Schistosomiasis is an important neglected tropical disease caused by digenean helminth parasites of the genus Schistosoma. Schistosomes are unusual in that they are dioecious and the adult worms live in the blood system. MicroRNAs play crucial roles during gene regulation and are likely to be important in sex differentiation in dioecious species. Here we characterize 112 microRNAs from adult Schistosoma mansoni individuals, including 84 novel microRNA families, and investigate the expression pattern in different sexes. By deep sequencing, we measured the relative expression levels of conserved and newly identified microRNAs between male and female samples. We observed that 13 microRNAs exhibited sex-biased expression, 10 of which are more abundant in females than in males. Sex chromosomes showed a paucity of female-biased genes, as predicted by theoretical evolutionary models. We propose that the recent emergence of separate sexes in Schistosoma had an effect on the chromosomal distribution and evolution of microRNAs, and that microRNAs are likely to participate in the sex differentiation/maintenance process.  相似文献

To see whether male worms within the gynecophoral canal of another male worm would become feminized (i.e. express vestigial female-associated genes), we established homosexual pairs by twice exposing mice to male cercariae with a 4 or 6-week interval, and perfusing 3–5 weeks later. From 13 to 34% of these worms were found in pairs, compared with 0 to 7% in singly exposed controls. ‘Inner’ males in homosexual pairs showed no histological evidence of female reproductive structures, but were stunted, had poorly developed testes, and the high nuclear density characteristic of mature females. More vitelline follicles occurred in unpaired unisexual males than in homosexually paired males, fewest in bisexually paired males. Uptake of tyrosine, an indicator of vitelline development, occurred in the same relative order. The gynecophoral microenvironment often led to stunting, probably through starvation induced by the relative inaccessibility of host blood to homosexually clasped males.  相似文献

Garden asparagus (Asparagus officinalis L.) is a dioecious species with male and female flowers on separate unisexual individuals. Since B- and C-functional MADS-box genes specify male and female reproductive organs, it is important to characterize these genes to clarify the mechanism of sex determination in monoecious and dioecious species. In this study, we isolated and characterized AODEF gene, a B-functional gene in the development of male and female flowers of A. officinalis. Southern hybridization identified a single copy of AODEF gene in asparagus genome. Northern blot analysis showed that this gene was specifically expressed in flower buds and not in vegetative tissues. In situ hybridization showed that during early hermaphrodite stages, AODEFgene was expressed in the inner tepal and stamen whorls (whorls 2 and 3, respectively), but not in the outer tepals (whorl 1), in both male and female flowers. In late unisexual developmental stages, the expression of AODEF gene was still detected in the inner tepals and stamens of male flowers, but the expression was reduced in whorls 2 and 3 of female flowers. Our results suggest that AODEF gene is probably not involved in tepal development in asparagus and that the expression of AODEF gene is probably controlled directly or indirectly by sex determination gene in the late developmental stages.  相似文献

1,2,6,7-Tetraoxaspiro[7.11]nonadecane (N-89) is a chemically synthesized compound with good efficacy against malaria parasites. We observed strong anti-schistosomal activities of N-89 both in vitro and in vivo. In a murine model with experimental infection of Schistosoma mansoni, orally administered N-89 at the dose of 300 mg/kg resulted in a significant reduction in worm burden (63%) when mice were treated at 2-weeks postinfection. Strong larvicidal effects of N-89 were confirmed in vitro; schistosomula of S. mansoni were killed by N-89 at an EC50 of 16 nM. In contrast, no significant reduction in worm burden was observed when N-89 was administered at 5 weeks postinfection in vivo. However, egg production was markedly suppressed by N-89 treatment at that time point. On microscopic observation, the intestine of N-89-treated female worms seemed to be empty compared with the control group, and the mean body length was significantly shorter than that of controls. Nutritional impairment in the parasite due to N-89 treatment was possible, and therefore quantification of hemozoin was compared between parasites with or without N-89 treatment. We found that the hemozoin content was significantly reduced in N-89 treated parasites compared with controls (P < 0.001). The surface of adult worms was observed by scanning and transmission electron microscopy, but there were no apparent changes. Taken together, these observations suggested that N-89 has strong antischistosomal effects, probably through a unique mode of drug efficacy. As N-89 is less toxic to mammalian host animals, it is a possible drug candidate against schistosomiasis.  相似文献

Sexual dimorphisms are primary regulated by sex‐biased gene expression. In the present study, using real‐time polymerase chain reaction, we determined the expression profiles of nine genes associated with development, metabolism, stress, and defense throughout adulthood of the Indian meal moth Plodia interpunctella, a global pest of stored food products. Most genes were differentially expressed in a sex‐biased manner during the adult lifespan of the moth. Expression of the heat shock protein genes hsp25 and hsp90 and the antioxidant gene thioredoxin peroxidase (Tpx) was highly female biased, whereas the expression of a gene related to host development (ecdysone receptor [EcR]) and two genes associated with immunity (β‐glycan recognition protein [βgrp] and prophenoloxidase [ProPO]) was male biased. In contrast, the expression of hsp70, glucose‐regulated protein 78 (grp78) and ultraspiracle (USP) was not sex biased. The results of the present study provide important insights into the role of sex‐biased genes in the physiology and behavior of P. interpunctella.  相似文献

While infecting a vertebrate host, blood flukes (Schistosoma mansoni) must continually resist adhesions by immune effector cells. However, the male and female schistosomes must adhere to one another in order to establish and maintain the sexual pairing process after 4 wk postinfection. Using a contact angle method, the relative adhesiveness of male and female parasites were determined. Results indicate that schistosomes restrict effector cell adhesion through developmental, sexual, and regional differences in adhesive properties.  相似文献

Here we describe the cloning and characterization of the Schistosoma mansoni Alkaline Phosphatase (SmAP), previously identified in the tegument of adult worms. SmAP encodes a complete sequence composed of 536 amino acids containing an N-terminal signal peptide, five N-glycosylation sites, and a GPI anchor signal, similar to that described for mammalian orthologs. Real-time RT-PCR and Western blot experiments suggest a rapid translation as soon as cercariae are transformed into schistosomula. Immunolocalization analysis shows that the protein is widely distributed in the worm tissues, with increased concentration in the vitelline glands of female parasites. Furthermore, the surface localization of this enzyme was quantitatively supported by its enzymatic activity in live ex vivo or cultured parasites throughout the life cycle stages. The fact that cercariae accumulate large amounts of SmAP mRNA, which rapidly translates into protein upon schistosomula transformation, indicates it may have an important role in host invasion.  相似文献

Is the genetic diversity of parasites infecting male and female hosts equal or different? This is the question we address in this paper by studying the neutral genetic variability of the plathyhelminth trematode Schistosoma mansoni within males and females of its natural murine host Rattus rattus in the marshy forest focus of Guadeloupe (French West Indies). Using seven microsatellite markers, we demonstrate that parasites from male hosts are genetically more diversified than parasites from female hosts. Three hypotheses are discussed that could explain this pattern: 1) a host sex-specific duration of cercariae recruitment; 2) a difference in the behaviour of male and female hosts that would lead to the exposure of males to a greater diversity of parasites; and 3) a host sex-biased immunocompetence that would lead to the selection of more genetically diversified individuals in male than in female rats. This finding is the first empirical evidence that each host sex may play different roles in the maintenance of parasite genetic diversity and so in their evolutionary dynamics and epidemiology.  相似文献

Genetic studies have indicated that some parasite species formerly thought to be generalists are complexes of morphologically similar species, each appearing to specialize on different host species. Studies on such species are needed to obtain ecological and parasitological data to address whether there are fitness costs in parasitizing atypical host species. We examined whether lungworms from two anuran host species, Lithobates sylvaticus and Lithobates pipiens, differed in measures of infection success in L. pipiens recipient hosts. We also determined if the worms from the two host species were sources of genetically resolvable species of morphologically similar nematodes. Sequences of internal transcribed spacer and lsrDNA regions of adult lungworms from each host species indicated that worms from L. sylvaticus matched Rhabdias bakeri, whereas worms from L. pipiens matched Rhabdias ranae. Our work suggested that these morphologically similar species are distant non-sibling taxa. We infected male and female metamorphs experimentally with lungworm larvae of the two species. We observed higher penetration, higher prevalence and higher mean abundance of adult worms in lungs of male and female metamorphs exposed to R. ranae larvae than in lungs of metamorphs exposed to R. bakeri larvae. Furthermore, metamorphs exposed to R. ranae larvae carried larger adult female worms in their lungs. Some variation in infection measures depended on host sex, but only for one parasite species considered. Overall, the differential establishment and reproductive potential of R. ranae and R. bakeri in L. pipiens suggests co-adaptation.  相似文献
