Deschampsia antarctica Desv. (Poaceae) represents one of the two vascular plants that have colonized the Antarctic continent, which is usually exposed to extreme environmental conditions. In this work, we have characterized the endophytic fungi associated with the leaves of D. antarctica. Endophytic fungi were recovered from 91 individual plants from different points of Admiralty Bay at King George Island, Antarctica. A total of 26 fungal isolates were obtained from 273 leaf fragments. All isolates were identified by analysis of the sequences of the internal transcribed spacer region (ITS) of the rDNA. Alternaria and Phaeosphaeria were the most frequent genera associated with the plant. Other fungal isolates were identified as Entrophospora sp. and several undescribed Ascomycete species. An interesting result was obtained for the isolates UFMGCB 215 and UFMGCB 262, which were related to fungi associated with bryophytes present in boreal ecosystems. Some isolates showed low identity in the ITS sequences to sequences of fungal species deposited in GenBank, suggesting that these fungi could be new species. This work is the first report on fungal endophytes associated with leaves of the Antarctic grass D. antarctica.  相似文献

Genetic variation of the Antarctic anisakid Contracaecum radiatum from the Ross and Weddell Seas is studied at 24 enzyme loci. All polymorphic loci proved to be in Hardy-Weinberg equilibrium, with no significant differences between the samples tested. Several loci were found to be diagnostic between C. radiatum and the five known members of the C. osculatum complex, indicating that no gene exchange occurs between them and confirming their specific status. No F1 hybrids, recombinant or introgressed individuals, sharing radiatum and osculatum alleles, were detected in the Antarctic Contracaecum samples; this directly proves the reproductive isolation of C. radiatum. Biochemical keys are given for the identification of Antarctic C. radiatum, C. osculatum D and E, both for adults (males and females) and larvae. Higher values of genetic variability were observed in these Antarctic Contracaecum species than in the Arctic-Boreal C. osculatum members, possibly related to a lower degree of habitat disturbance (i.e. by pollution, fishing and hunting) in the Antarctic region. On the basis of genetic distances, the evolutionary divergence between C. radiatum and C. osculatum (sensu lato) started about 5 million years ago, possibly with the first colonisation of the Antarctic region by seals. Data on paratenic (fish) and definitive (seal) hosts of C. radiatum and C. osculatum D and E are given.  相似文献

The heterotrophic dinoflagellate Pfiesteria piscicida was detected in Ace Lake in the Vestfold Hills, eastern Antarctica by using real-time PCR based on 18S rDNA sequences. Antarctic water samples collected in 2004 were tested by species-specific real-time PCR assays for the identification of P. piscicida and P. shumwayae. Positive results were shown with P. piscicida-specific real-time PCR, and PCR products were examined by sequence analysis for confirmation. A phylogenetic tree made from partial 18S rDNA sequences showed that the Antarctic clone clustered with P. piscicida. This result suggests that P. piscicida is present in the extreme conditions of an Antarctic saline lake which has not contained fish for thousands of years.  相似文献

The Antarctic krill (Euphausia superba Dana 1852) is widely distributed throughout the Southern Ocean, where it provides a key link between primary producers and upper trophic levels and supports a major commercial fishery. Despite its ecological and commercial importance, genetic population structure of the Antarctic krill remains poorly described. In an attempt to illuminate genetic markers for future population and phylogenetic analysis, five E. superba mitogenomes, from samples collected west of the Antarctic Peninsula, were sequenced using new 454 next-generation sequencing techniques. The sequences, of lengths between 13,310 and 13,326 base pairs, were then analyzed in the context of two previously-published near-complete sequences. Sequences revealed relatively well-conserved partial mitochondrial genomes which included complete sequences for 11 of 13 protein-coding genes, 16 of 23 tRNAs, and the large ribosomal subunit. Partial sequences were also recovered for cox1 and the small ribosomal subunit. Sequence analysis suggested that the cox2, nad5, and nad6 genes would be the best candidates for future population genetics analyses, due to their high number of variable sites. Future work to reveal the noncoding control region remains.  相似文献

Solieria chordalis (C. Agardh) J. Agardh and S. tenera (J. Agardh) Wynne et Taylor exhibit multiaxial growth from a cluster of four to eight obconical apical cells. A single periaxial cell is cut off from each axial cell and successive periaxial cells are rotated 120° in a zig-zag pattern along each axial filament. Periaxial cells produce branched, laterally diverging filaments which form the cortex. The medulla is composed of axial cells, elongate cells of lateral filaments, stretched interconnecting cells, and secondary rhizoids. The two species are nonprocarpic. Carpogonial branches are 3-celled, inwardly directed, with a reflexed trichogyne. The auxiliary cell together with associated darkly-staining inner cortical cells form an association, the auxiliary cell complex, that is recognizable prior to diploidization. A single, unbranched, non-septate connecting filament issues from the fertilized carpogonium and fuses with the inner, lateral side of an auxiliary cell. Production of an involucre from surrounding vegetative cells is stimulated and a gonimoblast initial is cut off toward the interior of the thallus which divides to form a compact cluster of gonimoblast cells. A fusion cell is produced through fusion of inner gonimoblast cells with the auxiliary cell that, in turn, fuses progressively with cells of the lateral file bearing the auxiliary cell. Mature cystocarps have terminal carposporangia cut off from gonimoblast cells at the periphery of the fusion cell and are surrounded by an involucre with a distinct ostiole. Tetrasporangia are cut off laterally from surface cortical cells which then cut off one or two additional derivatives toward the outside. A lectotype is designated for Solieria chordalis, but the lectotypification of S. tenera is questioned. We conclude that Solieria is closely related to Rhabdonia and place the Rhabdoniaceae in synonomy with the Solieriaceae.  相似文献

We present a phytogeographical comparison between polar (Arctic and Antarctic) and non-polar strains of the cyanobacterial genus Phormidium, which plays a key role in Arctic and Antarctic ecosystems as primary producer. A total of 26 Phormidium strains were studied using a polyphasic approach, 18 from Arctic (Svalbard, Ellesmere Island and Scandinavian Arctic—Abisko) and Antarctic (Antarctic Peninsula—King George and James Ross Island) regions, and 8 from temperate sites (mostly situated in Central Europe). A phylogenetic tree was constructed and compared with similar 16S rRNA sequences retrieved from Genbank. Within the Phormidium autumnale cluster, genetic similarity of 16S rDNA was more related to geographical proximity of strain origin than to morphological similarity. No genetic identity of Phormidium strains from north and south polar regions was found. The cluster Phormidium autumnale apparently belongs to generic entities in which geographical limitation plays a prominent role. However, the cyanobacterial strains found in Europe suggest that the distribution areas of some Phormidium cyanobacteria overlap. The Phormidium autumnale cluster is evidently a very characteristic type and represents an isolated clade within the traditional genus Phormidium. According to morphological features and the structure of trichomes, it is most similar and thus probably belongs to the genus Microcoleus.  相似文献

We characterized six tintinnid ciliates from Antarctic waters using molecular markers and morphological traits: Amphorellopsis quinquealata, Codonellopsis gaussi, Cymatocylis convallaria, Cy. calyciformis, Cy. drygalskii, and Laackmanniella prolongata. The 100% similarity in SSU‐ITS1‐5.8S rDNA‐ITS2‐partial LSU rDNA sequences among Cy. convallaria, Cy. calyciformis, and Cy. drygalskii is supportive of synonymy. Codonellopsis gaussi and L. prolongata also showed high levels of similarity in SSU rDNA (99.83%) and the D2 domain of LSU rDNA (95.77%), suggesting that they are closely related. Phylogenetic analysis placed Cymatocylis in the Rhabdonellidae, Amphorellopsis in the Tintinnidae and L. prolongata/Co. gaussi within the Dictyocystidae.  相似文献

Populations of both native higher Antarctic plants, Deschampsia antarctica and Colobanthus quitensis, increased during the last decades. However, for D. antarctica, previous population studies on the South Shetland Islands and the Antarctic Peninsula have been too sporadic, patchy, and methodologically different to allow general conclusions. Our aim was to compare sites with D. antarctica along a north–south latitudinal transect with an integral census method to assess the possible impact of climatic change on grass population dynamics. During two summer seasons (2009–2010), plant populations were censed on Fildes and Coppermine Peninsula and several localities on the west coast of the Antarctic Peninsula. Largest plant populations were found on Fildes Peninsula with vegetation cover (VC) of 44–46%. Six out of eleven stands of D. antarctica on Coppermine Peninsula were new records, with increasing plant number and VC (0.1–22%). In the Antarctic Peninsula, contrarily to our expectation, only at Forbes Point, D. antarctica VC was relatively high (ca. 2%) and a new stand of C. quitensis was found. At three previously reported sites, plants had disappeared. Our monitoring confirms that northern D. antarctica populations are expanding, but that this expansion is not continuous along the Antarctic Peninsula and inconsistent with the gradient of relative temperature increase in north–south direction. We suggest that other abiotic and biotic factors are influencing the colonization and expansion of vascular plants in this particular ecosystem.  相似文献

Antarctic toothfish (Dissostichus mawsoni) are the target of an important commercial fishery in the Southern Ocean, yet age data used for management have not been comprehensively tested for accuracy. In this study, Antarctic toothfish were aged using counts of otolith growth zones based on criteria established for Patagonian toothfish, D. eleginoides, a closely related species. To validate these ages, the radioactive disequilibrium of lead-210 and radium-226 in otolith cores was measured and used as an independent chronometer to accurately determine age across the range of fish caught in large numbers by the fishery. Growth-zone counts indicated Antarctic toothfish live to at least 39 years of age, and were in close agreement with the chronometer, validating the age estimation criteria and the accuracy of age estimates. Von Bertalanffy growth function parameters indicated Antarctic toothfish were relatively slow-growing (k = 0.111), especially in relation to their maximum size (L  = 158.9 cm).  相似文献

Cryptococcus adeliensis sp. nov. (CBS 8351) is described based on phenotypic characteristics and molecular sequence analysis of the D1/D2 large subunit and internal transcribed spacer regions of the ribosomal DNA. Molecular comparisons include species closely related to Cryptococcus albidus and several species isolated from the Antarctic. C. adeliensis, which has a cold-adapted xylanase, was isolated from Terre Adelie, Antarctica. ATCC 34633, which has a mesophilic xylanase, was identified as Cryptococcus albidosimilis.  相似文献

Monothalamous (single-chambered) foraminifers are a major component of the benthic meiofauna in high latitude regions. Several morphologically similar species are common in the Arctic and Antarctic. However, it is uncertain whether these morphospecies are genetically identical, or whether their accurate identification is compromised by a lack of distinctive morphological features. To determine the relationship between Arctic and Antarctic species, we have compared SSU rDNA sequences of specimens belonging to four morphotaxa: Micrometula, Psammophaga, Gloiogullmia, and one morphospecies Hippocrepinella hirudinea from western Svalbard (Arctic) and McMurdo Sound (Antarctic). Wherever possible, we include in our analyses representatives of these taxa from the deep Arctic and Southern Oceans, as well as from Northern European fjords. We found that in all cases, the bipolar populations were clearly distinct genetically. As expected, Arctic specimens were usually more closely related to those from Northern Europe than to their Antarctic representatives. The deep-sea specimens from Weddell Sea branched as a sister to the McMurdo Sound population, while those from the Arctic Ocean clustered with ones from Norwegian fjords. Our study has revealed a high number of cryptic species within each of the examined genera, and demonstrates the unexplored potential of monothalamous foraminifers for use as a tool to evaluate the origin and biogeography of polar meiofauna.  相似文献

The two giant notothenioid species, the Patagonian toothfish Dissostichus eleginoides and the Antarctic toothfish D. mawsoni, are important components of the Antarctic ichthyofauna and heavily exploited commercially. They have similar appearance and size, both are piscivorous and bentho-pelagic, but differ in their geographic distribution and absence/presence of the antifreeze trait. We karyotyped these two sister species by analyzing specimens collected from multiple Antarctic and sub-Antarctic sites. Both species have a diploid number of 48, but differ in karyotypic formula, (2m + 2sm + 44a) for D. eleginoides and (2m + 4sm + 42a) for D. mawsoni, due to an extra pair of submetacentric chromosomes in the latter. Chromosomal fluorescence in situ hybridization with rDNA probes revealed unexpected species-specific organization of rRNA genes; D. mawsoni possesses two rDNA loci (versus one locus in D. eleginoides), with the second locus mapping to its additional submetacentric chromosome. The additional rRNA genes in D. mawsoni may be a cold-adaptive compensatory mechanism for growth and development of this large species in freezing seawater.  相似文献

Phylogenetic relationships in the Desmarestiales (Phaeophyceae) were inferred among the monotypic Arthrocladia (Arthrocladiaceae) and 27 isolates from Desmarestiaceae, representing 17 taxa of Desmarestia and the monotypic Antarctic genera Himantothallus and Phaeurus. Phaeurus and Arthrocladia were used as outgroups. Parsimony analyses of nuclear ribosomal DNA internal transcribed spacer (ITS1 and ITS2) sequences, in which gaps were both included and excluded, yielded well-resolved trees with a consistent general branching pattern. A parallel analysis of nine morphological and life-history characters and three ecological characters yielded a similar tree but provided little resolution in the terminal clades. The position of the monotypic Arthrocladia villosa within the Desmarestiales is consistent with monophyly for the order, but its position as the most primitive desmarestialean is not resolvable from the molecular data set. The basal position of Phaeurus, the Antarctic Desmarestia species, and Himantothallus is consistent with the hypothesis of a Southern Hemisphere origin for the family Desmarestiaceae. The more recent Northern Hemisphere “aculeata” clade evolved from an Antarctic ancestor. A “D. aculeata-like” species was ancestral to a lineage characterized by annual sporophytes with high sulfuric acid content, which radiated into many species, widely distributed in both hemispheres. Mapping of morphological and ecological characters onto the molecular tree confirm the informativeness of sulfuric acid-containing vacuoles and unilocular sporangial types. There is good congruence between phylogenetic tree topology and temperature impints in relation to biogeographic distribution, supporting the theory that temperature tolerance is a conservative trait.  相似文献

Exopolysaccharides (EPS) may have an important role in the Antarctic marine environment, possibly acting as ligands for trace metal nutrients such as iron or providing cryoprotection for growth at low temperature and high salinity. Ten bacterial strains, isolated from Southern Ocean particulate material or from sea ice, were characterized. Whole cell fatty acid profiles and 16S rRNA gene sequences showed that the isolates included representatives of the genera Pseudoalteromonas, Shewanella, Polaribacter, and Flavobacterium as well as one strain, which constituted a new bacterial genus in the family Flavobacteriaceae. The isolates are, therefore, members of the “Gammaproteobacteria” and Cytophaga-Flexibacter-Bacteroides, the taxonomic groups that have been shown to dominate polar sea ice and seawater microbial communities. Exopolysaccharides produced by Antarctic isolates were characterized. Chemical composition and molecular weight data revealed that these EPS were very diverse, even among six closely related Pseudoalteromonas isolates. Most of the EPS contained charged uronic acid residues; several also contained sulfate groups. Some strain produced unusually large polymers (molecular weight up to 5.7 MDa) including one strain in which EPS synthesis is stimulated by low temperature. This study represents a first step in the understanding of the role of bacterial EPS in the Antarctic marine environment.  相似文献

The derived amino-acid sequences of the proteins encoded by E. coli genes sbcC and sbcD have been compared with other protein sequences using computer assisted methods. This work has shown that SbcC and D, which inhibit the propagation of replicons containing long palindromic DNA sequences, are distantly related to two putative bacteriophage nucleases. These nucleases both comprise two polypeptide chains which are the products of genes 46 and 47 of bacteriophage T4 (gp 46 and gp 47) and genes D13 and D12 of bacteriophage T5 (gp D13 and gp D12). The comparisons reveal that SbcC, gp 46 and gp D13 are more closely related to each other than are SbcD, gp 47 and gp D12. SbcC appears to have undergone a partial duplication of an ancestral sequence. These proteins all contain motifs common to the superfamily of nucleotide-binding proteins that includes UvrA and the cystic fibrosis transmembrane regulator CFTR.  相似文献

We isolated 59 strains of cyanobacteria from the benthic microbial mats of 23 Antarctic lakes, from five locations in two regions, in order to characterize their morphological and genotypic diversity. On the basis of their morphology, the cyanobacteria were assigned to 12 species that included four Antarctic endemic taxa. Sequences of the ribosomal RNA gene were determined for 56 strains. In general, the strains closely related at the 16S rRNA gene level belonged to the same morphospecies. Nevertheless, divergences were observed concerning the diversity in terms of species richness, novelty, and geographical distribution. For the 56 strains, 21 operational taxonomic units (OTUs, defined as groups of partial 16S rRNA gene sequences with more than 97.5% similarity) were found, including nine novel and three exclusively Antarctic OTUs. Sequences of Petalonema cf. involvens and Chondrocystis sp. were determined for the first time. The internally transcribed spacer (ITS) between the 16S and the 23S rRNA genes was sequenced for 33 strains, and similar groupings were observed with the 16S rRNA gene and the ITS, even when the strains were derived from different lakes and regions. In addition, 48 strains were screened for antimicrobial and cytotoxic activities, and 17 strains were bioactive against the gram‐positive Staphylococcus aureus, or the fungi Aspergillus fumigatus and Cryptococcus neoformans. The bioactivities were not in coincidence with the phylogenetic relationships, but rather were specific to certain strains.  相似文献

The study of Antarctic cyanobacterial diversity has been mostly limited to morphological identification and traditional molecular techniques. High‐throughput sequencing (HTS) allows a much better understanding of microbial distribution in the environment, but its application is hampered by several methodological and analytical challenges. In this work, we explored the use of HTS as a tool for the study of cyanobacterial diversity in Antarctic aquatic mats. Our results highlight the importance of using artificial communities to validate the parameters of the bioinformatics procedure used to analyze natural communities, since pipeline‐dependent biases had a strong effect on the observed community structures. Analysis of microbial mats from five Antarctic lakes and an aquatic biofilm from the Sub‐Antarctic showed that HTS is a valuable tool for the assessment of cyanobacterial diversity. The majority of the operational taxonomic units retrieved were related to filamentous taxa such as Leptolyngbya and Phormidium, which are common genera in Antarctic lacustrine microbial mats. However, other phylotypes related to different taxa such as Geitlerinema, Pseudanabaena, Synechococcus, Chamaesiphon, Calothrix, and Coleodesmium were also found. Results revealed a much higher diversity than what had been reported using traditional methods and also highlighted remarkable differences between the cyanobacterial communities of the studied lakes. The aquatic biofilm from the Sub‐Antarctic had a distinct cyanobacterial community from the Antarctic lakes, which in turn displayed a salinity‐dependent community structure at the phylotype level.  相似文献

Hydroidolinan hydrozoans are widely represented in the benthic Antarctic ecosystem, mainly by some endemic and putative monophyletic groups, never included in molecular phylogenetic analyses. 38 partial sequences of the mitochondrial 16S rRNA gene were obtained for 38 species belonging to 14 families (six anthoathecates and eight leptothecates) and 20 genera (7 anthoathecates and 13 leptothecates). These sequences were combined with 108 additional sequences retrieved from the GenBank to investigate both the hypothetical monophyletism and the phylogenetic relationships of those endemic Antarctic groups; the potential use of the marker for barcoding was also investigated. Our results uphold the monophyly of some important hydroidolinan groups, such as the superfamily Plumularioidea (together with all its families, including Schizotrichidae, fam. nov.) and the Aplanulata. Concerning the Antarctic endemic groups, most results as monophyletic (Oswaldella, Schizotricha and Staurotheca), some genera form part of the expectable groups (e.g. Abietinella, located into the monophyletic Zygophylacinae clade) and, finally, others have shown a surprising position (e.g. Stegella, closely related to Lafoeinae, or Billardia and Stegopoma, allied with Hebellidae). Finally, our study has shown the utility of the marker to recognize the Antarctic species considered, but the low genetic divergence in some of the most important Antarctic groups suggests being careful when using it for DNA barcoding in the case of the original Antarctic hydroid fauna.  相似文献

Deschampsia antarctica Desv. (Poaceae) and Colobanthus quitensis (Kunth) Bartl. (Cariophyllaceae) are the only two vascular plants that have colonized the Maritime Antarctic. The primary purpose of the present work was to determine cold resistance mechanisms in these two Antarctic plants. This was achieved by comparing thermal properties of leaves and the lethal freezing temperature to 50% of the tissue (LT50). The grass D. antarctica was able to tolerate freezing to a lower temperature than C. quitensis. The main freezing resistance mechanism for C. quitensis is supercooling. Thus, the grass is mainly a freezing‐tolerant species, while C. quitensis avoids freezing. D. antarctica cold acclimated; thus, reducing its LT50. C. quitensis showed little cold‐acclimation capacity. Because day length is highly variable in the Antarctic, the effect of day length on freezing tolerance, growth, various soluble carbohydrates, starch, and proline contents in leaves of D. antarctica growing in the laboratory under cold‐acclimation conditions was studied. During the cold‐acclimation treatment, the LT50 was lowered more effectively under long day (21/3 h light/dark) and medium day (16/8) light periods than under a short day period (8/16). The longer the day length treatment, the faster the growth rate for both acclimated and non‐acclimated plants. Similarly, the longer the day treatment during cold acclimation, the higher the sucrose content (up to 7‐fold with respect to non‐acclimated control values). Oligo and polyfructans accumulated significantly during cold acclimation only with the medium day length treatment. Oligofructans accounted for more than 80% of total fructans. The degrees of polymerization were mostly between 3 and 10. C. quitensis under cold acclimation accumulated a similar amount of sucrose than D. antarctica, but no fructans were detected. The suggestion that survival of Antarctic plants in the Antarctic could be at least partially explained by accumulation of these substances is discussed.  相似文献

The aims of this work were to study cyanobacterial isolates resembling the genus Hydrocoryne using a combination of morphology and phylogeny of 16S rRNA and nifH sequences and to investigate genes involved in cyanotoxin and protease inhibitor production. Four new cyanobacterial strains, isolated from biofilm samples collected from King George Island, Antarctica, were studied. In terms of morphology, these new strains share traits similar to true Anabaena morphotypes (benthic ones), whereas phylogenetic analysis of their 16S rRNA gene sequences grouped them with the sequence of the type species Hydrocoryne spongiosa (H. Schwabe ex Bornet and Flahault 1886–1888), but not with sequences of the type species from the genus Anabaena. This cluster is the sister group of Anabaena morphotypes isolated only from the Gulf of Finland. In addition, this cluster is related to two other clusters formed by sequences of Anabaena isolated from different sites. Partial nifH genes were sequenced from two strains and the phylogenetic tree revealed that the Antarctic nifH sequences clustered with sequences from Anabaena. Furthermore, two strains were tested, using PCR with specific primers, for the presence of genes involved in cyanotoxins (microcystin and saxitoxin) and protease inhibitor (aeruginosin, and cyanopeptolin). Only cyanopeptolin was amplified using PCR. These four Hydrocoryne strains are the first to be isolated and sequenced from Antarctica, which improves our knowledge on this poorly defined cyanobacterial genus.  相似文献
