Hill reaction and noncyclic photophosphorylation of isolated class C chloroplasts of spruce (Picea abies (L.) Karst.), as well as 14CO2 fixation by whole needles at constant laboratory conditions proceeded at high rates during spring and early summer, declined during late summer and autumn by about 60%, remained at this level during winter, and recovered quickly in early spring. During summer, the whole needles proved to be frost labile, since after exposure to-20°C and careful thawing, fast chlorophyll degradation occurred. In addition, only photosynthetically inactive chloroplasts could be isolated from those precooled needles. On the contrary, during winter the photochemical activities of plastids from freshly harvested needles did not differ from those of artificially frozen-thawed needles. When isolated spruce chloroplasts were exposed to the same subfreezing temperatures as the whole needles, no influence of freezing on the photochemical activities was observed, irrespective of whether the plastids were isolated from frost sensitive or frost hardened needles. It is concluded that frost damage to spruce chloroplasts is due to an attack of membrane toxic compounds or lytic enzymes which were liberated upon freezing from more labile compartments. Frost hardening of the chloroplasts, as determined by the stability of chlorophyll after exposure of the needles to low temperatures, as well as by the isolation of photosynthetically active chloroplasts from such precooled needles, appeared to depend at least on 2 processes: (i) an alteration of the composition of the photosynthetically active membranes and (ii) and additional stabilization of these membranes by protecting substances. The first process was indicated by a large increase (decrease) of the capability of isolated chloroplasts for PMS-mediated photophosphorylation which accompanied natural or artificial frost hardening (dehardening). Production of cryoprotecting compounds was suggested by a significant higher stability against NaCl observed with class C chloroplasts isolated from frost hardened needles as compared to that of plastids from frost labile material. The decrease of the capability for both, the ferricyanide dependent photoreactions of the plastids and the CO2 fixation by whole needles, which was observed during the frost hardening phase, cannot be due to freezing injuries; it rather appears to be a consequence of the frost hardening process.  相似文献

Soybean plants grown in controlled environment cabinets under light intensities of 220 w/m2 or 90 w/m2 (400–700 nm) and day to night temperatures of 27.5–22.5 C or 20.0–12.5 C in all combinations, exhibited differences in growth rate, leaf anatomy, chloroplast ultrastructure, and leaf starch, chlorophyll, and chloroplast lipid contents. Leaves grown under the lower light intensity at both temperatures had palisade mesophyll chloroplasts containing well-formed grana. The corresponding leaves developed under the higher light intensity had very rudimentary grana. Chloroplasts from high temperature and high light had grana consisting of two or three appressed thylakoids, while grana from the low temperature were confined to occasional thylakoid overlap. Spongy mesophyll chloroplasts were less sensitive to growth conditions. Transfer experiments showed that the ultrastructure of chloroplasts from mature leaves could be modified by changing the conditions, though the effect was less marked than when the leaf was growing.  相似文献

Abstract The effect that the massive presence of lichen thalli growing on the branches of Quercus pyrenaica and Q. rotundifolia leaves has on their chloroplasts been studied. In both species there were significant decreases in the amount of chlorophylls in the leaves of twigs with a dense cover of lichens in comparison with the leaves from thallus-free twigs. The areas and perimeter of chloroplasts in leaves from twigs with epiphytes did not differ significantly from those in leaves without epiphytes. However, in leaves with epiphytes the percentage of chloroplast area occupied by starch was higher. In Q. pyrenaica the number of grana per chloroplast section and per μm2, the percentage of chloroplast stroma occupied by grana, the average number of thylakoids forming grana and the grana width was significantly smaller in leaves near lichen populations. These results are discussed and related to the great chelating capacity of the lichen's substances.  相似文献

Wheat seedlings (Triticum aestivum L.) develop plastids (etioplasts and chloroplasts) which exhibit alterations in inner membrane organisation after treatment with Norflurazon (NF), an inhibitor of carotenoid biosynthesis. In dark-grown plants, it results in a decreased amount of partitions (contact zones) between prothylakoids. Under weak red radiation (WRR), plants contain chloroplasts devoid of grana. Using the fluorescent probe 9-amino acridine (9-AA), the average surface charge density of isolated prothylakoids (PTs) was –21.8±3.2 mC m–2 and –27.4±2.6 mC m–2 in the control and after treatment, respectively. Thylakoid membranes isolated from plants grown under WRR exhibited slightly more negative values, –23.5±2.9 mC m–2 and –29.0±2.1 mC m–2, in control and after NF treatment, respectively. The surface charge density of de-stacked thylakoids from greenhouse-grown untreated plants, containing extensive grana stacking, was –34.3±2.5 mC m–2. Assays using the fluorescent probe of DPH (1,6-diphenyl-1,3,5-hexatriene) showed a higher polarisation value when incorporated into thylakoids from NF-treated plants compared to untreated plants grown under WRR. The highest polarisation value was found in untreated plants grown in the greenhouse. This indicates a lower rotation transition of the probe in the lipid environment of thylakoids after NF treatment, which can be interpreted as more rigid membranes. Hence the surface charge density and the mobility of membrane components may play a major role for the formation of partitions in dark-grown plants and in the formation of grana in plants grown under WRR.  相似文献

We examined morphological and ultrastructural differences in chloroplasts of cotton leaves and the fruiting organs, bract, and capsule wall to advance our understanding of their commonly observed differences in photosynthetic efficiency. Chloroplasts from leaves were large (7.1 μm long in cross section), lens shaped with a well developed membrane system differentiated into grana and stroma lamellae that occupied the large cross-sectional area (12.3 μm2) of the organelle. A few small plastoglobuli and starch grains were scattered in the stroma region. The bract chloroplasts were correlative of leaf chloroplasts in size (6.8 μm in length) and shape with the exception that the bract chloroplasts exhibited greater thylakoid number per granum (15.8) than the leaf chloroplasts (10.5). In contrast to leaf and bract, the capsule wall chloroplasts were smaller in size (4.3 μm) and cross sectional area (6.8 μm2) than either the leaf or bract. The most intriguing feature of the capsule wall chloroplasts was its domination by large starch granules (5.3 μm2) in the stroma which filled the whole chloroplast coercing the membrane system to move towards the periphery of the organelle. It has been reported in quite a number of literatures that doubled CO2 concentration increased the photosynthetic rate and dry matter production of C3 plants, but substantially affected C4 plants little. However, why may CO2 enrichment promote growth and either no change or decrease reproductive allocation of the C3 species, but havinag no effects on growth characteristics of the C4 plants? So far, there has been no satisfactory explanation on that mentioned above, except the differences in their CO2 compensatory points. In the past, although some studies on ultrastructure of the chloroplasts under doubled CO2 concentration were limitedly conducted. Almost all the relevant experimental materials were only from C3 plants not from C4 plants, and even though the results were of inconsistancy. Thereby, it needs to verify whether the differences in photosynthesis of C3 and C4 plants at doubled CO2 level is caused by the difference in their chloroplast deterioration. Experiments to this subject were conducted at the Botanical Garden of Institute of Botany, Academia Sinica in 1993 and 1994. Both experimental materials from C3 plant alfalfa (Medicago sativa) and C4 plant foxtail millet (Setaria italica) were cultivated in the cylindrical open-top chambers (2.2 m in diameter × 2.4 m in height) with aluminum frames covered by polyethylene film. Natural air or air with 350× 10-6 CO2 were blown from the bottom of the chamber space with constant temperature between inside and outside of the chamber 〈0.2℃〉. Electron microscopic observation revealed that the ultrastructure of the chloroplasts from C3 plant Medicago sativa and C4 plant Seteria italica growing under the same doubled CO2 concentration were quite different from each other. The differential characteristics in ultrastructure of chloro plasts displayed mainly in the configuration of thylakoid membrances and the accumulation of starch grains. They were as follows: 1. The most striking feature was the building up of starch grains in the chloroplasts of the bundle sheath cells (BSCs) and the mesophyll cells (MCs) at doubled CO2 concentra tion. The starch grains appeared centrifugally first in the BSCs and then in the chloroplast of the other MCs. It was worthy to note that the starch grains in the chloroplasts of C4 plant Setaria ira/ica were much more than those of the C3 plant Medicago sativa . The decline of photosynthesis in the doubled CO2-grown C4 plants might be caused by an over accumulation of starch grains, that deformed the chloroplast even demaged the stroma thylakoids and grana. There might exsist a correlation between the comformation of thylakoid system and starch grain accumulation, namely conversion and transfer of starch need energy from ATP, and coupling factor (CF) for ATP formation distributed mainly on protoplastic surface (PSu) of stroma thylakoid membranes, as well as end and margin membranes of grana thylakoids. Thereby, these results could provide a conclusive evidence for the reason of non effectiveness on growth characteristics of C4 plant. 2. Under normal condition , the mature chlolroplats of higher plants usually develop complete and regularly arranged photosynthetic membrane systems . Chloroplasts from the C4 plant Setaria italica, however, exerted significant changes on stacking degree, grana width and stroma thylakoid length under doubled CO2 concentration; In these changes, the grana stacks were smaller and more numerous, and the number of thylakoids per granum was greatly increased, and the stroma thylakoid was greatly lengthened as compared to those of the control chloroplasts. But the grana were mutually intertwined by stroma thylakoid. The integrity of some of the grana were damaged due to the augmentation of the intrathylakoid space . Similarly, the stroma thylakoids were also expanded. In case. the plant was seriously effected by doubled CO2 concentration as observed in C4 plant Setaria italica , its chloroplasts contained merely the stroma (matrix) with abundant starch grains, while grana and stroma thylakoid membranes were unrecognizable, or occasionally a few residuous pieces of thylakoid membranes could be visualized, leaving a situation which appeared likely to be chloroplast deterioration. However, under the same condition the C3 plant Medicago sativa possessed normally developed chloroplasts, with intact grana and stroma thylakoid membranes. Its chloroplasts contained grana intertwined with stroma thylakoid membranes, and increased in stacking degree and granum width, in spite of more accumulated starch grains within the chloroplasts. These configuration changes of the thylakoid system were in consistant with the results of the authors another study on chloroplast function, viz. the increased capacity of chloroplasts for light absorption and efficiency of PSⅡ.  相似文献

Photosynthetic activity, the content of various photosynthetic pigments, and the chloroplast ultrastructure were examined in the leaves of cucumber (Cucumis sativus L.) and pea (Pisum sativum L.) plants of different ages grown under red light (600–700 nm, 100 W/m2). In pea leaves tolerant to red-light irradiation, chloroplast ultrastructure did not essentially change. In the first true leaves of cucumber plants susceptible to red-light irradiation, we observed a considerable increase in the number and size of plastoglobules, the appearance of chloroplasts lacking grana or containing only infrequent grana, and stromal thylakoids. In the upper leaves of 22-day-old cucumber plants, the chloroplast structure was essentially similar to that of the control chloroplasts in white light, and we therefore suppose that these plants have acclimated to red light.  相似文献

Mung bean (Phaseolus aureus Roxb.) is grown after harvest of wheat during the fallow period. Herbicides such as metsulfuron, atrazine and isoxaflutole are recommended to control weeds in wheat–rice cropping system including weeds of fallow crop. The effects of three herbicides with different modes of action—atrazine, photosystem II inhibitor; metsulfuron, acetolactate synthase inhibitor; and isoxaflutole, 4-hydroxyphenylpyruvatedioxygenase inhibitor—on shoot height, chlorophyll concentrations and cellular damage in herbicide-treated mung bean were studied. While isoxaflutole inhibited shoot growth and chlorophyll concentration of mung bean, atrazine and metsulfuron did not cause reduction in the shoot growth of mung bean. Metsulfuron (226, 452, 1356 and 2260 μg/kg soil) and isoxaflutole (452, 1356 and 2260 μg/kg soil) in soil reduced the concentration of leaf chlorophyll of mung bean compared to the control. Atrazine in soil did not affect the total chlorophyll concentration of mung bean leaves. Electron micrographs showed that untreated mung bean had elongated chloroplasts, thylakoids organized as intact grana, distinct starch grains and a small number of plastoglubuli. Mesophyll cells of atrazine-treated mung bean leaves had swollen chloroplasts and thylakoids with disorganized grana. Leaves of metsulfuron-treated mung bean had swollen chloroplasts with a large number of starch grains. Starch grains were not observed in leaves of mung bean treated with either atrazine or isoxaflutole. Complete disruption of thylakoids was observed in isoxaflutole-treated mung bean leaves. Leaves of atrazine-treated mung bean showed detached microfibrils along with distorted and degenerated secondary walls. Metsulfuron-treated mung bean leaves showed aggregated microfibrils with completely dissolved secondary walls, while isoxaflutole-treated leaves had completely degenerated secondary walls with complete loss of microfibrils. We conclude that isoxaflutole at higher doses, influence mung bean at the morphological, physiological and cellular levels.  相似文献

Soybean ( Glycine max (L.) Merr. ) plants were grown under ambient and elevated CO2 (plus 350 μL/L) concentration in cylindrical open-top chamber to examine their effects on the ultra- structure of chloroplasts. The upper, lower and mid-node leaves were harvested after 7 days full expansion under different CO2 concentrations and ultrathin section were prepared for transmission electron microscopy. In general, the average content of starch grains and thylakoid membranes in the chloroplasts under the elevated CO2 concentration were always higher than the control. Under higher CO2 concentration, there were smaller and less starch grains in the chloroplasts from upper-node leaves than those from mid-node leaves. The shape of their starch grains changed from elliptical to oval,and their thylakoid membranes and grana remained normal. At lower-node leaves, one or two oval, or three timer starch grains accumulated in the chloroplasts. In the mid-node leaves,however, some chloroplasts under higher CO2 concentration had rather large tim elliptical starch grains which could consequently cause disruption of grana and stroama thylakoids in the chloroplasts, whereas in other chloroplasts, the thylakoid membranes and grana were not deformed as the starch grains were smaller and elliptical. On the other hand, under higher CO2 concentration, the stacking degree of thylakoid membranes and starch grains accumulation in the mid-node leaves were significantly higher than those in the lower-node leaves,and slightly higher than the upper-node leaves. These results, in agreement with the chlorophyll contents and photosynthetic rate which reported by other authors in the past, indicated that the ultrastmcture response of the chloroplasts from different leaf nodes of soybeen under elevated CO2 coneentration were different. The seed yield of soybean at different nodes was decreased gradually from mid-nodes towards both upper- and lower-nodes. The greatest effect of elevated CO2 eoneentrafion on seed yeild was at the mid-node leaves. The variation of seed yields of soybean at different nodes under elevated CO2 concentration was in eoneert with the change in the ultrastmcture of chloroplasts and in turn the change in their photosynthetic rates of leaves at different nodes.  相似文献

The localization of photosynthetic activity in developing maize (Zea mays L.) chloroplasts was studied in situ by two electron-microscopic-cytochemical methods. The activity of photosystem I was detected by photooxidation of 3,3-diaminobenzidine (DAB) and the activity of the photosystem II by photoreduction of thiocarbamyl nitrotetrazolium blue (TCNBT). During the transformation of proplastids into chloroplasts, at the base of the leaf blade the DAB reaction appeared before the TCNBT reaction. A positive DAB reaction was observed in the single thylakoids of plastids in cells located only about 0.5 mm above the base. Dark, osmiophilic DAB polymers accumulated in the lumina of the thylakoids. Plastid envelopes and tubules of the prolamellar bodies in immature chloroplasts were DAB-negative. In fully differentiated leaf tissue the DAB reaction was intense in the thylakoids of bundle-sheath chloroplasts, as well as in the stroma thylakoids and the peripheral grana thylakoids of mesophyll chloroplats. The photoreduction of TCNBT started in leaf tissue about 1 mm above the base. Dark granular material of reduced TCNBT appeared mostly in the partitions of grana, i.e. interthylakoidally, but some granules were also attached to the stroma thylakoids. The membranes of plastid envelopes and the tubules of prolamellar bodies showed a negative TCNBT reaction. Young bundle-sheath chloroplasts contained some reduced TCNBT in their grana; these deposits largely disappeared in the course of further differentiation. Ultrastructure of leaf mesophyll and two-layer trap wall of Utricularia vulgaris L. was studied after 24-h and 48-h treatment with different concentrations of cadmium. The treatment of plants with 0.25 g(Cd2+) m−3 caused different changes in the chloroplasts of the two organs. The thylakoids swelled in chloroplasts of leaf and the starch content increased in chloroplasts of traps. Higher concentrations of Cd (0.5 and 1.5 g m−3) caused osmiophilisation of thylakoids in the chloroplasts of both studied organs. The secretory cells in the trap wall was resistant to Cd. 对高CO_2浓度下生长的大豆(Glycine max(L.)Merr.)不同叶位的叶片进行了电镜观察,揭示出大豆不同叶位叶片的叶绿体对倍增的CO_2浓度反应不一。其显著的超微结构差异特征是:1.叶位居中的叶片叶绿体积累的淀粉粒不仅很大,而且最多,有的叶绿体中的淀粉粒可达20个,几乎充满着叶绿体的基质空间。2.下位叶叶绿体的淀粉粒积累较多,通常为2~5个;3.上位叶叶绿体所含淀粉粒既小又少,虽然有的叶绿体中也积累有3~4个淀粉粒,但大多数叶绿体中所含淀粉粒仅有1~2个。以上结果联系到大豆中位叶的光合作用速率较高及对籽粒产量起作用最大来讨论是很有意义的。  相似文献

Investigations on heat resistance of spinach leaves   总被引:4,自引:0,他引:4  
Exposure of spinach plants to high temperature (35° C) increased the heat resistance of the leaves by about 3° C. This hardening process occurred within 4 to 6 h, whereas dehardening at 20°/15° C required 1 to 2 days. At 5° C dehardening did not take place. Hardening and dehardening occurred in both the dark and the light. The hardiness was tested by exposure of the leaves to heat stress and subsequent measurements of chlorophyll fluorescence induction and light-induced absorbance changes at 535 nm on the leaves and of the photosynthetic electron transport in thylakoids isolated after heat treatment. Heat-induced damage to both heat-hardened and non-hardened leaves seemed to consist primarily in a breakdown of the membrane potential of the thylakoids accompanied by partial inactivation of electron transport through photosystem II. The increase in heat resistance was not due to temperature-induced changes in lipid content and fatty acid composition of the thylakoids, and no conspicuous changes in the polypeptide composition of the membranes were observed. Prolonged heat treatment at 35° C up to 3 days significantly decreased the total lipid content and the degree of unsaturation of the fatty acids of membrane lipids without further increase in the thermostability of the leaves. Intact chloroplasts isolated from heat-hardened leaves retained increased heat resistance. When the stroma of the chloroplasts was removed, the thermostability of the thylakoids was decreased and was comparable to the heat resistance of chloroplast membranes obtained from non-hardened control plants. Compartmentation studies demonstrated that the content of soluble sugars within the chloroplasts and the whole leaf tissue decreased as heat hardiness increased. This indicated that in spinach leaves, sugars play no protective role in heat hardiness. The results suggest that changes in the ultrastructure of thylakoids in connection with a stabilizing effect of soluble non-sugar stroma compounds are responsible for acclimatization of the photosynthetic apparatus to high temperature conditions. Small particles derived from the digitonin treatment of chloroplast thylakoid membranes in either the stacked (grana-containing) or unstacked condition, as determined by cation concentration, have been used to study the aggregation of thylakoid membranes. At pH values above 5, the small particles from stacked chloroplasts do not aggregate in the presence of Mg2+ or other screening cations at concentrations sufficient to cause the restacking of thylakoids from low-salt chloroplasts. However, the small particles from stacked chloroplasts are aggregated either by lowering the pH to 4.6 or adding the binding cation La3+. In contrast, the small particles obtained on digitonin treatment of unstacked chloroplasts were aggregated by cations at neutral pH. Large particles (mainly grana) derived from digitonin treatment of stacked chloroplasts could not be unstacked by transfer to media of low cation concentration. It is concluded that the nonappressed regions of the chloroplast thylakoid membranes under stacking conditions carry higher than average negative surface charge densities under physiological pH conditions. Transfer of chloroplasts to media of low cation concentration causes a time-dependent lateral redistribution of charge between the appressed and nonappressed regions, but this redistribution is prevented by prior digitonin treatment of stacked chloroplasts.  相似文献

Structural variation in the stroma‐grana (SG) arrangement of the thylakoid membranes, such as changes in the thickness of the grana stacks and in the ratio between grana and inter‐grana thylakoid, is often observed. Broadly, such alterations are considered acclimation to changes in growth and the environment. However, the relation of thylakoid morphology to plant growth and photosynthesis remains obscure. Here, we report changes in the thylakoid during leaf development under a fixed light condition. Histological studies on the chloroplasts of fresh green Arabidopsis leaves have shown that characteristically shaped thylakoid membranes lacking the inter‐grana region, referred to hereafter as isolated‐grana (IG), occurred adjacent to highly ordered, large grana layers. This morphology was restored to conventional SG thylakoid membranes with the removal of bolting stems from reproductive plants. Statistical analysis showed a negative correlation between the incidences of IG‐type chloroplasts in mesophyll cells and the rates of leaf growth. Fluorescence parameters calculated from pulse‐amplitude modulated fluorometry measurements and CO2 assimilation data showed that the IG thylakoids had a photosynthetic ability that was equivalent to that of the SG thylakoids under moderate light. However, clear differences were observed in the chlorophyll a/b ratio. The IG thylakoids were apparently an acclimated phenotype to the internal condition of source leaves. The idea is supported by the fact that the life span of the IG thylakoids increased significantly in the later developing leaves. In conclusion, the heterogeneous state of thylakoid membranes is likely important in maintaining photosynthesis during the reproductive phase of growth.  相似文献

Abstract Stacking of thylakoid membranes in vitro was assessed using electron microscopy. Grana stacks of spinach thylakoids formed when 5 mol m?3 MgCl2 was present, but no stacking of thylakoids from the mangrove Avicennia marina occurred in the presence of 10 mol m?3? MgCl2. Isolation of mangrove thylakoids with a high osmotic strength medium did not induce grana formation if the medium consisted only of sorbitol or glycinebetaine. Addition of cations to the high osmotic strength medium did induce some loose-grana formation, with divalent cations being more effective than monovalent cations. Glycinebetaine was a better osmoticum than sorbitol for grana formation provided divalent cations had been added. Oxygen evolution activity of the preparations was influenced by the amount of membrane stacking, with the preparations with the greatest amount of stacked membrane having the highest activity. Isolation with sorbitol or glycinebetaine based media did not alter this pattern, nor did assay in sorbitol or glycinebetaine. Mangrove thylakoids have a requirement for both a high osmotic strength and divalent cations for grana formation in vitro which may be related to the low water potential of the plant environment in vivo.  相似文献

The amount and distribution of proteins of the light-harvesting complex associated with photosystem II (PS II) were investigated using immunogold labelling of chloroplasts of wheat ( Triticum aestivum L. cv. Walde). The seedlings were grown in weak red light (16 mW m−2) after imbibition of grains with SAN-9789 (Norflurazon, 0.028 to 28 mg I−1). Chloroplasts of these plants exhibited thylakoids with different degrees of stacking. Thylakoids of untreated plants grown in a greenhouse had most gold particles per unit membrane length in both appressed and non-appressed regions compared to red light grown plants. The ratios of labelling between appressed and non-appressed membranes were fairly constant in red light- and greenhouse-grown plants. The labelling densities were 2.5–3 times higher in the appressed thylakoids compared to the non-appressed thylakoids. However, at a SAN concentration of 2.8 mg I−1 there was a sharp decrease in thylakoid appressions and in labelling density of both appressed and non-appressed membranes. The total amount of particles per chloroplast was also much lower as compared to that at lower SAN concentrations. Plants treated with the highest concentration of SAN (28 mg I−1) contained chloroplasts devoid of normal grana structures. In these plastids, the thylakoids were elongated and single. The labelling density in these membranes was ca 50% of that observed at 2.8 mg I−1. This paper thus supports earlier observations that proteins of the light-harvesting complex of PS II (LHC II) are mainly localized in the appressed regions of the grana membranes, and may be involved in the formation of grana.  相似文献

Effects of water stress on needle ultrastructure of 2-year-old Scots pine (Pinus sylvestris L.) and 5-year-old Norway spruce [Picea abies (L.) Karst.] seedlings were studied in greenhouse experiments. Drought stress was induced by leaving seedlings without watering, and waterlogging stress was produced by submerging the seedling containers in water. Needle samples for ultrastructural analyses were collected several times during the experiments, and samples for nutrient analyses at the end of the experiments. In drought stress, plasmolysis of mesophyll and transfusion parenchyma tissues, aggregation of chloroplast stroma and its separation from thylakoids and decreased size and abundance of starch grains in needles of both species were observed. The concentration of lipid bodies around the chloroplasts were detected in pine needles. Calcium and water concentrations in spruce needles were lower by the end of the experiments compared to controls. In waterlogging treatment, swelling of phloem cells in pine needles and large starch grains, slight swelling of thylakoids and increased translucency of plastoglobuli in chloroplasts of both species studied were observed. The phosphorus concentration in pine needles was higher while phosphorus, calcium and magnesium concentrations in spruce needles were lower in the waterlogging treatments compared to controls. Typical symptoms induced by drought stress, e. g. aggregation of chloroplast stroma and its separation from thylakoids, were detected, but, in waterlogging stress, ultrastructural symptoms appeared to be related to the developing nutrient imbalance of needles.  相似文献

Membrane-bound ribosomes of chloroplasts, isolated from pea seedlings during grana formation, can be partially liberated by 0.5 M KCl and 0.001 M puromycin. In case of mature chloroplasts, after the completion of grana formation process these agents are inefficient, and liberation of ribosomes and polyribosomes may be achieved only after solubilization of thylakoid membranes by 1% Triton X-100. Electron microscopic study of the heavy membrane fraction of young chloroplasts reveals electron-transparent membranes, containing rings and discs of thylakoids with a diameter of about 2 mum. These rings are liberated together with ribosomes under the action of 0.5 M KCl; Triton X-100 liberates equally-sized annular polyribosomes. The rings detected in chloroplast membranes at early stages of development are regarded as structures, precursor grana thylakoids, and the annular polyribosomes included into them as immediate participants of thylakoid morphogenesis.  相似文献

The sea anemoneCereus pedunculatus was artificially UV-irradiated to test the effect of UV-light on the number of endosymbiotic dinoflagellates in its gastrodermis and on their ultrastructure. Anemones were kept in the laboratory in a light: dark cycle (LD 12∶12; 13 W m−2) at 18 °C and briefly (2, 5 and 9 d) exposed to UV radiation at quasisolar intensities, 0.5 or 1 W m−2. Their tentacles were then examined in the electron microscope for qualitative and quantitative changes in the zooxanthellae. There was an intensity-dependent decrease in the number of symbionts, which in some cases were lost altogether (bleaching). Irradiated anemones contained a larger proportion of symbionts with ultrastructural abnormalities, namely diminished starch, some mitochondria with altered matrix and, in particular, characteristic changes in the chloroplasts; instead of being densely stacked, the thylakoids were spread apart and swollen at the ends of their membranes to form vesicle-like structures. Relatively large vesicles also appeared in the cytoplasm. The resulting enlargement of the whole dinoflagellate cell was documented morphometrically.
