Summary Electroretinograms (ERG) were recorded from dark- and chromatic-adapted compound eyes in the dusk-active firefly,Photinus pyralis , at different wavelengths ranging from 320 to 700 run and over 4.5 log units change in stimulus intensity. ERG waveforms differed in the short (near-UV and violet) and long (yellow) wavelengths (Fig. 1). Waveform differences were quantitated by analysis of rise and fall times as a function of the amplitude of the response. Rise times were found to be relatively constant for all stimulus wavelengths. However, variations in the fall times were detected and followed characteristically different functions for short and long wavelengths (Fig. 2).No significant differences in the slopes of the Vlog-I curves at different stimulus wavelengths were observed (Fig. 3).Spectral sensitivity curves obtained from the ventral sector in dark- and chromatic-adapted conditions revealed peaks in the short ( max 400 nm: Fig. 4; max 430 nm: Fig. 5 A; and max 380 nm; Fig. 5B) and long ( max 570 nm: Figs. 4, 5) wavelengths, suggesting the presence of two spectral mechanisms. A neural net model of discrete populations of formal neurons have been constructed to study evoked potentials based on our previous simulation studies (Anninos, 1972). Some interesting results came up from the examination of our findings regarding the latencies and the period of the cyclic activity of the evoked potentials. In fact, the different successive latencies for the five identical stimuli and the different periods for each of the cyclic activities, all are consequences of inhibitory and excitatory influences from a large neuronal population. Furthermore, such behavior of the system is not only related to the unknown neuronal population but was also substantially altered by what occurred in other systems at the time of stimulus, or prior to it.Computation assistance was provided by the Health Sciences Computing Facility, UCLA, sponsored by NIH Special Research Resources Grant RR-3. The depolarization of a passive nerve cylinder or dendritic tree in the equivalent cylinder representation is assumed to satisfy the cable equation. We consider in detail the effects of white noise current injection at a given location for the case of sealed end boundary conditions and for an initial resting state. The depolarization at a point is a Gaussian random process but is not Markovian. Expressions (infinite series) are obtained for the expectation, variance, spatial and temporal covariances of the depolarization. We examine the steady state expectation and variance and investigate how these are approached in time over the whole neuronal surface. We consider the relative contributions of various terms in the series for the expectation and variance of the depolarization at x=0 (soma, trigger zone, recording electrode) for various positions of the input process. It is found that different numbers of terms must be taken to obtain a reasonable approximation depending on whether the stimulus is at proximal, central or distal parts of the dendritic tree. We consider briefly the interspike time problem and see in an approximate way how spatial effects are important in determining the mean time between impulses.The research was supported by Canadian National Research Council Operating Grant No. A9259 and No. A4559. An escape learning situation is discussed in terms of a neural model in which a stimulus can result in a conditioned excitement and a specific conditioned response. By using the simplest relations between the strengths of conditioning and the number of reinforcements and by introducing a distribution of fluctuations occurring regularly in time, one can calculate the probabilities of various responses, as well as the various latencies, in successive trials. The results are in moderately satisfactory agreement with the data of R. L. Solomon and L. C. Wynne (Psychol. Monogr.,67, No. 4, 1953). Consequences of the model for various experimental situations are discussed. Pre-cue activity, the neuronal modulation that precedes a predictable stimulus, was studied in the premotor cortex of three rhesus monkeys. In one condition, a directional cue dictated the timing and target of a forelimb movement. In another condition, a non-directional cue provided identical timing information but did not indicate the target. Of 501 task-related neurons recorded in premotor cortex, 168 showed pre-cue activity. The onset time of pre-cue activity varied markedly from trial to trial and cell to cell, ranging from trial initiation to 4.8 sec later. No pre-cue activity reflected the direction of limb movement; thus, the data argue against the hypothesis that pre-cue activity reflects preparation for specific limb movements. A small number of cells showed greater pre-cue activity before directional than before nondirectional cues, and this difference may reflect anticipation of the cue's directional information. However, the vast majority (84%) of neurons lacked such differences. We therefore hypothesize that most pre-cue activity reflects or contributes to a facet of behavior common to the two conditions: anticipation of the time and/or nature of events.  相似文献

Three neuronal models of the spike initiating process were investigated with respect to their ability to show adaptation to a current step: (i) the perfect integrator model (PIM), (ii) the leaky integrator model (LIM), and (iii) the Hodgkin-Huxley (HH-) model. It was found that although each neuronal model will generate different response spike trains to a given stimulus, all responses fulfilled the criteria of a deterministic neural response (Awiszus 1988). The results show that both PIM and LIM are unable to show adaptation regardless of the choice of model parameters whereas the HH-model shows a clear rate of discharge adaptation. The reason for this adaptation lies in the fact that there are conditions for the HH-model where a step stimulus is highly effective. These conditions are investigated by means of a phase plane analysis. Consequences of these results for the explanation of neuronal adaptation and the validity of the neuronal models investigated are discussed.  相似文献

ST0838 (designed stRad55B) is one of the four RadA paralogs (or Rad55 homologues) in the genome of the hyperthermophilic crenarchaeon Sulfolobus tokodaii. The gene is induced by UV irradiation, suggesting that it is involved in DNA recombinational repair in this organism. However, this protein could not be expressed normally in vitro. In this study, thermostable and soluble stRad55B was obtained by co-expression with S. tokodaii RadA (stRadA) in E. coli, and the enzymatic properties were examined. It was found that stRad55B bound ssDNA preferentially and had a very weak ATPase activity that was not stimulated by DNA. The recombinant protein inhibited the strand exchange activity promoted by stRadA, indicating that stRad55B might be an inhibitor to the homologous recombination in this archaeon. The results will be helpful for further functional and interaction analysis of RadA paralogs and for the understanding of the mechanism of recombinational repair in archaea. Screening and breeding of high taxol producing fungi by genome shuffling   总被引:4,自引:0,他引:4  
To apply the fundamental principles of genome shuffling in breeding of taxol-producing fungi, Nodulisporium sylviform was used as starting strain in this work. The procedures of protoplast fusion and genome shuffling were studied. Three hereditarily stable strains with high taxol production were obtained by four cycles of genome shuffling. The qualitative and quantitative analysis of taxol produced was confirmed using thin-layer chromatography (TLC), high performance liquid chromatography (HPLC) and LC-MS. A high taxol producing fungus, Nodulisporium sylviform F4-26, was obtained, which produced 516.37 μg/L taxol. This value is 64.41% higher than that of the starting strain NCEU-1 and 31.52%―44.72% higher than that of the parent strains.  相似文献

In this study, our previous results on the important relation between EEG and EPs were extended by experiments with chronically implanted and freely moving cats, which had electrodes at the acoustical cortex, inferior colliculus and reticular formation. During the experiments the frequency stabilization upon sound stimulation was shown in the frequency domain by comparison of the pre-stimulus power spectra and post-stimulus amplitude frequency characteristics. Comparative frequency domain analysis of about 75 EEG-EPograms (sample of spontaneous activities just prior to stimulation and single evoked potentials following the stimulation), which were recorded from all the brain nuclei mentioned above and from each of the 11 cats, was performed as follows: 1) Power spectra of the EEG-records prior to stimulus were evaluated. 2) Instantaneous frequency characteristics of single EPs were obtained by the Fourier transform. 3) Distribution of the amplitude maxima of the EP-frequency characteristics and the distribution of the EEG-spectral peaks were compared by plotting two types of histograms containing relevant spectral peaks before and after the stimulation. In a frequency range between 1–1000 Hz, the frequency distribution of the EP records from RF and IC were accumulated in narrow discrete frequency channels, whereas, the distribution of the spectral peaks of the EEG depicted frequency spread in broad channels. The frequency stabilization of the EP records from GEA, in the alpha frequency range, was also observed. This effect was described by a factor which we called as the Frequency Stabilization Factor. The results presented in this study showed that the frequency stabilization of the brain's electrical activity induced by sensory stimulation displayed a fluctuation leading to frequency stabilization factors between 0.95 and 5.00. The frequency stabilization and relevant power enhancement upon stimulation strongly support our contention that evoked potential results from the frequency stabilization of the spontaneous activity, triggered by stimulation.Supported by Grant No. TAG-345 of the Scientific and Technical Research Council of Turkey  相似文献

Thirty-one polymorphic decamer primers were selected to genotype 92 progenies from the cross between Yiben No.4, a monoembryonic diploid F1 hybrid of Citrus reticulata Blanco cv Huanongbendizao tangerine and C. ichangensis Swingle, and [Hamlin sweet orange + Rough lemon], an allotetraploid somatic hybrid of Citrus sinensis Osbeck cv. Hamlin and C. jambhiri Lush cv. Rough Lemon. χ2 (Chi-square) analysis of RAPD markers in the progenies indicated they were randomly transmitted from the four donor parents, without significant difference between the diploids and triploids. However, these progenies were clustered into three major groups using dendrogram constructed by UPGMA, skewed to three parents in certain degrees, 15 (13 triploids and 2 diploids) to Hamlin, 16 (9 and 7) to Yiben No. 4, and 61 (57 and 4) to [Hamlin sweet orange + Rough Lemon] from which genomic contribution was predominant in progenies, respectively  相似文献

DNA polymerase (POL) λ plays an important role during DNA repair and DNA nonhomologous recom-bination processes. A novel POL λ variant was cloned from a human liver cDNA library and named POL λ2 (GenBank Accession No. AY302442). POL λ2 has 2206 base pairs in length with an open reading frame of 1452 base pairs encoding a 482-amino-acids protein. Bioinformatics analysis reveals that POL λ2 spans 7.9 kb on human chromosome 10q24 and is composed of 8 exons and 7 introns. It has the specific domain of DNA polymerase X family-POL Xc at the C-terminus and BRCT domain at the N-terminus. POL λ2 was localized predominantly in nucleus in transfected L0-2 cells. It was expressed abundantly in liver and testis, weakly in ovary, and undetectably in other tested human tissues. In comparison with the expression ratio between POL λ and POL λ2 in normal liver tissues and hepato-cellular carcinoma (HCC) adjacent tissues, the ratio was aberrant in 80% of those 15 HCC specimens examined due to the up-regulated expression of POL λ. This abnormality might be involved in hepato-carcinogenesis. The recombinant POL λ2 with His-tag was expressed as a soluble active protein in E. coli BL21 (DE3)CONDON Plus and purified by Ni-NTA resin and then desalted by Superdex-75 chro-matography in an FPLC system. The analysis using isotope α-32P-dCTP incorporation in vitro showed that the purified recombinant POL λ2 exhibited DNA polymerase activity.  相似文献

Summary Using operant conditioning methods, the pigeon's wavelength discrimination abilities were assessed in two experiments to generate discrimination functions. Both these functions showed three minima at 460, 530 and 595 nm. In the second wavelength discrimination experiment, extending measurements into the UV spectral region, pigeons also maintained good discrimination between wavelengths within the UV range tested. A fourth minimum was indicated at the lower end of the spectral range tested (365–385 nm). The results point to the complexity of the pigeon's chromatic system, which must be at least tetrachromatic, probably pentachromatic.Abbreviations S+ positive stimulus - S– negative stimulus We thank the technical staff of the Psychology Department at Durham University for their assistance and. in particular, Mr. D. L6 skeletal muscle myoblasts stably overexpressing glucose transporter GLUT1 or GLUT4 with exofa- cial myc-epitope tags were characterized for their response to insulin. In clonally selected cultures, 2-deoxyglucose uptake into L6-GLUT1myc myoblasts and myotubes was linear within the time of study. In L6-GLUT1myc and L6-GLUT4myc myoblasts, 100 nmol/L insulin treatment increased the GLUT1 content of the plasma membrane by 1.58±0.01 fold and the GLUT4 content 1.96±0.11 fold, as well as the 2-deoxyglucose uptake 1.53±0.09 and 1.86±0.17 fold respectively, all by a wortmannin-inhibitable manner. The phosphorylation of Akt in these two cell lines was increased by insulin. L6-GLUT1myc myoblasts showed a dose-dependent stimulation of glucose uptake by insulin, with unaltered sensitiv- ity and maximal responsiveness compared with wild type cells. By contrast, the improved insulin re- sponsiveness and sensitivity of glucose uptake were observed in L6-GLUT4myc myoblasts. Earlier studies indicated that forskolin might affect insulin-stimulated GLUT4 translocation. A 65% decrease of insulin-stimulated 2-deoxyglucose uptake in GLUT4myc cells was not due to an effect on GLUT4 mobi- lization to the plasma membrane, but instead on direct inhibition of GLUT4. Forskolin and dipyridamole are more potent inhibitors of GLUT4 than GLUT1. Alternatively, pentobarbital inhibits GLUT1 more than GLUT4. The use of these inhibitors confirmed that the overexpressed GLUT1 or GLUT4 are the major functional glucose transporters in unstimulated and insulin-stimulated L6 myoblasts. Therefore, L6-GLUT1myc and L6-GLUT4myc cells provide a platform to screen compounds that may have differ- ential effects on GLUT isoform activity or may influence GLUT isoform mobilization to the cell surface of muscle cells.  相似文献

Precise structural identification of photosynthetic polar glycerolipids in microalga Tetraselmis chuii has been established using Ultra Performance Liquid Chromatography-Electrospray ionization-Quadrupole-Time of Flight Mass Spectrometry (UPLC-ESI-Q-TOF-MS) by direct analysis of the total lipids extract. The mass spectrometry was performed in reflective time-of-flight using electron spraying ionization in both positive and negative modes. The structural determination was based on the characteristic product ions yielded by different glycerolipids under ESI-MS/MS mode, and confirmed the molecular species by the carboxylate anions produced by glycerolipids in the negative mode. As a result, more than 40 lipid molecular species, including 11 monogalactosyldiacylglycerols (MGDG), 7 digalactosyldiacylglycerols (DGDG), 16 sulfoquinovosyldiacylglycerols (SQDG), and 9 phosphatidylglycerols (PG), were detected in Tetraselmis chuii, which had never been identified before in this microalga. Furthermore, some intact lipid molecules with hydroxylated fatty acids that could not be detected by the traditional GC-MS method were found this time, providing novel information for the photosynthetic lipidome of Tetraselmis chuii. Comparative studies on fatty acids at the sn-2 position showed that SQDG and MGDG are dominantly biosynthesized through the prokaryotic pathway, PG is a typically mixed biosynthetic pathway, while DGDG is somewhat peculiar with C14:0 and C16:0 at its sn-2 position. This method could provide a full structural profile of intact photosynthetic lipid molecular species, which may be applied to study the physiological and ecological functions of lipid by monitoring their individual changes.  An intracellular recording made from the retinal luminosity horizontal cell (LHC) demonstrated that repetitive red flashes enhanced the cell's responsiveness to red stimulus and depressed its responsiveness to green stimulus and that repetitive green flashes suppressed the cell's red response but produced little change in its green response. Based on the idea that the spectral plasticity of LHCs may reflect some synaptic efficacy changes between the LHC and various cones, a self-organizing system is proposed to investigate the possible manner of information processing and storage within the synapses. The results of model analysis suggest that the stimulus-pattern-related spectral plasticity is attributable to the excitatory interaction within the same kinds of synapses and the inhibitory interaction between different kinds of synapses. This system is able to encode and store the history of signal transmission in a graded and cumulative fashion. Background

The perception of global form requires integration of local visual cues across space and is the foundation for object recognition. Here we used magnetoencephalography (MEG) to study the location and time course of neuronal activity associated with the perception of global structure from local image features. To minimize neuronal activity to low-level stimulus properties, such as luminance and contrast, the local image features were held constant during all phases of the MEG recording. This allowed us to assess the relative importance of striate (V1) versus extrastriate cortex in global form perception.

Methodology/Principal Findings

Stimuli were horizontal, rotational and radial Glass patterns. Glass patterns without coherent structure were viewed during the baseline period to ensure neuronal responses reflected perception of structure and not changes in local image features. The spatial distribution of task-related changes in source power was mapped using Synthetic Aperture Magnetometry (SAM), and the time course of activity within areas of maximal power change was determined by calculating time-frequency plots using a Hilbert transform. For six out of eight observers, passive viewing of global structure was associated with a reduction in 10–20 Hz cortical oscillatory power within extrastriate occipital cortex. The location of greatest power change was the same for each pattern type, being close to or within visual area V3a. No peaks of activity were observed in area V1. Time-frequency analyses indicated that neural activity was least for horizontal patterns.

Conclusions

We conclude: (i) visual area V3a is involved in the analysis of global form; (ii) the neural signature for perception of structure, as assessed using MEG, is a reduction in 10–20 Hz oscillatory power; (iii) different neural processes may underlie the perception of horizontal as opposed to radial or rotational structure; and (iv) area V1 is not strongly activated by global form in Glass patterns.  相似文献

The method of isoelectric focusing in polyacrylamide gel was used to separate HGPRT isoenzymes in crude hemolysates of human and rat erythrocytes. HGPRT from erythrocytes of a normal human male donor consistently revealed three peaks of activity. Their mean isoelectric points, using pH 5–7 range ampholytes, were, peak I, pI 6.00; peak II, pI 5.83; and peak III, pI 5.71. Peak III was wide and tailed. It always had a shoulder with a mean pI of 5.62. HGPRT from rat erythrocytes revealed two peaks of activity, corresponding to isoelectric points of 5.90 and 5.80. Summary The crustaceanDaphnia magna responds to a flash of light with a ventral rotation of its compound eye; this behavior is termed eye flick. We determined the spectral sensitivity for the threshold of eye flick in response to light flashes having three different spatial characteristics: (1) full-field, extending 180° from dorsal to ventral in the animal's field of view; (2) dorsal, 30° wide and located in the dorsal region of the visual field; (3) ventral, same as dorsal but located ventrally. All three stimuli extended 30° to the right and to the left of the animal's midplane. We found that spectral sensitivity varies with the spatial characteristics of the stimulus. For full-field illumination, the relative sensitivity was maximal at 527 nm and between 365 nm and 400 nm, with a significant local minimum at 420 nm. For the dorsal stimulus, the relative sensitivity was greatest at 400 nm, but also showed local maxima at 440 nm and 517 nm. For the ventral stimulus, the relative sensitivity maxima occurred at the same wavelengths as those for the full-field stimulus. At wavelengths of 570 nm and longer, the responses to both dorsal and ventral stimuli showed lower relative sensitivity than the full-field stimulus. No circadian or other periodic changes in threshold spectral sensitivity were observed under our experimental conditions. Animals which had their nauplius eyes removed by means of laser microsurgery had the same spectral sensitivity to full-field illumination as normal animals. Our results are discussed in terms of our current knowledge of the spectral classes of photoreceptors found in theDaphnia compound eye.  相似文献
