Raman spectroscopy was used to determine the conformation of the disulfide linkage between cysteine residues in the homodimeric construct of the N-terminal alpha helical domain of surfactant protein B (dSP-B1-25). The conformation of the disulfide bond between cysteine residues in position 8 of the homodimer of dSP-B1-25 was compared with that of a truncated homodimer (dSP-B8-25) of the peptide having a disulfide linkage at the same position in the alpha helix. Temperature-dependent Raman spectra of the S-S stretching region centered at ∼ 500 cm− 1 indicated a stable, although highly strained disulfide conformation with a χ(CS-SC) dihedral angle of ± 10° for the dSP-B1-25 dimer. In contrast, the truncated dimer dSP-B8-25 exhibited a series of disulfide conformations with the χ(CS-SC) dihedral angle taking on values of either ± 30° or 85± 20°. For conformations with χ(CS-SC) close to the ± 90° value, the Raman spectra of the 8-25 truncated dimers exhibited χ(SS-CC) dihedral angles of 90/180° and 20-30°. In the presence of a lipid mixture, both constructs showed a ν(S-S) band at ∼ 488 cm− 1, corresponding to a χ(CS-SC) dihedral angle of ± 10°. Polarized infrared spectroscopy was also used to determine the orientation of the helix and β-sheet portion of both synthetic peptides. These calculations indicated that the helix was oriented primarily in the plane of the surface, at an angle of ∼ 60-70° to the surface normal, while the β structure had ∼ 40° tilt. This orientation direction did not change in the presence of a lipid mixture or with temperature. These observations suggest that: (i) the conformational flexibility of the disulfide linkage is dependent on the amino acid residues that flank the cysteine disulfide bond, and (ii) in both constructs, the presence of a lipid matrix locks the disulfide bond into a preferred conformation.  相似文献

Running is a popular form of recreation, but injuries are common and may be associated with abnormal joint motion. The objective of this study was to determine the effect of three footwear conditions – barefoot (BF), an ultraflexible training shoe (FREE), and a motion control shoe (MC) – on 3D foot and ankle motion. Dynamic, biplane radiographic images were acquired from 12 runners during overground running. 3D rotations of the tibiotalar and subtalar joints were quantified in terms of plantarflexion/dorsiflexion (PF/DF), inversion/eversion (IN/EV) and internal/external rotation (IR/ER). Across the early stance phase (defined as footstrike to heel-off), BF running demonstrated greater tibiotalar joint range of motion for PF/DF (28.2±8.3°) and IR/ER (7.0±1.4°) than the shod conditions (FREE: PF/DF=15.1±5.9°, IR/ER=4.8±2.1°; MC: PF/DF=15.0±6.2°, IR/ER=4.3±0.7°). Also at the tibiotalar joint, BF running resulted in a position significantly more plantarflexed (BF: 2.0±12.5°, FREE: 15.7±12.2°, MC: 16.5±9.3°) and internally rotated (BF: 12.9±4.5°, FREE: 10.7±4.3°, MC: 10.6±3.9°) at footstrike compared to both shod conditions. No differences were detected between the shod conditions at any point in the early stance phase at the tibiotalar joint. The MC condition demonstrated significant differences compared to FREE at several points throughout the early stance phase at the subtalar joint, with the greatest differences seen at 30% in PF/DF (MC −1.4±8.8°: FREE: −0.5±9.0°), IN/EV (MC −8.1±5.7°: FREE −6.3±5.5°) and IR/ER (MC −9.5±5.3°: FREE: −8.7±5.2°). These findings indicate that footwear has subtle effects on joint motion mainly between BF and shod conditions at the tibiotalar joint and between shod conditions at the subtalar joint.  相似文献

The peformance of the parasitoid Anagyrus kamali Moursi [Hymenoptera: Encyrtidae], as a function of host density, temperature, and photoperiod was investigated with the objective to optimize a mass-rearing system in the context of a biological control program. The number of hosts parasitized at densities varying from 2–100 hibiscus mealybug (HMB), Maconellicoccus hirsutus Green [Homoptera: Pseudococcidae], corresponded to a type II-III functional response in fixed-time conditions and a type III in variable-time conditions. Twenty-six percent of the oviposited eggs led to progeny emergence with a sex ratio of 0.49±0.102 (M/F), regardless of host density. Fecundity and oviposition period under six abiotic combinations (i.e., two temperatures (26±2 °C and 32±2 °C) and three photoperiods (L0:D24, L12:D12, L24:D0)) were measured. Lifetime fecundity and reproductive life were significantly affected by temperature and photoperiod conditions. Optimum female parasitoid lifetime fecundity was attained at 26±2 °C, L0:D24 with an average of 116.1±17.43 eggs. At 32±2 °C, L24:D0 and L12:D12, an average of 79.4±34.57 and 85.8±35.81 eggs were laid, respectively. Reproductive longevity was maximal at 26±2 °C, L0:D24 with 12±4.85 days of oviposition. Because the parasite A. kamali can be reared optimally without light, this may save tremendous energy costs.  相似文献

A software algorithm has been developed to investigate the folding process in B-DNA structures in vacuum under a simple and accurate force field. This algorithm models linear double stranded B-DNA sequences based on a local, sequential minimization procedure. The original B-DNA structures were modeled using initial nucleotide structures taken from the Brookhaven database. The models contain information at the atomic level allowing one to investigate as accurately as possible the structure and characteristics of the resulting DNA structures. A variety of DNA sequences and sizes were investigated containing coding and non-coding, random and real, homogeneous or heterogeneous sequences in the range of 2 to 40 base pairs. The force field contains terms such as angle bend, Lennard-Jones, electrostatic interactions and hydrogen bonding which are set up using the Dreiding II force field and defined to account for the helical parameters such as twist, tilt and rise. A close comparison was made between this local minimization algorithm and a global one (previously published) in order to find out advantages and disadvantages of the different methods. From the comparison, this algorithm gives better and faster results than the previous method, allowing one to minimize larger DNA segments. DNA segments with a length of 40 bases need approximately 4 h, while 2.5 weeks are needed with the previous method. After each minimization the angles between phosphate–oxygen-carbon A1, the oxygen–phosphate–oxygen A2 and the average helical twists were calculated. From the generated fragments it was found that the bond angles are A1=150°±2°and A2=130°±10°, while the helical twist is 36.6°±2° in the A strand and A1=150°±6° and A2=130±6° with helical twist 39.6°±2° in the B strand for the DNA segment with the same sequence as the Dickerson dodecamer.Figure The final minimized DNA segment of the Dickerson dodecamer sequence represented by ball drawings and viewed (left) perpendicular and (right) down the helical axis  相似文献

Homonuclear 3D NOESY-NOESY has shown great promise for the structural refinement of large biomolecules. A computationally efficient hybrid-hybrid relaxation matrix refinement methodology, using 3D NOESY-NOESY data, was used to refine the structure of a DNA three-way junction having two unpaired bases at the branch point of the junction. The NMR data and the relaxation matrix refinement confirm that the DNA three-way junction exists in a folded conformation with two of the helical stems stacked upon each other. The third unstacked stem extends away from the junction, forming an acute angle (60° ) with the stacked stems. The two unpaired bases are stacked upon each other and are exposed to the solvent. Helical parameters for the bases in all three strands show slight deviations from typical values expected for right-handed B-form DNA. Inter-nucleotide imino-imino NOEs between the bases at the branch point of the junction show that the junction region is well defined. The helical stems show mobility (± 20° ) indicating dynamic processes around the junction region. The unstacked helical stem adjacent to the unpaired bases shows greater mobility compared to the other two stems. The results from this study indicate that the 3D hybrid-hybrid matrix MORASS refinement methodology, by combining the spectral dispersion of 3D NOESY-NOESY and the computational efficiency of 2D refinement programs, provides an accurate and robust means for structure determination of large biomolecules. Our results also indicate that the 3D MORASS method gives higher quality structures compared to the 2D complete relaxation matrix refinement method.  相似文献

Bovine chromaffin secretory granules were purified by isopycnic Metrizamide gradient centrifugation and their Ca2+ sequestration pathways were characterized. The rate of Ca2+ sequestration at 37°C was first order, with a maximal uptake of 26.9 ±0.46 (mean ± S.D., n = 3) nmol Ca2+/mg protein and a first order rate constant (k) of 0.046 ± 0.002 min–1. At 4°C the rate of uptake was substantially attenuated, with only 2.47 ± 0.2 (mean ± S.D, n = 3) nmol Ca2+/mg protein sequestered in 60 min. Ca2+ sequestration was 93% inhibited by 180 mM NaCl [I50% of 78.7 ± 9.3 mM NaCl (mean ± S.D., n = 11)] but only slightly inhibited by KCl or MgCl2. Ca 2+ sequestration was not stimulated by incubation with MgATP but was inhibited by 57% after incubation with 30 M monensin. Ca 2+ sequestration was dependent on extravesicular Ca 2+ with half-maximal sequestration at pCa2+ 6.81 ± 0.028 (mean ± S.D., n = 3). Sequestered Ca2+ could be exchanged with external 45Ca2+, the exchange rate was first order (k of 0.042 ± 0.004: mean ± S.D., n = 3) and saturated at 27.7 ± 1.1 nmol Ca2+/mg (mean ± S.D., n = 3). The Ca2+/Ca2+ exchange system was totally inhibited by NaCl or KCl but only slightly by MgCl2. About 75% of sequestered 45Ca2+ could be released by incubation with NaCl, but only 8% was released by incubation with KCI. Half-maximal release of sequestered 45Ca2+ required 69.3 ± 12.2 mM NaCl (mean ± S.D., n = 3). The Na+-induced release of sequestered 45Ca2+ was rapid, t0.5 of 2.80 ± 0.63 min (mean ± S.D., n = 3) and inhibited at 4°C. The concurrent incubation of chromaffin granules with 45Ca2+ and either annexin proteins V or VI resulted in attenuated uptake of 45Ca2+. We report molecular dynamics simulations in the explicit membrane environment of a small membrane-embedded protein, sarcolipin, which regulates the sarcoplasmic reticulum Ca-ATPase activity in both cardiac and skeletal muscle. In its monomeric form, we found that sarcolipin adopts a helical conformation, with a computed average tilt angle of 28 ± 6° and azymuthal angles of 66 ± 22°, in reasonable accord with angles determined experimentally (23 ± 2° and 50 ± 4°, respectively) using solid-state NMR with separated-local-field experiments. The effects of time and spatial averaging on both 15N chemical shift anisotropy and 1H/15N dipolar couplings have been analyzed using short-time averages of fast amide out-of-plane motions and following principal component dynamic trajectories. We found that it is possible to reproduce the regular oscillatory patterns observed for the anisotropic NMR parameters (i.e., PISA wheels) employing average amide vectors. This work highlights the role of molecular dynamics simulations as a tool for the analysis and interpretation of solid-state NMR data.  相似文献

In natural ecosystems, differences often exist in the relative abundanceof stable S isotopes (°34S) that can provide clues as tothe source, nature, and cycling of S. Values of °34S inprecipitation, throughfall, soils, soil solution, and stream waters weremeasured at the Hubbard Brook Experimental Forest (HBEF), New Hampshire.Values of °34S in precipitation and throughfall weresimilar to each other but differed seasonally. Precipitation°34S values were higher in the dormant season[°34S = 5.9±0.6 (17)][Mean + SE(N)]than in the growing season [°34S = 5.0±0.6(40)] but throughfall growing-season values were higher[°34S = 5.6±0.6(68)] than for the dormantseason [°34S = 4.9±0.7 (9)]. Different treespecies did not affect throughfall °34S values. In soilsolution, °34S values were higher in the growing season(°34S = 8.9±2.8; 8.8±1.7;and 4.0±0.6 for Oa, Bh, and Bs horizons, respectively) thanin the dormant season (°34S = 5.6±1.5;3.7±2.4; and 3.4±1.2 for Oa, Bh, and Bshorizons, respectively). These seasonal differences in°34S were probably caused by biological isotopicfractionation. The °34S values in streams were generally2 lower and more variable than those in precipitation andthroughfall, suggesting fractionation and/or different isotopic sources inthe soil.  相似文献

Although muscle–tendon slack length is a crucial parameter used in muscle models, this is one of the most difficult measures to estimate in vivo. The aim of this study was to determine the onset of the rise in tension (i.e., slack length) during passive stretching in both Achilles tendon and gastrocnemius medialis. Muscle and tendon shear elastic modulus was measured by elastography (supersonic shear imaging) during passive plantarflexion (0° and 90° of knee angle, 0° representing knee fully extended, in a random order) in 9 participants. The within-session repeatability of the determined slack length was good at 90° of knee flexion (SEM=3.3° and 2.2° for Achilles tendon and gastrocnemius medialis, respectively) and very good at 0° of knee flexion (SEM=1.9° and 1.9° for Achilles tendon and gastrocnemius medialis, respectively). The slack length of gastrocnemius medialis was obtained at a significantly lower plantarflexed angle than for Achilles tendon at both 0° (P<0.0001; mean difference=19.4±3.8°) and 90° of knee flexion (P<0.0001; mean difference=25.5±7.6°). In conclusion, this study showed that the joint angle at which the tendon falls slack can be experimentally determined using supersonic shear imaging. The slack length of gastrocnemius medialis and Achilles tendon occurred at different joint angles. Although reporting this result is crucial to a better understanding of muscle–tendon interactions, further experimental investigations are required to explain this result.  相似文献

This study was performed to clarify how weather and current dynamics affect the resistance to temperature change in the oceanic sea skaters, Halobates. Heat coma temperature (HCT) was measured for the adults and 5th instar larvae of four Halobates species collected from a fixed sampling location (12°00′N, 135°00′E ) in western tropical Pacific Ocean and from 13 locations in the eastern area of the India Ocean ranging from 08°00′N-06°00′S and 86°00-76°00′E. Both the gap temperature for heat coma (GTHC, mean±SD: 7.83±1.86 °C, n=32) and the heat coma temperature (HCP, 35.03±1.80 °C, n=32) of individuals collected from the Pacific Ocean, during the first half (10 days) of the sampling period at the fixed sampling point, were significantly higher than those during the second half (GTHC: 5.10±2.05 °C, n=63; HCP: 34.03±2.02 °C, n=63). The reduction in heat tolerance shown in the second half of the 20 day period may have been caused by a decrease in air temperature due to rainfall that occurred around the sampling point accompanied with the arrival of Typhoon No. 6.In the study of individuals collected from the Indian Ocean, significantly higher average GTHCs of >8 °C were recorded for the adult H. micans collected at 02°00′S and 06°00′S (89°00′E) than those at 0°00-8°00′N in the eastern Indian Ocean. Dynamic mixture of water from northern and southern currents occurs at 02°00-6°00′S of the Indian Ocean and might relate to such high heat tolerance.Temperature dynamics in the ocean habitat might directly affect the temperature resistance of the oceanic sea skaters.  相似文献

The occurrence of summer mortalities of the commercially important Pacific oyster, Crassostrea gigas, has increased in recent years. These mortality events occur during the late summer when water temperatures are at their highest. Many theories have been proposed concerning the causes including reproductive stress, environmental stress, disease, or synergistic interactions of these factors. C. gigas are grown intertidally and are exposed to the air (emersed) for hours at a time. These organisms can experience extreme changes in temperature during the course of a day. An oyster closed during emersion depletes the oxygen stores to near zero within the shell and builds up CO2 causing a decrease in tissue pH. The focus of this study is to determine the respiratory (pH, Po2, Pco2 and total CO2) and immune responses of oysters exposed to air at normal seasonal temperatures, and to determine whether these stresses associated with emersion inhibit the immune system of the oyster and contribute to the summer mortalities. The respiratory variables of the hemolymph of oysters submerged at 18 °C (pH = 7.52 ± 0.04 S.E.M., Po2 = 7.09 ± 0.53 S.E.M. kPa and Pco2 = 0.20 ± 0.03 S.E.M. kPa) varied significantly from oysters emersed for four hours at 22°C (pH = 7.11 ± 0.03 S.E.M., Po2 = 3.83 ± 0.15 S.E.M. kPa, Pco2 = 0.36 ± 0.03 S.E.M. kPa) and those emersed for four hours at 30 °C (pH = 6.84 ± 0.02 S.E.M., Po2 = 3.10 ± 0.12 S.E.M. kPa, Pco2 = 1.31 ± 0.06 S.E.M. kPa). The ability of hemocytes to kill the bacterium Vibrio campbellii was assessed using an in vitro assay to generate a killing index. There was no significant difference in the killing index between pH treatment groups (p = 0.856): at pH 7.6 killing index = 50.2% ± 2.33 S.E.M., at pH 6.6 killing index = 52.3% ± 3.67 S.E.M.. Temperature was the only factor to significantly affect the killing indices among temperature and oxygen treatment groups. The killing index was lowest (29.3% ± 3.25 S.E.M.) at 30 °C and 7% oxygen, simulating in vivo oxygen pressure in well-aerated conditions and 30 °C and 3% oxygen, simulating in vivo oxygen pressure in hypoxia (30.5% ± 3.25 S.E.M.), compared with the index in 7% oxygen at low temperature (18 °C) (44.4% ± 4.50 S.E.M.) or compared with low oxygen (3%) at low temperature (18 °C) (39.7% ± 2.51 S.E.M.). The seasonal and diurnal rise in temperature may, therefore, be an important factor contributing to summer mortalities of C. gigas.  相似文献

Both thelytokous and arrhenotokous Trichogramma minutum were collected from eggs of the spruce budmoth, Zeiraphera canadensis in New Brunswick, Canada and their phenotypic traits compared for use in biological control. The lower threshold temperature for development of thelytokous and arrhenotokous parasitoids was 9.7 and 10.1 °C, respectively; the former required significantly higher degree-days for development from egg to adult (–x±SE=165.1±5.8 °D) than the latter (128.1±4.9 °D). Thelytokous and arrhenotokous parasitoids had similar forewing length (0.49±0.01 vs. 0.49±0.01 mm) and adult lifespan (13.3±0.7 vs. 14.0±1.1 days), but significantly different fecundity and sex ratios. Thelytokous females produced fewer offspring (89.5±6.6 vs. 173.9±6.4) and fewer female progeny (77.2±5.4 vs. 109.8±3.3) despite an overall higher proportion of females (91.6±1.1% vs. 65.4±2.8%) than their arrhenotokous counterparts. Ovarian dissections showed that the number of eggs increased with parasitoid age in arrhenotokous parasitoids but remained steady in thelytokous parasitoids. The variation in ovarian development of the two forms was the major factor contributing to the differences in fecundity. Thelytokous parasitoids were more host-specific than arrhenotokous ones; when offered eight host species, the former rejected three whereas the latter rejected only one. Thelytokous parasitoids survived better than arrhenotokous ones when stored from 30 to 150 days at 4 °C. Thelytokous females were slower at initiating flight after emergence than arrhenotokous females but maintained flight activity longer (6 h). These results indicate that thelytokous T. minutum are different from their arrhenotokous counterparts physiologically, biologically and ecologically and that they may play different roles in the field. The potential for using thelytokous parasitoids in biological control programs is discussed.  相似文献

Nematode strains of the entomopathogenic family Steinernematidae differ in their ability to infect insects at different temperatures. Survival and infectivity of infective juveniles (IJs) of Steinernema rarum (OLI) were studied after their storage at 23 ± 2 °C and at 5 ± 1 °C. Survival at 23 ± 2 °C was always above 95%. At 5 ± 1 °C, survival decreased at week 5, but infectivity did the same after week 2. Unlike other steinernematids, both infectivity and survival of IJs would be higher for S. rarum (OLI) when stored at 23 ± 2 °C.  相似文献

Diaeretiella rapae (M'Intosh), a polyphagous and cosmopolitan parasite of aphids, was imported from China to the USA for biological control of Diuraphis noxia (Mordwilko), a pest of small grains. We studied several aspects of its biology on D. noxia hosts in the laboratory at 3 constant temperatures, 10.0, 21.1, and 26.7 °C. Females lived significantly longer than males at all 3 temperatures, and longevity was greatest at 10 °C, followed by 21.1 and 26.7 °C. Fecundity (= number of mummies) did not differ significantly among the 3 temperatures studied. The ovarian egg-load was 129.1 ± 9.3, and was significantly affected by the size of adult females. A pre-oviposition period ± 0.26 days) was detected at 10 °C, but not at the other temperatures. Pre-imaginal survivorship was similar among temperatures, while R o and T c decreased with temperature, and r m increased with temperature. The offspring sex ratio (proportion females) was lowest at 26.7 °C, and similar between 10.0 and 21.1 °C. In addition, the offspring sex ratio significantly declined with the age of the female parent. The suitability of D. rapae for colonization against D. noxia in North America is discussed in relation to its responses to temperature and the climate of its home range.  相似文献

The development and application of an algorithm to assess the ability of an infrared thermography (IRT) device to predict cows in estrus and about to ovulate was investigated. Twenty cows were synchronized using a controlled internal drug release and PGF2α. Vulval and muzzle temperatures were measured every 12 hours from controlled internal drug release insertion to 32 hours after PGF2α treatment and then every 4 hours until ovulation occurred or until 128 hours after PGF2α treatment (whichever occurred first). Thermal images obtained with a FLIR T620 series infrared camera were analyzed using ThermaCAM Researcher Professional 2.9 software. Cows were also monitored for behavioral signs of estrus and color changes of an Estrotect applied to the tail head of each cow 36 hours after PGF2α treatment. Algorithms were developed by adjusting body surface temperature of individual animals for ambient temperature and humidity during each observation period, and were expressed as a deviation from the baseline temperature. Of the 20 cows enrolled in this study, 12 (60%) ovulated. An IRT estrus alert was defined using different thresholds (D = 1 °C, 1.25 °C, and 1.5 °C). Sensitivity and specificity to predict estrus depended upon the chosen threshold level. At a threshold D = 1 °C, the highest sensitivity (92%; n = 11) and the lowest specificity (29%) and positive predictive value (64%) were observed. Conversely, D = 1.5 °C resulted in sensitivity of 75%, specificity of 57%, and positive predictive value of 69%. The mean ± standard deviation intervals between onset and the end of IRT estrus alert to ovulation were 30.7 ± 8.2 and 13.3 ± 7.7 hours, respectively. Ovulation occurred 24 to 47 hours after the onset of the IRT estrus alert for eight out of the 11 ovulated cows (73%). Although the sensitivity of the IRT alert was greater than visual observation (67%) and Estrotect activation (67%), the specificity and positive predictive value were lower than these two aids (i.e., the IRT overpredicted the incidence of ovulation). Results presented indicate that IRT shows some potential as an estrus detection aid; however, further studies investigating the potential to improve the specificity and capturing data throughout entire 21-day reproductive cycles would be worthwhile.  相似文献

Mean (± S.E.) developmental times of eggs, larvae and pupae were 4.0 ± 0.06, 14.9 ± 0.19 and 14.6 ± 0.11 days, respectively, the mean (± S.E.) survival of the original egg cohort to the start of larval, pupal and adult stage being 77.2 ± 3.2%, 54.5 ± 2.6% and 47.9 ± 3.5%, respectively. Females and males displayed approximately straight survivorship curves during adult life, implying constant mortality rates. Mean (± S.E.) adult longevity was 41.6 ± 2.98 days for females and 51.2 ± 3.91 days for males. Assuming a stable age distribution the population consisted of 56% eggs, 31% larvae, 6% pupae and 7% adults. Oviposition peaked when females were 25 days old, and the highest reproductive values (RVx) (mean ± S.E.) ranged from 129.1 ± 7.57 to 138.5 ± 6.83 for individuals 17–27 days old. A female deposited a mean (± S.E.) of 614.7 ± 35.9 female eggs over a maximum life span of 93 days. The basic reproductive rate (R 0) (mean ± S.E.) was 173.0 ± 14.2 female offspring per female and the intrinsic rate of natural increase of female individuals (r) (mean ± S.E.) was 0.088 ± 0.001 day-1. The mean (± S.E.) generation time (T) was 57.8 ± 0.78 days. In cultures with equal numbers of first instar S. fusca larvae and predacious third instar larvae of Hydrotaea aenescens (synonymous Ophyra aenescens) Wiedemann, mean (± S.E.) survival to the adult stage of S. fusca (16.7 ± 8.8%) was significantly lower than in controls with S. fusca alone (58.3 ± 7.4%). The potential significance of predation by H. aenescens on S. fusca in pig farms is discussed.  相似文献

The experiment was carried out with the aim of investigating the effect of ascorbic acid (AA) on fluctuations in rectal temperature (RT) of pigs during the harmattan season. Sixteen pigs administered with AA at the dose of 250 mg/kg orally and individually served as experimental animals, and 13 others administered orally with sterile water were used as control animals. The RT was measured from all the pigs at 06:00, 13:00 and 18:00 h twice in a week for three consecutive weeks. The lowest overall mean RT value of 37.52±0.90 °C was obtained at 06:00 h in the experimental pigs, while the corresponding value in control pigs was 37.63±0.90 °C (P>0.05). The maximum RT values of 38.75±0.18 °C and 39.27±0.11 °C were recorded at 13:00 h in experimental and control pigs, respectively (P<0.05). The results indicate that AA modulates the body temperature by decreasing the maximum RT value in pigs exposed to harmattan stress, and may alleviate the risk of adverse effects of the stress on health and productivity of pigs during the cold-dry season.  相似文献

Summary Angular sensitivity functions for light and dark-adapted locust retinula cells have been measured. The angular sensitivity for light-adapted cells follows a Gaussian function but in the dark-adapted condition the measured function is broader than Gaussian at its base. Acceptance angles for light-adapted cells are 1.5°±0.2° (horizontal), 1.4°±0.1° (vertical) and for dark-adapted cells, 2.4°±0.5° (horizontal) and 2.5°±0.4° (vertical). These values are significantly smaller than those previously reported and show the locust eye to be closer to its theoretical limit of resolution than was previously indicated.The angular sensitivity of light-adapted cells agress well with the minimum resolvable stripe period reported by those authors refuting anomalous resolution.  相似文献

Three experiments were conducted to optimize the protocol for cryopreservation of emu sperm. Ejaculates were collected from trained male emus then diluted 1:1 and pooled before allocation to treatments and measured for sperm viability, motility, egg membrane penetration ability, membrane stability, and morphology. In Experiment 1, semen was either cooled to 5 °C after dilution or diluted with a precooled to 5 °C diluent before cooling to 5 °C and then frozen at liquid nitrogen vapor temperatures of −140 °C and −35 °C, with 6% or 9% dimethylacetmide (DMA; a permeating cryoprotectant) and compared for sperm functions. The percentages of viable (42.8 ± 1.1%), normal (39.0 ± 1.3%), and motile (29.8 ± 1.3%) sperm were higher (P < 0.001) for semen frozen at −14 °C with 9% DMA (path 2) than for all other combinations. In Experiment 2, we assessed the value of combining DMA and trehalose in the diluent. Combining trehalose (3% to 9%) with DMA (3% to 9%) prior to freezing reduced (P < 0.001) the percentages of postthaw viable (by 4 to 9 ± 1.2%), normal (by 5 to 11 ± 1.3%), and motile sperm (by 13 to 17 ± 2.5%) and the number of holes on the perivitelline layer (by 27 to 29 holes/mm2). Postthaw function was best preserved with 9% DMA alone. In experiment 3, we investigated the possibility of increasing DMA concentrations from 6% to 24%. Postthaw sperm viability (52 to 55 ± 2.3%) and morphology (48 to 51 ± 1.7%) were higher (P < 0.05) with 18% and 24% than with 6% to 12% DMA and did not differ between 18% and 24% DMA. However, sperm motility (36 to 43 ± 2.9%) and the number of perivitelline holes were similar (P > 0.05) for 9% to 18% DMA (36 to 55 ± 12%). We concluded that adding 6% to 9% trehalose to the diluent offered no advantage, and that the current best practice for preserving postthaw function in emu sperm is to dilute semen with a precooled to 5 °C diluent and use 18% DMA.  相似文献
