Thein vitro susceptibilities of propolis and antifungal drugs were determined against some yeasts isolated from patients with superficial mycoses. The agents tested included fluconazole, itraconazole, ketoconazole, terbinafine and propolis. MICs were determined by the broth microdilution technique following National Committee for Clinical Laboratory Standards document M27-P. For allCandida albicans isolates from the patients with superficial mycoses, ketoconazole presented higher (P<0.05) efficiency than that of the other antifungal agents tested. The geometric mean MIC values of antifungal drugs and propolis against the yeasts tested ranged from 0.087 to 12.69 μg/mL and 0.4–0.6 μg/mL, respectively. Propolis also showed an important antifungal activity against the yeasts tested, MIC ranges of the propolis were between 0.01–1.65 μg/mL. Based on these results, propolis requires further investigation as a potential agent for the treatment of superficial mycoses.  相似文献

AIMS: To evaluate the fungitoxic activity of Larrea divaricata Cav. extract and one of its components against yeasts and fungi. This activity was compared with the action of ketoconazole, a known synthetic antimycotic. METHODS AND RESULTS: Antifungal activity of Larrea divaricata extract and of a fraction (Fr. B) purified by thin layer chromatography, was investigated using different methodologies. Both exhibited strong activity against the majority of the assayed fungi. Only Fusarium oxysporum and Schizophyllum commune growth was not affected with the assayed conditions. The fungitoxic and cytotoxic activity of the ethanolic extract and ketoconazole were compared. CONCLUSIONS: Ethanolic extracts of L. divaricata Cav. produce growth inhibition of several fungi. One of its constituents with the same activity was purified and identified as a glycoside of a flavanone. A comparison with the action of ketoconazole, which is currently used as antimycotic and can cause adverse health effects was made. SIGNIFICANCE AND IMPACT OF THE STUDY: Our data suggest that L. divaricata extract contains, at least, one compound of phenolic nature, with fungitoxic potency against yeasts and fungi.  相似文献

Ten propolis samples from Bulgaria, Italy and Switzerland were analyzed by GC-MS. As expected, most samples displayed the typical chemical pattern of "poplar" propolis: they contained pinocembrin, pinobanksin and its 3-O-acetate, chrysin, galangin, prenyl esters of caffeic and ferulic acids. Two samples differed significantly: one from the Graubünden Alpine region, Switzerland, rich in phenolic glycerides, and one from Sicily which contained only a limited number of phenolics and was rich in diterpenic acids.  相似文献

Propolis, a multifunctional substance used by bees to maintain the safety of their hives, is popular for its therapeutic potential against some micro-organisms. The aim of this study was to evaluate the ethanolic extract of propolis originated from northern Turkey for its antiproliferative, apoptotic and cell cycle arrest promoting effects on MCF7, HGC27, A549 cancer cell lines and a healthy cell line (HUVEC) in terms of DNA content, morphological features, expression of cell cycle checkpoint proteins p21, p53, Cyclin D1 and immune checkpoint protein PD‐L1. The extract showed moderate antiproliferative activity against all tested cancer cell lines with IC50 values in the range of 58.6–90.7 μg/mL in MTS assay. Further studies indicated that propolis extract exerted apoptotic effect on cancer cell lines, promoted cell cycle arrest through activation of p21 and resulted in accumulation at G0/G1 phase of cancer cells. Propolis treatment caused increased cell size, according to fluorescent imaging except for MCF7. HPTLC analysis revealed that 3‐O‐methylquercetin, chrysin, caffeic acid, CAPE, galangin and pinocembrin were the main components of the extract. The amounts of caffeic acid and CAPE in the extract were found to be 5.5 and 11.1 mg/g, respectively, by a validated HPLC method. Our study is the first one, revealing effect of propolis on PD‐L1 expression on certain cancer cell lines.  相似文献

AIM: The flavonol galangin, an antimicrobial constituent of the traditional medicines propolis and Helichrysum aureonitens, is being assessed as part of an ongoing investigation into the antibacterial activity of flavonoids. The present study sought to establish whether galangin has any aggregatory effect on bacterial cells. METHODS AND RESULTS: In preparatory time-kill assays, 50 microg ml(-1) of galangin was found to reduce colony counts of c. 5 x 10(7) CFU ml(-1)Staphylococcus aureus NCTC 6571 by approximately 15 000-fold during 60 min of incubation. Subsequent light microscopy studies demonstrated significant increases in the number of large clusters of bacterial cells in populations treated with the flavonol. CONCLUSION: Data presented here show that galangin causes aggregation of bacterial cells. SIGNIFICANCE AND IMPACT OF THE STUDY: The finding that galangin causes bacterial cells to clump together may implicate the cytoplasmic membrane as a target site for this compound's activity. More importantly, this observation indicates that decreases in CFU numbers detected in time-kill and minimum bactericidal concentration (MBC) assays in previous investigations were at least partially attributable to this aggregatory effect. This raises the possibility that galangin is not genuinely bactericidal in action, and calls into question the suitability of time-kill and MBC assays for determining the nature of activity of naturally occurring flavonoids.  相似文献

The validation of rapid, low-cost spectrophotometric procedures for the quantification of the three main groups of bioactive substances (flavones and flavonols, flavanones and dihydroflavonols, and total phenolics) in poplar-type propolis has been performed. A spectrophotometric assay based on the formation of an aluminium chloride complex was applied for the quantification of total flavones and flavonols using galangin as standard. Because of the high amount of flavanones and dihydroflavonols in "poplar type" propolis, the introduction of a distinct procedure for their quantification was considered of special significance and the DAB9 colorimetric method was applied for the purpose. Total phenolic content was measured by the Folin-Ciocalteu procedure using a mixture of pinocembrin and galangin as a reference. The procedures were validated using a model mixture of compounds representing the poplar-type propolis composition as found in previous studies. The accuracy (recovery) varied in the range 84-109%, and the relative standard deviation was 0.5-6.2%. The developed spectrophotometric procedures were applied to six poplar type propolis samples. The results were verified independently by a HPLC procedure. The two sets of results agreed satisfactory, as proven by Student's t-test.  相似文献

The effect of an ethanolic extract of propolis, with and without CAPE, and some of its components on cyclooxygenase (COX-1 and COX-2) activity in J774 macrophages has been investigated. COX-1 and COX-2 activity, measaured as prostaglandin E2 (PGE2) production, were concentration-dependently inhibited by propolis (3 x 10(-3) - 3 x 10(2) microgml(-1)) with an IC50 of 2.7 microgml(-1) and 4.8 x 10(-2) microgml(-1), respectively. Among the compounds tested pinocembrin and caffeic, ferulic, cinnamic and chlorogenic acids did not affect the activity of COX isoforms. Conversely, CAPE (2.8 x 10(-4) - 28 microgml(-1); 10(-9) - 10(-4) M) and galangin (2.7 x 10(-4) - 27 microgml(-1); 10(-9) - 10(-4) M) were effective, the last being about ten-twenty times less potent. In fact the IC50 of CAPE for COX-1 and COX-2 were 4.4 x 10(-1) microgml(-1) (1.5 x 10(-6) M) and 2 x 10(-3) microgml(-1) (6.3 x 10(-9) M), respectively. The IC50 of galangin were 3.7 microgml(-1) (15 x 10(-6) M) and 3 x 10(-2) microgml(-1) (120 x 10(-9) M), for COX-1 and COX-2 respectively. To better investigate the role of CAPE, we tested the action of the ethanolic extract of propolis deprived of CAPE, which resulted about ten times less potent than the extract with CAPE in the inhibition of both COX-1 and COX-2, with an IC50 of 30 microgml(-1) and 5.3 x 10(-1) microgml(-1), respectively. Moreover the comparison of the inhibition curves showed a significant difference (p < 0.001).These results suggest that both CAPE and galangin contribute to the overall activity of propolis, CAPE being more effective.  相似文献

Antimicrobial Activity of Propolis on Oral Microorganisms   总被引:8,自引:0,他引:8  
Formation of dental caries is caused by the colonization and accumulation of oral microorganisms and extracellular polysaccharides that are synthesized from sucrose by glucosyltransferase of Streptococcus mutans. The production of glucosyltransferase from oral microorganisms was attempted, and it was found that Streptococcus mutans produced highest activity of the enzyme. Ethanolic extracts of propolis (EEP) were examined whether EEP inhibit the enzyme activity and growth of the bacteria or not. All EEP from various regions in Brazil inhibited both glucosyltransferase activity and growth of S. mutans, but one of the propolis from Rio Grande do Sul (RS2) demonstrated the highest inhibition of the enzyme activity and growth of the bacteria. Galangin, an active flavonoid present at high concentration in Alpinia officinarum Hance and propolis, shows cytotoxicity towards several cancer cell lines, including melanoma. However, the specific cellular targets of galangin-induced cytotoxicity in melanoma are still unknown. Here, we investigated the effects of galangin in B16F10 melanoma cells and explored the possible molecular mechanisms. Galangin significantly decreased cell viability of B16F10 cells, and also induced cell apoptosis shown by Hoechst 33342 staining and Annexin V-PI double staining flow cytometric assay. Furthermore, upon galangin treatment, disruption of mitochondrial membrane potential was observed by JC-1 staining. Western blotting analysis indicated that galangin activated apoptosis signaling cascades by cleavage of procaspase-9, procaspase-3 and PARP in B16F10 cells. Moreover, galangin significantly induced activation of phosphor-p38 MAPK in a time and dose dependent manner. SB203580, an inhibitor of p38, partially attenuated galangin-induced apoptosis in B16F10 cells. Taken together, this work suggests that galangin has the potential to be a promising agent for melanoma treatment and may be further evaluated as a chemotherapeutic agent.  相似文献

AIMS: This study compared in vitro activities of Actichelated propolis (a multicomposite material obtained with mechano-chemichal activation) and of a hydroalcoholic extract of propolis. METHODS AND RESULTS: Minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC), determined by means of microdilution broth method, against five strains of Staphylococcus aureus, Streptococcus pyogenes, Haemophilus influenzae, Enterococcus spp., Escherichia coli, Proteus mirabilis and Pseudomonas aeruginosa, showed a greater potency of Actichelated propolis (MIC range: 0.016-4 mg flavonoids ml(-1)) in respect to the hydroalcoholic extract (MIC range: 0.08-21.4 mg flavonoids ml(-1)). Concentrations of Actichelated propolis active against adenovirus, influenza virus, parainfluenza virus and herpes virus type 1 were at least 10 times lower than those of the hydroalcoholic extract. Preincubation of Strep. pyogenes and H. influenzae with subinhibitory concentrations of Actichelated propolis (1/4 and 1/8 x MIC) significantly reduced the number of bacteria that adhered to human buccal cells. CONCLUSIONS: Actichelated propolis has proven to possess antibacterial and antiviral activity higher than a hydroalcoholic extract, being also able to interfere on bacterial adhesion to human oral cells. SIGNIFICANCE AND IMPACT OF THE STUDY: This new formulation of propolis showing better antimicrobial and physical characteristics could improve the application of propolis in respiratory tract infections.  相似文献

Trichophyton species susceptibility to green and red propolis from Brazil   总被引:1,自引:0,他引:1  
Aims:  The in vitro antifungal activity of Brazilian green and red propolis was tested against different species of Trichophyton .
Methods and Results:  The antifungal activity of the Brazilian aqueous and alcoholic extracts of the green propolis and the alcoholic extract of red propolis was observed against Trichophyton rubrum , Trichophyton tonsurans and Trichophyton mentagrohytes samples, using as controls itraconazole and terbinafine. The minimal inhibitory concentration was determined following the microdilution method indicated by the 'Clinical and Laboratory Standards Institute'. The minimal fungicide concentration was determined by the absence of growth in liquid sabouraud culture medium. The data obtained showed that the green propolis alcoholic extract's antifungal activity was from 64 to 1024 μg ml−1, whereas the red propolis alcoholic extract was from 8 to 1024 μg ml−1.
Conclusions:  The antifungal activity of the red propolis alcoholic extract was more efficient than the green propolis alcoholic extract for all three species studied. The T. rubrum samples were shown to be more sensitive to the antifungal activity of the alcoholic extracts of the propolis.
Significance and Impact of the Study:  The antifungal potential of the alcoholic extracts of green and red propolis demonstrated suggest an applicable potential as an alternative treatment for dermatophytosis caused by these species.  相似文献

In recent years, many papers have provided information concerning its composition. This review compiles data from most studies of propolis, focusing on the chemical composition of ethanol extracts of propolis (EEP), water extracts of propolis (WEP), and volatile oils from propolis (VOP). The characteristic compounds of EEP are polyphenols including flavonoids and related phenolic acids, and flavonoids are the most abundant and effective parts. They are considered to contribute more to the antibacterial, antiviral, and antioxidant effects than the other constituents. The main flavonoids in EEP are pinocembrin, galangin, chrysin, quercetin, kaempferol, and naringenin. The constituents reported to be in WEP include phenolic acids, caffeoylquinic acid, 3-mono-O-caffeoylquinic acid, caffeic acid, flavonoids, etc. The propolis volatile compounds are benzyl alcohol, benzyl acetate, cinnamic alcohol, vanillin, eudensmol, cyclohexyl benzoate, and benzyl benzoate, which are responsible for several biological properties. As a natural mixture, propolis is widely used in medicine and cosmetics, as well as being a constituent of health foods. Since propolis has been used extensively, information on its composition is not only of interest to the academic field, but also to propolis users.  相似文献

Three spectrophotometric methods for the quantitative determination of different flavonoid groups and total phenolics in Croatian propolis samples were optimised and validated. The assay based on the formation of aluminium chloride complex (with galangin as a standard) was applied to the quantification of flavones and flavonols, while the 2,4-dinitrophenylhydrazine method (with pinocembrine as a reference) was used for the quantification of flavanones. Total phenolic content was measured by the Folin-Ciocalteau method using reference solution of caffeic acid:galangin:pinocembrine (1:1:1). Through analytical validation, the most suitable extraction conditions (with respect to time, temperature and concentration of extraction solvent) were determined, and final conditions for the extraction were established (80% ethanol, 1 h at the room temperature). The appropriate ratio between the mass of raw propolis and the extraction solvent volume was also established. By the application of the optimised method of extraction, 10 propolis tinctures were prepared and subjected to the analysis of general pharmacopoeial parameters, which are fundamental for the creation of quality specification (relative density, dry residue of extract, content of ethanol, methanol and 2-propanol). Additionally, the content of waxes as the main inactive constituents was determined in order to observe the level of their migration from crude propolis to the prepared tinctures.  相似文献

Propolis was extracted using water and various concentrations of ethanol as solvents. The extracts were investigated by measurement of absorption spectrum with a UV spectrophotometer, reversed phase-high pressure thin-layer chromatography and reversed phase-HPLC. Maximum absorption of all extracts was 290 nm, resembling flavonoid compounds, and the 80% ethanolic extract showed highest absorption at 290 nm. The most isosakuranetin, quercetin, and kaempferol were extracted from mixtures of propolis and 60% ethanol, while 70% ethanol extracted the most pinocembrin and sakuranetin, but 80% ethanol extracted more kaempferide, acacetin, and isorhamnetin from propolis. The 60 to 80% ethanolic extracts of propolis strongly inhibited microbial growth and 70 and 80% ethanolic extracts had the greatest antioxidant activity and 80% ethanolic extract strongly inhibited hyaluronidase activity.  相似文献

从山杨 (PopulusdavidianaDode.)树皮的氯仿部分得到了 7个黄酮类化合物 ,经理化数据和波谱分析鉴定为 :高良姜素 (Galangin)、白杨素 (Chrysin)、7 甲氧基白杨素 (Tectochrysin)、芫花素(Genkwanin)、商陆素 (Ombuin)、乔松素 (Pinocembrin)、3 乙酰基乔松素 (trans 3 acetoxy 5 ,7 dihydrox yflavanone)。商陆素和 3 乙酰基乔松素为首次从该属植物中分离得到。高良姜素、7 甲氧基白杨素、商陆素有较强的抗炎活性。  相似文献

In the continuing search for new compounds with trypanocidal activity for use in blood banks to prevent the transmission of Chagas' disease, a trypanocidal extract of Lychnophora staavioides Mart. (Vernonieae, Asteraceae) was fractionated using several chromatographic techniques and afforded the following flavonoids: tectochrysin, pinostrobin, pinobanksin, pinobanksin 3-acetate, pinocembrin, chrysin, galangin 3-methyl ether, quercetin 3-methyl ether, chrysoeriol and vicenin-2. The most active compound was quercetin 3-methyl ether, which showed no blood lysis activity and which represents a promising compound for use against T. cruzi in blood banks.  相似文献

Propolis is a multi-functional bee product rich in polyphenols. In this study, the inhibitory effect of Anatolian propolis against SARS-coronavirus-2 (SARS-CoV-2) was investigated in vitro and in silico. Raw and commercial propolis samples were used, and both samples were found to be rich in caffeic acid, p-coumaric acid, ferulic acid, t-cinnamic acid, hesperetin, chrysin, pinocembrin, and caffeic acid phenethyl ester (CAPE) at HPLC-UV analysis. Ethanolic propolis extracts (EPE) were used in the ELISA screening test against the spike S1 protein (SARS-CoV-2): ACE-2 interaction for in vitro study. The binding energy values of these polyphenols to the SARS-CoV-2 spike and ACE-2 protein were calculated separately with a molecular docking study using the AutoDock 4.2.6 program. In addition, the pharmacokinetics and drug-likeness properties of these eight polyphenols were calculated according to the SwissADME tool. The binding energy value of pinocembrin was highest in both receptors, followed by chrysin, CAPE, and hesperetin. Based on the in silico modeling and ADME (absorption, distribution, metabolism, and excretion) behaviors of the eight polyphenols, the compounds exhibited the potential ability to act effectively as novel drugs. The findings of both studies showed that propolis has a high inhibitory potential against the Covid-19 virus. However, further studies are now needed.  相似文献
