Among the drimane compounds tested, the dialdehydes polygodial and warburganal were the most active as antifeedants against Colorado potato beetle larvae, Leptinotarsa decemlineata Say (Coleoptera: Chrysomelidae), in a dual-choice assay with potato, Solanum tuberosum L., leaf discs. Lactones were less effective. Direct observations showed that decreased feeding on leaf discs treated with polygodial and warburganal was accompanied by increased locomotry activity. Topical application of these two compounds on the insect's cuticle decreased food intake of untreated leaf discs, indicating that besides deterrent effects, toxic properties of these molecules influence feeding behaviour.  相似文献

Extracellular electrophysiological recordings were made from individual type-A trichoid sensilla on the antenna of the female sphinx moth Manduca sexta. A single annulus of the antenna bears about 1,100 of these sensilla, and each is innervated by two olfactory receptor cells. We tested the responses of these receptor cells to a panel of 102 volatile compounds, as well as three plant-derived odor mixtures, and could discern three different functional types of type-A trichoid sensilla. One subset of receptor cells exhibited an apparently narrow molecular receptive range, responding strongly to only one or two terpenoid odorants. The second subset was activated exclusively by aromatics and responded strongly to two to seven odorants. The third subset had a broad molecular receptive range and responded strongly to odorants belonging to several chemical classes. We also found receptor cells that did not respond to any of the odorants tested but were spontaneously active. Certain odorants elicited excitatory responses in some sensilla but inhibitory responses in others, and some receptor cells were strongly excited by certain odorants but inhibited by others. Impregnation of groups of receptor cells in type-A trichoid sensilla with rhodamine-dextran demonstrated that their axons project mainly to the large female glomeruli of the antennal lobe.  相似文献

1. Silica in the leaves of grasses can act as a defence against both vertebrate and invertebrate herbivores. The mechanisms by which silica affects herbivore performance are not well characterized. Here we expose an insect herbivore Spodoptera exempta to high-silica diets and test two mechanisms by which silica has been proposed to act as a defence. First, that silica reduces the digestibility of leaves and second, that silica causes wear to insect mandibles, both of which could potentially impact on herbivore performance. 2. Silica reduced the efficiency with which S. exempta converted ingested food to body mass and the amount of nitrogen absorbed from their food, leading to reduced insect growth rates. The measure of how efficiently herbivores utilize digested food (ECD) was unaffected by silica. 3. These effects occurred even with short-term exposure to silica-rich diets, but they also increased markedly with the duration of exposure and affected late instars more than early instar larvae. This appears to be due to the progressive impacts of silica with longer exposure times and suggests that herbivores cannot adapt to silica defences, nor do they develop a tolerance for silica with age. 4. Exposure to silica-rich diets caused increased mandible wear in S. exempta. This effect was extremely rapid, occurring within a single instar, further reducing feeding efficiency and growth rates. These effects on insect growth and feeding efficiency are nonreversible, persisting after the herbivore has switched diets. Up to a third of this residual impact can be explained by the degree of mandible wear caused by previous silica-rich diets. 5. The impacts of silica on S. exempta larvae were progressive with exposure time and could not be compensated for, even by switching to a different diet. Thus, herbivores cannot easily adapt to physical defences such as silica, suggesting this defence will have major implications for herbivore fitness.  相似文献

The lip structures termed labial probolae, characteristic for the subfamily Acrobelinae, and the anterior sense organs have been studied by electron microscopy. The labial probolae consist of an amorphous tissue with denser strengthenings. They do not contain any sensory structures or musculature. The anterior sense organs consists of: (1) six inner labial sensilla, each with one receptor ending in a pore on a papilla; (2) six outer labial sensilla, each with one receptor ending in the cuticle; (3) four cephalic sensilla, each with two receptors, one ending in a pore on a papilla and the other in the cuticle; (4) two amphids, each with 12 sensory neurons basally; (5) accessory sensilla ventro-laterally. The combination of several chemoreceptive anterior sensilla and the morphological diversity of the labial probolae within the subfamily may aid in the utilization of different soil microhabitats.  相似文献

The head of Austroperipatus aequabilis bears five types of sensilla. which were examined by electron microscopy. They differ from each other in position, shape of outer sensory elements and cuticular socket structures. Thus, we distinguish sensilla with sensory hairs, sensilla with sensory bulbs, cone-shaped sensilla. sensilla with sensory bristles, and sensilla of the lips. They are composed of up to 15 cells, which can he separated into four cell types. The most frequent cell type is the bipolar receptor cell that occurs in all sensilla. The apical surface of this primary receptor cell is characterized by one or two partly branched cilia with a basal 9 × 2 + 0 pattern of microtubules. A modified bipolar receptor cell was found in all sensilla bearing a sensory peg except for the sensilla equipped with sensory bristles. The apical dendrite extends to a long pale process which exclusively contains mitochondria and single microtubules. In all sensilla examined in this study at least one supporting cell occurs which is characterized by parallel microvilli. An additional function of this cell type as a part of the stimulus-conducting system is possible. In the sensillum with a sensory bulb two kinds of supporting cells occur. A unique cell type with an upside down position has regularly been found in all sensilla bearing a sensory peg. Apart from the sensilla they also occur within the labial epidermis. Electrophysiological studies on female An. gambiae s.s. mosquitoes revealed a receptor neuron within a subpopulation of the antennal grooved-peg sensilla sensitive to the odour of incubated sweat, but not responding to fresh sweat. This receptor neuron was sensitive to ammonia as well, a sweat-borne component which attracts female An. gambiae in a windtunnel bioassay. Neurons innervating a different subpopulation of grooved-peg sensilla did not show a response to incubated sweat. In the latter sensilla, however, one type of neuron responded to water or water containing solutions, while another receptor neuron was inhibited when stimulated with dry air, ether or ethanol. Neurons innervating sensilla trichodea, a more abundant antennal type of olfactory sensillum, did not respond to fresh or incubated sweat at the doses offered. However, receptor neurons within the sensilla trichodea responded with excitation to several sweat-borne components. A subpopulation of the sensilla trichodea was innervated by neurons sensitive to geranyl acetone. A second subpopulation housed receptor neurons sensitive to indole. 3-Methyl-1-butanol and 6-methyl-5-hepten-2-one evoked excitation of receptor neurons within both subpopulations of sensilla trichodea. Neurons were most sensitive to indole and geranyl acetone with a threshold of 0.01%. These findings are discussed in the context of host-seeking behaviour.  相似文献

利用扫描电镜和透射电镜对苹毛丽金龟Proagopertha lucidula (Faldermann)雌雄成虫触角嗅感器超微结构特征进行研究。结果表明:嗅感器集中于触角鳃片上,分布在触角鳃片表皮内陷形成的凹腔里,有5种感器,以板状感器为主,以外还有锥状感器、腔状感器、腔锥状感器和毛状感器。板状感器根据盘体形状的不同可分为4种类型,锥状感器根据锥体形状的差异可分为2种类型,腔锥状感器根据形状分为2种类型。嗅感器表皮有微孔和孔道微管。嗅感器内神经元的数目并不一致,1~2个不等。雄虫鳃片嗅感器总数显著多于雌性,是雌虫嗅感器总数1.8倍。其中雄性锥状感器的数目是雌性2倍,雌虫腔锥状感器数量是雄虫的4倍,雄虫板状感器数量是雌虫的2倍,雌雄腔状感器的数目无显著性差异。  相似文献

The nipa palm hispid beetle, Octodonta nipae (Maulik) has been killing palm trees since its introduction into Hainan province, China, from Malaysia in 2001. It continues to spread within Hainan province, northeast to Fujian province, and northwest to Yunnan province within China. Knowledge on signals involved in mate location and recognition could help develop effective integrated pest management programs. In the present study, we first experimentally proved that antennae were essential in success of O. nipae mating. We then excised various segments/flagellomeres of adult male and female antennae and observed their mating behavior. Results revealed that the 5th to 9th flagellomeres, especially those of males, were important for the mating success. Finally, in an attempt to elucidate the types of antennal sensilla accountable for the mating success, morphology of O. nipae antennae was studied in detail with scanning electron microscopy. Six types of sensilla were distinguished: aporous sensilla trichodea (T1), multiporous sensilla trichodea (T2), aporous sensilla chaetica (Ch1), uniporous sensilla chaetica (Ch2), multiporous sensilla basiconica (B), and Böhm sensilla (Bm). Aporous sensilla trichodea is the most abundant; multiporous sensilla trichodea and sensilla basiconica are considered as olfactory receptors, and uniporous sensilla chaetica as gustatory receptor. Importance of flagellomeres 5–9 in mating success seemed to correspond to the abundance of sensilla on these segments.  相似文献

Warburganal, a unique dialdehyde sesquiterpene isolated from East African Warburgia plants, showed a strong antifungal activity. However, this growth inhibition in Saccharomyces cerevisiae was reversed with L-cysteine. In addition, warburganal inhibited the alcoholic fermentation of S. cerevisiae while L-cysteine reversed this inhibition. When alcohol dehydrogenase, a sulfhydryl enzyme, was incubated with warburganal, the enzyme activity decreased with time. The decrease was more rapid at alkaline pH. L-Cysteine prevented this enzyme inhibition by warburganal but could not restore the enzyme activity lost already due to warburganal. Warburganal lost its characteristic ultraviolet absorption spectrum in the presence of L-cysteine. The change in absorbance was favored at alkaline pH, indicating Michael reaction type addition of L-cysteine to warburganal. Based on these observations, a variety of physiological activities due to warburganal appear to result from its irreversible reactivity with sulfhydryl groups.  相似文献

Ultrastructural examination of grooved-peg (GP) sensilla on the antenna of fifth instar Triatoma infestans nymphs by scanning electron microscopy and transmission electron microscopy reveal that they are 8–18 μm long with a diameter of about 2–2.8 μm at the non-articulated base. Some pegs have a terminal pore. These double-walled wall-pore (dw-wp) sensilla have an outer cuticular wall with 13–18 longitudinal grooves at the distal part of the peg. Groove channels are present at the bottom of the grooves from which radial spoke channels lead into the inner sensillum-lymph cavity. A dendrite sheath connects the tip of the thecogen cell to the inner cuticular wall thus forming separated outer and inner sensillum-lymph cavities. Four or five bipolar receptor cells are ensheathed successively within the GP sensilla by the thecogen cell, trichogen and tormogen cells. The inner dendritic segments of each sensory cell give rise at the ciliary constriction to an unbranched outer dendritic segment which can reach the tip of the sensillum.Electrophysiological recordings from the GP sensilla indicate that they house NH3, short-chain carboxylic acid and short-chain aliphatic amine receptor cells and can be divided into three functional sub-types (GP 1–3). All GP sensilla carry a receptor cell excited by aliphatic amines, such as isobutylamine, a compound associated with vertebrate odour. GP type 1 and 2 sensilla house, in addition, an NH3-excited cell whereas the type 2 sensilla also contains a short-chain carboxylic acid receptor. No cell particularly sensitive to either NH3 or carboxylic acids was found in the grooved-peg type 3 sensilla. GP types 1, 2 and 3 represent ca. 36, 10 and 43% of the GP sensilla, respectively, whereas the remaining 11% contain receptor cells that manifest normal spontaneous activity but do not respond to any of the afore mentioned stimuli.  相似文献

The allocation, external morphology, and ultrastructure of various types of sensilla on the antennae have been studied in the larvae of the Colorado potato beetle (Leptinotarsa decemlineata Say) using scanning and transmission electron microscopy. It has been found that the larval antenna bears 11 sensilla of four morphological types: trichoid (five), basiconic (four), styloconic (one), and antennal cone (one). It has been shown that the sensilla are innervated by 2?C7 receptor cells: trichoid, by 2?C4 receptor cells; basiconic, by 4?C7 receptor cells; and styloconic, by 6 receptor cells. Judging by the specifics of their ultrastructural organization, the function of these sensilla is gustatory. The antennal cone is innervated by two receptor cells, and its function is olfactory.  相似文献

Single-cell electrophysiological recordings were obtained from olfactory receptor neurons housed in sensilla trichodea along the adult antennae arising from transplantation of the antennal imaginal discs between larval male Helicoverpa zea and Heliothis virescens. The olfactory receptor neurons from the majority of type C sensilla sampled on transplanted antennae displayed response characteristics consistent with those of the species that donated the antennae. However, some of the sensilla type C sampled in either transplant type contained olfactory receptor neurons that responded in a manner typical of the recipient species or other neurons that have not previously been found in the type C sensilla of either species. The single-cell data help to explain behavioral results showing that some transplant males do fly upwind to both species' pheromone blends, an outcome not expected based on known antennal sensory phenotypes. Our results suggest that host tissue can influence antennal olfactory receptor neuron development, and further that because of a common phylogenetic ancestry the donor tissue has the genetic capability to produce a variety of sensillar and receptor types.  相似文献

The third antennal segment (funiculus) and the maxillary palp are the main and accessory olfactory sense organs of Drosophila melanogaster. Cryofixed antennae and palps revealed a superior preservation of the sensory dendrites and other cellular details as compared to conventional chemical fixation. Extensive cross-section series through funiculus and palp were studied in order to obtain as complete an evaluation as possible of the sensillar complement on these appendages. About 75% of all sensilla on the male and female funiculus were individually studied and their position on the antennal surface mapped. Dimensions of the cuticular apparatus of the various types of sensilla are provided as well as the number of innervating receptor neurons. Particular attention has been paid to the cuticular pores important for stimulus transport and to the sensory dendrites. On the funiculus surface, all sensilla have multiple wall pores: sensilla (s.) trichodea and s. basiconica are single-walled, s. coeloconica are double-walled. The distribution of s. trichodea and s. basiconica follows opposing gradients along a diagonal axis parallel to the axis of the arista from proximo-medial to disto-lateral. In this disto-lateral direction the density of s. trichodea increases while that of the s. basiconica decreases. S. trichodea occur in three subtypes with one, two or three receptor neurons. Basiconic sensilla can be subdivided into three subtypes of large s. basiconica (with two or four receptor neurons), three subtypes of thin s. basiconica (with mostly two, rarely four neurons), and one subtype of small s. basiconica with two receptor neurons. Large s. basiconica occur only in the most proximal region (the ‘LB-zone’); thin s. basiconica are most common in a belt that borders the LB-zone distally, while small s. basiconica are most numerous even further distally along the mentioned diagonal axis in between the s. trichodea. S. intermedia are single-walled, multiporous sensilla which combine features of s. trichodea and s. basiconica; they are found in two subtypes with two or three receptor neurons, in the same region where s. trichodea abound. The s. coeloconica are irregularly distributed over the funicular surface, and occur in two subtypes with two or three receptor neurons. Sexual dimorphism on the antenna is moderate, the female funiculus is a bit longer and carries a larger number of small s. basiconica and large s. basiconica of the LB-I subtype; the male funiculus, however, has more s. trichodea than the female. On the maxillary palp, besides mechanoreceptive s. chaetica, there are only s. basiconica with two receptor neurons. According to the fine structure of their sensory dendrites, three subtypes can be discriminated which are distributed in a random pattern. The functional significance of the described structures and distribution awaits future immunocytochemical and electrophysiological experiments.  相似文献

Insect olfactory receptor neurons are compartmentalized in sensilla. In a sensillum, typically two receptor neurons are in close contact and can influence each other through electrical interaction during stimulation. This interaction is passive, non-synaptic and a consequence of the electrical structure of the sensillum. It is analysed in a sensillum model and its effects on the neuron receptor potentials are investigated. The neurons in a sensillum can be both sensitive to a given odorant compound with the same sensory threshold or with different thresholds, or only one neuron be sensitive to the odorant. These three types of sensilla are compared with respect to maximum amplitude, threshold and dynamic range of the potentials. It is found that gathering neurons in the same sensillum is disadvantageous if they are identical, but can be advantageous if their thresholds differ. Application of these results to actual recordings from pheromone and food-odour olfactory sensilla is discussed.  相似文献

Summary The digitiform sensilla on the distal segment of the maxillar palps ofAgabus bipustulatus (L.) andHydrobius fuscipes (L.) were investigated by electron microscopic methods. Each sensillum is innervated by a single bipolar sensory cell. The sensilla ofHydrobius are associated with three enveloping cells, which enclose an inner and outer receptor lymph cavity. A single enveloping cell only is found in the completely differentiated sensilla ofAgabus. These sensilla do not form an outer lymph cavity. The area beneath the hair base is filled by the distal process of the enveloping cell and by extensions of epidermal cells. Only one extra-cellular space exists, which seems to be homologous to an inner receptor lymph cavity.The outer dendritic segment surrounded by a dendritic sheath runs to the tip of the hair shaft. In the hair shaft the outer dendritic segment divides into several branches. The poreless hair shaft does not rise over the surface of the cuticle, but it is positioned in a narrow shallow groove. Special socket structures or a tubular body do not exist. The digiti-form sensilla possess neither the typical feature of mechanosensitive, nor gustatory or olfactory sensilla. The functional significance of the structural divergences in the sensilla of both species and the presumed function of the sensilla are discussed referring to hygro- and thermo-receptors.

Male Utetheisa ornatrixhave a pair of eversible glandular brushes (coremata) which are displayed during precopulatory interactions with the female. Earlier studies have shown that a pheromone associated with the coremata, hydroxydanaidal (HD), is derived by the males from pyrrolizidine alkaloids (PAs) that they sequester as larvae from their foodplants (Crotalariaspp.) The PAs impart a distastefulness upon Utetheisathat protects both larvae and adults against predation. The receptor neurons specialized for detection of HD are housed in sensilla whose morphological features, as revealed by scanning electron microscopy, classify them as sensilla basiconica. The sensitivity and dynamic range of these receptor neurons were largely unaffected by whether the females were raised on an alkaloid-free diet or on a diet supplemented with Crotalariaseeds. Acetylation of the hydroxyl group of HD substantially reduced the activity of the molecule. None of the antennal sensilla examined contained receptor neurons sensitive to a PA (monocrotaline) or its N-oxide.  相似文献

ABSTRACT. Female Spodoptera exempta (Walker) (Lepidoptera: Noctuidae) moths require access to water to achieve hydration and maturation of their oocytes if they are to achieve their potential fecundity. For moths provided with water, the main factor limiting fecundity is moth weight. There is some evidence that sucrose in the adult diet can increase fecundity, particularly in lighter moths from a suboptimal larval feeding regime. Moths fed sucrose live longer, but complete oviposition at about the same age as moths provided only with water. Dietary protein and amino acids have no effect on fecundity or longevity. Although- multiple matings are frequent in the laboratory, female S.exempta only need to mate once to complete oviposition. Differences are apparent in the relative contribution of larval and adult feeding to reproduction in Noctuidae. Some species, like S.exempta , require only water to achieve their reproductive potential, while others (e.g. Heliothis spp.) are dependent on sugars in the adult diet. Female S.exempta denied access to water or food until night 3 after eclosion and then provided with water or sucrose, commence oviposition on night 4 and have fecundities comparable with those moths fed from emergence.  相似文献

The aim of this work was to investigate the olfactory system of the walking stick insect, Carausius morosus. Morphological, ultrastructural and immunocytochemical studies of adult female antennae were conducted by scanning and transmission electron microscopy. Extensive cross-section series were made through the last antennal segment to define the cuticular apparatus, wall pore distribution and the number of innervating receptor neurons of each sensillum type. Single-walled wall pore sensilla occur in three subtypes: (i) with 27 or 28 branched receptor neurons, (ii) with two branched neurons and (iii) with one or two unbranched neurons, respectively. Double-walled wall pore sensilla were found in two subtypes with spoke channels, one with four unbranched neurons, the other with two unbranched neurons. One terminal pore sensillum was found, showing two cavities within the hair and being innervated by six sensory cells. Immunocytochemical experiments were performed to show the localization of a 19 kDa soluble protein found in the chemosensory organs of C. morosus. This protein shows an amino acid sequence homologous to the family of chemosensory proteins (CSP). The polyclonal antibody raised against the purified protein (CSP-cmA) showed, for the first time in CSPs, a strong labeling in olfactory sensilla, specifically in the sensillum lymph surrounding the dendritic branches of SW-WP sensilla and in the uninnervated lumen between the two concentric walls of DW-WP type 1 sensilla.  相似文献

Summary Coeloconic sensilla on the antenna of Locusta migratoria were investigated electrophysiologically, labeled and then examined under the scanning or transmission electron microscope. They can be categorized into two main morphological types: 1) sensilla with wall pores and two concentric cuticular walls (wp-dw sensilla), and 2) sensilla lacking wall pores (np-sensilla).Hygroreceptors were observed only in np-sensilla. Olfactory receptors were observed in wp-dw sensilla, but not in np-sensilla. Cold-sensitive units were found in both types. Wp-dw sensilla with a cold-sensitive unit also manifest a fatty acid olfactory receptor. This cold-sensitive unit appears to be less sensitive to drops in temperature than the cold receptor combined with hygroreceptors in np-sensilla. Recordings from both types revealed up to three or sometimes four units that could be distinguished on the basis of impulse amplitude.The number of np-sensilla on each antennal segment ranges from 0 to 6. These sensilla are pegs 4–5 m long with a terminal swelling. The dendritic outer segments of two sensory cells fit tightly into these pegs. The dendrite of the third sensory cell ends at its base. All three are up to 1 m thick, unbranched, and densely filled with microtubules. The cuticular wall and socket of the peg are characterized by (1) an electron-dense inner layer surrounding the dendrites and continuous with electron dense clusters at the base of the peg, and (2) clefts in the cuticle at the insertion of the peg.The present observations favor the hypothesis that insect sensilla combining the triad of moist air, dry air and cold receptors generally lack pores.
