Systematics, conservation and morphology of the spider genus Tayshaneta (Araneae, Leptonetidae) in Central Texas Caves

The spider genus Tayshaneta is revised based on results from a three gene phylogenetic analysis (Ledford et al. 2011) and a comprehensive morphological survey using scanning electron (SEM) and compound light microscopy. The morphology and relationships within Tayshaneta are discussed and five species-groups are supported by phylogenetic analyses: the anopica group, the coeca group, the myopica group, the microps group and the sandersi group. Short branch lengths within Tayshaneta contrast sharply with the remaining North American genera and are viewed as evidence for a relatively recent radiation of species. Variation in troglomorphic morphology is discussed and compared to patterns found in other Texas cave invertebrates. Several species previously known as single cave endemics have wider ranges than expected, suggesting that some caves are not isolated habitats but instead form part of interconnected karst networks. Distribution maps are compared with karst faunal regions (KFR's) in Central Texas and the implications for the conservation and recovery of Tayshaneta species are discussed. Ten new species are described: Tayshaneta archambaultisp. n., Tayshaneta emeraldaesp. n., Tayshaneta fawcettisp. n., Tayshaneta grubbsisp. n., Tayshaneta madlasp. n., Tayshaneta oconnoraesp. n., Tayshaneta sandersisp. n., Tayshaneta sprouseisp. n., Tayshaneta vidriosp. n. and Tayshaneta whiteisp. n. The males for three species, Tayshaneta anopica (Gertsch, 1974), Tayshaneta devia (Gertsch, 1974) and Tayshaneta microps (Gertsch, 1974) are described for the first time. Tayshaneta furtiva (Gertsch, 1974) and Tayshaneta uvaldea (Gertsch, 1974) are declared nomina dubia as the female holotypes are not diagnosable and efforts to locate specimens at the type localities were unsuccessful. All Tayshaneta species are thoroughly illustrated, diagnosed and keyed. Distribution maps are also provided highlighting areas of taxonomic ambiguity in need of additional sampling.

Potassium stimulated calcium dependent release of immunoreactive somatostatin from incubated rat hypothalamus

S omatostatin (somatotropin release inhibiting factor, SRIF) is present in the median eminence of the hypothalamus in high concentration (K ronheim et al., 1976), is visualized in nerve endings (H ökfelt et al., 1974) and has been found to be concentrated in the synaptosome fraction of hypothalamic homogenates (E pelbaum et al., 1977; B erelowitz et al., 1978), suggesting a true neurosecretory role. To further explore this possibility we have studied the release of immunoreactive SRIF from the incubated rat hypothalamus (B radbury et al., 1974: R otsztein et al., 1977), basally and in response to depolarising concentrations of potassium, and have assessed the calcium dependence of this release.     

An enzymatic assay for acetate in spent bacterial culture supernatants

A method is presented for the rapid enzymatic determination of acetate in spent bacterial culture supernatants. The assay is based on a previously published assay for acetate kinase [Bergmeyer et al. (1974) in Methods of Enzymatic Analysis (Bergmeyer, H. V., ed.), Vol. 1, pp. 425-426, Verlag Chemie-Academic Press, New York/London], and is sufficiently sensitive to detect acetate levels of 50 microM. The assay is cheaper than commercially available assays and is particularly useful for occasional use by laboratories not equipped for routine acetate analysis using gas chromatography. The application of the assay to the measurement of acetate in bacterial cultures is described, though it should also be applicable to other biological fluids and foodstuffs.

Purification and partial characterization of alkaline phosphatase of matrix vesicles from fetal bovine epiphyseal cartilage. Purification by monoclonal antibody affinity chromatography

Alkaline phosphatase of matrix vesicles isolated from fetal bovine epiphyseal cartilage was purified to apparent homogeneity using monoclonal antibody affinity chromatography. The enzyme from the butanol extract of matrix vesicles bound specifically to the immobilized antibody-Sepharose in the presence of 2% Tween 20 whereas the major portion of nonspecific protein was removed by this single step. Of various agents tested, 0.6 M 2-amino-2-methyl-1-propanol, pH 10.2, was the most effective in eluting 80-100% of the enzyme initially applied. Both Tween 20 and 2-amino-2-methyl-1-propanol associated with the eluted enzyme were effectively removed by the sequential application of DEAE-cellulose and Sepharose CL-6B chromatography. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis of the enzyme preparation treated with sodium dodecyl sulfate and mercaptoethanol showed the presence of a dominant band (using silver staining) corresponding to a molecular weight of 81,000. This molecular weight was nearer reported values for rat liver (Ohkubo, A., Langerman, N., and Kaplan, M. M. (1974) J. Biol Chem. 249, 7174-7180) and porcine kidney (Cathala, G., Brunel, C., Chapplet-Tordo, D., and Lazdunski, M. (1975) J. Biol. Chem. 250, 6040-6045) alkaline phosphatase, than to previously reported values for chicken (Cyboron, G. W., and Wuthier, R. E. (1981) J. Biol. Chem. 256, 7262-7268) and fetal calf (Fortuna, R., Anderson, H. C., Carty, R. P., and Sajdera, S. W. (1980) Calcif. Tissue Int. 30, 217-225) cartilage matrix vesicle alkaline phosphatase. The purified alkaline phosphatase was activated by micromolar Mg2+. The amino acid composition of cartilage alkaline phosphatase was found to be similar to that previously described for porcine kidney (Wachsmuth, E. D., and Hiwada, K. (1974) Biochem. J. 141, 273-282). Double immunoprecipitation data indicated that monoclonal antibody against cartilage alkaline phosphatase cross-reacted with fetal bovine liver or kidney enzyme but failed to react with calf intestinal or rat cartilage enzyme. Thus these observations suggest that alkaline phosphatase of matrix vesicles from calcifying epiphyseal cartilage is a liver-kidney-bone isozyme.

New cretaceous radiolarian species of the genus <Emphasis Type="Italic">Afens</Emphasis> Riedel et Sanfilippo

Diverse radiolarians from the Upper Cretaceous Perapedhi and Moni formations (southern Cyprus) are studied. Many representatives of the genus Afens Riedel et Sanfilippo, 1974 are recognized. The diagnosis of the genus Afens is emended and three new species of this genus, Afens perapediensis sp. nov., Afens moniensis sp. nov., and Afens concinnus sp. nov., are described. In addition, the species Afens liriodes Riedel et Sanfilippo, 1974 is redescribed.     

The conformation of the polar group of lysophosphatidylcholine in H2O: conformational changes induced by polyvalent cations

The conformation of the polar group of egg lysophosphatidylcholine and 1-myristoyl-sn-glycer-3-phosphorylcholine present as micelles in aqueous solution has been studied using NMR methods. In the absence of polyvalent cations the preferred conformation derived from spin-spin coupling constants is similar, but not identical, to that of phosphatidylethanolamine in the crystal structure (cf. Hitchcock, P.B., Mason, R., Thomas, K.M. and Shipley, G.F. (1974) Proc. Natl. Acad. Sci. U.S. 71, 3036--3040). The presence of lanthanides induces a conformational change involving primarily the phosphorylcholine group, e.g. torsion angle alpha5 changes from an all gauche to an approximate trans disposition. The gauche leads to trans transitions observed with torsion angles alpha3 and alpha5 produce a more extended orientation of the polar group (relative to the hydrocarbon chain axis). In the presence of lanthanides the conformation of lysophosphatidylcholine is very similar to that of the diacyl phosphatidylcholines observed in fully hydrated bilayers (cf. Hauser, H., Phillips, M.C., Levine, B.A. and Williams, R.J.P. (1976) Nature 261, 390--394) with the P-N vector at an angle of about 45 degrees to the bilayer.
