目的:利用酵母双杂交技术筛选人肝cDNA文库中与类固醇激素合成急性调节蛋白(STAR)相互作用的蛋白质。方法:将全长STAR基因克隆到酵母表达载体pDBLeu中,形成诱饵;将构建好的诱饵质粒转化至AH109酵母感受态中,利用酵母双杂交技术筛选人肝cDNA文库中与其相互作用的蛋白质,并进行相互作用的回转验证。结果:构建了酵母诱饵蛋白表达质粒pDBLeu-STAR,并筛选到6个猎物,其中有5对相互作用回转验证阳性。结论:为进一步研究STAR的功能和作用机制提供了新的线索。  相似文献

次生乳管是天然橡胶合成和贮存的场所,是由橡胶树树干树皮中维管形成层细胞分裂分化而来。次生乳管的数量与天然橡胶产量直接相关,而这些乳管的数量取决于形成层分化次生乳管的频率(乳管分化能力),是橡胶树产量育种的主要指标。前期研究中,我们发现组蛋白去乙酰化酶(HDA)抑制剂曲古抑菌素A(TSA)能诱导橡胶树乳管分化且组蛋白去乙酰化酶基因(HbHDA6)能够参与橡胶树乳管分化调控。由于组蛋白乙酰化修饰调控橡胶树次生乳管分化的分子机制尚未阐明,因此该文使用冠菌素(COR)诱导橡胶树形成层分化产生次生乳管的实验系统,以分离形成层组织为材料,构建酵母双杂交cDNA文库,以HbHDA6基因为诱饵来筛选酵母双杂交文库,确定与HbHDA6相互作用的蛋白。结果表明:(1)利用Gateway技术构建的均一化COR诱导橡胶树形成层组织的酵母双杂交cDNA文库,初级文库的容量为6.34×106 CFU·mL-1,总单克隆数为1.27×107,文库重组率为100%;次级文库的容量为7.72×106 CFU·mL-1  相似文献

酵母RNA聚合酶ⅡRpb2和Rpb3两亚基间相互作用位点的定位   总被引:2,自引:0,他引:2  
为研究S .pombeRNApolⅡ各亚基间体内装配成复合体的机制 ,本文首次用酵母双杂交系统鉴定了Rpb2和Rpb3两亚基间体内相互作用的位点。首先将Rpb2的 4个片段克隆至Gal4BD表达载体pAS2上 ,构建BD Rpb2片段融合蛋白重组质粒 ;同时将Rpb3克隆至Gal4AD表达载体pGADGH上 ,构建AD Rpb3融合蛋白重组质粒。其次 ,将pGADGHRpb3分别与pAS2Rpb2各片段重组质粒共转化到受体酵母菌Y1 90感受态细胞内 ,筛选并鉴定β gal活性阳性 (β gal+)的共转化子。最后 ,将β gal+共转化子中的Rpb2片段进行序列分析并进行同源序列比较确定其在Rpb2中的位置。结果表明 ,Rpb2与Rpb3相互作用的位点位于Rpb2的 90 2~ 989aa肽段内  相似文献

目的 利用酵母双杂交技术从人脑cDNA文库中筛选与人GATA-1相互作用的蛋白质.方法 从人K562细胞中扩增出全长GATA1基因,设计引物将其3段截断体亚克隆入酵母表达载体pDBLeu中,转化至AH109感受态酵母中,利用酵母双杂交技术筛选人脑cDNA文库中与其相互作用的蛋白质,阳性克隆通过回转及免疫共沉淀试验进行验证,利用3xGATA荧光素酶报告基因对相互作用蛋白质进行功能验证.结果 成功构建出酵母诱饵蛋白表达质粒pDBLeu-GATA1(1),pDBLeu-GATA1(2),pDBLeu-GATA1(3),筛到34个阳性克隆,用生物信息学分析及回转验证得到5个与GATA-1相互作用的候选蛋白,通过免疫共沉淀试验进一步验证,获得3个蛋白质能与GATA-1相互作用,分别是ECSIT,EFEMP1和GPS2.荧光素酶试验表明这3个蛋白质均能对GATA1的转录活性产生影响,证实它们之间的相互作用具有影响GATA1转录的功能.结论 应用酵母双杂交技术及免疫共沉淀试验,从人脑cDNA文库中成功获得3个与GATA-1相互作用并对其转录活性具有调节作用的蛋白质,为研究GATA1蛋白质的功能提供了新的线索.  相似文献

Thrombopoietin (TPO) is the major cytokine involved in platelet production and exerts its effects via the receptor c-Mpl. The yeast two-hybrid system has been used to screen the proteins interacting with c-Mpl. First, the cDNA fragment of c-Mpl intracellular domain was cloned into two-hybrid vector pAS2, and the resulting plasmid is designated as pASMM. Then a human placenta cDNA library was screened using the pASMM as a target plasmid. Seven positive clones were isolated from 150 000 independent transformants. Sequence analysis of one of the positive clones demonstrates that a part of coding sequence of vimentin from 611 bp to 3′ end and flanking non-translation region was obtained. Therefore, there is an interaction between vimentin and TPO receptor. The results suggest that cytoskeletal protein may play an important role in TPO signal transduction pathway.  相似文献

目的:利用酵母双杂交系统从人心肌cDNA文库中筛选与热激蛋白70(HSP70)相互作用的蛋白质。方法:从人心脏cDNA文库扩增Hsp70基因,克隆于pGBKT7载体上,酶切鉴定及序列分析,并检测pGBKT7-Hsp70酵母细胞AH109中的自激活活性;将构建的酵母表达诱饵质粒载体pGBKT7-Hsp70转化AH109酵母细胞,与转化有人心脏cDNA文库的酵母Yl87进行交配实验,筛选与HSP70相互作用的蛋白质,通过一对一的回复杂交实验排除假阳性,对阳性克隆进行序列测定和生物信息学分析。结果:构建了"诱饵"质粒栽体pGBKT7-Hsp70,并证明其在酵母双杂交系统中无自激活活性,筛选得到多个与Hsp70相互作用的阳性转化子,并最终得到HSP70的1个相互作用蛋白质HIP。结论:应用酵母双杂交系统筛选出与HSP70相互作用的1个蛋白质,它们的相互作用可能与HSP70发挥细胞分子伴侣作用有关。  相似文献

With yeast two-hybrid methods, we used a C-terminal fragment (residues 1697–2145) of non-erythroid beta spectrin (βII-C), including the region involved in the association with alpha spectrin to form tetramers, as the bait to screen a human brain cDNA library to identify proteins interacting with βII-C. We applied stringent selection steps to eliminate false positives and identified 17 proteins that interacted with βII-C (IPβII-C s). The proteins include a fragment (residues 38–284) of “THAP domain containing, apoptosis associated protein 3, isoform CRA g”, “glioma tumor suppressor candidate region gene 2” (residues 1-478), a fragment (residues 74–442) of septin 8 isoform c, a fragment (residues 704–953) of “coatomer protein complex, subunit beta 1, a fragment (residues 146–614) of zinc-finger protein 251, and a fragment (residues 284–435) of syntaxin binding protein 1. We used yeast three-hybrid system to determine the effects of these βII-C interacting proteins as well as of 7 proteins previously identified to interact with the tetramerization region of non-erythroid alpha spectrin (IPαII-N s) [1] on spectrin tetramer formation. The results showed that 3 IPβII-C s were able to bind βII-C even in the presence of αII-N, and 4 IPαII-N s were able to bind αII-N in the presence of βII-C. We also found that the syntaxin binding protein 1 fragment abolished αII-N and βII-C interaction, suggesting that this protein may inhibit or regulate non-erythroid spectrin tetramer formation.  相似文献

目的 利用酵母双杂交技术在活细胞内筛选并回转验证与PML-C结构域相互作用的蛋白质.方法 通过诱饵质粒pGBKT7-PML-C,利用酵母双杂交系统从白血病细胞cDNA文库中筛选与PML-C结构域相互作用的蛋白质.结果 利用酵母双杂交技术筛选到43个能与PML-C结构域相互作用的克隆;经进一步的归类与酵母回转试验得到9个阳性克隆.结论 在细胞内PML-C结构域能与多种蛋白质有相互作用.中性粒细胞弹性蛋白酶(neutrophil elastase,NE)介导的急性早幼粒细胞白血病的发生可能与这些相互作用所致的生物学功能改变有关.  相似文献

禽流感病毒核蛋白 (NP) 在病毒的转录、复制以及决定病毒的宿主特异性方面都具有重要作用。通过酵母双杂交系统筛选与核蛋白相互作用的蛋白,为进一步了解NP蛋白与细胞内蛋白质的相互关系以及流感病毒与宿主的相互关系奠定基础。应用酵母双杂交系统,构建NP诱饵质粒,进而筛选人脑cDNA文库,寻找可能与禽流感病毒NP相互作用的蛋白质。经过酵母双杂交共验证,得到7个与NP相互作用的阳性克隆。该结果为深入了解病毒复制的分子机理及其在蛋白质水平上与宿主蛋白的相互作用关系提供了线索。  相似文献

Collectrin是在小鼠 5 6肾切除后 ,在肾小球的高滤过、高增生期分离克隆的一个新基因 .通过酵母双杂交系统从人肾脏cDNA文库中筛选与collectrin相互作用的蛋白 ,可以为该基因的功能研究提供线索 .构建collectrin的真核表达载体collectrin pGBKT7 c myc ,转化酵母菌AH10 9.Western印迹证实 ,collectrin蛋白能够在酵母中正常表达 ,对酵母细胞无毒性 ,不存在自激活现象 .将AH10 9 collectrin pGBKT7 c myc与转化了成人肾脏cDNA文库的酵母菌Y187接合 ,共筛选到 5个与细胞代谢有关的蛋白 ,包括鞘磷脂激活蛋白、精氨琥珀酸合成酶、氨基酸转运蛋白XAT2、NADH脱氢酶 1和金属硫蛋白 2A .由此推论 ,collectrin可能通过与细胞内某些酶类相互作用而影响细胞代谢 ,为新基因collectrin的功能研究提供了重要线索 .  相似文献

The vertebrate Six3 gene a homeobox gene of the Six-family, plays a crucial role in early eye and forebrain development. Here we report the isolation of candidate factors that interact with Six3 in a yeast two-hybrid screen. Among these are two basic helix loop helix (bHLH) domain containing proteins. Biochemical analysis reveals that the bHLH proteins ATH5, ATH3, NEUROD as well as ASH1 interact specifically with XSix3. By defining the interacting domains we show that the bHLH domain of NEUROD interacts with the SIX domain of XSix3. The co-expression of the interacting molecules during late retina determination/differentiation suggests a new role for Six3 and the respective interaction partner also in these late steps of eye development.  相似文献

Candida shehatae gene xyl1 and Pichia stipitis gene xyl2, encoding xylose reductase (XR) and xylitol dehydrogenase (XD) respectively, were amplified by PCR. The genes xyl1 and xyl2 were placed under the control of promoter GAL in vector pYES2 to construct the recombinant expression vector pYES2-P12. Subsequently the vector pYES2-P12 was transformed into S. cerevisiae YS58 by LiAc to produce the recombinant yeast YS58-12. Background  

The membrane-bound cell-surface precursor and soluble forms of heparin-binding epidermal growth factor-like growth factor (HB-EGF) contribute to many cellular developmental processes. The widespread occurrence of HB-EGF in cell and tissue types has led to observations of its role in such cellular and tissue events as tumor formation, cell migration, extracellular matrix formation, wound healing, and cell adherence. Several studies have reported the involvement of such extracellular matrix proteins as latent transforming growth factor β-binding protein, TGF-β, and fibulin-1 in some of these processes. To determine whether HB-EGF interacts with extracellular matrix proteins we used the extracellular domain of proHB-EGF in a yeast two-hybrid system to screen a monkey kidney cDNA library. cDNA clones containing nucleotide sequences encoding domains of two proteins were obtained and their derived amino acid sequences were evaluated.  相似文献

目的:获得组蛋白去乙酰化酶1(HDAC1)的cDNA并构建为酵母双杂交系统中的饵基因,筛选出与其相互作用的蛋白质。方法:利用RT-PCR方法从人HeLa细胞中克隆出1446bp的cDNA片段,重组入载体pGBKT7得到pGBKT7-HDAC1,转化至酵母菌AH109,检测其对酵母细胞无毒性也无自激活报告基因活性,然后将其与已转入HeLa细胞基因组cDNA文库的酵母菌融合,进行筛选和验证工作。结果:筛选出3个阳性克隆,经免疫共沉淀试验进一步验证,发现仅有1个蛋白能与HDAC1发生相互作用,测序结果表明该蛋白为FHL2。结论:应用酵母双杂交方法筛选出HDAC1的相互作用蛋白FHL2,为进一步研究HDAC1的作用提供了新线索。  相似文献
