In this study, we reported the isolation and analysis of new polymorphic microsatellites in mungbean (Vigna radiata (L.) Wilczek). Twelve out of 210 primer pairs screened in 30 mungbean accessions gave polymorphism. The polymorphic markers detected two to three alleles per locus with an average of 2.08. Observed heterozygosity varied from 0 to 0.133, while expected heterozygosity ranged from 0.095 to 0.498. Tests for Hardy-Weinberg equilibrium (HWE) and pairwise linkage disequilibrium of the polymorphic loci revealed that all loci except MB-SSR14 significantly departed from HWE and four pairwise combinations, viz. MB-SSR14 vs. MB-SSR42, MB-SSR42 vs. MB-SSR87, MB-SSR114 vs. MB-SSR121, and MB-SSR175 vs. MB-SSR231 significantly deviated from linkage disequilibrium. The markers are being used to study genetic diversity and genome mapping of mungbean.  相似文献

Greengram or mungbean (Vigna radiata (L.) Wilczek), which originated in the Indian sub-continent, is an important pulse crop in India. Genetic diversity and patterns of variation among 111 accessions representing the better agronomic types from the entire collection of the National Bureau of Plant Genetic Resources were studied. Wide variation in morphological and agronomic traits viz. number of pods per plant, yield per plant, 100-seed weight, fruit setting capacity, flowering period, maturity, number of pod bearing peduncles, plant height, number of primary branches, length of branch, nodulation and leafiness was observed. Grain yield was found to be significantly correlated with 100-seed weight and pod length. The data were subjected to cluster and principal components analysis. The accessions were grouped into six discrete and well-defined clusters. The study demonstrated the patterns of variation at the population level. The multivariate analysis was useful in identifying a group of accessions with yield enhancing traits within a highly diverse group of accessions and their potential value in greengram improvement is suggested.  相似文献

Abstract. Storage parenchyma, vascular parenchyma and phloem companion cells are found adjacent to sieve tubes in the vascular bundles of cotyledon leaves of mung bean ( Vigna radiata L.) seedlings. The paramural bodies of storage parenchyma cells are characterized by flask shaped invaginations of the plasmalemma whereas the plasmalemmasomes of the adjacent vascular parenchyma and companion cells consist of numerous finger-like evaginations which are not enclosed in plasmalemma pockets. Phloem associated transfer cells are not present and it is suggested that the fine tubular plasmalemmasomes may act as the interface between apoplast and symplast in the transport of rapidly mobilized reserves during germination. Tubular structures observed within the protoplasts of storage cells close to vascular tissue were also observed in vascular parenchyma and companion cells between the plasmalemmae and cell walls.  相似文献

Summary Interspecific hybrids of the mungbean, Vigna radiata (L.) Wilczek (2n=22) and V. glabrescens (2n=44) were generated with the aid of embryo culture. V. glabrescens x V. radiata hybrids were recovered via germination of the immature embryos. Reciprocal hybrids were obtained via shoot formation from embryonic callus. The authenticity of the hybrids was determined by morphological characteristics, chromosome number, and isozyme patterns. The hybrids were highly sterile upon selfing, but backcrossing to the diploid parent yielded viable seeds. Some of the plants resembled the diploid parent morphologically while others resembled neither parent. The backcross plants were sufficiently fertile to give a large number of mature, selfed seeds. Plants obtained differed morphologically and in their isozyme patterns from either parent, indicating introgression. These progeny populations will be used as bridging materials to transfer pest resistance from the wild tetraploid to the cultivated mungbean.  相似文献

The present work reports the isolation and characterization of new polymorphic microsatellites in mung bean (Vigna radiata L.). Of 93 designed primer pairs, seven were found to amplify polymorphic microsatellite loci, which were then characterized using 34 mung bean accessions. The number of alleles ranged from two to five alleles per locus with an average of three alleles. Observed and expected heterozygosity values ranged from 0 to 0.088 and from 0.275 to 0.683, respectively. All seven loci showed significant deviations from Hardy–Weinberg equilibrium, whereas only one pairwise combination (GBssr‐MB77 and GBssr‐MB91) exhibited significant departure from linkage disequilibrium. These newly developed markers are currently being utilized for diversity assessment within the mung bean germplasm collection of the Korean Gene Bank.  相似文献

Ten mungbean cultivars were evaluated for their resistance to iron deficiency in view of chlorosis symptoms, plant growth and seed yield under field conditions on a calcareous soil in Thailand. The KPS2 cultivar was highly susceptible; the KPS1, PSU1 and Pag-asa 1 cultivars were somewhat susceptible; the VC1163B cultivar was moderately tolerant; the CN36, CN60, UT1 and CNM-I cultivars were tolerant; and the CNM8509B cultivar was very tolerant to iron deficiency. Foliar application of a solution of 5 g L-1 ferrous sulphate was effective in correcting chlorosis that was induced by iron deficiency, and it enhanced both the growth and the yield of susceptible cultivars. Compared with the susceptible cultivar KPS2, the tolerant cultivar UT1 had a greater ability to lower the pH of the nutrient solution in response to iron deficiency. The root-associated Fe3+-reduction activity of UT1 that had been grown in -Fe medium was similar to that of the plants grown in +Fe medium when the acidification of the medium occurred. Acidification of the medium in response to iron deficiency might contribute to the efficient solubilization of iron from calcareous soils, and it related more closely to the resistance to iron deficiency than Fe3+ reduction by roots in mungbean cultivars.  相似文献

The morphogenetic responses of seedling explants of mung bean (Vigna radiata L. Wilczek cv ML-5) were studied in vitro. Direct induction of shoots/plants was possible from shoot tip, cotyledon and cotyledonary node explants. Dedifferentiation of the explants viz; Shoot tip, cotyledons, cotyledonary node, primordial leaves and roots was obtained on basal medium supplemented with auxin and cytokinin. Shoot regeneration was limited to primary calli while rhizogenesis was of common occurrence in established calli. In addition to differences in hormonal requirements, the various explants showed preferential growth in different basal media.  相似文献

Summary The inheritance of the dwarf plant type was studied in blackgram (V. mungo (L.) Hepper). Type 9 has erect plant type with normal internode length. The mutant line, EMSD has reduced internode length. The F1, F2 and F3 generations of a cross between Type 9 and EMSD and its reciprocal were studied. Greengram [Vigna radiata (L.) Wilczek], also known as mung bean, widely cultivated in a large number of countries, is an important pulse crop of Asia and is considered one of the ancestral species of the genus Vigna. Since yields of greengram have remained low across subtropical and tropical Asia, it is important to estimate genetic diversity in existing cultivars in order to see if the lack of genetic variability might be a constraining factor. In this study, 32 Indian cultivars of greengram were subjected to random amplified polymorphic DNA (RAPD) analysis using 21 decamer primers. A total of 267 amplification products were formed at an average of 12.71 per primer with an overall polymorphism of 64%. The extent of polymorphism was moderate to low. Jaccard similarity coefficient values ranged from 0.65 to 0.92. The cluster analysis resulted in mainly three clusters revealing greater homology between cultivars released from the same source. The results of principal components analysis also substantiated this conclusion. The close genetic similarity between the cultivars could be explained due to the high degree of commonness in their pedigrees. The narrow genetic base of the greengram cultivars revealed in the present analysis emphasises the need to exploit the large germplasm collections having diverse morphoagronomic traits in cultivar improvement programs. We studied the effects of temperature, carbon dioxide and abscisic acid on mung bean (Vigna radiata). Plants were grown under 26/22°C or 32/28°C (16?h?light/8?h?dark) at 400 or 700?μmol?mol?1 CO2 and received ABA application of 0 or 100?μl (10?μg) every other day for three weeks, after eight days of initial growth, in growth chambers. We measured 24 parameters. As individual factors, in 16 cases temperature; in 8 cases CO2; in 9 cases ABA; and as interactive factors, in 4 cases, each of temperature?×?CO2, and CO2?×?ABA; and in 2 cases, temperature?×?ABA were significant. Higher temperatures increased growth, aboveground biomass, growth indices, photochemical quenching (qP) and nitrogen balance index (NBI). Elevated CO2 increased growth and aboveground biomass. ABA decreased growth, belowground biomass, qP and flavonoids; increased shoot/root mass ratio, chlorophyll and NBI; and had little role in regulating temperature–CO2 effects.

Until now no 'early-methionine-labelled' (Em) proteins have been reported in the Fabaceae. To check whether a previously isolated low-molecular mass albumin from dry mung bean embryonic axes possibly corresponded to an Em-like protein, the protein was purified, sequenced and its cDNA clone isolated and characterized. N-terminal sequencing of cyanogen bromide cleavage products of the protein revealed homology with previously described Em-like proteins from other species. Analysis of cDNA clones encoding the mung bean Em protein revealed the presence of two classes of Em proteins and confirmed their homology to the previously characterized Em-like proteins. In vivo labelling and northern blot analysis further demonstrated that the mung bean protein is synthesized during early germination of the axes and that abscisic acid (ABA) extends its synthesis. It appears, therefore, that legumes also contain maturation-specific, ABA-responsive Em-like proteins.  相似文献

Summary The inheritance of resistance to mungbean yellow mosaic virus (MYMV) was studied in blackgram (Vigna mungo (L.) Hepper). The highly resistant donors Pant U-84 and UPU-2 and a highly susceptible line, UL-2, their F1's, F2's and backcrosses were grown with spreader located every 5 to 6 rows. The resistance was found to be digenic and recessive in all the crosses and free from cytoplasmic effect.  相似文献

Cytosolic fructose-1,6-P(2) (FBP) aldolase (ALD(c)) from germinated mung beans has been purified 1078-fold to electrophoretic homogeneity and a final specific activity of 15.1 micromol FBP cleaved/min per mg of protein. SDS-PAGE of the final preparation revealed a single protein-staining band of 40 kDa that cross-reacted strongly with rabbit anti-(carrot ALD(c))-IgG. The enzyme's native M(r) was determined by gel filtration chromatography to be 160 kDa, indicating a homotetrameric quaternary structure. This ALD is a class I ALD, since EDTA or Mg(2+) had no effect on its activity, and was relatively heat-stable losing 0-25% of its activity when incubated for 5 min at 55-65 degrees C. It demonstrated: (i) a temperature coefficient (Q(10)) of 1.7; (ii) an activation energy of 9.2 kcal/mol active site; and (iii) a broad pH-activity optima of 7.5. Mung bean ALD(c) is bifunctional for FBP and sedoheptulose-1,7-P(2) (K(m) approximately 17 microM for both substrates). ATP, ADP, AMP and ribose-5-P exerted inhibitory effects on the activity of the purified enzyme. Ribose-5-P, ADP and AMP functioned as competitive inhibitors (K(i) values=2.2, 3.1 and 7.5mM, respectively). By contrast, the addition of 2mM ATP: (i) reduced V(max) by about 2-fold, (ii) increased K(m)(FBP) by about 4-fold, and (iii) shifted the FBP saturation kinetic plot from hyperbolic to sigmoidal (h=1.0 and 2.6 in the absence and presence of 2mM ATP, respectively). Potent feedback inhibition of ALD(c) by ATP is suggested to help balance cellular ATP demands with the control of cytosolic glycolysis and respiration in germinating mung beans.  相似文献

Summary Selective intermating and pedigree selection methods were applied simultaneously to highly heterogeneous and heterozygous base populations of greengram in order to compare their relative efficacy in terms of evolving the number of productive lines as well as their production potential. Selection after two cycles of selective intermating was found to be a better method than traditional pedigree selection. The demerits of pedigree selection and merits of selective intermating are discussed. An osmotically (mannitol) tolerant callus line of Vigna radiata (L.) Wilczek has been isolated from callus cultures grown on modified PC-L2 medium supplemented with increasing concentrations of mannitol. The tolerance was stable and retained after growth in the absence of mannitol selection for 2 months. The growth of the tolerant line, in the presence of mannitol (540 mol m-3) was comparable to that of a sensitive callus line growing in the absence of mannitol. This line not only grew well on media containing up to 720 mol m-3 mannitol, but also required 450 mol m-3 mannitol for its optimal growth. Osmotically tolerant callus also showed increased tolerance to NaCl (0–250 mol m-3) stress as compared to sensitive callus. Accumulation of Na+ was lower, and the level of K+ was more stable in osmotically tolerant than in sensitive calli, when both were exposed to salt. The free proline content of both tolerant and sensitive calli increased on media supplemented with mannitol or NaCl. Five strains of Streptomyces sp. were evaluated in vitro for their ability of inhibiting the mycelial growth of Macrophomina phaseolina, the causal agent of root rot of mung bean (Vigna radiata L.). Among the Streptomyces sp. strains tested, PDK showed the maximum in vitro inhibition of mycelial growth of M. phaseolina and recorded an inhibition zone of 21?mm. The strains CBE, MDU, SA and ANR recorded inhibition zones of 18, 16, 13 and 11?mm, respectively. These Streptomyces sp. strains were tested for their growth-promoting efficiency on mung bean seedlings. Among them, CBE and PDK recorded the maximum increase in shoot length, root length and seedling vigour compared with control, followed by MDU. Three Streptomyces sp. strains (CBE, MDU and PDK) that showed higher levels of inhibition of growth of M. phaseolina in dual culture assay and plant growth-promoting activity were tested for their biocontrol activity against root rot under greenhouse and field conditions. Seed treatment or soil application with powder formulation of Streptomyces sp. strains CBE, MDU and PDK was effective in controlling root rot disease; but, combined application through seed and soil increased the efficacy in both the greenhouse and field trials. Among the treatments, seed treatment plus soil application with powder formulation of Streptomyces sp. strain CBE proved to be most effective, which reduced the root rot incidence from 26.8% (with non-bacterised seeds) to 4.0% in Trial I and from 32.0 to 4.9% in Trial II. The above treatment recorded the highest yield in both the field trials, and the yield increase was 78 and 74% over control in Trial I and Trial II, respectively. Isozyme analysis of the Streptomyces sp.-treated plants indicates that seed treatment plus soil application strongly induce the activities of peroxidase (PO-1 and PO-2) and polyphenol oxidase (PPO-2 and PPO-3) in mung bean. Among the three strains tested, Streptomyces sp. strain MDU- treated plants showed higher levels of activities of PO and PPO. Based on the above findings, it can be concluded that both the direct inhibition of pathogen and induced resistance might be involved in the control of root rot of mung bean by Streptomyces sp.  相似文献

A comparative chromosomal evaluation was carried out between Vigna unguiculata (cowpea) and V. radiata (mung bean) with chromomycin A3 (CMA3)/4’,6-diamidino-2-phenylindole (DAPI) banding and fluorescent in situ hybridization (FISH) using 5S/45S ribosomal DNA (rDNA) probes. Both species had symmetric karyotypes (2n = 22), with prevalence of centromeres in chromosomes at median (m) and submedian (sm) regions and chromosomes ranging in size from 2.1 to 1.25 μm (V. unguiculata) and 2.18 to 0.93 μm (V. radiata). Three different banding patterns were identified for V. unguiculata: CMA3+/DAPI0, CMA3++/DAPI, and CMA3+/DAPI. The CMA3+/DAPI0 bands were observed in the pericentromeric regions of all chromosomes, while the CMA3++/DAPI and CMA3+/DAPI bands were co-localized with the 45S rDNA in the subtelomeric position (chromosomes B, G, and D, J, respectively) and in the proximal position in chromosome F. Two pairs of chromosomes (D and I) bearing interstitial 5S rDNA have been also identified. Vigna radiata displayed CMA30/DAPI+ bands distributed in the centromeric region of chromosomes B, C, and F, while CMA3++/DAPI bands were co-localized with the 45S rDNA sites in the subtelomeric position of the short arm in the F and K chromosome pairs. Three pairs of 5S rDNA sites were identified, the first in the proximal region of the long arm in chromosome E and the two others in the proximal and subterminal positions in the long arm of chromosome J. These data highlight some divergences regarding the amount and composition of the heterochromatin in both species, allowing the identification of individual chromosomes in V. unguiculata and V. radiata, and a comparison with other members of the Phaseoloid clade.  相似文献

The present experiment was undertaken to examine whether shading affects the critical boron (B) concentration for leaf blade elongation in black gram. Six days after germination (D6), black gram seedlings were transferred to 8 pairs of pots containing basal nutrient solution: one pot of each pair contained 1 M H3BO3 and the other 10 M H3BO3. On D10, one day after the emergence of the first trifoliolate leaf blade (TF1) and one day before the emergence of TF2, four pairs of pots were shaded, decreasing the light intensity they received in the glasshouse to about 35% of full sunlight compared with 70% received by the unshaded plants. The response to B supply of dry matter (DM) and elongation rate (LBER) of TF2 were less pronounced in shaded than in unshaded plants. Critical B concentrations for LBER in TF2 of black gram were 10 mg B kg-1DM in shaded and 15 mg B unshaded plants, suggesting that shading may have decreased the B requirement for LBER. Thus the present results suggest that light may need to be considered when setting critical values for the diagnosis of B deficiency in black gram.  相似文献

Eight natural populations ofVigna radiata var.sublobata— wild relative of cultivated urd (V. mungoj and mung (V. radiata,) beans—were sampled from different ecozones of Palney Hills, an eastward offshoot of Western Ghats of Tamilnadu, India. Photosynthetic efficiency, protein content, seed weight, and amino acid composition were determined for it and the cultigens. Some populations ofV. radiata var.sublobata are as good as or even superior to the cultigens. The wild relative is a potential donor of desirable traits to urd and mung beans.  相似文献

Summary The connections and structural inter-relations of dictyosomes and endoplasmic reticulum (ER) in cotyledons of germinating mung beans were studied using thick (0.3 m) sections of aldehyde fixed, zinc iodide-osmium tetroxide (ZIO) impregnated tissue. The sections were examined by conventional (100 kV), rather than high voltage, transmission electron microscopy.Continuity of cisternal ER with tubular ER was confirmed and a direct connection of tubular ER totrans dictyosome cisternae was observed as were GERL networks associated withtrans dictyosome cisternae.Dictyosomes also gave rise to an extensive system of very fine tubules (10–20 nm diam) which have not been described previously in plant tissue. These tubules, which originated at thetrans dictyosome face, extended throughout the cytoplasm and were found connected to cisternal ER and tubular ER.The implications of these observations are discussed with regard to present ideas concerning endomembrane flow and protein sorting by the Golgi apparatus.  相似文献
