The aging is associated with alterations in the hypothalamic-pituitary-thyroidal axis which can lead to hypothyreosis. Our previous investigations has shown that polyphenol curcumin can enhance the manifestation of hypothyreosis in rats simultaneous treated with propylthiouracil. The aim of the study was to investigate the relationship between age-related changes and curcumin action in the thyroid of old rats. To this end, morphometric and radioimmunological methods were used. The study was conducted on 3- and 18-month-old male Wistar rats. The experimental rats were treated daily for 30 days by gavage with 100 mg/kg b.w. of curcumin. There were observed age-related changes in morphology and endocrine function of the thyroid. It was increase in the percentages of large follicles and significant decrease in FT3 level in 18-month-old rats in comparison to 3-month ones. Curcumin treatment lead to significant increase in FT3 and FT4 levels in 3-month-old experimental rats, but the level of FT3 significantly decreased in 18-month-old rats after curcumin administration. Our results show that curcumin activity depends on the functional condition of the rat thyroid which changes with age. This compound exerts stimulatory influence on the secretory function of the thyroid gland in young rats, but has rather weak antithyroid activity in old animals.  相似文献

The investigation was carried out on 65 adult white male rats (12-month-old) after normal development or chemical sympathectomy by guanethidine and subtotal thyroidectomy. By means of electronic microscopic, autoradiographic and radioimmunological methods the increased functional activity in thyroid gland of chemically sympathectomized rats was found. This is mainly due to thyrotropin hyperproduction and intensification of proliferative potential of the organ. Partial resection of sympathectomized rats thyroid gland leads to exhaustion of adenohypophysis compensatory potential manifested in disjunction of integrative connections in hypothalamus-hypophysis-thyroid gland system and delay of thyroid status normalization.  相似文献

A considerable decrease in the amount of sympathetic nervous cells in neonatal rats injected guanethidine resulted in the depression of thyroid gland functional activity in 1-month-old animals, there was an apparent parallel increase in the proliferative activity of the gland. A 2/3 thyroidectomy led to the functional tension of the remaining part of the organ. A comparison of compensatory-reparative processes after thyroidectomy has shown similarity of reparative changes in experimental and control rats, however, the degree of repair potential was much higher in the sympathectomized thyroid gland.  相似文献

A study of the morphological structure and functional activity of the rat thyroid gland was carried out after 22 months following a single exposure to external radiation. The 3-month-old animals were irradiated with doses of 0.25, 0.5, 1.0, 2.0 and 5.0 Gy. Blood was assayed for thyroxin (T4) and triiodothyronine (T3) levels, while liver tissue--for NADP-MDH activity and thyroid tissue--for thyroperoxidase activity. The thyroid was studied histologically, morphometrically and by electron microscope. The decreased T4 concentrations 2.59-fold in the 5.0 Gy group, the increased T3/T4 in the 2.0 and 0.25 Gy groups, the reduced diameter of cellular nuclei and follicles, the flat follicular epithelium and diminished number of thyrocyte ultrastructures indicate thyroid hypofunction in the irradiated animals. The morphological changes are characterized by enhanced diffuse and focal sclerotic changes in thyroid, most pronounced at high irradiation doses (1.0-5.0 Gy), whereas the hemosiderosis foci suggest that the structural changes are consequences of radiation-induced destructive injuries in the gland parenchyma. Two of the thyroids (0.5 Gy) demonstrate foci with pronounced lymphoid infiltration, while follicular carcinomas were detected in 4 thyroids (2.0 Gy), and in one thyroid (0.5 Gy) in one thyroid (5.0 Gy). The remote effects of radiation were dose-dependent destructive, sclerotic and atrophic processes, decreased functional activity, stimulation of development of autoimmune aggression and carcinogenesis in thyroid.  相似文献

The authors observed signs of premature maturation and activation of the hypophysio-adrenocortical system at the early postnatal period (the 70--90th hour) in the progeny of rats adrenalectomized 1 to 3 and 7 to 9 days before labour. A more pronounced in comparison with control (progeny of the sham-adrenalcetomized and intact rats) response to stress pointed to the early maturation of the suprahypophysial control mechanisms as well. Signs of functional exhaustion in the hypophysio-adrenocortical system and also in the thyroid gland were revealed in mature (6--10-month-old) rats born of adrenalectomized rats.  相似文献

The morphological and functional changes in the thyroid tissue of white rats injected lithium chloride at doses 0.5 mekv/kg and 1.0 mekv/kg (groups 1 and 2, respectively) during three weeks were studied by the radiometric, histological and biochemical methods. The radiometric, histological and biochemical methods. The inhibitory action of lithium chloride on hormonogenesis in the thyroid gland and secretion of thyroid hormones into the blood was proved to be directly related to the lithium dose and concentration in the blood. The data of intravital radiometric and morphological analysis of the organs of the 1st group animals suggest some activization of the gland function, with the secretion of the hormones into the blood being suppressed. Increased concentration of the drug inhibits hormonogenesis and secretion of thyroid hormones into the blood.  相似文献

The functional state of the thyroid gland and the concentration of thyroid hormones in the peripheral blood were studied in 20 mature female albino rats during their estral cycle. Evaluation of the thyroid functional state was made according to data of histological, morphological (the diameter of folliculi, the height of the thyroid epithelium) and histochemical analysis (determination of NAD and NADP-dehydrogenase, succinatedehydrogenase, lactate dehydrogenase, peroxydase, acid and alkaline phosphatase) as well as biochemical determination of iodine bound with protein (IBP) in the blood plasma and investigation of the ratio of the parameters in question under conditions of the sex cycle. The cyclic changes of the morphological state of the thyroid gland attended by the phases of the estral cycle were revealed. The activation of the organ was observed in proestrus and estrus which was evidenced by high levels of activity of the enzymes under study, high concentration of IBP in the blood and increased height of thyreocytes. A decreased function of the thyroid parenchyma was observed at the period of metaestrus-diestrus.  相似文献

Population of the thyroid C-cells, normal and at sympathectomy has been analyzed in 75 white male rats at the age of 1, 3 and 6 months by means of electron microscopical, morphometrical and radioimmunological methods. Partial sympathectomy has been performed using subcutaneous injection of guanethidine (15 mg/kg of body mass) during 14 days after birth. During the period from 1 up to 6 months of life in intact rats a decrease in C-cells functional activity is observed. Under conditions of sympathectomy in 30-day-old animals decreasing extrusion processes of the secretory material are observed. During successive periods of life (3 and 6 months) mechanisms of paracrinic evacuation of hormonal products enhance considerably, nuclear volume of the cells and number of C-cells in the field of vision increase. Their hyperplastic alterations in the sympathectomized thyroid gland are more pronounced in 3-month-old animals.  相似文献

The changes of the thyroid gland and neurocytes of the cranial sympathetic ganglia were followed in rats of different ages after guanethidine injections with the use of radioimmunological assay, electron microscopy and morphometry. The injection of 15 mg of the drug per kg of body weight within the first two weeks after birth caused the death of over 80% of the cells in the sympathetic ganglion. In the sympathectomized 15-day- and 1-month-old rats the functional activity of the thyroid gland was markedly reduced. Later on, intrathyroid hormonogenesis somewhat increases due, apparently, to partial recovery of the organ adrenergic innervation and increase in the production of thyrotropic hypophysial hormone and calcitonin.  相似文献

The effect was studied of biochemical and morphological changes induced by antithyroid substances (PTU, C10(-4)) on proton spin-relaxation properties of rat thyroid gland. It was found that thyroid stimulated by PTU (0.05%) or C10(-4) (1.0%) exhibit marked morphological changes (hyperplasia and epithelial hypertrophy) with alteration of the soluble iodoprotein pattern (content and composition.). Both relaxation times spin-lattice (T1) and spin-spin (T2) were increasing with the lenght of treatment with antithyroid drugs. Reversibility of the process was noted in accordance with biochemical and morphological data. The relaxation rate (formula: see text) for thyroid tissue water was in positive correlation with the suluble protein concentration and particularly with the TG content in the gland. There was no difference in relaxation times between normal thyroid and gland of rats treated chronically with excess iodide. The observed difference in T1 between normal glands and glands of PTU,-C10(-4)--treated rats was comparable with that found in cases of human thyroid cancer. This finding is of importance when the diagnostic potential of NMR in the detection of malignancy is considered. In conclusion, a strong correlation was found between microstructural and biochemical changes of the thyroid gland and proton magnetic relaxation of tissue water. The striking difference between the proton spin-relaxation times in normal and in goiter thyroid glands of rats suggests that pulsed NMR spectroscopy could be a method for evaluation of some disturbances in thyroid gland.  相似文献

In 172 white male rats 1- and 3-month-old, using electron microscopical, autoradiographical, morphometrical and radioimmunological methods, analysis of structural-functional changes of the thyroid gland has been carried out both under normal conditions and at guanethidine desympathization (15 mg/kg for 14 days after birth). Resection of 2/3 of the organ has been performed on the 3d, 5th, 7th and 15th days before sacrifice. In the 1-month-old desympathized rats hypofunctional state of the gland is observed. By the 3d month at the expense of an increased production of thyrotropin and calcitonin the thyroid status becomes normal. The dynamics of the restorative processes in the thyroid gland after resection of 1/3 of the organ in the control and test rats occurs with a similar intensity. Proliferative potency of the desympathized thyroid tissue is much higher.  相似文献

The total protein glycosylation profile and specific activity of lysosomal enzymes were investigated in rat submandibular glands isolated from very young (1-month), young (1.5-months) and adult rats (3-months) rats. The specific activity of lysosomal hydrolases (i.e. acid phosphatase, arylsulfatases A and B, beta-N-acetyl-D-glucosaminidase, beta-galactosidase and beta-glucuronidase) decreased in parallel to increasing age of the animals. Furthermore, the thermal stability of acid phosphatase and beta-N-acetyl-D-glucosaminidase was influenced by the age of rats. Age-related changes in protein profile regarding the intensity of particular bands as well as the appearance of certain proteins limited to special age groups were also demonstrated as revealed by Coomassie and lectin staining. Moreover, the marked age-related increase in structures Man (alpha1-2, alpha1-3, alpha1-6) Man, Fuc (alpha1-6) GlcNAc as well as Gal (beta1-3) GlcNAc was observed, whereas staining with terminal NeuAc and GlcNAc showed an inverse correlation. The reaction with (beta1-6) branched N-glycans and Gal (beta1-3) Gal structures was limited to 1-month-old rats. No significant changes in a specific reaction with NeuAc (alpha2-3) Gal were observed. We speculate that the observed differences with respect to protein and glycosylation profiles between 1-month-old rats and older ones could be caused by a modification of the diet composition as well as by the functional and morphological maturation of the rat submandibular gland.  相似文献

In Wistar rats a study was made of a functional condition of the thyroid gland 1, 6 and 24 hours after partial pancreatectomy. The following signs-served as criteria of a functional condition of the gland: the amount of neutrone-activated iodine in iron, iodine-absorptive capacity of the gland by I131, the amount of protein-bound stable and radioactive iodine in the blood, a morphological condition of the gland structures. It was demonstrated that, despite the initial intensification of the process of synthesis and secretion in the thyroid gland, its secretory activity fell by the end of the experiment. This reduction is regarded as an adaptive response to partial insulin deficiency.  相似文献

A study of the morphological structure of the thyroid was made at the acute (1, 3, 6 hours) and remote periods (1, 4, 24, 48 weeks) following the external gamma-irradiation at doses of 0.25-5.0 Gy using histologic and electron microscopic methods. At the acute period, morphological changes prevailed, which reflect the functional alterations (changes of the nucleic and of the follicular diameters and of the colloid density). Frequently they hare a non-synchronous character. At more remote periods, destructive (colloidorrhage, desquamation, hemorrhage) and reparative changes reveal in thyroid, which and by the development of the sclerotic processes and of the morphological signs of decreased functional activity of the gland (increased follicular size and colloidal density, flatness of follicular epithelium and pronounced decreasing of the thyrocytes ultra structure number). The presence of such morphological changes after high and low-dose irradiation (<1.0 Gy) shows high sensitivity of the thyroid to single external radiation.  相似文献

The aim of this study was to examine the effects of dexamethasone (Dex) on functional properties of the rat insulin receptor (IR). Male Mill Hill hooded rats, 3, 6, 12, 18 and 21 months old, were injected with Dex (4 mg/kg) and rat liver and erythrocytes were used for experiments 18 h after Dex administration. Treatment with Dex lowered the specific binding (SB) of insulin (INS) in the liver of 3- and 18-month-old rats and concentration of INS binding sites (N1, N2) and the dissociation constant of low-affinity binding sites (Kd2) in the liver of 6- and 18-month-old rats. In addition, Dex treatment lowered the liver IR protein level in all analyzed groups, except 21-month-old rats where it remained unchanged, but raised the IR mRNA level in 18-month-old rats. In erythrocytes, treatment with Dex decreased SB and Kd2 (in animals 3 and 6 months old) and N1 (in ones 3 and 18 months old). Following Dex treatment, the INS plasma level increased (in rats 3, 18 and 21 months old), while glucose (Glu) concentration increased in 3 and 12 months old, but decreased in 6- and 21-month-old rats. In summary, Dex exerts the strongest effect on the erythrocyte IR of 3- and 6-month-old rats and the hepatic IR of 18-month-old rats. IR in both tissues is almost insensitive to Dex in 12- and 21-month-old rats. The pattern of age-related changes of IR induced by Dex does not correlate with changes of plasma Glu and INS.  相似文献

45Ca2+ uptake and cytosolic Ca2+ concentrations [( Ca2+]i) were measured in synaptosomes prepared from the cerebral cortex of 3-, 16-, and 24-month-old male Charles River Wistar rats. Electron-microscopic examination demonstrated no morphological differences between the synaptosomes prepared from 3- and 24-month-old rats. The fast phase of Ca2+ uptake was reduced in the 24-month-old animals as compared to the 3-month-old ones (-23%, p less than 0.001), whereas no difference was found between the 16- and the 3-month-old rats. Age did not modify [Ca2+]i, as measured by the quin 2 technique, both at rest and immediately after depolarization with 50 mM K+. The Ca2+ load following depolarization was cleared in about 13 min in the 3-month-old rats. The rate of clearance was significantly slower both in the 16- (p less than 0.01) and in the 24-month-old rats (p less than 0.0001). The addition of verapamil (60 microM) after depolarization restored [Ca2+]i to resting level in aged rats at the same rate as in young rats. A prolonged Ca2+ influx, therefore, may be responsible for the slower clearance of Ca2+ load in aged rats.  相似文献

The ultrastructure of mitochondria of cross-striated muscles during aging was studied by electron microscopy. Mitochondrial ultrastructure was analyzed in the flight muscle of D. melanogaster (1- and 36-day-old) and in the cardiomyocytes and skeletal muscle of young and senile Wistar and OXYS rats (3- and 25-month-old). The mitochondria in the flight muscle samples of senile D. melanogaster flies were shown to have several types of peculiar age-related mitochondrial abnormalities corresponding to those described previously. Previously unknown changes were revealed in the ultrastructure of cardiomyocyte mitochondria in senile rats (both Wistar and OXYS). Substantial changes in the ultrastructure of subsarcolemmal mitochondria were found in the fibers of red skeletal muscle of senile OXYS rats. It has been shown that the subsarcolemmal mitochondria of red muscle fibers are a peculiar population of mitochondria with atypical ultrastructure. Initial changes in the ultrastructure of subsarcolemmal mitochondria were revealed even in 3-month-old OXYS rats. At the same time, the skeletal muscle mitochondria of senile Wistar rats maintain their morphological characteristics, and their ultrastructure corresponds to that of skeletal muscle mitochondria in 3-month-old Wistar rats.  相似文献

Effect of thyrotropic hormone (TTH) on membrane potential (MP) of thyroid cells and thyroid hormone secretion was studied in experiments on male rats of two age groups (7--12- and 27--32-month-old animals). It was found that during the first 3 hours after TTH administration (5 U/100 g i. v.) the depolarization of secretory cell membranes of adult rats was done pronounced and developed more rapidly than in old ones and that an increase in free thyroxin (T4) correlation with MP changes with time. In a dose of 0.5 U/100 g TTH caused a significant rise in T4 secretion only in old rats. The cAMP level in the thyroid gland declined with aging. In a dose of 5 U/100 g TTH provoked a significant increase in the cAMP content in adult rats and had no effect on its content in old ones. A relationship between the MP level of thyroid secretory cells and thyroid hormone secretion is discussed.  相似文献

Thyroxin was administered daily to male Wistar rats aged 1, 3, 12 and 24 months in a dose of 250 g per 100 g body weight. After 9 days there was a significant increase in the content of cytochromes c and a (a(3)). An appreciable rise in the cytochrome c content was recorded as long as after one day, whereas the content of cytochromes a (a(3)) did not exceed normal even after 2 days. Following one day the content of cytochromes a (a(3)) in 3-, 12-, and 24-month-old rats was sightly lower than in normal rats. A significant temporary increase in the c/a (a(3)) ratio was observed after 1-2 days only in 12- and 24-month-old rats. The c/a (a(3)) ratio increased with age. It is suggested that application of thyroid hormones may be promising in studies on the regulation of mitochondrial biogenesis.  相似文献
