Changes in the sugar and amino acid contents of potato tubers during short-term storage and the effect on the acrylamide level in chips after frying were investigated. The acrylamide content in chips began to increase after 3 days of storage at 2 degrees C in response to the increase of glucose and fructose contents in the tubers. There was strong correlation between the reducing sugar content and acrylamide level, R(2)=0.873 for fructose and R(2)=0.836 for glucose. The sucrose content had less correlation with the acrylamide content because of its decrease after 4 weeks of storage at 2 degrees C, while the reducing sugar in potato tubers and the acrylamide in chips continued to increase. The contents of the four amino acids, i.e., asparatic acid, asparagine, glutamic acid and glutamine, showed no significant correlation with the acrylamide level. These results suggest that the content of reducing sugars in potato tubers determined the degree of acrylamide formation in chips. The chip color, as evaluated by L* (lightness), was correlated well with the acrylamide content.  相似文献

A new defect of potato, Solanum tuberosum L., "zebra chip," so named for the characteristic symptoms that develop in fried chips from infected potato tubers, has recently been documented in several southwestern states of the United States, in Mexico, and in Central America. This defect is causing millions of dollars in losses to both potato producers and processors. Zebra chip plant symptoms resemble those caused by potato purple top and psyllid yellows diseases. Experiments were conducted to elucidate the association between the psyllid Bactericera cockerelli (Sulc) (Homoptera: Psyllidae) and zebra chip by exposing clean potato plants to this insect under greenhouse and field conditions. Potato plants and tubers exhibiting zebra chip symptoms were tested for phytoplasmas by polymerase chain reaction. Potato psyllids collected from infected potato fields also were tested. Results indicated that there was an association between the potato psyllid and zebra chip. Plants exposed to psyllids in the greenhouse and field developed zebra chip. In the greenhouse, 25.8 and 59.2% of tubers exhibited zebra chip symptoms in the raw tubers and fried chips, respectively. In the field, 15 and 57% of tubers showed symptoms in raw tubers and chips, respectively. No zebra chip was observed in tubers from plants that had not been exposed to psyllids, either in the greenhouse or field. No phytoplasmas were detected from potato plants or tubers with zebra chip symptoms, suggesting that these pathogens are not involved in zebra chip. Of the 47 samples of potato psyllids tested, only two tested positive for the Columbia Basin potato purple top phytoplasma.  相似文献

To clarify the effects of storage temperature on potato components and acrylamide in chips, tubers from five cultivars were stored at various temperatures (2, 6, 8, 10, and 18 degrees C) for 18 weeks, and the contents of sugars, free amino acids in tubers, and acrylamide in chips after frying were analyzed. At temperatures lower than 8 degrees C, the contents of reducing sugars increased markedly in all cultivars, with similar increases in the acrylamide level and dark brown chip color. Free amino acids showed little change at the storage temperatures tested and varied within certain ranges characteristic of each cultivar. The contents of reducing sugars correlated well with the acrylamide level when the fructose/asparagine molar ratio in the tubers was <2. When the fructose/asparagine ratio was >2 by low-temperature storage, the asparagine content, rather than the reducing sugar content, was found to be the limiting factor for acrylamide formation.  相似文献

SUMMARY: Acrylamide is produced in starchy foods that are baked, roasted or fried at high temperatures. Concerns about the potential health issues associated with the dietary intake of this reactive compound led us to reduce the accumulation of asparagine, one of its main precursors, in the tubers of potato (Solanum tuberosum). This metabolic change was accomplished by silencing two asparagine synthetase genes through 'all-native DNA' transformation. Glasshouse-grown tubers of the transformed intragenic plants contained up to 20-fold reduced levels of free asparagine. This metabolic change coincided with a small increase in the formation of glutamine and did not affect tuber shape or yield. Heat-processed products derived from the low-asparagine tubers were also indistinguishable from their untransformed counterparts in terms of sensory characteristics. However, both French fries and potato chips accumulated as little as 5% of the acrylamide present in wild-type controls. Given the important role of processed potato products in the modern Western diet, a replacement of current varieties with intragenic potatoes could reduce the average daily intake of acrylamide by almost one-third.  相似文献

Factors affecting the detection of potato leafroll virus (PLRV) by enzyme-linked immunosorbent assay (ELISA) in tubers of field-grown potato plants with primary or secondary infection were studied. The reactions of extracts of virus-free potato tubers were minimised by pre-incubating the extracts at room temperature and by careful choice of the dilution of enzyme-conjugated globulin. PLRV was reliably detected in tubers produced by secondarily infected plants of all six cultivars tested. PLRV concentration was greater in heel-end than in rose-end vascular tissue of recently harvested tubers but increased in rose-end tissue when tubers stored at 4°C for at least 5 months were placed at 15–24°C for 2 wk. PLRV occurred at greater concentration in tubers from plants of cv. Maris Piper with natural or experimentally induced primary infection than in tubers from secondarily infected plants; again PLRV concentration was greater in heel-end than in rose-end vascular tissue. Plants whose shoots were infected earliest in the growing season were invaded systemically and produced the greatest proportion of infected tubers; plants infected late in the season also produced infected tubers but PLRV was not detected in their shoot tops. PLRV concentration in tubers from the earliest-infected plants was less than in tubers from later-infected plants. PLRV was detected reliably by ELISA in tubers from progenies that were totally infected but was not detected in all infected tubers from partially infected progenies. ELISA is suitable as a routine method of indexing tubers for PLRV, although the virus will not be detected in all infected tubers produced by plants to which it is transmitted late in the growing season.  相似文献

The acceptability of potatoes for processing chips and French fries is largely dependent on the color of the finished product. Most potato cultivars and varieties stored at temperatures below 9–10 °C are subjected to low temperature sweetening (LTS) which result in the production of bitter-tasting, dark colored chips and French fries which are unacceptable to consumers. However, storing tubers at low temperatures (i.e., <10 °C) has many advantages such as lowered weight loss during storage, natural control of sprouting, and reduction/elimination of chemical sprout inhibitors. Our earlier research results on LTS suggested a role for pyruvate decarboxylase (PDC) in LTS-tolerance. In the present study, the role of PDC was examined whereby the potato variety Snowden was transformed with Arabidopsis cold-inducible pyruvate decarboxylase gene 1 (AtPDC1) under the control of promoter rd29A. Two transgenic plants were selected and storage studies were conducted on tubers harvested from one of the transgenic lines grown under green house conditions. Transgenic tubers showed higher Agtron chip color score indicating lighter chip and lower reducing sugar and sucrose concentrations compared to the untransformed tubers during the storage periods studied at 12 °C and 5 °C. These results suggest that overexpression of pyruvate decarboxylase gene resulted in low temperature sweetening tolerance in the transgenic Snowden.  相似文献

The improvement of processing quality of potato products (fries and chips) demands less accumulation of reducing sugars (glucose and fructose) in cold-stored potato (Solanum tuberosum) tubers. Control of gene expression to achieve this requires promoters with specificity to tubers as well as inducible activity under low temperatures. Here we use overlapping extension PCR to construct two chimeric promoters, pCL and pLC, to control gene expression in a tuber-specific and cold-inducible pattern. This combined different combinations of the LTRE (low-temperature responsive element) from Arabidopsis thaliana cor15a promoter and the TSSR (tuber-specific and sucrose-responsive sequence) from potato class I patatin promoter. The cold-inducible and tuber-specific activities of the chimeric promoters were investigated by quantitative analysis of GUS activity in transgenic potato cultivar E3 plants. The results showed that the cis-elements, LTRE and TSSR, played responsive roles individually or in combination. pCL with the TSSR closer to the TATA-box showed substantially higher promoter activity than pLC with the LTRE closer to the TATA-box at either normal (20°C) or low temperature (2°C), suggesting that the promoter activity was closely associated with the position of the two elements. The chimeric promoter pCL with tuber-specific and cold-inducible features may provide valuable tool for controlling the expression of gene constructs designed to lower the formation of reducing sugars in tubers stored at low temperature and to improve the processing quality of potato products. The incidence of potato virus Y (PVY) infection was assessed in samples of potato tubers, cv. Record, taken from Scottish seed stocks and English ware crops grown from some of these seed stocks. PVY was readily detected by ELISA of tuber sprouts. PVY-infected tubers were found in 10 seed stocks of 84 tested. The mean level of virus infection was 0.23%, 0.76% and 0.56% in Super Elite, Elite and AA stocks respectively. In 46 commercial ware crops grown from some of these seed stocks, a substantial proportion of the harvested tubers in all but one of the crops were infected with PVY, the mean percentage of infected tubers was 58.5%. Ware crops grown from seven seed stocks in which PVY had been detected (mean 6.2% infection in seed) contained a mean of 70% infected tubers, compared with 56% infection in crops grown from 39 stocks in which PVY was not detected in the seed tubers. The predominant PVY strain detected in the ware crops was the veinal necrosis strain (PVYvn).  相似文献

A larger proportion of tubers of Arran Pilot potato growing at the surface of soil infested with potato mop-top virus (PMTV) showed spraing symptoms (brown rings) at harvest than of tubers from below the surface. Infected tubers with or without spraing developed a spraing ring when stored in darkness, first for 1–2 wk at 18 d?C and then for 1–2 wk at any constant temperature between 5 and 13 d?C. Only a faint surface ring developed when either of these periods was decreased to 1 day; 4-day periods were needed to induce distinct symptoms. Internal tuber symptoms developed more slowly than surface symptoms, and their formation was favoured by cutting the tubers in half. Additional pigmented surface rings were produced outside the first ring by successive cycles of treatment at 18 and 9d?. Spraing did not develop when the first stage of treatment was at 22–25d?, when the tubers were kept first at 10d? and then at 5d?, when the treatment at 5–13d? preceded that at 18d?, or when the tubers were kept at constant temperatures ranging from 5 to 25d?. When tubers of six potato varieties were grown in PMTV-infested soil and then stored at temperatures designed to induce symptoms, the varieties known to be the most susceptible in the field were those which had the greatest tendency to develop spraing during storage. When infected tubers were exposed to light, typical spraing symptoms were not induced, but greening of the tuber surface was much delayed in localized ring-shaped areas, so that pale weals appeared. Spraing symptoms were produced, in favourable conditions, by the reaction of cells at the periphery of the PMTV-invaded zone. Internal spraing did not prevent PMTV invading tissue outside the brown arcs; its rate of spread was about 10 μm/h at 14–18d?.  相似文献

Summary Out of 720 field-grown potato first year seedlings plants with best tuber apearance and large and medium size tubers with best shape were respectively selected. The tuber progeny of each group was evaluated in field experiments. It was demonstrated that the selection of individual tubers was very effective in the elimination of clones with irregular tuber shape and deep eyes. The tuber progeny of selected tubers was not inferior to the tuber progeny of selected plants. Possible applications of the results to practical potato breeding are discussed.  相似文献

The possibility of practical application of biosensors based on pH-sensitive field-effect transistors and butyrylcholinesterase to glycoalkaloid analysis in potato tubers was studied. The main analytical features of the designed biosensors and measurement conditions were optimized. The biosensor was applied to quantitative analysis of glycoalkaloids in tubers of different potato varieties. The results proved to be in good agreement with those obtained by conventional protocols. Experiments on glucose assay were performed. An inverse correlation between the contents of glucose and glycoalkaloids in potato tubers was demonstrated.  相似文献

In the variety Arran Victory, which has purple tubers, there was found a tuber half purple and half pink. The pink half produced a plant which had pink tubers with small purple patches. X-ray irradiation of these tubers gave plants which had about five per cent of pink tubers without the purple patches. Breeding and eye-excision experments showed that the pink tubers with small purple patches were periclinal chimeras in which geneP had been lost from L1, L2 plus L3 still having geneP as in normal Arran Victory. The pink tubers without the purple patches had no geneP in L1, L2 and L3. The chimerical pink tubers had purple sprouts, the full pink tubers pink sprouts. The pigmented layer in the tubers of Arran Victory is the phelloderm (secondary cortex), not the primary outer cortex.  相似文献

Nature of enhanced respiration during sprouting of aged potato seed-tubers   总被引:2,自引:0,他引:2  
Respiration of 18-month-old Solarium tuberosum L. tubers was about 53% greater than that of 6-month-old tubers during sprouting at 23°C; yet, a significant loss of sprout vigor in the older tubers was apparent. Involvement of alternative oxidase (AO) in the age-induced difference in tuber respiration was assessed. AO was only detected in immunoblots if tissue disks from tubers were pre-incubated for 24 h prior to isolation of submitochondrial membrane particles (SMPs). No AO1 was detected in SMPs from nonincubated tuber tissue of either age, indicating that it was not contributing to tuber respiration during sprouting as previously thought. Respiratory control and ADP/O ratios indicated that oxidative phosphorylation was fully coupled to electron transport in mitochondria isolated from 6- and 18-month-old tubers. Cytochrome c oxidase (EC 1.9.3.1) activities of intact mitochondria were also not affected by tuber age. The difference in respiration during sprouting was unique to whole tubers, as oxygen consumption by mitochondria from young and oid tubers was equal on a milligram protein basis. Sprouting 18-month-old tubers had 15% more mitochondrial protein per gram fresh weight than did 6-month-old tubers. Older tubers also produced more ATP than younger tubers prior to and during sprouting, through a fully coupled, Cyt-mediated respiratory pathway, reduced sprout vigor notwithstanding. From 5 to 10 days of sprouting, coinciding with development of the age-induced difference in whole-tuber respiration, ATP concentration in 18-month-old tubers increased to become 52% higher than that in 6-month-old tubers. ATP synthase (EC 3.6.1.34), assessed by SDS-PAGE and immunoblots of β- and oligomycin-sensitivity conferring protein-subunits, also increased as a proportion of SMP protein in older tubers during this period. Relative to 6-month-old tubers, the increased respiration and associated oxidative phosphorylation of 18-rnonth-old tubers during sprouting were probably in response to a lower adenylate energy charge (AEC) prior to sprouting (from 0 fo 5 days). From 5 to 10 days of sprouting, AEC of 18-rnonth-old tubers increased to equal that of 6-month-old tubers and the two tuber ages maintained the same AEC for the remainder of the 20-day sprouting interval. Higher respiration and lower AEC of older tubers in storage at 4°C, along with the fact that older tubers respired at a higher rate to achieve the same AEC as younger tubers during sprouting, indicate greater utilization of ATP by older tubers.  相似文献

Stolons attached to developing potato tubers were inoculated with the soft rot bacterium Erwinia carotovora var. atroseptica. Almost all the stolons rotted, but soft rots developed in less than 10% of new tubers; the bacterium was isolated later from these tubers. No rots developed in the other tubers but the bacterium was later isolated from about half of them. It could not be isolated from tubers attached to inoculated stolons where the rot on them did not extend to the tuber or from tubers attached to stolons that were not inoculated though many of these rotted. The bacterium was reisolated from almost all arrested lesions in tubers inoculated 8 month earlier with E. carotovora var. atroseptica. Blackleg did not develop from plants grown fom these tubers under various soil conditions. It did develop in a large proportion of plants from tubers inoculated shortly before planting and grown in cool, wet soil. Less than 1% blackleg developed in plants grown from tubers from plants with blackleg or from plants immediately adjacent. The presence of pectolytic bacteria and E. caratovora var. atroseptica in seed and new tubers was investigated during June, July and August. Although E. caratovora var. atroseptica was obtained from c. 40% tubers, only c. 0·3% of c. 8400 plants developed blackleg. The bacterium was isolated from only three of 160 new tubers sampled during the summer.  相似文献

The aim of this work was to investigate the effect of decreased activity of lactate dehydrogenase (EC 1.1.1.27; LDH) on lactate metabolism in potato tubers. By expressing a cDNA‐encoding potato tuber LDH in the antisense orientation, we generated transgenic potato plants with a preferential decrease in two of the five isozymes of LDH. Surprisingly, transgenic tubers grown under normoxic conditions did not contain less lactate, but rather instead contained approximately two‐fold more lactate than control tubers. This result is explicable if the decreased isozymes of LDH are responsible for the oxidation of lactate to pyruvate in vivo. This was confirmed by measurements of the rate of metabolism of lactate supplied to tuber discs: the rate in transgenic tubers was approximately half that of control tubers. The decrease in LDH activity had no measurable effect on the accumulation of lactate in cold‐stored tubers under anoxia, nor during the subsequent utilization of this lactate upon return to normoxia. In both control and transgenic tubers, the accumulation of lactate during anoxia was not accompanied by an induction of LDH activity or a change in isozyme distribution. In contrast, the metabolism of lactate after a period of anoxia was accompanied by a two‐fold increase in LDH activity and the induction of two isozymes that were distinct from those which had been decreased in the transgenic plants.  相似文献
