The prefrontal cortex (PFC) is involved in mediating important higher-order cognitive processes such as decision making, prompting thereby our actions. At the same time, PFC activation is strongly influenced by emotional reactions through its functional interaction with the amygdala and the striatal circuitry, areas involved in emotion and reward processing. The PFC, however, is able to modulate amygdala reactivity via a feedback loop to this area. A role for serotonin in adjusting for this circuitry of cognitive regulation of emotion has long been suggested based primarily on the positive pharmacological effect of elevating serotonin levels in anxiety regulation. Recent animal and human functional magnetic resonance studies have pointed to a specific involvement of the 5-hydroxytryptamine (5-HT)2A serotonin receptor in the PFC feedback regulatory projection onto the amygdala. This receptor is highly expressed in the prefrontal cortex areas, playing an important role in modulating cortical activity and neural oscillations (brain waves). This makes it an interesting potential pharmacological target for the treatment of neuropsychiatric modes characterized by lack of inhibitory control of emotion-based actions, such as addiction and other impulse-related behaviors. In this review, we give an overview of the 5-HT2A receptor distribution (neuronal, intracellular, and anatomical) along with its functional and physiological effect on PFC activation, and how that relates to more recent findings of a regulatory effect of the PFC on the emotional control of our actions. 相似文献

Abstract: In the rat parotid gland, the neuropeptide substance P (SP), as well as SP(4–11), and septide elicited inositol phosphate production (EC50 values 0.44, 2, and 20 n M , respectively). No additivity of the maximal response to the three agonists was observed. SP, SP(4–11), and septide also stimulated protein secretion; for SP, two EC50 were determined (0.5 and 160 n M ), whereas a single one could be determined for SP(4–11) and septide (EC50 values 15 and 20 n M , respectively). The selective tachykinin NK1 receptor antagonist RP67580 acted as a competitive inhibitor of both SP- and SP(4–11)-induced inositol phosphate production. Its effect on septide-induced inositol phosphate production was noncompetitive. RP67580 is apparently as potent at antagonizing septide, SP, or SP(4–11) (in all cases K B = 3 n M ). These results show that in parotid gland, only NK1 receptors are activated by SP, SP(4–11), and septide. We also showed that the protein secretion stimulated by SP was inhibited competitively by RP67580, whereas the effect of RP67580 was noncompetitive on protein secretion when SP(4–11) or septide was used. Our data indicate that in rat parotid gland, the existence of a specific "septide-sensitive" receptor can be ruled out and that only the NK1 receptor is present and mediates cellular responses. Taken together, these results show that in this tissue the NK1 receptor would present at least two different binding sites that could be coupled to different transduction pathways and that would regulate protein secretion. 相似文献

It is generally accepted that in most insects adults are sexually immature when they initiate migration and that migratory behaviour terminates with the onset of sexual maturation. However, a few studies examining the mating status of field collected moths have suggested that sexually mature individuals may continue migrating, but in these cases it was impossible to completely eliminate the possibility that the mated females captured came from local, non-migrant populations. In this study we examined the ovarian development of Mythimna separata females captured using a vertical pointing searchlight trap on Beihuang Island in the Bohai Gulf, China, a site >40 km from land. Moths were collected from May to October from 2003 to 2008 in order to test the hypothesis that the onset of sexual maturation resulted in the termination of migratory behaviour. While females at the end of the summer had little ovarian development and were unmated, a significant proportion of those migrating northward in the early summer had developed ovaries and often had at least one spermatophore. Given that theses insects were captured while flying up to 500 m above sea level, at a site with no local populations, the findings would not support the hypothesis and suggest that both ovarian development and mating may occur during migration. 相似文献

Background
The interaction of tumor necrosis factor-α (TNF-α) with its receptors: TNFRSF1A and TNFRSF1B is critical for the promotion of tumor growth, invasion and metastasis. To better understand the roles of single nucleotide polymorphisms (SNPs) in the TNF-α, TNFRSF1A and TNFRSF1B genes in the development of breast cancer, we explored the associations between SNPs in these three genes and breast cancer susceptibility in northeast Chinese Han women.Methodology/Principal Findings
This case-control study was conducted among 1016 breast cancer patients and 806 age-matched healthy controls. Seven SNPs in the TNF-α (rs1800629, rs361525), TNFRSF1A (rs767455, rs4149577 and rs1800693) and TNFRSF1B (rs1061622 and rs1061624) genes were genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. In TNFRSF1B, the rs1061622 GT genotype and the G allele conferred a reduced susceptibility to breast cancer (P = 0.000662, OR = 0.706, 95% CI: 0.578–0.863; P = 0.002, OR = 0.769, 95% CI; 0.654–0.905, respectively). Moreover, the AG genotype, the AA genotype and the A allele in rs1061624 conferred an increased risk of breast cancer (P = 0.007, OR = 1.470, 95% CI:1.112–1.943; P = 0.00109, OR = 1.405 95% CI:1.145–1.724; P = 0.001, OR = 1.248 95% CI:1.092–1.426, respectively). These two SNPs also had associations with breast cancer risk under the dominant model. In haplotype analysis, the CTA (rs767455 C-rs4149577 T-rs1800693 A) haplotype in TNFRSF1A and the TA (rs1061622 T-rs1061624 A) haplotype in TNFRSF1B had higher frequencies in breast cancer patients (P = 0.00324; P = 0.000370, respectively), but the frequency of GG (rs1061622 G-rs1061624 G) haplotype in TNFRSF1B was lower in breast cancer patients (P = 0.000251). The associations of the three haplotypes remained significant after correcting for multiple testing. In addition, significant associations were also observed between TNFRSF1A polymorphisms and lymph node metastasis, P53, estrogen receptor (ER) and progesterone receptor (PR) statuses.Conclusions
Our results suggest that rs1061622 and rs1061624 in TNFRSF1B may affect breast cancer risk, and SNPs in TNFRSF1A are associated with the clinical features of breast cancer. 相似文献8.
In 1719 Morgagni described a condition, today known as hyperostosis frontalis interna (HFI), as one sign within a triad consisting of HFI, virilism, and obesity. Today, HFI is predominantly found in older women. Although the etiology of HFI has not yet been determined precisely, the condition has been linked to metabolic disorders. HFI is reported to be rare in the archaeological record and the frequency of the condition is thought to have increased during the 19th and 20th centuries. We present preliminary results on the occurrence of HFI in the commingled human bone assemblage from “tomb VII” discovered underneath the Bronze Age royal palace of the ancient city of Qatna, Syria. A preliminary minimal number of individuals of 70 has been estimated for the as yet not fully analyzed skeletal remains. Skull fragments of nine individuals exhibit endocranial bone formations consistent with HFI. Rarity of stress indicators in the skeletons, the rich grave goods, and the burial place within the area of the Royal palace are suggestive of a high social status and an economically favorable situation of the buried individuals. Assuming that their life style included a high calorie diet in combination with little physical activity, acquired metabolic disorders may have been present in many individuals. The comparatively high number of individuals presenting HFI in the studied sample might therefore be viewed as being related to their high social status. Multiple occurrences of HFI in archaeological skeletal assemblages might serve as a proxy for social status. 相似文献

The C1 domains of classical and novel PKCs mediate their diacylglycerol-dependent translocation. Using fluorescence resonance energy transfer, we studied the contribution of different negatively charged phospholipids and diacylglycerols to membrane binding. Three different C1B domains of PKCs were studied (the classical γ, and the novel δ and ?), together with different lipid mixtures containing three types of acidic phospholipids and three types of activating diacylglycerols. The results show that C1Bγ and C1B? exhibit a higher affinity to bind to vesicles containing 1-palmitoyl-2-oleoyl-sn-phosphatidic acid, 1-palmitoyl-2-oleoyl-sn-phoshatidylserine, or 1-palmitoyl-2-oleoyl-sn-phosphatidylglycerol, with C1B? being the most relevant case because its affinity for POPA-containing vesicles increased by almost two orders of magnitude. When the effect of the diacylglycerol fatty acid composition on membrane binding was studied, the C1B? domain showed the highest binding affinity to membranes containing 1-stearoyl-oleoyl-sn-glycerol or 1,2-sn-dioleoylglycerol with POPA as the acidic phospholipid. Of the three diacylglycerols used in this study, 1,2-sn-dioleoylglycerol and 1-stearoyl-oleoyl-sn-glycerol showed the highest affinities for each isoenzyme, whereas 1,2-sn-dipalmitoylglycerol; showed the lowest affinity. Rat liver peroxisomes contain in their matrix the α-subunit of the mitochondrial F1-ATPase complex. The identification of this protein in liver peroxisomes has been achieved by immunoelectron microscopy and subcellular fractionation. No β-subunit of the mitochondrial F1-ATPase complex was detected in the peroxisomal fractions obtained in sucrose gradients or in Nycodenz pelletted peroxisomes. The consensus peroxisomal targeting sequence (Ala-Lys-Leu) is found at the carboxy terminus of the mature α-subunit from bovine heart and rat liver mitochondria. Due to the dual subcellular localization of the α-subunit and to the structural homologies that exist between this protein and molecular chaperones [(1990) Biol. Chem. 265, 7713-7716] it is suggested that the protein should perform another functional role(s) in both organelles, plus to its characteristic involvement in the regulation of mitochondrial ATPase activity. 相似文献

Summary Electron microscopy study shows that cytochalasin treatment of the mullet damages the microfilaments system in the apex of gill ionocytes: the microfilaments are reduced in number and shortened. Cytochalasin causes a reduction of transgill potential difference and an increase of the Na+ and Cl– blood concentration, of the diffusional water permeability of the gill, of the Na+ branchial influx and of Cl– efflux. The increase of the Na+ influx may result in a reduction of the Na+ net excretion flux compared to the control. The increased permeability in cytochalasin treated fish facilitates the Cl– entry probably leading to a reduction of the net Cl– excretion. The partial inhibition of the K+ dependent components of Na+ and Cl– effluxes also contributes to the reduction of Na+ and Cl– excretion. The role of microfilaments in the mechanisms of ionic excretion by the gill is discussed. 相似文献

The Apolipoprotein A5 (APO A5) ?1131T/C, fibrinogen β (FgB) ?455G/A, ?148C/T, and cholesteryl ester transfer protein (CETP) TaqIB gene polymorphisms have been indicated to be associated with the coronary artery disease (CAD) risk, but the individual study results are still inconsistent. To explore the relationship between APO A5 ?1131T/C, FgB ?455G/A, ?148C/T, and CETP TaqIB gene polymorphisms and CAD in the Chinese population, the current meta-analysis involving 15,055 subjects from 40 individual studies was conducted. The pooled odds ratio (OR) for the association between APO A5 ?1131T/C, FgB ?455G/A, ?148C/T, and CETP TaqIB gene polymorphisms and CAD and its corresponding 95 % confidence interval (95 % CI) were evaluated by random or fixed effect model. A significant association between APO A5 ?1131T/C gene polymorphism and CAD in the Chinese population was found under an allelic (OR: 1.33, 95 % CI: 1.22–1.44, P < 0.00001), recessive (OR: 1.67, 95 % CI: 1.25–2.25, P = 0.0006), dominant (OR: 0.820, 95 % CI: 0.767–0.876, P = 1.0 × 10?10), homozygous (OR: 2.36, 95 % CI: 1.55–3.58, P < 0.0001) and heterozygous genetic models (OR: 1.136, 95 % CI:1.075–1.200, P = 1.0 × 10?10). A significant association between FgB ?455G/A gene polymorphism and CAD was also detected in the Chinese population under an allelic (OR: 1.50, 95 % CI: 1.25–1.81, P < 0.0001), dominant (OR: 0.864, 95 % CI: 0.819–0.912, P = 1.0 × 10?10), homozygous (OR: 1.616, 95 % CI: 1.213–2.152, P = 0.001) and heterozygous genetic models (OR: 1.245, 95 % CI:1.138–1.361, P = 1.0 × 10?10). No significant association was found between them under a recessive genetic model (OR: 1.124, 95 % CI: 0.844–1.497, P = 0.424). A significant association was also found between FgB ?148C/T gene polymorphism and CAD in the Chinese population under an allelic (OR: 1.34, 95 % CI: 1.06–1.71, P = 0.02), recessive (OR: 1. 65, 95 % CI: 1.02–2.69, P = 0.04), dominant (OR: 0.924, 95 % CI: 0.872–0.978, P = 0.007) and homozygous genetic models (OR: 0.968, 95 % CI: 0.942–0.995, P = 0.018). No significant association was found between them under a heterozygous genetic model (OR: 0.979, 95 % CI: 0.937–1.023, P = 0.342). In the whole Chinese population, no significant association between the CETP TaqIB gene polymorphism and CAD was found under an allelic (OR: 1.17, 95 % CI: 0.94–1.45, P = 0.15), dominant (OR: 1.46, 95 % CI: 0.80–2.67, P = 0.22) or recessive genetic models (OR: 0.68, 95 % CI: 0.32–1.44, P = 0.31). However, in the subgroup analysis stratified by ethnicity, there was a significant association between them under an allelic (OR: 1.27, 95 % CI: 1.07–1.52, P = 0.007) and dominant genetic model (OR: 2.04, 95 % CI: 1.49–2.79, P < 0.00001) in the Han subgroup. In the Chinese population, the APO A5 ?1131T/C and FgB ?455G/A, ?148C/T gene polymorphisms were implied to be associated with CAD susceptibility. The APO A5 ?1131C, FgB ?455A, and ?148T alleles might confer susceptibility to CAD. CETP TaqIB gene polymorphism was suggested to be associated with CAD susceptibility in the Chinese Han population. Carriers with B1 allele of CETP TaqIB gene might be predisposed to CAD in the Chinese Han population. 相似文献

The new ligand 4′-(4?-pyridyl-N-oxide)-2,2′:6′,2″-terpyridine (pyNoxterpy) and its homoleptic iron(II) complex have been synthesised, and structural and spectroscopic studies have been carried out. The obtained results have been compared with the reported data for the parent ligand 4′-(4?-pyridyl)-2,2′:6′,2″-terpyridine (pyterpy) and its homoleptic iron(II) complex. In this study we isolate a heptameric peptide from the marine microalga Pavlova lutheri and examine its inhibitory effect and mechanism of action on the matrix metalloproteinase-9 (MMP-9) protein in human fibrosarcoma HT1080 cells. Through consecutive rounds of purification we isolated a potent MMP-9 inhibitory peptide composed of seven amino acids: Leu-Leu-Ala-Pro-Pro-Glu-Arg (796.4 Da). We show that the mRNA and protein expression levels of MMP-9 are reduced in the presence of this purified peptide. Furthermore, we show that this peptide inhibits PMA-induced translocation of nuclear factor-κB (NF-κB) by suppressing IκB-α phosphorylation and that it suppresses the phosphorylation of the c-Jun N-terminal (JNK) and p38 mitogen-activated protein kinases (MAPKs). Our results indicate that this peptide, purified from P. lutheri, has the potential to be a valuable natural preventive agent against MMP-related diseases. 相似文献

A series of carboxamide and sulphonamide alkyl(ethyl to hexyl)piperazine analogues were prepared and tested for their affinity to bind to a range of receptors potentially involved in psychiatric disorders. These chemical modifications led us to explore the impact of homology and bioisosteric replacement of the amide group. All of these compounds possessed a high affinity for 5-HT1A receptors, irrespective of the size of the linker, the carboxamide derivative with a pentyl linker had the highest affinity for α2A receptor sites and also a high affinity for 5-HT1A and D3 receptors. Recent studies in receptor-transfected cell lines have demonstrated that
extracellular signal-regulated kinase (ERK) activation by angiotensin type 1A
receptor and other G protein-coupled receptors can be mediated by both G
protein-dependent and β-arrestin-dependent mechanisms. However, few
studies have explored these mechanisms in primary cultured cells expressing
endogenous levels of receptors. Accordingly, here we utilized the
β-arrestin biased agonist for the angiotensin type 1A receptor,
SII-angiotensin (SII), and RNA interference techniques to investigate
angiotensin II (ANG)-activated β-arrestin-mediated mitogenic signaling
pathways in rat vascular smooth muscle cells. Both ANG and SII induced DNA
synthesis via the ERK activation cascade. Even though SII cannot induce
calcium influx (G protein activation) after receptor stimulation, it does
cause ERK activation, although less robustly than ANG. Activation by both
ligands is diminished by depletion of β-arrestin2 by small interfering
RNA, although the effect is more complete with SII. ERK activation at early
time points but not later time points is strongly inhibited by those protein
kinase C inhibitors that can block protein kinase Cζ. Moreover, ANG- and
SII-mediated ERK activation require transactivation of the epidermal growth
factor receptor via metalloprotease 2/9 and Src kinase. β-Arrestin2
facilitates ANG and SII stimulation of Src-mediated phosphorylation of Tyr-845
on the EGFR, a known site for Src phosphorylation. These studies delineate a
convergent mechanism by which G protein-dependent and
β-arrestin-dependent pathways can independently mediate ERK-dependent
transactivation of the EGFR in vascular smooth muscle cells thus controlling
cellular proliferative responses.G protein-coupled receptors, also known as seven transmembrane
(7TM)2 receptors,
control virtually all known physiological processes in mammals
(1). The various functions of
these receptors are mediated and modulated by three families of proteins,
which share the property that they interact virtually universally with the
receptors in a strictly stimulus-dependent way
(1). These three families of
proteins are the heterotrimeric G proteins, the G protein-coupled receptor
kinases (GRKs), and the β-arrestins. Activation of the receptors
stimulates classical G protein-dependent signaling, often involving regulation
of levels of second messengers such as cAMP and diacyglycerol. However, as has
been known for many years, interaction of activated receptors with GRKs
leading to their phosphorylation, and subsequent interaction with
β-arrestins leads to desensitization of G protein signaling.In recent years, however, it has become increasingly clear that the
β-arrestin-GRK system is in fact bifunctional
(2). Thus, even as it
desensitizes G protein signaling by the receptors, it also serves as a signal
transduction system in its own right, activating a growing list of signaling
pathways. These positive signaling functions are often mediated by the ability
of β-arrestin to serve as an adaptor or scaffold molecule, bringing
elements of diverse signaling pathways into proximity with one another and the
receptors and thereby facilitating their activation. This new paradigm for
understanding the previously unrecognized signaling properties of the
β-arrestin-GRK system has been explored in a wide variety of transfected
cultured cell systems.However, to date, relatively little investigation of these novel signaling
pathways has been carried out in primary cell culture systems expressing
endogenous levels of 7TM receptors. In seeking such a system in which to
characterize and compare β-arrestin and G protein-mediated signaling
pathways from a typical 7TM receptor, our attention was drawn to cultured rat
vascular smooth muscle cells (VSMCs). Several features of rat VSMCs suggest
this to be a relevant system for these purposes. Rat VSMCs express a variety
of physiologically important 7TM receptors including the angiotensin II type
1A receptor (AT1R) (3). This
receptor has been the focus of extensive study in transfected cell systems
with respect to its β-arrestin-mediated signaling to a variety of
pathways, most particularly extracellular signal-regulated kinase (ERK).
Moreover, the AT1R mediates the physiologically important effects of
angiotensin II (ANG) on vascular tone as well as on proliferation and
chemotaxis (4,
5). Pathophysiologically, ANG
stimulation of this receptor has been implicated in VSMC proliferation and
chemotaxis, which are thought to play an important role in such important
disease processes as atherosclerosis and restenosis after angioplasty
(6,
7). Moreover, a ligand has been
characterized
[Sar1,Ile4,Ile8](SII)-angiotensin (SII), a
triply mutated angiotensin octapeptide that, in transfected cell systems, acts
as a specific agonist for β-arrestin-mediated signaling, although not
activating G protein-mediated signaling
(8).Accordingly, in the studies described here, we set out to investigate the
characteristics of activation of ERK in rat VSMCs that might be mediated
through G protein as well as β-arrestin signaling. The results not only
demonstrate the importance of β-arrestin-mediated signaling in
ERK-mediated proliferative responses of these cells, but also shed new light
on the molecular mechanisms and interrelationships between the β-arrestin
