The roles of juvenile hormone III (JH III) on phase changes and pheromone production were examined in laboratory-reared gregarious desert locust, Schistocerca gregaria (Forskal). The hormone was applied to 5th instar nymphs and newly emerged adult locusts. Generally, the 5th instar nymphs exhibited a higher sensitivity to hormone treatments than the adults. Hormone applications inhibited pheromone production (as measured by the amounts of phenylacetonitrile released). In addition, JH III had a significant effect on the external colouration and absorbance ratios of the haemolymph pigments. It is concluded that the effects of exogenous JH III on gregarious locusts represent a shift towards the solitarious phase.  相似文献

The effects of crowding and isolation on body color and behavior were observed for the mid‐instar nymphs of the desert locust, Schistocerca gregaria. Some of the solitarious (isolation‐reared) nymphs that were crowded for 1 or 4 h during the third instar developed black patterns at the fourth instar, but most individuals remained unaffected. Black patterns appeared in all individuals that were crowded for 1 day or longer, but even after 4 days of crowding the black patterning for some individuals was not as intense as that for the gregarious (crowd‐reared) controls. Isolation of gregarious nymphs caused the black patterns to recede or disappear at the last (fifth) nymphal instar, but it was necessary to isolate the nymphs from the beginning of the first instar to obtain body coloration looking like solitarious nymphs in most individuals. Solitarious nymphs that were allowed to see gregarious nymphs developed different intensities of black patterns depending on the body size and number of nymphs shown. The behavioral phase shift from one phase to another was observed when the nymphs were crowded or isolated for 2 days or longer, as previously reported for the last nymphal instars of the same strain. Behavioral gregarization was induced for isolated nymphs that were allowed to see a group of nymphs through a transparent double wall. These results suggested that body‐color phase shift occurred more rapidly for mid‐instar nymphs than for late instar nymphs but the rate of behavioral phase shift was similar for the two instars.  相似文献

Extrusion technology was used to produce maize-starch 'contact bait' carriers, and vegetable-oil suspensions of Metarhizium flavoviride were incorporated into the baits after extrusion. Laboratory bioassays using the locust, Schistocerca gregaria, demonstrated mortality and reduction of 8 1 feeding after exposure to the baits, and gave an LC 50 of 1.5 10 spores l slope 0.63 for fifth instar nymphs. In laboratory trials, maize-starch contact baits showed improved efficacy relative to wheat-bran edible bait formulations of M. flavoviride. Field cage experiments, undertaken in Mali, using fourth and fifth instar nymphs of the grasshopper, Hieroglyphus dagenensis, demonstrated significant mortality with both fresh and weathered maize-starch contact bait formulations of M. flavoviride, relative to untreated controls.  相似文献

The soybean aphid, Aphis glycines Matsamura, is an eastern Asian soybean [Glycine max (L.) Merr.] pest which can reduce soybean yield. We determined the effects of pyriproxyfen, a juvenile hormone analogue, on development, mortality, longevity and fecundity of A. glycines under laboratory conditions. Distance Insect Growth Regulator, containing ∼11.2% pyriproxyfen, was applied at two concentrations, 50 and 150 mg/l, to first and fourth instar nymphs. When first or fourth instar A. glycines were treated with pyriproxyfen, some nymphs became supernumerary‐molted nymphs with 1–3 extra molts or were sterilized. Mortality of treated first instar nymphs was >68% greater than the control group and longevity was reduced by >40%. The higher concentration of pyriproxyfen reduced fecundity of first instar nymphs when they reached adulthood by ∼79%. Pyriproxyfen similarly affected fourth instar nymphs. Mortality of treated fourth instar nymphs was ≥15% greater than the control group and longevity was reduced by >24%. Both concentrations of pyriproxyfen lowered the fecundity of fourth instar nymphs by >27%. Pyriproxyfen also had other sublethal effects on fourth instar nymphs which became apparent when they molted to adulthood. In a few instances they developed wing pads and many produced dead, deformed or abnormal neonates that lacked appendages.  相似文献

To study developmental response of the brown-winged green bug, Plautia stali Scott, to food shortage we reared nymphs under restricted feeding conditions produced by shortening the feeding period after molt or withholding food from second to fifth instars. For second instars, molting rates were significantly reduced as the feeding period was shortened. Shortening the feeding periods for third to fifth instars also reduced molting rates, but less so; some nymphs were able to complete their developments even if no food was given to any of the instars. Compared with controls for which feeding was not restricted, nymphs that successfully reached the next instar had reduced postmolt body size as a result of restricted feeding for all instars except the fifth (2-day feeding for second instar, and 1-day feeding for third and fourth instars), whereas instar duration was unchanged or only slightly prolonged for all instars. These results suggest that only nymphs with nutritional accumulation over a specific threshold in each instar can progress to the next instar, and that, particularly for second to fourth instars, nymphs develop on schedule without prolonging instar durations to compensate for reduced growth under conditions of food shortage.  相似文献

Small brown planthopper, Laodelphax striatellus (Fallén) numbers usually drop sharply in the summer and revive quickly in the autumn. However, it is unclear whether and how the high temperature plays a role in this process. The effects of durations of heat exposure (33°C) on life‐history traits were examined here. Exposure of adults for 1 day during the oviposition stage led to a very low survival of nymphs. The average longevity of L. striatellus exposed for 1–31 days from oviposition was significantly longer than that of the control (27°C). Short‐term (1–5 days) heat exposure of the third instar nymphs did not significantly influence eclosion, but exposure of the fourth instar nymphs significantly increased eclosion. Lifespan from egg to adult was significantly lengthened when the third instar nymphs were exposed to heat for 2–15 days, or the fourth instar were exposed for 10 days. The preoviposition period was prolonged by heat exposure of the third or fourth instar nymphs. Short‐term heat exposure of less than 3 days of the third or fourth instar nymphs did not restrict fecundity, but when the exposure duration exceeded 5 days the total eggs per female and hatchability decreased. Exposure to high temperature increased the brachypter rate of adults. In summary, low survival and slowing development under heat exposure resulted in population decline in the summer, and the relatively high fecundity and brachypter rate led to quick revival in autumn. Temperature in the summer determines the rise and fall in numbers of L. striatellus.  相似文献

Feeding trials were conducted on three (young, mid-fill and mature) developmental stages of cowpea Vigna unguiculata ssp. unguiculata pods in the screenhouse using fourth instar nymphs and adults of Anoplocnemis curvipes (Fabricius), Riptortus dentipes (Fabricius), Mirperus jaculus (Thunberg), Clavigralla tomentosicollis St?l and C. shadabi Dolling. Anoplocnemis curvipes was observed to be the most damaging coreoid species causing a yield reduction of 26.4-51.7% followed by R. dentipes (24.4-29.4%), M. jaculus (21.9-26.9%), C. tomentosicollis (17.9-22.4%) and C. shadabi (15.9-20.4%). The fourth instar nymphs of each pod-sucking bug species caused a significantly higher cowpea yield reduction than their respective adults. Similarly, infestation on young pods compared to mid-fill and mature stages resulted in significantly higher yield reduction. The results suggest that infestation levels of two fourth instar nymphs of A. curvipes or three fourth instar nymphs of the other four pod-sucking bug species per young pod should be adequate for screening of cowpea varieties for resistance to the coreoid bugs.  相似文献

We examined the effects of the presence of plant allelochemicals in prey diet, prey availability and supplemental plant material on the growth of the generalist predator Podisus maculiventris (Hemiptera: Pentatomidae). We tested two different nymphal stages of this predator. Third to fourth instar nymphs and fifth instar nymphs were fed a diet of prey (Manduca sexta larvae, Lepidoptera: Sphingidae) without allelochemicals in their diet or prey fed maximal levels of allelochemicals (tomatine, rutin and chlorogenic acid) found in their host plant (Lycopersicon esculentum). The nymphs were fed prey ad libitum, once every three days, or once every five days. They were given either no supplemental plant material or a 2 cm slice of green bean pod (Phaseolus vulgaris). We also conducted another experiment with fifth instar nymphs using the same conditions, except that mean levels of allelochemicals found in the host plant were fed to prey instead of maximal levels and the prey were provided either once a day or once every five days. For all experiments, prey scarcity depressed developmental rate, weight gain and relative growth rate. Overall, there was no negative effect of allelochemicals in the diet of the prey on these variables when predators were supplied with an excess of prey, but allelochemicals in the prey diet negatively affected these predators when prey were scarce. The addition of plant material to the diet of third to fourth instar nymphs did not have any effect on developmental rate, final dry weight, or relative growth rate. However, for fifth instar nymphs, the addition of plant material negatively affected these variables. Thus, the addition of plant material to the diet of the nymphs did not alleviate the negative effects of prey scarcity or allelochemicals in prey diet.  相似文献

A new procedure for the measurement of adipokinetic hormone (AKH) concentrations in locust (Schistocerca gregaria) haemolymph is described: Haemolymph is extracted with chloroform/methanol/water and the aqueous layer is fractionated with reverse-phase cartridges and HPLC. The fractions corresponding to AKH-I (Lom-AKH-I) and AKH-II (Scg-AKH-II) are then measured in a competitive binding assay using specific antibodies and [3H]AKHs. The procedures could be applied to any peptides containing N-terminal pyroglutamate residues including all members of the adipokinetic/hyperglycaemic/red pigment concentrating hormone family. Results show that the concentrations of both AKH-I and AKH-II increase within 5 min of initiation of flight and are maintained at approx. 15-fold (AKH-I) and 6-fold (AKH-II) the resting levels over flights of at least 60 min. Poisoning of locusts with either the insecticide deltamethrin or with potassium chloride also caused release of hormones. Starvation for 6 h caused elevation of hormone levels in 5th instar nymphs, but starvation for 6 or 20 h had little effect on hormones in adults, despite an increase in haemolymph diacylglycerols at 20 h.  相似文献

The effects of injection of [His(7)]-corazonin and juvenile hormone (JH) III on the body color in L. migratoria were investigated using albino and normal (pigmented) nymphs. Most albino nymphs turned green in the fourth instar if injected with JH III during the last 2 days of the previous instar. When albino third instar nymphs injected with 10 pmol of [His(7)]-corazonin on different days were treated with 100 μg of JH III on day 3.5, they developed various body colors in the following nymphal instar: those injected with [His(7)]-corazonin during the first 2 days developed very dark green or black color, whereas some of those injected after this period turned green and their legs and ventral side of the body were variously pigmented, the coloration being similar to green solitary individuals often found in the field. Field-collected brown solitary nymphs injected with 1 nmol of [His(7)]-corazonin and kept individually, turned reddish without any black spots in the following nymphal instar when the ecdysis occurred within 1 day after injection. Injection of [His(7)]-corazonin 2 days before the following ecdysis induced black patterns on an orange background color, the coloration characteristic of gregarious forms. Similar injections into field-collected green solitary nymphs also induced black patterns but the rest of their body remained green. These results may indicate that the temporal changes in the hemolymph titers of [His(7)]-corazonin and JH play an important role in the control of body-color polymorphism in this locust.  相似文献

We tested the effects of Paranosema locustae spores in wheat bran formulation on the immature stages of Schistocerca gregaria and Oedaleus senegalensis under laboratory conditions. Younger instars were the most sensitive to the pathogen. While 100% infection was recorded in younger instar nymphs, older instars were less sensitive, with 16–27% of the inoculated nymphs remaining uninfected at the end of the experiment. Mortality of each instar increased with increased spore concentration. Immature survival time was significantly reduced by the pathogen and none of the nymphs inoculated as first, second, and third instar nymphs developed to adulthood (6–30% and 55–74% of nymphs inoculated as fourth and fifth instar, respectively). Sublethal effects such as delayed host growth, reduced host size, and abnormal wing and leg development (37% of emerging adults) were noted. Almost half the infected adults showed morphological abnormalities at emergence. Moreover, infection in S. gregaria and O. senegalensis by P. locustae did not affect female oviposition. However, 60% of S. gregaria and 52% of O. senegalensis progeny clearly showed infection by P. locustae with infection intensity of 1.08±0.27×101 and 1.19±0.32×102 spores/nymph, respectively. In view of the mortality rates, immature survival, host growth, and abnormal development in the P. locustae treatments, and the high prevalence of the pathogen in offspring from infected parents, it can be expected that the reduction in the impact of the two acridid species in the field will be considerable.  相似文献

It has been demonstrated that an apparently good, nutrient-rich plant can be relatively short of phenylalanine for fifth-instar nymphs of Schistocerca gregaria. Supplementary phenylalanine invariably increased the efficiency of conversion of ingested food to body substance, showing that this was the limiting nutrient. Relatively large quantities of phenylalanine are required for cuticle protein over a few days in the first half of the instar, which is when this amino acid is at a premium.  相似文献

Incorporation of cortisol, or various derivatives and analogues of cortisol, into the diet had no effect on the rate of growth of Tenebrio molitor. Injections of cortisol, or its derivatives, into fourth instar larvae of Schistocerca americana gregaria, had no effect on their rate of growth. The moult to fifth instar was normal. Administration in the diet, or injections of cortisol had no effect on the rate of growth of fourth and fifth instar larvae of Manduca sexta, or on the insect's uptake and utilization of food. The development of larvae to pupae and adults was unaffected. It is concluded, contrary to the findings of two previously published reports, that the vertebrate hormone cortisol does not affect the growth and development of insects.  相似文献

Changes in prothoracic gland morphology were correlated to developmental events and ecdysteroid titres (20-hydroxyecdysone equivalents) during the last-larval instar in Spodoptera littoralis. After ecdysis to the last-larval instar the haemolymph ecdysteroid titre remained at about 45 ng/ml, when the prothoracic glands appeared quiescent. The first signs of distinct gland activity, indicated by increased cell size and radial channel formation, were observed at about 12 h prior to the cessation of feeding (36 h after the last-larval moult), accompanied by a gradual increase in ecdysteroid titre to 110 ng/ml haemolymph, at the onset of metamorphosis. During this phase ecdysteroid titres remained at a constant level (140–210 ng/ml haemolymph) and prothoracic gland cellular activity was absent for a short period. The construction of pupation cells occurred when haemolymph ecdysteroids titres increased to 700 ng/ml. A rapid increase in ecdysteroids began on the fourth night (1600 ng/ml haemolymph) reaching a maximal level (4000 ng/ml haemolymph) at the beginning of the fourth day. In freshly moulted pupae a relatively high ecdysteroid titre (1100 ng/ml haemolymph) was still observed, although during a decrease to almost negligible levels. The increase in ecdysteroid level during the third and the fourth nights of the last-larval instar was correlated with the period when almost all the prothoracic gland cells showed signs of high activity. Neck-ligation experiments indicated the necessity of head factors for normal metamorphosis up to the second to third day of the instar. The possibility that the prothoracic glands are under prothoracicotropic hormone regulation at these times is discussed.  相似文献

Abstract:  An experimental study on the effect of isolation and crowding of the desert locust, Schistocerca gregaria (Forskål) on haemolymph pigment composition (as measured by absorbance ratios at 460 and 680 nm) was carried out at the International Centre of Insect Physiology and Ecology, Nairobi. The results showed that haemolymph absorbance ratios of gregarious and solitarious locusts were different (P < 0.05) and that there was no phase-specific sex differentiation with regard to this measure (P < 0.05). The haemolymph pigment composition changes during isolation and grouping showed contrasting trends. It changed rapidly in nymphs, but slowly and erratically in adults both resulting from isolation and grouping at hopper stage. Significant shifts in locust phase status were recorded in both situations in nymphs in F0 generation, and in the adults by the end of the F2 and F3 generations of solitary and gregarious conditions, respectively. Shifting of fledglings did not induce significant changes in their mean absorbance ratios in both situations, i.e. isolation and crowding within F0 generation. The ratio of length of posterior femur (F) to greatest width of the head capsule (C) changed either at a similar rate or more often slower than the haemolymph pigment ratio and both required more than a generation to transform fully. The study has shown that haemolymph pigment composition could be a suitable measure for monitoring phase changes at nymphal stages of the desert locust.  相似文献

ABSTRACT. Behavioural habituation† of Schistocerca gregaria Forskål fifth instar larvae to a feeding deterrent, nicotine hydrogen tartrate (NHT), is described. An attempt was made to differentiate between various factors possibly involved in the induction of this behavioural habituation. Sensory stimulation (through maxillary palp), cannulation of NHT into the crop, and its administration by gelatine capsules placed into the oesophagus each resulted in behavioural habituation, but in the latter two cases a sole induction via post-ingestional input is not completely proven. Injection of NHT into the haemolymph did not induce habituation. It is concluded that habituation via sensory stimulation is a central phenomenon.  相似文献

Two isolates of Metarhizium anisopliae var acridum were tested for their effects on the locust immune system and for comparison with the effects of challenge by injection with laminarin. Isolate IMI 330189 (referred to hereafter as Met 189) is highly pathogenic whether applied topically as conidia or injected as blastospores. However, isolate ARSEF 728 (referred to hereafter as Met 728) is pathogenic only when injected as blastospores, suggesting that the lack of pathogenicity of topically applied conidia from this isolate is due to a failure to penetrate the insect cuticle and gain access to the haemocoel. After topical application of conidia from Met 189, no activation of prophenoloxidase is detected, but injection of blastospores from Met 189 brings about a transient increase in phenoloxidase activity in the haemolymph in both adult locusts and 5th instar nymphs, although this does not prevent fungal-induced mortality. Co-injection of adipokinetic hormone-I (AKH-I) with blastospores prolongs the activation of prophenoloxidase in the haemolymph of adult locusts, and enhances it in nymphs. It is argued that the lack of activation of prophenoloxidase in nymphs shown previously (Mullen, L., Goldsworthy, G., 2003. Changes in lipophorins are related to the activation of phenoloxidase in the haemolymph of Locusta migratoria in response to injection of immunogens. Insect Biochemistry and Molecular Biology 33, 661-670), reflects differences in the sensitivity of the immune system between adults and nymphs rather than distinct qualitative differences, and this is confirmed in this study by the demonstration that doses of laminarin higher than those used previously (>or=100 microg) do activate the prophenoloxidase cascade in 5th instar nymphs. Nodules are formed in locusts of all ages in response to fungal infection or injection of laminarin, although there is wide variation in the number, size and distribution of nodules formed. During the examination of 5th instar nymphs for nodule formation, a previously unknown phenomenon was observed in which the salivary glands melanise in response to injections of blastospores or high doses of laminarin. In c. 85% of such nymphs, this reaction is so strong that the whole salivary gland is intensely black. Such a response is not observed in the salivary glands of mature adult locusts.  相似文献

Thin layer chromatography separation of 80% ethanol extracts of adult Elatobium abietinum revealed the presence of the polyhydric alcohol mannitol in aphids overwintering outdoors but not in aphids kept permanently indoors at 15°C. After 3 days at 15°C no traces of mannitol were left in overwintering aphids. Mean freezing temperatures of outdoor, unfed instar I nymphs were about 4°C lower than those of unfed instar I nymphs produced at 15°C. Mean freezing temperatures of overwintering adults were considerably higher than those of unfed instar I nymphs and showed no changes associated with time at 15°C following transference indoors. Similarly, mean freezing temperatures of Sitka spruce needles transferred to 15°C did not change. It was concluded that, although freezing was mainly avoided by supercooling, the presence of mannitol lowered the true freezing temperature of aphid haemolymph and, consequently, the actual freezing temperatures of nymphs produced under cold conditions. However, the considerable increase in freezing point temperatures caused by imbibition of plant sap masked these acclimatisation changes in feeding aphids.  相似文献

Brain (median or lateral regions) or suboesophageal ganglion (SOG) homogenates of Day 1 fifth instar larvae of Trichoplusia ni induced the appearance of haemolymph juvenile hormone esterase (JHE) when injected into Day 1, Day 2 or early Day 4 fifth instar ligated hosts. Brain and SOG homogenates of late fourth instars also induced JHE when injected into Day 1 hosts, whole late fifth instar and pupal tissue did not. The pattern of JHE induction by early fourth through Day 3 fifth instar brain and SOG homogenates correlated with natural haemolymph JHE activity occurring at these times. Implantation of late fourth and Day 1 fifth instar brains and/or SOG into similar age hosts similarly induced JHE activity while prothoracic and abdominal ganglia did not. The relative levels of induction following implantation were SOG<brain<brain+SOG. JHE activity which appears in the haemolymph following injection of brain homogenates appears to be largely due to a single enzyme which has an isoelectric point indistinguishable from that of the natural haemolymph enzyme. Evidence is presented which suggests that inhibitory as well as stimulatory brain factors are involved in JHE regulation.  相似文献

该文报道外源蜕皮激素(ED)和保幼激素类似物(JHA)对褐飞虱Nilaparvata lugens (Stal)翅型分化及体内酯酶活性、可溶性糖和游离氨基酸含量等的影响。褐飞虱在3、4龄若虫期经JHA处理后,所羽化出的成虫短翅型比例明显升高;而在5龄若虫期处理,所羽化出成虫的短翅型比例无明显变化。ED处理能增加3、4龄若虫体内酯酶活性、可溶性糖及游离氨基酸含量,而JHA处理则能有效地抑制ED的这些生物活性,这可能是JHA处理后褐飞虱成虫短翅型比例提高的生理生化基础。  相似文献
