Peritoneal macrophages (PM) of Wistar rats harvested after the intraperitoneal injection of paraffin oil were destroyed by repeated freezing-thawing. When injected intratracheally to control rats or to those after 4 daily exposured to TiO2 dust, these macrophage destruction products (MDP) caused a significant rise of both the alveolar macrophages (AM) and the neutrophilic leukocytes (NL) counts in the pulmonary washing-outs; the mean NL/AM ratio increased several times as compared to rats injected with normal saline intratracheally. Thus, the response to the inert dust particles plus the exogenous MDP became similar to the one observed after the cytotoxic (for instance silica) particles inhalation. Enhancing the NL contribution to the inhaled particles phagocytosis, the MDP led to a significant decrease of the mean "Dust load" of a single AM, although the total number of the engulfed particles increased. The predominant attraction of granulocytes and particularly of the NL as compared to the peritoneal macrophages was also found in the peritoneal exudates of rats injected with the MDP or silica suspension intraperitoneally, while the alveolar phagocytosis was not influenced. In vitro the MDP was shown to stimulate the NL migration and to facilitate the O2 consumption by PM. A possible role of the MDP as a multipotent controlling factor of phagocytosis response is briefly discussed.  相似文献

A study was made of the effect of the antilysosomal sera on various phases of phagocytosis Macrophages fluorochromized with acridine orange served as a model for the assessment of the effect of the antilysosomal sera on the phagocytosis: the fluorescent-microscopic method permitted to evaluate quantitatively the activity of the antilysosomal sera and to study the intracellular changes in the phagocytized antigen associated with its interaction with the lysosomes. The data obtained showed the antisera to the enzymes and the lysosome membranes to inhibit the phagocytosis both in the presence of a complement and without it. It was also demonstrated that the antilysosomal sera influenced the activity of the acid phosphatase and its distribution in the macrophages.  相似文献

CC chemokine receptor 7 (CCR7) is selectively expressed on mature dendritic cells (DC). The CCR7 ligands, CC chemokine ligand (CCL) 19 and CCL21, facilitate migration of mature DC from the peripheral tissues to regional lymph nodes. We previously demonstrated that CCR7 ligands induced rapid receptor-mediated endocytosis of dextran in mature DC. In the present study, we further examined the effects of CCR7 ligands on endocytosis of other kinds of antigen, mannosilated bovine serum albumin (Mann-BSA), Escherichia coli(E. coli), or ovalbumin-containing immune complex (OVA-IC), by mature DC. We found that CCR7 ligands enhanced the endocytosis of Mann BSA, E. coli, and OVA-IC in mature DC but not in immature DC. The endocytosis of BSA was not enhanced by CCR7 ligands. Furthermore, the phagocytosis of OVA-IC was significantly inhibited by anti-Fcgamma receptor III/II antibody. These results demonstrate that CCR7 ligands enhance only receptor-mediated endocytosis by mature DC. When rapidly phagocytosed E. coli were traced in CCL19-treated mature DC, most of the phagocytosed E. coli did not colocalize with the lysosomal marker: lysosome-associated membrane protein-1 (Lamp-1), whereas most of E. coli taken up relatively slowly by immature DC colocalized with Lamp-1. These results suggest that phagocytosis of antigens by immature and mature DC plays different functional roles.  相似文献

We studied the attachment of MRBC, 1.1 μm latex (foreign surface receptor), hemocyanin-DNP antiDNP Ab and sensitized SRBC (Fc receptor) to mouse peritoneal macrophages in culture, and the phagocytosis of 1.1 μm latex, AbAg complex, and sensitized SRBC by macrophages. Both types of receptors appeared over the whole cell surface. The particles attached mostly in the central area of the cell, but peripheral attachment was also observed. Internalization of 1.1 μm latex beads appeared to be mostly perinuclear, by sinking in of the cytoplasm with the attached particles, rather than a flow of membrane process over the beads. Also in the case of the AbAg complex attached to the Fc receptor internalization seemed mostly perinuclear and similar in mode to the latex ingestion. When attachment occurred in the periphery, active ruffling was observed. Sensitized SRBC internalization was seen mostly in the extreme periphery of the cells after the attached SRBC were moved from the perinuclear area where many of them first attached.  相似文献

Macrophages display a large variety of surface receptors that are critical for their normal cellular functions in host defense, including finding sites of infection (chemotaxis) and removing foreign particles (phagocytosis). However, inappropriate regulation of these processes can lead to human diseases. Many of these receptors utilize tyrosine phosphorylation cascades to initiate and terminate signals leading to cell migration and clearance of infection. Actin remodeling dominates these processes and many regulators have been identified. This review focuses on how tyrosine kinases and phosphatases regulate actin dynamics leading to macrophage chemotaxis and phagocytosis.  相似文献

Background

Bacteria are frequently cultured from sputum samples of severe asthma patients suggesting a defect in bacterial clearance from the airway. We measured the capacity of macrophages from patients with asthma to phagocytose bacteria.

Methods

Phagocytosis of fluorescently-labelled polystyrene beads, Haemophilus influenzae or Staphylococcus aureus by broncholaveolar lavage alveolar macrophages (AM) and by monocyte-derived macrophages (MDM) from non-asthmatics, mild-moderate and severe asthmatic patients was assessed using fluorimetry.

Results

There were no differences in phagocytosis of polystyrene beads by AMs or MDMs from any of the subject groups. There was reduced phagocytosis of Haemophilus influenzae and Staphylococcus aureus in MDMs from patients with severe asthma compared to non-severe asthma (p < 0.05 and p < 0.01, respectively) and healthy subjects (p < 0.01and p < 0.001, respectively). Phagocytosis of Haemophilus influenzae and Staphylococcus aureus by AM was also reduced in severe asthma compared to normal subjects (p < 0.05). Dexamethasone and formoterol did not suppress phagocytosis of bacteria by MDMs from any of the groups.

Conclusions

Persistence of bacteria in the lower airways may result partly from a reduced phagocytic capacity of macrophages for bacteria. This may contribute to increased exacerbations, airway colonization and persistence of inflammation.  相似文献

Elimination of apoptotic bodies is one of the important functions of macrophages. The aim of this work was to study the role of macrophage lectins in this process. Macrophage lectins were probed with neoglycoconjugates Glyc-PAA-fluo where carbohydrate is linked to fluorescein-labeled polyacrylamide (MW 30 kD). It was shown that neoglycoconjugates containing a Neu5Ac2-3Gal fragment bound to macrophages isolated from blood of healthy donors. Besides, carbohydrate chains containing the same fragment were revealed on apoptotic bodies. Phagocytosis of apoptotic bodies by macrophages was inhibited with sialooligosaccharide ligands of siglec-5 and MAbs to siglec-5. Thus, siglec-5 expressed on macrophages could participate in phagocytosis of apoptotic bodies. In addition, the role of siglecs in engulfment of apoptotic bodies by tumor-associated macrophages was studied. The phagocytic potency of macrophages isolated from blood of breast cancer patients was lower than engulfment ability of macrophages obtained from healthy donors and depended on tumor degree. Cyclodextrin glycosyltransferases (CGT-ase, 1.4-alpha-glucanotransferase, cyclizing, EC 2.4.1.19) produced by some thermophilic, alkalophilic and mesophilic bacterial strains, were isolated and characterized. It was shown that thermophilic and mesophilic CGT-ases represent a mixture of alpha-, beta- and gamma-cyclodextrins (CD), alpha-cyclodextrin being the predominant component. Alkalophilic enzymes produce only beta-CD and are able to produce CD not only from starch but also from maltose, melibiose, maltotriose, amylose and amylopectin. The optimal conditions for the catalytic activity of the enzymes were determined. It was found that calcium, magnesium and zinc ions have a beneficial effect on the specific activity of these enzymes. The amino acid composition of the enzymes was studied.  相似文献

The acid phosphatase (APh) was studied in the skin macrophage lysosomes of animals with delayed-type hypersensitivity (DTH) to the antigens of group A streptococci or tuberculoproteins. The histochemical procedure for the APh determination was employed in the skin sections without any preliminary tissue fixation. Intradermal injection of specific antigens in DTH proved to lead to the increase of the lysosomal membrane permeability in the skin macrophages. These data support the supposition that the lysosome enzymes of macrophages can act as factors causing tissue destruction in DTH.  相似文献

Macrophages stimulated by various substances exhibit altered morphology, metabolism, and enhanced phagocytosis. The present studies were done to show if peroxidative enzymes would affect macrophage spreading and phagocytosis. Resident peritoneal macrophages, collected from C57BI mice were exposed to various concentrations of peroxidases and compared with appropriate controls. Results indicated that 0.01 microM myeloperoxidase (MyPO), 0.09 microM horseradish peroxidase (HRP), 0.16 microM lactoperoxidase (LPO) and 70 microM microperoxidase (MPO) significantly enhanced macrophage spreading. It was also noted that peroxidases were able to stimulate phagocytosis by increasing the number of cells with internalized zymosan at least twofold. Stimulation of these functions suggests a possible role of endogenous peroxidases as natural cell activators.  相似文献

Role of H2-calponin in regulating macrophage motility and phagocytosis   总被引:1,自引:0,他引:1  
The actin cytoskeleton plays a major role in cell motility that is essential for the function of phagocytes. Calponin is an actin-associated regulatory protein. Here we report the finding of significant levels of the h2 isoform of calponin in peripheral blood cells of myeloid lineage. To study the functional significance, h2-calponin gene (Cnn2) interrupted mice were constructed. Germ line transmission of the Cnn2-flox-neo allele was obtained in chimeras from two independent clones of targeted embryonic stem cells. The insertion of the neo(R) cassette into intron 2 of the Cnn2 gene resulted in a significant knockdown of h2-calponin expression. Removing the frt-flanked neo(R) cassette by FLP1 recombinase rescued the knockdown effect. Cre recombinase-induced deletion of the loxP-flanked exon 2 eliminated the expression of h2-calponin protein. H2-calponin-free mice showed reduced numbers of peripheral blood neutrophils and monocytes. H2-calponin-free macrophages demonstrated a higher rate of proliferation and faster migration than that of h2-calponin-positive cells, consistent with a faster diapedesis of peripheral monocytes and neutrophils. H2-calponin-free macrophages showed reduced spreading in adhesion culture together with decreased tropomyosin in the actin cytoskeleton. The lack of h2-calponin also significantly increased macrophage phagocytotic activity, suggesting a novel mechanism to regulate phagocyte functions.  相似文献

Iron-exposed murine macrophages have a modified bactericidal activity as shown by previous observations. In order to assess the role of iron in macrophage activation, as measured by free radical production and by intracellular bacterial killing, murine peritoneal macrophages were cultivated in the presence of various sources of iron, human iron-saturated transferrin and ammonium ferric citrate, or iron chelators, Desferal, and human Apo-transferrin, and were infected with an enteropathogenic strain ofE. coli. The release of nitrite (NO2 ?), and the production of superoxide anion (O2 ?) and hydrogen peroxide (H2O2) by the phagocytes were measured and compared to the production by uninfected macrophages. The synergistic action with murine r.IFN-γ was also studied in the radical production reaction and for the bactericidal activity of macrophages. Our results show that in vitro phagocytosis ofE. coli induced elevated production of NO2 ? and H2O2 by macrophages, and that oxygen derivatives were released independently of the presence of added iron or chelator. Despite a phagocytosis-related enhancement of NO2 ? release, reactive nitrogen intermediates (RNI) are not directly involved in the bactericidal mechanism, as revealed by increased intracellular killing owing to RNI inhibitors. Moreover, bacterial killing may depend on oxygen derivatives, as suggested by the effect of the antioxidant sodium ascorbate leading to both a diminished H2O2 production and a decreased bactericidal activity of macrophages.  相似文献

Splenomegaly characterised by myeloid metaplasia was induced in BALB/c mice by the injection of antigens and immunostimulants. In difference from leukemoid reaction, this syndrome was transmissible both by the plasma and by the splenic cells. Small virus-like particles (30--50 nm) were revealed in the plasma, and RNA-containing viruses of type C--in the splenic cells.  相似文献
