2 novel methods of coprophagy prevention were used in iron-deficient rats to measure their effect on haemoglobin regeneration during the feeding of ferrous sulphate or several soya proteins. Neck collars produced considerable weight loss and depression in food intake compared to aluminium anal cups which were tolerated well. Both methods of coprophagy prevention reduced iron availability from a range of soya proteins and ferrous sulphate and it is suggested that the practice of coprophagy in iron-deficient and possibly iron-replete rats may overestimate iron (and possibly other mineral) absorption (retention) studies.  相似文献

At many breeding facilities, rats are housed at relatively high densities until they are 5 weeks old, at which point they are either shipped for research or rehoused at standard cage densities according to weight. The authors carried out a pilot study in Long Evans and in Fischer 344 rats to investigate whether continuing to house rats at high densities (24 in(2) floor space per rat) past the age of 5 weeks, through puberty and into adulthood would alter behavioral or physiological parameters compared with raising rats at standard densities (about 72 in(2) floor space per rat). After rats reached puberty, the authors rehoused them with unfamiliar cagemates. The researchers evaluated clinical and behavioral signs of stress, weight, blood glucose concentration, white blood cell count and serum corticosterone concentration. Overall, cage density had little effect on the parameters measured, though gender seemed to affect stress in Long Evans rats. The results suggest that rats of these strains can be raised at the higher densities tested until any age and regrouped with unfamiliar cagemates without compromising rats' welfare or subsequent experimental data.  相似文献

Bacteriol. 92: 13-16. 1966.-Lactobacilli were enumerated in the feces of rats prevented from coprophagy by tail-cupping. No differences were found when numbers of these organisms were compared with lactobacilli in feces of control rats, without tail-cups. High and similar numbers of lactobacilli were found in the stomachs of rats with and without tail-cups. The effect of coprophagy on fecal lactobacilli was therefore negligible.  相似文献

In order to investigate coprophagy from the viewpoint of nutrition, fecal constituents were analyzed in freeze-dried samples. Feces were collected from 7:00 to 11:00 and from 19:00 to 23:00. Inorganic elements and crude fibers per unit weight were 3-4 times more concentrated in feces than in basal diet, whereas, crude proteins, crude fats and nitrogen-free extract showed various degrees of reduction. There were no differences in these tendencies with sampling time. As for some B vitamins, feces collected from 7:00 to 11:00 contained 22-92% more vitamins than feces collected from 19:00 to 23:00. In comparison with the dietary concentration, vitamin B12 was increased by 124-197 times (520-730 micrograms/100 g) in feces collected between 7:00 and 11:00. Folic acid in feces collected between 7:00 and 11:00 was 10 times greater than that in the diet. On the basis of the findings on vitamins, the effect of a vitamin B12 fortified diet (1,350 micrograms/100 g) on coprophagy was examined. Mean frequency of coprophagy per animal per day was 9.6 when animals were fed on the basal diet, whereas the frequency was immediately and significantly (p less than 0.05 approximately p less than 0.01) reduced to 4.7 after the diet had been replaced by the fortified one. However, coprophagy was not completely inhibited by vitamin B12 fortification. This indicates that some nutrient(s) in feces other than vitamin B12 might be of use to the host, and that otherwise, coprophagy might be a basically habitual form of behavior. The aim of the study was to assess whether positioning of the operant cost would affect blue foxes’ (Vulpes lagopus) behaviour and time budget between a mesh floor and sand floor. We built fourteen test apparatuses, six consisting of two fox cages with a mesh floor in both cages and eight with a mesh floor in one cage and a sand floor in the other cage. A cost could be imposed to pass through one-way doors between the cages. The effect of changing the cost to move from the mesh floor to the sand floor or from the sand floor to the mesh floor or between the two mesh floors was examined in 28 juvenile male blue foxes. The foxes’ time allocation between the cages was recorded for four different costs. The foxes’ behaviour was analysed in more detail at the lowest and highest costs. The visit duration increased considerably when leaving the cage became costly, but did not markedly change when entering was costly. The foxes spent less time on the sand floor than on the mesh floor. The foxes preferred to rest on the mesh floor, but rested also on the sand floor when there was a cost associated with leaving the sand floor. The sand floor stimulated digging, rooting and play. In conclusion, both positioning of the cost and the floor material affected blue foxes’ time budget between the two cages.  相似文献

A device for intragastric nutrition of unsedated and minimally restrained rats under complete alimentary abstinence has been developed. The cannulation system consists of an infusion pump, modified glass syringe as flow swivel, rat-harness and a silicone-tube-gastrostomy. The animals were kept in special cages and coprophagy was prevented by an own model of faecal collection cup. Methionine and Ca-glycerophosphate had to be added to a commercial elementary diet. The rats were allowed to move freely during intragastric infusion, which was applied for 14 to 28 days in 118 Wistar-rats (350-400 g). They maintained weight on an energy supply of 86.4 kcal/day.  相似文献

1. Nicotinic acid-deficient rats were given a dose of niacytin or an equivalent one of free nicotinic acid or hydrolysed niacytin. 2. The excretion of N'-methylnicotinamide and of tertiary nicotinoyl derivatives in urine showed that niacytin was not metabolized as free nicotinic acid, although hydrolysed niacytin was equivalent to free nicotinic acid. 3. Little or none of the niacytin dose was recovered as tertiary nicotinoyl derivatives in faeces. This result was not affected by fitting rats with tail-cups to prevent coprophagy. 4. At the high doses used niacytin restored the growth rate of the deficient animals because of a small degree of hydrolysis of the bound nicotinic acid.  相似文献

The microclimate in two types of rat cages (a Makrolon type IV with a solid floor and a stainless steel cage with a wire mesh floor (five rats per cage)) placed in the same macro-environment was compared. The temperature, relative humidity and ammonia concentration in the cages were measured twice a day for 8 days. The cages were cleaned every 4 days. The greatest difference between the cage types was in the ammonia build-up. In Makrolon cages the ammonia concentration never reached 5 ppm, whereas in steel cages it showed a constant increase and already exceeded the threshold limit for man (25 ppm for 8 h per day) on the third day after cleaning.  相似文献

Abstract

Most animal studies conducted to determine the bioavailability of lead have, in the past, employed rodents or lagomorphs as experimental models. In this paper issues and data are presented which raise questions and uncertainties about employing rodents or lagomorphs for investigations into the bioavailability of lead. These issues include: (1) the possible role of coprophagy and feeding behavior in reducing estimates of lead bioavailability; (2) anatomical and physiological differences related to coprophagy which may influence estimates of lead bioavailability derived in rats or rabbits; (3) evidence for relatively high biliary excretion of lead by rats and rabbits; (4) the possibility of a strong developmental component to the active transport of lead. The importance of addressing these and other questions in studies designed to determine the bioavailability of lead is discussed.  相似文献

Farmed blue foxes are willing to work to gain access to a sand floor from a wire mesh floor. It is not clear whether the foxes work for the sand floor because of its solidity or because it enables them to perform certain behaviours, e.g. exploration and digging. Here, we measured blue foxes’ motivation to gain access from a wire mesh floor to a floor with 15–30 cm deep sand, a floor with 3–4 cm deep sand, a solid concrete floor, and another wire mesh floor. In addition, we analysed the foxes’ behaviour on these floor materials. Seven male blue foxes were trained and tested in self-constructed operant apparatuses. In an apparatus, the fox could move a bottomless test cage from a wire mesh floor to a neighbouring, alternative floor material for a 4-min visit by pressing a lever in the test cage for a fixed number of times (Fixed Ratio, FR). The foxes worked for each floor material for 12 days. In each daily test session, the foxes were exposed to work on one of the four workloads (FR 6, 12, 24, 48), for 3 h. The behaviour of the foxes was analysed during the 4-min visits on each floor material. The results showed that there was no difference between the floor materials, either in the demand elasticity of the fragment of the demand curve (ranging from −0.46 to −0.33), or in the intensity of the demand. However, the foxes’ behaviour varied between the floor materials. More digging, play, rooting (exploration with the muzzle), and vole jumping were observed on the floor materials with sand, than on the concrete floor and the wire mesh floor. Both the presence and the depth of the sand layer stimulated these behaviours. It is concluded that juvenile blue foxes do not value solid floor materials more than a wire mesh floor. However, the sand floor stimulates more digging, play, vole jumping, and exploration than the concrete floor or wire mesh floor. Furthermore, the depth of sand may be an important factor in eliciting these behaviours. Access to a floor material with sand may improve the welfare of farmed blue foxes by providing the possibility to perform species-specific behaviours.  相似文献

Cage space requirements for laboratory animals have been established by Government Regulation and Recommendations. In order to test the adequacy of these space allocations, the use of cage floor area by breeding groups of guineapigs was studied. A computer-coupled video tracking system capable of imaging in low light intensity as well as total darkness was used to determine the average per cent occupancy by guineapigs in all portions of a cage over 12-h light and dark cycles. Simultaneous time synchronized slow motion video recordings permitted an analysis of activity to be coordinated with cage use data. Results of the study revealed that breeding groups of guineapigs utilize the periphery of the cage almost to the total exclusion of the centre of the cage. Approximately 75-85% of all occupancy in both the day and evening hours occurred in 47% of the cage floor area located along the periphery. Analysis of video recordings revealed that the animals remained active throughout the day and night with no prolonged period of quiescence that could be associated with sleep. Results of this study suggest that while guidelines for housing guineapigs based on area allocation per animal can be formulated and are easy to administer, they cannot be supported by the behavioural characteristics of these animals or careful quantitation of their pattern of cage space utilization.  相似文献

Farmed silver foxes (Vulpes vulpes) were allowed to balance their known preference for an elevated floor against their presumed preference for a sand floor. In Experiment 1, foxes had to choose between two identical cages, connected with an opening. One cage had a wire floor and the other had a sand floor, but the cages either were on the same (low or elevated) or on different levels (one cage 40 cm higher than the other). In Experiment 2, the cage pairs were connected with a 1.2 m long wire-mesh tunnel, one cage was always on a higher level (50 cm) than the other. In Experiment 1, foxes always preferred the sand floor during their active time. They also preferred the sand floor for resting, if it was on the same level as wire floor, but did not show any genuine preference if the floors were on different levels. In Experiment 2, foxes never preferred the lower floor. They preferred the elevated sand floor for activity and the elevated wire floor for lying. When two floors were identical they preferred the elevated one. Their rest consisted of 11-22 bouts, a major part of these being spent in the preferred cage. They also preferred a previous lying site to a new one, often exclusively and independently of floor material. In Experiment 1 foxes preferred the sand floor whereas in Experiment 2 they preferred the elevated floor indicating that the ability of a trade-off situation to rank resources depends on the method it is inflicted.  相似文献

The present work investigated whether the number of rats housed in a cage affects exploration of an elevated plus-maze. Male Wistar-derived rats were kept 1, 2, 3, 4, 6, 8, 12, 16, or 24 to same size cages either for 1 or 14 days and tested in the elevated plus-maze. Rats kept 6 to a cage were arbitrarily considered controls because this is the housing condition adopted in many laboratories, ours included. In comparison to controls, 1-day housed rats kept 1, 2, 16, and 24 to a cage decreased the percentage of entries into the open arms. Similar decreases were also found in the time spent in the open arms, the only exception being the group with rats kept 16 to a cage which failed to show significant differences from the control group. Fourteen-day housed rats kept 1, 2, 16, or 24 to a cage decreased the percentage of entries and time spent in the open arms. We found plus-maze exploration to be similar in groups in which rats were kept from 4 to 12 to a cage. The present data indicate that anxiogenic effects resulting from under- and overcrowding should be taken into consideration in behavioral studies.  相似文献

In the first experiment, farmed blue foxes were allowed to choose for 1 week between four standard farm cages equipped with different floor materials: plastic-coated wire mesh, dry wood, dry sand and wet (summer) or icy (winter) sand. Resting consisted of 10-15 separate bouts/day occupying 50-60% of the total 24-h. There were no other differences in the use of the cages except that the time spent on, and the duration and number of resting bouts were lower on wet or icy sand, resting periods being more affected than activity. In the second experiment, two cages were connected with a 1.2 m tunnel. One cage was always elevated (50 cm) compared to that one which was lower. One cage of each pair had a wire floor whereas the other cage either had sand or wire floor. Sand floor was preferred for active behaviours and wire floor for resting if these were on elevated level. Of two identical wire-floored cages, the elevated one had the priority. Foxes preferred to rest on the same floor where they had finished their previous resting bout, often exclusively and independently of floor material or floor level.  相似文献

Males rats of the Wistar strain (Institute of Physiology, CSAV ) were divided into six groups from the 30th to the 200th day of age. Rats (n = 54) were put into cages in various numbers (1, 2, 3, 4, 6 and 8 animals in a cage) and were weighed regularly and the consumption of water and food was measured. At the end of the experiment the animals were killed and the weight of some organs and haematological values were determined. During the experiment the highest weight was attained in animals with 3 or 4 members in one cage. The differences in weight were significant from the 80th day of age. The consumption of diets and water was not influenced significantly by the number of animals in a cage. On the 200th day, the differences between the groups were found in the weight of some organs, and in some haematological values.  相似文献

The present study was designed to measure the use of various, simultaneously available resources in a complex housing environment in juvenile blue foxes. Twelve blue fox sibling (male–female) pairs were housed in two-section experimental cages from the age of 8 weeks until the age of 7 months (from June to December). Each experimental cage was furnished with two platforms, a nest box, a sand box and a wooden block. This housing set-up provided the foxes with social contact, and an opportunity for oral manipulation, scratching and nesting, as well as the choice of staying on a solid floor material or on an elevated location. The foxes’ behaviour was recorded at three time points during autumn (September, November and December). The foxes used all available resources. The most utilised resource was the nest box, possibly because it could be utilised in several ways (as a shelter, an elevated location, an object for scratching and for oral manipulation). The foxes also stayed more in the cage section containing the nest box than in the cage section containing a sand box. The foxes rested much on the cage floor, but they also used the interior of the nest box and elevated locations for resting. Social contact often occurred during resting. Thus, the nest box and elevated location, in conjunction with social contact seem to be valuable while resting. While active, the foxes utilised the cage floor and roof of the nest box instead of the platforms. Scratching, digging and an interaction with the wooden block were seldom observed. Activity occurred mainly on the ‘empty’ cage area. In conclusion, all studied resources provided blue foxes with a distinct value, as they all were used in the complex housing environment. The improvement of housing conditions for mice by using environmental enrichment materials is of high concern for the scientific community. Plastic, autoclavable nest boxes are commercially available and ready to use for specific cases such as in individually ventilated cages, metabolic cages, or during toxicological studies. The aim of this study was to see if the location of the nest box within the cage could influence the mice to prefer and use it. Located on the cage floor or hung from the cage lid, a nest box (MPLEX, Otto Environmental, Milwaukee, Wisconsin), enriched the cages. The study concluded that the location of the nest boxes in the individually ventilated cage plays a significant role in the mice preferring to use it or to avoid it. It is also important to use environmental enrichment items that provide animals with the possibility of expressing their preferences and manipulating them in a way to cope better with their environmental conditions.  相似文献

The cage systems commonly used for housing laboratory rats often result in sedentary and overweight animals, as a consequence of restricted opportunities for physical activity combined with ad libitum feeding. This can have implications both for animal well-being and for the experimental outcome. Physical activity has several known positive effects on health and lifespan, and physical fitness might therefore be incorporated into the animal welfare concept. The aim of this study was to investigate if and how pen housing affects the physical activity and fitness of rats. Thirty-two juvenile male Sprague-Dawley rats were randomly assigned to two different housing systems for a 4-week period. Sixteen rats were kept individually in standard Makrolon type III cages (42x26x18 cm) furnished with black plastic tubes (singly-housed, SI). The remaining rats were kept in groups of eight, housed in large floor pens (150x210 cm), which were furnished with various objects to increase environmental complexity (pen-housed, PH). The body weight gain, and food and water intake of the rats were measured. During weeks 3 or 4, home cage behaviour, urinary cortiosterone/creatinine ratios (CO/CR), and muscle strength on an inclined plane, were measured. Enzyme activities and glycogen content were measured in tissue samples from m. triceps brachii taken after euthanization at the end of the study. There were no significant differences between groups for food and water intake, but PH rats weighed 14% less than SI rats after 4 weeks, and PH rats also had a more diverse behavioural pattern than SI rats. PH rats had significantly higher oxidative capacity (28% more citrate synthase (CS)) and greater glycogen content (28%) in their muscle samples than SI rats. The PH rats performed significantly better on the inclined plane, both in the muscle strength test (mean angle 75+/-0.5 degrees for PH rats and 69+/-0.4 degrees for SI rats) and the endurance strength test (mean time 233+/-22 s for PH rats and 73+/-14 s for SI rats). There was a negative correlation between body weight and results on the inclined plane for the PH rats. There were no significant differences between housing types with respect to CO/CR ratios. In conclusion, the large pen represents an environment that stimulates physical activity and more varied behaviour, which should be beneficial for the welfare of the animal.  相似文献

The International Space Station will allow extended habitation in space and long-term exposure to microgravity (microG). A concern is the impact of long-term microG exposure on the ability of species to reproduce. The model often used to simulate microG is rat hindlimb suspension (HLS), where the hindlimbs are elevated above the cage floor with a tail harness. Experiments described here are the first to examine the effect of long-term HLS on testicular function in adult male rats. Free-roaming (controls), animals with only the tail harnessed but hindlimbs in contact with the cage floor (TO), and HLS animals were tested for 6 wk. Cryptorchidism was prevented in TO and HLS animals by partial constriction of the inguinal canal with sutures. All parameters were compared at the end of the 6-wk experiment. Testicular weights and spermatogenesis were significantly reduced by HLS, such that no spermatogenic cells beyond round spermatids were present and epididymides were devoid of mature sperm. In many tubules, loss of all germ cells, except a few spermatogonia, resulting in histopathology similar to the Sertoli cell, was observed. Spermatogenesis appeared unaffected in control and TO animals. Sertoli and Leydig cell appearance, testosterone, luteinizing hormone, and follicle-stimulating hormone levels, and epididymal and seminal vesicle weight were unchanged by HLS. Cortisone was not elevated by HLS; thus stress may not be a factor. These results demonstrate that spermatogenesis is severely inhibited by long-term HLS, whereas testicular androgen production is not. These results have significant implications regarding serious effects of long-term exposure to microG on the reproductive capability of scrotal mammals, including humans.  相似文献
