No study has yet been carried out on seed development in a cold desert sand dune papilionoid legume. Thus, our primary aims were to (i) monitor seed development in the cold desert sand dune species Eremosparton songoricum from the time of pollination to seed maturity, and (ii) compare seed development in this species with that in other species of papilionoid legumes. Fruit and seed size, mass and seed moisture content, and seed imbibition, germination, desiccation tolerance and water retention during development (pollination to seed maturity) were monitored in the papilionaceous shrub E. songoricum in the Gurbantunggut Desert of northwest China. The duration of seed development was 40 days. Seeds reached physiological maturity 28 days after pollination (DAP), at which time 58% of them germinated and they had developed desiccation tolerance. Seeds became impermeable 36–40 DAP, when their moisture content was about 10%. The final stage of maturation drying occurred via loss of water through the hilum. The developmental stages and their timing (DAP) in seeds of E. songoricum are generally similar to those reported for other papilionaceous legumes with a water‐impermeable seed coat (physical dormancy). In general, the developmental features of seeds with water‐impermeable coats at maturity do not appear to be specific to habitat or phylogeny.  相似文献

Investigations were undertaken on the desiccation and freezingsensitivity of recalcitrant seeds of three species: tea [Camelliasinensis (L.) O. Kuntze], cocoa (Theobroma cacao L.) and jackfruit(Artocarpus heterophyllus Lamk.). All species showed changesin the physiological characteristics, desiccation and freezingsensitivity of both the seed and the embryonic axes with increasingseed maturity. Fully mature seeds of tea, cocoa and jackfruitsurvived desiccation to 24, 35 and 31% moisture content, respectively,but at these moisture levels seeds were not able to toleratefreezing in liquid nitrogen (-196 °C). Some survival ofcryopreservation was, however, achieved for excised embryonicaxes of partially and fully-mature tea and jackfruit seeds afterdrying to 14% moisture content; cocoa axes were totally freezingsensitive at all three stages of physiological maturity studied.Biochemical investigations on fully mature axes after desiccationand freezing showed that the decline in viability with moisturelevel was associated with increased leachate conductivity, lipidperoxidation products and/or soluble carbohydrates. The fatty acid composition and content of membrane and storage lipids of two transgenic laurate-producing rape (Brassica napus L.) lines were monitored during seed development. The two lines, the medium-laurate (ML) line and the high-laurate (HL) line, accumulated 34 mol% and 55 mol% of laurate in their seed triacylglycerols, respectively. The diacylglycerols contained about 17 and 33 mol% of laurate in the ML- and HL-lines, respectively, from the mid-stage of seed development up to seed maturity. The ML-line showed a maximal relative laurate content in phosphatidylcholine (17 mol%) at the mid-stage of seed development whereafter the content decreased to 2.7 mol% with seed maturity. The laurate content in phosphatidylcholine was observed to remain high (26 mol%) in the HL-line from the mid-stage to the end of triacylglycerol deposition. Thereafter, the relative content decreased and reached 6.6 mol% in the mature seeds. There was an enhanced activity of lauroyl-phosphatidylcholine- metabolizing enzymes in the seed membranes from laurate-producing lines compared with control lines, which might explain the decrease seen in laurate content in phosphatidylcholine during seed maturation. A comparison of the laurate distribution in the lipids from developing laurate-producing rape seeds and developing seeds from three species naturally accumulating laurate at similar levels revealed differences in laurate metabolism compared with these species. The results suggest that phospholipids and triacylglycerols are synthesized from the same diacylglycerol pool in rape seeds and that rape lysophosphatidic acid acyltransferase and diacylglycerol acyltransferase do not have the same preference for laurate substrates as the corresponding enzymes in seed tissues naturally accumulating this acyl group. In addition, the mechanisms that specifically remove or exclude laurate from membrane lipids appear less effective in rape seed than in tissues naturally evolved to synthesize laurate-rich oils. Biochemical properties, i.e. endogenous abscisic acid, proline,sugars, respiration, adenosine phosphates and adenylate energycharge, and growth and moisture content were measured duringthe development of seeds of Machilus thunbergii. As dry matteraccumulated in the embryo during development, moisture content,ABA, proline, respiration and sugars all declined. At maturity,the dry mass of the seeds failed to attain a plateau beforethe period of natural seed shedding; the axis and cotyledonsreached moisture contents of 58 and 45%, respectively. Dryingof immature seeds at 73% relative humidity and 25 °C for30 d resulted in a complete loss of viability at all developmentalstages tested with the exception of mature seeds that were ableto tolerate a 5% decrease in moisture content before germinationdeclined. ABA was detected in all embryos tested, with a maximum value16.·16 µg g-1 d. wt about midway through development.Although the presence of ABA induced no tolerance to desiccationof mature seeds, it did coincide with decreased content of waterin the developing seeds and decreased respiration. Desiccationdamage of M. thunbergii seeds occurred when moisture contentwas still high (45%) and this damage was not related to theabsence of oligosaccharides in the mature seeds. We concludethat developing embryos and mature seeds of M. thunbergii haveproperties common to many recalcitrant seeds, with seeds beingsensitive to desiccation at all stages, having a prominent ABApeak, little proline, lacking oligosaccharides, and specifically,little dormancy and a moderate rate of respiration of matureseeds (0·9 µmol O2 min-1 g-1 f. wt). Adenosinetriphosphate content and energy charge decreased from stagefour to stage eight of seed development, then increased againto 103 nmol g-1 d. wt and 0·73, respectively, in matureseeds. Seeds of bean (Phaseolus vulgaris cv. Vernel) were collected throughout their development on the plant and dried at 15 degrees C and 75% relative humidity to a final moisture content of about 16% (fresh weight basis) to determine whether the onset of tolerance to this drying condition was related to changes in soluble sugars or the activities of the main antioxidant enzymes, namely superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), and glutathione reductase (GR). Measurements of soluble sugars and enzyme activities were made after drying the seeds, and drying tolerance was evaluated by the ability of dried seeds to germinate and to produce normal seedlings. Seeds became tolerant to drying at 45 d after anthesis, a time marking physiological maturity. At physiological maturity, the moisture content of seeds was about 50-55% (fresh weight basis) and seed dry matter reached about 190 mg per seed. Seed vigour, evaluated by controlled deterioration and conductivity measurements, continued to increase after seed mass maturity, but decreased when seeds remained thereafter for more than 7 d on the plant. Acquisition of drying tolerance was coincident with an accumulation of raffinose and stachyose. Dried-tolerant seeds were also characterized by a high amount of sucrose, the most abundant sugar, and by a low content of monosaccharides. The (raffinose+stachyose)/sucrose ratio increased during seed filling, reaching a value close to 1 when all the seeds became tolerant to drying, and maintaining this proportion during the final stages of maturation. Acquisition of drying tolerance was also related to a reorientation of the enzymatic antioxidant defence system. Drying-tolerant dried seeds displayed high CAT and GR activities and low SOD and APX activities, while the opposite condition was observed in immature dried seeds. The shift in antioxidant enzymes corresponded to the beginning of the maturation-drying phase. These results suggest that oligosaccharide metabolism and enzymatic antioxidant defences may be involved in acquisition of drying tolerance during bean seed development, but are not related to seed vigour.  相似文献

‘Physiological maturity’, i.e. the time when seedsreach their maximum dry weight during development, occurredwhen maturation drying on the parent plant in the field hadreduced seed moisture content to approximately 60 per cent infaba bean (Vicia faba L.), lentil (Lens culinaris Medic.), chickpea(Cicer arietinum L.), white lupin (Lupinus albus L.), soya bean(Glycine max [L.] Merr.) and pea (Pisum sativum L.) The onsetof desiccation-tolerance, i.e. the ability of seeds to germinatefollowing harvest and rapid artificial drying, coincided withphysiological maturity, except in pea where it occurred a littleearlier at about 70 per cent moisture content. Maximum seedquality as determined by maximum viability, minimum seedlingabnormalities and maximum seedling size occurred in pea, chickpeaand lupin when seeds were harvested for rapid drying at physiologicalmaturity; but for maximum seed quality in the other speciesmaturation drying had to proceed further - to about 45 per centmoisture content in soya bean and to about 30 per cent moisturecontent in lentil and faba bean seed crops. Much of this variationamongst the six species, however, was due to differences inthe variation in maturity within each seed crop. Results forindividual pods showed that peak maturity, i.e. maximum seedquality following harvest and rapid artificial drying, was achievedin all six species once maturation drying had reduced the moisturecontent of the seeds to 45–50 per cent. In pea, faba beanand soya bean there was a substantial decline in viability andan increase in seedling abnormalities when harvest was delayedbeyond the optimal moisture content for harvest.  相似文献

Precise knowledge of seed quality after harvest and during storageis of particular importance for seed producers. We analyseddifferent sunflower seed lots (Helianthus annuusL.) characterizedby extremes of germination ability. We used RNA analysis tostudy possible changes in gene expression in seeds unable togerminate. Total RNA content was very small in dry seeds showinga low germination ability. Capacity for total RNA synthesisat the onset of imbibition was also reduced in these seeds.In addition, correlations were found between these parametersand germination ability at 19 °C. We demonstrated a highcorrelation between the amount of total RNA in the dry seed,the capacity of RNA synthesis at the onset of imbibition andthe seed moisture content at the time of the harvest. The abilityof dry seed mRNAs to be translatedin vitrowas also reduced andseven polypeptides, from stored mRNAs, were characteristic ofthe cotyledons from high germinability seeds. Seed growth characteristics of Aesculus hippocastanum were examinedin detail during development from about 70 to 140 d after anthesis(DAA), mainly in 1988 and 1989. Mean fresh and dry weights increasedfor both the axis and the whole seed up to the time of peakseed fall at 135 DAA with no cessation before fruit abscission.Water per seed increased up to 100 DAA, after which no furtherincrease occurred; moisture content declined for the embryonicaxis and whole seed respectively from above 75 and 65% at 95DAA to 65 and 50% at 130 DAA. At fruit shedding in 1990 waterpotential values of -1·2, -2·6 and -1·1MPa were observed for the testa, cotyledon and axis tissuesrespectively; relevant sorption isotherms are presented. Decreases in seed moisture content during development were accompaniedby increases in desiccation tolerance and in germinability,both reaching their maximum at the time of peak seed fall. Atmaturity, only about 10% viability was retained on drying seedto 20% moisture content; it is confirmed that the seeds are'recalcitrant'. The exact relationship between moisture contentand germination during development was dependent on the deptof dormancy, as judged by the period of chilling required; eachduration of chilling at 2°C within the range 3-12 weeksyields a curve of sigmoid shape. No germination occurred at26°C without chilling, but nearly full germination can beobserved for samples collected at 6 weeks before maximum seedfall with 12 weeks chilling. Summary Seed set and fruit development in cucumber (Cucumis sativus L.) were studied in relation to female flower receptivity from day — 2 before anthesis to day + 2 after anthesis. The female cucumber flower is protogynous. The pistil was receptive 2 days before anthesis. The iso-electric focusing (IEF) patterns of the stigma/style proteins, were identical from day -5 to day +2. In pollinated flowers in vivo germination and pollen-tube growth in the ovary were affected by pistil age from day -2 to day +2. In addition, differences in sectorial filling in full seeds were observed within the fruits. A negative correlation was observed between the frequency of fertilized ovules in the pedoncular part of the fruit and ovary length at the time of pollination. In the whole fruit, significant differences in the number of full seeds and fruit size at maturity were found, and these were observed to be correlated with the various stages of female flower maturation at pollination. The day -2 and day +2 stages yielded the smallest fruits with few full seeds compared to the day -1, day 0 and day +1 stages, which had the biggest fruits and a large number of full seeds. A strong positive correlation was found between total seed number (including full and empty seeds), fruit length and weight at maturity. All these results suggest that both seed set in the different parts of the fruit and fruit development are controlled by ovular receptivity rather than by stigma/style receptivity.  相似文献

Irrigation of rapid-cycling brassica (Brassica campestris [rapa]L.)plants either ended 16 or 24 days after pollination (DAP) orcontinued throughout the experiment (control). Seeds were harvestedserially from these plants during their development and maturation.The earlier irrigation to the mother plant ended, the earliermass maturity (end of seed-filling phase) occurred, the lowerthe final seed dry weight, and the more rapid the decline inseed moisture content. The onset of ability to germinate normallyoccurred as early as 12 DAP, when seeds were less than half-filled.The onset of ability to tolerate rapid enforced desiccation(to 10% moisture content) occurred at 16 DAP. Desiccation tolerancedeveloped within most seeds in both populations about 5 d soonerin seeds harvested from plants in which irrigation was stoppedat 16 DAP than in control plants, but maximum desiccation toleranceoccurred at about 28 DAP in all treatments. The growth substances of the seeds of Cassia fistula were studied and the changes in the relative levels in the endosperm and embryo (plus cotyledons) with development of the seed were noted. Indoleacetic acid was found to be the major auxin component of the seed almost throughout its growth and development, while acidic inhibitors possibly belonging to β-complex were also noted in bioassay tests. The main source of the IAA in the seed is the endosperm, although measurable amounts are also present in the embryo. While this IAA activity in the endosperm is detectable till maturity of the fruit, it decreases relatively in the embryo to fall to insignificance at maturity of the seed. However, there is indication of the binding of such IAA in the embryo or the cotyledon, which can be released by alkaline hydrolysis but not before the seeds are matured. No such bound auxin could be detected in the endosperm. The inhibitors, on the other hand, are more prominent in the embryo than in the endosperm, particularly with ageing of the fruit. The possible significance of these changes in the growth factors has been discussed in relation to the age of the seed and the development of the embryo inside it.  相似文献

Survival curves of four seed lots of onion held at 15%, 20%and 25% seed moisture content (me) and 45 ° C for 24 h showeda fall in seed germination at all moisture contents. The rateof loss of viability was less for seeds held at 20% and 25%me than that of seeds held at 15% me, with low vigour seedsshowing the greatest reduction in the rate of deteriorationat the high moisture contents. Summary The changes in the level of indole-3-acetic acid (IAA) were investigated in seeds and fruit tissues-placenta and mesocarp-during tomato (Lycopersicon esculentum Mill.) zygotic embryogenesis, which was characterized through eight morphological embryo stages [from globular (stage 1) to mature embryo (stage 8)]. In whole seeds, IAA levels increased mainly at stage 3 (young torpedo) and at stage 5 (late torpedo stage). As the seed matured and dehydrated, IAA levels decreased and showed a new distribution pattern within seed structures, preferentially in endosperm tissue. IAA contents in fruit tissues were lower but followed the same pattern as those of seeds. These data support the hypothesis of IAA biosynthesis in seeds with a transient role of the endosperm at the end of embryo development and suggest a role of IAA in fruit and seed growth. We examined the effects of seed formation andpara-chlorophenoxyacetic acid (p-CPA)treatment on the growth and endogenous indole acetic acid (IAA) content ofmuskmelon fruit. The growth of parthenocarpic muskmelon fruit induced by 1-(2-chloro-4-pyridyl)-3-phenylurea (CPPU) declined 15 days after anthesis (DAA),resulting in smaller fruit than those pollinated at harvest.p-CPA improved the growth of parthenocarpic fruit thatweretreated between 10 and 25 DAA. Endogenous IAA levels in the seedsof artificially pollinated fruit were at their highest at 10 DAA,then decreased, and increased again after 30 to 45 DAA, whereas,the levels in the empty seeds of parthenocarpic fruit were significantly lowerthroughout development. Although endogenous IAA levels in the placenta ofpollinated fruit were lower than those in the seeds, the changing patterns werevery similar to those in the seeds. Endogenous IAA levels in the mesocarp ofpollinated fruit remained lower than those in the placenta throughout fruitgrowth, and the pattern of change was similar to that of the placenta. Levelsinthe seed, placenta and mesocarp of p-CPA-nontreatedparthenocarpic fruit stayed lower than those in pollinated fruit.p-CPA increased the levels of IAA in the seeds, placenta,and mesocarp of parthenocarpic fruit after the first treatment (10DAA) to 15 DAA, while those in the mesocarp increasedsignificantly after the second treatment (25 DAA), but did notincrease in empty seed and placenta.  相似文献

Seeds of Hancornia speciosa germinated best at a temperatureof 20–30 °C. The viability of the seeds during storagewas short and the best storage conditions for viability entailedkeeping the seeds in polyethylene bags. Seed viability was maintainedonly when the seeds were stored at a moisture content above30%; storage conditions which allowed dehydration resulted ina rapid loss of viability (the seeds showed recalcitrant behaviour). Low temperature during storage did not improve longevity. Arelationship between germination and moisture content was established,but when the moisture content fell below 25% there was a drasticreduction of germination. After 9 weeks of storage, even athigh moisture content, seeds lost viability. Loss of seed viability during seed dehydration was associatedwith increased leakage of electrolytes and organic solutes,and reduced tetrazolium staining during subsequent imbibition. In seeds of Trifolium repens, T. pratense, and Lupinus arboreus,the hilum is a hygroscopically activated valve in the impermeableepidermis of the testa. When relative humidity was low the fissurein the hilum opened permitting the seed to dry out; when therelative humidity was high the fissure closed obstructing theabsorption of moisture. During seed-ripening the moisture contentfell readily to approximately 25 per cent., and thereafter moreslowly until the epidermis became impermeable at approximately14 per cent, moisture content. Further drying of the seed tookplace only by diffusion of water vapour through the hilum. ‘Hard’seeds tended to have a moisture content in equilibrium withthe lowest relative humidity to which they had been exposed.They absorbed moisture under conditions of gradually increasingrelative humidity such that the hilar fissure remained open.The duration of the impermeable condition increased with thedegree of desiccation brought about by loss of water throughthe hilum.  相似文献

The correlation between desiccation tolerance and soluble sugars was investigated in seeds of a number of rice cultivars belonging to the Asian rice Oryza sativa L. They were dried or ultradried to various low moisture content and then imbibed for germination testing. Few or no changes on germination percentage and vigor index were found in Indian rice seeds even after their moisture content fell to 3.5%, indicating that Indian rice exhibited a strong desiccation tolerance. On the contrary, Japonica rice seed germination percentage rapidly decreased, after their moisture content fell to 4.5%. The capacity for desiccation tolerance in Japonica (cv. Chunjiang 15) and Indian (cv. Zhongzu 1) developing seeds increased on 23–40 and 15–25 days after pollination, respectively. Though the level of monosaccharides declined, the content of sucrose has increased during desiccation. These results suggest that desiccation tolerance might be associated with the increase in seed viability and the changes in sugar level, and that raffinose could be capable of contributing to the desiccation tolerance to ultradrying. We propose herein a novel single seed descent protocol that has application across a broad phenotypic range of pea genotypes. Manipulation of key in vivo growing conditions, including light, photoperiod and temperature, combined with precocious in vitro germination of the embryo at full physiological maturity substantially shortened the pea lifecycle. We define full embryo physiological maturity as the earliest point in seed development when precocious in vitro germination and robust seedling growth can be reliably achieved without supply of exogenous hormones. Under our optimised conditions for accelerated plant growth, embryo physiological maturity was attained at c. 18 days after pollination, when seed moisture content was below 60?% and sucrose level under 100 mg g?1 DW. No delay penalty in terms of time to flowering and plant development was caused by the culture of immature seeds 18 days after pollination compared to the used of mature ones. Determining the role embryo maturity plays in the fitness of the germinated plant has facilitated the truncation of the lifecycle across pea genotypes. The accelerated single seed descent system proposed within this research will benefit complex genetic studies via the rapid development of recombinant inbred lines (RIL) and multi-parental advanced generation intercrosses (MAGIC) populations.  相似文献

Changes in seed quality during ripening were studied in sixteencultivars of rice, representing the three ecogeographic racesofOryza sativa, and one cultivar ofO. glaberrima, grown duringone dry season (Nov. –May) 1992 –1993 at Los Baños, Philippines. Mass maturity (defined as the end ofseed filling period) among the cultivars was attained between18.5 and 21.6d after anthesis (DAA). The seed moisture contentat mass maturity varied between 24 and 40%. Germination abilityof seeds in the early stages of development varied significantly,but as mass maturity approached, germination increased to themaximum and no significant differences were found among cultivars.The seeds were stored hermetically at 35 °C with 15±0.2%moisture content and the resultant seed survival data were analysedby probit analysis. Potential longevity (quantified by the valueof seed lot constantKiof the seed viability equation) was greatestbetween 33 and 37 DAA, i.e. about 2 weeks after mass maturity.The stage during development at which seeds achieve maximumpotential longevity is described by the term storage maturity.Lowlandjaponicacultivars, large seeded accessions (seed mass40mg) andO. glaberrimahad shorter storage longevity ( , standarddeviation of the frequency of seed deaths in time=1.47 weeks)while cultivars with purple pericarp survived longer than othercultivars ( =2.33 weeks). The initial germination of thejaponicacultivarsat storage maturity was high (99 –100%) and the estimatesof maximum potential longevity (Ki) which ranged between 3.3(Shuang cheng nuo) and 4.4 (Minehikare) were close to thoseof theindicacultivars. This research suggests that seed production environment betweenNov. and May at Los Ba ños is benign for the temperatejaponicacultivars.The implications of these results on management of rice geneticresources are discussed. Sunflower seeds behaved as chilling and freezing sensitive and also exhibited acclimation under low seed moisture content (< 1 %). At high seed moisture content (approx. 22 %) they tolerated chilling stress but failed to acclimate under freezing temperatures. Pre-imbibitional chilling (5 °C) or freezing (−5 or −10 °C) stress significantly enhanced total soluble protein (TSP) content. Chilling treatment after imbibition (in contrast to pre-imbibition) enhanced germination and this was accompanied by increase in 30, 24 and 21.9 kDa TSPs content (3 d after germination). Freezing at −5 and −10 °C suppressed seed germination and increased content of 78 and 56.2 kDa wall bound proteins. Chilling acclimation decreased 35.4, 33.9, 29.5, 23.4 and 21.4 kDa TSPs.  相似文献
