Members of the grass subfamily Pooideae are characterized by their adaptation to cool temperate climates. Vernalization is the process whereby flowering is accelerated in response to a prolonged period of cold. Winter cereals are tolerant of low temperatures and flower earlier with vernalization, whereas spring cultivars are intolerant of low temperatures and flower later with vernalization. In the pooid grasses wheat (Triticum monococcum, Triticum aestivum) and barley (Hordeum vulgare), vernalization responsiveness is determined by allelic variation at the VERNALIZATION1 (VRN1) and/or VRN2 loci. To determine whether VRN1, and its paralog FRUITFULL2 (FUL2), are involved in vernalization requirement across Pooideae, we determined expression profiles for multiple cultivars of oat (Avena sativa) and wheat with and without cold treatment. Our results demonstrate significant up-regulation of VRN1 expression in leaves of winter oat and wheat in response to vernalization; no treatment effect was found for spring or facultative growth habit oat and wheat. Similar cold-dependent patterns of leaf expression were found for FUL2 in winter oat, but not winter wheat, suggesting a redundant qualitative role for these genes in the quantitative induction of flowering competency of oat. These and other data support the hypothesis that VRN1 is a common regulator of vernalization responsiveness within the crown pooids. Finally, we found that up-regulation of VRN1 in vegetative meristems of oat was significantly later than in leaves. This suggests distinct and conserved roles for temperate cereal grass VRN1/FUL-like genes, first, in systemic signaling to induce flowering competency, and second, in meristems to activate genes involved in the floral transition.  相似文献

The life cycle of Podophyllum can be divided into two phases, a subterranean phase during which a conspicuous winter mixed terminal bud forms at the end of a rhizome, and an aerial phase, during which the primordia of the structures within the winter bud give rise the next spring to an aerial shoot composed of a stem, 2 leaves, and a single flower. The transition from a vegetative to a floral apex occurs at the end of July, when the apical meristem becomes a globoid structure. During the first and second weeks of August, the floral organs are laid down along the sides of an elongated floral apex. The order of initiation of the floral organs is sepals, petals, stamens, gynoecium, and stamens. Petal primordia are initiated in early August, but growth ceases after they attain a height of about 2 mm. This inhibition persists until the middle of May in the next growing season, when the petals grow to 12 mm within 2 weeks. At anthesis the petals have enlarged to a length of 2 cm or more. The gynoecium is usually composed of a single terminal carpel. The ovules are chiefly supplied by branches from a ventral bundle complex, but that is supplemented by medullary bundles that are formed in the base of the gynoecium, below the loculus. It could be argued that these medullary bundles are surviving remnants of the vascular supply to a second carpel, no longer extant. A transmitting tract extends from the stigma about half the distance to the loculus. The tract is lined with unicellular glandular cells and is open from the stigma to the loculus.  相似文献

The genomes of grass family species have three paralogs of APETALA1/FRUITFULL (AP1/FUL)-like genes (FUL1, FUL2 and FUL3) that are derived from the FUL lineage. In this study, we focus on the different roles of the wheat AP1/FUL-like genes, WFUL1 (identical to VRN1), WFUL2 and WFUL3, during the transition from vegetative to reproductive growth. Sequence analysis indicated that there was a high level of variability in the amino acid sequence of the C-domain among three WFUL genes. Expression analyses using the spring wheat cultivar Chinese Spring indicated that WFUL1/VRN1 was expressed in leaves as well as spike primordia of non-vernalized plants at the vegetative stage just before phase transition, while WFUL2 and WFUL3 were not expressed in leaves. This result indicates that WFUL1/VRN1 performs a distinct role in leaves before phase transition. In young spikes, WFUL1/VRN1 and WFUL3 were expressed in all developing floral organs, whereas WFUL2 expression was restricted in the floral organs to the lemma and palea. Furthermore, yeast two-hybrid and three-hybrid analyses revealed that WFUL2, but not WFUL1/VRN1 or WFUL3, interacted with class B and class E proteins. These results suggest that WFUL2 of wheat has class A functions in specifying the identities of floral meristems and outer floral organs (lemma and palea) through collaboration with class B and class E genes.  相似文献

Background  

Evolution of unisexual flowers entails one of the most extreme changes in plant development. Cultivated spinach, Spinacia oleracea L., is uniquely suited for the study of unisexual flower development as it is dioecious and it achieves unisexually by the absence of organ development, rather than by organ abortion or suppression. Male staminate flowers lack fourth whorl primordia and female pistillate flowers lack third whorl primordia. Based on theoretical considerations, early inflorescence or floral organ identity genes would likely be directly involved in sex-determination in those species in which organ initiation rather than organ maturation is regulated. In this study, we tested the hypothesis that sexual dimorphism occurs through the regulation of B class floral organ gene expression by experimentally knocking down gene expression by viral induced gene silencing.  相似文献

The life cycles of plants are characterized by two major life history transitions—germination and the initiation of flowering—the timing of which are important determinants of fitness. Unlike annuals, which make the transition from the vegetative to reproductive phase only once, perennials iterate reproduction in successive years. The floral repressor PERPETUAL FLOWERING 1 (PEP1), an ortholog of FLOWERING LOCUS C, in the alpine perennial Arabis alpina ensures the continuation of vegetative growth after flowering and thereby restricts the duration of the flowering episode. We performed greenhouse and garden experiments to compare flowering phenology, fecundity and seed traits between A. alpina accessions that have a functional PEP1 allele and flower seasonally and pep1 mutants and accessions that carry lesions in PEP1 and flower perpetually. In the garden, perpetual genotypes flower asynchronously and show higher winter mortality than seasonal ones. PEP1 also pleiotropically regulates seed dormancy and longevity in a way that is functionally divergent from FLC. Seeds from perpetual genotypes have shallow dormancy and reduced longevity regardless of whether they after‐ripened in plants grown in the greenhouse or in the experimental garden. These results suggest that perpetual genotypes have higher mortality during winter but compensate by showing higher seedling establishment. Differences in seed traits between seasonal and perpetual genotypes are also coupled with differences in hormone sensitivity and expression of genes involved in hormonal pathways. Our study highlights the existence of pleiotropic regulation of seed traits by hub developmental regulators such as PEP1, suggesting that seed and flowering traits in perennial plants might be optimized in a coordinated fashion.  相似文献

Rapeseed (Brassica napus L.), one of the most important sources of vegetable oil and protein‐rich meals worldwide, is adapted to different geographical regions by modification of flowering time. Rapeseed cultivars have different day length and vernalization requirements, which categorize them into winter, spring, and semiwinter ecotypes. To gain a deeper insight into genetic factors controlling floral transition in B. napus, we performed RNA sequencing (RNA‐seq) in the semiwinter doubled haploid line, Ningyou7, at different developmental stages and temperature regimes. The expression profiles of more than 54,000 gene models were compared between different treatments and developmental stages, and the differentially expressed genes were considered as targets for association analysis and genetic mapping to confirm their role in floral transition. Consequently, 36 genes with association to flowering time, seed yield, or both were identified. We found novel indications for neofunctionalization in homologs of known flowering time regulators like VIN3 and FUL. Our study proved the potential of RNA‐seq along with association analysis and genetic mapping to identify candidate genes for floral transition in rapeseed. The candidate genes identified in this study could be subjected to genetic modification or targeted mutagenesis and genotype building to breed rapeseed adapted to certain environments.  相似文献

蝴蝶兰花非常独特且高度进化,如萼片瓣化、瓣片特化为唇瓣、雌雄蕊合生成合蕊柱及子房发育须由授粉启动等,是单子叶植物花发育研究的理想材料。近年来蝴蝶兰花发育分子生物学取得了重要进展。该文就近年来国内外有关蝴蝶兰开花转换及花器官发育相关基因研究以及B类基因与兰花花被的进化发育关系方面的研究进展进行综述。研究表明:MADS基因在蝴蝶兰开花转换及花器官发育过程中起重要作用,推测其中的DEF(DE-FICIENS)-like基因早期经过2轮复制,形成了4类不同的DEF-like基因,进而决定兰花花被属性。蝴蝶兰花发育分子生物学的深入研究,将极大地利于通过基因工程手段提高蝴蝶兰花品质如花色改良及花期调控等,推动分子育种进程。  相似文献

Investigation of low-temperature (LT) tolerance in cereals has commonly led to the region of the vyn-A1 vernalization gene or its homologue in related genomes. Two cultivars, one a non-hardy spring wheat and one a very cold-hardy winter wheat, whose growth habits are determined by the Vrn-A1 (spring habit) and vrn-A1 (winter habit) alleles, were chosen to produce reciprocal near-isogenic lines (NILs). These lines were then used to determine the relationship between rate of phenological development and the degree and duration of LT tolerance gene expression. Each allele was isolated in the genetic backgrounds of the non-hardy spring wheat 'Manitou' and the very cold-hardy winter wheat 'Norstar'. The effects of each allele on phenological development and low-temperature tolerance (LT50) were determined at regular intervals over a 4 degrees C acclimation period of 0-98 d. The vegetative/reproductive transition, as determined by final leaf number (FLN), was found to be a major developmental factor influencing LT tolerance. Possession of a vernalization requirement increased both the length of the vegetative growth phase and LT tolerance. Similarly, increased FLN in spring Norstar and winter Manitou NILs delayed their vegetative/reproductive transition and increased their LT tolerance relative to Manitou. Although the winter Manitou NILs had a lower FLN than the spring Norstar NILs, they were able to extend their vegetative stage to a similar length by increasing the phyllochron (interval between the appearance of successive leaves). Cereal plants have four ways of increasing the length of the vegetative phase, all of which extend the time that low-temperature tolerance genes are more highly expressed: (1) vernalization; (2) photoperiod responses; (3) increased leaf number; and (4) increased length of the phyllochron.  相似文献

Estimating the timing of flower bud formation in plants is essential to identify environmental factors that regulate floral transition. The presence of winter dormancy between the initiation of flowers and anthesis, characteristic of most trees in the temperate forests, hampers accurate estimation of the timing of floral transition. To overcome this difficulty, expression levels of flowering-time genes could be used as indicators of the timing of floral transition. Here, we evaluated the usefulness of molecular markers in estimating the timing of floral transition in Fagus crenata, a deciduous tree that shows intermittent and synchronized flowering at the population level. We selected FLOWERING LOCUS T (FT) as a candidate molecular marker and quantified the expression levels of its ortholog in F. crenata (FcFT). Subsequently, we analyzed the relationship between morphogenetic changes that occur between the vegetative state of the buds and the initiation of floral organs, and compared the FcFT expression levels in reproductive and vegetative buds, collected from spring to autumn. FcFT expression in leaves peaked at least two weeks before the morphological changes associated with flowering were visible in the buds in late July. FcFT expression levels were significantly higher in the reproductive buds than in the vegetative buds in July. These results suggest that the FcFT expression in July is a reliable indicator of the timing and occurrence of floral transition. This study highlights the utility of molecular tools in unraveling reproductive dynamics in plants, in combination with ecological and physiological approaches.  相似文献

Larger and longer lived flowers receive more pollinators, but may also involve increased water maintenance costs under hot, dry environments. Hence, smaller and/or short-lived flowers may buffer such costs. We surveyed floral longevity in three large-flowered Mediterranean Cistus species. We hypothesize that: (1) in Cistus, floral longevity decreases with increasing air temperature and flower size; (2) in C. ladanifer, flower size and longevity increase along an altitudinal gradient; (3) floral longevity is differentially affected by temperature rather than flower size along the gradient; (4) under similar temperature, floral longevity decreases with flower size. For each species, we evaluated the effects of flower size and air temperature on floral longevity. Specifically, floral longevity was surveyed along an altitudinal gradient in the largest flowered species Cistusladanifer. Floral longevity in Cistus species lasted < 1 d and was affected by air temperature, independently of flower size. In C. ladanifer, flower size increased along the gradient but floral longevity decreased. Still, floral longevity decreased with increasing air temperature and, to a lesser extent, with flower size. Together, our findings show a triangular relationship among air temperature, flower size and floral longevity with margins for plasticity to accommodate pollinator attraction with the costs of large-flowered Mediterranean plants.  相似文献
