Summary Wheat accessions lacking some of the - and -gliadin components encoded by the Gli-1 loci on the short arm of chromosome 1D in bread wheat and chromosome 1A in durum wheat were studied by two-dimensional polyacrylamide gel electrophoresis and restriction fragment analysis. Digested genomic DNAs of normal and null forms were probed with a cDNA clone related to -/-gliadins and with a genomic clone encoding an LMW subunit of glutenin. The hybridisation patterns with the -/-gliadin probe were similar to those of cvs Chinese Spring and Langdon used as standards for bread and durum wheats, respectively, but several restriction fragments located on the 1D chromosome of bread wheat and the 1A chromosome of durum wheat were absent in the null forms. In addition, specific LMW glutenin fragments encoded by the same chromosomes were also absent in the null forms, suggesting that simultaneous deletions of blocks of genes for both -/-gliadins and LMW glutenins had occurred. Comparisons of the protein and RFLP patterns enabled some proteins to be mapped to specific restriction fragments.  相似文献

A novel syrup containing neofructo-oligosaccharides was produced from sucrose (Brix 70) by whole cells of Penicillium citrinum. The efficiency of fructo-oligosaccharides production was more than 55% and those of the main carbohydrate components, 1-kestose (Fruf 21Fruf 21 Glc), nystose (Fruf 21Fruf 21 Fruf 21 Glc) and neokestose (Fruf 26 Glc12 Fruf), were 22, 14 and 11%, respectively.  相似文献

Summary The B factors of Schizophyllum commune are of 3 classes: The high recombining class I has 7 alleles and 7 alleles; the low recombining classes are class II with 7 allels and probably 2 alleles and class III with probably 2 (or also 2) alleles and 7 allels. A fourth hypothetical class (-) was not found and either does not exist or is indistinguishable from class III by the tests employed. When a flat figure of uniform color and with an elliptic contour is slowly rotated around an axis orthogonal to the plane of the image, an observer set in the frontal position will perceive it first as a rotating ellipse. After a few seconds of inspection, the ellipse appears to deform with an amoeba-like movement until it appears as a rigid, circular disk tilting back and forth in 3-D space; finally it is seen as a rotating ellipsoid tilted in depth at a constant inclination angle with respect to the rotating platform. In an attempt to provide an explanation for the apparent ellipsoid, the authors present a mathematical model based on an hypothesis of velocity differences minimization, successfully used to describe other stereokinetic phenomena, such as the rotating cone and the tilted bar. This technique does not make use of Ullman's ridigity hypothesis for extracting depth from 2-D moving stimuli. A comparison with experimental results supports the validity of the model.  相似文献

Zusammenfassung Kegel und Säulen der Schwanen-Eischale hinterlassen am Querschliff nach Entkalkung mit EDTA organisches (mucoproteides) Material als ein zusammenhängendes Gerüst, das sich mit Thionin metachromatisch färbt; ohne Demineralisierung oder wenigstens Anätzung bleibt Thionin an Schliffen und Bruchkanten der Schale wirkungslos. Das Lichtmikroskop zeigt an Schliffen nichts von dem organischen Material, es wurde während des Kristallwachstums fein zerteilt in Gitterlücken des Schalencalcits eingeschlossen. Es findet sich am stärksten angehäuft an den äueren und inneren Oberflächen der Kristall-individuen. In den Kegeln ist das Gerüst radial ausgebildet als die Loculi der Keile, und konzentrisch geschichtet, entsprechend den Lagen der Globularinklusionen, um deren jede herum Verdichtung der organischen Substanz statthat. In den inneren Säulen folgt das organische Gerüst dem Rhombenmuster; die äueren Säulen sind arm an organischer Substanz, hier verbleibt nach der Entkalkung eine dünne laterale Oberflächenschicht.
Summary The cones and columns of the swans egg shell leave behind after decalcification with EDTA an organic (mucoproteid) material in form of a continuous frame work stainable metachromatically with thionine. Without demineralisation or at least etching, thionine proves ineffectual in ground sections or breaking edges of the shell. In ground sections the light microscope demonstrates nothing of the organic material: it was inclosed during the crystal growth in submicroscopical lattice gaps of the calcite individuals. The organic material is chiefly accumulated in the outer and inner surfaces of the crystals. In the cones the organic frame work is developed radially as the loculi of the wedges and concentrically layered corresponding with the globular inclusions, concentrated in the circumference of each. In the inner columns the organic material follows to the rhomb pattern. The outer columns after decalcification only leave behind a thin lateral organic sheath.

It is shown in the preceding paper that neurons with two-dimensional spatio-temporal properties to linear acceleration behave like one-dimensional rate sensors: they encode the component of angular velocity (associated with a rotating linear acceleration vector) that is normal to their response plane. During off-vertical axis rotation (OVAR) otolith-sensitive neurons are activated by the gravity vector as it rotates relative to the head. Unlike one-dimensional linear accelerometer neurons which exhibit equal response magnitudes for both directions of rotation, two-dimensional neurons can be shown to respond with unequal magnitudes to clockwise and counterclockwise off-vertical axis rotations. The magnitudes of the sinusoidal responses of these neurons is not only directionally selective but also proportional to rotational velocity. Thus, responses from such two-dimensional neurons may represent the first step in the computations necessary to generate the steady-state eye velocity during OVAR. An additional step involving a nonlinear operation is necessary to transform the sinusoidally modulated output of these neurons into a signal proportional to sustained eye velocity. Similarly to models of motion detection in the visual system, this transformation is proposed to be achieved through neuronal operations involving mathematical multiplication followed by a leaky integration by the velocity storage mechanism. The proposed model for the generation of maintained eye velocity during OVAR is based on anatomical and physiological properties of vestibular nuclei neurons and capable of predicting the experimentally observed steady-state characteristics of the eye velocity.  相似文献

In this paper we analyse a stochastic model for invertebrate predation taking account of the predator's satiation. This model approximates Holling's hungry mantid model when handling time is negligible (see Part I). For this model we derive equations from which we can calculate the functional response and the variance of the total catch. Role of mitochondria in ethanol tolerance of Saccharomyces cerevisiae   总被引:7,自引:0,他引:7  
The presence of active mitochondria and oxidative metabolism is shown to be essential to maintain low inhibition levels by ethanol of the growth rate (), fermentation rate (v) or respiration rate () of Saccharomyces cerevisiae wild type strain S288C. Cells which have respiratory metabolism show K i (ethanol inhibition constant) values for , v and , higher (K i>1 M) than those of petite mutants or grande strains grown in anaerobiosis (K i=0.7 M). In addition, the relationship between or v and ethanol concentration is linear in cells with respiratory metabolism and exponential in cells lacking respiration. When functional mitochondria are transferred to petite mutants, the resulting strain shows K i values similar to those of the grande strain and the inhibition of and v by increasing ethanol concentrations becomes linear.  相似文献

Zusammenfassung Uferschwalben kehren aus den afrikanischen Winterquartieren in Trupps beiderlei Geschlechts zurück. Erste Beringungsergebnisse belegen, daß zunächst mehrjährige, vermutlich untereinander bekannte Vögel eintreffen, die den Brutplatz aus vergangenen Brutperioden her kennen. Die Masse der später ankommenden Vögel dürfte weitgehend aus einjährigen oder ortsfremden Uferschwalben bestehen, die sich größtenteils erst während der Paarbildung persönlich kennenlernen. Der anfängliche Schwarmzusammenhalt der nacheinander eintreffenden Trupps führt zur Bildung von Subkolonien, die für Brutplätze ab einer bestimmten Größenordnung typisch sind. Uferschwalben- gründen nacheinander mehrere Reviere, d. h. sie besetzen Steilwandbereiche, in denen sie ausschließlich mit den Füßen eine Röhre oder Mulde graben, singen und Bogenflüge starten. Bis auf singende oder bekannte werden Artgenossen im Revier geduldet. Uferschwalben- suchen besetzte Reviere auf. Ohne Röhrenbindung verhalten sie sich still und unauffällig, ihre Grabungsaktivitäten sind von untergeordneter Bedeutung. Die Bindung an ein bestimmtes Revier entwickelt sich individuell verschieden und entscheidet über den Abschluß des Röhrenbaues (Herstellung der Nistkammer). Reviere ohne dauerhafte -Bindung werden von den aufgegeben. Aktivitäten, die auf wachsende Revierbindung eines hindeuten, sind: häufige oder/und länger dauernde Aufenthalte des in einem besetzten Revier und sporadisches Mitgraben; aggressives Verhalten gegenüber Artgenossen (i. d. R. fremde ), die im Revier landen wollen; gemeinschaftlicher, leiser Gesang von und im Röhrenbereich. Aktivitäten, die für eine vollzogene Paarbildung sprechen, sind: Fertigstellen der Röhre durch Grabung der Nestkammer; längere gemeinsame Aufenthalte innerhalb und außerhalb der Röhre; Voranfliegen des beim Röhrenanflug; Übernachten von und in der Röhre; Nestbau; ausdauernde Verfolgungsflüge während der Kopulationsphase. Die Paarbildung ist demnach ein individueller Prozeß, bei dem die Aktivitäten der im Revier als Werbung, die der als Revierwahl interpretiert werden.
On pair-formation in the Sand Martin,Riparia riparia
Summary European Sand Martins arrive at their breeding sites in flocks of usually unmated and . Ringing results of a large population in NW-Germany and own observations indicate that the first flocks about a dozen individuals with an approximately balanced sex ratio appear at traditional breeding places and consist of older, experienced resident birds (presumably acquainted with one another). The birds arriving over the next several weeks are mainly first-year or non-resident individuals. The flocks arrive separately in areas with suitable sandcliffs, synchronize the pair-formation activities and avoid disturbances among paired and unpaired birds. This behaviour causes the formation of subcolonies, which are typical for all densely occupied breeding places. Each settles on a fixed area on the sandcliff (territory) in order to excavate a burrow, to sing the territory-song (fig. 2 b) and to perfor the territory-circle-flight (fig. 2 c, 4 a). Silent birds (normally ) are welcomed or tolerated by the resident . The sexes are monomorphic and therefore courtship displays of the are non-aggressive until establishment of pair-bonds. Only intruding singing or individually known neighbouring are driven away, usually at early stages of territory occupation. Unmated are normally shy and very sensitive to protracted disturbances. visit several occupied territories of the colony (fig. 1–3) in order to choose a burrow. leave territories which do not attract a . They settle new territories on the sandcliff, causing a surplus of burrows compared to breeding pairs in the colony. The seed storage globulins from sixHelianthus and four hybrids were studied using mono and bidimensional gel SDS electrophoresis (+ 2 mercaptoethanol). The polypeptide composition of each subunit was determined. Different pairs are specifically expressed according to the species studied. Three typical patterns were discriminated. All the studied species exhibit five subunits: two of them are expressed in all the species (11 and 22). The subunit corresponding to the 11 pair is present inH. petiolaris and in the three populations ofH. annuus studied. The 2b2 pair is common toH. annuus andH. argophyllus. H. petiolaris presents two specific 2a2 and 44 pairs andH. annuus a specific 33 pair. InH. argophyllus 11 33 or 44 are never observed but are replaced by 13 and 31 pairs. Some globulins, poorly represented, are of forms but present chains of higher molecular weights (in the range 54–56 kDa). Expressing variations in the banding patterns between these species by the use of a similarity index reveals complete identity between the three populations ofH. annuus. Identity between the twoH. petiolaris studied is also observed.H. annuus andH. argophyllus appear to be closer to each other thanH. petiolaris concerning the seed storage globulins.  相似文献

Summary Films of cleaving embryos of the axolotl,Ambystoma mexicanum, taken by the double camera technique, were used in order to arrive at a more detailed staging based on quantitative criteria. Drawings were made of the animal hemispheres just prior to the start of each cleavage cycle from the 6th to the 15th cleavage. The number of cells intersected either by the (apparent) egg diameter (up to the 10th cleavage) or by half the diameter (from the 10th cleavage onwards) was determined. The cell numbers for each cleavage cycle did not overlap with those of the previous or succeeding cycles. On the basis of these cell numbers, in place of the four Harrison stages 6–9, 10 successive stages were established each of which corresponds to one cleavage cycle.  相似文献

Liver homogenate-supernatants from most Japanese exhibit an atypical pH optimum for ethanol oxidation at pH 8.8 instead of 10.5, the typical pH-activity optimum. It has been proposed that atypical livers contain alcohol dehydrogenase isozymes with 2 subunits while typical livers contain isozymes with 1 subunits, both produced by the ADH 2 gene. Because it is difficult to differentiate the atypical ADH2 2-2 phenotype from the ADH2 2-1 phenotype by starch gel electrophoresis, an agarose isoelectric focusing procedure was developed that clearly separated the atypical Japanese livers into two groups, A1 and A2. The isozymes in A1 and A2 livers were purified. Type A1 livers contained a single isozyme with an atypical pH-rate profile; it was designated 22. Three isozymes were isolated from A2 livers, two of which corresponded to 11 and 22. A third, absent from the typical and the atypical A1 livers, had an intermediate mobility; it was designated 21. Type A1 livers are, therefore, the homozygous ADH2 2-2 phenotype, and type A2 livers, the heterozygous ADH2 2-1 phenotype. The ADH2 2-2 phenotype was found in 53% of 194 Japanese livers, and the ADH2 2-1 phenotype, in 31%. Results of this investigation demonstrate that exposure to 17 -estradiol differentially and significantly regulates cortical nerve cell outgrowth depending on the cortical region. Parietal and occipital neurons treated with 1 nM 17 -estradiol showed a greater magnitude of neuronal outgrowth whereas outgrowth of temporal cortex neurons was decreased in the presence of 1 nM 17 -estradiol. Frontal cortex neurons showed a consistent enhancement of neuronal outgrowth that did not reach statistical significance. The dose response profile for 17 -estradiol regulation of the macromorphological features exhibited a bimodal dose response relationship whereas the dose response profile for 17 -estradiol regulation of the micromorphological features exhibited a dose response more characteristic of an inverted V-shaped function. An antagonist to the NMDA receptor antagonist, AP5, abolished the growth promoting effect of 17 -estradiol whereas the nuclear estrogen receptor antagonist ICI 182,780 did not. Lastly, neocortical neurons exposed to 17 -estradiol exhibited greater viability and survival than control neurons over a two week period. These data indicate that 17 -estradiol can enhance the growth and viability of select populations of neocortical neurons and that the growth promoting effects of 17 -estradiol can be blocked by an antagonist to the NMDA glutamate receptor and not by an antagonist to the estrogen nuclear receptor.  相似文献

Summary A cytochemical method for the detection of -galactosidase (-Gase) in mouse peritoneal macrophages was used to study the ultrastructural localization of this enzyme in these cells. It was found that the reaction product for -Gase was localized in the perinuclear cisternae, the endoplasmic reticulum, the Golgi complex, lysosomes, vesicles and on the cell surface of peritoneal macrophages from untreated C57BL mice. When examined by X-ray microanalysis the crystalline reaction product was found to contain bromine, an element present in the indolyl substrate which was used to identify -Gase. In log-phase cells of staphylococci, cultivated under high, non-lytic concentrations of penicillin G, there occurred a novel killing process hitherto hidden behind seemingly bacteriostatic effects. Two events are essential for the apprearance of this hidden death: (i) the failure of the dividing cell to deposit enough fibrillar cross-wall material to be welded together, and (ii) a premature ripping up of incomplete cross walls along their splitting system. Hidden death started as early as 10–15 min after drug addition, already during the first division cycle. It was the consequence of a loss of cytoplasmic constituents which erupted through peripheral slit-like openings in the incomplete cross walls. The loss resulted either in more or less empty cells or in cell shrinkage. These destructions could be prevented by raising the external osmotic pressure. In contrast, the conventional non-hidden death occurred only much later and exclusively during the second division cycle and mainly in those dividing cells, whose nascent cross walls of the first division plane had been welded together. These welding processes at nascent cross walls, resulting in tough connecting bridges between presumptive individual cells, were considered as a morphogenetic tool which protects the cells, so that they can resist the otherwise fatal penicillin-induced damages for at least an additional generation time (morphogenetic resistance system). Such welded cells, in the virtual absence of underlying cross-wall material, lost cytoplasm and were killed via ejection through pore-like wall openings or via explosions in the second division plane and after liberation of their murosomes, as it was the case in the presence of low, lytic concentrations of penicillin. Summary The and subunit of RNA polymerase are thought to be controlled by a translational feedback mechanism regulated by the concentration of RNA polymerase holoenzyme. To study this regulation in vivo, an inducible RNA polymerase overproduction system was developed. This system utilizes plasmids from two incompatibility groups that carry RNA polymerase subunit genes under lac promoter/operator control. When the structural genes encoding the components of core RNA polymerase (, and ) or holoenzyme (, , and 70) are present on the plasmids, induction of the lac promoter results in a two fold increase in the concentration of functional RNA polymerase. The induction of RNA polymerase overproduction is characterized by an initial large burst of synthesis followed by a gradual decrease as the concentration of RNA polymerase increases. Overproduction of RNA polymerase in a strain carrying an electrophoretic mobility mutation in the rpoB gene results in the specific repression of synthesis off the chromosome. These results indicate that RNA polymerase feedback regulation controls synthesis in vivo.  相似文献

Using the technique of allele-specific priming of the polymerase chain reaction (PCR), the C-T substitution in codon 39 was identified as the cause of -thalassaemia in an Irish family. Analysis of the restriction fragment length polymorphisms (RFLPs) in the -globin gene cluster established linkage of the -thalassaemia mutation to a particular -haplotype but indicated that a recombinational event had occurred in the paternal chromosome in the younger of two affected children. Non-paternity was excluded by DNA fingerprinting analysis with hypervariable minisatellite probes. This is the fourth case of recombination in the -globin gene cluster to be reported. The event has occurred 5 of the polymorphic RsaI site at position-550 bp upstream of the -globin gene mRNA Cap site, within the 9.1-kb region that has been shown to be a hot spot for recombination in the -globin gene cluster.  相似文献

Experimental equipment has been developed to make possible continuous recordings of intensity distribution curves as a function of angle of incidence of light in single ommatidia of arthropod compound eyes. — As preliminary experiment, the visual fields of ommatidia in Limulus, Apis and Cataglyphis have been ascertained without a light measuring instrument. A point source was moved through the visual field and the angular position of the light source on both sides of the visual field at which the rhabdome in the cut-off eye viewed from proximal turned dark was determined. In the dorsal part of the Limulus eye, the cranio-caudal width of the visual field is 42.6±0.3°. The width of the visual field of central ommatidia in Apis is 6.68±0.14° in the dorsoventral plane and 7.42±0.24° in the cranio-caudal direction, in Cataglyphis 15.5±0.5° and 12.5±0.4° respectively.— Similar Gaussian curves for light and dark adapted ommatidia resulted from the continuous recordings of intensity distribution curves in Apis. The rotatory symmetric curve of the worker bee has a half-width of =2.57°. The standardized curve of amplitude one may be approximated by the function y=exp (-0.41 [degree–2] 2). In Cataglyphis, the half-width of the intensity distribution curve is =8.8° in the dorso-ventral plane and =6.8° in the cranio-caudal plane. Near the axis, the functions y=exp(-0.04 [degree–2]2) and y=exp(-0.06 [degree–2] 2) approximate the standardized curves. — Knowledge of intensity distributions in connection with interommatidial angles allows estimation of contrast transfer and resolving power of the array of the dioptric systems. Correlations with behavioural findings are attempted.

The primary structure of the bglA gene region encoding a -glucosidase of Thermotoga maritima strain MSB8 was determined. The bglA gene has the potential to code for a polypeptide of 446 amino acids with a predicted molecular mass of 51545 Da. The T, maritima -glucosidase (BglA) was overexpressed in E. coli at a level comprising approximately 15–20% of soluble cellular protein. Based on its amino acid sequence, as deduced from the nucleotide sequence of the gene, BglA can be classified as a broad-specificity -glucosidase and as a member of the -glucosidase family BGA, in agreement with the results of enzymatic characterization of the recombinant protein. Comparative sequence analysis revealed distant amino acid sequence similarities between BGA family -glucosidases, a -xylosidase, -1,4-glycanases of the enzyme family F (mostly xylanases), and other families of -1,4-glycosyl hydrolases. This result indicates that BGA -glucosidases may comprise one enzyme family within a large enzyme order of retaining -glycosyl hydrolases, and that the members of these enzyme groups may be inter-related at the level of active site architecture and perhaps even on the level of overall three-dimensional fold.  相似文献
