic capacity of tissue from younger tubers increased about 1. 7-fold during sprout development. Despite the higher respiratory activity and faster total sprout dry matter accumulation from older seed-tubers, protein synthesis remained at a low and constant level through all stages of sprouting. Protein-synthetic capacity thus declines with advancing tuber age, and this may contribute to reduced growth potential during the latter stages of establishment by affecting the ability of seed-tubers to synthesize enzymes involved in mobilization and translocation of tuber reserves to developing plants.  相似文献

Seasonal changes in the fatty acid composition of phospholipids (PL), monoglycerides (MG), diglycerides (DG), free fatty acids (FA) and triglycerides (TG) separated from oleosomes (lipid bodies) of perennial root nodules of beach pea (Lathyrus maritimus) were analysed. Thin layer chromatography (TLC) revealed that PL and MG are the major lipids in nodule oleosomes. The fatty acid profile and overall double bond index (DBI) varied among lipid classes depending upon the season. High DBI in PL and MG found during late winter and early spring indicated that they may play a major role in winter survival and regeneration of perennial nodules. The DBI of DG was high at the end of the fall season and the DBI of FA and TG was high in summer months. The dominant fatty acids are C16:0 followed by C18:0 and C18:1. The levels of many unsaturated fatty acids such as C18:1, C18:2 and C18:3 increased while saturated fatty acid C18:0 decreased during winter. These unsaturated fatty acids possibly play an important role in the protection of nodule cells from cold stress. Nodules seem to retain some fatty acids and selectively utilize specific fatty acids to survive the winter and regenerate in spring.  相似文献

The membrane of vesicular stomatitis virus (VSV) contains two distinct pools of phosphatidylethanolamine molecules which reside in the inner and outer phospholipid monolayers, respectively. 36% of the total membrane phosphatidylethanolamine is found in the outer monolayer while 64% is found in the inner. The two pools of VSV phosphatidylethanolamine can be distinguished operationally by the fact that only outer phosphatidylethanolamine is reactive in intact virions with the membrane-impermeable reagent trinitrobenzenesulfonate (TNBS). We have made use of this property to separate inner from outer VSV phosphatidylethanolamine and to determine the fatty acyl chain compositions of the two phosphatidylethanolamine pools separately. The results show that compared to outer phosphatidylethanolamine, inner phosphatidylethanolamine molecules contain a significantly higher proportion of unsaturated fatty acyl chains. Furthermore, whereas the proportion of unsaturated fatty acyl chains was found to be quite similar at the 1 and 2 glycerol carbon atoms in inner phosphatidylethanolamine, a marked dissimilarity was observed in outer phosphatidylethanolamine; outer phosphatidylethanolamine was enriched in saturated fatty acyl chains at the 1 position and in unsaturated fatty acyl chains at the 2 position. The differential fatty acyl chain composition of inner compared to outer phosphatidylethanolamine indicates that rapid, random transmembrane migration (flip-flop) of phosphatidylethanolamine does not occur in the VSV membrane. The gene encoding a 10-kDa acyl-CoA-binding protein (ACBP) from Brassica napus was over-expressed in developing seeds of Arabidopsis thaliana . Biochemical analysis of T2 and T3 A. thaliana seeds revealed a significant increase in polyunsaturated fatty acids (FAs) (18:2 cis Δ9,12 and 18:3 cis Δ9,12,15) at the expense of very long monounsaturated FA (20:1 cis Δ11) and saturated FAs. In vitro assays demonstrated that recombinant B. napus ACBP (rBnACBP) strongly increases the formation of phosphatidylcholine (PC) in the absence of added lysophosphatidylcholine in microsomes from ΔYOR175c yeast expressing A. thaliana lysophosphatidylcholine acyltransferase ( AthLPCAT ) cDNA or in microsomes from microspore-derived cell suspension cultures of B. napus L. cv. Jet Neuf. rBnACBP or bovine serum albumin (BSA) were also shown to be crucial for AthLPCAT to catalyse the transfer of acyl group from PC into acyl-CoA in vitro . These data suggest that the cytosolic 10-kDa ACBP has an effect on the equilibrium between metabolically active acyl pools (acyl-CoA and phospholipid pools) involved in FA modifications and triacylglycerol bioassembly in plants. Over-expression of ACBP during seed development may represent a useful biotechnological approach for altering the FA composition of seed oil.  相似文献

The growth-associated development of the m. pectoralis superficialis (MPS) phospholipid (PL) and triacylglycerol (TAG) fatty acyl (FA) chain composition was determined in BUT8 meat-type turkeys. Samples (3 d, 8, 12, 16 and 20 wk) of each 6 males were analysed by lipid fractionation and subsequent gas chromatography. Results were interpreted on an allometric basis. The MPS mass increased linearly (MPS weight = 0.2787 BW- 123.67; R2 = 0.9935, P<0.001, n = 30). In the total phospholipids 62-63% unsaturated fatty acids were found irrespective of the diet. A negative allometric alteration was found for the total saturated acyl chains (B = -0.012), while a positive value for the calculated unsaturation index (B = 0.026) was obtained. Within the PUFA chains, the n3- n6 balance was markedly changed, on the favour of the n3 fatty acyl chains, namely competitive allometric trends were found for the total n3 (B = 0.087) and n6 (B = 0.032) fatty acid groups. The alterations of the TAG FA chain composition were diet-dependent. The serum creatine kinase activity increased by over one class of magnitude during the trial. The allometric approach was found to be powerful in the characterization of the basic, non diet-dependent ontogenic alterations of the phospholipid fatty acyl chain composition.  相似文献

The effects of high alpha-linolenate content on lipid peroxidation, oxidative stress and loss of plant growth potential during ageing of potato (Solanum tuberosum L.) seed-tubers was examined. Endoplasmic reticulum (FAD3) and plastidal (FAD7) 18:2 fatty acid desaturases were upregulated in potato (cv. Desiree), resulting in a 2-fold average increase in mol percentage 18:3 in the total lipid fraction across all transgenic clones. In double-transformed (FAD3+7) tubers, high alpha-linolenate phenotype effected accelerated ageing, resulting in growth responses characteristic of older seed-tubers. Although respiration rates of wild-type (WT) and FAD3+7 tubers were equal at 7 months of storage, rates had increased by 23% and 50% in WT and FAD3+7 tubers, respectively, by 19 months of storage. Electrolyte leakage of tissue from 19-month-old FAD3+7 tubers was significantly greater than that from WT tubers of the same age, indicating that the high alpha-linolenate phenotype was detrimental to membrane integrity during long-term storage. On average, indices of lipid peroxidation (malondialdehyde, ethane, C-6 aldehydes) were higher in older FAD3+7 tubers, relative to WT tubers. Activities of glucose-6-phosphate dehydrogenase, peroxidase, glutathione reductase, ascorbate peroxidase and monodehydroascorbate reductase increased in tubers with advancing age and were higher, on average, in FAD3+7 tubers. Dehydroascorbate reductase activity decreased with age, with no difference between transgenic and WT lines. Collectively, these results indicate that FAD3+7 tubers underwent a higher degree of oxidative stress during ageing. The age-induced increase in respiration of FAD3+7 tubers was at least partly a response to fuel increased free radical scavenging through the ascorbate-glutathione antioxidant pathway. By affecting the susceptibility of lipids to peroxidation, the degree of fatty acid unsaturation influenced the development of oxidative stress and the overall rate at which growth potential was lost from seed-tubers during ageing. Thus, oxidative stress plays an integral role in modulating the ageing process to affect growth potential from potato seed-tubers.  相似文献

The thermotropic behavior of a series of synthetic fatty acyl ethylesters (FAEE) in multilamellar liposomes has been studied by differential scanning calorimetry and monitoring the changes in polarization emitted by the fluorescent probe 1,6-diphenyl-1,3,5-hexatriene. Their thermotropic behaviour has been compared to that of the homologous fatty acids (FA) from which they are synthesized in vivo in the presence of ethanol. Compared to the correspondent FA, saturated FAEE show, depending on the chain length, a minor rigidifying effect or even a slight fluidizing effect on phospholipid bilayers. Unsaturated FAEE show, compared to the homologous FA, slightly greater fluidizing properties. The difference between FA and FAEE is more evident in single component phospholipid liposomes in the gel phase, and in mixed liposomes of two lipids at temperatures at which microdomains of gel and liquid zones coexist. The calorimetric data suggest that FAEE are completely miscible with phospholipids both in the gel and liquid phases; they appear to destabilize the bilayer wherein the ethoxy head group interferes with the intrinsic phospholipid interactions.  相似文献

Seven day old wheat plants (Triticum aestivum L.), cv. Fredrick,were exposed to an ozone atmosphere of 0.5 ppm for a periodof 6 h. The leaves of the control plants showed typical symptomsof ozone injury, i.e. chlorosis, necrotic lesions, increasedsolute leakage and reduced Hill activity. These phytotoxic symptomswere prevented in the plants grown from seeds which had beentreated at 0.03 g.kg–1 seed with the triazole S-3307.Ozone treatment also induced a decline in phospholipid contentand a concomitant increase in the concentrations of free fattyacids. There was no evidence of fatty acid peroxidation in thatthere was no significant decrease in fatty acid unsaturationwith ozone fumigation. This supports the hypothesis that themechanism of ozone toxicity may be partially mediated througha free radical cleavage of the ester linkages between the glycerolmoiety and the fatty acid tails of phospholipids. The cleavageof phospholipids was not evident in the plants pretreated withS-3307 and it is suggested that this may be due in part to increasesin lipid soluble antioxidants found in the triazole treatedplants. Age-dependent changes in the concn (nmol g d. wt–1) andrelative proportion of polar lipid fatty acids in potato tubertissue were characterized over a 32-month storage period. Astubers advanced in age from 2 to 14 months, a decrease (approximately22%) in concn of both saturated (16:0, 18:0) and unsaturated(18:2, 18:3) fatty acids was evident. This decrease was followedby an exponential increase through 32 months. Electrolyte leakagestudies with excised tissue revealed age-dependent differencesin membrane permeability. The relationship between maximum electrolyteleakage (% total) and tissue age was described by a cubic polynomial.Leakage declined approximately 4% as tissues aged from 2 to7 months; however, from 7 to 24 months, leakage increased 21%.These trends were followed by partial restoration of membranefunction in 24 to 32-month-old tissues, as indicated by attenuatedleakage. Similarly, the trend in double bond index (DBI) withage was defined by a cubic polynomial; however, the minima andmaxima were the opposite of those for electrolyte leakage vs.age. The regression of DBI upon leakage was inverse and linear(r = –0·97, P < 0.01). The physical properties and chemical composition of microsomal membranes were examined during a 7 day period of ice encasement in crown tissue of winter wheat (Triticum aestivum L. cv Norstar). Membrane damage, detected as an increase in microviscosity and electrolyte leakage, began between 1 and 3 days of icing, and was associated with a reduction in the recovery of microsomal membranes from stressed tissue, an increase in the microsomal free fatty acid:total fatty acid ratio, and a decrease in the phospholipid:total fatty acid ratio. These trends were amplified between 3 and 7 days of ice encasement. Examination of the free and total fatty acid fractions showed there was a slight, but not statistically significant (P = 0.05) reduction in the degree of unsaturation of the total fatty acid fraction. The composition of the free and total fatty acid fractions were very similar during ice encasement. Furthermore, analysis of phospholipid classes revealed no significant change in the relative amounts of phosphatidylcholine, phosphatidylethanolamine, phosphatidic acid, or lysophospholipids in microsomal membranes during icing. Membrane injury during ice encasement apparently involves hydrolysis of the ester bond between glycerol and the acyl groups of the phospholipid resulting in loss of the phosphate-containing polar head group and a concomitant accumulation of free fatty acids in the bilayer.  相似文献

In septic shock patients, alterations of plasma phospholipid fatty acid profile have never been described. The purpose of this monocentric, non-interventional, observational prospective study was to describe this fatty acid profile in the early phase of septic shock in intensive care unit. Thirty-seven adult patients with septic shock were included after the first day of stay in intensive care unit, before any form of artificial nutritional support. Plasma phospholipid fatty acid composition was determined by gas chromatography. All biological data from patients with septic shock were compared with laboratory reference values. Patients presented hypocholesterolemia and hypertriglyceridemia. They had low concentrations of phospholipid fatty acids specifically n-6 and n-3 polyunsaturated fatty acids (PUFAs) with a high n-6/n-3 ratio. Plasma phospholipid PUFA concentrations were strongly correlated with cholesterolemia. PUFAs/SFAs (saturated fatty acids) and PUFAs/MUFAs (monounsaturated fatty acids) ratios were low because of low percentage of n-6 and n-3 PUFAs and high percentage of SFAs and MUFAs. Low levels of plasma long chain PUFAs (≥20 carbons) were significantly associated with mortality at 28th day. In conclusion, plasma phospholipid FA profile of septic patients is very characteristic, close to that of acute respiratory distress syndrome and mortality is associated with long chain PUFA decrease. This profile could be explained by numerous non-exclusive physio-pathological processes 1) an activation of hepatic de novo lipogenesis that could contribute to hepatic steatosis, 2) an elevated adipose tissue lipolysis, 3) an increased free radical attack of FA by oxidative stress, 4) an over-production of inflammatory lipid mediators.  相似文献

Biosynthetic pathways of acyl moieties of capsaicinoid in intactCapsicum fruits and spheroplasts prepared from placentas ofCapsicum fruits were examined using a radioisotopic technique.In intact Capsicum fruits, L-[U-14C] valine was incorporatedinto capsaicin and dihydrocapsaicin, the acyl constituents ofwhich are even-number branched chain fatty acids, while L-[U-14C]leucine was incorporated into nordihydrocapsaicin and homodihydrocapsaicin,which have odd-number branched chain facty acids as the acylmoieties. The intermediates of the odd- and even-number branchedchain fatty acids were identified with GLC/GPC after the spheroplastshad been incubated with L-[U-14C] valine or L-[U-14C] leucine.After incubation with L-[U-14C] valine, isobutyric acid and8-methyl nonanoic acid were detected, while isopentanoic acidand 9-methyl decanoic acid were found after incubation withL-[U-14C] leucine. The involvement of a-ketoisovalerate or a-ketoisocaproatein the biosynthesis of acyl moieties of capsaicinoid was alsodemonstrated in vitro using cell-free extracts of the placentasof Capsicum fruits.
