Immature male chickens were treated with testosterone (1 mg/day), Δ4-androstenedione (1 mg/day), 5α-dihydrotestosterone (5α-DHT; 1 mg/day), 5α-androstanedion (1 mg/day), or estradiol (100 μg/day) in order to assess the effects of these steroids on copulatory behavior, agonistic behavior, and attentional processes. Testosterone, estradiol, and 5α-DHT were most effective in stimulating male copulatory behavior above that of oil-treated controls; whereas Δ4-androstenedione and 5α-androstanedione had less, but nevertheless significant, effects on this behavior. Testosterone and 5α-DHT facilitated agonistic behavior; however, estradiol, 5α-androstanedione, and Δ4-androstenedione were ineffective in this capacity. The persistence of response to a given stimulus type was increased by testosterone and decreased by 5α-DHT: 5α-Androstanedione had no discernible effect on this behavior. These findings suggest that in the male chicken the neural structures regulating male copulatory and aggressive behavior as well as attentional processes are differentially sensitive to sex steroids. The effects of all these steroids on somatic structures were assessed.  相似文献

The mechanism of action of androgenic steroids on erythropoiesis is not well understood. In order to assess whether the site of action of androgens is on the early erythroid committed stem cell compartment, the invitro effects of testosterone (T), 5α-dihydrotestosterone (5α-DHT) and 5β-dihydrotestosterone (5β-DHT) on the so-called erythropoietic burst forming unit (BFU-E) in normal rabbit bone marrows were studied. Even though all of the steroids studied increased the number of BFU-E in the presece of Ep, 5β-DHT was the most potent in stimulating BFU-E. Testosterone was moderately effective in increasing BFU-E. Even though 5α-DHT produced a significant increase in BFU-E, it was the least effective of the 3 steroids studied. Preincubation (2 hrs) of normal rabbit bone marrow cells with testosterone followed by removal of T from the culture system resulted in a significant increase in BFU-E when compared with that of non-treated marrow cells in the presence of Ep. These data suggest that testosterone and 5β-DHT and possibly 5α-DHT act on an early uncommitted stem cell, perhaps the CFU-S, to increase the numbers of erythroid committed stem cells to eventually cause an increase in erythropoiesis in combination with Ep.  相似文献

Recently, the development of selective androgen receptor modulators (SARMs) has been suggested as a means of combating the deleterious catabolic effects of hypogonadism, especially in skeletal muscle and bone, without inducing the undesirable androgenic effects (e.g., prostate enlargement and polycythemia) associated with testosterone administration. 17β-Hydroxyestra-4,9,11-trien-3-one (trenbolone; 17β-TBOH), a synthetic analog of testosterone, may be capable of inducing SARM-like effects as it binds to androgen receptors (ARs) with approximately three times the affinity of testosterone and has been shown to augment skeletal muscle mass and bone growth and reduce adiposity in a variety of mammalian species. In addition to its direct actions through ARs, 17β-TBOH may also exert anabolic effects by altering the action of endogenous growth factors or inhibiting the action of glucocorticoids. Compared to testosterone, 17β-TBOH appears to induce less growth in androgen-sensitive organs which highly express the 5α reductase enzyme (e.g., prostate tissue and accessory sex organs). The reduced androgenic effects result from the fact that 17β-TBOH is metabolized to less potent androgens in vivo; while testosterone undergoes tissue-specific biotransformation to more potent steroids, dihydrotestosterone and 17β-estradiol, via the 5α-reductase and aromatase enzymes, respectively. Thus the metabolism of 17β-TBOH provides a basis for future research evaluating its safety and efficacy as a means of combating muscle and bone wasting conditions, obesity, and/or androgen insensitivity syndromes in humans, similar to that of other SARMs which are currently in development.  相似文献

Sex steroids act on the developing and adult telencephalon of songbirds to organize and activate the neural circuits required for the learning and production of song. Presumably, the availability of active androgens and estrogens to steroid-sensitive neural circuits controlling song is modulated by the local expression of androgen-metabolizing enzymes. Two enzymes, 5α- and 5β-reductase, are expressed widely in the songbird telencephalon, as they are in the telencephalons of other avian species. These enzymes convert circulating testosterone (T) into the active and inactive metabolites, 5α- and 5β-dihydrotestosterone (DHT), respectively. A third enzyme, aromatase, converts T into estradiol (E2) and is expressed at unusually high levels in several regions of the songbird telencephalon. In many tissues, including the brain, the regulation of expression of one or more of these enzymes can be a critical feature of their ability to control the production of active sex steroids. We have used primary cell cultures to examine factors that might regulate the expression of these enzymes in developing zebra finch telencephalon. Cultures were treated for 0-72 h with sex steroids (T, E2, 5α-DHT, and 5β-DHT) or with dibutyryl cAMP. Afterward, activities of aromatase, 5α-, and 5β-reductase were determined or total RNA was extracted for Northern analysis. Treatments with cAMP increased both aromatase activity and aromatase mRNA levels by 220%. E2 significantly reduced aromatase activity by an average of 65%, whereas 5α- and 5β-DHT had no effect on aromatase activity. Compared to untreated controls, E2 treatment decreased aromatase mRNA levels by 56%. None of these treatments consistently affected either 5α- or 5β-reductase activities. These results suggest that telencephalic E2 may regulate its own synthesis by repression of aromatase expression, whereas factors that upregulate cAMP in the telencephalon can increase the local concentrations of E2. © 1998 John Wiley & Sons, Inc. Female stumptail macaques continue to copulate at moderate to high levels for years after gonadectomy. This study examined the extent to which sexual behavior of ovariectomized stumptail females was maintained by steroids of adrenal origin, and second, considered the possibility that ovarian fragments might have been left in situ following surgery. Daily injections of 0.1 mg of dexamethasone sodium phosphate suppressed serum cortisol, estradiol, and testosterone by at least 85% in three of four ovariectomized females, but dihydrotestosterone was suppressed by only 50 to 70%. The fourth female showed maximal suppression of cortisol but maintained much higher levels of the other steroids, in particular estradiol, and therefore it was strongly suspected that this animal had an ovarian fragment.Within the limits to which sex steroids were depressed with dexamethasone, no correlation was found between steroid levels and sexual performance. Ejaculatory frequencies and measures of attractivity, proceptivity, and receptivity collected during heterosexual pair tests remained unaffected in all four females during 4 weeks of dexamethasone treatment. Thus it was concluded that the maintenance of copulatory activity after ovariectomy in this species was largely due to nonsteroidal mechanisms.  相似文献

The mechanisms regulating sexual behaviours in female vertebrates are still poorly understood, mainly because in most species sexual displays in females are more subtle and less frequent than displays in males. In a sex-role reversed population of a teleost fish, the peacock blenny Salaria pavo, an external fertilizer, females are the courting sex and their sexual displays are conspicuous and unambiguous. We took advantage of this to investigate the role of ovarian-synthesized hormones in the induction of sexual displays in females. In particular, the effects of the sex steroids oestradiol (E2) and testosterone (T) and of the prostaglandin F2α (PGF2α) were tested. Females were ovariectomized and their sexual behaviour tested 7 days (sex steroids and PGF2α) and 14 days (sex steroids) after ovariectomy by presenting females to an established nesting male. Ovariectomy reduced the expression of sexual behaviours, although a significant proportion of females still courted the male 14 days after the ovary removal. Administration of PGF2α to ovariectomized females recovered the frequency of approaches to the male''s nest and of courtship displays towards the nesting male. However, E2 also had a positive effect on sexual behaviour, particularly on the frequency of approaches to the male''s nest. T administration failed to recover sexual behaviours in ovariectomized females. These results suggest that the increase in E2 levels postulated to occur during the breeding season facilitates female mate-searching and assessment behaviours, whereas PGF2α acts as a short-latency endogenous signal informing the brain that oocytes are mature and ready to be spawned. In the light of these results, the classical view for female fishes, that sex steroids maintain sexual behaviour in internal fertilizers and that prostaglandins activate spawning behaviours in external fertilizers, needs to be reviewed.  相似文献

Mongolian gerbils (Meriones unguiculatus) display a territorial scent marking response associated with a ventral sebaceous gland. In males, both the gland and behavior are androgen dependent and hormone control of marking is delimited to the preoptic area. The present study examines the effects of eleven steroids, injected subcutaneously or implanted into the preoptic area, on territorial marking in adult male castrates. Steroid effects on ventral gland function are also described. The neural target cells that mediate marking respond to a narrower range of steroids than do peripheral target cells in the ventral gland. Testosterone appears to be the only endogenous steroid capable of eliciting marking in males. Other steroids which share molecular commonalities with testosterone (a 17β-hydroxyl group and a double bond involving carbon 4) also induce marking behavior. Central receptors may recognize the relevant endogenous steroid, testosterone, by these features.  相似文献

Evidence is accumulating that sex steroids in the eggs, besides affecting progeny phenotype and behavior in the short term, also have enduring effects until adulthood, when they may translate into differences in reproductive strategies and success. Maternal steroids transfer may therefore affect both agonistic behavior and mate choice decisions, either through the promotion of body size and condition or through a priming effect on the neuroendocrine system. However, owing to the prevalence of a short-term perspective, relevance of maternal transfer of sex steroids to sexual selection processes has been seldom studied. Here we investigate the effects of an experimental increase in egg testosterone on male dominance and copulation success in the ring-necked pheasant, Phasianus colchicus, a polygynous galliform with multiple male ornamental traits, in captivity. We found that females from testosterone (T) injected eggs copulated less than control females. Males from T-injected eggs obtained more copulations than control males, specifically with control females. The effect of male ‘ordinary’ and secondary sexual traits on either dominance or copulation frequency did not depend on early exposure to T, nor did T treatment affect male dominance. Present results demonstrate that variation in the early hormonal environment set up by mothers affects sexual behavior of the offspring, which might translate into fitness differences.  相似文献

Morphological and behavioural effects of testosterone (T) and of 5 α-dihydrotestosterone (5α-DHT) injected daily for a 3-week period at dosages of 0.5, 1, 5 and 10 mg for T and 1 and 5 mg for α-DHT were studied in the adult male castrated Japanese quail. Injections of 0.5 or 1.0 mg T produced only slight development of the cloacal gland while the other four treatments stimulated growth which reached maximal or submaximal values. Testosterone injections stimulated sexual activities; some such effects were also observed after treatment with 5α-DHT. Although both steroids elicited crowing, there were qualitative differences between quails given 5α-DHT and those given T and intacts. These differences were not due to the development of the sternotracheal (syringeal) muscles, the weights of which were increased and reached similar values in the 5α-DHT and T (5 and 10 mg) treated males. These results are discussed in the context of our present knowledge of the mechanisms of regulation of reproduction processes by testosterone and its metabolites in birds.  相似文献

Changes in consecutive estimates of milk progesterone concentrations and serum steroid hormone and sex hormone-binding globulin (SHBG) concentrations in the postpartum period were examined in Finnish Ayrshire and Friesian dairy cows which were divided according to feeding into a hay group and a silage group. Milk progesterone concentrations rose above 10 nmol/1, indicating the start of ovarian luteal activity, slightly earlier in the silage group (28.4 ± 8.7 (S.D.) days, n = 19) than in the hay group (33.4 ± 10.3, n = 28) after calving. Likewise, the first normal oestrous cycles began slightly earlier in cows fed with silage. On the other hand, no differences in the beginning of ovarian luteal activity were observed between the breeds. Serum oestradiol-17β, oestrone, testosterone, 5α-dihydrotestosterone (5α-DHT), pregnenolone and progesterone concentrations were fairly unchanged during postpartum anoestrus after uterine involution and before ovarian cyclic activity. After first ovulation, considerable increases in milk and serum progesterone concentrations were observed. The increase was accompanied by elevations in serum pregnenolone and 5α-DHT concentrations. In the late luteal phase, progesterone, 5α-DHT and pregnenolone concentrations rapidly declined, leading to low hormone levels in pro-oestrus. Thereafter, serum pregenolone and 5α-DHT concentrations slightly increased during the follicular phase. On the other hand, oestradiol-17β concentrations were elevated in pro-oestrus and decreased after that, being lowest at met-oestrous. Serum testosterone concentrations appeared to be unchanged during postpartum anoestrus and over the oestrous cycle. Serum SHBG concentrations were unchanged during postpartum anoestrus and over the oestrous cycle, as well as in pregnant animals. The serum SHBG concentrations were about double those found in women with normal menstrual cycles, whereas oestradiol concentrations were much lower. At present, it cannot be explained how the biological effects of oestradiol become evident under such conditions.  相似文献

Genetic male Japanese quail were administered sex hormones or the oil vehicle on Day 10 of incubation and were caponized 3 weeks after hatching. As adults, the capons were injected with testosterone propionate daily for 2 weeks and then were tested for masculine sexual behavior in response to sexually receptive females. Males that had received as little as 2 μg of estradiol-17β in ovo failed to exhibit the head grabbing and mounting typical of the normal masculine sexual response to females. In a second experiment, this demasculinization was produced by prenatal treatment with 2 μg of estradiol-17α, estrone, estriol, or diethylstilbestrol, but not by this quantity of testosterone. These data suggest that an estrogen is the agent of behavioral demasculinization in the normal female, and that endogenous testosterone poses no difficulty for proper sexual development in the normal male.  相似文献

Prehatch treatment of domestic chicks with 17 beta-estradiol dipropionate (EDP) reduces later testosterone-facilitated sexual behavior, crowing, and suppression of peeping. Prehatch treatment with 5 alpha-dihydrotestosterone propionate (DHTP) reduces later testosterone-facilitated aggressive behavior and produces greatly enhanced avoidance that interferes with sexual behavior in some test situations. It also disturbs crowing by removing trilled call elements and increasing the number of elements/crow but has little effect on crowing posture. The majority of these results can be understood as "demasculinizing" effects of gonadal steroids on the developing male fetus, but the possibility that some result from more extreme pathological changes is also discussed. Posthatch treatment with DHTP facilitates attack and cackling but has no effect on sexual behavior or crowing; the synthetic androgen R1881 has similar effects on cackling and also has no effect on sexual behavior or crowing. Posthatch treatment with EDP enhances sexual behavior but has no effect on attack; the aromatase inhibitor 1,4,6-Androstatrien-3,17-dione (ATD) blocked the suppression of peeping by testosterone but had no effect on testosterone-facilitated sexual behavior. Combined posthatch treatment with EDP and DHTP facilitated crowing although treatment with either steroid alone was ineffective.  相似文献

Experiments in which avian embryos are treated with sex steroidsor steroid antagonists suggest that sexual differentiation ofreproductive behavior (and thus differentiation of the brainmechanisms for such behavior) is controlled by steroids producedby the embryonic gonads. In chickens and Japanese quail, maleshatched from eggs treated with estradiol or testosterone duringincubation are feminized (demasculinized); they fail to exhibitmasculine sexual behavior as adults, and no longer are behaviorallydistinguishable from females. Some evidence suggests that testosteronemay mimic the feminizing action of estradiol by being convertedto an estrogen in the embryonic brain. Genetic female quailexposed to an antiestrogen during embryonic development aremasculinized; they exhibit an increased ability to display themasculine copulatory pattern. Thus the behavior of these speciesis feminized by embryonic exposure to sex steroids, the anhormonal(neutral) sex for behavioral differentiation appears to be themale, and females appear to result from estrogen produced bythe embryonic ovaries. In contrast, sex steroid treatment ofmammals early in development masculinizes behavior, the femaleis the neutral sex, and males result from fetal androgen secretion.These opposite patterns of psychosexual differentiation in birdsand mammals are correlated with a major difference between theavian and mammalian sex-determining mechanism. Daily administration over a period of 6 weeks of increasing doses of dexamethasone sodium phosphate (DEX) to seven long-term ovariectomized female stumptail monkeys significantly lowered circulating levels of testosterone without reducing any aspect of the females' sexual behavior or that of their male partners. Since treatment with DEX failed to suppress serum testosterone levels completely an additional experiment was performed in which the sexual behavior of five ovariectomized stumptails was compared before and after bilateral adrenalectomy, combined with chronic administration of both gluco- and mineralocorticoids. Serum levels of both testosterone and estradiol were reduced to very low levels in females after ovariectomy and adrenalectomy, yet no significant depression of females' sexual performance or that of their male partners occurred. Subsequent sc administration of estradiol or estradiol + testosterone in Silastic capsules to ovariectomized, adrenalectomized stumptails had little effect on sexual interaction. In a third experiment five ovariectomized stumptails which initially were relatively unreceptive and unattractive to males were given first testosterone and then testosterone + estradiol sc in Silastic capsules. One of the three indexes of females' receptivity increased significantly after testosterone; however, no other essential aspect of sexual interaction was affected. These findings suggest that sex steroids are normally not required in the female stumptail macaque for activation of preceptive and receptive sexual behaviors or for maintenance of sexual attractivity.  相似文献

Testosterone and other anabolic-androgenic steroids enhance athletic performance in men and women. As a result, exogenous androgen is banned from most competitive sports. However, due to variability in endogenous secretion, and similarities with exogenous testosterone, it has been challenging to establish allowable limits for testosterone in competition. Endogenous androgen production is dynamically regulated by both exercise and winning in competition. Furthermore, testosterone may promote athletic performance, not only through its long-term anabolic actions, but also through rapid effects on behavior. In women, excess production of endogenous testosterone due to inborn disorders of sexual development (DSD) may convey a competitive advantage. For many years, female competitors have been subject to tests of sexual genotype and phenotype known as gender verification. Although gender verification has not identified any normal man competing as a woman, this process has identified women athletes with DSD. As understanding of DSD has expanded in recent years, women with DSD are increasingly able to continue athletic competition.  相似文献

Castrated male Japanese quail were implanted with Silastic capsules containing testosterone (T), estradiol-17β (E2), 5β-dihydrotestosterone (5β-DHT), Δ4-androstenedione (Δ4) 5α-androstanedione (A), 5α-dihydrotestosterone (5α-DHT) or with empty capsules. Calling, monitored continuously and automatically, was induced significantly by T and Δ4. Locomotor activity, also monitored continuously by floor deflection, was enhanced by T, Δ4, and E2. Additional data concerning heterosexual and homosexual behavior were obtained from castrated quails after implantation of T, Δ4, E2, or 5α-DHT. T and Δ4 restored hetero- and homosexual behavior as did E2 but to a lesser extent. 5α-DHT did not induce either sexual behavior. Growth of the cloacal protrusion was induced in birds implanted with T, Δ4, A, and 5α-DHT but not with 5β-DHT and E2. These results indicate that calling and locomotor activity enhancement (including sexual behavior) are two different components of reproductive behavior which require different androgens or their metabolites to be activated.  相似文献

Precocious adult sexual behavior was determined for chicks from two lines of chickens selected bidirectionally for adult mating frequency. Three-day-old chicks were injected with either 0 or 12.5 mg testosterone cypionate (TC) in sesame oil, and their sexual responses to a hand thrust test (HTT) measured at 10, 12, 14, 16 and 18 days of age. TC stimulated copulation in both the high and low mating line, while birds receiving injections of the vehicle only, failed to copulate. There were differences in precocious sexual behavior between lines at this young age, although further differential maturation of neural tissues mediating mating behavior may occur.Courts, mounts, treads and matings were determined for these same birds at 7 weeks of age, following treatment with 25 mg TC. Differences were found between the lines, with high mating line birds scoring higher than low mating line birds for all behaviors measured. Early treatment with TC, however, had no direct effect on sexual behaviors at this age. Correlations between 7-week behaviors and HTT scores were determined and their possible meanings discussed.  相似文献

Castration significantly lowers serum testosterone in sexually active male Triturus cristatus. Replacement therapy by implants of testosterone in silastic capsules elevates the serum testosterone level to higher values than normal. Sex behavior is depressed by castration and partially reinstated by replacement therapy with testosterone. 5α-dihydrotestosterone was the only testosterone metabolite showing some behavioral effectiveness in castrates; estradiol and 5β-dihydrotestosterone failed to elicit sex behavior.  相似文献

It was reported recently that vertebrate-type steroids exist and control reproduction in several groups of invertebrates, including molluscs. Sexually reproductive freshwater planarians of the species Bdellocephala brunnea have a limited breeding season in their natural habitat. This phenomenon suggests that some endogenous reproductive hormones might play a role in vivo. However, to date, sex steroids such as androgen, estrogen, and progesterone have not been found in planarians. The goal of the present study was to determine whether androgen is present in sexual planarians such as B. brunnea. The presence of testosterone was detected by high-pressure liquid chromatography and, in sexually reproductive individuals in which no seminal vesicles were visible, the level of testosterone was about twice than that in individuals with visible seminal vesicles. An enzyme-linked immunosorbent assay revealed that the levels of testosterone during terminal spermatogenesis were three times higher than during the spermatocyte-building phase. Our results indicate that sexually reproductive freshwater planarians such as B. brunnea might have vertebrate-type steroids and show variation in testosterone levels during spermatogenesis.  相似文献

The profiles of sex steroids (estradiol-17β, testosterone and 11-ketotestosterone) and the mRNA levels of gonadotropins (luteinizing hormone and follicle-stimulating hormone) were investigated before and after downstream migration in females of the Japanese eel species Anguilla japonica, which were collected in the brackish Hamana Lake and its inlet freshwater rivers. Eels were separated into three groups using otolith microchemistry: 'migrants' that grew in the inlet rivers and then made a downstream migration to Hamana Lake mainly in October and November; 'non-migrant' yellow eels caught in rivers during the same season; and 'residents,' which were yellow eels caught in rivers in August. Sex steroid levels, especially those of testosterone and 11-ketotestosterone, were higher in migrants than in non-migrants and residents. Real-time quantitative PCR analysis indicated that mRNA levels of luteinizing hormone (LH) β-subunits were significantly higher in migrants than in other groups, whereas those of follicle-stimulating hormone β-subunits did not show significant changes during downstream migration. The high levels of these hormones during downstream migration raise the question about if they also play a role in motivating the migratory behavior of eels.  相似文献
