PAF-acether is a phospholipid synthesized by most animal tissues and exerting a strong decrease on the heart’s contractile force and coronary flow. PAF-acether (10−9 and 10−10 M) was administered to isolated guinea pig hearts perfused via the Langendorff apparatus with Chenoweth solution. Zinc (1.5 μM) is known to benefit heart function thus, Zn2+ (1.5, 7.5, and 30 μM) was added in the perfusing solution before or after PAF-acether administration. Contractile force, coronary flow, and heart rate were recorded by means of a Narco MK-IV Physiograph throughout all modes of perfusion. Calcium inhibitor (Verapamil 10−10 M) and Pb2+ Co2+ (1.5×10−6 M) were used subsequently in the perfusing solutions in order to elucidate some of the Zn and PAF interactions observed. All hearts were analyzed for their Zn and Ca content by means of an Atomic Absorption Spectrophotometry (AAS). Our data suggest that low concentrations of zinc (1.5 μM) can strongly inhibit PAF-induced decrease of contractile force and coronary flow. Zinc-inhibiting effects on PAF's negative inotropic action (myocytic level) is not exerted through Zn−Ca antagonism. Nevertheless, a Zn−Ca antagonism in the arteriolar level cannot be excluded. Zinc inhibits PAF selectively only if it is administered before PAF injection and this strongly suggests a receptor interaction between the metal and the phospholipid at the heart level.  相似文献

In aqueous alkaline Zn batteries (AZBs), the Co3+/Co4+ redox pair offers a higher voltage plateau than its Co2+/Co3+ counterpart. However, related studies are scarce, due to two challenges: the Co3+/Co4+ redox pair is more difficult to activate than Co2+/Co3+; once activated, the Co3+/Co4+ redox pair is unstable, owing to the rapid reduction of surplus Co3+ to Co2+. Herein, CoSe2?x is employed as a cathode material in AZBs. Electrochemical analysis recognizes the principal contributions of the Co3+/Co4+ redox pair to the capacity and voltage plateau. Mechanistic studies reveal that CoSe2?x initially undergoes a phase transformation to derived CoxOySez, which has not been observed in other Zn//cobalt oxide batteries. The Se doping effect is conducive to sustaining abundant and stable Co3+ species in CoxOySez. As a result, the battery achieves a 10 000‐cycle ultralong lifespan with 0.02% cycle?1 capacity decay, a 1.9‐V voltage plateau, and an immense areal specific capacity compared to its low‐valence oxide counterparts. When used in a quasi‐solid‐state electrolyte, as‐assembled AZB delivers 4200 cycles and excellent tailorability, a promising result for wearable applications. The presented effective strategy for obtaining long‐cyclability cathodes via a phase transformation‐induced heteroatom doping effect may promote high‐valence metal species mediation toward highly stable electrodes.  相似文献

Scale and sludge deposits formed during oil production can contain elevated levels of Ra, often coprecipitated with barium sulfate (barite). The potential for sulfate-reducing bacteria to release 226Ra and Ba (a Ra analog) from oil-field barite was evaluated. The concentration of dissolved Ba increased when samples containing pipe scale, tank sludge, or oil-field brine pond sediment were incubated with sulfate-reducing bacteria Desulfovibrio sp., Str LZK1, isolated from an oil-field brine pond. However, Ba release was not stoichiometric with sulfide production in oil-field samples, and < 0.1% of the Ba was released. Potential for the release of 226Ra was demonstrated, and the 226Ra release associated with sulfate-reducing activity was predictable from the amount of Ba released. As with Ba, only a fraction of the 226Ra expected from the amount of sulfide produced was released, and most of the Ra remained associated with the solid material.  相似文献

A bioautographic assay based on thin layer chromatography was developed for phosphoenolpyruvate (PEP) detecting as a known but rarely studied inhibitor of phosphoglucose isomerase (PGI). The protocol with NADP+/NBT/PMS (β-nicotinamide adenine dinucleotide phosphate/nitrotetrazolium blue chloride/phenazine methosulfate) staining was capable of detecting Mycobacterium tuberculosis H37Ra PGI inhibition using PEP. According to this method, visibly brighter spots (zones) against purple background are observed in the area of inhibition of the above-mentioned enzyme activity. The detection limit for PEP as an inhibitor of Mycobacterium tuberculosis H37Ra PGI was 226?μg per spot/zone. Noteworthy is that we are the first authors to have successfully used a bioautographic assay to detect Mycobacterium tuberculosis H37Ra PGI inhibition by PEP.  相似文献

Snapping turtles Elseya dentata (Gray) from Magela Creek, Northern Territory, were exposed under laboratory conditions for up to 30 days to waters resembling the inorganic composition of Magela Creek water during the Wet season, with background and elevated Ca and Mg concentrations, that were labelled with 226Ra and 45Ca. The resulting concentrations of 45Ca in muscle, skin, gut, liver, shell bone and leg bone of E. dentata equilibrated or approached equilibrium by 12–18 days. Among the experiments, the concentrations of 45Ca in all six tissues were inversely related to turtle mass. An increase in the Ca water concentration by a factor of 15 increased the 45Ca concentration in all six tissues. The arithmetic factors of increase in the concentration in each tissue were proportional or more than proportional to the factor of increase in Ca water concentration; this factor was highest for muscle tissue (26.6). An increase in the Mg water concentration by a factor of 48 reduced the 45Ca concentration in all tissues except skin where it increased. The concentration of 226Ra in each tissue (except the gut) was positively related to its 45Ca concentration and inversely related to turtle mass in muscle, skin and liver. With the exception of the skin, the increased Ca water concentration did not reduce the 226Ra in the tissues but increased the 226Ra concentration in bone and muscle. The increased Mg water concentration had an inverse effect on the 226Ra concentrations in all tissues except shell. With the exception of the skin the effects of increased Ca and Mg water concentrations and turtle size on 226Ra concentrations in the tissues of E. dentata were similar to their effects on 45Ca tissue concentrations, indicating the similar metabolic behaviour of 226Ra and 45Ca in E. dentata.Exposures of the species Elseya latisternum (Gray), Emydura signata (Ahl) and Chelodina longicollis (Shaw), which are the same or closely related to species reported to occur in Magela Creek, to 45Ca-labelled Sydney tap water for 7 days demonstrated their ability to also accumulate 45Ca from their aquatic medium. The patterns of 45Ca concentrations in the tissues of these species indicated that they were inversely related to turtle mass, as demonstrated in E. dentata. The concentrations of 45Ca accumulated in the tissues were also comparable to those found in single specimens of E. dentata and E. victoriae (Gray) that were exposed for 7 days to simulated Magela Creek water. The data also indicated the larger long-necked C. longicollis accumulated less 45Ca per gram of tissue than similar-sized, short-necked species E. signata and E. latisternum, suggesting that long-necked turtles from Magela Creek would accumulate less 226Ra from their aquatic medium than similar-sized short-necked species.The capacity of E. dentata to accumulate 226Ra from the aquatic medium is about two orders of magnitude less than that of the tissue of the freshwater mussel Velesunio angasi (Sowerby) exposed under similar experimental conditions.  相似文献

The second chromosome Co-122 (Corato-122) extracted from a natural population of Drosophila melanogaster caught in Corato (Apulia) and maintained in the laboratory over the SM5 balancer chromosome, proved to carry: (1) a Segregation distorter factor, named Sd Co; (2) a recessive lethal mutation, termed mle-Co (maleless-Corato), which causes the lethality of only males; (3) another recessive lethal mutation, l(2)Co (lethal (2) Corato), probably arisen in the laboratory by mutation. This mutation accounts for the diminished recovery of homozygous females observed in the stock.The genetic features and the cytological analysis of the SD chromosome are reported, as well as the genetic localization of mle-Co and 1(2)Co and their cytogenetic mapping. An allelism test has ascertained that mle-Co is allelic to mle, a male-specific mutation described by Fukunaga et al., 1975. The tight linkage of mle-Co and 1(2)Co with Sd is discussed from the standpoint of population genetics.  相似文献

Pulsatile spiral blood flow in a modelled three-dimensional arterial stenosis, with a 75% cross-sectional area reduction, is investigated by using numerical fluid dynamics. Two-equation k-ω model is used for the simulation of the transitional flow with Reynolds numbers 500 and 1000. It is found that the spiral component increases the static pressure in the vessel during the deceleration phase of the flow pulse. In addition, the spiral component reduces the turbulence intensity and wall shear stress found in the post-stenosis region of the vessel in the early stages of the flow pulse. Hence, the findings agree with the results of Stonebridge et al. (2004). In addition, the results of the effects of a spiral component on time-varying flow are presented and discussed along with the relevant pathological issues.  相似文献

Flow regimes are believed to be of major evolutionary significance in fish. The flow regimes inhabited by cyprinids vary extensively from still flow regimes to riptide flow regimes. To test (i) whether flow‐driven swimming performance and relevant morphological differentiation are present among fish species and (ii) whether evolutionary shifts between high‐flow and low‐flow habitats in cyprinids are associated with evolutionary trade‐offs in locomotor performance, we obtained data on both steady and unsteady swimming performance and external body shape for 19 species of cyprinids that typically occur in different flow regimes (still, intermediate and riptide). We also measured the routine energy expenditure (RMR) and maximum metabolic rate (MMR) and calculated the optimal swimming speed. Our results showed that fish species from riptide groups tend to have a higher critical swimming speed (Ucrit), maximum linear velocity (Vmax) and fineness ratio (FR) than fish from the other two groups. However, there was no correlation between the reconstructed changes in the steady and unsteady swimming performance of the 19 species. According to the phylogenetically independent contrast (PIC) method, the Ucrit was actively correlated with the MMR. These results indicated that selection will favour both higher steady and unsteady swimming performance and a more streamlined body shape in environments with high water velocities. The results suggested that steady swimming performance was more sensitive to the flow regime and that for this reason, changes in body shape resulted more from selective pressure on steady swimming performance than on unsteady swimming performance. No evolutionary trade‐off was observed between steady and unsteady swimming performance, although Ucrit and MMR were found to have coevolved. However, a further analysis within each typically occurring habitat group suggested that the trade‐off that may exist between steady and unsteady swimming performance may be concealed by the effect of habitat.  相似文献

nhlF and hoxN, the genes encoding a cobalt transporter of Rhodococcus rhodochrous J1 and a nickel permease of Alcaligenes eutrophus H16, respectively, were expressed in Escherichia coli. 57Co2+ and 63Ni2+ transport of the recombinants was examined by means of a previously described physiological assay. Although the transporters are highly similar, different preferences for divalent transition metal cations were observed. HoxN was unable to transport 57Co2+, but mediated 63Ni2+ uptake. The latter activity was unaffected by a tenfold excess of other divalent cations, showing the specificity of HoxN for Ni2+. In contrast, NhlF transported both 57Co2+ and 63Ni2+ ion. NhlF-mediated 63Ni2+ uptake was markedly reduced in the presence of Co2+, while 57Co2+ uptake was only slightly lower in the presence of Ni2+. These results indicate different affinities of NhlF for Co2+ and Ni2+ and identified Co2+ ion as the preferred substrate. Received: 8 September 1998 / Accepted: 30 November 1998  相似文献

Summary Mycelial biomass of wild type and a Co2+-resistant N.crassa (cor) was used to remove Co2+ from aqueous media. Mycelia obtained from growth in nitrate N-medium (NaNO3) was more effective than ammonium N-medium (NH4NO3), in removing Co2+. Co2+-resistant N.crassa cor was more efficient than wild type in removing Co2+ from medium containing higher concentrations (500 mg/L). Metal removal was linear up to 12 h at which 35–60% Co2+ is depleted from medium, reaching near saturation by 24 h (90% removal). Co2+ removal was as efficient even from pure solutions and sodium azide inhibited the process up to 60%. Cell walls prepared from nitrate N-medium grown mycelia bound 3–5 fold more Co2+ when compared to ammonium N-medium. The importance of bioaccumulation and biosorption in bioremediating toxic metal ions from effluents is discussed.  相似文献

Abstract

Monitoring of biofilms subjected to different operating conditions was performed using a flow cell system. The system was fed by chlorine-free tap water, with and without added nutrients (0.5 mg l?1 carbon, 0.1 mg l?1 nitrogen and 0.01 mg l?1 phosphorus), and biofilms were grown on polyvinyl chloride (PVC) and stainless steel (SS) coupons, both in laminar and turbulent flow. The parameters analysed were culturable cells, using R2A, and total bacteria, which was assessed using the 4,6-diamino-2-phenylindole (DAPI) staining method. The impact of the different operating conditions in the studied parameters was established using Multivariate Analysis of Variance (MANOVA). From the most relevant to the least relevant factor, the total and culturable bacteria in biofilms increased due to the addition of nutrients to water (F = 20.005; p < 0.001); the use of turbulent (Re = 11000) instead of laminar (Re = 2000) hydrodynamic flows (F = 9.173; p < 0.001); and the use of PVC instead of SS as the support material (F = 2.848; p = 0.060). Interactions between these conditions, namely between surface and flow (F = 8.235; p < 0.001) and also flow and nutrients (F = 5.498; p < 0.05) have also proved to significantly influence biofilm formation. This work highlights the need for a deeper understanding of how the large spectrum of conditions interact and affect biofilm formation potential and accumulation with the final purpose of predicting the total and culturable bacteria attached to real drinking water distribution pipes based on the system characteristics.  相似文献

The genome of Mycobacterium tuberculosis encodes two paralogous P1B4‐ATPases, CtpD (Rv1469) and CtpJ (Rv3743). Both proteins showed ATPase activation by Co2+ and Ni2+, and both appear to be required for metal efflux from the cell. However, using a combination of biochemical and genetic studies we found that these proteins play non‐redundant roles in virulence and metal efflux. CtpJ expression is induced by Co2+ and this protein possesses a relatively high turnover rate. A ctpJ deletion mutant accumulated Co2+, indicating that this ATPase controls cytoplasmic metal levels. In contrast, CtpD expression is induced by redox stressors and this protein displays a relatively low turnover rate. A ctpD mutant failed to accumulate metal, suggesting an alternative cellular function. ctpD is cotranscribed with two thioredoxin genes trxA (Rv1470), trxB (Rv1471), and an enoyl‐coA hydratase (Rv1472), indicating a possible role for CtpD in the metallation of these redox‐active proteins. Supporting this, in vitro metal binding assays showed that TrxA binds Co2+ and Ni2+. Mutation of ctpD, but not ctpJ, reduced bacterial fitness in the mouse lung, suggesting that redox maintenance, but not Co2+ accumulation, is important for growth in vivo.  相似文献

The influence of medication on salivary flow of the elderly: preliminary study Objective: This study was to evaluate the influence of medications on unstimulated and stimulated salivary flow in elderly men and women. Background: Several diseases and conditions are associated with decreasing salivary flow. Medications can be risk factors for hyposalivation due to the effect particular drug categories. Methods: Seventy‐five elderly of both gender (sixty years old or over) from the Geriatric Dental Clinic at the Federal Fluminense University Dental School, were interviewed about their health status and chronic use of medication. After the interview, unstimulated and stimulated saliva were collected from each subject. The collection time was five minutes, and the flow rate was calculated as ml/min. Results: The mean unstimulated salivary flow was 0.25 ml/min for women and 0.30 ml/min for men, while the mean for stimulated salivary flow was 1.23 ml/min for women and 1.31 ml/min for men, without both differences being non‐significant (p > 0.05). The difference between the mean production of unstimulated and stimulated salivary flow was statistically significant, regardless of gender (p < 0.01). A significant reduction of stimulated salivary flow was observed with the usage of cardiovascular agents (p < 0.05). Conclusion: Drugs used in cardiovascular disease influence the flow of stimulated saliva.  相似文献

Conditions for the preparation of an aminoacylase column using DEAE-Sephadex as a carrier were investigated. The aminoacylase column having the highest activity was obtained when 7500 μmoles/hr. of partially purified aminoacylase was charged into a column packed with 9 ml. of DEAE-Sephadex A-25 (bead type, hydroxy form). By employing a DEAE-Sephadex–aminoacylase column, conditions for continuous optical resolution of acyl-DL -amino acids were investigated. When a solution of 0.2M acetyl-DL -methionine (pH 7.0, containing 5 × 10?4M Co2+) or 0.2M acetyl-DL -phenylalanine (pH 6.0, containing 5 × 10?4M Co2+) was passed through the aminoacylase column at the flow rate of SV = 2.5 or 2.0, respectively, at 50°C., the highest rate of hydrolysis of both substrates was attained. From the column effluents, enzymatically hydrolyzed L -methionine and L -phenylalanine were isolated in a good yield.  相似文献
