In male rats treated with pargyline, reserpine or pyridostigmine during neonatal life significant permanent changes of sexual behaviour and conditioned learning behaviour were observed in juvenile and/or adult life. Male sexual activity and learning capacity were permanently decreased in neonatally pargyline- or reserpine-treated animals, but permanently increased in neonatally pyridostigmine-treated rats. These findings suggest that nonphysiological concentrations and/or turnover rates of neurotransmitters, if produced during a critical period of brain differentiation, are able to induce lifelond effective behavioural changes, i.e. teratopsychogenetic effects.  相似文献

The aim of this study was to assess, whether a sublethal dose of gamma-rays will influence the avoidance behaviour and anxiety in rats and whether the response to radiation depends on time of day of its application. Adult male Wistar rats were tested in elevated plus-maze, in hot plate test and in the light/dark box in 4 regular intervals during a day. After two weeks the animals were irradiated with a whole-body dose 6 Gy of gamma-rays. One day after irradiation the animals were repeatedly tested in the same way, as before irradiation. In the plus-maze test an increased level of anxiety was established. The irradiation significantly decreased the locomotor activity of rats, but the extent of exploratory and comfortable behaviour were not altered. After irradiation, an elevated aversion to the thermal stimulus was observed in the hot plate test. The effects of radiation were more pronounced in the light period of the day, than in the dark one. No significant differences in aversion to light were detected after irradiation. The obtained results indicate, that sublethal doses of ionizing radiation can markedly influence the reactivity of animals to adverse stimuli, their motoric activity and emotional status, as well.  相似文献

Wistar female rats were subjected to a 3.5-day water deprivation once a week in the period of 1.5 to 3 months of age. Their progeny was subjected to the same influences during the same period of life. A week later, reproductive function of female rats was evaluated by mating them with normal males. In the experimental groups of both generations, no significant changes were found in number of neonates and their body mass, in maternal behaviour during the lactation period, in postnatal mortality of pups, in their growth and development, in motor activity, physical endurance and behaviour.  相似文献

Chronic administration of cycloheximide to young rats from 15-th to 30-th postnatal day before daily 3-hour seances of their contacts, disturbed the formation of animals intraspecies aggressivity. In adults it was manifested by hyperaggressivity and by changes of proportions of different reactions in the integral act of aggressive behaviour. By their character these disturbances were close to those observed in the young rats completely deprived of social contacts in this period of ontogenesis.  相似文献

In this account we report in vivo effects of a plant lipid preparation (MMPL) on brain cholesterol and the activity and learning performance of aging male rats. Three-month-old rats were fed for 3 months with a diet that was enriched with 3% MMPL. Another group of 18 month-old rats was fed for 6 months with a 3% MMPL-enriched diet. This food regime lowered markedly the cholesterol level in the hippocampal and cortical regions and increased their lipid membrane fluidity. The animals of both age groups also responded to MMPL with a higher activity and their learning performances, compared to normal diet-fed animals, improved notably. This improvement continued at least 4 months after terminating the supply of MMPL. Significant inverse correlationships were obtained between the length of the training period required to attain proper criteria and cholesterol levels of the hippocampal and cortical brain fractions.  相似文献

The stem barks and the leaves from Bridelia ferruginea (BF, Euphorbiaceae), a medicinal plant, are used in traditional medicine for the treatment of several ailments including male impotency. The present study was aimed at investigating the effects of the dried stem bark of BF on the sexual behaviour of normal and castrated sexually inexperienced male Wistar rats. Animals were orally administered with 100 mg/kg of either the aqueous or the ethanol extracts of BF whilst the neutral control group received in the same way 10 ml/kg of distilled water. The positive control group was treated with a subcutaneous injection of testosterone propionate (20 mg/kg/day/3days) prior to the experiment. The sexual behaviour of all rats was monitored on days 0, 1 and 7 by measuring frequencies of penile erection, mount, intromission and ejaculation. In a separate group of normal sexually inexperienced rats, the pro-sexual effects induced by a single dose of the aqueous extract (100 mg/kg) were measured after pre-treatment with either haloperidol (10 mg/kg), atropine (10 mg/kg) or L-omega-nitro-arginine methyl ester (Lω-NAME, 10 mg/kg). Results obtained showed a significant influence of the duration of treatment on the frequencies of penile erection, mount and intromission of both normal and castrated rats. An increase in all sexual performance parameters was observed when compared to respective controls. The intromission frequency of normal animals was significantly increased (P < 0.05–0.01) on day 1 of treatment and the effect was more expressed in rats receiving the aqueous extract. In castrated animals, a statistical increase was noticed on day 7 for rats treated with testosterone. The sexual effects induced by the aqueous extract of BF were completely abolished after pre-treatment of rats with atropine or haloperidol while pre-administration of Lω-NAME did not produce any significant effects. Flavonoids and sterols revealed in the aqueous and ethanolic extracts from the barks of BF may account for the enhancement of sexual activity in naive rats which could be expressed through dopaminergic and/or cholinergic receptor(s). Results of this work also give value to the traditional use of the plant for the improvement of male sexual behaviour.  相似文献

The single housing of laboratory rats may be recommended in some situations such as hypothesis-driven or test-specific studies, during electroencephalogram recording of phases of sleep and after surgical procedures. However, as single housing of laboratory rats has been shown to be stressful, modification of the housing environment is needed to improve the welfare of these animals. This experiment was carried out to investigate the long-term effects of environmental enrichment on some behavioural, physiological, pathological and psychological measures of welfare. With two batches of animals, 24 rats were housed singly in either enriched cages (EC) (n = 12 cages) or unenriched cages (UC) (n = 12 cages). Behaviour was sampled every week and so was body weight and weight gain over a six-week observation period. Behaviours of the rats in the elevated plus-maze were recorded on the seventh week, whereas organ weights were recorded postmortem. The results revealed that long-term single housing of rats in super-enriched cages increased levels of indicators of good welfare including sleep, exploration, movement and feeding behaviour, body weights, weight gains and the relative weights of the thymus gland and spleen, and decreased levels of indicators of poor welfare such as stationary behaviour and the relative weight of adrenal glands. Thus, enrichment of conventional cages of newly weaned singly-housed laboratory rats with multiple physical structures appeared to improve their ability to control the environment and to promote their species-specific behaviour; changes that can ultimately result in good welfare.  相似文献

To explore the role of the glutathione oxidation-reduction cycle in altering the sensitivity of rats to the effects of hyperbaric hyperoxia, we administered N,N-bis(2-chloroethyl)-N-nitrosourea (BCNU) to decrease tissue glutathione reductase activity. We then exposed these animals and their matched vehicle-treated controls to 100% O2 at 4 ATA. Animals that received BCNU and were immediately exposed to hyperbaric O2 showed enhanced toxicity by seizing earlier in the exposure than controls. Animals that received BCNU 18 h before the hyperbaric O2 exposure were paradoxically protected from the effects of the exposure with a prolongation of their time to initial seizure and a marked increase in their survival time during the exposure. Tissue glutathione concentrations were also measured in the various groups and the hyperbaric O2 exposure produced marked decreases in hepatic glutathione levels in all control animals. In animals treated with BCNU 18 h before exposure, hepatic glutathione concentrations also decreased, but the concentrations had significantly increased during the 18-h waiting period, allowing these animals to maintain hepatic levels in the normal range even during their hyperbaric exposures. We conclude that treatment of rats with BCNU 18 h before exposure to hyperbaric hyperoxia results in enhanced protection of the animals during the exposure.  相似文献

Pronouncement of stress-induced disturbance of searching behaviour (using "open field" test) and stomach ulceration were compared for the first time with activity of the catecholamine system in hypothalamus and striatum and also with activity of the stress-limiting system of nitric oxide (NO) in the rats of two strains August and Wistar, which differ in their resistance against stress-induced cardiovascular disorders. The effect of prior adaptation to hypobaric hypoxia on these disorders was also studied. August rats appeared to be more resistant than Wistar rats against stress-induced disturbance of the searching behaviour and stomach ulceration. Results of measuring the content of catecholamines in brain structures and the content of NO stable metabolites nitrate/nitrite in plasma suggested that these differences could be due to the stress activation of the nigro-striatal dopaminergic system in August rats, which was not observed in Wistar rats, and also to the higher production of NO in August than in Wistar rats. Adaptation to hypoxia considerably restricted these stress disorders in rats of both strains. Importantly, the protective effects were associated with activation of the nigro-striatal dopaminergic system in all the animals. In the result, adapted Wistar rats, as distinct from non-adapted Wistar rats, displayed a stress activation of this system. The protective effects of adaptation were also accompanied by an increased NO synthesis. Taken together, the data suggest an important role of the responsiveness of the brain dopaminergic system and NO system in the mechanism of resistance against stress-induced disturbances.  相似文献

In operant conditioning experiments, two methods are commonly used to motivate laboratory rats to perform designated tasks. The first is restricting food so that rats are forced to lose 20% of body weight within one week, followed by maintenance at 80% of the baseline weight for the remainder of the experiment. The second is restricting access to water to 15 min in each 24 h period. These methods are effective in motivating the animals. There is, however, little information available on the effects on performance in tests of behaviour that are not related to operant conditioning. In addition, it is not clear if these commonly used methods of food and water restriction will lead to physiological stress as indicated by an elevation of serum corticosterone. Male rats were either food-restricted to reduce and maintain their weight at 80% of baseline weight, or were restricted to 15 min access to water every 24 h. Activity in the open field was significantly greater in food-restricted rats than in water-restricted or control rats, but freezing behaviour was similar in all experimental groups. Food-restricted rats had a higher mean serum corticosterone level than water-restricted and control rats 37 days after the start of the experimental period. These data suggested that chronically restricting food and maintenance of body weight at 80% of baseline body weight led to significant behavioural changes and physiological stress. In contrast, water restriction did not lead to changes in behaviour or corticosterone levels. A second experiment was conducted to compare the effects of food restriction to 80% of baseline body weight, as described above, with a less stringent protocol in which test rats were initially reduced to 80% of baseline weight, but were then maintained at 80% of an ad libitum fed control rat's weight. Serum corticosterone levels and adrenal gland weights were measured after the initial week of forced weight loss and after maintenance for 21 days. Forced loss of 20% of body weight in the first week led to significantly increased serum corticosterone levels and adrenal gland weights compared to ad libitum fed controls. Serum corticosterone levels and adrenal gland weights in rats maintained at 80% of their initial body weight for 21 days remained higher than ad libitum fed control rats. However, rats maintained at 80% of an ad libitum fed control rat's weight did not differ from control rats in serum corticosterone levels or adrenal gland weights at the end of the 21-day study period. Adjustment of the feeding regimen in this manner eliminated physiological evidence of chronic stress.  相似文献

A report account of the effects of cerebral circulatory drugs viz., nicotinic acid, Cyclospasmol, Ronicol on electrocortical activity and automatic movement behaviour of rats, has been presented. Observations ware made of the tats kept in a sound proof, well aerated (ventilated) dimly lighted chamber with one way glass. Electrocortical activity was recorded from the unanaesthetised, unrestrained rats chronically implanted with electrodes. Results obtained showed that the administration of nicotinic acid resulted in sleep-like condition lasting for about one and a half hours, with concomittant changes in electrical activity and behaviour. Cyclospasmol and Ronicol did not produce any change in the electrical activity and behaviour of the animals. Cyclospasmol, however, prevented the effects of nicotinic acid. Ronicol appeared to be somewhat less effective in this respect. Autocorrelation and Fowler analysis of electrocortical activity were also performed. Actions of the drugs used have been discussed and explained with respect to their known effects on cerebral blood flow.  相似文献

Considering the frequent use of mobile phones, we have directed attention to possible implications on cognitive functions. In this study we investigated in a rat model the long-term effects of protracted exposure to Global System for Mobile Communication-900 MHz (GSM-900) radiation. Out of a total of 56 rats, 32 were exposed for 2 h each week for 55 weeks to radio-frequency electromagnetic radiation at different SAR levels (0.6 and 60 mW/kg at the initiation of the experimental period) emitted by a (GSM-900) test phone. Sixteen animals were sham exposed and eight animals were cage controls, which never left the animal house. After this protracted exposure, GSM-900 exposed rats were compared to sham exposed controls. Effects on exploratory behaviour were evaluated in the open-field test, in which no difference was seen. Effects on cognitive functions were evaluated in the episodic-like memory test. In our study, GSM exposed rats had impaired memory for objects and their temporal order of presentation, compared to sham exposed controls (P = 0.02). Detecting the place in which an object was presented was not affected by GSM exposure. Our results suggest significantly reduced memory functions in rats after GSM microwave exposure (P = 0.02).  相似文献

Laboratory rabbits maintained under controlled lighting and fed ad libitum exhibit a weak but consistent diurnal fluctuation in feed intake. There are two major periods of eating; at the beginning and the end of the light period. This results in similar feed consumption for the light and dark periods. Water intake shows a similar diurnal variation to that of feed intake. If the normal lighting cycle is retarded by 6 hours, the animals adjust their diurnal rhythm of feeding behaviour to the new lighting cycle within 8 to 15 days. The behaviour of the granulation of the myo-epithelioid cells of the juxtaglomerular apparatus of the kidney and its relationship with the plasma concentrations of renin, sodium and potassium, and plasma osmolality have been studied in male and female Wistar rats that have been subjected to a programme of moderate physical exercise throughout their period of growth. In the trained rats a significant increase in the juxtaglomerular granulation index and a significant decrease in plasma renin concentration has been observed, the changes being more prominent in the female animals. Moreover in the trained rats significant increases in plasma sodium, potassium and osmolality have been observed.  相似文献

Summary Macrophages of the adrenal cortex were studied in normal male and female, ovariectomized and estradiol-injected rats. In normal male rats few macrophages with numerous granules were observed in the zona fasciculatazona reticularis border, and in the zona reticularis. Granules, identified as lysosomes, were limited by a single membrane with a heterogeneous matrix; they exhibited acid phosphatase- and aminotriazole-resistant peroxidatic activities. A larger number of macrophages had identical distributions in normal female rats. In ovariectomized and estradiol-injected rats the number and distribution of adrenal macrophages were similar to those in normal females; however, in spayed animals the number of these cells in the zona reticularis was higher than in the other experimental groups. Lysosomes in macrophages of treated animals were more numerous and their contents more complex than in normal male animals. These results indicate that the adrenal macrophage system is stimulated in experimental conditions involving high levels of circulating estrogens.  相似文献

ABSTRACT: BACKGROUND: It is well known that axotomy in the neonatal period causes massive loss of motoneurons, which is reflected in the reduction of the number of motor units and the alteration in muscle properties. This type of neuronal death is attributed to the excessive activation of the ionotropic glutamate receptors (glutamate excitotoxicity). In the present study we investigated the effect of the NMDA antagonist DAP5 [D-2-amino-5-phosphonopentanoic acid] in systemic administration, on muscle properties and on behavioural aspects following peripheral nerve injury. METHODS: Wistar rats were subjected to sciatic nerve crush on the second postnatal day. Four experimental groups were included in this study: a) controls (normal saline injection), b) crush c) DAP5 treated and d) crush and DAP5 treated. Animals were examined with isometric tension recordings of the fast extensor digitorum longus and the slow soleus muscles, as well as with locomotor tests at four time points, at P14, P21, P28 and adulthood (2 months). RESULTS: 1. Administration of DAP5 alone did not provoke any side-effects. 2. In all age groups, animals with crush developed significantly less tension than the controls in both muscles and had a worse performance in locomotor tests (p<0.01). Crush animals injected with DAP5 were definitely improved as their tension recordings and their kinetic behaviour were significantly improved compared to axotomized ones (p<0.01). 3. The time course of soleus contraction was not altered by axotomy and the muscle remained slow-contracting in all developmental stages in all experimental groups. EDL, on the other hand, became slower after the crush (p<0.05). DAP5 administration restored the contraction velocity, even up to the level of control animals 4. Following crush, EDL becomes fatigue resistant after P21 (p<0.01). Soleus, on the other hand, becomes less fatigue resistant. DAP5 restored the profile in both muscles. CONCLUSIONS: Our results confirm that contractile properties and kinetic behaviour of animals are severely affected by axotomy, with a differential impact on fast contracting muscles. Administration of DAP5 reverses these devastating effects, without any observable side-effects. This agent could possibly show a therapeutic potential in other models of excitotoxic injury as well.  相似文献

This study examined patterns of consummatory behavior in normal and vasopressin-deficient rats under ad lib and food-restricted conditions. Differing patterns of intake of food, water, sucrose, and saccharin were found in the two groups. During the ad lib period, the normal rats gained more weight and ate significantly more food than vasopressin-deficient rats. The vasopressin-deficient animals consumed significantly more sucrose than normal animals during this period. No significant differences in body weight, food intake, sucrose intake, or saccharin intake were noted during the food-restricted period between the two groups. During the food-restricted condition the normal animals showed a cyclic pattern of food, sucrose, and saccharin intake. This cyclic pattern of intake was absent in vasopressin-deficient animals. A vasopressin-serotonin interaction is hypothesized as a possible mechanism producing the differences in the two strains of animals. In addition, the data support the position that food consumption is regulated primarily by caloric and/or nutritional factors.  相似文献

Effects of the intraperitoneal injection of quercetin in streptozocin-induced diabetic and normal rats were investigated and compared. Although quercetin had no effect on plasma glucose level of normal animals, it significantly and dose-dependently decreased the plasma glucose level of streptozocin-induced diabetic rats. Glucose tolerance tests of the diabetic animals approached those of normal rats, their plasma cholesterol and triglycerides were reduced significantly, while their hepatic glucokinase activity was significantly increased upon quercetin treatment. In normal rats, quercetin did not affect the glucose tolerance test, but resulted in an increase of plasma cholesterol and triglycerides and a decrease in hepatic glucokinase activity. No significant pathologic changes were noted in hepatocytes or kidney tubules and glomeruli, while the number of pancreatic islets significantly increased in both treated normal and diabetic groups. It is concluded that quercetin, a flavonoid with antioxidant properties brings about the regeneration of the pancreatic islets and probably increases insulin release in streptozocin-induced diabetic rats; thus exerting its beneficial antidiabetic effects. However, it may be of little value in normoglycemic animals.  相似文献

In this paper we describe the effects of manipulating two kinds of sensory input in neonatal rats upon the development of the macrovibrissae--that movable subset of the rodent mystacial vibrissae. In an initial study of normal whisker development, data on whisker size were obtained from neonatal, perinatal, and adult rats. Data on whisker size were also obtained from rats sustaining either neonatal sensory or motor denervation of the whiskers and from both rats and mice bilaterally enucleated as neonates (BEN). In normally reared rats, most whiskers attain their final size over the first three postnatal weeks but development of rows 6 and 7 are not completed until after the first month. In normal animals we found a significant correlation both between body weight and whisker size and between the size of a whisker and the size of its corresponding cortical barrel. Rats sustaining neonatal denervation of the whiskers have shorter and thinner whiskers as adults than normally reared animals. In both rats and mice bilaterally enucleated as neonates a subset of the macrovibrissae are significantly larger than those of normal controls but no such effect is seen if the enucleation is carried out in adults. Moreover, BEN rats exposed to a novel stimulus environment whisk at a significantly higher frequency than normally reared animals. Mechanisms which might mediate these effects are discussed.  相似文献
