Summary In the moss Physcomitrella patens, single-cell protonemata and multicellular gametophores respond to reorientation relative to the gravity vector by growing negatively gravitropically. A mutant class in which the protonemata, but not the gametophores, respond by growing towards gravity has been identified. In this paper, we describe the isolation of additional mutants of this class. Complementation and segregation ratio analyses were carried out on these mutants, which indicate that a single gene may mutate to switch the polarity of gravitropism.  相似文献

The phosphoinositide signalling pathway is important in plant responses to extracellular and intracellular signals. To elucidate the physiological functions of phosphoinositide-specific phopspholipase C, PI-PLC, targeted knockout mutants of PpPLC1, a gene encoding a PI-PLC from the moss Physcomitrella patens, were generated via homologous recombination. Protonemal filaments of the plc1 lines show a dramatic reduction in gametophore formation relative to wild type: this was accompanied by a loss of sensitivity to cytokinin. Moreover, plc1 appeared paler than the wild type, the result of an altered differentiation of chloroplasts and reduced chlorophyll levels compared with wild type filaments. In addition, the protonemal filaments of plc1 have a strongly reduced ability to grow negatively gravitropically in the dark. These effects imply a significant role for PpPLC1 in cytokinin signalling and gravitropism.  相似文献

In order to examine a possible role of calcium in graviperception, the calcium ionophore A23187 was used to elevate the concentration of free cytoplasmic calcium in statocytes of the roots of Lepidium sativum L. After a brief incubation (30 min) in a medium that contained 10 micromoles A23187 and 5.5 micromoles CaCl2, 50% of the roots bent gravitropically during a subsequent 2 h of horizontal exposure, with an angle of curvature that varied from 5 degrees to 70 degrees. The corresponding statocytes exhibited a polar arrangement of cell organelles as did the controls. However, in statocytes from 50% of the roots which were not curved after gravistimulation a portion of the distal endoplasmic reticulum (ER) complex was displaced in the direction of gravity within 30 min of horizontal exposure. After washing of the briefly treated roots for 24 h with 1% dimethylsulfoxide the percentage of gravitropically bending roots increased to approximately 80%, but the angle of curvature amounted to only 5 degrees-10 degrees. Longer treatment (2 h) with A23187 caused a complete loss of graviresponsiveness which was accompanied by disintegration of statocyte polarity. We concluded from these results that i) calcium is involved in graviperception and ii) gravisensitivity depends on the integrity of statocytes.  相似文献

Axon regeneration is crucial for recovery after trauma to the nervous system. For neurons to recover from complete axon removal they must respecify a dendrite as an axon: a complete reversal of polarity. We show that Drosophila neurons in vivo can convert a dendrite to a regenerating axon and that this process involves rebuilding the entire neuronal microtubule cytoskeleton. Two major microtubule rearrangements are specifically induced by axon and not dendrite removal: 1) 10-fold up-regulation of the number of growing microtubules and 2) microtubule polarity reversal. After one dendrite reverses its microtubules, it initiates tip growth and takes on morphological and molecular characteristics of an axon. Only neurons with a single dendrite that reverses polarity are able to initiate tip growth, and normal microtubule plus-end dynamics are required to initiate this growth. In addition, we find that JNK signaling is required for both the up-regulation of microtubule dynamics and microtubule polarity reversal initiated by axon injury. We conclude that regulation of microtubule dynamics and polarity in response to JNK signaling is key to initiating regeneration of an axon from a dendrite.  相似文献

Abstract This time course and location of gravitropically induced curvatures in stems of goosegrass (Galium aparine L.), a member of the Rubiaceae, have been investigated. In the early stages of the response (0–5 h), curvature develops throughout the growing region, and is followed by an autotropic straightening which affects the internodes only, leading to the production of essentially straight internodes some 15 h after the onset of gravistimulation. Curvatures developing in the nodal regions, however, continue to increase over this period, and are not subject to reversal by autotropism. The nodal curvatures are not entirely dependent on the presence of any other part of the plant, since marked curvatures can be induced in isolated nodal segments. This pattern of response leads ultimately to correction of the growth direction of the plant by means of curvature responses confined exclusively to the nodes, despite the initial participation of both nodes and internodes in the gravitropic reaction.  相似文献

CYP78 is a plant-specific family of cytochrome P450 monooxygenases, some members of which regulate the plastochron length and organ size in angiosperms. The CYP78 family appears to be highly conserved in land plants, but there have been no reports on the role of CYP78s in bryophytes. The moss, Physcomitrella patens, possesses two CYP78As, CYP78A27 and CYP78A28. We produced single and double mutants and overexpression lines for CYP78A27 and CYP78A28 by gene targeting to investigate the function of CYP78As in P. patens. Neither the cyp78a27 nor cyp78a28 single mutant showed any obvious phenotype, while the double mutant exhibited severely retarded protonemal growth and gametophore development. The endogenous levels of some plant hormones were also altered in the double mutant. Transgenic lines that overexpressed CYP78A27 or CYP78A28 showed delayed and reduced bud formation. Our results suggest that CYP78As participate in the synthesis of a critical growth regulator in P. patens.  相似文献

Background: The frizzled (fz) gene of Drosophila encodes the founding member of the large family of receptors for the Wnt family of signaling molecules. It was originally studied in the adult epidermis, where it plays a key role in the generation of tissue polarity. Mutations in components of the fz signal transduction pathway disrupt tissue polarity; on the wing, hairs normally point distally but their polarity is altered by these mutations.Results: We devised a method to induce a gradient of fz expression with the highest levels near the distal wing tip. The result was a large area of proximally pointing hairs in this region. This reversal of polarity was seen when fz expression was induced just before the start of hair morphogenesis when polarity is established, suggesting that the gradient of Fz protein acted fairly directly to reverse hair polarity. A similar induction of the dishevelled (dsh) gene, which acts cell autonomously and functions downstream of fz in the generation of tissue polarity, resulted in a distinct tissue polarity phenotype, but no reversal of polarity; this argues that fz signaling was required for polarity reversal. Furthermore, the finding that functional dsh was required for the reversal of polarity argues that the reversal requires normal fz signal transduction.Conclusions: The data suggest that cells sense the level of Fz protein on neighboring cells and use this information in order to polarize themselves. A polarizing signal is transmitted from cells with higher Fz levels to cells with lower levels. Our observations enable us to propose a general mechanism to explain how Wnts polarize target cells.  相似文献

Using an ultrasensitive extracellular vibrating electrode, I have studied the membrane-generated electrical currents around the egg of the brown alga, Pelvetia, between fertilization and germination. During this period, the egg chooses an elongation axis and moves wall-precursor vesicles to the prospective growth region where they are secreted. This results in visible oöplasmic segregation which appears under the light microscope as a 1- to 2-μm-thick clear band at the cortex of the growth region. A steady electrical current enters a small region of the membrane and leaves the remainder of the egg's surface as early as 30 min after fertilization. This early spatial current pattern is unstable and shifts position, often with more than one inward current region. However, current enters mainly on the side where germination will occur and is usually largest at the prospective cortical clearing region. The average measured early current density is 0.06 μA/cm2 at 50 μm from the egg's surface, implying a surface current density of between 0.2 and 1 μA/cm2 due to the extrapolation uncertainty. At germination the current increases about twofold, resulting in a total transcellular current on the order of 100 pA. Unilateral growth-orienting light reversal stimulates inward current on the new dark side, and subsequent morphological polarity reversal is preceded by electrical polarity reversal. The steady current tends to increase when the external Ca2+ concentration is increased or the external Na+ concentration is decreased, suggesting that the current is carried in part by Ca2+. This current will generate a transcellular electrical field which may be the force driving the observed oöplasmic segregation.  相似文献

The seven subunit Arp2/3 complex is a highly conserved nucleation factor of actin microfilaments. We have isolated the genomic sequence encoding a putative Arp3a protein of the moss Physcomitrella patens. The disruption of this ARP3A gene by allele replacement has generated loss-of-function mutants displaying a complex developmental phenotype. The loss-of function of ARP3A gene results in shortened, almost cubic chloronemal cells displaying affected tip growth and lacking differentiation to caulonemal cells. In moss arp3a mutants, buds differentiate directly from chloronemata to form stunted leafy shoots having differentiated leaves similar to wild type. Yet, rhizoids never differentiate from stem epidermal cells. To characterize the F-actin organization in the arp3a-mutated cells, we disrupted ARP3A gene in the previously described HGT1 strain expressing conditionally the GFP-talin marker. In vivo observation of the F-actin cytoskeleton during P. patens development demonstrated that loss-of-function of Arp3a is associated with the disappearance of specific F-actin cortical structures associated with the establishment of localized cellular growth domains. Finally, we show that constitutive expression of the P. patens Arp3a and its Arabidopsis thaliana orthologs efficiently complement the mutated phenotype indicating a high degree of evolutionary conservation of the Arp3 function in land plants.  相似文献

The displacement of resin spheres attached to the tip of the gravitropically bending Chara rhizoid was measured by means of time-lapse photography. The relative growth of the flanks which at the beginning of stimulation were opposite to each other, was calculated during the earliest response time. The physically upper subapical flank section continues growing after stimulation at a decreasing rate. The growth of the opposite lower flank section is inhibited immediately after stimulation by the position of the statoliths. As soon as the statoliths are displaced in a basal direction, the growth of this section is transiently promoted. The gravitropic downward bending of the Chara rhizoid is by bowing, not by bulging.  相似文献

Influences underlying the direction of nematocyte migration in hydra were studied. Nematocytes arise by interstitial cell differentiation in the body column, and then up to 80% migrate into the ectodermal epithelial cells of the tentacles. The migration of these cells, which is clearly apically directed, may be due either to a chemotactic attraction into the hypostome and tentacles, or to a property inherent in the tissue of the body column, such as the regeneration polarity. To distinguish between these two possibilities, the rates of accumulation of 3H-proline-labeled desmoneme and stenotele nematocytes in unlabeled heads (hypostome and tentacles) grafted either basally or apically to the labeled body column were compared. Basally grafted heads, if left in place for an appropriate length of time, reversed the regeneration polarity of the tissue. In all experiments the direction of desmoneme migration was correlated with the direction (apical or basal) of the regeneration polarity of the tissue. Further, the kinetics of polarity reversal were modified by varying the grafting procedure or the environmental conditions. In every case the kinetics of reversal of desmoneme migration also paralleled the kinetics of reversal of tissue polarity. The results suggest that the direction of desmoneme migration is influenced by the regeneration polarity of the tissue. Stenotele migration was largely unaffected by tissue polarity, but behaved as though chemotactically attracted to the head.  相似文献

Cell biological, structural, and genetic approaches have demonstrated the presence of arabinogalactan proteins (AGPs) in the moss Physcomitrella patens and provided evidence for their function in cell expansion and specifically in the extension of apical tip-growing cells. Inhibitor studies indicated that apical cell expansion in P. patens is blocked by synthetic AGP binding beta-glucosyl Yariv reagent (betaGlcYR). The anti-(1-->5)-alpha-L-arabinan monoclonal antibody LM6 binds to some AGPs in P. patens, to all plasma membranes, and to the cell wall surface at the most apical region of growing protonemal filaments. Moreover, LM6 labeling of cell walls at the tips of apical cells of P. patens was abolished in the presence of betaGlcYR, suggesting that the localized movement of AGPs from the plasma membrane to the cell wall is a component of the mechanism of tip growth. Biochemical and bioinformatic analyses were used to identify seven P. patens ESTs encoding putative AGP core proteins from homology with Arabidopsis thaliana, Brassica napus, and Oryza sativa sequences and from peptide fragments isolated from betaGlcYR-precipitated AGPs. Gene knockout by homologous recombination of one of these genes, P. patens AGP1, encoding a classical AGP core protein, resulted in reduced cell lengths in protonemal filaments, indicating a role for AGP1 in apical cell expansion in P. patens.  相似文献

Control of tissue composition and organization will be a key feature in the development of successful products through tissue engineering. However, the mechanism of collagen fibril formation, growth, and organization is not yet fully understood. In this study we have examined collagen fibril formation in a wound healing model in which the newly formed fibrils were kept distinct from preexisting tissue through use of a porous tubular biomaterial implant. Samples were examined after 4, 6, 14, and 28 days by light microscopy, in situ hybridization, and immunofluorescence microscopy. These showed a normal wound healing response, with significant collagen formation at 14 and 28 days. Individual collagen fibrils were isolated from these samples by gentle extraction in a gentamicin-containing buffer which allowed extraction of a large proportion of intact fibrils. Examination by transmission electron microscopy showed that approximately 80% of the intact fibrils showed a single polarity reversal, with both ends of each fibril comprising collagen amino-terminal domains; the remaining fibrils had no polarity reversal. All fibrils had similar diameters at both time points. Immunoelectron microscopy showed that all labeled fibrils contained both type I and III collagens. These data indicate that this wound healing model provides a system in which collagen fibril formation can be readily followed.  相似文献

1 We examined the effects of nutrient availability on the competitive interactions of the New England salt marsh perennials that occupy discrete vegetational zones parallel to the shoreline.
2 Fertilized and unfertilized plots of pair-wise mixtures and monocultures of Spartina alterniflora, S. patens and Juncus gerardi were compared in order to assess the effects of nutrient addition on the competitive dynamics of these species in the field. In addition, we examined competition between some of these species and Distichlis spicata , a species common to disturbed marsh habitats.
3 After two growing seasons, changes in above-ground biomass of the species indicated that in fertilized plots, S. alterniflora outcompeted S. patens, S. patens outcompeted J. gerardi, and D. spicata outcompeted both J. gerardi and S. patens. This was the reverse of the interactions seen under ambient marsh conditions, and suggested that, under conditions of nutrient limitation, competitive dominance may result from efficient competition for nutrients.
4 Using a conceptual model of salt marsh zonation as a function of competition, physical stress and nutrient limitation, we hypothesize that a nutrient-induced reversal in the competitive dynamics among salt marsh perennials may result in modification of the pattern of plant zonation in this and similar marshes.  相似文献

A model is presented, based on this finding and previous studies, which proposes an electrochemical mechanism for the control of polarity and possibly for the maintenance of tissue organization in planarians. It is proposed that a bioelectric field exists and moves the electronegative brain-inhibiting substance in a posterior direction, establishing polarity. This model explains the polarity reversal experiments using external fields and many of the previously unexplained classical planarian experiments. Data are presented demonstrating the existence, magnitude, and polarity of this bioelectric field, which is not greatly altered upon decapitation, all in accord with predictions of the model.  相似文献

In darkness, protonemal filaments of Ceratodon purpureus (Brid.) grow negatively gravitropically (upwards). Red light induces a positive phototropic response mediated by the photoreceptor phytochrome. A red light treatment also has an inhibitory effect on the gravitropic response, an effect also mediated by phytochrome. In this study the effects of blue light on phototropism and on gravitropism were analysed. Unilateral blue light resulted in only a weak phototropic response, but markedly randomised growth direction. Blue light given together with a gravitropic stimulus reversed the gravitropism, changing it from negative to positive (filaments grow downward). The effect of blue light was also analysed with the mutant ptr116, which is defective in the biosynthesis of the phytochrome chromophore, and in a newly isolated mutant wwr2, which is positively gravitropic in darkness. Blue light induced the same reversal of gravitropism in ptr116 as in the wild type, indicating that phytochrome is not involved in this process. In wwr2 the direction of gravitropism was unaltered by the blue light treatment. Light also affects chlorophyll content and the size of plastids, potential statoliths for gravitropism. Red light induced an increase in plastid size and chlorophyll content in the wild type but not in ptr116. Blue light induced a similar change in wild type plastids. It seems as though light-induced alterations of gravitropism are not simply mediated by alterations in plastid properties, and that red light and blue light evoke fundamentally different responses. Diverse methods for the disruption of cell entanglements and pellets of the moss Physcomitrella patens were tested in order to improve the homogeneity of suspension cultures. The morphological characterization of the moss was carried out by means of image analysis. Selected morphological parameters were defined and compared to the reduction of the carbon dioxide fixation, and the released pigments after cell disruption. The size control of the moss entanglements based on the rotor stator principle allowed a focused shear stress, avoiding a severe reduction in the photosynthesis. Batch cultures of P. patens in a 30.0-l pilot tubular photo-bioreactor with cell disruption showed no significant variation in growth rate and a delayed cell differentiation, when compared to undisrupted cultures. A highly controlled photoautotrophic culture of P. patens in a scalable photo-bioreactor was established, contributing to the development required for the future use of mosses as producers of relevant heterologous proteins.  相似文献

Gravicurvature in water- and auxin (IAA)-incubated coleoptiles of rye ( Secale cereale L.) is similar, despite a general strongly enhancing effect of exogenous IAA on the overall (cell) elongation of these organs. Longitudinally split coleoptiles or isolated longitudinally halved coleoptiles (horizontally positioned as upper or lower halves) respond gravitropically in the same way as water-incubated intact coleoptiles, irrespective of whether the halves are incubated in distilled water or IAA. A new model for the principal mechanism of regulation of gravitropic growth is proposed which depends on, yet does not involve, the redistribution of IAA as the means for gravistimulated differential growth, as postulated by the Cholodny-Went hypothesis (CWH). It is based on a gravimediated temporarily restrained infiltration of IAA-induced wall-loosening factors into the growth-limiting outer epidermal walls of the concave organ flank.  相似文献
