Summary Absorption of nitrate and ammonium was studied in water culture experiments with 4 to 6 weeks old plants of barley (Hordeum vulgare L.), buckwheat (Fagopyrum esculentum L. Moench) and rape (Brassica napus L.). The plants were grown in a complete nutrient solution with nitrate (5.7±0.2 mM) or nitrate (5.6±0.2 mM) + ammonium (0.04±0.02 mM). The pH of the nutrient solution was kept at 5.0 using a pH-stat. It was found that phosphorus deficiency reduced the rate of nitrate uptake by 58±3% when nitrate was the sole N source and by 83±1% when both nitrate and ammonium were present. The reduction occurred even before growth was significantly impeded by P deficiency. The inhibition of the uptake of ammonium was less,i.e. ammonium constituted 10±1% of the total N uptake in the P sufficient plants and 30±5% in the P deficient plants. The reduction of nitrate absorption greatly decreased the difference between the uptake of anions and cations. It is suggested that P deficiency reduced the assimilation of NO 3 into the proteins, which might cause a negative feedback on NO 3 influx and/or stimulate NO 3 efflux.  相似文献

Deprivation of nitrogen (N) increases assimilate partitioning towards roots at the expense of that to shoots. This study was done to determine the physiological basis of increased root growth of tea (sCammellia sinensis L.) under N shortage. Nine-month-old clonal tea (clone TRI2025) was grown in quartz sand under naturally lit glasshouse conditions. Three levels of N (0, 3.75 and 7.5 mM N) were incorporated in to the nutrient solution and applied daily. Plant growth, photosynthesis, root respiration and plant N contents were measured at 10-day intervals over a 45-day period. Root dry weight showed a sharp increase during the first 15 days after the plants were transferred to 0 mM N, whereas no such increase was shown in plants transferred to 7.5 mM N. In contrast, shoot dry weight increased at 7.5 mM N and was significantly greater than at 0 mM N, where no increase was observed. Due to the above changes, root weight ratio increased and leaf weight ratio decreased during the first 15 days of N deprivation. Leaf photosynthetic rates did not vary between N levels during the initial 15-day period. Thereafter, photosynthetic rates were greater at 7.5 mM and 3.75 mM N than at 0 mM N. Root respiration rate decreased at 0 mM N, whereas it increased at 3.75 and 7.5 mM N, probably because of the greater respiratory cost for nitrate uptake. Root respiratory costs associated with maintenance (R m) and nitrate uptake (R u) were calculated to investigate whether the sharp increase of root growth observed upon nitrogen deprivation was solely due to the reduced respiratory costs for nitrate uptake. The estimated values for R m and R u were 3.241 × 10–4 mol CO2 g–1 (root dry matter) s–1 and 0.64 mol CO2 (mol N)–1, respectively. Calculations showed that decreased respiratory costs for nitrate uptake could not solely account for the significant increase of root biomass upon N deprivation. Therefore, it is concluded that a significant shift in assimilate partitioning towards roots occurs immediately following N deprivation in tea.  相似文献

Ammonia-oxidizing bacteria (AOB) populations were studied on the root surface of different rice cultivars by PCR coupled with denaturing gradient gel electrophoresis (DGGE) and fluorescence in situ hybridization (FISH). PCR-DGGE of the ammonium monooxygenase gene (amoA) showed a generally greater diversity on root samples compared to rhizosphere and unplanted soil. Sequences affiliated with Nitrosomonas spp. tended to be associated with modern rice hybrid lines. Root-associated AOB observed by FISH were found within a discrete biofilm coating the root surface. Although the total abundance of AOB on root biofilms of different rice cultivars did not differ significantly, there were marked contrasts in their population structure, indicating selection of Nitrosomonas spp. on roots of a hybrid cultivar. Observations by FISH on the total bacterial community also suggested that different rice cultivars support different bacterial populations even under identical environmental conditions. The presence of active AOB in the root environment predicts that a significant proportion of the N taken up by certain rice cultivars is in the form of NO3 -N produced by the AOB. Measurement of plant growth of hydroponically grown plants showed a stronger response of hybrid cultivars to the co-provision of NH4 + and NO3 . In soil-grown plants, N use efficiency in the hybrid was improved during ammonium fertilization compared to nitrate fertilization. Since ammonium-fertilized plants actually receive a mixture of NH4 + and NO3 with ratios depending on root-associated nitrification activity, these results support the advantage of co-provision of ammonium and nitrate for the hybrid cultivar.  相似文献

Soluble aluminium (Al) is a major factor limiting plant growth in acid mineral soils. Aluminium concentrations in soil solutions are mainly determined by soil pH. However, pH also affects the ratio between activities of protons and cationic Al species and the equilibrium between mono-and polynuclear hydroxy-Al species. The phytotoxicity of these species is not yet clear. The objective of the present study was to clarify the role of minor changes of pH in the rhizosphere on Al phytotoxicity in two Al-tolerant plant species by direct control of the pH in the nutrient solution (4.1, 4.3, 4.5) and in addition by varying the pH in the root apoplast using either nitrate or ammonium as N source. The plants were grown in solution culture at constant external pH. Whereas the Al-sensitive plant species barley and horse bean were damaged at very low Al supplies (1.85 μM and 9.3 μM respectively), 222 μM had to be applied to rye and yellw lupin for a comparable inhibition of root elongation. Yellow lupin was initially severely inhibited in root growth by Al, but then gradually recovered from this ‘Al shock’ within 3 days. In contrast to lupin, rye was hardly affected by Al initially, and it took about 16 h until maximum inhibition of root elongation. In the presence of nitrate, raising the pH from 4.1 to 4.5 aggravated root-growth depression by Al in rye and lupin. Whereas rye roots were severely damaged by ammonium especially at low pH, lupin was rather indifferent to the N source. Aluminium toxicity was less severe in presence of ammonium compared to nitrate N. This effect was less clear with rye at lower pH, because of it's higher proton sensitivity compared to lupin. Less Al injury at lower pH and in presence of ammonium was related to lower Al concentrations in the 1 cm root tips. The results are compatible with data showing high phytotoxicity of mononuclear and polynuclear hydroxy-Al species. However, they could also be interpreted in the light of proton amelioration of Al toxicity owing to competition for Al-sensitive binding sites in the root apoplast.  相似文献

The experiment was set up to examine the influence of different nitrogen forms: (NH4)2SO4, Ca(NO3)2 or NH4NO3 on growth response, root induced pH changes in the rhizosphere, root-borne acid phosphatase activity in strawberry plants cv. Senga Sengana. The plants grown on sandy mineral soil were fertilized with 3 forms of nitrogen, in concentrations of 46 mg N·kg−1 soil. The plants were grown in rhizoboxes with removable plexiglass lids. To ensure the root growth along the plexiglass lids, the rhizoboxes were placed at an angle of about 50° with the lid on the lower side. In case of ammonium supply, the nitrification inhibitor DIDIN was added (10 mg·kg−1 of moist soil) to prevent conversion of ammonium into nitrate. The growth response (roots and shoots) of strawberry plants were determined after 11 weeks of treatment with different N forms. The best development of the root system and shoots (root and shoot dry weight and root length) was obtained, when ammonium nitrate was supplied. It is suggested therefore, that NH4NO3 stimulates vegetative growth of strawberry plants cv. Senga Sengana. However, there were no statistical differences in a leaf and flower number of the plants grown under different forms of N-fertilization. Determination of rhizosphere pH, and acid phosphatase activity were executed using non-destructive techniques, which enabled weekly measurement of chemical changes in the rhizosphere. The results revealed that the form of nitrogen supplied had a predominant effect on chemical changes in the rhizosphere of strawberry plants. The highest pH values (average pH 6.8) were measured in the rhizosphere of individual plants supplied with Ca(NO3)2. Whereas the lowest pH values (average pH 5.8) were detected in the presence of (NH4)2SO4. The curve of rhizosphere pH measured along individual roots of the plants treated with Ca(NO3)2 represents the highest pH values whereas the curve of rhizosphere pH under (NH4)2SO4 treatment had the lowest pH values. The highest activity of acid phosphatase were observed in the rhizosphere of strawberry plants grown in the presence of (NH4)2SO4, at pH 5.8.  相似文献

Our previous work indicated that salinity caused a shift in the predominant site of nitrate reduction and assimilation from the shoot to the root in tomato plants. In the present work we tested whether an enhanced supply of dissolved inorganic carbon (DIC, CO2+ HCO3) to the root solution could increase anaplerotic provision of carbon compounds for the increased nitrogen assimilation in the root of salinity-stressed Lycopersicon esculentum (L.) Mill. cv. F144. The seedlings were grown in hydroponic culture with 0 or 100mM NaCl and aeration of the root solution with either ambient or CO2-enriched air (5000 μmol mol?1). The salinity-treated plants accumulated more dry weight and higher total N when the roots were supplied with CO2-enriched aeration than when aerated with ambient air. Plants grown with salinity and enriched DIC also had higher rates of NO?3 uptake and translocated more NO?3 and reduced N in the xylem sap than did equivalent plants grown with ambient DIC. Incorporation of DIC was measured by supplying a 1 -h pulse of H14CO?3 to the roots followed by extraction with 80% ethanol. Enriched DIC increased root incorporation of DIC 10-fold in both salinized and non-salinized plants. In salinity-stressed plants, the products of dissolved inorganic 14C were preferentially diverted into amino acid synthesis to a greater extent than in non-salinized plants in which label was accumulated in organic acids. It was concluded that enriched DIC can increase the supply of N and anaplerotic carbon for amino acid synthesis in roots of salinized plants. Thus enriched DIC could relieve the limitation of carbon supply for ammonium assimilation and thus ameliorate the influence of salinity on NO?3 uptake and assimilation as well as on plant growth.  相似文献

Eggplants (Solanum melongena L. cv. Bonica) were grown in a glasshouse during summer under natural light with one unbranched shoot or one shoot with 3 to 4 branches and with or without fruit in quartz sand buffered and not buffered with 0.5% CaCO3 (w : v), respectively. Nutrient solutions supplied contained nitrate or ammonium as the sole nitrogen source. Compared with nutrient solutions containing nitrate (10 mM), solutions containing ammonium (10 mM) caused a decrease in net photosynthesis of eggplants during early stages of vegetative growth when grown in quartz sand not buffered with CaCO3. The decrease was not observed before leaves showed interveinal chlorosis. In contrast, net photosynthesis after bloom at first increased more rapidly in eggplants supplied with ammonium than with nitrate nitrogen. However, even in this case, net photosynthesis decreased four weeks later when ammonium nutrition was continued. The decrease was accompanied by epinasty and interveinal chlorosis on the lower leaves and later by severe wilting, leaf drop, stem lesions, and hampered growth of stems, roots, and fruits. These symptoms appeared later on plants not bearing fruits than on plants bearing fruits. If nutrient solutions containing increasing concentrations of ammonium (0.5–30 mM) were supplied after the time of first fruit ripening, shoot growth and set of later flowers and fruits were promoted. In contrast, vegetative growth and reproduction was only slightly affected by increasing the concentration of nitrate in the nutrient solutions. In quartz sand buffered with CaCO3 ammonium nutrition caused deleterious effects only under low light conditions (shade) and on young plants during rapid fruit growth. If eggplants were supplied with ammonium nitrogen before bloom, vegetative growth was promoted, and set of flowers and fruit occurred earlier than on plants supplied with nitrate. Furthermore, the number of flowers and fruit yield increased. These effects of ammonium nutrition were more pronounced when plants were grown with branched shoots than with unbranched shoots. The results indicate that vegetative and reproductive growth of eggplants may be manipulated without causing injury to the plants by supplying ammonium nitrogen as long as the age of the plants, carbohydrate reserves of the roots, quantity of ammonium nitrogen supplied, and pH of the growth medium are favourable. Increased levels of rhizospheric dissolved inorganic carbon have repeatedly been demonstrated to enhance plant growth by up to 80%, although carbon from dark fixation accounts for only 1–3% of total plant carbon gain. This study, therefore, aimed at investigating the effects of bicarbonate on nitrate uptake, assimilation and translocation to shoots. Clonal saplings of poplar (Populus canescens(Ait.) Sm.) and elder (Sambucus nigraL.) were grown hydroponically for 35 days in a nutrient solution containing 0, 0.5 and 1 mM bicarbonate and 2 mM nitrate as the sole nitrogen source at pH 7.0. Net nitrate uptake, root nitrate accumulation and reduction, and export of nitrogenous solutes to shoots were measured after incubating plants with 15N-labelled nitrate for 24 h. Net nitrate uptake increased non-significantly in plant species (19–61% compared to control plants) in response to 1 mM bicarbonate. Root nitrate reduction and nitrogen export to shoots increased by 80 and 95% and 15 and 44% in poplar and elder, respectively. With enhanced root zone bicarbonate, both species also exhibited a marked shift between the main nitrate utilising processes. Poplar plants increasingly utilised nitrate via nitrate reduction (73–88% of net nitrate uptake), whereas the proportions of export (20–9%) and storage in roots (7–3%) declined as plants were exposed to 1 mM external bicarbonate. On the other hand, elder plants exhibited a significant increase of root nitrate reduction (44–66%) and root nitrate accumulation (6–25%). Nitrate translocation to elder shoots decreased from 50 to 8% of net nitrate uptake. The improved supply of nitrogen to shoots did not translate into a significant stimulation of growth, relative growth rates increased by only 16% in poplar saplings and by 7% in elder plants. The modelling of ion uptake by plants requires the measurement of kinetic and growth parameters under specific conditions. The objective of this study was to evaluate the effect of nine NH inf4 sup+ :NO inf3 sup− ratios on onions (Allium cepa L.). Twenty-eight to 84 day-old onion plants were treated with NH inf4 sup+ :NOf3/sup− ratios ranging from 0 to 100% of each ionic species in one mM solutions in a growth chamber. Maximum N influx (Imax) was assessed using the N depletion method. Except at an early stage, ionic species did not influence significantly Imax, the Michaelis constant (Km) and the minimum concentration for net uptake (Cmin). Imax for ammonium decreased from 101 to 59 pmole cm-2 s-1 while Imax for nitrate increased from 26 to 54 pmole cm-2 s-1 as the plant matured. On average, Km and Cmin values were 14.29 μM, and 5.06 μM for ammonium, and 11.90 μM and 4.54 μM for nitrate, respectively. In general, the effect of NH4 +:NO3 - ratios on root weight, shoot weight and total weight depended on plant age. At an early stage, maximum plant growth and N uptake were obtained with ammonium as the sole source of N. At later stages, maximum plant growth and N uptake were obtained as the proportion of nitrate increased in the nutrient solution. The was no apparent nutrient deficiency whatever NH4 +:NO3 - ratio was applied, although ammonium reduced the uptake of cations and increased the uptake of phosphorus. Mesquite plants (Prosopis glandulosa var. Torreyana) were grown in 2-m long columns 20 cm in diameter, and provided with a constant, stable ground water source 10 cm above the sealed base of the column. Ground water contained 0, 1 or 5 mM nitrate, or a mixed salt solution (1.4, 2.8, or 5.6 dS m-1) with the ionic ratios of ground water found in a field stand of Prosopis at Harper's Well (2.8 dS m-1). Water uptake in the highly salinized columns began to decrease relative to low salt columns when soil salinity probes 30 cm above the column base read approximately 28 dS m-1, a potential threshold for mesquite salt tolerance. Prosopis growth increased with increasing nitrate, and decreased with increasing salinity. Water use efficiency was little affected by treatment, averaging approximately 2 g dry matter L-1 water used. Most fine roots were recovered from a zone about 25 cm above the ground water surface where water content and aeration appeared to be optimal for root growth. Root-shoot ratio was little affected by nitrate, but increased slightly with increasing salinity. Plant tissue P concentrations tended to increase with increasing salinity and decrease with increasing N, approaching potentially deficient foliage concentrations at 5 mM nitrate. The whole-plant leaf samples increased in sodium concentration both with added salt and with added nitrate. Foliar manganese concentrations increased with increasing salt in the absence of nitrate. Concentrations of sodium in leaves were low (<10 g kg-1), considering the high salt concentrations in the ground water. Prosopis appears to exclude sodium very effectively, especially from its younger leaves. Although Prosopis is highly salt tolerant, the degree to which it utilizes soil nitrate in place of biologically fixed N may lower its salinity tolerance and affect its nutrient relations in phreatic environments.  相似文献

Possible Involvement of Cytokinin in Nitrate-mediated Root Growth in Maize   总被引:1,自引:1,他引:0  
Response of root system architecture to nutrient availability in soils is an essential way for plants to adapt to soil environments. Nitrate can affect root development either as a result of changes in the external concentration, or through changes in the internal nutrient status of the plant. Nevertheless, less is known about the physiological mechanisms. In the present study, two maize (Zea mays L.) inbred lines (478 and Wu312) were used to study a possible role of cytokinin in nitrate-mediated root growth in nutrient solutions. Root elongation of 478 was more sensitive to high nitrate supply than that of Wu312. Medium high nitrate (5 mM) inhibited root elongation in 478, while, root elongation in Wu312 was only inhibited at high NO 3 supply (20 mM). Under high nitrate supply, the root elongation zone in 478 became swollen and the site of lateral root elongation was close towards the root tip. Both of the phenomena are typical of root growth induced by exogenous cytokinin treatments. Correspondingly, zeatin and zeatin nucleotide (Z + ZR) concentrations were increased at higher nitrate supply in 478, whereas they were constant in Wu312. Furthermore, exogenous cytokinin 6-benzylaminopurine (6-BA) completely reversed the stimulatory effect of low nitrate on root elongation. Therefore, it is supposed that the inhibitory effect of high concentration of nitrate on root elongation is, at least in part, mediated by increased cytokinin level in roots. High nitrate supply may have negative influences on root apex activity by affecting cytokinin metabolism so that root apical dominance is weakened and, therefore, root elongation is suppressed and lateral roots grow closer to the root apex. Nitrate suppressed lateral root elongation in Wu312 at concentration higher than 5 mM. In 478, however, this phenomenon was not significant even at 20 mM nitrate. Although exogenous 6-BA (20 nM) could suppress lateral root elongation as well, the inhibitory effect of high NO 3 concentration of nitrate on lateral root growth cannot be explained by changes in endogenous cytokinin alone.  相似文献

The effects of nitrogen form and phosphorus source on the growth, nutrient uptake and rhizosphere soil property of tea (Camellia sinensis L.) were investigated in a pot experiment. The experiment was performed with a compartmental cropping device, which enables the collection of rhizosphere soil at defined distances from the root of tea plant. Nitrogen was supplied as nitrate or ammonium in combination with soluble phosphorus as Ca(H2PO4)2 or insoluble P as rock phosphate. The leaf dry matter production of tea was significantly greater in the treatments with NH4 + than NO3 -, whereas dry matter production of root and stem was not significantly affected. Addition of phosphorus as either source did not influence the dry matter production. The concentrations of K in root, Mg and Ca in both the shoot and root supplied with NO3 - were significantly higher than in NH4 + and influence of P sources was minor. On the contrary, Al and Mn concentrations were significantly larger in NH4 --fed plants which could be attributed to remarkably increased availability of Al and Mn caused by acidification of the rhizosphere soil (the first 1-mm soil section from the root surface) with NH4–N nutrition. The concentration of N in shoot was also significantly higher in NH4- than in NO3-fed plants, indicating higher use efficiency of NH4–N. Whatever the phosphate source, rhizosphere pH declined in ammonium compared to in nitrate treatment. The pH decrease was much larger when no P or soluble P were applied and reached 0.85–1.30 units which extended to 3–5 mm away from the root surface. Exchangeable acidity, content of exchangeable Al and Mn were also considerably higher in the rhizosphere soils of NH4 + fed tea plants. Significant amounts of P dissolved from rock phosphate accumulated in rhizosphere of NH4 +, not NO3 -, suggesting that the dissolution of rock phosphate was induced by the proton excreted by tea root fed with ammonium. With soluble P addition, shoot and root P concentrations were greater in NH4 + than in NO3 - treatment and it appeared that this difference could not be sufficiently explained by the available P content in soil which was only slightly higher in NH4 + treatment. With rock phosphate addition, the shoot and root P concentrations were hardly affected by nitrogen form, although the available P content was much higher and accumulated in the rhizosphere soil supplied with ammonium. 采用自制根盒试验,主要研究了脲酶抑制剂氢醌(HQ),硝化抑制剂双氰胺(DCD)及二者组合对离水稻根际不同距离处NH4^--N和NO3^-N分布的影响,结果表明,DCD及其与HQ组合均能显著促进稻株地上部分生长,始终显著降低水稻根际与近根际土中NH4^ -N含量直至施肥后60d,施肥后20d时,DCD及其与HQ组合可使非根际土中NH4^ -N含量显著增加,随后,却出现相反现象,施肥后20d时,距根际不同距离的土壤中,配施DCD或DCD+HQ处理均能显著降低NO3^-N含量,随后,近根际和非根际仍保持上述现象直至施肥后40d;同未施DCD处理相比,根际土壤却较早出现NO3^--N含量高峰,正好与水稻N营养需求时期相一致,因此,DCD及其与HQ组合可减少水稻根际环境下尿素N损失潜势,通过不种稻土壤和距根际3cm处的土壤中尿素无机氮形态分布的差异,充分显示了研究水稻根际土壤氮素转化及相关抑制剂对其影响时,以取离根际3cm外的土壤作为非根际明显优于不种稻土壤。  相似文献

One-year old nectarine trees [Prunus persica, Batsch var. nectarina (Ait.) Maxim.], cv Nectaross grafted on P.S.B2 peach seedlings [Prunus persica (L.) Batsch] were grown for five months in 4-litre pots filled with two alkaline soils, one of which was also calcareous. Soils were regularly subjected to fertigation with either ammonium sulphate or calcium nitrate providing a total of 550 mg N/tree. Trees were also grown in such soils receiving only deionized water, as controls. Rhizosphere pH, measured by the use of a microelectrode inserted in agar sheet containing a bromocresol purple as pH indicator and placed on selected roots, was decreased by about 2–3 units compared to the bulk soil pH in all treatments. This decrease was slightly less marked when plants were supplied with calcium nitrate rather than ammonium sulphate or control. Measurements conducted during the course of the experiment indicated that ammonium concentration was similar in the solution of soils receiving the two N fertilizers. During the experiment, soil solution nitrate-N averaged 115 mg L–1 in soil fertilized with calcium nitrate, 68 mg L–1 in those receiving ammonium sulphate and 1 mg L–1 in control soils. At the end of the experiment nitrate concentrations were similar in soils receiving the two N sources and bulk soil pH was decreased by about 0.4 units by ammonium sulphate fertigation: these evidences suggest a rapid soil nitriflcation activity of added ammonium. Symptoms of interveinal chlorosis in apical leaves appeared during the course of the experiment in trees planted in the alkaline-calcareous soil when calcium nitrate was added. The slightly higher rhizosphere pH for calcium nitrate-fed plants may have contributed to this. The findings suggest that using ammonium sulphate in a liquid form (e.g. by fertigation) in high-pH soils leads to their acidification and the micronutrient availability may be improved.  相似文献

Douglas-fir, grown on strongly acid soil (pH-H2O 3.87), was fertilized with ammonium, nitrate or ammonium nitrate as N-source. Rhizosphere pH along the root axis was measured with microelectrodes. Pure ammonium supply resulted in acidification of the rhizosphere, almost along the entire root axis; only the extreme root tip was slightly alkaline compared with the bulk soil. With nitrate supply, the alkalization of the rhizosphere at the root tip was considerably stronger than with ammonium supply and the length of the alkalization zone greater, extending over the entire growth zone of the root. Acidification of the rhizosphere along the older parts of the root was less pronounced in the case of nitrate. It is concluded that nitrate nutrition enables the plant to protect its most essential root zone from the adverse effects of strong acidity by locally raising the rhizosphere pH.  相似文献

The impact of two constant non-toxic levels of Al stress (0.2 and 0.4 mM) on growth and 32P uptake capacity on sub-optimal (P-limited) Betula pendula seedlings grown in sand culture was examined.Seedling growth was under optimum controlled conditions in a growth chamber where nutrient additions were made at a predetermined relative addition rate (RA) of 10% day-1. Three treatment groups of seedlings 0, 0.2 and 0.4 mM Al were harvested at 15, 29 and 42 days. The excised roots were exposed to a 32P-labelled solution for 15 minutes to measure their capacity for P uptake. Growth was determined by weighing the roots, stems and leaves of the seedlings.Growth data showed that relative growth rate (RG) should equal the RA of P the most limiting nutrient, which was supplied at P/N 3% instead of an optimal 15%. Therefore, Ingestad's theory can also be used succesfully in sand culture and this may be particularly important for future studies of root and rhizosphere exudates. Low levels of Al (< 0.2 mM) in combination with low P supply significantly lowered the RG of the birch seedlings by further reducing P supply. However, previous studies of birch seedling growth and nutrient uptake using Ingestad's solution culture technique with optimumal P supply did not show any effect of Al on growth untill the Al was in excess of 3 mM. Aluminium was not directly toxic to the plants and therefore roots could respond to the 32P bioassay.  相似文献

 Three-year-old Norway spruce trees were planted into a low-nitrogen mineral forest soil and supplied either with two different levels of mineral nitrogen (NH4NO3) or with a slow-release form of organic nitrogen (keratin). Supply of mineral nitrogen increased the concentrations of ammonium and nitrate in the soil solution and in CaCl2-extracts of the rhizosphere and bulk soil. In the soil solution, in all treatments nitrate concentrations were higher than ammonium concentrations, while in the soil extracts ammonium concentrations were often higher than nitrate concentrations. After 7 months of growth, 15N labelled ammonium or nitrate was added to the soil. Plants were harvested 2 weeks later. Keratin supply to the soil did not affect growth and nitrogen accumulation of the trees. In contrast, supply of mineral nitrogen increased shoot growth and increased the ratio of above-ground to below-ground growth. The proportion of needle biomass to total above-ground biomass was not increased by mineral N supply. The atom-% 15N was higher in younger needles than in older needles, and in younger needles higher in plants supplied with 15N-nitrate than in plants supplied with 15N-ammonium. Ammonium sulphate is a major component of the air pollutants deposited on forests in the Netherlands. Different amounts of NH4 + were added to Douglas-fir seedlings grown in tall containers of sand, to study the influence of high concentrations of NH4 + in the soil on the development of fine roots and the effects of nitrogen uptake on rhizosphere pH. At the end of this eight-month experiment part of the ammonium appeared to have nitrified into nitrate. High doses of ammonium negatively affected root length and root length per unit of dry matter (specific root length). Although Douglas fir shows a preferential ammonium uptake in nutrient solutions the increases in the pH of the rhizosphere in this experiment indicate that nitrogen was mostly taken up as nitrate. When the ammonium concentration in the soil is low, it cannot be taken up readily because of its low mobility in soil. Shoot growth was stimulated by high availability of nitrogen. The possible effects of high doses of ammonium on long-term forest vitality are discussed.  相似文献

Bacterial growth rates on the rhizoplane of rape seedlings grown in sand were determined using 3H-thymidine incorporation into DNA. Axenic roots incorporated thymidine into DNA, which had to be subtracted from values for roots with associated bacteria. Thymidine incorporation into rhizoplane bacterial DNA ranged between 0.6 and 1.4 pmol thymidine h–1 root–1 for 6 to 26-day-old plants. Using a conversion factor, the turnover time of bacteria was calculated to decrease from 9.2 h for 6-day-old plants to 160h for 26-day-old plants. A similar value was found for rhizosphere bacteria of plants grown for 26 days in natural soil.  相似文献

Nodulation in pea (Pisum sativum L.) grown in hydroponic and sand culture systems is stimulated by low concentrations (<1.0 mM) of ammonium, but the physiological mechanisms underlying this stimulation are unknown. The current study involves a series of experiments, which investigate if the ammonium‐induced stimulation of nodulation involves changes in endogenous hormone (auxin and cytokinin) levels. P. sativum L. cv. Express was grown in growth pouches for 1 week with mineral N (0.5 and 2.0 mM NH4+ or NO3) or for 3 weeks exposed to exogenous indole‐3‐acetic acid (IAA) or 6‐benzylaminopurine (BAP) at a range of concentrations (10‐9?10‐5 M). Ammonium enhanced nodulation on the basis of both early whole plant (nodules plant?1) and specific nodulation (nodules g?1 root DW), especially in 0.5 mM treatment in which nodulation was approximately 4‐fold of the mineral‐N‐free control 1 week after inoculation. Correspondingly, the roots treated with ammonium contained much higher levels of t‐zeatin (Z) and lower t‐zeatin riboside (ZR) than that the control or nitrate‐treated plants. There was no significant difference in IAA levels between the control and ammonium treatments. Exogenous application of BAP for 3 weeks at concentrations of 10‐7?10‐5 M strongly inhibited nodulation. However, 10?9 M BAP, but not IAA, significantly enhanced nodulation. These data support the theory that a relatively high ratio of cytokinin:auxin in roots is favourable for nodule initiation, but that an excessively high level of cytokinin inhibits nodulation. Based on these results we propose that stimulation of nodulation by low concentrations of ammonium may be mediated through increasing Z level in roots, which alters the balance of cytokinin and auxin, which in turn induces cortical cell divisions leading to nodule initiation.  相似文献
