Over the last 20 years, global production of Persian walnut (Juglans regia L.) has grown enormously, likely reflecting increased consumption due to its numerous benefits to human health. However, advances in genome‐wide association (GWA) studies and genomic selection (GS) for agronomically important traits in walnut remain limited due to the lack of powerful genomic tools. Here, we present the development and validation of a high‐density 700K single nucleotide polymorphism (SNP) array in Persian walnut. Over 609K high‐quality SNPs have been thoroughly selected from a set of 9.6 m genome‐wide variants, previously identified from the high‐depth re‐sequencing of 27 founders of the Walnut Improvement Program (WIP) of University of California, Davis. To validate the effectiveness of the array, we genotyped a collection of 1284 walnut trees, including 1167 progeny of 48 WIP families and 26 walnut cultivars. More than half of the SNPs (55.7%) fell in the highest quality class of ‘Poly High Resolution’ (PHR) polymorphisms, which were used to assess the WIP pedigree integrity. We identified 151 new parent‐offspring relationships, all confirmed with the Mendelian inheritance test. In addition, we explored the genetic variability among cultivars of different origin, revealing how the varieties from Europe and California were differentiated from Asian accessions. Both the reconstruction of the WIP pedigree and population structure analysis confirmed the effectiveness of the Applied Biosystems? Axiom? The Persian walnut (Juglans regia L.), a diploid species native to the mountainous regions of Central Asia, is the major walnut species cultivated for nut production and is one of the most widespread tree nut species in the world. The high nutritional value of J. regia nuts is associated with a rich array of polyphenolic compounds, whose complete biosynthetic pathways are still unknown. A J. regia genome sequence was obtained from the cultivar ‘Chandler’ to discover target genes and additional unknown genes. The 667‐Mbp genome was assembled using two different methods (SOAPdenovo2 and MaSuRCA), with an N50 scaffold size of 464 955 bp (based on a genome size of 606 Mbp), 221 640 contigs and a GC content of 37%. Annotation with MAKER‐P and other genomic resources yielded 32 498 gene models. Previous studies in walnut relying on tissue‐specific methods have only identified a single polyphenol oxidase (PPO) gene (JrPPO1). Enabled by the J. regia genome sequence, a second homolog of PPO (JrPPO2) was discovered. In addition, about 130 genes in the large gallate 1‐β‐glucosyltransferase (GGT) superfamily were detected. Specifically, two genes, JrGGT1 and JrGGT2, were significantly homologous to the GGT from Quercus robur (QrGGT), which is involved in the synthesis of 1‐O‐galloyl‐β‐d ‐glucose, a precursor for the synthesis of hydrolysable tannins. The reference genome for J. regia provides meaningful insight into the complex pathways required for the synthesis of polyphenols. The walnut genome sequence provides important tools and methods to accelerate breeding and to facilitate the genetic dissection of complex traits.  相似文献

  1. A preference experiment was set up with two planthopper species (Hemiptera: Delphacidae) to test the influence of competition on host plant choice.
  2. The delphacid Javesella pellucida was chosen as a generalist and the rarer Ribautodelphax imitans as a monophagous specialist, which feeds on the grass, tall fescue Schedonorus arundinaceus.
  3. In the absence of the specialist, the generalist showed a marked preference for tall fescue. In some experiments, however, the introduction of the specialist resulted in a shift of preference to an alternative plant if the specialist was established prior to the introduction of the generalist.
  4. This experiment supports the hypothesis that specialist herbivores can potentially alter the host plant choices of generalists, which may lead to differing host plant use patterns in insect communities.

  1. Pissodes castaneus represents an emerging pest species for Pinus spp. production in Latin America. This species attacks all the cultivated pine species in Patagonia Argentina causing wood damage and tree death. The objectives of this work were to describe the host preference behaviour of P. castaneus and the influence of wood damage by conspecifics on its host selection.
  2. In two-choice bioassays, P. castaneus showed a feeding preference for Pinus contorta over P. ponderosa. However, percent weight gain when feeding on either species was similar.
  3. In other bioassays, P. castaneus spent more time in areas with twigs on which other conspecifics had recently fed, although they were able to successfully feed on twigs without previous damage.
  4. These results show that while P. castaneus can successfully colonize different pine species, P. contorta may be more susceptible to attack than P. ponderosa. Silvicultural management should be prioritized considering the weevil's preference for attacking damaged hosts.

Life history traits, historic factors, and human activities can all shape the genetic diversity of a species. In Italy, walnut (Juglans regia L.) has a long history of cultivation both for wood and edible nuts. To better understand the genetic variability of current Italian walnut resources, we analyzed the relationships among the genetic structure of local walnut populations (inferred by SSR markers) and human migrations along ancient routes, using the territory of Royal Tratturo Candela-Pescasseroli (RT) as a case study. Sixteen J. regia provenances were collected along RT and compared with 13 Italian provenances and the landrace Sorrento. Although the level of SSR polymorphism we observed was moderately high, AMOVA revealed that most of the diversity was located within individuals (92.58%), and geographical differentiation was low (D est = 0.076). Evidence for human-mediated domestication bottleneck events was detected in about 95% of walnut provenances. A Bayesian approach divided 456 walnut samples into three clusters: (1) Sorrento genotypes, (2) trees from the island of Sicily, and (3) the remaining germplasm. The UPGMA tree based on Nei's distances distinguished northeastern provenances and weakly grouped 12 of 16 provenances of RT. The observed genetic differences derived mainly from gradations in allele frequencies. Separation of the Sicilian provenance from the mainland may be explained in terms of founder effects and prolonged geographic isolation. Two contrasting forces, selection, and frequent inter-regional transfer of propagules, appear to drive the patterns of genetic variability for J. regia.  相似文献

  1. Host selection behaviour of the walnut twig beetle (WTB) among hardwood trees was investigated in a riparian forest in northern California by monitoring the landing rate of the beetle with sticky traps on branches baited with 3-methyl-2-buten-1-ol, the male-produced aggregation pheromone.
  2. The assay was conducted over 7 days (22 May to 29 May 2017) and compared landing rates on branches of six nonhost species paired with northern California black walnut, Juglans hindsii (the host).
  3. A total of 2242/1192 WTB were collected on branches of host/nonhost pairs, and more WTB landed on J. hindsii than on nonhosts in 42 of 58 instances. Female landing rate generally exceeded male landing rate, which underscores the influence of the male-produced synthetic pheromone in this system.
  4. Landing rates of WTB males, females, and the combined sexes on boxelder, Acer negundo, and valley oak, Quercus lobata, did not differ significantly from the landing rates on J. hindsii, suggesting that these two nonhost riparian hardwoods do not repel WTB (in the context of the aggregation pheromone).
  5. Significantly fewer WTB landed on Oregon ash, Fraxinus latifolia, river red gum, Eucalyptus camaldulensis, Fremont cottonwood, Populus fremontii, and red willow, Salix laevigata, than on J. hindsii, which suggests that these four nonhosts may repel one or both sexes of WTB in the context of the aggregation pheromone. Future analysis of the volatiles from these four hardwood species may lead to the discovery of semiochemical repellents for WTB.

The olfactory stimuli from the maturation feeding‐plant, Rosa multiflora Thunb., and larval‐host plant, Populus deltoides, of the longhorned beetle, Batocera horsfieldi (Hope), were investigated by using TCT‐GC/MS (Thermal desorption and cold trap, and GC/MS) and electroantennogram recordings (EAG). A total of 20 plant compounds were identified from plant headspace volatile, including aliphatic, aromatic and terpenoid compounds. Five compounds were common to both plants. Eight compounds eliciting strong EAG response were E‐3‐hexenyl acetate, 3‐carene, 1‐penten‐3‐ol, 3‐pentanol, Z‐2‐penten‐1‐ol, hexanal and E‐2‐hexenal. Female and male B. horsfieldi exhibited broad overlap in their EAG responses to individual plant odour, and there was no clear pattern of difference between responses of female and male antennae to different compounds. Mating status had little effect on the EAG responses of females.  相似文献

Abstract 1. Plant traits (e.g. nutrition, allelochemistry) are an important determinant of the feeding preferences and performance of insect herbivores. Recent evidence suggests that plant inbreeding can affect plant–insect interactions by impacting host‐plant quality and resistance to herbivory. 2. The effect of inbreeding on host‐plant quality for, and resistance against, the tobacco hornworm, Manduca sexta L., was assessed in the wild solanaceous weed horsenettle, Solanum carolinense L. Caterpillar preference, relative growth rate (RGR), total leaf consumption (TC), and per cent total nitrogen in leaves were examined using selfed and outcrossed progeny of eight maternal plants. 3. Inbreeding significantly influenced insect preference, with caterpillars preferring leaf discs from selfed versus outcrossed plants. There was also a breeding effect for RGR and TC, with both higher on selfed plants. No breeding effect for per cent total nitrogen was observed. 4. The results of this study indicate that inbreeding decreased resistance against the tobacco hornworm, but did not affect plant quality. Decreased plant resistance will likely alter interactions with the herbivore community and could also have important consequences for plant–herbivore–natural enemy interactions.  相似文献

为鉴定引起四川盆地地区核桃黑斑病的病原菌,采用组织分离法对病原菌进行分离,利用柯赫氏法则验证其致病性,依据菌株形态学和基于16S rDNA基因序列分析对病原菌进行鉴定;同时,利用分离的菌株对18个栽培品种(无性系)进行抗病性评价。结果表明,分离菌株的菌落形态与黄单胞杆菌属(Xanthomonas)相似,其16S rDNA序列与树生黄单胞杆菌(X. arboricola)的(登录号为KP340804.1)同源性高达99%,因此,将引起四川盆地地区核桃黑斑病的病原菌鉴定为树生黄单胞杆菌。18个核桃栽培品种(无性系)的田间侵染发病率和病情指数分别为35.07%~78.57%和17.71%~51.96%,变异系数分别为17.62%和28.78%,并以此为基础评价出5个高抗病品种(无性系)。这为核桃黑斑病致病机理研究和抗病新品种的选育奠定基础。  相似文献

应用16对SSR引物对国内不同地理生态型的44个核桃样品、8个铁核桃样品和7个美国核桃样品进行了分析。结果显示,在16个SSR位点上共获得87个等位基因,每个位点扩增等位基因4~9个,平均5.4个。各SSR位点的观察杂合度为0~0.931 0,平均为0.389 4。各位点PIC值在0.376 2~0.786 3之间,平均为0.663 2。59个核桃样品的杂合度在0.133~0.813之间,平均0.387。聚类分析表明,来自秦岭山地的核桃样品与华北核桃属于同一生态类型;巩留的野核桃属于与新疆生态型和华北生态型不同的一个独立的生态型;美国核桃和铁核桃属于核桃种下不同的生态类型。  相似文献

  • Citrus plants are host to several plant parasitic nematodes (PPNs), which are microscopic organisms. Among PPNs, the citrus root nematode, T. semipenetrans (Cobb 1913) (Tylenchida: Tylenchulidae), causes significant damage to citrus plantations worldwide. Understanding citrus nematode populations, precise identification, host preference among citrus species, and damage threshold are crucial to control T. semipenetrans. The minutiae of citrus plant–nematode interactions, nematode density and molecular nematode identification are not well understood. In this study, nematode species and density in citrus orchards, host specialization, molecular and morphological characteristics of nematodes were assessed.
  • Molecular and morphological methods, host–nematode interactions, host (citrus species) preference, damage economic threshold (ET), and economic injury level (EIL) were determined using laboratory methods and field sampling. Citrus plantations in different provinces in the Mediterranean region of Turkey were investigated.
  • Nematode species were identified molecularly and morphologically. ITS sequences revealed that samples were infected by citrus root nematode T. semipenetrans. The lowest nematode density was in C. reticulata in Mersin (53 2nd stage juveniles (J2s) 100 g−1 soil), while the highest density was from Hatay in C. sinensis (12173 J2s 100 g−1 soil). Highest citrus nematode population density was on roots of C. reticulata, followed by C. sinensis, C. limon, and C. paradisi.
  • The citrus nematode is more common than was thought and population fluctuations change according to specific citrus species. Environmental conditions, host and ecological factors, such as temperature, soil pH, and soil nutrients, might influence nematode populations in citrus orchards. Investigating nematode density in diverse soil ecologies and the responses of different resistant/tolerant citrus species and cultivars to nematode populations is essential in future studies.

English walnut (Juglans regia L.) is the most economically important species from all the 21 species belonging to the genus Juglans and is an important and healthy food as well as base material for timber industry. The aim of this study was to develop a simple technique for specific characterization of English walnut using DNA method. The first and second internal transcribed spacers (ITS1 and ITS2) as well as the intervening 5.8S coding region of the rRNA gene for 18 cultivars of J. regia L. isolated from different geographic origins were characterized. The size of the spacers sequences ranged from 257 to 263 bases for ITS1 and from 217 to 219 bases for ITS2. Variation of GC contents has also been observed and scored as 55–56.7 and 57.1–58.9% for ITS1 and ITS2, respectively. This data exhibited the presence of polymorphism among cultivars. Alignment of the ITS1-5.8S-ITS2 sequences from 18 walnut cultivars showed that there were 244 single nucleotide polymorphisms (SNPs) and 1 short insertion–deletion (indel) at 5′ end ITS1. Amplification refractory mutation system strategy was successfully applied to the SNP markers of the ITS1 and ITS2 sequences for the fingerprinting analysis of 17 on 18 walnut cultivars. The prediction of ITS1 and ITS2 RNA secondary structure from each cultivar was improved by detecting key functional elements shared by all sequences in the alignments. Phylogenetic analysis of the ITS1-5.8S-ITS2 region clearly separated the isolated sequences into two clusters. The results showed that ITS1 and ITS2 region could be used to discriminate these walnut cultivars.  相似文献

The cabbage whitefly [Aleyrodes proletella L. (Hemiptera: Aleyrodidae)] is becoming a serious pest in Brassica oleracea L. (Brassicaceae) crops. However, almost nothing is known about the interaction of this insect with its host plants. Previous studies have shown differences in the natural occurrence of adults, eggs, and nymphs on the closely related B. oleracea cultivars Christmas Drumhead and Riviera grown in the field. In this study, we aimed to identify the nature of these differences and to gain insight into the resistance mechanisms against A. proletella. We used no‐choice experiments on field‐ and greenhouse‐grown plants to show that the differences between the two cultivars are mainly based on antibiosis (traits that reduce herbivore performance) and not on antixenosis (traits that deter herbivory). This was further supported by laboratory choice experiments that indicated little or no discrimination between the two cultivars based on plant volatiles. We showed that resistance is dependent on plant age, that is, resistance increased during plant development, and is mainly independent of environmental factors. Analysis of probing behaviour revealed that the resistance trait affects A. proletella at the phloem level and that morphological differences between the two cultivars are most likely not involved. We suggest that compounds present in the phloem reduce sap ingestion by the whitefly and that this explains the observed resistance.  相似文献

Thirty-two low-copy-number genomic DNA clones from a walnut (Juglans sp.) Pst I genomic library were used to establish a molecular-marker linkage map for walnut. The clones were hybridized to restriction-endonuclease-digested DNA from parent walnut trees involved in an interspecific backcross of (J. hindsii x J. regia) x J. regia in order to identify parental polymorphism. Sixty-three backcross progeny were analyzed to determine the inheritance and linkage of 48 RFLP loci. Sixty-six percent of the walnut cloned sequences detected duplicated, but unlinked, loci. Twelve linkage groups were identified by 42 of the RFLP loci. A Poisson probability method for estimating genome size was utilized to calculate the approximate walnut genome length as 1660 cM and to estimate that 138 markers would be needed to cover 95% of the walnut genome within 20 cM of each marker.  相似文献

  • 1 The host‐odour preferences of the rice leaf bug Trigonotylus caelestialium between the rice plant Oryza sativa L. and four species of graminaceous weeds, Poa annua, Alopecurus aequalis, Digitaria ciliaris and Eleusine indica, were investigated with an olfactometer aiming to clarify the influence of these odours on invasion of the bug to paddy fields at the flowering stage of rice.
  • 2 Both female and male adults significantly preferred the graminaceous weed A. aequalis in the flowering stage to rice in the fifth‐leaf stage. The bugs also significantly preferred flowering P. annua and A. aequalis to rice in the panicle‐formation stage. However, the bugs showed no preferences between rice in the flowering and grain‐filling stages and the flowering graminaceous weeds P. annua, D. ciliaris and E. indica.
  • 3 The preference of the rice leaf bug for the flowering graminaceous weeds before rice flowering coincides with the fact that these bugs mainly live on these weeds before rice flowering. It is considered that the bug's similar preference for flowering rice panicles as the flowering graminaceous weeds causes the intense invasion of the bug into paddy fields at this rice developmental stage.
