以灵芝(Ganoderma lucidum)之赤芝(Ganoderma lucidum Karst)、韩芝(Ganoderma luci- dum H.G)为试验品种,培养基以棉壳98%、石膏1%、蔗糖1%为对照,另外以棉壳、麸皮、木屑、氨基酸不同含量配制4种培养基作比较栽培试验。结果表明:2号培养基出菇产量高、菇形好,对赤芝、韩芝都适宜;赤芝、韩芝对培养基的要求略有差异,韩芝在含麸皮量高的基质上栽培更为适合。 相似文献

This technique is very useful where pollen is readily available and when roots or microspore mother cells are difficult to obtain or to process. It provides a relatively uniform means of studying chromosomes in a great number of species. Pollen is collected from buds at anthesis or the day before and sown on a medium containing H3BO3, 100 ppm; colchicine, 0.04%; lactose, 12%; gelatin, 5%; egg albumen, 1 drop in 10 ml. The gametic chromosome complement is studied at mitosis of the generative cell in the pollen tube. In species which have sufficiently large chromosomes it is possible to construct idiograms for comparative studies. All palm species here reported have a haploid complement of n = 18, and the chromosomes range in length from 0.5-3.5 μ. 相似文献

It is possible to grow pollen tubes routinely for cytological analysis of nuclei at the pollen tube division. Pollen has been grown successfully after temperature, pressure, gas, moisture, and radiation treatments. The technic for growing Tradescantia pollen is described, but any method is satisfactory which ensures that: (a) the pollen is kept dry before sowing, (b) a minimum of time elapses between sowing pollen on culture medium and placing in a moist growing box, and (c) the growing pollen tubes are not allowed to dry out. We propose a method to construct adaptive tests based on a bootstrap technique. The procedure leads to a nearly exact adaptive test depending on the size of the sample. With the use of the estimated Pitman's relative efficacy as selector statistic, we show that the adaptive test has a power that is asymptotically equal to the power of it's better component. We apply the idea to construct an adaptive test for two-way analysis of variance model. Finally, we use simulations to observe the behaviour of the method for small sample sizes. 相似文献

采用液体培养法研究不同培养基组分和培养条件对蜡梅花粉萌发和花粉管生长的影响。结果表明:(1)PEG-4000是蜡梅花粉离体培养所必需的培养基成分,当培养基中无PEG-4000时,花粉不能正常萌发。(2)培养基内低浓度蔗糖对花粉萌发和花粉管的生长无显著影响,但随着蔗糖浓度的升高,则对花粉萌发和花粉管生长表现出强烈的抑制作用,且浓度越高,抑制效应越强。(3)培养基内其它组分分别在一定浓度范围(0~250g/L PEG-4000、0~50mg/L硼酸、0~30mg/L硝酸钙)内对花粉萌发及花粉管生长有促进作用,但超过上述高限值时则起抑制作用。(4)培养基内镁和钾的浓度对花粉萌发及花粉管生长影响不显著。研究表明,蜡梅最适花粉液体培养基组分为250g/L PEG-4000+50mg/L H3BO3+30mg/L Ca(NO3)2.4H2O,且在pH 5.5、温度15℃和600lx的光照培养条件下蜡梅花粉萌发和花粉管生长最佳。 相似文献

为提高金黄色葡萄球菌(Staphylococcus aureus)发酵产生的菌体及荚膜多糖产量,对4种不同培养基(THB肉汤、哥伦比亚液体培养基、营养肉汤和脑心浸出液)进行了筛选,利用单因素及正交试验对培养条件进行了优化,并通过透射电镜对优化前后细菌的荚膜进行了比较观察。结果表明,添加2%乳清的哥伦比亚液体培养基最适合金黄色葡萄球菌的生长,优化后的培养条件为发酵液p H 7.0,接种量1%,37℃、200 r/min培养至16 h。与优化前相比,菌体产量提高了20%;电镜观察表明,有荚膜菌体的数量增加且荚膜层增厚。本研究为后期多糖疫苗的研究提供参考。 相似文献

在遗传力的估计过程中,需将多种非遗传因素的影响从公畜间方差或者母畜间方差中剔除。在我国常使用的是盛志廉教授提出的单元内同胞相关法。本文对该法从理论上进行了更详细的证明,并将其推广到两层分类方差分析时的情况。同时还给出了当公母畜彼此间有亲缘关系时,利用单元内方差分析估计遗传力的方法。这些方法既可使遗传力的估计简便,又具有多因方差分析的功用。 相似文献
