ABSTRACT. Oxygen consumption (O2) in six species of adult tiger beetles Cicindela spp. (Coleoptera: Cicindelidae) was correlated with body mass and temperature during rest. In beetles forced to run and/or right themselves continuously for 5–10 min at 25°C, O2 was approximately 7–12 times as high as in resting individuals; the difference increased with increasing mass. Resting and active VO2 were similar to previous results for other beetles, although the slope of log O2 on log mass was lower. Detailed analysis suggests the existence of taxonomic and ecological correlates of resting metabolism. The possible ecological implications and adaptive advantages of these results for adult tiger beetles are discussed.  相似文献

Process pasteurization values for reference temperature 70°C (P70) were calculated from the temperature profiles of 250 g luncheon meat chubs cooked under experimental conditions. A simple equation relating Process P70-value and the time and temperature of cooking was derived. With minimal cooking (P70= 40) the surviving microflora (103/g) was dominated by species of Lactobacillus, Brochothrix and Micrococcus. These organisms were destroyed by more intensive cooking (P70= 105), leaving a flora (102/g) composed of Bacillus and Micrococcus species. The spoilage that developed after 14 d storage at 25°C reflected the severity of the heat treatment received by each chub: with P70 between 40 and 90, a Streptococcus spoilage sequence occurred; with P70 between 105 and 120, a Bacillus/Streptococcus spoilage sequence occurred; with P70 of 135 and above, a Bacillus spoilage sequence occurred. Cooking to a P70= 75 was adequate to reduce the surviving microflora to the 102/g level associated with current good manufacturing practice.  相似文献

Endurance and swimming speed were measured in mackerel, herring and saithe when they were induced by the optomotor response to swim at prolonged speeds along a 28-m circular track through still water in a 10-m diameter gantry tank. The maximum sustained swimming speed ( U ms was measured as body lengths per second ( b.l.s −1) for each species and for saithe of different size groups. Herring with U ms of 4.06 b.l.s −1 (25.3 cm, 13.5°C) were the fastest, mackerel U ms was 3.5 b.l.s 1 (33 cm, 11.7°C) and saithe (14.4°C) showed a size effect where U ms at 25 cm was 3.5 b.l.s 1 and at 50 cm 2.2 b.l.s 1. When swimming at speeds higher that U ms, all three species showed reduced endurance as speed increased. How the curved track reduces the swimming speed is discussed.  相似文献

1. The influence of leaf thickness on internal conductance for CO2 transfer from substomatal cavity to chloroplast stroma ( g i) and carbon isotope ratio (δ13C) of leaf dry matter was investigated for some evergreen tree species from Japanese temperate forests. g i was estimated based on the combined measurements of gas exchange and concurrent carbon isotope discrimination.
2. Leaves with thicker mesophyll tended to have larger leaf dry mass per area (LMA), larger surface area of mesophyll cells exposed to intercellular air spaces per unit leaf area ( S mes) and smaller volume ratio of intercellular spaces to the whole mesophyll (mesophyll porosity).
3. g i of these leaves was correlated positively to S mes but negatively to mesophyll porosity. The variation in g i among these species would be therefore primarily determined by variation of the conductance in liquid phase rather than that in gas phase.
4. δ13C was positively correlated to mesophyll thickness and leaf nitrogen content on an area basis. However, g i values did not correlate to δ13C. These results suggest that difference in δ13C among the species was not caused by the variation in g i, but mainly by the difference in long-term photosynthetic capacity.
5. Comparison of our results with those of previous studies showed that the correlation between leaf thickness and g i differed depending on leaf functional types (evergreen, deciduous or annual). Differences in leaf properties among these functional types were discussed.  相似文献

To develop further the methods for estimation of NOx absorption by plants supplied with 15N-labelled fertilizer, we proposed a new calculation method, total N fixed method (TNF), and compared with the 15N dilution method and the classical mass balance method (MB).
Hydroponically grown soybean plants were supplied with 15N-labelled nitrate and exposed to 200–250 nl l−1 NO2 for 7 d. The proportions of the N derived from NO2 to total N in exposed plants were estimated by the three methods.
The reported rates of NO2 absorption by several plant species, estimated by the 15N dilution method, were recalculated using the TNF method. The results of the two methods were compared and showed that: (1) The 15N dilution method overestimated the content of NO2-N in exposed plants compared with the MB method whilst the TNF method produced estimations of NO2-N closer to those by the MB method when the plants were supplied with 5 m M nitrate. (2) The differences in estimations between the MB method and either the 15N dilution method or the TNF method increased with decreasing supply of 15N-labelled nitrate to roots.  相似文献

The immune response of chickens to goat erythrocytes has been examined. The H line selected for high competence of graft-versus-host reaction (GVHR) showed higher immune responses than the L line selected for low GVHR competence. It appeared also that immune responses were controlled by the B blood group locus, which is the major histocompatibility locus in chickens. The relative immune responsiveness of B genotypes were B 11/11> B 9/ B 11> B 9/B9.
Treatment of antiserum with 2-mercaptoethanol (2-ME) proved that the difference in immune responses between lines was due mainly to the 2-ME resistant antibody and that the difference between the B genotypes was due to the 2-ME sensitive antibody.  相似文献

Basal metabolic rate (BMR), commonly used as a measure of the cost of living, is highly variable among species, and sources of the variation are subject to an enduring debate among comparative biologists. One of the hypotheses links the variation in BMR with diversity of food habits and life-history traits. We test this hypothesis by asking how BMR of a particular species, the bank vole Myodes (= Clethrionomys ) glareolus , would change under selection for high growth rate (measured as a postweaning body mass change; MDPW ) and the ability to cope with a low-quality herbivorous diet (measured as body mass change during a four-day test; MDLQD ). We show that both of the traits are heritable in the narrow sense ( MDPW : h 2= 0.30; MDLQD : h 2= 0.19), and are genetically correlated with mass-independent BMR (additive genetic correlation, rA = 0.28 for MDPW and 0.37 for MDLQD ). Thus, both of the traits could change in response to a selection, and the selection would also result in a correlated evolution of the level of metabolism. The results are consistent with the hypothesis that a part of the interspecific variation in BMR evolved in response to selection for life-history and ecological traits such as food habits.  相似文献

Abstract: Techniques are described for measuring the isotope distribution in dissolved nitrate and N2 using membrane inlet mass spectrometry, which allows several gases to be measured in a water sample without the need for any separation steps. The isotope distribution in dissolved nitrate was measured using denitrifying Pseudomonas nautica to reduce the nitrate to N2 which was then measured by mass spectrometry. Pseudomonas nautica NCIMB 1967 was easily grown in nitrate-limited continuous culture minimising intra- or extracellular nitrate or nitrite pools, and the bioassay was tolerant of a range of salinities. The precision of the bioassay when measuring samples with high 15NO3 contents (0.5 μmol) was 0.05 atom%; with 0.1 μmol 15NO3, the precision was around 0.2 atom%. Differences in labelling of N2 in preserved samples obtained from 15NO3 incubations of water-covered sediment cores were measured on parallel samples with membrane inlet MS and GC-MS. The membrane inlet technique was accurate but the precision on ratio measurements was lower than by GC-MS.  相似文献

Water-use efficiency and stable isotope composition were studied in three tropical tree species. Seedlings of Tectona grandis , Swietenia macrophylla and Platymiscium pinnatum were grown at either high or low water supply, and with or without added fertilizer. These three species previously exhibited low, intermediate and high whole-plant water-use efficiency ( TE ) when grown at high water supply in unfertilized soil. Responses of TE to water and nutrient availability varied among species. The TE was calculated as experiment-long dry matter production divided by cumulative water use. Species-specific offsets were observed in relationships between TE and whole-plant 13C discrimination (Δ13Cp). These offsets could be attributed to a breakdown in the relationship between Δ13Cp and the ratio of intercellular to ambient CO2 partial pressures ( c i/ c a) in P. pinnatum , and to variation among species in the leaf-to-air vapour pressure difference ( v ). Thus, a plot of v · TE against c i/ c a showed a general relationship among species. Relationships between δ 18O of stem dry matter and stomatal conductance ranged from strongly negative for S. macrophylla to no relationship for T. grandis . Results suggest inter-specific variation among tropical tree species in relationships between stable isotope ratios ( δ 13C and δ 18O) and the gas exchange processes thought to affect them.  相似文献

Ten species of ungulate were filmed, galloping in their natural habitat. They ranged in size from Thomson's gazelle (about 20 kg) to giraffe (about 1000 kg). They were pursued to make them run as fast as possible. The films have been analysed to determine speed, stride frequency, stride and step lengths, and duty factors. The dependence of these quantities on body size is discussed.  

Summary:


Fast locomotion of zebra, giraffe, warthog and seven species of Bovidae has been studied. The animals were filmed from a pursuing vehicle while galloping in their natural habitat.
Stride frequency was more closely correlated with limb length (represented by hip height) than with body mass. Mean stride frequency was proportional to (hip height)-0·51 and maximum stride frequency to (hip height) -0·63.
Maximum speed was between 10 and 14 m s -1 for all species except buffalo (7 m s -1). It was not significantly correlated with body mass.
Since the small species ran at least as fast as the large ones they attained higher Froude numbers. Relative stride length was approximately 1·8 (Froude number)0·39 for all species, irrespective of size. Relative step length was approximately 0·65 (Froude number)0·2, both for the fore feet and for the hind ones. The vertical forces exerted by the feet are proportional to (body weight)×(Froude number)0·2 so the forces at maximum speed are larger multiples of body weight for small species than for large ones.  相似文献

Aflatoxin content and number of fungi in poultry feedstuffs from Indonesia   总被引:1,自引:1,他引:0  
The content of aflatoxin and associated fungi was determined in 56 samples, including 34 of corn, 10 of soybean meal, nine of rice bran and three of broken rice, collected from different poultry farms and poultry feedmills situated around Jakarta-Bogor, Indonesia.
Ninety-one per cent of the corn samples contained aflatoxins and the total concentration ranged from 22 to 6171 μ g/kg. With rice bran, 100% of the samples were positive for aflatoxin B1, ranging from 36 to 71 μ g/kg. No aflatoxin was detected in samples of soybean meal or broken rice. All the samples were contaminated by several fungi (8 times 103–5 times 106cfu/g) and further identification was limited to Aspergillus flavus and A. parasiticus. The dominant species was A. flavus (2 times 103–4 times 106cfu/g in corn samples, 1·0 times 103–1·0 times 105cfu/g in soybean meal, 2 times 104–4·4 times 105cfu/g in rice bran and 2 times 104–6 times 104cfu/g in broken rice). Some of the corn samples also contained A. parasiticus (2 times 103–9·5 times 104cfu/g).  相似文献

The generally rhizotoxic ion Al3+ often enhances root growth at low concentrations. The hypothesis that Al3+ enhances growth by relieving H+ toxicity was tested with wheat seedlings ( Triticum aestivum L.). Growth enhancement by Al3+ only occurred under acidic conditions that reduced root elongation. Al3+ increased cell membrane electrical polarity and stimulated H+ extrusion. Previous investigations have shown that Al3+ decreases solute leakage at low pH and that the alleviation of H+ toxicity by cations appears to be a general phenomenon with effectiveness dependent upon charge (C3+>C2+>Cl+). Alleviation of one cation toxicity by another toxic cation appears to be reciprocal so that Al3+ toxicity is relieved by H+. It has been argued previously that this latter phenomenon accounts for the apparent toxicity of ALOH2+ and Al(OH)+2. Reduction of cell-surface electrical potential by the ameliorative cation may reduce the cell-surface activity of the toxic cation.  相似文献

Rates of oxygen consumption were measured in the geothermal, hot spring fish, Oreochromis alcalicus grahami by stopped flow respirometry. At 37° C, routine oxygen consumption followed the allometric relationship: V o2=0.738 M 0.75, where V o2 is ml O2 h −1 and M is body mass (g). This represents a routine metabolic rate for a 10 g fish at 37° C of 0.415 ml O2 g−1 h −1 (16.4 μmol O2 g −1 h −1). Acutely increasing the temperature from 37 to 42° C significantly elevated the rate of O2 consumption from 0.739 to 0.970 ml O2 g −1 h −1 ( Q 10=l.72). In the field, O. a. grahami was observed to be 'gulping' air from the surface of the water especially in hot springs that exceeded 40° C. O. a. grahami may utilize aerial respiration when O2 requirements are high.  相似文献

Determining body composition in gray seals ( Halichoerus grypus ) is important when studying their physiology and life history. In this study we investigated the predictability of total body fat (TBF) and protein (TBP) in postweaned gray seal pups from morphometric measurements, blubber thickness using ultrasonographs and bioelectrical impedance analysis (BIA). In postweaned pups, TBF (kg) could be estimated from girth measurements and sex ( n = 45, r 2= 0.878) using hydrogen isotope dilution methods as the reference. However, TBP could not be reliably estimated from morphometric data. TBF (kg) in yearlings was best predicted from mass ( n = 6, r 2= 0.776) and TBP (kg) from mass/length ( r 2= 0.949). Dorsal blubber thickness using B-mode ultrasound was also a significant predictor of TBF (kg) in postweaned pups ( r 2= 0.725) but BIA was not. Marked pups were recaptured during their first few months of life ( n = 48) and body composition changes investigated. Animals lost mass and TBF after leaving the breeding beach, largely during the first 5–6 mo of life. Postweaned pups were ∼40% TBF and ∼13% TBP whereas yearlings were ∼12% TBF and ∼20% TBP. Pups that survived beyond 6 mo of age then regained mass as protein. Morphometric measurements are a useful field indication of body condition when isotope dilution is impractical.  相似文献

Abstract: 4-Hydroxy-3-methoxyphenylglycol (HMPG) labelled with 14C was used to study the metabolic fate of HMPG in six healthy volunteers. Besides conjugation and oxidation to 4-hydroxy-3-methoxymandelic acid (HMMA, VMA) a minor portion, 8.4 ± 1.1% (mean ± SEM) was excreted as 14C-labelled vantllic acid (VA). To study if VA was formed from HMPG or HMMA (VMA), deuterium-labelled HMPG ([2H3]HMPG) and HMMA ([2H6]HMMA) were simultaneously injected intravenously to seven healthy volunteers. The recovery of [2H3]VA from [2H3]HMPG was 8.3 ± 2.1% and the recovery of [2H6]VA from [2H6]HMMA was 9.0 ± 2.1%. The 2H-labelled VAs were probably formed by a decar boxylation reaction, in the case of HMPG after previous oxidation to HMMA.  相似文献

N 5 N 10-Methylenetetrahydromethanopterin reductase was purified 13-fold to apparent homogeneity from methanol grown Methanosarcina barkeri . The colourless enzyme was found to be composed of four identical subunits of apparent molecular mass 36 kDa. It catalysed the reduction of methylenetetrahydromethanopterin ( K m=15 μM) to methyltetrahydromethanopterin with reduced coenzyme F420 ( K m=12 μM) at a specific rate ( V max) of 2200 μmol min−1· mg protein−1 ( K cat=1320 s−1). With respect to coenzyme specificity, molecular properties and catalytic mechanism the enzyme was found to be similar to CH2=H4MPT reductase of Methanobacterium thermoautotrophicum which phylogenetically is only distantly related to M. barkeri .  相似文献

The in vitro conversion of [14C]-tryptophan to [14C]-indole-3-acetaldoxime (IAOX) by microsomal membranes of Chinese cabbage (Brassica campestris ssp. pekinensis cv. Granat) has been studied. The reaction product was identified by thin-layer chromatography (TLC) and high performance liquid chromatography (HPLC). Furthermore. IAOX was identified as an endogenous compound of Chinese cabbage by mass spectroscopy. The tryptophan-oxidizing enzyme (TrpOxE) was characterized. MnCl2 was required as cofactor, H2O2, and 2,4-dichlorophenol (DCP) stimulated the reaction. The enzyme showed a pH optimum at pH 8–9 and a Km for l -tryptophan of 20 μ M . The membranes containing TrpOxE activity were identified as plasma membranes by means of aqueous polymer two-phase partitioning. The TrpOxE from Chinese cabbage was purified 3-fold from plasma membranes by solubilization followed by (NH4)2SO4-fractionation, affinity-chromatography with concanavalin A, and native gel electrophoresis. Enzyme activity was reduced by a tunicamycin pretreatment. Several other plant species, e.g. maize (Zea mays L. Inrakorn), sunflower (Helianthus annuus L. cv. Hohes Sonnengold), tobacco (Nicotiana tabacum L. cv. White Burley), and pea (Pisum sativum L. cv. Krombeck) showed a similar conversion of [14C]-tryptophan to [14C]-IAOX by phase-partitioned plasma membranes.  相似文献

We show that ecosystem-specific aboveground net primary productivity (SANPP, g g−1 day−1, productivity on a per gram basis) can be predicted from species-level measures of potential relative growth rate (RGRmax), but only if RGRmax is weighted according to the species' relative abundance. This is in agreement with Grime's mass-ratio hypothesis. Productivity was measured in 12 sites in a French Mediterranean post-agricultural succession, while RGRmax was measured on 26 of the most abundant species from this successional sere, grown hydroponically. RGRmax was only weakly correlated ( r 2 = 0.12, P  < 0.05) with field age when species abundance was not considered, but the two variables were strongly correlated ( r 2 = 0.81, P  < 0.001) when the relative abundance of species in each field was taken into account. SANPP also decreased significantly with field age. This resulted in a tight relationship ( r 2 = 0.77, P  < 0.001) between productivity and RGRmax weighted according to species relative biomass contribution. Our study shows that scaling-up from the potential properties of individual species is possible, and that information on potential and realized species traits can be integrated to predict ecosystem functioning.  相似文献

Abstract: The ability of adenosine agonists to modulate K+-evoked 4D†-[3H]aminobutyric acid ([3H]GABA) and acetylcholine (ACh) release from rat striatal synaptosomes was investigated. The A2a receptor-selective agonist CGS 21680 inhibited Ca2+-dependent [3H]GABA release evoked by 15 m M KCI with a maximal inhibition of 29 ± 4% (IC50 of ∼4 ± 10 −12 M ). The relative order of potency of three agonists was CGS 21680 ± 5'- N -ethylcarboxamidoadenosine > R-phenylisopropyladenosine (R-PIA), with the inhibition being blocked by A2a receptor-selective antagonists (CP 66,713 and CGS 15943A) but not by the A1-selective antagonist 8-cyclopentyl-1,3-dipropylxanthine (DPCPX). When release of [3H]GABA was evoked by 30 mM KCI, no significant inhibition was observed. In contrast, CGS 21680 stimulated the release of [3H]ACh evoked by 30 m M KCI, with a maximal stimulation of 26 ± 5% (IC50 of ∼10−11 M ). This effect was blocked by CP 66,713 but not by DPCPX. The A1 agonist R -PIA inhibited [3H]ACh release, an effect blocked by DPCPX. It is concluded that adenosine A2a receptors are present on both GABAergic and cholinergic striatal nerve terminals where they inhibit and stimulate transmitter release, respectively. Abstract The relationship between the cytotoxic effect and binding to different cell lines of Clostridium perfringens enterotoxin was investigated. The enterotoxin released 51Cr from Vero and MDCK cells labeled with Na2-51CrO4. The effect varied depending upon the dose of enterotoxin and the duration and temperature of the interaction. The enterotoxin gave no effect on FL, KB, or L-929 cells. [125I]Enterotoxin bound specifically to Vero and MDCK cells via a binding site of distinct nature, but not to FL, KB, or L-929 cells. The number of the binding sites located on one MDCK cell (1.98 × 106 sites/cell) was three times that on one Vero cell (5.64 × 105 sites/cell), although the binding affinity of MDCK cell ( K a/ 3.76 × 107 M−1) was 0.1 that of Vero cells ( K a/ 3.23 × 108 M−1). Binding of the enterotoxin to susceptible cells was temperature-independent.  相似文献
