Changes in antioxidant systems in soybean and associated weeds (Ambrosia artemisiifolia L., Chenopodium album L., Convolvulus arvensis L and Sinapis arvensis L.) were studied in relation to treatment with herbicides linuron and dimethenamid in the field experiment. Differences in the total superoxide dismutase (SOD) and catalase (Cat) activities were observed in plants after application of herbicide formulation. Quantities of superoxide (O2.-) and hydroxyl (·OH) radicals and malonyldialdehyde (MDA), reduced glutathione (GSH) and total polyphenols content were also determined. In addition to this, potential antioxidant activity of the plant ethanolic extracts were assessed based on the scavenging activity of stable DPPH free radicals. Results obtained suggest that plants investigated 1) expressed different antioxidant systems in response to herbicide treatment; 2) enzymatic and non-enzymatic protective mechanisms were complementary; 3) some weed species showed distinctive and combined activity of several biochemical parameters, such as Ambrosia artemisiifolia.  相似文献

Crystals in 16 species of poisonous plants growing naturally in Saudi Arabia were studied with light microscopy. Three types of crystals were observed: druses, prismatics, and crystal sand. Raphides and styloids were not observed in any of the species studied. Druses occur more frequently in the leaf midrib and in the cortex and pith of the stem. In contrast, crystal sand and prismatic crystals are rare and occur in the leaf, intercostal lamina, and in the vascular tissues. The preliminary results show the absence of the three types of calcium oxalate crystals in the stem and leaf of seven species: Ammi majus L., Anagallis arvensis L., Calotropis procera Ait., Citrullus colocynthis (L.) Schard, Euphorbia peplis L., Hyoscyamus muticus L., and Solarium nigrum L., and the presence of druses, prismatic crystals, and crystal sand either in the leaf and stem or in the leaves or stems of nine species: Anabasis articulata (Forssk.) Moq. in DC., Chenopodium album L., Convolvulus arvensis L., Datura stramonium L., Nerium oleander L., Ricinus communis L., Rumex nervosus Vahl., Pergularia tomentosa L., and Withania somnifera (L.) Dun. in DC. These observations indicate that there is no apparent relationship between the distribution of calcium oxalate crystals and the toxic organs of the plants, and supports the view that the presence of calcium oxalate crystals may not be related to plant toxicity.  相似文献

During the induction of DNA synthesis in Jerusalem artichoke (Helianthus tuberosus L.) tuber by 2,4-D, the 2-14C-2, 4-D from the agar medium rapidly incorporated into the ethanol soluble and insoluble fractions. Although the 2,4-D level in the ethanol soluble fraction decreased on transplantation of the tissue from the 2-14C-2,4-D medium to medium without the auxin, its level in the buffer-soluble and -insoluble macromolecular fractions increased. The purified, buffer-insoluble macromolecules were chromatin. The 2,4-D binding to chromatin particularly increased during DNA synthesis. The histone contents of chromatin decreased as DNA synthesis progressed. The polyacrylamide gel electrophoretic patterns of the histones showed a decrease in the moderately lysine-rich histone fraction as compared to other fractions. Thus, the decrease in the histone level caused by 2,4-D and the presence of the 2,4-D moderately lysine-rich histone complex may be closely related to the induction of DNA synthesis by 2,4-D in cells.  相似文献

Summary In vitro, selection is a viable method of selecting herbicide-tolerant crops. This research was to evaluate in vitro selection techniques for enhancing 2,4-D [(2,4-dichlorophenoxy) acetic acid] tolerance in red clover (Trifolium pratense L.). In vivo and in vitro responses to 2,4-D of eight diverse red clover populations were correlated (r=0.77), justifying in vitro selection for 2,4-D tolerance. Suspension cultures of a red clover genotype capable of regeneration were plated onto agar-based nutrient media supplemented with 0.18 mM 2,4-D for selection experiments. After two cycles of selection, 16 2,4-D tolerant callus lines were identified based on visual growth assessment. These lines were evaluated for 2,4-D tolerance (based on 2,4-D content), using a 2,4-D bioassay procedure which consisted of placing selected callus tissue pieces on top of oat (Avena sativa L.) coleoptile or internode sections. The relative amount of 2,4-D in the callus tissue was estimated by the amount of oat section elongation after 24 h. Two of the more tolerant callus lines had 61% and 83% less 2,4-D in their tissues than the susceptible control tissue. The flea beetle, Phyllotreta nemorum L. (Coleoptera: Chrysomelidae), is an intermediate specialist feeding on a small number of plants within the family Brassicaceae. The most commonly used host plant is Sinapis arvensis L., whereas the species is found more rarely on Cardaria draba (L.) Desv., Barbarea vulgaris R.Br., and cultivated radish (Raphanus sativus L.). The interaction between flea beetles and Barbarea vulgaris ssp. arcuata (Opiz.) Simkovics seems to offer a good opportunity for experimental studies of coevolution. The plant is polymorphic, as it contains one type (the P‐type) that is susceptible to all flea beetle genotypes, and another type (the G‐type) that is resistant to some genotypes. At the same time, the flea beetle is also polymorphic, as some genotypes can utilize the G‐type whereas others cannot. The ability to utilize the G‐type of B. vulgaris ssp. arcuata is controlled by major dominant genes (R‐genes). The present investigation measured the frequencies of flea beetles with R‐genes in populations living on different host plants in 2 years (1999 and 2003). Frequencies of beetles with R‐genes were high in populations living on the G‐type of B. vulgaris ssp. arcuata in both years. Frequencies of beetles with R‐genes were lower in populations living on other host plants, and declining frequencies were observed in five out of six populations living on S. arvensis. Selection in favour of R‐genes in populations living on B. vulgaris is the most likely mechanism to account for the observed differences in the relative abundance of R‐genes in flea beetle populations utilizing different host plants. A geographic mosaic with differential levels of interactions between flea beetles and their host plants was demonstrated.  相似文献

Translocation and complex formation of 14C-labelled 2,4-dichlorophenoxyacetic acid (2,4-D) and 4-amino-3,5,6-trichloropicolinic acid (picloram) in seedlings of sunflower (Helianthus annuus L. var. uniflorus), rape (Brassica napus L. cv. Nilla), wheat (Triticum aestivum L. cv. Starke), and Norway spruce (Picea abies (L.) H. Karst.) were studied. The herbicides were absorbed through the roots from the nutrient solution, Picloram was well translocated to the shoots of the four species; while the acropetal translocation of 2,4-D was small except in rape. In 2,4-D-susceptible sunflower and rape and in picloramsusceptible sunflower and spruce the herbicides were recovered mainly in the uncomplexed form. In 2,4-D tolerant wheat and spruce most of the absorbed 2,4-D was converted into water-soluble or TCA-insoluble complexes. In picloram-tolerant wheat and in relatively picloram-tolerant rape, the absorbed picloram was also converted into complexes recovered predominantly in the water-soluble fraction. Most of the complexes released free herbicides by hydrolyzing in NaOH or HCI. The results further support the hypothesis that complex formation counteracts herbicide toxicity.  相似文献

Summary 2,4-Dichlorophenoxyacetic acid (2,4-D) resistant plants of transgenic cotton (Gossypium hirsutum L.) were produced using Agrobacterium tumefaciens containing a plasmid carrying the neomycin phosphotransferase II (npt II) and 2,4-D monooxygenase (tfd A) genes. An in vitro assay was performed to determine the sensitivity of seed germination, and the growth of seedlings of transgenic and non-transgenic cotton to various concentrations of kanamycin and 2,4-D. The results indicated that kanamycin caused the cotyledons of non-transgenic plants to turn white, but transgenic plants grew normally. Seed germination and seedling growth of non-transgenic plants were strongly inhibited by 2,4-D, but only slightly for transgenic plants. Transgenic plants and non-transgenic plants can be clearly distinguished by the use of 2 mg l−1 2,4-D in seed germination medium. There was a high correlation between the response of seed germination and the growth of seedlings to kanamycin or 2,4-D, based on the germination ration, albino ratio, dry weight or fresh weight. On this basis, we development a rapid method for identifying transgenic plants that has been verified in the field. These findings will allow identification of cotton transformants at an early stage of plant development, saving time and improving cultivars containing the 2,4-D resistance trait.  相似文献

All hybrid plants from interspecific crosses among the three species, Sonchus asper (L.) Hill, S. oleraceus L., and S. arvensis L., were male-sterile, but the progeny of the cross S. arvensis X S. oleraceus and reciprocal cross produced seed after backcrossing to the male parent. Hybrid plants from crosses between S. oleraceus and S. asper lacked vigor. Cytological data indicated that S. arvensis has one genome in common with S. asper and that S. oleraceus is an autotetraploid (2n = 36) with no genomes in common with S. arvensis or S. asper.  相似文献

Plants of the cross Datura stramonium L. var. stramonium×Datura stramonium L. var. inermis (Juss. ex Jacq.) Timm. were sprayed either with a water solution of the sodium salt of 2,4-dichlorophenoxyacetic acid (2,4-D) or with a combination of 2,4-D (sodium salt) and the vitamin K compound menadione sodium bisulfite in water solution. The plants were found to differ in one of their responses to the treatments, namely the development of necrotic spots on the leaf blades. Many necroses appeared on the plants treated with 2,4-D plus menadione sodium bisulfite while only a few necroses or none at all developed on the plants treated with 2,4-D alone. Furthermore, 2,4-D in combination with menadione sodium bisulfite induced much larger necrotic spots than did 2,4-D by itself. Spraying with menadione sodium bisulfite in water solution did not produce any observable effects upon Datura plants. The great increase in necrotic lesion formation was probably due to an interaction between 2,4-D and menadione sodium bisulfite (or a metabolite of this compound) within leaf cells.  相似文献

The diamondback moth Plutella xylostella (L.) (Lepidoptera: Plutellidae) is an important pest of cultivated brassicaceous crops worldwide. The host plant preferences, developmental biology and survival and longevity of P. xylostella are relatively well understood on commercial crop species; however, its relationship with brassicaceous weeds is poorly known. Sinapis arvensis L., Erysimum cheiranthoides L. and Capsella bursa‐pastoris (L.) Medicus are among the most common brassicaceous weeds worldwide and can serve as important bridge hosts of P. xylostella. In this study, preference and performance of P. xylostella were compared on these weed species. In free‐choice situations, females deposited 5.5 and 18.8 times more eggs on S. arvensis than on E. cheiranthoides and C. bursa‐pastoris, respectively. Survival from neonate to pupa and from pupa to adult was highest on S. arvensis and E. cheiranthoides and lowest on C. bursa‐pastoris. Development was fastest, foliage consumption was greatest, pupae and silk cocoons were heaviest, adult body masses and longevities were highest and forewings were largest for both females and males when reared as larvae on S. arvensis. Realized fecundity of new generation adults was highest for individuals reared on S. arvensis compared to those reared on E. cheiranthoides or C. bursa‐pastoris. Relative growth rates of pupae and adults were highest on S. arvensis, suggesting that this plant species is a high‐quality host for P. xylostella compared with other species tested. Potential impacts of these wild brassicaceous species on P. xylostella populations are discussed.  相似文献

Auxin autotrophy was studied in cultured carrot (Daucus carota L.), tobacco (Nicotiana tabacum L.), and potato (Solanum tuberosum L.) cell lines. Of 10 carrot lines resistant to 5-methyltryptophan (5MT), which accumulate free tryptophan (trp) because of an altered control enzyme, 5 were auxinautotrophic while the normal parent line was not. Carrot lines selected from the same parent line as resistant to other amino-acid analogs were not auxinautotrophic, like the parent. The only 5MT-resistant potato line studied was also auxin-autotrophic while the normal line was only partially auxin-autotrophic. The tobacco lines which accumulated free trp were not auxin-autotrophic, and no auxin-autotrophic tobacco lines were selected by screening for growth in medium lacking 2,4-dichlorophenoxyacetic acid (2,4-D). Several auxin-autotrophic carrot and potato lines were selected from the normal lines and none of these lines were resistant to 5MT. Length of time in culture and difficulty in selecting auxin-autotrophic lines were correlated on the 3 normal carrot lines studied. Uptake, translocation and complex formation of 14C-labelled 4-amino-3,5,6-trichloropicolinic acid (picloram) and 2,4-dichlorophenoxyacetic acid (2,4-D) in seedlings of rape (Brassica napus L. cv. Nilla) and sunflower (Helianthus annuus L. var. uniflorus) were studied. Sunflower is susceptible both to 2,4-D and picloram, while rape is susceptible to 2,4-D but more tolerant to picloram. The uptake of the herbicides through the leaves was almost complete in both species. Translocation of 2,4-D into the roots took place more readily than that of picloram. In sunflower about 50 per cent of the applied 2,4-D was extruded through the roots into the nutrient solution after 9 days. In the picloram-treated sunflower most of the activity was found in the aerial parts, while in picloram-treated rape most of the activity still occurred in the treated leaf after 9 days. No activity at all was found in the roots or in the nutrient solution of the picloram-treated rape seedlings. While the major part of 2,4-D always was found in the state of free herbicide, a large fraction of picloram was rapidly bound into water-soluble complexes. This binding was especially pronounced in rape. Separation by paper chromatography showed that different radioactive compounds were formed. Most of these could be hydrolyzed, thereby releasing free herbicide. The results support the hypotheses that complex formation could counteract herbicide translocation and toxicity of auxin herbicides.  相似文献

White mustard (Sinapis alba L.) plants, susceptible to 2,4-D, and scentless mayweed (Tripleurospermum maritimum L.) plants, resistant to 2,4-D, markedly differ in the distribution and metabolism of 2,4-D. When 2,4-D-14C was applied onto the leaf, radioactivity was found in mustard after 8 days in the entire plant, whereas in scentless mayweed radioactivity occurred mainly in the leaf onto which it was applied and in another one or two leaves, with the roots showing only traces of radioactivity. The two plant species differed both in the character and in the number and amount of metabolites detected in aqueous and ether fractions. The metabolite with Rf 0.63 -0.65, soluble in ethylether (39% of the total radioactivity applied), and that with Rf 0.35 -0.36 (23% of the total radioactivity) were present exclusively in the roots of scentless mayweed plants. The Rf values established indicate that the former metabolite may be a conjugate with the amino acids alanine and valine and the latter a hydroxylated derivate of 2,4-D. In the shoots of both plants, 2,4-D-14C was metabolized to identical metabolites with Rf 0.59 -0.61, which occurred in mustard plants only in trace amounts. Free 2,4-D-14C occurred in the shoots of both plants and in considerable amounts also in mustard roots; it could not be demonstrated in the roots of scentless mayweed. The two species did not differ in the uptake rate or in the amount of absorbed 2,4-D-14C.  相似文献

The host range of the knapweed nematode, Subanguina picridis (Kirjanova) Brzeski, under controlled environmental conditions was extended to include, in addition to Russian knapweed, Acroptilon repens (L.) DC., plant species within the Centaureinae, and Carduinae subtribes of the Cynareae tribe of the Asteraceae family. Examination of host response to nematode infection revealed that Russian knapweed was the only highly susceptible host plant. Diffuse knapweed (Centaurea diffusa Lam.) was moderately susceptible, and other plants which formed galls were resistant to S. picridis.  相似文献

Diclofop-methyl (DM) sprayed onto 6–8-week-old plants of leafy spurge ( Euphorbia esula L.) caused senescence and abscission of older leaves, while the young leaves and apex remained attached. The phytotoxicity of DM was reversed by the antioxidant, α -tocopherol (vitamin E), in leafy spurge and DM-susceptible oat ( Avena sativa L. cv. Gary). DM and 2,4-dichlorophenoxyacetic acid (2,4-D) increased ethylene evolution in mature leaves of leafy spurge. Vitamin E reduced the DM-induced ethylene by ampproximately 50%, but had no effect on the 2,4-D-induced ethylene. DM did not increase ethylene in DM-resistant pea or tobacco, but 2,4-D induced a 3-fold increase in ethylene evolution over controls in DM-resistant tobacco. 2,4-D amppears to act at a site different from that of DM in the pathway of ethylene formation. Ethylene evolution increased in DM-treated susceptible biotypes of annual ryegrass ( Lolium rigidum L.) and wild oat ( Avena fatua L.), but not in resistant biotypes of these species. DM reduced root and shoot formation and dry weight in hypocotyl segments of etiolated leafy spurge seedlings grown in vitro. Organogenesis and dry weights were increased by the combination of DM+antioxidants. Vitamin E was a more effective antioxidant than ascorbic acid. These results sumpport the hypothesis that DM induces oxidative stress in susceptible plant tissues and that antioxidants reduce the damaging action of the phytotoxic free radicals.  相似文献

The influences of nutrient supply and plant growth regulators on the phytotoxicity of imazamethabenz in wild oat (Avena fatua L.) were evaluated in the greenhouse. Wild oat plants supplied with half-strength rather than one-eighth-strength Hoagland solution were more susceptible to imazamethabenz, showing greater growth reduction in main shoot and tillers. The improved herbicide efficacy at higher nutrient levels appeared related to increased herbicide interception by the greater leaf surface available. Leaves developing at either nutrient level did not differ significantly in epicuticular wax, so differential absorption appeared unlikely. Wild oat plants supplemented with nutrient, switching from low to high levels at the time of herbicide application, were as susceptible to imazamethabenz or even more so than plants growing with a constant high level of nutrition. The wild oat pure-line Montana 73, a strongly tillering line, was more susceptible to imazamethabenz than the limited-tillering line, Crop Science 40. Both 2,4-D and GA3 reduced imazamethabenz-induced tillering. Imazamethabenz efficacy was increased by GA3 but not by 2,4-D. These results support the hypothesis that lowering apical dominance of wild oat increases imazamethabenz activity in tillers, and that increased tillering following sublethal doses of imazamethabenz treatment is associated with the release of apical dominance.  相似文献

Nectar-carbohydrate production and composition were investigated by high-performance liquid chromatography and enzymology in nine species from five tribes of the Brassicaceae. In six species (Arabidopsis thaliana (L.) Heynh., Brassica napus L., B. rapa L., Lobularia maritima (L.) Desv., Raphanus sativus L., Sinapis arvensis L.) that produced nectar from both lateral nectaries (associated with the short stamens) and median nectaries (outside the long stamens), on average 95% of the total nectar carbohydrate was collected from the lateral ones. Nectar from these glands possessed a higher glucose/fructose ratio (usually 1.0–1.2) than that from the median nectaries (0.2–0.9) within the same flower. Comparatively little sucrose was detected in any nectar samples except from Matthiola bicornus (Sibth. et Sm.) DC., which possessed lateral nectaries only and produced a sucrose-dominant exudate. The anatomy of the nectarial tissue in nectar-secreting flowers of six species, Hesperis matronalis L., L. maritima, M. bicornus, R. sativus, S. arvensis, and Sisymbrium loeselii L., was studied by light and scanning-electron microscopy. Phloem alone supplied the nectaries. However, in accordance with their overall nectar-carbohydrate production, the lateral glands received relatively rich quantities of phloem that penetrated far into the glandular tissue, whereas median glands were supplied with phloem that often barely innervated them. All nectarial tissue possessed modified stomata (with the exception of the median glands of S. loeselii, which did not produce nectar); further evidence was gathered to indicate that these structures do not regulate nectar flow by guard-cell movements. The numbers of modified stomata per gland showed no relation to nectar-carbohydrate production. Taken together, the data on nectar biochemistry and nectary anatomy indicate the existence of two distinct nectary types in those Brassicacean species that possess both lateral and median nectaries, regardless of whether nectarial tissue is united around the entire receptacle or not. It is proposed that the term “nectarium” be used to represent collectively the multiple nectaries that can be found in individual flowers. Esters of substituted phenoxy-phenoxy propionic acid constitute a new class of herbicides that are effective against gramineous weed and crop species. Slight changes in chemical structure alter drastically the spectrum of weeds controlled by this class of herbicides. Wheat (Triticum aestivum L.) is resistant to diclofop-methyl (methyl 2-[4-(2′,4′-dichlorophenoxy)phenoxy] propanoate) (DM) and clofop-isobutyl (iso-butyl 2-[4-(4′-phenoxy)phenoxy] propanoate) (CI), oat (Avena sativa L.) and wild oat (Avena fatua L.) are susceptible to DM but resistant to CI, and corn (Zea mays L.) is susceptible to both compounds. The antagonism of IAA-induced elongation in the coleoptile straight growth test was determined to measure biological activity of the herbicides. The basis for the differential responses by gramineous species was related to the metabolism and deioxication of the herbicides in coleoptiles. Growth of wheat coleaptiles was relatively unaffected by both compounds, oat coleoptile growth was inhibited by DM but not by CI. but corn coleoptile growth was inhibited equally by both compounds. Coleoptiles and excised shoots of the three species rapidly hydrolyzed both compounds to their respective acids (diclofop, clofop). The acids were conjugated to a water-soluble ester conjugate or they were hydroxylated in the chlorine-substituted phenyl ring and conjugated as a phenolic conjugate. Aryl hydroxylation is a detoxication mechanism in resistant plants. Plants resistant to DM or CI contained low concentrations of the parent ester and the free or bound (ester conjugate) acid and a high concentration of free or bound (phenolic conjugate) aryl hydroxylated acid in coleoptile and shoot tissues, Differential responses by the three gramineous species to DM and CI axe due apparently to differences in their detoxication mechanism. The enzyme for aryl hydroxylation in oat appears to have a higher affinity for the 4-chloro- than for the 2,4-dichloro-substituted moiety. Therefore, oat hydroxylated clofop rapidly and was tolerant to CI but the limited ability of oat to hydroxylate diclofop resulted in oat being extremely susceptible to DM.  相似文献

The essential oils from the aerial parts of catmint (Nepeta meyeri Benth.) were analyzed by hydrodistillation with GC–MS. Fourteen compounds were identified in the yellowish essential oil of the plant, representing more than 99.07% of the oil, of which the major components were found to be 4aα,7α,7aβ-nepetalactone (83.4%) and 4aα,7α,7aα-nepetalactone (8.83%). The oils were characterized by relatively high content of oxygenated monoterpenes, and were tested on the germination and antioxidative systems in early seedlings of seven weed species (Amaranthus retroflexus L., Bromus danthoniae Trin., Bromus intermedius Guss., Chenopodium album L., Cynodon dactylon L., Lactuca serriola L., and Portulaca oleracea L.) and autotoxicity. The essential oil of N. meyeri inhibited seed germination by more than 50% in three weed species (B. danthoniae, B. intermedius, and L. serriola) when applied at a concentration of 0.01%. When the same oils were applied at 0.02% concentration, the inhibition of germination was more than 70% in two weeds (C. album and C. dactylon) and was 100% in four weeds (A. retroflexus, B. danthoniae, B. intermedius, and L. serriola). The essential oils increased CAT activity in all the weed species and decreased SOD activity, except in A. retroflexus. POX activity did not exhibit a revealing situation in the weed species tested. The essential oils increased the level of lipid peroxidation and hydrogen peroxide (H2O2) concentration in all the weeds studied. Our results show that the essential oils of N. meyeri have an important phytotoxic effect on seed germination and, consequently, seedling growth by exhausting antioxidative system of the weeds. The phytotoxic activity of the essential oils may be attributed to their relatively high content of oxygenated monoterpenes, especially 4aα,7α,7aβ-nepetalactone. It can be suggest that the essential oils of N. meyeri have the potential to be used as a bioherbicide.  相似文献

Summary Mutant lines of Arabidopsis thaliana resistant to the artificial auxin 2,4-dichloro phenoxyacetic acid (2,4-D) were isolated by screening for growth of seedlings in the presence of toxic levels of 2,4-D. Genetic analysis of these resistant lines indicated that 2,4-D resistance is due to a recessive mutation at a locus we have designated Axr-1. Mutant seedlings were resistant to approximately 50-fold higher concentrations of 2,4-D than wild-type and were also resistant to 8-fold higher concentrations of indole-3-acetic acid (IAA) than wild-type. Labelling studies with (14C)2,4-D suggest that resistance was not due to changes in uptake or metabolism of 2,4-D. In addition to auxin resistance the mutants have a distinct morphological phenotype including alterations of the roots, leaves, and flowers. Genetic evidence indicates that both auxin resistance and the morphological changes are due to the same mutation. Because of the pleiotropic morphological effects of these mutations the Axr-1 gene may code for a function involved in auxin action in all tissues of the plant.  相似文献
