We studied Holocene speleothems and tufa samples collected in numerous caves and rivers in the Dinaric Karst of Croatia, Slovenia, Bosnia and Herzegovina, as well as Serbia and Montenegro. Differences in the formation process of tufa and speleothems are discussed in the context of their isotopic composition (14C, 13C and 18O), as well as the chemistry of surface water (rivers, lakes) and drip water (in caves). The physical and chemical parameters monitored in the surface water (tufa precipitation) and drip water (speleothem precipitation) show that more stable conditions accompany speleothem rather than tufa formation. This is particularly obvious in the water temperature variations (2-22°C in surface water and 7-12°C in drip water) and in saturation index variation (3-11 in surface water and 1-6 in drip water). The range of 14C ages recorded by Holocene speleothems (∼12?000 yr) is wider by several thousands years than that of Holocene tufa samples (∼6000 yr). δ13C values for tufa samples range from −12‰ to −6‰ and for speleothem samples from −12‰ to +3‰ reflecting higher soil carbon and/or vegetation impact on the process of tufa than on speleothem formation. The differences in δ18O values of tufa and speleothem samples from different areas reflect different temperature conditions and differing isotopic composition in the water. The study shows that speleothems from the Dinaric Karst can be used as global palaeoclimatic records, whereas tufa records changes in the local palaeoenvironment.  相似文献

Using an immunoblotting technique and goat antihuman C4, we observed five distinct electrophoretic variants of C4 in a panel of 60 random dogs. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis of immunoprecipitated C4 showed that dog C4 is composed of three polypeptide subunit chains (, , and ) and that structural variability occurs within the - and -chain regions. Two distinct molecular weight forms of both the C4- ( A and B) and C4-( A and B) chain were detected. The variant forms of C4 and C4 were found in association with particular C4 allotypes.  相似文献

Four populations of Mollugo verticillata L. were compared on the basis of their photosynthetic products, photosynthetic rates, enhancement under low oxygen concentration, and CO2 compensation points. In addition, pulse-chase labeling experiments were conducted using one of the four populations. Depending on the plant population, C4 acids ranged from 40% to 11% of the primary products under short-term exposure to 14CO2. These compounds were also metabolized during pulse-chase experiments. All four populations had significantly different photosynthetic rates and those rates were correlated with the amounts of labelled C4 acids produced and C4-acid turnover. Three populations of M. verticillata had similar compensation points (40 l/l) and degrees of photosynthetic enhancement under low [O2] (20%), the fourth population was much lower in both characteristics (CO2 compensation, 25 l/l; low-O2 enhancement, 12%). The 13C/12C ratios of leaves of the conifer morphotype Frenelopsis were measured to decipher the influences of water and salt stress on stomatal density (SD), epidermal cell density (ECD) and stomatal index (SI). Three morphospecies were analyzed: F. ugnaensis from freshwater fluvio-lacustrine deposits (Upper Barremian), F. turolensis and alata from coastal deposits (Lower-Middle Albian and Upper Albian respectively). The cuticle δ13C values show a large variation from ? 28‰ to ? 21‰. Comparison with previously published marine carbonate δ13C records indicate that the difference in cuticle δ13C between the different deposits are mainly due to difference in CO2-plant isotope fractionation rather than to change in isotopic composition of inorganic carbon in the atmosphere and ocean. The less negative δ13C and wide range in δ13C of F. turolensis and alata (? 27.5 to ? 21‰), compared to F. ugnaensis, (? 28 to ? 25‰) are interpreted as a result of salt and/or water stress. The data as a whole yield a good relationship between the 13C/12C ratio and SD (r = 0.67, n = 42, p < 0.001), SI (r = 0.53, n = 41, p < 0.001), hence suggesting that the differences in SD and SI between the three morphospecies are related to freshwater/saline environment. Looking at single morphospecies, the SD of F. ugnaensis decreases with increasing δ13C value (r = ? 0.57, n = 15, p = 0.026) as well as a decrease of SI (r = ? 0.62, n = 15, p = 0.013), possibly reflecting warmer and drier conditions. Average SI of F. alata does not significantly change with δ13C and inferred soil salinity in contrast to SD (p < 0.01).  相似文献

Albinism is a heterogeneous group of genetic disorders resulting from deficiencies in pigmentation. Clinically, it is divided into ocular (OA) and oculocutaneous albinism (OCA). OCA involves lack of pigment in the skin, hair, and eyes and results from mutations in the tyrosinase gene or in the P gene. OA mainly affects pigmentation in the visual system and may be a mild form of OCA or may be caused by other genetic defects. Clinical diagnosis of albinism type is difficult, because of the observed range of phenotypic variation. Thus, genetic analysis may be helpful with respect to a more accurate diagnosis. Here, we report the mutational profile, determined by genetic analysis of the tyrosinase and P genes, of a large German albino population. We have revealed a total of 42 distinct mutations, 19 of which are novel. Of the 74 unrelated patients screened, 32 (43%) had mutations in the tyrosinase gene, 16 (22%) had P gene mutations, and 26 (35%) patients had no detectable genetic abnormalities. This defines a population of albino patients who are tyrosinase-gene- and P-gene-negative and who thus may represent a good study group for searching for additional genes associated with albinism.  相似文献

Purpose

Inflammation may contribute to the pathogenesis of specific cardiovascular diseases, but it is uncertain if mediators released during the inflammatory process will affect the continued efficacy of drugs used to treat clinical signs of the cardiac disease. We investigated the role of the complement 5a receptor 1 (C5aR1/CD88) in the cardiac response to inflammation or atenolol, and the effect of C5aR1 deletion in control of baseline heart rate in an anesthetized mouse model.

Methods

An initial study showed that PMX53, an antagonist of C5aR1 in normal C57BL6/J (wild type, WT) mice reduced heart rate (HR) and appeared to have a protective effect on the heart following induced sepsis. C5aR1 knockout (CD88-/-) mice had a lower HR than wild type mice, even during sham surgery. A model to assess heart rate variability (HRV) in anesthetized mice was developed to assess the effects of inhibiting the β1-adrenoreceptor (β1-AR) in a randomized crossover study design.

Results

HR and LF Norm were constitutively lower and SDNN and HF Norm constitutively higher in the CD88-/- compared with WT mice (P< 0.001 for all outcomes). Administration of atenolol (2.5 mg/kg) reduced the HR and increased HRV (P< 0.05, respectively) in the wild type but not in the CD88-/- mice. There was no shift of the sympathovagal balance post-atenolol in either strains of mice (P> 0.05), except for the reduced LF/HF (Lower frequency/High frequency) ratio (P< 0.05) at 60 min post-atenolol, suggesting increased parasympathetic tone of the heart due to the effect of atenolol administration. The HR of the WT mice were lower post atenolol compared to the CD88-/- mice (P = 0.001) but the HRV of CD88-/- mice were significantly increased (P< 0.05), compared with WT mice.

Conclusion

Knockout of the C5aR1 attenuated the effect of β1-AR in the heart, suggesting an association between the β1-AR and C5aR1, although further investigation is required to determine if this is a direct or causal association.  相似文献

We evaluated the effects of ecosystem composition and structure (species richness and diversity, cover and spatial attributes of vegetation), abiotic factors (climate and topographical features) and the condition of the bare-ground areas (evaluated using soil-surface indicators) on the performance of Stipa tenacissima [evaluated using foliar 13C, 15N, nitrogen concentration and the carbon-to-nitrogen (C:N) ratio] in 15 steppes of SE Spain. Foliar 13C values of S. tenacissima showed a low degree of variation in the studied steppes, with average values ranging from –24.1 to –22.9. Higher variation was found in the 15N values, which ranged from –5.5 to –2.4. The nitrogen concentration and the C:N ratio varied between 5.0 and 8.0 mg g–1, and between 55.4 and 85.3, respectively. The 13C values became less negative with increasing spatial aggregation of perennial vegetation, while the C:N values increased with increasing perennial vegetation cover. The 15N values became more negative with increasing infiltration in the bare-ground areas, but the nitrogen concentration was not related to any of the environmental variables measured. Our results suggest that the relative importance of ecosystem structure and soil-surface conditions in the bare ground areas was higher than that of abiotic factors as determinants of the performance of S. tenacissima. The results also show that even subtle changes in these ecosystem features may lead to modifications in plant performance in semiarid S. tenacissima steppes, and thus to modifications in the associated ecosystem functions in the mid- to long-term.  相似文献

Modelling of the carbon dynamics in arable soils is complex and the accuracy of the predictions is unknown before the model is applied to each specific site. Objectives were (i) to test the accuracy of predictions of the carbon dynamics using the Rothamsted Carbon (RothC) Model in a field trial in Quzhou, North China Plain, using different methods for initialization and estimation of carbon input into the soil and (ii) to test the applicability of the RothC model for plots with either conventional tillage (CT) or no-tillage (NT) systems. A field trial was conducted with applications of differing amounts of N (0, 112 or 187 kg N ha?1 year?1), P (0, 75 or 150 kg P2O5 ha?1 year?1) and wheat straw (0, 2.25 or 4.5 t DM ha?1 year?1) in differing combinations with either CT or NT for 18 years. CT and NT affected stocks of soil organic carbon (SOC) similarly. Carbon inputs from crops were either estimated from published regression functions that relate C inputs to crop yield including rhizodeposition (models 1 and 2) or published root:aboveground biomass ratios (model 3). Model 1, which was not calibrated to the site conditions, was successful in predicting the carbon dynamics in seven out of nine treatments (model efficiencies EF ranged from 0.28 to 0.87), whereas for two treatments, EF (?0.35 and?2.3) indicated an unsuccessful prediction. The prediction of the C dynamics in NT experiments using model 1 was generally successful, but this may have been due to the fact that NT did not have a specific effect on SOC stocks for this trial. Model 2, which was the same as model 1 except for an optimization of the stock of inert organic matter using one treatment, predicted SOC stocks in the remaining eight treatments overall better than model 1. Model 3 was less successful than models 1 and 2 in all treatments (?19 ≤ EF ≤ 0.56). The results indicate that the RothC model may successfully predict C dynamics—for the site studied even without prior calibration as in model 1—, but care should be taken in choosing an appropriate approach for estimating C inputs into the soil.  相似文献

The bond dissociation energies of the Co–C bonds in the cobalamin cofactors methylcobalamin and adenosylcobalamin were calculated using the hybrid quantum mechanics/molecular mechanics method IMOMM (integrated molecular orbital and molecular mechanics). Calculations were performed on models of differing complexities as well as on the full systems. We investigated the origin of the different experimental values for the Co–C bond dissociation energies in methylcobalamin and adenosylcobalamin, and have provided an explanation for the difficulties encountered when we attempt to reproduce this difference in quantum chemistry. Additional calculations have been performed using the Miertus–Scrocco–Tomasi method in order to estimate the influence of solvent effects on the homolytic Co–C bond cleavage. Introduction of these solvation effects is shown to be necessary for the correct reproduction of experimental trends in bond dissociation energies in solution, which consequently have no direct correlation with dissociation processes in the enzyme.  相似文献

This study reports on the flowering phenology of 12 Poaceae species in different types of plant cover (scrub, riverbank and pasture), with a view to creating a phenological calendar for the major species contributing most to airborne pollen concentrations. The study has been carried out from March to June between 2000 and 2013. Seventeen sampling sites in the study area were selected by accessibility, and predominantly, grasses grow in different land covers for phenological monitoring. Airborne pollen was collected using a Hirst-type volumetric spore trap. Results showed that longer phenological ranges were recorded in the wettest year and shorter ranges in the driest year. Over the study period, certain species displayed changes in the timing of flowering phenology. In pastureland and scrubland, a slight phenological delay over the study period has been recorded. In pastureland, some species—i.e., Bromus madritensis, B. diandrus and B. hordeaceus, Hordeum leporinum and Stipa capensis—displayed a delay in the timing of phenophases over the study period, including B. hordeaceus and B. madritensis on riverbank. Dactylis glomerata and Trisetaria panicea in scrub have been the only species with a clear advance; B. diandrus and B. madritensis a clear flowering delay. Lolium rigidum, Dactylis glomerata, Aegilops geniculata, in scrub, riverbank and pasture, and Arrhenatherum album, only in scrub, full flowering coincided with pollen peak in some populations during several years.  相似文献

Abstract

Major efforts are currently underway to dissect the phenotype–genotype relationship in plants and animals using existing populations. This method exploits historical recombinations accumulated in these populations. However, linkage disequilibrium sometimes extends over a relatively long distance, particularly in genomic regions containing polymorphisms that have been targets for selection. In this case, many genes in the region could be statistically associated with the trait shaped by the selected polymorphism. Statistical analyses could help in identifying the best candidate genes into such a region where an association is found. In a previous study, we proposed that a fragment of the PHYTOCHROME C gene (PHYC) is associated with flowering time and morphological variations in pearl millet. In the present study, we first performed linkage analyses using three pearl millet F2 families to confirm the presence of a QTL in the vicinity of PHYC. We then analyzed a wider genomic region of ~100 kb around PHYC to pinpoint the gene that best explains the association with the trait in this region. A panel of 90 pearl millet inbred lines was used to assess the association. We used a Markov chain Monte Carlo approach to compare 75 markers distributed along this 100-kb region. We found the best candidate markers on the PHYC gene. Signatures of selection in this region were assessed in an independent data set and pointed to the same gene. These results foster confidence in the likely role of PHYC in phenotypic variation and encourage the development of functional studies.  相似文献

The genus Lippia comprises herbs, shrubs, and small trees, including many species with medicinal properties. The species are distributed throughout South and Central America and Tropical Africa, but the majority of them occur in Brazil, Paraguay, and Argentina. The DNA?C value of 28 Brazilian species has been estimated by flow cytometry. Estimated DNA?C values ranged from 0.825?pg (L. corymbosa) to 2.150?pg (L.?brasiliensis). In addition, new chromosome numbers of 12 species have also been described, and meiotic cells with 12, 13, and 14 chromosome pairs were observed. A straightforward correlation between chromosome number and DNA?C value was not observed, probably due to two outlier species of Lippia that have been transferred from the genus Lantana. In general, the data confirm previous reports regarding the variation within the taxonomic sections and also suggest a new revision in section Zapania. Aspects of karyotypic evolution of the genus are also discussed.  相似文献

Wounded cells such as Xenopus oocytes respond to damage by assembly and closure of an array of actin filaments and myosin-2 controlled by Rho GTPases, including Rho and Cdc42. Rho and Cdc42 are patterned around wounds in a characteristic manner, with active Rho concentrating in a ring-like zone inside a larger, ring-like zone of active Cdc42. How this patterning is achieved is unknown, but Rho and Cdc42 at wounds are subject to regulation by other proteins, including the protein kinases C. Specifically, Cdc42 and Rho activity are enhanced by PKCβ and inhibited by PKCη. We adapt a mathematical model of Simon and coworkers to probe the possible roles of these kinases. We show that PKCβ likely affects the magnitude of positive Rho–Abr feedback, whereas PKCη acts on Cdc42 inactivation. The model explains both qualitative and some overall quantitative features of PKC–Rho GTPase regulation. It also accounts for the previous, peculiar observation that ∼20% of cells overexpressing PKCη display zone inversions—that is, displacement of active Rho to the outside of the active Cdc42.  相似文献

The objective of this investigation was to evaluate the frequency of chromosomal aberrations expressed as micronuclei (MN) in 4–8 cell embryos generated by gamma-irradiation of female mice in the absence and in the presence of vitamin C. Female NMRI mice were whole body exposed to 4 Gy gamma-irradiation after intraperitoneal (i.p.) injection of pregnant mare’s serum gonadotrophin (PMSG) followed by injection of human chorionic gonadotrophin (HCG) and mating with non-irradiated NMRI male mice. Pregnant animals were sacrificed and embryos flushed from the oviducts and fixed on slides. Cells were treated for MN observation using standard method. To investigate the protective effect of vitamin C (ascorbic acid) on the frequency of MN, 100 mg/kg vitamin C was i.p. injected 1 h before irradiation. Results show that the frequency of MN generated in the embryos of irradiated mother compared to those of control in the non-irradiated group increased dramatically (P < 0.001). Frequency of MN in embryos generated in irradiated female mice treated with vitamin C dramatically and statistically decreased relative to the frequency observed in the irradiation only group (P < 0.001). This decrease returned the combined treatment group to a level that was not statistically different from the controls (P > 0.05). Thus, irradiation of preovulatory stage oocytes leads to stable chromosome abnormalities expressed as micronuclei in successive preimplantation embryos. Vitamin C reduces these clastogenic effects of radiation in preovulatory oocytes and thus the reduced frequency of MN in embryos is probably due to its antioxidation and radical scavenging properties.  相似文献

The diversification and distribution of Abies species throughout the Mediterranean region has led to a complex of species which are difficult to classify. An open question is whether these mainly allopatric taxa have exchanged genetic information via secondary contact. We studied the variation and geographic distribution of paternally inherited chloroplast DNA markers in nine Mediterranean Abies taxa. Markers with high and low mutation rates were applied in order to differentiate among a scenario of secondary genetic contact and a scenario of complete isolation after speciation. The observed molecular variation was analysed using statistical parsimony, geostatistics, and measures of population genetic differentiation. Ancient paternal lineages, represented by markers with low mutation rates, were shared among species. The central and widespread A. alba retained all ancient lineages whereas other species exhibited fewer, down to a single lineage. In contrast, modern lineages, depicted by markers with high mutation rates, were largely separated among species. The western Mediterranean A. pinsapo and A. numidica were clearly separated from each other and from the remaining Abies species. This indicates the absence of secondary contact. The same scenario applies to the eastern Mediterranean Abies species. An exception is the parapatric complex of A. alba, A. cephalonica, and their supposed hybrid A. borisii regis, which exhibited evidence of secondary contact.  相似文献

Summary The 13C values of whole body samples of the beetle Tribolium castaneum are closely correlated with the 13C values of the plant carbon in its diet. The correlation is always high for diets ranging from 100% C4 to 100% C3 plant material. The degree of correlation is independent of the growth rate of the animals.  相似文献
