Waterlogging tolerance, root porosity and root anatomy wereevaluated for 20 Trifolium accessions (species and sub-species,all annuals) selected from the eight Sections of the genus.Nine accessions were sensitive [relative growth rate (RGR) reducedby up to 80%] to waterlogging, nine accessions were tolerant(RGR not reduced), and in two accessions RGR increased (up to1.9-fold), when compared to drained controls. Growth of themain (i.e. tap) root axis was severely reduced in all accessionswhen waterlogged. Lateral roots formed the bulk of the rootsystem of tolerant accessions when grown in waterlogged soil.Lengths of the longest lateral roots were up to three-timeslonger than the main root axis. Root porosity varied from 0.7–12%among accessions when grown in aerated solution and from 1.1–15.5%in plants grown in hypoxic (0.031–0.045 mol O2m-3) solution.In some accessions aerenchyma formed by cell lysigeny; in othersit formed by schizogenous cell separation, or a combinationof both processes. O2consumption rates of expanded lateral roottissues varied by up to 1.7-fold (on a mass basis) among thesix accessions tested and was reduced by an average of 24% forroots of plants grown in hypoxic solution prior to measurements.Accessions with the highest root porosity tended to have longerroots when grown in waterlogged soil. Three accessions formed‘aerotropic roots’ and the lateral root lengthsof these plants exceeded those of all other accessions, suggestingenhanced O2movement to the submerged lateral root axis via theaerotropic roots. Waterlogging-tolerant accessions were identifiedin seven of the eight Sections in Trifolium, and the tolerantaccessions tended to be those with extensive lateral root systemsof relatively high porosity. Stagnant nutrient solution containing 0.1% agar and with anextremely low oxygen level (‘stagnant agar solution’)was used to simulate the gaseous composition and slow gas diffusionof waterlogged soils. Comparisons were made between the growthof two wheat cultivars(Triticum aestivum,cvs. Gamenya and Kite)and one triticale cultivar(Triticosecale,cv. Muir) grown instagnant relative to aerated solution. For all genotypes tested,immersion of roots in stagnant agar solution resulted in thedeath of the entire seminal root system and led to profuse branchingof the laterals of the nodal roots. In the stagnant agar solutionaerenchyma, as a percentage of the total cross sectional areaof nodal roots, was 18% for Muir, 14% for Kite and 12% for Gamenya;the roots of species with more aerenchyma also attained a longermaximum root length as predicted by the model of Armstrong (in:Woolhouse HW, ed.Advances in botanical research, vol. 7. London:Academic Press, 1979). Muir also had a nodal root/shoot freshweight ratio of 0.5 compared with 0.2–0.3 in Kite andGamenya. Nodulated white clover plants (Trifolium repens L. cv. Huia)were grown as simulated swards for 71 d in flowing nutrientsolutions with roots at 11 C and shoots at 20/15 C, day/night,under natural illumination. Root temperatures were then changedto 3, 5, 7, 11, 13, 17 or 25 C and the total N2, fixation over21 d was measured in the absence of a supply mineral N. Alltreatments were subsequently supplied with 10 mmol m–2NO2 in the flowing solutions for 14 d, and the relativeuptake of N by N2, fixation and NO3 uptake was compared.Net uptake of K+ was measured on a daily basis. Root temperature had little effect on root d. wt over the 35-dexperimental period, but shoot d. wt increased by a factor of3.5 between 3 and 25 C, with the sharpest increase occurringat 7–11 C. Shoot: root d. wt ratios increased from 25to 68 with increasing temperature at 7–25 C. N2-fixationper plant (in the absence of NO2 ) increased with roottemperature at 3–13C, but showed little change above13 C. The ratios of N2 fixation: NO2 uptake over 14d (mol N: mol N) were 0.47–0.77 at 3–7 C, 092–154at 11–17 C, and 046 at 25 C, reflecting the dominanceof NO3 uptake over N2 fixation at extremes of high andlow root temperature. The total uptake of N varied only slightlyat 11–25 –C (095–110 mmol N plant–1),the decline in N2 fixation as root temperature increased above11 C was compensated for by the increase in NO 3 uptake.The % N in shoot dry matter declined with decreasing root temperature,from 32% at 13 C to 15% at 3 C. In contrast, concentrationsof N expressed on a shoot water content basis showed a modestdecrease with increasing temperature, from 345 mol m–3at 3 C to 290 mol m–3 at 25 C. Phaseolus vulgaris L. grown at a range of external concentrationsof NaCl (0 to 80 mM) responded differently to gaseous anaerobiosis(N2 gas) in nutrient solution or stagnant waterlogging of theroot-zone. With similar patterns of distribution of Na+ andCl- occurring in the plants with comparable NaCl treatments,and similar final concentrations of Na+ and Cl- in plants grownunder both root-zone conditions, rates of uptake of Na+ andCl- were much higher in plants with the stagnant waterloggedrootzones. After 72 h stagnant waterlogging, plant tops fromplants grown at 40 mM NaCl contained 1.42 per cent Na+ and 3.44per cent Cl- (d. wt basis) while after 9 days exposure to NaClwith gaseous anaerobiosis, leaf tissue contained 1.49 per centNa+ and 4.28 per cen Cl- (d. wt basis). Plants exposed to 40mM external NaCl were severely damaged within 72 h when grownwith stagnant waterlogged root-zones; those grown with N2 anaerobiosiscontinued growth and development over the 9 d period. The present study evaluated waterlogging tolerance, root porosityand radial O2 loss (ROL) from the adventitious roots, of sevenupland, three paddy, and two deep-water genotypes of rice (Oryzasativa L.). Upland types, with the exception of one genotype,were as tolerant of 30 d soil waterlogging as the paddyand deep-water types. In all but one of the 12 genotypes, thenumber of adventitious roots per stem increased for plants grownin waterlogged, compared with drained, soil. When grown in stagnantdeoxygenated nutrient solution, genotypic variation was evidentfor root porosity and rates of ROL, but there was no overalldifference between plants from the three cultural types. Adventitiousroot porosity increased from 20–26 % for plants grownin aerated solution to 29–41 % for plants grown instagnant solution. Growth in stagnant solution also induceda ‘tight’ barrier to ROL in the basal regions ofadventitious roots of five of the seven upland types, all threepaddy types, and the two deep-water types. The enhanced porosityprovided a low resistance pathway for O2 movement to the roottip, and the barrier to ROL in basal zones would have furtherenhanced longitudinal O2 diffusion towards the apex, by diminishinglosses to the rhizosphere. The plasticity in root physiology,as described above, presumably contributes to the ability ofrice to grow in diverse environments that differ markedly insoil waterlogging, such as drained upland soils as well as waterloggedpaddy fields.  相似文献

This paper deals with the responses of roots of wheat {Triticumaestivum L.) to hypoxia with special emphasis on the effectsof severe O2 deficiency on membrane integrity, loss of K+ fromthe root and root membrane potentials. Seminal and crown roots of 26-d-old plants exposed to severehypoxia (0.003 mol O2 m–3) for 3 h or 10 d prior to excisionand subsequently exposed to hypoxic solutions, had slightlylower rates of sorbitol influx and a slightly smaller apparentfree space than roots in aerated solutions. These results indicatethat neither a few hours nor a 10-d exposure to hypoxia hadadverse effects on the membrane integrity of the bulk of thecells in the roots. However, both 6-d-old seedlings and 26-d-oldplants lost K+ from the roots following their transfer fromaerated to hypoxic nutrient solutions. In the 26-d-old plants,which were of high nutritional status, there was a net K+ effluxfrom the roots to the external solution. In contrast, with the6-d-old seedlings, which were of low nutritional status, thedecrease in the K+ content of the roots was smaller than thenet K+ uptake to the shoots. Exposure of excised roots to 0.008 mol O2 m3caused arapid and reversible membrane depolarization from –120to ––80 mV. These data and the magnitude of thenet effluxes strongly suggest that K+ losses during the earlystages of hypoxia are due to membrane depolarization ratherthan to increases in the permeability of membranes to K +. The relationship between coleoptile elongation and survivalvs. alcoholic fermentation of rice under anoxia is examinedusing eight cultivars differing in submergence tolerance. Anoxiawas imposed on either 1 or 4 d aerated seeds either by N2 flushingsubmerged tissues or by incubating tissues in stagnant deoxygenatedagar at 0·1% w/v; the latter simulated the stagnant conditionsof waterlogged soil. Two cultivars that were most tolerant tosubmergence also had the greatest tolerance to anoxia, whilea submergence intolerant cultivar was also intolerant to anoxia. Coleoptile growth under anoxia was related to rates of ethanolsynthesis (RE), however differences between growth during anoxiaand survival after anoxia indicated that post-anoxic injurymay also be important in rice seeds exposed to anoxia. The correlationbetween coleoptile growth and RE measured on a tissue basisusing intact seeds was r2 = 0·67 among six varietiesover 0-3 d anoxia. This correlation improved to about r2 = 0·85using RE of (embryos plus coleoptiles) over 0-3 d, or coleoptilesat 3 d after anoxia. Coleoptile growth of individual seeds wasusually poorly correlated to RE in these cultivars at 2-3 dafter anoxia. When coleoptiles of similar lengths were obtainedfrom different cultivars using 4 d aerated seeds, there weredifferences in RE and coleoptile growth which were related tocoleoptile growth during 3 or 5 d anoxia, either on a tissue(r2 = 0·85) or a fresh weight basis (r2 = 0·70-0·97respectively). Chickpea cultivar ILC 482 was inoculated with salt-tolerantRhizobium strain Ch191 in solution culture with different saltconcentrations added either immediately with inoculation or5 d later. The inhibitory effect of salinity on nodulation ofchickpea occurred at 40 dS m–1 (34.2 mol m–3 NaCl)and nodulation was completely inhibited at 7 dS m–1 (61.6mol m–3 NaCl); the plants died at 8 dS m–1 (71.8mol m–3 NaCl). Chickpea cultivar ILC 482 inoculated with Rhizobium strain Ch191spcstrwas grown in two pot experiments and irrigated with saline water.Salinity (NaCl equivalent to 1–4 dS m–1) significantlydecreased shoot and root dry weight, total nodule number perplant, nodule weight and average nodule weight. The resultsindicate that Rhizobium strain Ch191 forms an infective andeffective symbiosis with chickpea under saline and non-salineconditions; this legume was more salt-sensitive compared tothe rhizobia, the roots were more sensitive than the shoots,and N2 fixation was more sensitive to salinity than plant growth. The effects of excess salinity and oxygen deficiency on growthand solute relations in Zea mays L. cv. Pioneer 3906 were examinedin greenhouse experiments. The roots of plants 14 d old growingin nutrient solution containing additions of NaCl in the range1.0–200 mol m–3 were either exposed to a severedeficiency of O2 by bubbling with nitrogen gas (N2 treatment),or maintained with a supply of air (controls), for a periodof 1–7 d. The threshold NaCl concentration resulting inappreciable inhibition of leaf extension, and shoot f. wt gainin controls was between 10 and 25 mol m–3. At 25 mol m–3NaCl the ratio of Na+/K+ transported to shoots was about 20times greater than in plants in 1.0 mol m–3 NaCl. Theeffect of addition of NaCl to the nutrient solution was to enhanceNa+ movement but simultaneously depress the rate of K+ transportto shoots (per g f. wt roots). Interactions between NaCl levels and aeration treatment wereshown by analyses of variance to be statistically significantfor leaf extension, shoot and root f. wt gains, Na+ and K+ concentrationsin shoots and roots. When roots were N2-treated, shoot and rootgrowth were depressed, the effect of aeration treatment beinggreatest at NaCl concentrations of 50 mol m–3 or less.Additionally, N2-treatment greatly accelerated Na- transportto shoots while depressing K+ transport still further, so thatat 10 mol m–3 NaCl the ratio Na+/K+ acquired by the shootswas 230 times greater than in controls. Over the concentrationrange 1.0 to 50 mol m–3 NaCl, the ratio Na+/K+ transportedto shoots by anoxic roots increased by a factor of 860. The relationship between coleoptile elongation and alcoholicfermentation of rice under anoxia is examined using seeds either:(a) N2 flushed during submergence, (b) incubated in stagnantdeoxygenated agar at 0·1% w/v to simulate the stagnantconditions of waterlogged soil, or (c) incubated in waterloggedsoil. Coleoptile elongation growth was greater for N2 flushing> stagnant agar > soil; seed survival was also greatestin this order over 1-5 d. Ethanol concentrations in coleoptiles and intact seeds (cv.IR42) were approximately 300 and 100 mol m-3 respectively whenseeds were grown 3 d in stagnant agar, however 92% of the ethanolin seeds diffused into the external medium when solutions weremixed for 5-10 s. Coleoptile growth under anoxia was relatedto rates of ethanol synthesis (RE) in different treatments;there was greater coleoptile growth and RE for seeds in N2 flushedsolutions than in stagnant deoxygenated agar. Coleoptile growthof individual seeds was also related to the RE of each seedat 2-3 d after anoxia (r2 = 0·46). Analysis of different tissues was important in evaluating growthand metabolism of coleoptiles. Although the coleoptile onlyaccounted for 0·7% of seed dry weight at 3 d after anoxia,it contained 21% of the ethanol produced by rice seeds. Therewere also three-fold higher rates of RE on a fresh weight basisin expanding tissues in the base of the coleoptile relativeto the elongated tissues at the apex. Root growth of 7-d-old wheat (Triticum aestivum cv. Gamenya)seedlings was impaired at dissolved O2 concentrations of 0.01and 0.055 mol m–3 O2, while growth at 0.115 mol m–3O2 was the same as that in continuously aerated controls (0.26mol m–3 O2). Oxygen uptake by apical (0–2 mm), expanding (2–4mm) and expanded (10–12 mm) tissues of the roots decreasedbelow 0.16, 0.09 and 0.05 mol m–3 O2, respectively. Thishierarchy is consistent with the metabolic rates of these tissues.There was a small (c. 9%) inhibition of O2 uptake and some netsynthesis of ethanol and alanine in root apices at 0.115 molm–3 O2. Significant amounts of anaerobic end-productsaccumulated at 0.055 mol m–3 O2 and even more so at 0.01mol m–3 O2, indicating that oxidative phosphorylationwas strongly inhibited. Net alanine synthesis increased in fully expanded (10–16mm) tissues exposed to <0.003–0.01 mol m–3 O2,and this increase was accompanied either by a proportionallysmaller increase in the concentration of other free amino acidsor by a net decrease in free amino acid levels excluding alanine.This suggests that alanine was synthesized as an end-productof anaerobic catabolism and did not accumulate simply becauseof decreased net protein synthesis. Comparing the carbon flow to CO2, ethanol, lactate and alaninein roots at 0.01 mol m–3 O2 with carbon loss as CO2 inaerated roots suggests that carbon flow to products of metabolismwas not greatly enhanced due to O2 deficiency. This study investigated aerenchyma formation and function in adventitious roots of wheat (Triticum aestivum L.) when only a part of the root system was exposed to O2 deficiency. Two experimental systems were used: (1) plants in soil waterlogged at 200 mm below the surface; or (2) a nutrient solution system with only the apical region of a single root exposed to deoxygenated stagnant agar solution with the remainder of the root system in aerated nutrient solution. Porosity increased two‐ to three‐fold along the entire length of the adventitious roots that grew into the water‐saturated zone 200 mm below the soil surface, and also increased in roots that grew in the aerobic soil above the water‐saturated zone. Likewise, adventitious roots with only the tips growing into deoxygenated stagnant agar solution developed aerenchyma along the entire main axis. Measurements of radial O2 loss (ROL), taken using root‐sleeving O2 electrodes, showed this aerenchyma was functional in conducting O2. The ROL measured near tips of intact roots in deoxygenated stagnant agar solution, while the basal part of the root remained in aerated solution, was sustained when the atmosphere around the shoot was replaced by N2. This illustrates the importance of O2 diffusion into the basal regions of roots within an aerobic zone, and the subsequent longitudinal movement of O2 within the aerenchyma, to supply O2 to the tip growing in an O2 deficient zone.  相似文献

Roots of intact wheat plants were grown for 7-12 d in stagnant nutrient solution, containing 0.1% agar, to mimic the lack of convection in waterlogged soil. Net K+ and P uptakes by seminal and nodal roots were measured separately using a split root system. For seminal roots in stagnant solution, net uptakes as a percentage of aerated roots were between 0% and 16% for P, while K+ ranged between 15% uptake and 54% loss. For the more waterlogging-tolerant nodal roots, net uptakes in stagnant nutrient solution, as a percentage of aerated roots, were 31-73% for P and 69-115% for K+. Elongation rates of nodal roots in stagnant nutrient were about 35-43% of those for roots in aerated solution. This partial inhibition occurred in these nodal roots despite their 15% porosity (v/v). Elevation of O2 partial pressures around the shoots to 40 kPa and then to 80 kPa substantially accelerated nodal root elongation in stagnant solution, demonstrating that most of the inhibition seen with ambient O2 around the shoots was associated with a restricted O2 supply to these nodal roots. Thus, in wheat nodal roots, with a partial pressure of 20 kPa O2 around the shoots, O2 diffusion from the shoots did not completely relieve the restrictions on elongation resulting from stagnancy in the nutrient solution. These results contrast with those in the literature for rice, in which roots function efficiently in stagnant solutions (0.1% agar). So, when wheat roots are aerenchymatous there are still restrictions to O2 diffusion in the gas space continuum between the atmosphere and the functional tissues of the roots. This poor acclimation must have been due to inefficiency of the aerenchymatous axes, which may include persistence of anoxic steles, and/or restricted O2 diffusion in other parts of the gas space continuum, in either the shoots and shoot-root junction or in the root tip.  相似文献

Nine species from the tribe Triticeae – three crop, three pasture and three ‘wild’ wetland species – were evaluated for tolerance to growth in stagnant deoxygenated nutrient solution and also for traits that enhance longitudinal O2 movement within the roots. Critesion marinum (syn. Hordeum marinum) was the only species evaluated that had a strong barrier to radial O2 loss (ROL) in the basal regions of its adventitious roots. Barriers to ROL have previously been documented in roots of several wetland species, although not in any close relatives of dryland crop species. Moreover, the porosity in adventitious roots of C. marinum was relatively high: 14% and 25% in plants grown in aerated and stagnant solutions, respectively. The porosity of C. marinum roots in the aerated solution was 1·8–5·4‐fold greater, and in the stagnant solution 1·2–2·8‐fold greater, than in the eight other species when grown under the same conditions. These traits presumably contributed to C. marinum having a 1·4–3 times greater adventitious root length than the other species when grown in deoxygenated stagnant nutrient solution or in waterlogged soil. The length of the adventitious roots and ROL profiles of C. marinum grown in waterlogged soil were comparable to those of the extremely waterlogging‐tolerant species Echinochloa crus‐galli L. (P. Beauv.). The superior tolerance of C. marinum, as compared to Hordeum vulgare (the closest cultivated relative), was confirmed in pots of soil waterlogged for 21 d; H. vulgare suffered severe reductions in shoot and adventitious root dry mass (81% and 67%, respectively), whereas C. marinum shoot mass was only reduced by 38% and adventitious root mass was not affected.  相似文献

Shoot specific growth rates (d. wt)doubled with increasing temperature between 7 and 17°C,whilst root specific growth rates showed little response; shoot:root ratios increased with root temperature, and over time at11°C. Net uptake of total N per plant (N2 fixation + NH4++ NO3) over 14 d increased three-fold between 3 and 17°C.The proportion contributed by N2 fixation decreased with increasingtemperature from 51% at 5°C to 18% at 25°C. Uptake ofNH4+ as a proportion of NH4+ + NO3 uptake over 14 d variedlittle (55–62%) with root temperature between 3 and 25°C,although it increased with time at most temperatures. Mean ratesof total N uptake per unit shoot f. wt over 14 d changed littlebetween 9 and 25°C, but decreased progressively with temperaturebelow 9°C, due to the decline in the rates of NH4+ and NO3uptake, even though N2 fixation increased. The results suggestthat N2 fixation in the presence of sustained low concentrationsof NH4+ and NO4 is less sensitive to low root temperaturethan are either NH4+ or NO3 uptake systems. Acclimation of NO3 transport fluxes (influx, efflux)in roots of oilseed rape (Brassica napus L. cv. Bien venu) andtheir sensitivity to growth at low root temperature was studiedin relation to external NO3 supply, defined by constantconcentrations ranging from sub- to supra-optimal with respectto plant growth rate. Plants were grown from seed in flowingnutrient solutions containing 250 mmol m–3 NO3at 17°C for 20d, and solution temperature in half the cultureunits was then lowered decrementally over 3 d to 7°C. Threedays later plants were supplied with NO3 at 1, 10, 100or 1000 mmol m–3 maintained for 18 d. Dry matter productionwas decreased more by low root zone temperature than low [NO3]e. Root specific growth rates were inversely related to [NO3]eand shoot:root ratios increased with time at [NO3]e between10–1000 mmol m–3. Net uptake of NO3 at 17°Cwas twice that at 7°C, and at both temperatures it doubledwith increasing [NO3]e between 1–10 mmol m–3with further small increases at higher [NO3]e. Mean unitabsorption rates of NO3 between 0–6 d and 6–14d were linearly related (r2 of 0.79–0.99) to log10[NO].Steady-state Q10 (7–17°C) for uptake between 0–6d were 0.91, 1.62, 1.27, and 1.10, respectively, at [NO3]eof 1, 10, 100, and 1000 mmol m–3, compared with correspondingvalues of 0.98, 1.38, 1.68, and 1.89 between 6–14 d. Thedata indicated that net uptake rates at 7 and 17°C divergedover time at high [NO3]e. Short-term uptake rates from1 mol m–3 NO3 measured at 17°C were higherin plants grown with roots at 7°C than at 17°C; for7°C plants there was a strong inverse linear relationship(r2=0.94) between uptake rate and treatment log10 [NO3]ewhilst rates in 17°C plants were independent of prior [NO3]e. Rates of NO3 influx and efflux under different steady-stateconditions of NO3 supply and root temperature were calculatedfrom dilution of 15N added to culture solutions. Efflux wassubstantial relative to net uptake in all treatments, and wasinversely related to [NO3]e at 17°C but not at 7°C.Ratios of influx: efflux ranged from 1.6–2.9 at 17°Cand 1.3–1.8 at 7°C, indicating the proportionatelygreater impact of efflux at low root temperature. Ratios ofefflux: net uptake were 0.53–1.56 at 17°C and 1.21–3.58at 7°C. The apparent sensitivities of influx and effluxto steady-state root temperature varied with [NO3]e.Both fluxes were higher at 17°C than 7°C in the presenceof 100–1000 mmol m–3 NO3 but the trend wasreversed at 1–10 mmol m–3 NO. Concentrations oftotal N measured in xylem exudate were at least 2-fold higherat 7°C compared with 17°C, attributable mainly to higherconcentrations of NO3 glutamine and proline. The resultsare discussed in terms of acclimatory and other responses shownby the NO3 transport system under conditions of limitingNO3 supply and low root temperature. When young plants of Macroptilium atropurpureum, cv. Siratrowere deprived of external sulphate (-S plants) growth of shootsand roots continued at rates comparable to those in plants wellsupplied with sulphate (control) for 3 d and 5 d respectively.Dilution of internal sulphur therefore took place and redistributionof sulphur occurred between inorganic and organic forms andbetween roots and younger leaves. Even when S-deficiency limitedgrowth, plants contained 16% of their total sulphur as sulphate,but most of this was retained in old leaves and redistributedslowly to growing zones. The capacity for sulphate uptake increased in roots of –Splants very soon after they were deprived of external sulphate;within 24 h the absorption from 0.25 mol m–3 SO42–was more than five times that of control roots. Maximum increasedcapacity was reached after 2–3 d stress when the Vmaxof system 1 was 1948 nmol h–1g–1root fr. wt. in–S plants and 337 nmol h–1g–1root fr. wt.in controls. The Kmfor system 1 did not change significantlywith S-stress being between 5–8 µM in both setsof plants. Absorption of L-cysteine was not stimulated by S-stress. There was a close, positive relationship between plant growthrate and the rate at which sulphate uptake capacity was enhancedby withholding sulphate from culture solutions. When –S plants were replaced in sulphate-containing solutiontheir capacity for SO42– declined to the control levelwithin 24 h. Very marked repression of capacity was also foundwhen –S plants were treated with L-cysteine, but therewas no immediate effect with methionine. Roots of this species appear to have a very active system fordegrading L-cysteine to sulphate, 30% of the label in 35S-cysteineabsorbed by roots was recovered in 35SO42– after 20 minor 2 h incubation. By contrast, roots had a very weak abilityto reduce sulphate. When part of the root system was in solution lacking sulphatethere was enhanced uptake of sulphate by other parts which themselveswere amply supplied with sulphate. This is seen as an exampleof compensatory absorption. The response to S-stress is specific and there were no positiveinteractions between S-stress and the absorption of phosphate,or P-stress and the uptake of sulphate. The results are discussed in relation to the close control ofsulphate uptake by internal sulphate concentration, redistributionof forms of sulphur during stress and mobility of sulphate inthe phloem. Barley plants (Hordewn vulgare L. cv. Atem) were grown fromseed for 28 d in flowing solution culture, during which timeroot temperature was lowered decrementally to 5?C. Plants werethen subjected to root temperatures of 3, 5, 7, 9, 11, 13, 17or 25 ?C, with common air temperature of 25/15 ?C (day/night).Changes in growth, plant total N, and NO3 levels, andnet uptake of NH4+ and NO3 from a maintained concentrationof 10 mmol m–3 NH4NO3 were measured over 14 d. Dry matterproduction increased 6-fold with increasing root temperaturebetween 3–25 ?C. The growth response was biphasic followingan increase in root temperature. Phase I, lasting about 5 d,was characterized by high root specific growth rates relativeto those of the shoot, particularly on a fresh weight basis.During Phase I the shoot dry weight specific growth rates wereinversely related to root temperature between 3–13 ?C.Phase 2, from 5–14 d, was characterized by the approachtowards, and/or attainment of, balanced exponential growth betweenshoots and roots. Concentrations of total N in plant dry matterincreased with root temperature between 3–25 ?C, moreso in the shoots than roots and most acutely in the youngestfully expanded leaf (2?l–6?9% N). When N contents wereexpressed on a tissue fresh weight basis the variation withtemperature lessened and the highest concentration in the shootwas at 11 ?C. Uptake of N increased with root temperature, andat all temperatures uptake of NH4+, exceeded that of NO3,irrespective of time. The proportions of total N uptake over14 d absorbed in the form of NH4+ were (%): 86, 91, 75, 77,76, 73, 77, and 80, respectively, at 3, 5, 7, 9, Il, 13, 17,and 25 ?C. At all temperatures the preference for NH4+ overNO3 uptake increased with time. An inverse relationshipbetween root temperature (3–11 ?C) and the uptake of NH4+as a proportion of total N uptake was apparent during PhaseI. The possible mechanisms by which root temperature limitsgrowth and influences N uptake are discussed. Rice seedlings (Oryza saliva L.) were grown in the dark forup to 4 d in solutions containing various concentrations ofO2. Compared with seedlings grown at 0·250 mol O2 m–3,the dry weight of the growing seedling was 14% lower at 0·110mol O2 m–3 and 60% lower at 0 mol O2 m–3. Decreasesin fresh weight were similar but not identical to decreasesin dry weight, possibly because leaf growth was suppressed evenabove 0·110 mol O2 m–3. Oxygen deficiency inhibitedroot growth more severely than coleoptile growth. Coleoptiles from seedlings grown in aerated solution were exposedto an atmosphere of pure N2 for 30 min. Anoxia caused a declinein ATP content and energy charge, suggestive of decreased oxidativephosphorylation. It is not clear whether the decline in oxidativephosphorylation was solely responsible for impaired growth inhypoxia. In seedlings growing at O2 concentrations less than 0·110mol O2 m–3, significant amounts of ethanol were synthesized.The rate of O2 uptake decreased markedly below 0·06 molO2 m–3; this was presumably near the external O2 concentrationat which oxidative phosphorylation became limited by the supplyof O2. The stage of development of the seedlings appeared toinfluence O2 uptake, possibly through changes in conductanceof the tissue to O2. Uncouplers were used to confirm that thecritical O2 concentration was dependent on O2 diffusion ratherthan enzyme kinetics. Impaired growth above 0·110 molO2 m–3 may have been due to a decreased activity of oxygenasesof relatively low affinity for O2, which in turn altered cellmetabolism. Soybean [Glycine max (L.) Merrill] plants that had been subjectedto 15 d of nitrogen deprivation were resupplied for 10 d with1.0 mol m–3 nitrogen provided as NO3, NH4+, orNH4++NO3 in flowing hydroponic culture. Plants in a fourthhydroponic system received 1.0 mol m–3 NO3 duringboth stress and resupply periods. Concentrations of solublecarbohydrates and organic acids in roots increased 210 and 370%,respectively, during stress. For the first day of resupply,however, specific uptake rates of nitrogen, determined by ionchromatography as depletion from solution, were lower for stressedthan for non-stressed plants by 43% for NO3- resupply, by 32%for NH4+ + NO3 resupply, and 86% for NH4+ resupply. Whenspecific uptake of nitrogen for stressed plants recovered torates for non-stressed plants at 6 to 8 d after nitrogen resupply,carbohydrates and organic acids in their roots had declinedto concentrations lower than those of non-stressed plants. Recoveryof nitrogen uptake capacity of roots thus does not appear tobe regulated simply by the content of soluble carbon compoundswithin roots. Solution concentrations of NH4+ and NO3 were monitoredat 62.5 min intervals during the first 3 d of resupply. Intermittent‘hourly’ intervals of net influx and net effluxoccurred. Rates of uptake during influx intervals were greaterfor the NH4+ -resupplied than for the NO3 -resuppliedplants. For NH4+ -resupplied plants, however, the hourly intervalsof efflux were more numerous than for NO3 -resuppliedplants. It thus is possible that, instead of repressing NH4+influx, increased accumulation of amino acids and NH4+ in NH4+-resupplled plants inhibited net uptake by stimulation of effluxof NH4+ absorbed in excess of availability of carbon skeletonsfor assimilation. Entry of NH4+ into root cytoplasm appearedto be less restricted than translocation of amino acids fromthe cytoplasm into the xylem.
