Female two‐spotted spider mite Tetranychus urticae are grown under different photoperiods and the photoperiodic regulation of diapause is examined. The photoperiodic response curve for diapause induction was of the long day–short day type, with critical day lengths (CDLs) of 2 and 12.5 h; diapause was induced between these CDLs. The preimaginal period is significantly longer in diapausing females than in non‐diapausing females; moreover, a significant positive correlation is detected between diapause incidence and deutonymphal period. Diapause incidence is high when long‐night photoperiods are applied against a background of continuous darkness in the stages including the deutonymph; this stage appears to be the most sensitive to photoperiod. These observations suggest that diapause‐inducing conditions inhibit nymphal development, particularly in the deutonymphal stage when photoperiodic time measurement for determination of reproduction or diapause is carried out.  相似文献

Plant regeneration through somatic embryogenesis was achieved using immature zygotic embryos (ZE) of Sorbus pohuashanensis as explants. Over 50% of immature ZEs from immature seed collected at 30 days after pollination produced direct somatic embryos (SEs) on Murashige and Skoog (MS) medium supplemented with 0–0.44 μM 6-benzyladenine (BA) in combination with 5.73 μM naphthaleneacetic acid (NAA) or with 0.91–2.26 μM 2,4-dichlorophenoxyacetic acid (2,4-D) alone. Fourteen to 23 SEs per explant were regenerated on MS medium supplemented with BA 0.44 μM in combination with NAA 5.73 μM. SE formation decreased when sucrose concentrations were higher than 40 g L−1. Repetitive embryogenesis occurred following culture on solid MS medium containing 12 μM abscisic acid, 75 g L−1 polyethylene glycol, and 20 g L−1 sucrose at 25 ± 1°C under a 16-h photoperiod with a light intensity of 40 μmol m−2 s−1. Over 40% of the mature SEs germinated on solid MS medium under light condition described previously. Up to 40% of the regenerated plantlets were successfully acclimatized under greenhouse conditions. Plantlets derived from SEs grew vigorously with similar morphology as those germinated from ZEs. Histological studies of explants at various developmental stages of somatic embryogenesis revealed that SEs passed through globular, heart, torpedo, and mature stages. Similar to ZE suspensors, similar structures of SE degenerated in later stages of embryo development. ZE and SE are a effective means of regenerating tissue culture plantlets for S. pohuashanesis.  相似文献

The inhibitory effects of the timing, intensity (II) and period (IT) of night-interrupting light on diapause induction of the Kanzawa spider mite (Tetranychus kanzawai) were investigated in a series of laboratory experiments. During a light and dark period of 8 and 16 h d−1, respectively, a single 1-h night-interrupting light was applied at early (E), middle (M), and late (L) parts of the dark period: i.e., at 3, 7.5, and 12 h after the start of the dark period, respectively. No interrupting light was applied in the control treatment. The incidence of diapause was significantly lower in the M treatment (63%) compared to the control treatment (100%). In the E and L treatments, more than 90% of females entered diapause, which was comparable to the control treatment. Since the longest consecutive dark period during the E and L treatments was longer than the critical dark period (CDP) of 10.5-11 h d−1, during which 50% of females entered diapause, the night-interrupting light probably failed to prevent diapause induction. However, in the M treatment, the longest consecutive dark period was shorter than the CDP; therefore, the night-interrupting light inhibited diapause induction. Moreover, the inhibitory effects of night-interrupting light in the M treatment increased as II and IT increased. The dose of night-interrupting light (II × IT) was significantly negatively related to the incidence of diapause. The median effective dose for 50% disturbance of diapause induction was 2.5 kJ m−2 at wavelengths between 350 and 1050 nm. Our results suggest that the longest consecutive dark period and the dose of night-interrupting light should both be considered when a lighting-based physical control is applied to inhibit diapause induction and consequent overwintering of T. kanzawai in commercial agricultural fields.  相似文献

Summary A characteristic phenotype of highly embryogenic explants along with the location of embryogenesis- and transformation-competent cells/tissues on immature cotyledons of soybean [Glycine max (L.) Merrill.] under hygromycin selection was identified. This highly embryogenic immature cotyledon was characterized with emergence of somatic embryos and incidence of browning/necrotic tissues along the margins and collapsed tissues in the mid-region of an explant incubated upwards on the selection medium. The influences of various parameters on induction of somatic embryogenesis on immature cotyledons following Agrobacterium tumefaciens-mediated transformation and selection were investigated. Using cotyledon explants derived from immature embryos of 5–8 mm in length, a 1∶1 (v/v; bacterial cells to liquid D40 medium) concentration of bacterial suspension and 4-wk cocultivation period significantly increased the frequency of transgenic somatic embryos. Whereas, increasing the infection period of explants or subjecting explants to either wounding or acetosyringone treatments did not increase the frequency of transformation. An optimal selection regime was identified when inoculated immature cotyledons were incubated on either 10 or 25 mgl−1 hygromycin for a 2-wk period, and then maintained on selection media containing 25 mgl−1 hygromycin in subsequent selection periods. However, somatic embryogenesis was completely inhibited when inoculated immature cotyledons were incubated on a kanamycin selection medium. These findings clearly demonstrated that the tissue culture protocols for transformation of soybean should be established under both Agrobacterium and selection conditions.  相似文献

To investigate the physiology of Chrysopa pallens, the effect of photoperiod on diapause and development was examined in a Japanese population (33.4°N). The response stage for diapause of C. pallens was considered to be the prepupal stage. The critical photoperiod for diapause induction at 20.0°C was between 13 h light : 11 h dark (LD 13:11) and LD 14:10. The larval developmental period was affected by photoperiod: larvae in diapause took longer to complete their development. This difference of larval developmental period in relation to photoperiod was considered to be an adjustment of larval diapause timing.  相似文献

Jatropha curcas L. (Physic nut) is a commercially important non-edible oil seed crop known for its use as an alternate source of biodiesel. In order to investigate the morphogenic potential of immature embryo, explants from four developmental stages were cultured on medium supplemented with combinations of auxins and cytokinins. It was found that the size of embryo is critical for the establishment of callus. Immature embryos (1.1–1.5 cm) obtained from the fruits 6 weeks after pollination showed a good response of morphogenic callus induction (85.7%) and subsequent plant regeneration (70%) with the maximum number of plantlets (4.7/explant) on Murashige and Skoog’s (MS) medium supplemented with IBA (0.5 mg l−1) and BA (1.0 mg l−1). The above medium when supplemented with growth adjuvants such as 100 mg l−1 casein hydrolysate + 200 mg l−1 l-glutamine + 8.0 mg l−1 CuSO4 resulted in an even higher frequency of callus induction (100%). Plant regeneration (90%) with the maximum number of plantlets (10/explant) was achieved on MS medium supplemented with 500 mg l−1 polyvinyl pyrrolidone + 30 mg l−1 citric acid + 1 mg l−1 BA + 0.5 mg l−1 Kn + 0.25 mg l−1 IBA. It was observed that plantlet regeneration could occur either through organogenesis of morphogenic callus or via multiplication of pre-existing meristem in immature embryos. The age of immature embryos and addition of a combination of growth adjuvants to the culture medium appear to be critical for obtaining high regeneration rates. Well-developed shoots rooted on half-strength MS medium supplemented with 0.5 mg l−1 IBA and 342 mg l−1 trehalose. The rooted plants after acclimatization were successfully transferred to the field in different agro-climatic zones in India. This protocol has been successfully evaluated on five elite lines of J. curcas.  相似文献

In this paper we examine the hypothesis that diapause induction in the polyvoltine pod midge Dasyneura brassicae Winn. (Diptera: Cecidomyiidae) is controlled by cumulative global solar radiation during the larval stage. The correlation between field observations of relative diapause for 3 years and four environmental factors (daylength, temperature, cumulative daylight time, and cumulative global solar radiation) was investigated. The cumulative global solar radiation during the larval stage clearly showed the strongest correlation with the observed relative diapause. It was estimated that larvae which cumulated less than 71.1 kWh m−2 entered diapause. Compared to former theories on diapause induction in insects, this hypothesis explains how temperature and light can function together and, furthermore, it removes the requirement for biological clock involvement in diapause induction in insects. The willow leaf beetle, Plagiodera versicolora (Coleoptera: Chrysomelidae) overwinters in adult diapause. In this study, the photoperiodic responses for diapause induction and developmental time were examined in the Ishikari (Hokkaido, Japan) population of P. versicolora. All females entered reproductive diapause under short daylength (L10:D14), but 31.7% of females did not enter diapause under long daylength (L16:D8). The developmental time from oviposition to adult emergence was significantly longer at L10:D14 than that at L16:D8. Norm of reaction curves illustrated variation among families in the photoperiodic responses for diapause induction and for developmental time. ANOVA indicated significant family × photoperiod interactions in the developmental time. At L16:D8, developmental time was positively correlated with the incidence of diapause in females. This means that a female having a longer developmental time tends to have a longer critical photoperiod. Such variation may be maintained by differences in selection pressures on the growth rate and the critical photoperiod for diapause induction between univoltine and bivoltine genotypes because Ishikari is located in a transitional area between populations with univoltine and bivoltine life cycles.  相似文献

Seasonal activities of the digestive enzyme trypsin were measured between August 1998 and May 1999 to study different nutritional strategies of the two copepods Pseudocalanus minutus and Oithona similis in the Arctic Kongsfjorden (Svalbard) using a highly sensitive fluorescence technique. Stage-, depth- and season-specific characteristics of digestive activity were reflected in the trypsin activity. P. minutus females and stage V copepodids (C) had highest trypsin activities in spring during reproduction (197.5 and 145.7 nmol min−1 ng C−1, respectively). In summer stages CIII–V and in autumn stages CIV and V had high activities (80–116 nmol min−1 ng C−1) in the shallow layer (< 100 m) presumably as a consequence of prolonged feeding before descending to overwintering depth. Trypsin activities at depth (> 100 m) in summer and autumn were low in stages CIII and CIV (29–60 nmol min−1 ng C−1) and in winter in all stages in both layers (20–43 nmol min−1 ng C−1). Based on low trypsin activity, males most likely did not feed. In O. similis, the spring phytoplankton bloom did not significantly affect trypsin activity as compared to the other seasons. O. similis CV and females had high trypsin activities in summer in the deep stratum (304.5 nmol min−1 ng C−1), which was concomitant with reproductive processes and energy storage for overwintering. In autumn, stage CV and female O. similis had significantly higher activities than stage CIV (130–152 versus 78 nmol min−1 ng C−1), which is in accordance with still ongoing developmental and reproductive processes in CVs and females. Comparisons of both species revealed different depth-related responses emphasizing different nutritional preferences: the mainly herbivorous P. minutus is more actively feeding in the shallow layer, where primary production occurs, whereas the omnivorous O. similis is not as much restricted to a certain depth layer, when searching for food. P. minutus had lower levels of trypsin activity during all seasons. In contrast to P. minutus, higher enzyme activities in males of O. similis suggest that they continue to feed and survive after fertilization of females.  相似文献

We investigated the effect of carbon/nitrogen (C/N) ratio on astaxanthin synthesis in Haematococcus pluvialis during photoautotrophic induction by continuous input of both CO2–air mixture and intense light. When H. pluvialis was induced by constant irradiance induction at 200 μmol photon m−2 s−1, there was a positive correlation with astaxanthin content and C/N ratio, which was similar to the case for heterotrophic induction. Lower C/N ratios did not retard Haematococcus encystment, but did increase culture biomass, resulting in a decrease in astaxanthin production because of light limitation. However, induction using variable irradiance showed that reduction of astaxanthin production at low C/N ratios was successfully overcome by simply increasing the light intensity from 200 to 300 μmol photon m−2 s−1 to overcome the light limitation. This resulted in a greatly enhanced astaxanthin synthesis in proportion to cell density in cultures with low C/N ratios. Our results indicate that light intensity is more critical than C/N ratio in astaxanthin production by H. pluvialis during photoautotrophic induction.  相似文献

The predatory mite Typhlodromus bagdasarjani Wainstein and Arutunjan (Acari: Phytoseiidae) is an indigenous and widespread species of the Middle East fauna. In this paper we assess the effect of temperature on developmental rate and reproduction potential of T. bagdasarjani under laboratory conditions. The development of this species was determined at 15, 20, 25, 30, 35 and 37.5 ± 1°C, 60 ± 10% RH and L16:D8 h photoperiod. The total developmental time averaged 28.2, 15.0, 8.9, 7.6, 7.2 and 7.4 days at 15–37.5°C, respectively, when feeding on immature stages of two-spotted spider mite, Tetranychus urticae Koch. The lower developmental threshold (T 0 ) and thermal constant (K) for the development of this predator were estimated 9.2°C and 162 degree-days by the Ikemoto linear model. The life table parameters were estimated at 15–35°C. The shortest life span of females at 35°C was 45.0 days, followed by 50.7, 50.9, 103.3 and 136.8 days at 30, 25, 20 and 15°C, respectively. Mated females laid on average 19.9, 26.3, 41.1, 39.6 and 31.3 eggs per female at 15–35°C, respectively. The intrinsic rate of increase (r m ) and finite rate of increase (λ) increased significantly with increasing temperature. The r m values ranged from 0.021 (15°C) to 0.186 (35°C) days−1. The highest value of net reproductive rate (R 0) was 13.6 females progeny/female/generation at 25°C. The results demonstrated that T. bagdasarjani is well adapted to high temperatures. However, the efficiency to control spider mites may be affected by behavioral characteristics of the predator and its prey under real conditions.  相似文献

Side branch formation in the moss, Physcomitrella patens, has been shown to be light dependent with cryptochrome 1a and 1b (Ppcry1a and Ppcry1b), being the blue light receptors for this response (Imaizumi et al. in Plant Cell 14:373, 2002). In this study, detailed photobiological analyses were performed, which revealed that this response involves multiple photoreceptors including cryptochromes. For light induction of branches, blue light of a fluence rate higher than 6 μmol m−2 s−1 for period longer than 3 h is required. The number of branches increased with the increase in fluence rate and in the irradiation period. The number of branches also increased when red light was applied together with the blue light, although red light alone had a very few effect. By partially irradiating a cell, both receptive sites for blue and red light were found to be located around the nucleus. Further, both red and blue light determine the positions of branches being dependent upon the vibration plane of polarized light. Red light control of branch position was nullified by simultaneous far-red light irradiation. A blue light effect on branch position was not found in lines with disrupted phototropin genes. Thus, dichroic phytochrome and phototropin, possibly on the plasma membrane, regulate branch position. These results indicate that at least four distinct photoreceptor systems, namely, cryptochromes and red light receptor around or in the nucleus, dichroic phytochrome and phototropin around the cell periphery, are involved in the light induction of side branches in the moss Physcomitrella patens.  相似文献

Nothofagus nitida (Phil.) Krasser (Nothofagaceae) regenerates under the shade. Nonetheless, older seedlings are commonly found at full sun. We tested the hypothesis that light capture and photochemical and non-photochemical energy dissipation of both photosystems PSI and PSII adjust with ontogeny and brighter environment. Light energy partitioning in both photosystems was studied in seedlings of different developmental stages (small 9.7 cm, tall 36 cm) under contrasting light environments (8–200 and 1,800–2,043 μmol photons m−2 s−1) in the Chilean evergreen temperate forest. Higher A max, dark respiration, and light compensation and saturation points in sun seedlings of both developmental stages were accompanied by higher rates of electron transport. These seedlings also showed a high fraction of open PSII reaction centres and similar non-photochemical quenching at high-light in both photosystems, showing no effect of developmental stage in these parameters. Conversely, light capture, total thermal dissipation after photoinhibition, active down-regulation of antenna efficiency, and state transitions were higher in smaller seedlings than in taller ones. These changes maintain photostasis, preventing photodamage, while favouring a more oxidized quinone pool. There is an independent effect of seedling development and light acclimation on this transition from shade to sun during early ontogeny. This transition reflects short-term responses of the photosynthetic apparatus to light and longer term responses that depend on seedling developmental stage.  相似文献

In temperate areas, population dynamics of the invasive Asian tiger mosquito Aedes albopictus are strongly affected by winter. The work we present here analyzes the adaptive synchronization of the diapause process in the wintry generation of A. albopictus, where the egg stage is exposed to adverse winter conditions. The seasonal pattern of egg laying activity of a French Mediterranean population of the Asian tiger mosquito was monitored weekly for 2 years with ovitraps. The field diapause incidence and the critical photoperiod (CPP, i.e. the maternal day length inducing diapause in 50% of the eggs), were determined by hatching experiments on the collected eggs. The period of diapause termination was estimated by a field survey of the first hatchings for both years. The CPP is equal to 13.5 hours of light and occurs in the field on the 25th of August. Thus, it is on September 11th, 17 days after the CPP, that 50% of the eggs are in a prediapause stage in the field. The egg diapause rate increases rapidly during September, whereas the mean number of eggs laid decreases sharply after mid-September. Surprisingly, after having reached a peak of 95% at the end of September, from mid-October the diapause incidence declined and stayed below 50%. Indeed, both years the diapause initiates before the rapid decrease of the environmental temperature. This leaves a sufficient period of time to the complete development of one generation of A. albopictus with effective induction of diapause in the laid eggs. The very first larvae hatched were sampled both years in the first half of March. With 20 to 26 weeks in the egg stage and about 7 weeks in the larval stages, the first annual generation spends a long time in immature stages. On a practical point of view, this long development time represents a wide window for eggs and larvae control in early spring.  相似文献

A protocol for plant regeneration via somatic embryogenesis was developed in two chickpea (Cicer arietinum L.) cultivars ICCV-10 and Annigeri. Somatic embryos were induced from immature cotyledons on Murashige and Skoog’s (MS) medium supplemented with different concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D), 2,4,5-trichlorophenoxyacetic acid (2,4,5-T), α-naphthaleneacetic acid (NAA) and picloram alone or in combination with 0.5 — 2.0 mg dm−3 N6-benzylaminopurine (BA) or kinetin (KIN). NAA was better for somatic embryo induction compared to other auxins. The well formed, cotyledonary shaped embryos germinated into plantlets with 36.6 % frequency on MS medium supplemented with 2.0 mg dm−3 BA + 0.5 mg dm−3 abscisic acid (ABA). The frequency of embryogenesis and plantlet regeneration was higher in cv. ICCV-10 as compared to cv. Annigeri. Regenerated plants were transferred to soil (40 % survival) and grown to maturity. Histological studies of explants at various developmental stages of somatic embryogenesis reveled that somatic embryos developed directly from the cotyledon cells and they were single cell origin.  相似文献

β-Glucans of Agaricus brasiliensis fruiting bodies in different stages of maturity were isolated and characterized by FTIR and NMR. These fractions had greater amount of (1→6)-β-glucan and the (1→3)-β-glucan increased with fruiting bodies maturation. Yields of β-glucans increased from 42 mg β-glucans g−1 fruiting bodies (dry wt) in immature stage to 43 mg g−1 in mature stage with immature spores, and decreased to 40 mg g−1 in mature stage with spore maturation. Mature fruiting bodies, which included these glucans, have potential therapeutical benefits for use in nutraceutical products.  相似文献

The effect of temperature on the development and fecundity of Sancassania polyphyllae fed on tissues of Polyphylla fullo larvae was studied at 15, 20, 25, 30, and 35 ± 1°C and 65 ± 10% RH in a dark incubator. Mean developmental period of immature stages decreased significantly with increasing temperatures from 15 to 30°C. Developmental periods at 30–35°C were not significantly different. The estimated lower developmental thresholds of the various immature stages ranged between 10.1 and 11.5°C. The thermal constant for the egg-to-female adult was 93.5 degree-days. The pre-oviposition, oviposition, and post-oviposition periods and female longevity were significantly longer at 15°C than at higher temperatures. Mean total and daily fecundity were the highest at 25°C, which were significantly different from those obtained at 15, 20 and 30°C. The net reproductive rate (R 0) was the highest at 25°C (588.3 ♀/♀). The longest mean generation time (T 0) occurred at 15°C (36 days) and the shortest occurred at 30°C (9.2 days). The highest intrinsic rate of increase (r m) for S. polyphyllae was observed at 25 (0.61 ♀/♀/day) and 30°C (0.62 ♀/♀/day).  相似文献

Embryogenic cultures were induced from immature avocado zygotic embryos representing different botanical races and complex hybrids. The optimum induction medium consisted of B5 major salts, MS minor salts, 0.4 mg l−1 thiamine HCl, 100 mg l−1 myo-inositol, 30 g l−1 sucrose, 0.41 μM picloram and 8 g l−1 TC agar. Somatic embryogenesis occurred directly from the explants on induction medium, and secondary embryos and proembryonic masses proliferated in liquid and on semisolid maintenance medium. Embryogenic culture maintainance was optimized in liquid, filter-sterilized MS medium, supplemented with 30–50 mg l−1 sucrose, 4 mg l−1 thiamine HCl and 0.41 μM picloram. Two types of embryogenic cultures were recognized: –genotypes that proliferated as proembryonic masses in the presence of auxin (PEM-type) and; –genotypes in which the heart stage and later stages of somatic embryos developed in the presence of auxin(SE-type). Embryogenic suspension cultures became increasingly disorganized over time, and this was associated with progressive loss of embryogenic potential. Seashore paspalum (Paspalum vaginatum Swartz) is a salt tolerant, fine textured turfgrass used on golf courses in coastal, tropical, and subtropical regions. A callus induction and plant regeneration protocol for this commercially important turfgrass species has been developed. Induction of highly regenerable callus with approximately 400 shoots per cultured immature inflorescence (1 cm in length) was achieved by culturing 0.2 cm segments on media with 3 mg l−1 3,6-dichloro-2-methoxybenzoic acid (dicamba) and 0.1 or 1.0 mg l−1 benzylaminopurine (BA). A multifactorial experiment demonstrated the combination of 3 mg l−1 dicamba and 1.0 mg l−1 BA for induction of callus resulted in 12 times higher plant regeneration frequency compared to 3 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) alone or ten times higher plant regeneration frequency than the combination of 3 mg l−1 2,4-D and 1.0 mg l−1 BA. These results are expected to support the development of a genetic transformation protocol for seashore paspalum.  相似文献

ABSTRACT. Reproductive diapause was induced in A. brassicae by short photo-period at 15 and 20°C but was almost prevented at 25°C. The critical photoperiod was LD 153/4: 81/4 at 15°C, giving 60% diapause. Photoperiodic sensitivity was confined to the early immature stages. Food plant quality appeared not to influence the induction or prevention of diapause. Chilling facilitated diapause termination. Overwintering females brought into the laboratory as adults or pupae from the field from September to December took longer before laying their first eggs than females collected in January and February.  相似文献
