To address the questions of whether allocation of carbohydrates to roots is influenced by ionic form of nitrogen absorbed and whether allocation of carbohydrates to roots in turn influences proportionality between NH4+ and NO3? uptake from mixed sources, NH4+ and NO3? were supplied separately to halves of a split-root hydroponic system and were supplied in combination to a whole-root system. Dry matter accumulation in the split-root system was 18% less in the NH4+-fed axis than in the NO3?-fed axis. This, however, does not indicate that partitioning of carbohydrate between the two axes was different. Most of the reduction in dry matter accumulation in the NH4+-fed axis can be accounted for by the retransport of CH2O equivalents from the root back to the shoot with amino acids produced by NH4+ assimilation. Uptake of NH4+ or NO3? by the respective halves of the split-root system was proportional to the estimated allocation of carbohydrate to that half. When NH4+ and NO3? were supplied to separate halves of the split-root system, the cumulative NH4+ to NO3? uptake ratio was 0.81. When supplied in combination to the whole-root system, the cumulative NH4+ to NO3? uptake ratio was 1.67. Thus, while the shoot may affect total nitrogen uptake through the export of carbohydrates to roots, the shoot (common for halves of the split-root system) apparently does not exert a direct effect on proportionality of NH4+ and NO3? uptake by roots. For whole roots supplied with both NH4+ and NO3?, the restriction in uptake of NO3? may involve a stimulation of NO3? efflux rather than an inhibition of NO3? influx. While only the net uptake of NH4+ and NO3? was measured by ion chromatography, monitoring at approximately hourly intervals during the first 3 days of treatment revealed irregularly occurring intervals of both depletion (net influx) and enrichment (net efflux) in solutions. In the case of NH4+, numbers of net efflux events were similar (21 to 24 out of 65 sequential sampling intervals) whether NH4+ was supplied with NO3? to whole-root systems or separately to an axis of the split-root system. In the case of NO3?, however, the number of net efflux events increased from 8 when NO3? was supplied to a separate axis of the split-root system to between 19 and 24 when NO3? was supplied with NH4+ to whole-root systems.  相似文献

The carbon and nitrogen partitioning characteristics of wheat (Triticum aestivum L.) and maize (Zea mays L.) grown hydroponically at a constant pH on either 4 mM or 12 mM NO3 - or NH4 + nutrition were investigated using either 14C or 15N techniques. Greater allocation of 14C to amino-N fractions occurred at the expense of allocation of 14C to carbohydrate fractions in NH4 +-compared to NO3 --fed plants. The [14C]carbohydrate:[14C]amino-N ratios were 1.5-fold and 2.0-fold greater in shoots and roots respectively of 12 mM NO3 --compared to 12 mM NH4 +-fed wheat. In both 4 mM and 12 mM N-fed maize the [14C]carbohydrate:[14C]amino-N ratios were approximately 1.7-fold and 2.0-fold greater in shoots and roots respectively of NO3 --compared to NH4 +-fed plants. Similar results were observed in roots of wheat and maize grown in split-root culture with one root-half in NO3 --and the other in NH4 +-containing nutrient media. Thus the allocation of carbon to the amino-N fractions occurred at the expense of carbohydrate fractions, particularly within the root. Allocation of 14N and 15N within separate sets of plants confirmed that NH4 --fed plants accumulated more amino-N compounds than NO3 --fed plants. Wheat roots supplied with 15NH4 + for 8 h were found to accumulate 15NH4 + (8.5 g 15N g-1 h-1) whereas in maize roots very little 15NH4 + accumulated (1.5 g 15N g-1 h-1)It is proposed that the observed accumulation of 15NH4 + in wheat roots in these experiments is the result of limited availability of carbon within the roots of the wheat plants for the detoxification of NH4 +, in contrast to the situation in maize. Higher photosynthetic capacity and lower shoot: root ratios of the C4 maize plants ensure greater carbon availability to the root than in the C3 wheat plants. These differences in carbon and nitrogen partitioning between NO3 --and NH4 +-fed wheat and maize could be responsible for different responses of wheat and maize root growth to NO3 - and NH4 + nutrition.  相似文献

为了提高西洋参不定根的诱导率和生长速度,该研究以西洋参鲜根为外植体,在基本培养基的基础上优化IBA、碳源、氮源和磷源等营养成分。结果表明:西洋参不定根诱导过程可以明显分为外植体脱分化(愈伤化)、再分化(根形成)和根伸长等三个阶段; MS基本培养基更有利于西洋参不定根的诱导,可能与MS培养基中矿质元素含量高有关;当培养基中IBA浓度达到2 mg·L-1时,外植体表面上不定根分布密度大,诱导率达到(96±3.5)%;培养基中添加蔗糖到30 g·L-1时,不定根的诱导效果最好,但继续提高浓度后不定根变短、直径变粗;培养基中NO3-∶NH4+和PO43-浓度分别为20∶10(总氮量30 mmol·L-1)和25.0mmol·L-1时,西洋参不定根诱导率达到最大。结果提示优化培养条件可以显著改善西洋参不定根的诱导和生长,为后续西洋参不定根规模化培养提供理论支持。  相似文献

The phytotoxicity of aluminium (Al) ions can be alleviated by ammonium (NH4+) in rice and this effect has been attributed to the decreased Al accumulation in the roots. Here, the effects of different nitrogen forms on cell wall properties were compared in two rice cultivars differing in Al tolerance. An in vitro Al‐binding assay revealed that neither NH4+ nor NO3? altered the Al‐binding capacity of cell walls, which were extracted from plants not previously exposed to N sources. However, cell walls extracted from NH4+‐supplied roots displayed lower Al‐binding capacity than those from NO3?‐supplied roots when grown in non‐buffered solutions. Fourier‐transform infrared microspectroscopy analysis revealed that, compared with NO3?‐supplied roots, NH4+‐supplied roots possessed fewer Al‐binding groups (‐OH and COO‐) and lower contents of pectin and hemicellulose. However, when grown in pH‐buffered solutions, these differences in the cell wall properties were not observed. Further analysis showed that the Al‐binding capacity and properties of cell walls were also altered by pHs alone. Taken together, our results indicate that the NH4+‐reduced Al accumulation was attributed to the altered cell wall properties triggered by pH decrease due to NH4+ uptake rather than direct competition for the cell wall binding sites between Al3+ and NH4+.  相似文献

In order to investigate the effects of homogeneous and localized supply of different nitrogen forms (nitrate, NO3 ? vs ammonium, NH4 +) on the growth of tomato seedlings, root morphology and six cytokinin (CTK) fractions in xylem sap were analyzed. Whole roots were supplied with different ratios of NO3 ? to NH4 + (100:0, as 100-0NA; 75:25, as 75-25NA; 50:50, as 50-50NA) under homogeneous supply. In split-root experiments, three treatments were compared: a sole NO3 ? supply (N|N), a spatially separated supply of NO3 ? and NH4 + (N|A), and a spatially separated supply of NO3 ? and a mixture of NO3 ? and NH4 + nutrition at a ratio of 75:25 (N|AN). All concentrations of total N were set at 5 mM. The results showed that (1) homogeneous 75% NO3 ? plus 25% NH4 + supply to the whole root zone led to maximum shoot and root dry matter (DM), root surface area (RS) and root volume (RV). The spatially separated supply of NO3 ? and NH4 + (N|A) resulted in a contrasting effect on root morphology: in comparison to N|N, root DM in the NO3 ?-containing pot was increased by 50% whereas it was depressed by 50% in the NH4 +-containing pot. The 75% NO3 ? plus 25% NH4 + supply in the split-root experiment led to no significant effects either on shoot DM and root DM, or on RS and RV when compared to N|N. (2) The presence of NH4 + in the external medium led to a significantly reduced total xylem-CTK concentration, and a close negative correlation was found between xylem NH4 + and total CTK concentration irrespective of culture mode. A relatively high level of zeatin riboside (ZR) was maintained both in 75-25NA and N|A treatments. It was concluded that, in addition to the percentage of NH4 + to NO3 ? in the nutrient solution, whether NH4 + was supplied to the whole root system or to only part of the root system was also an important factor affecting plant growth. The fact that the 75-25NA and N|A treatments resulted in optimal growth of tomato seedlings might be attributed to the higher ZR concentration in xylem.  相似文献

Ammonia-oxidizing bacteria (AOB) populations were studied on the root surface of different rice cultivars by PCR coupled with denaturing gradient gel electrophoresis (DGGE) and fluorescence in situ hybridization (FISH). PCR-DGGE of the ammonium monooxygenase gene (amoA) showed a generally greater diversity on root samples compared to rhizosphere and unplanted soil. Sequences affiliated with Nitrosomonas spp. tended to be associated with modern rice hybrid lines. Root-associated AOB observed by FISH were found within a discrete biofilm coating the root surface. Although the total abundance of AOB on root biofilms of different rice cultivars did not differ significantly, there were marked contrasts in their population structure, indicating selection of Nitrosomonas spp. on roots of a hybrid cultivar. Observations by FISH on the total bacterial community also suggested that different rice cultivars support different bacterial populations even under identical environmental conditions. The presence of active AOB in the root environment predicts that a significant proportion of the N taken up by certain rice cultivars is in the form of NO3 -N produced by the AOB. Measurement of plant growth of hydroponically grown plants showed a stronger response of hybrid cultivars to the co-provision of NH4 + and NO3 . In soil-grown plants, N use efficiency in the hybrid was improved during ammonium fertilization compared to nitrate fertilization. Since ammonium-fertilized plants actually receive a mixture of NH4 + and NO3 with ratios depending on root-associated nitrification activity, these results support the advantage of co-provision of ammonium and nitrate for the hybrid cultivar.  相似文献

A study was conducted to elucidate the effect of N form, either NH4 + or NO3 , on growth and solute composition of the salt-tolerant kallar grass [Leptochloa fusca (L.) Kunth] grown under 10 mM or 100 mM NaCl in hydroponics. Shoot biomass was not affected by N form, whereas NH4 + compared to NO3 nutrition caused an almost 4-fold reduction in the root biomass at both salinity levels. Under NH4 + nutrition, salinity had no effect on the biomass yield, whereas under NO3 nutrition, increasing salinity from 10 mM to 100 mM caused 23% and 36% reduction in the root and shoot biomass, respectively. The reduced root growth under NH4 + nutrition was not attributable to impaired shoot to root C allocation since N form did not affect the overall root sugar concentration and the starch concentration was even higher under NH4 + compared to NO3 nutrition. The low NH4 + (2 mM) and generally higher amino-N concentrations in NH4 +- compared to NO3 -fed plants indicated that the grass was able to effectively detoxify NH4 +. Salinity had no effect on Ca2+ and Mg2+ levels, whereas their concentration in shoots was lower under NH4 + compared to NO3 nutrition (over 66% reduction in Ca2+; over 20% reduction in Mg2+), but without showing deficiency symptoms. Ammonium compared to NO3 nutrition did not inhibit K+ uptake, and the K+-Na+ selectivity either remained unaffected or it was higher under NH4 + than under NO3 nutrition. Results suggested that while NH4 + versus NO3 nutrition substantially reduced root growth, and also strongly modified anion concentrations and to a minor extent concentrations of divalent cations in shoots, it did not influence salt tolerance of kallar grass.  相似文献

Lolium perenne L. cv. 23 (perennial ryegrass) plants were grown in flowing solution culture and acclimatized over 49 d to low root temperature (5°C) prior to treatment at root temperatures of 3, 5, 7 and 9°C for 41 d with common air temperature of 20/15°C day/night and solution pH 5·0. The effects of root temperature on growth, uptake and assimilation of N were compared with N supplied as either NH4 or NO3 at 10 mmol m?3. At any given temperature, the relative growth rate (RGR) of roots exceeded that of shoots, thus the root fraction (Rf) increased with time. These effects were found in plants grown with the two N sources. Plants grown at 3 and 5°C had very high dry matter contents as reflected by the fresh weight: freeze-dried weight ratio. This ratio increased sharply, especially in roots at 7 and 9°C. Expressed on a fresh weight basis, there was no major effect of root temperature on the [N] of plants receiving NHJ but at any given temperature, the [N] in plants grown with NHJ was significantly greater than in those grown with NO3. The specific absorption rate (SAR) of NH+4 was greater at all temperatures than SAR-NO3. In plants grown with NH+, 3–5% of the total N was recovered as NH+4, whereas in those grown with NO?3 the unassimilated NO?3 rose sharply between 7 and 9°C to become 14 and 28% of the total N in shoots and roots, respectively. The greater assimilation of NH+4 lead to concentrations of insoluble reduced N (= protein) which were 125 and 20% greater, in roots and shoots, respectively, than in NO?3-grown plants. Plants grown with NH+4 had very much greater glutamine and asparagine concentrations in both roots and shoots, although other amino acids were more similar in Concentration to those in NO?3 grown plants. It is concluded that slow growth at low root temperature is not caused by restriction of the absorption or assimilation of either NH+4 or NO?3. The additional residual N (protein) in NH+4 grown plants may serve as a labile store of N which could support growth when external N supply becomes deficient.  相似文献

Preference for NH4+ or NO3 nutrition by the perennial legume Sesbania sesban (L.) Merr. was assessed by supplying plants with NH4+ and NO3 alone or mixed at equal concentrations (0.5 mM) in hydroponic culture. In addition, growth responses of S. sesban to NH4+ and NO3 nutrition and the effects on root nodulation and nutrient and mineral composition of the plant tissues were evaluated in a hydroponic setup at a range of external concentration of NH4+ and NO3 (0, 0.1, 0.2, 0.5, 2 and 5 mM). Seedlings of S. sesban grew equally well when supplied with either NH4+ or NO3 alone or mixed and had high relative growth rates (RGRs) ranging between 0.19 and 0.21 d−1. When larger plants of S. sesban were supplied with NH4+ or NO3 alone, the RGRs and shoot elongation rates were not affected by the external concentration of inorganic N. At external N concentrations up to 0.5 mM nodulation occurred and contributed to the N nutrition through fixation of gaseous N2 from the atmosphere. For both NH4+ and NO3-fed plants the N concentration in the plant tissues, particularly water-extractable NO3, increased at high supply concentrations, and concentrations of mineral cations generally decreased. It is concluded that S. sesban can grow without an external inorganic N supply by fixing atmospheric N2 gas via root nodules. Also, S. sesban grows well on both NH4+ and NO3 as the external N source and the plant can tolerate relatively high concentrations of NH4+. This wide ecological amplitude concerning N nutrition makes S. sesban very useful as a N2-fixing fallow crop in N deficient areas and also a candidate species for use in constructed wetland systems for the treatment of NH4+ rich waters.  相似文献

In short-term water culture experiments with different 15N labeled ammonium or nitrate concentrations, citrus seedlings absorbed NH4 + at a higher rate than NO3 . Maximum NO3 uptake by the whole plant occurred at 120 mg L–1 NO3 -N, whereas NH4 + absorption was saturated at 240 mg L–1 NH4 +-N. 15NH4 + accumulated in roots and to a lesser degree in both leaves and stems. However, 15NO3 was mostly partitioned between leaves and roots.Adding increasing amounts of unlabeled NH4 + (15–60 mg L–1 N) to nutrient solutions containing 120 mg L–1 N as 15N labeled nitrate reduced 15NO3 uptake. Maximum inhibition of 15NO3 uptake was about 55% at 2.14 mM NH4 + (30 mg L–1 NH4 +-N) and it did not increase any further at higher NH4 + proportions.In a long-term experiment, the effects of concentration and source of added N (NO3 or NH4 +) on nutrient concentrations in leaves from plants grown in sand were evaluated. Leaf concentration of N, P, Mg, Fe and Cu were increased by NH4 + versus NO3 nutrition, whereas the reverse was true for Ca, K, Zn and Mn.The effects of different NO3 -N:NH4 +-N ratios (100:0, 75:25, 50:50, 25:75 and 0:100) at 120 mg L–1 total N on leaf nutrient concentrations, fruit yield and fruit characteristics were investigated in another long-term experiment with plants grown in sand cultures. Nitrogen concentrations in leaves were highest when plants were provided with either NO3 or NH4 + as a sole source of N. Lowest N concentration in leaves was found with a 75:25 NO3 -N/NH4 +-N ratio. With increasing proportions of NH4 + in the N supply, leaf nutrients such as P, Mg, Fe and Cu increased, whereas Ca, K, Mn and Zn decreased. Yield in number of fruits per tree was increased significantly by supplying all N as NH4 +, although fruit weight was reduced. The number of fruits per tree was lowest with the 75:25 NO3 -N:NH4 +-N ratio, but in this treatment fruits reached their highest weight. Rind thickness, juice acidity, and colour index of fruits decreased with increasing NH4 + in the N supply, whereas the % pulp and maturity index increased. Percent of juice in fruits and total soluble solids were only slightly affected by NO3 :NH4 + ratio.  相似文献

The effect of two N-forms (NH4 + and NO3 ) and NaCl on pattern of accumulation of some essential inorganic nutrients was examined in sunflower (Helianthus annuus L.) cv. Hisun 33. Eight-day-old plants of were subjected for 21 d to Hoagland's nutrient solution containing 8 mM N as NH4 + or NO3 ·, and salinized with and addition of NaCl to the growth medium had no significant effect on total leaf N. However, root N of NH4-supplied plants decreased significantly with increase in NaCl concentration, whereas that of NO3-supplied plants remained unaffected. There was no significant effect of NaCl on leaf or root P, but the NO3-supplied plants had significa concentration of leaf P than that of NH4-supplied plants at varying salt treatments. Salinity of the rooting med did not show any significant effect on Na+ concentrations of leaves or roots of plants subjected to two differen N. NH4-treated plants generally had greater concentrations of Cl in leaves and roots and lower K+ content in leaves than NO3-supplied plants. Ca2+ concentrations of leaves and roots and Mg2+ concentrations of leaves decreased in NH4-supplied plants due to NaCl, but they remained unaffected in NO3-treated plants.  相似文献

Alfalfa (Medicago sativa L.) N-sufficient plants were fed 1·5 mM N in the form of NO3, NH4+ or NO3 in conjunction with NH4+, or were N-deprived for 2 weeks. The specific activity of phosphoenolpyruvate carboxylase (PEPC) from the non-nodulated roots of N-sufficient plants was increased in comparison with that of N-deprived plants. The PEPC value was highest with NO3 nutrition, lowest with NH4+ and intermediate in plants that were fed mixed salts. The protein was more abundant in NO3-fed plants than in either NH4+- or N mixed-fed plants. Nitrogen starvation decreased the level of PEPC mRNA, and nitrate was the N form that most stimulated PEPC gene expression. The malate content was significantly lower in NO3-deprived than in NO3-sufficient plants. Root malate accumulation was high in NO3-fed plants, but decreased significantly in plants that were fed with NH4+. The effect of malate on the desalted enzyme was also investigated. Root PEPC was not very sensitive to malate and PEPC activity was inhibited only by very high concentrations of malate. Asparagine and glutamine enhanced PEPC activity markedly in NO3-fed plants, but failed to affect plants that were either treated with other N types or N starved. Glutamate and citrate inhibited PEPC activity only at optimal pH. N-nutrition also influenced root nitrate and ammonium accumulation. Nitrate accumulated in the roots of NO3- and (NO3 + NH4+)-fed plants, but was undetectable in those administered NH4+. Both the nitrate and the ammonium contents were significantly reduced in NO3- and (NO3 + NH4+)-starved plants. Root accumulation of free amino acids was strongly influenced by the type of N administered. It was highest in NH4+-fed plants and the most abundant amides were asparagine and glutamine. It was concluded that root PEPC from alfalfa plants is N regulated and that nitrate exerts a strong influence on the PEPC enzyme by enhancing both PEPC gene expression and activity.  相似文献

The effects of nitrogen form and phosphorus source on the growth, nutrient uptake and rhizosphere soil property of tea (Camellia sinensis L.) were investigated in a pot experiment. The experiment was performed with a compartmental cropping device, which enables the collection of rhizosphere soil at defined distances from the root of tea plant. Nitrogen was supplied as nitrate or ammonium in combination with soluble phosphorus as Ca(H2PO4)2 or insoluble P as rock phosphate. The leaf dry matter production of tea was significantly greater in the treatments with NH4 + than NO3 -, whereas dry matter production of root and stem was not significantly affected. Addition of phosphorus as either source did not influence the dry matter production. The concentrations of K in root, Mg and Ca in both the shoot and root supplied with NO3 - were significantly higher than in NH4 + and influence of P sources was minor. On the contrary, Al and Mn concentrations were significantly larger in NH4 --fed plants which could be attributed to remarkably increased availability of Al and Mn caused by acidification of the rhizosphere soil (the first 1-mm soil section from the root surface) with NH4–N nutrition. The concentration of N in shoot was also significantly higher in NH4- than in NO3-fed plants, indicating higher use efficiency of NH4–N. Whatever the phosphate source, rhizosphere pH declined in ammonium compared to in nitrate treatment. The pH decrease was much larger when no P or soluble P were applied and reached 0.85–1.30 units which extended to 3–5 mm away from the root surface. Exchangeable acidity, content of exchangeable Al and Mn were also considerably higher in the rhizosphere soils of NH4 + fed tea plants. Significant amounts of P dissolved from rock phosphate accumulated in rhizosphere of NH4 +, not NO3 -, suggesting that the dissolution of rock phosphate was induced by the proton excreted by tea root fed with ammonium. With soluble P addition, shoot and root P concentrations were greater in NH4 + than in NO3 - treatment and it appeared that this difference could not be sufficiently explained by the available P content in soil which was only slightly higher in NH4 + treatment. With rock phosphate addition, the shoot and root P concentrations were hardly affected by nitrogen form, although the available P content was much higher and accumulated in the rhizosphere soil supplied with ammonium. Knowledge of the preferred source of N for Eucalyptus nitens will lead to improved fertiliser management practices in plantations. Ion selective microelectrodes were used non-invasively to measure simultaneously net fluxes of NH4 +, NO3 and H+ along the tap root of solution-cultured E. nitens. Measurements were conducted in solutions containing 100 m NH4NO3. The pattern of fluxes was such that there was a large influx of NH4 +, a smaller influx of NO3 and large H+ efflux. The ratio of these fluxes was constant, according to the ratio 3:1:–6 (NH4 +:NO3 :H+). Within the region 20–60 mm from the root apex of E. nitens seedlings there was spatial and temporal variation in fluxes but flux patterns remained constant. Root hair density did not affect fluxes nor did proximity to lateral roots. Variation was less than that found in previous studies of localised root fluxes using similar high-resolution measurement techniques. It was concluded that small-scale spatial variation in fluxes may have confounded previous studies. There were associations between fluxes of all three ions, the strongest associations being between NH4 + and H+, and NH4 + and NO3 . Overall, these results are consistent with NH4 + being the preferred source N for E. nitens.  相似文献

Abstract Lolium perenne L. cv. S23 was grown in flowing culture solution, pH 5, in which the concentrations of NH4+, NO3? and K+ were frequently monitored and adjusted to set values. In a pre-experimental period, plants were acclimatized to a regime in which roots were treated at 5°C with shoots at 25°C. The root temperature was then changed to one of the following, 3, 7, 9, 11, 13, 17 or 25°C, while air temperature remained at 25°C. When root temperature was increased from 5X, the relative growth rate of roots increased immediately while that of shoots changed much less for a period of approximately 9 d (phase 1). Thus, the root: shoot ratio increased, but eventually approached a new, temperature-dependent, steady value (phase 2). The fresh: freeze-dried weight ratio (i.e. water content) in shoots (and roots) increased during the first phase of morphological adjustment (phase 1). In both growth phases and at all temperatures, plants absorbed more NH4+ than NO4+, the tendency being extreme at temperatures below 9° where more than 85% of the N absorbed was NH4+. Plants at different root temperatures, growing at markedly different rates, had very similar concentrations of total N in their tissues (cells) on a fresh weight basis, despite the fact that they derived their N with differing preference for NH4+. Specific absorption rates for NH4+, NOx?, K+ and H2PO4? showed very marked dependence on root temperature in phase 1, but ceased to show this dependence once a steady state root: shoot ratio had been established in phase 2. The results indicate the importance of relative root size in determining ion fluxes at the root surface. At higher temperatures where the root system was relatively large, ‘demand’ per unit root was low, whereas at low temperatures roots were small relative to shoots and ‘demand’ was high enough to offset the inhibitory effects of low temperature on transport processes.  相似文献

Adventitious roots were produced directly from root segments ofPanax ginseng seedlings when cultured on an MS solid medium containing 3.0 mg L-1 IBA. Omitting NH4NO3 from this medium greatly enhanced both the frequency of adventitious root formation and the number of roots per expiants. This frequency declined markedly with the age of the root, but could be increased through repeated sub-culturing events. A two-step procedure that included NH4NO3 free media for the first two weeks of culture, followed by transfer onto media containing NH4NO3 for another four weeks, greatly improved total fresh weights of these adventitious roots compared with a method of continuous culture over six weeks in media that always contained NH4NO3. Expression of the genes involved in triterpene biosynthesis was analyzed by RT-PCR. Ginsenoside contents were enhanced by the omission of NH4NO3 and were also greatly increased by treatment with methyl jasmonate.  相似文献

Black root rot of tobacco, caused by Thielaviopsis basicola, is generally severe at soil pH values >5.6 and suppressed under more acidic conditions (pH < 5.2). Soil acidifying fertilizers containing NH4–N are generally recommended for burley tobacco production in North Carolina, but the effects of N form and application rate on development of black root rot and on the population dynamics of T. basicola have not been determined. Greenhouse and laboratory studies were conducted to evaluate the effects of N form (NH4 + or NO3 ) and rate on pathogen and disease parameters at several initial soil pH levels. A moderately-conducive field soil, initial pH 4.7, was adjusted to a pH of 5.5 or 6.5 by the addition of CaOH2, then amended with the desired nitrogen form and rate. Pathogen populations were determined over time. In addition, spore production in extracts of roots from plants grown in the various nitrogen and pH treatments was determined. Finally, because tobacco responds to acidic soil conditions and exposure to NH4–N by accumulating high concentrations of the polyamine putrescine, the toxicity of putrescine on vegetative growth and reproduction of T. basicola was investigated. Low soil pH and high levels of NH4–N suppressed reproduction of T. basicola in soil and in root extract, while use of NO3–N and depletion of NH4–N resulted in rapid increases in populations of T. basicola. At 20 mM, putrescine inhibited hyphal growth by 60% and aleuriospore production by 98%. Fertilizers that reduced soil pH also reduced reproduction by T. basicola, and thus have potential for management of black root rot by suppressing populations of T. basicola over multiple years of crop production. The suppression of T. basicola and black root rot observed with NH4–N amendments may partially be due to development of an inhibitory environment in the root and not solely to changes in rhizosphere pH.  相似文献

Al stress and ammonium–nitrogen nutrition often coexist in acidic soils due to their low pH and weak nitrification ability. Rice is the most Al-resistant species among small grain cereal crops and prefers NH4 + as its major inorganic nitrogen source. This study investigates the effects of NH4 + and NO3 ? on Al toxicity and Al accumulation in rice, and thereby associates rice Al resistance with its NH4 + preference. Two rice subspecies, indica cv. Yangdao6 and japonica cv. Wuyunjing7, were used in this study. After treatment with or without Al under conditions of varying NH4 + and NO3 ? supply, rice seedlings were harvested for the determination of root elongation, callose content, biomass, Al concentration and medium pH. The results indicated that Wuyunjing7 was more Al-resistant and NH4 +-preferring than Yangdao6. NH4 + alleviated Al toxicity in two cultivars compared with NO3 ?. Both NH4 +-Al supply and pretreatment with NH4 + reduced Al accumulation in roots and root tips compared with NO3 ?. NH4 + decreased but NO3 ? increased the medium pH, and root tips accumulated more Al with a pH increase from 3.5 to 5.5. Increasing the NO3 ? concentration enhanced Al accumulation in root tips but increasing the NH4 + concentration had the opposite effect. These results show NH4 + alleviates Al toxicity for rice and reduces Al accumulation in roots compared with NO3 ?, possibly through medium pH changes and ionic competitive effects. Making use of the protective effect of NH4 +, in which the Al resistance increases, is advised for acidic soils, and the hypothesis that rice Al resistance is associated with the preferred utilization of NH4 + is suggested.  相似文献

We examined the effect of soil NH4+ and NO3? content upon the root systems of field-grown tomatoes, and the influence of constant, low concentrations of NH4+ or NO3? upon root growth in solution culture. In two field experiments, few roots were present in soil zones with low extractable NH4+ or NO3?; they increased to a maximum in zones having 2μg-N NO3? g?1 soil and 6 μg-N NO3= g?1 soil, but decreased in zones having higher NH4+ or NO3? levels. Root branching was relatively insensitive to available mineral nitrogen. Plants maintained in solution culture at constant levels of NH4+ or NO3?, had similar shoot biomass, but all root parameters – biomass, length, branching and area – were greater under NH4 nutrition than under NO3?. These results suggest that the size of root system depends on a functional equilibrium between roots and shoots (Brouwer 1967) and on the balance between soil NH4+ and NO3?.  相似文献
