Summary The vertical distributions of species of chaetognaths are described to a depth of 1043 m in Baffin Bay during the summer. The dominant species in the southern region of the Bay were Eukrohnia hamata and Sagitta maxima and, in the northern region, E. hamata and S. elegans. The younger stages of all chaetognath species were found at depths above the older more mature stages. There was no evidence of any diurnal vertical migration by any species. The population structure of E. hamata was bimodal suggesting two sub-populations and a two year life cycle. Sagitta maxima showed a multimodal population suggesting a life cycle of four or more years. The population structure of E. elegans was unimodal. Gut contents showed that E. hamata fed at a higher rate than did S. maxima. E. hamata fed at a higher rate during the night but there was no significant difference between day/night feeding rates for S. maxima. It was estimated that E. hamata consumed up to 1.8 prey items day-1 and S. maxima 0.5 prey items day-1. The daily ration was influenced by the water temperature. Calculations showed chaetognaths are major predators on copepods consuming between 1.2 and 1.3% of the standing crop of copepods per day. Generally the maximum concentration of chaetognaths was found at a depth below the maximum concentration of copepods.  相似文献

The taxonomic placement of strains belonging to the extremophilic red alga Galdieria maxima has been controversial due to the inconsistent phylogenetic position inferred from molecular phylogenetic analyses. Galdieria maxima nom. inval. was classified in this genus based on morphology and molecular data in the early work, but some subsequent molecular phylogenetic analyses have inferred strains of G. maxima to be closely related to the genus Cyanidioschyzon. To address this controversy, an isolated strain identified as G. maxima using the rbcL gene sequence as the genetic barcode was examined using a comprehensive analysis across morphological, physiological, and genomic traits. Herein are reported the chloroplast-, mitochondrion-, and chromosome-level nuclear genome assemblies. Comparative analysis of orthologous gene clusters and genome arrangements suggested that the genome structure of this strain was more similar to that of the generitype of Cyanidioschyzon, C. merolae than to the generitype of Galdieria, G. sulphuraria. While the ability to uptake various forms of organic carbon for growth is an important physiological trait of Galdieria, this strain was identified as an ecologically obligate photoautotroph (i.e., the inability to utilize the natural concentrations of organic carbons) and lacked various gene models predicted as sugar transporters. Based on the genomic, morphological, and physiological traits, we propose this strain to be a new genus and species, Cyanidiococcus yangmingshanensis. Re-evaluation of the 18S rRNA and rbcL gene sequences of the authentic strain of G. maxima, IPPAS-P507, with those of C. yangmingshanensis suggests that the rbcL sequences of “G. maxima” deposited in GenBank correspond to misidentified isolates.  相似文献

A shoot regeneration protocol was developed for five cultivars of theCucurbitaceae. The influence of the seed coat, an ethanol pre-treatment,different seed sources, NaOCl concentrations and treatment times ondecontamination were investigated. The effects of combinations of BA, kinetin,iP and TDZ with IAA in the culture medium on shoot regeneration of cotyledonaryexplants were tested. The cultivars Cucurbita maxima cv.A-line, C. maxima cv. Chicago Warted and C.pepo cv. Rolet, did not form shoots on any of the treatments.However, somatic embryos formed on C. maxima cv. ChicagoWarted explants. Cucumis melo cv. Hales Best 36 was highlyregenerable and shoots formed on all the treatments containing cytokinin. Theshoot regeneration response was significantly higher on BA-containing media thanwith the other cytokinins tested. Although Cucumis sativuscv. Ashley responded poorly, shoot development was observed on media containingBA or iP.  相似文献

Despite the fact that the stomach of turbot Psetta maxima is a curved tube that forms a half circle, it was demonstrated that gastric evacuation in this predatory flatfish fed natural prey closely followed the surface‐dependent cylinder model developed from studies on gadoids with a straight stomach. Evacuation experiments were performed on two size groups of P. maxima fed sandeel Ammodytes tobianus as well as on P. maxima fed brown shrimp Crangon crangon at three different temperatures. This enabled the provision of a gastric evacuation model for studies on P. maxima, which takes into account the effects of the explanatory variables predator size, temperature, prey energy density and resistance of prey to the digestive processes in the stomach. Basically, the cylinder model predicts that a square‐root function accurately describes gastric evacuation, which is inconsistent with the conclusion of a previous study on P. maxima that evacuation of A. tobianus is essentially linear with time. Use of the cylinder model to the values of the explanatory variables presented in the latter study, however, gave accurate predictions of the actually acquired evacuation data, which points to the generic value of the model.  相似文献

Summary 10 species of Mysidacea were sampled during the Polarstern and Walther Herwig SIBEX cruises to the Antarctic Peninsula region from 1983 to 1986. The commonest were Antarctomysis maxima, A. ohlini and Mysidetes posthon. For these species the size and maturity stage composition as well as the length-weight relationships are given. The species considered are growing slowly and attain a long life span, M. posthon reached age class 3+, A. ohlini at least lived as long as 5 years, while A. maxima was found to be of age 5+ in the Peninsula area and probably 6+ in the Weddell Sea. The generation time of Antarctomysis species was 4 years and 3 years in Mysidetes posthon. Fecundity was low, mean number of offspring was about 13 for M. posthon and 21 for A. maxima. Rematuration was observed for the species A. maxima, indicating several spawning events during its life span. Biomass production is low for A. maxima, shown by a P/B-index of 0.98. Mortality of this species was estimated to be Z=1.1, which indicates that 33% of the specimens of an age group survive until the next year. The distribution and spatial separation of the two Antarctomysis species is discussed.  相似文献

The purpose of this study was to construct a cosmid library from chromosomal DNA of a marine macroalga, Bryopsis maxima Okamura ex Segawa (Bryopsidales, Ulvophyceae), in a rapid, simple and inexpensive manner. In the DNA purification, polysaccharides were removed by covalently binding them to resin particles containing free boric acid groups. The DNA yield was 20 μg g?1 of B. maxima fresh weight. This DNA was 100–200 kb in length, and its A260/A280 and A230/A260 ratios were 1.8 and 0.4, respectively. It was of sufficient quality for molecular research. The cloning procedures were carried out in the following steps: controlled partial shearing of purified DNA through a microsyringe, optimal size separation of the DNA by biased sinusoidal field gel electrophoresis, ligation of the DNA to the cosmid vector in the gel, and in vitro packaging into the lambda phage. The library consisted of 2.0 × 103 independent clones with an average insert size of 40 kb. The fragment amplified by polymerase chain reaction in the library was hybridized with a DNA fragment (328 bp) encoding B. maxima glutamate dehydrogenase under high‐stringency conditions by Southern blot analysis, thus demonstrating that the library contained B. maxima chromosomal DNA. This cosmid library is the first to be constructed for any species of marine macroalgae.  相似文献

The accumulation of cadmium (Cd) by Tetraselmis chuii and Spirulina maxima was studied with dead and growing cells. Results indicated that the 2 microorganisms accumulated Cd by 2 different means according to the mechanisms involved—metabolism-dependent or metabolism-independent sorption. The mechanism involved in Cd accumulation on Tetraselmis chuii was restricted to surface phenomena, while in Spirulina maxima, Cd was accumulated on different layers of the cyanobacterium surface. In order to select a suitable immobilization support for the cells, several seaweeds were tested. Two types of seaweed were selected for experiments, using a small continuous pilot unit: Sargassum sp., a strong Cd adsorber, and Ulva sp., a poor one. The column reactors of the continuous system were filled with the algal supports and covered with dense microbial biofilms of Tetraselmis chuii or Spirulina maxima. The results obtained proved the success of the association between living microbial cells and dead seaweeds for operation of the continuous system.  相似文献

Testing whether rare species are limited by narrow habitat conditions from correlations between species occurrence and measured environmental factors is usually hampered by the lack of statistical power associated to the low number of observations. Aneura maxima is an exceedingly rare liverwort in Europe whose recent discovery precluded the inclusion within the Red Data Book of European bryophytes. A series of new observations resulting from intensive bryophyte grid-mapping at the border between France and Belgium allowed the statistical investigation of the factors accounting for its distribution at the landscape scale. The species was systematically observed in deep ravines on damp, loamy soils under light tree cover within broadleaf woodlands. These conditions differ from those of other European localities, suggesting that A. maxima is not limited by a narrow ecological range. Attempting to predict the occurrence of A. maxima at the landscape scale from data on land use and soil conditions resulted in a model where the species was actually observed in only a half of the squares where its probability of occurrence was the highest. The species thus does not seem to have colonized all its potential habitats, which may be explained by the poor dispersal ability of this dioecious, rarely fertile species. Given the relative frequency of A. maxima in the Semois river basin, but taking into account its seemingly low dispersal ability and its preference for swampy habitats that are threatened by draining and spruce plantations, a status as 'conservation dependent' to 'near threatened' within the category 'lower risk' is proposed.  相似文献

 Morphological and electrophoretic data were studied to examine species delimitation, patterns of morphological and genetic variation in three Korean Hepatica including two endemics, H.␣maxima and H. insularis. Based on a phenogram using 15 morphological characters, taxa were distinct; it was consistent with the phenogram based on genetic distance. In the enzyme electrophoresis study, the genetic identities suggested that three taxa were genetically divergent enough to be recognized as different species, falling within the range expected␣for congeners. The genetic identity between H.␣asiatica and H. insularis was higher than the values between these two taxa and H.␣maxima, a restricted endemic of Ulleung Island. The least genetic variation was found in H. maxima and the greatest in widespread H. asiatica. These data are consistent with theoretical expectations that small populations are more likely to be genetically depauperate. The biochemical pathways followed by a mixed bacterial culture and one of its constituent strains, Sarcina maxima, MTCC 5216 (hitherto unreported) during the degradation of mononitrophenol isomers was studied using extensive nuclear magnetic resonance (NMR) spectroscopy (One- and Two-Dimensional Heteronuclear Multiple Quantum Coherence Transfer-2D HMQCT NMR). NMR investigations revealed that o-nitrophenol (ONP) could be degraded by the consortium to metabolites such as catechol, cis, cis-muconic acid, γ-hydroxymuconic semialdehyde, maleylacetate and β-ketoadipate. The spectra of ONP reaction mixture degraded by S.␣maxima showed that formation of maleylacetate from γ-hydroxymuconic semialdehyde should go through a new metabolite γ-hydroxymaleylacetate, hitherto unreported. The consortium could breakdown m-nitrophenol (MNP) to 4-aminocatechol indicating that it came from 3-hydroxyaminophenol. However, S. maxima MTCC 5216, could convert MNP to hitherto unreported 2-nitrohydroquinone and the subsequent 2-hydroxylaminohydroquinone to 1,2,4-benzenetriol along with γ-hydroxymuconic semialdehyde, muconolactone and maleylacetate. The pathway followed by the consortium during p-nitrophenol (PNP) degradation was by the formation of 4-nitrocatechol, maleylacetate and β-ketoadipate. PNP reaction mixture of S.␣maxima, MTCC 5216 on the other hand, showed that the pathway could proceed through the formation of p-hydroquinone as the initial metabolite. The present study conclusively established the nitrophenol-degrading ability of both the consortium and S. maxima MTCC 5216, including exhibiting slight deviations from the pathways followed by the other reported microorganisms.  相似文献

Summary The AT-rich highly repeated satellite DNA of Cucurbita pepo (zucchini) and Cucurbita maxima (pumpkin) were cloned and their DNA structure was investigated. DNA sequencing revealed that the repeat length of satellite DNA in Cucurbita pepo is 349–352 base pairs. The percentage of AT-base pairs is about 61%. This satellite is highly conserved in restriction enzyme pattern and DNA sequence; sequence heterogeneity is about 10%. In contrast, the satellite DNA of Cucurbita maxima has a repeat length of 168–169 base pairs. This satellite is also rich in AT-base pairs (64%), existing in at least three different variants as revealed by restriction enzyme analysis and DNA sequencing. The sequence heterogeneity between these variants is about 15%. The two satellite DNAs showed no cross-hybridization to each other and sequence homology is only limited. Nevertheless, we found in the C. pepo genome a high amount of sequences resembling the satellite of C. maxima. In contrast, the satellite repeat of C. pepo is found in the C. maxima DNA only in a few copies. These observations were discussed with respect to satellite DNA evolution and compared to the data received from monocotyledonous species.  相似文献

The nucleotide (nt) sequence of the 5508-nt intergenic spacer (IGS), between the 25S- and the 18S-coding regions of Cucurbita maxima rDNA, was determined. The fragment sequenced is 6142 nt long and includes 472 nt of 25S- and 162 nt of 18S-coding regions. The IGS has a complex primary structure, composed of five repetitive families (A-E) and three unique domains. It is dominated by the presence of nine, tandemly-repeating units of approximately 250 nt (repeat D), each unit containing four copies of an internal subrepeat (repeat E). The repetitive units show sequence variability consisting of nt changes, insertions and deletions. Upstream of the nine D repeats and between two copies of the B repeat is a 575-nt region, highly G + C rich (83%) and heavily biased toward C (58%) in the sense strand. Within this region are six repetitive units, averaging 42 nt (repeat C) each, containing but a single A nt. Downstream from the terminus of the 25S-coding sequence, are two tandem copies of the 103-nt A repeat. The IGS of C. maxima is longer and more complex than that of other plant IGSs described to date. The 600 nt at the 5' portion of cucurbit IGS is more conserved in evolution than the remainder, as revealed by comparison of C. maxima and C. pepo IGS restriction maps and by nucleotide sequence comparison of C. maxima and Cucumis sativa IGSs.  相似文献

In South Africa, more than 7000 t (f wt) of kelp (Ecklonia maxima) fronds are harvested annually to feed cultured abalone. Carpoblepharis flaccida, Gelidium vittatum and Polysiphonia virgata are conspicuous red algal epiphytes on older kelps and provide habitat and food for numerous animals. Over 4.5 y, we examined the effects of one destructive harvest of E. maxima on these 3 epiphytes. Two 20 × 20 m plots of kelp with similar epiphyte loads were demarcated. In one, all E. maxima sporophytes with stipes longer than 50 cm were harvested. The other plot served as a control. After 2.5 y the biomass of E. maxima in the harvested plot had recovered to control levels, but the epiphyte load (g epiphytes. Kg kelp−1) was statistically lower in the harvested plot after 2.5 and 3.5 y, and only recovered after 4.5 y. While most commercial harvesters cut through the “heads” (primary blades) of the kelp, effectively killing them, a new, non-lethal method removes secondary blades 20–30 cm from their bases, leaving the meristems and primary blades intact. At 5 sites studied, G. vittatum and P. virgata were found almost entirely on stipes and primary blades, and harvesting only distal parts of secondary blades limited losses to about 50% of C. flaccida biomass. To protect epiphytes, non-lethal harvesting is recommended and permanent non-harvest zones have been established in addition to limiting kelp yields and disallowing harvesting in Marine Protected Areas.  相似文献

  • 1 The abundance and microdistribution of thirteen gastropod and ten macrophyte species were monitored over 2 years in a 1 ha shallow eutrophic pond near Oxford, England. While areas of allochthonous leaf litter supported a depauperate and nondistinctive gastropod fauna, three different macrophyte habitats supported more diverse and distinctive assemblages of gastropods.
  • 2 Most gastropod species were clearly more abundant (number m-2pond bottom) on one macrophyte type than on other substrates. One exception, the limpet Acroloxus lacustris (Linn.) was abundantion both the water lily Nymphaea alba L. and on emergent macrophyles. Planorbis vortex (Linn.) was associated with graminoid emergent macropytes, especially Glyceria maxima (Hartm.) Holmberg, while Lymnaea peregra (Mull.) was associated with submersed macrophytes, perhaps more particularly with Elodea canadensis Michx.
  • 3 The association of, 4. lacuxtris with N. alba and emergent macrophytes is perhaps explained by constraints of the morphology of the limpet requiring a relatively broad smooth substrate for attachment and locomotion.
  • 4 The reason for the associations of P. vortex with G. maxima, and of L. peregra with E.canadensis are less obvious. Experiments giving each snail species a choice between the two macrophytes showed that P. vortex regularly exhibited a preference for G. maxima but failed to reveal substrate selection by L. peregra. Neither snail species affected the macrophyte choice of the other. The results suggested that L. peregra did not behave naturally in experiments.
  • 5 The preference for G, maxima by P. vortex was not affected by prior conditioning of the plant by the snail, but was affected by the emergent nature of G. maxima and by the presence/absence of a natural periphyton assemblage and associated detritus.
  • 6 The distribution of A. lacustris is probably determined by the physical structure of the substrate, while that of P. vortex is determined by the presence and quality of epiphytic periphyton-detritus. The observations and experiments with L. peregra reported here leave the reasons for its distribution in doubt.

With the expected increase in the spread of invasive species, examination of factors controlling distributions at multiple spatial scales and ecological modelling of their potential distributions are important analyses for informed decision-making. The scale-dependence of mechanisms influencing invasion by non-native species has been shown previously, indicating that studies of key factors affecting invasive species distributions at multiple spatial scales are critical for successful management. Freshwater systems are particularly vulnerable to invasive species, yet few studies have examined the environmental factors influencing distributions of invasive species at multiple spatial scales. We examined the effect of environmental variables on the predicted distribution of the invasive aquatic grass Glyceria maxima over continental, regional and local scales in Australia. We undertook an initial critical evaluation of which predictor variables were most appropriate to use at each scale, largely considering prior knowledge. On a continental scale, climatic, topographic and hydrological variables predicted well the potential distribution of G. maxima, identifying temperate regions as most susceptible to invasion. The regional analysis found that dense, woody, riparian vegetation has a strong negative impact on the occurrence of G. maxima, especially at intermediate elevations. The invasive grass was found less often on biotite granite and on fluvial geology. At a local scale, occurrence of G. maxima was related positively to soil phosphorus and nitrogen, and negatively related to soil organic carbon. The identification of key factors affecting invasive species distributions at multiple spatial scales will inform prevention schemes, assist targeted field sampling for the development of monitoring programs, and allow prioritization of control methods.  相似文献

Antibodies were raised against lectin purified from the sieve-tube exudate of Cucurbita maxima. Immunocytochemistry, using peroxidase-labelled antibodies and Protein A-colloidal gold, was employed to determine the location of the lectin within the tissues and cells of C. maxima and other cucurbit species. The anti-lectin antibodies bound to P-protein aggregates in sieve elements and companion cells, predominantly in the extrafascicular phloem of C. maxima. This may reflect the low rate of translocation in these cells. Under the electron microscope, the lectin was shown to be a component of P-protein filaments and was also found in association with the sieve-tube reticulum which lines the plasmalemma. The anti-lectin antibodies reacted with sieve-tube proteins from other species of the genus Cucurbita but showed only limited reaction with other genera. Six di‐, tri‐ and tetranucleotide microsatellite loci were developed for the silver‐ or gold‐lipped pearl oyster Pinctada maxima using a linker‐ligated, magnetic bead enrichment protocol. Based on a minimum of 134 Indonesian pearl oyster samples, number of alleles and observed heterozygosity at each locus ranged from six to 17 alleles and from 0.172 to 0.813 (mean = 0.448), respectively. Mean polymorphic information content for the six loci was 0.562. These loci should be very useful in DNA parentage analyses and population differentiation of P. maxima in Australia and Indonesia.  相似文献
