White clover (Trifolium repens L.) was grown in controlled environmentsto determine the distinct effects of root and shoot temperatureon the accumulation of total and fixed (15 N dilution) nitrogenat two levels of nitrate (10 and 75 mM). Nitrogen fixation(BNF) showed a positive response to higher shoot temperature(23 vs. 13 C day temperature), irrespective of whether or notroot temperature was increased in parallel. Low root temperature(5 C) caused a marked reduction in the accumulation of totalnitrogen at both nitrate levels, and led to a lower proportionof N derived from BNF. The temperature response of BNF was attributedfor the major part to an adaptation to the demand for fixedN. It is therefore concluded that BNF is not primarily responsiblefor the reduced clover growth at low temperatures. Resource Optimization and Symbiotic Nitrogen Fixation   总被引:5,自引:1,他引:5  
In temperate forests, symbiotic nitrogen (N) fixation is restricted to the early phases of succession despite the persistence of N limitation on production late in succession. This paradox has yet to be explained adequately. We hypothesized that the restriction of N fixation to early stages of succession results from the optimization of resource allocation in the vegetation. Because of this optimization, N fixation should be restricted to periods when fixation is less costly than N uptake. Our analysis differs from others in the way we calculate the cost of N uptake; we assess the cost of N uptake as the amount of carbon (C) that could be assimilated if the resources necessary to acquire one gram of N from the soil were allocated instead to photosynthesis. We then simulate N fixation as an asymptotic function of the difference in cost between N uptake and N fixation and proportional to the abundance of host tissues for the N-fixing symbionts. The factors that contribute to conditions that favor N fixation are (a) elevated-carbon dioxide (CO2) concentrations, (b) an open canopy, (c) low available N in the soil, and (d) a soil volume already well exploited by roots. Our results indicate that changes in the relative cost of uptake vs fixation can explain most of the pattern in fixation through both primary and secondary succession, but that competitive interactions with nonfixing species play a role in the final exclusion of fixation in later stages of succession. Nodulated white clover plants (Trifolium repens L. cv. Huia)were grown as simulated swards for 71 d in flowing nutrientsolutions with roots at 11 C and shoots at 20/15 C, day/night,under natural illumination. Root temperatures were then changedto 3, 5, 7, 11, 13, 17 or 25 C and the total N2, fixation over21 d was measured in the absence of a supply mineral N. Alltreatments were subsequently supplied with 10 mmol m–2NO2 in the flowing solutions for 14 d, and the relativeuptake of N by N2, fixation and NO3 uptake was compared.Net uptake of K+ was measured on a daily basis. Root temperature had little effect on root d. wt over the 35-dexperimental period, but shoot d. wt increased by a factor of3.5 between 3 and 25 C, with the sharpest increase occurringat 7–11 C. Shoot: root d. wt ratios increased from 25to 68 with increasing temperature at 7–25 C. N2-fixationper plant (in the absence of NO2 ) increased with roottemperature at 3–13C, but showed little change above13 C. The ratios of N2 fixation: NO2 uptake over 14d (mol N: mol N) were 0.47–0.77 at 3–7 C, 092–154at 11–17 C, and 046 at 25 C, reflecting the dominanceof NO3 uptake over N2 fixation at extremes of high andlow root temperature. The total uptake of N varied only slightlyat 11–25 –C (095–110 mmol N plant–1),the decline in N2 fixation as root temperature increased above11 C was compensated for by the increase in NO 3 uptake.The % N in shoot dry matter declined with decreasing root temperature,from 32% at 13 C to 15% at 3 C. In contrast, concentrationsof N expressed on a shoot water content basis showed a modestdecrease with increasing temperature, from 345 mol m–3at 3 C to 290 mol m–3 at 25 C. Species differ in the relationship of nitrate reductase activityto nitrate uptake. In Capsicum annuum different diurnal patternsof leaf nitrate reductase activity and nitrate uptake have beenreported. As a consequence, the relationship of free nitratein the plant to nitrate supplied has a higher level of significancethan has reduced nitrogen to nitrate supplied. In Zea mays ithas been reported that leaf nitrate reductase activity respondsdirectly to nitrate translocation to the leaf and in this speciesthe relationship of greatest significance is reduced nitrogencontent to nitrate supplied. In both species, and also in Cucumis melo, the proportion oftotal plant free nitrate and reduced nitrogen in the roots decreases,and in the stem increases, with increasing nitrate supplied. The accumulation of free nitrate in leaves is accompanied bya quantitatively different relationship between reduced nitrogenand dry weight compared to leaves not accumulating nitrate. Simulated mixed swards of Perennial Ryegrass (Lolium perenneL.) cv. S23 and White clover (Trifolium repens L.) cv. S100were grown from seed under a constant 20 °C day/15 °Cnight temperature regime and harvested at intervals over and88 d growht period. The swards received a nutrient solutiondaily, which was either High (220 mg l1) or Low (10 mgl–1) in nitrate N. The nitrate was labelled with the 15Nisotope. An acetylene reduction assay was carried out on eachsward just prior to harvest. Rates of acetylene reduction agreed qualitatively with the l5Nanalyses but absolute values did not match (assuming a 4:1 C2H4:N2ratio) and errors in the acetylene assay are discussed. In theLow-N swards clover relied almost entirely on symbioticallyfixed N2, fixing more than ten times as much as the High-N cloverplants. In the Low-N treatment the grass was N-deficient despiteobtaining much more nitrate per unit root dry weight than clover.In the High-N swards, however, clover took up more nitrate perunit root weight than grass. The High-N clover plants also fixedsome N2 and maintained a higher total-N content than grass throughoutthe period. There was no evidence of transfer of symbioticallyfixed N from the clover to the grass in either treatment. Nodulated white clover plants (Trifolium repens L. cv. Huia)were grown for 71 d in flowing nutrient solutions containingN as 10 mmol m–3 NH4NO3, under artificial illumination,with shoots at 20/15°C day/night temperatures and root temperaturereduced decrementally from 20 to 5°C. Root temperatureswere then changed to 3, 7, 9, 11, 13, 17 or 25°C, and theacquisition of N by N2 fixation, NH4+ and NO3 uptakewas measured over 14 d. Shoot specific growth rates (d. wt)doubled with increasing temperature between 7 and 17°C,whilst root specific growth rates showed little response; shoot:root ratios increased with root temperature, and over time at11°C. Net uptake of total N per plant (N2 fixation + NH4++ NO3) over 14 d increased three-fold between 3 and 17°C.The proportion contributed by N2 fixation decreased with increasingtemperature from 51% at 5°C to 18% at 25°C. Uptake ofNH4+ as a proportion of NH4+ + NO3 uptake over 14 d variedlittle (55–62%) with root temperature between 3 and 25°C,although it increased with time at most temperatures. Mean ratesof total N uptake per unit shoot f. wt over 14 d changed littlebetween 9 and 25°C, but decreased progressively with temperaturebelow 9°C, due to the decline in the rates of NH4+ and NO3uptake, even though N2 fixation increased. The results suggestthat N2 fixation in the presence of sustained low concentrationsof NH4+ and NO4 is less sensitive to low root temperaturethan are either NH4+ or NO3 uptake systems. The effects of aluminium (Al3 +) on the growth of white cloverdependent upon symbiotic fixation of atmospheric nitrogen wereexamined at concentrations that may be encountered in solutionsfrom soils of low pH. Well established plants were grown inflowing solution culture with carefully maintained concentrationsof Al and of P and with pH held constant at a value (4.5) atwhich insoluble precipitates are unlikely to form. After 3 weeksof treatment, there were major visual differences between treatmentsin both shoots and roots. Although added Al at 25, 50 and 100mmol m–3 reduced dry weight, the differences between theplants were not significant. There were, however, some considerabledifferences in P and Ca contents between the treatments. Muchof the Al held by the roots was displaced when plants were transferredto solutions containing either scandium or gallium. As wellas the effects on the plant, Al had a considerable influenceon the fixation of atmospheric nitrogen. At concentrations of50 and 100 mmol m–3 Al3+ nodule initiation was reducedand there was a much reduced nodule nitrogenase activity perunit of root. Photosynthetic data collected from Pisum sativum L. and Phaseolus vulgaris L. plants at different stages of development were related to symbiotic N2 fixation in the root nodules. The net carbon exchange rate of each leaf varied directly with carboxylation efficiency and inversely with the CO2 compensation point. Net carbon exchange of the lowest leaves reputed to supply fixed carbon to root nodules declined in parallel with H2 evolution from root nodules. The decrease in H2 evolution also coincided with the onset of flowering but preceded the peak in N2 fixation activity measured by acetylene-dependent ethylene production. A result of these changes was that the relative efficiency of N2 fixation in peas increased to 0.7 from an initial value of 0.4. The data reveal that attempts to identify photosynthetic contributions of leaves to root nodules will require careful timing and suggest that the relative efficiency of N2 fixation may be influenced by source-sink relationships.  相似文献

Subterranean clover (Trifolium subterraneum L.) cv. ‘Woogenellup’ swards were grown at 10, 15, 20 and 25 Cwith a 12 h photoperiod of 500 or 1000 µmol m–2s–1 [low and high photosynthetic photon flux density (PPFD)].Nitrogen-fixing swards received nutrient solution lacking combinednitrogen while control swards received a complete nutrient solution.Growth was measured by infra-red analysis of carbon dioxideexchange and by accumulation of dry matter. Swards were harvestedat intervals between 95 and 570 g d. wt m–2 for estimationof nitrogenase activity by acetylene reduction and hydrogenevolution assays. Nitrogen fixation was also measured by increasein organic nitrogen. The growth rate was highest at 10 C at low PPFD, and at 10–15C at high PPFD. Nitrogen-fixing swards grew slower than thosereceiving combined nitrogen. Nitrogen fixation measured by increasein organic nitrogen responded similarly to the growth rate,as did acetylene reduction between 10 and 20 C. At 25 C therelationship between acetylene reduction and nitrogen fixationwas distrupted. The difference between the rates of acetylenereduction and hydrogen evolution, theoretically proportionalto nitrogen fixation, was not a reliable indicator of nitrogenfixation because hydrogen uptake developed. Single plants of white clover grown in controlled environments,and dependent for nitrogen on N, fixation, were defoliated at1 or 2 d intervals to 3, 2 and 1 expanded leaves per stolon(Expt 1), and to 1,0.5 (1 leaf on every alternate stolon) and0 expanded leaves per stolon (Expt 2), for 43–50 days Plants adapted to severe defoliation by developing much smallerleaves with a slightly reduced specific leaf area, more stolons,a smaller proportion of weight in leaf, root and nodules anda greater proportion of weight in stolons. The daily yield (materialremoved by defoliation) of d. wt and nitrogen generally decreasedwith severity of defoliation, as did the residual plant weight.However, the ‘efficiency’ of yield (daily yield/residualweight x 100) of dry matter and nitrogen was greater in themost severely defoliated treatments, attaining a maximum of5–6 % All plants adapted to the imposed defoliation regimes, howeversevere, with the result that even plants maintained withoutany fully expanded leaves invested a similar fraction of theirmetabolic resources in shoot and root as less severely defoliatedplants, and continued to grow and fix N2, albeit at a very reducedrate of 1–2 mg Nd–11. The energetic cost of N2 fixation(acetylene reduction) remained constant in all treatments at31 mole CO2 mole C2H4–1, but there was some evidence thatrate of N2 fixation per unit of nodule weight declined in themost harshly defoliated treatment. Single plants of white clover, grown in a controlled environmentand dependent for nitrogen on fixation in their root nodules,were defoliated once by removing approximately half their shoottissue. Their regrowth was compared with the growth of comparableundefoliated plants. Two similar experiments were carried out:in the first, plants were defoliated at 2.5 g, and in the secondat 1.2 g total plant d. wt. Defoliation reduced rate of N2 fixation by > 70 per cent,rate of photosynthesis by 83–96 per cent, and rate ofplant respiration by 30–40 per cent. Nodule weights initiallydeclined following defoliation as a result of loss of carbohydratesand other unidentified components. No immediate shedding ofnodules was observed but nodules on the most severely defoliatedplants exhibited accelerated senescence. The original rates of N2 fixation were re-attained after 5–6or 9 d regrowth, with increase in plant size at defoliation.In general, the rate of recovery of N2 fixation was relatedto the re-establishment and increase of the plant's photosyntheticcapacity. Throughout the growth of both defoliated and undefoliatedplants nodule respiration (metabolism) accounted for at least23 ± 2 per cent of gross photosynthesis. The unit ‘cost’of fixing N2 in root nodules, in terms of photosynthate, appearedto be unaffected by defoliation, except perhaps for plants veryrecently defoliated. Similarly, the percentage nitrogen contentsof shoot, root and nodules of defoliated plants became adaptedwithin a few days to those characteristic of undefoliated plants. Previous studies have shown that nitrogenase activity decreases dramatically after defoliation, presumably because of an increase in the O2 diffusion resistance in the infected nodules. It is not known how this O2 diffusion resistance is regulated. The aim of this study was to test the hypothesis that current N2 fixation (ongoing flux of N2 through nitrogenase) is involved in the regulation of nitrogenase activity in white clover (Trifolium repens L. cv Ladino) nodules. We compared the nitrogenase activity of plants that were prevented from fixing N2 (by continuous exposure of their nodulated root system to an Ar:O2 [80:20] atmosphere) with that of plants allowed to fix N2 (those exposed to N2:O2, 80:20). Nitrogenase activity was determined as the amount of H2 evolved under Ar:O2. An open flow system was used. In experiment I, 6 h after complete defoliation and the continuous prevention of N2 fixation, nitrogenase activity was higher by a factor of 2 compared with that in plants allowed to fix N2 after leaf removal. This higher nitrogenase activity was associated with a lower O2 limitation (measured as the partial pressure of O2 required for highest nitrogenase activity). In experiment II, the nitrogenase activity of plants prevented from fixing N2 for 2 h before leaf removal showed no response to defoliation. The extent to which nitrogenase activity responded to defoliation was different in plants allowed to fix N2 and those that were prevented from doing so in both experiments. This leads to the conclusion that current N2 fixation is directly involved in the regulation of nitrogenase activity. It is suggested that an N feedback mechanism triggers such a response as a result of the loss of the plant's N sink strength after defoliation. This concept offers an alternative to other hypotheses (e.g. interruption of current photosynthesis, carbohydrate deprivation) that have been proposed to explain the immediate decrease in nitrogenase activity after defoliation.  相似文献

Wrinkled-seeded pea mutants (Pisum sativum L., genotypes rrrbrb-, rrRbRb-, and RRrbrb-) have seeds with reduced, but different, starch content and modified starch properties. Analysis of these mutants revealed an enhanced capacity of root nodules for symbiotic nitrogen fixation and of host plant organs for assimilation of ammonium nitrogen. This observation was confirmed by morphological data on organization of symbiotic system, by elevated nitrogenase activity, high protein accumulation in plants due to nitrogen fixation, and by enhanced activity of glutamine synthase in leaves and glutamate dehydrogenase in roots of mutants, as compared with the organs of wild-type pea. It is supposed that the aforementioned advantages of mutants are related to accumulation in seeds of elevated protein reserves that satisfy their demand for nitrogen during formation of symbiotic systems.  相似文献

Rhizobium meliloti bacteroids carrying mutations in either fdxNor fixX isolated from alfalfa root nodules were shown to containthe nitrogenase proteins NifH, NifD and NifK. In contrast toan in vitro system of N2-fixation based on R. meliloti wild-typebacteroids, nitrogenase activity could not be restored in crudeextracts of these mutant bacteroids by the addition of an artificialelectron donor, indicating that the nitrogenase proteins werepresent but not functional. ESR-studies revealed that both mutantslacked the FeMo-cofactor of nitrogenase. To analyse the roleof free O2 on the damage of the nitrogenase components and theFeMo-cofactor in these mutant bacteroids, microelectrode studiesof O2 concentrations and gradients within alfalfa root noduleswere carried out. R. meliloti mutants defective in other genesnecessary for symbiotic N2-fixation were also included in thisstudy. Four distinct types of O2 gradients were defined by theapparent presence or absence of an O2 diffusion barrier andby the minimum internal O2 concentration. These data clearlydemonstrated the influence of the microsymbiont on the O2 gradientswithin the nodules. Nodules induced by Rm0540, an R. melilotimutant with altered exopolysaccharide production, which is notable to infect plant cells, did not contain an O2 diffusionbarrier. In contrast, nodules containing a mutant defectivein dicarboxylate transport (dctA-), produced an O2 gradientsimilar to the wild-type. Microelectrode measurements revealedH2 concentrations in alfalfa wild-type nodules comparable tosoyabean, whereas no hydrogen could be detected in nodules harbouringthe dctA mutant or any other mutant strain. Seedlings of Italian ryegrass (Lolium multiflorum Lam. cv. RVP)and clonal stolon cuttings of white clover (Trifolium repensL. cv. Blanca) were grown for 19 d in flowing solution culture,with N supplied as either 250 mmol m–3 NO3 or NH3+.Rates of net uptake, influx and translocation of NO3and NH4+ were then determined using 15N and 13N labelling techniques:between 3–5 h into the photoperiod following 8 h darknessfor white clover (CL), and for ryegrass plants that were eitherentire (IL) or with shoots excised 90 min prior to 13N influx(IC); and 75 min into the photoperiod following 37–39h darkness for ryegrass (ID). Rates of net uptake, influx andefflux of NH4+ exceeded those of NO3 in IL and IC ryegrassplants: the opposite occurred in white clover (CL). The decreasein net uptake following defoliation of ryegrass was greaterfor NH4+ (62%) than NO3 (40%). For NH4+ this was associatedwith a large decrease in influx from 110 to 6.0µmol h–1g–1 root fr. wt; but for NO3, influx only decreasedfrom 42 to 37 µmol h–1 g–1. Prolonged exposureto darkness (ID plants) also lowered net uptake of NO3and NH4+ by, respectively, 86% and 95% of IL levels. For NH4+this was characterized by a large decrease in influx and a smalldecrease in efflux; whilst for NO3 the effect of a largedecrease in influx was reinforced by a smaller increase in efflux. The data were used to estimate the translocatory fluxes of NO3(03–20µmol h–1 g–1) and NH4+ (003–0.4µmolh–1 g–1), assimilation in the roots of NO3(02–26µmol h–1 g–1) and NH+4 (05–89 µmolh–1 g–1), and the concentrations of NO3 (9–15mol m–3) in the cytoplasmic compartment of the roots.The relevance of variable influx and efflux to models for theregulation of N uptake is discussed. Oxygen Diffusion Pathways and Nitrogen Fixation in Legume Root Nodules   总被引:1,自引:0,他引:1  
The theoretical analysis presented in this paper suggests thatthe nature of the diffusion pathway from the surface of a noduleto the infected zone depends on the morphology of the nodule;in particular the cross-sectional area of the intercellularspaces in the inner cortex. If the diffusion barrier containscontinuous pathways there will be no pressure difference betweenthe atmosphere and the infected zone. The conditions under whichthe intercellular spaces of the inner cortex could be air-filled,water-filled, or a combination of both are explored. An experimentto resolve this issue is suggested. Information obtained usingcryo-scanning electron microscopy and oxygen electrode datahave been used to illustrate various points. Expressions arederived for the diffusion resistance of the nodule and its componentparts. To provoke further discussion a simple mechanism forthe control of diffusion is described in general terms. We evaluated soil phosphorus (P) fractions, other soil characteristics, and rates of symbiotic N2 fixation across a substrate-age gradient in Hawaii that was dominated by the leguminous tree Acacia koa (koa). Patterns of soil P observed on this gradient were compared to those on a slightly wetter gradient dominated by the nonfixer Metrosideros polymorpha (ohia). Along both gradients, concentrations of primary-mineral P fell sharply between the young and intermediate-aged sites, while labile inorganic P declined most steeply between the intermediate-aged and old sites. The most marked difference between the two gradients was that total soil carbon (C), nitrogen (N), and P, as well as nonoccluded organic P, were more variable across the ohia gradient, increasing to the intermediate-aged sites, then declining sharply at the old site. On the koa gradient, specific nitrogenase activity, measured by the acetylene-reduction (AR) assay, decreased three- to eightfold between the young site and the intermediate-aged and old sites, respectively. Nodule biomass showed no clear pattern. N2 fixation rates, estimated by combining AR activity and nodule biomass measurements, were up to 8 kg N · ha−1 · y−1 at the young site and no more than 2 kg N · ha−1 · y−1 at the older sites, suggesting that koa may be a modest source of N in these Hawaiian forests. The aim of this study was to test the effect of oxygen partialpressure as a possible limiting factor of nitrogen fixationfollowing defoliation. The response of nitrogenase activity(C2H2-reduction) of defoliated and undefoliated white and redclover plants (Trifolium repens L. and Trifolium pratense L.)to either 19 kPa oxygen or 55 kPa oxygen was investigated. Priorto defoliation, white clover plants were grown for five weeksin a growth chamber, and red clover plants for 7 or 11 weeksin a glasshouse. The results included measurements of 16N2-uptake. Increasing oxygen partial pressure from 19 to 55 kPa severelyrestricted nitrogenase activity of undefoliated white cloverplants; however, 2 h after complete defoliation, the same treatmentcaused a significant increase. A fivefold increase in nitrogenaseactivity upon exposure to the elevated oxygen partial pressurewas found at the end of a 24 h period. This beneficial effectdecreased gradually from 1 to 5 d after defoliation. The responseof recently defoliated red clover plants to 55 kPa oxygen partialpressure was similar to that of white clover, independentlyof plant age. The gradual recovery of nitrogenase activity duringthree weeks of regrowth was associated with a simultaneous changein the response to increased oxygen partial pressure, leadingagain to the response of undefoliated plants. These data suggested that lack of oxygen at the site of nitrogenfixation, resulting from a dramatic increase in oxygen-diffusionresistance, is the main factor limiting nitrogenase activityfollowing defoliation.
