Ethylene formation by loaves of the central stem zone of the short-day tobacco cv. Maryland Mammoth and long-davNicntiana sylvestris was followed for 40 days during in duction and transition to flowering. In SD tobacco Mammoth, ethylene formation rose between days 0-10, remained unchanged for the next 10 davs, rose slightly between days 20 - 30 and sharply within the last 10 days. The time-course of ethylene formation by the leaves of LD tobacco N.silvestris resembled that of Mammoth, but tho changes were less pronounced, especially at the beginning of the period. Generally, ethylene formation is much higher in SD tobacco Mammoth than in LDN. silvestris. Ethephon (0.02 %) application during flower induction significantly reduced flowering in SD tobacco Mammoth (by 47.5 %) and also reduced apical meristem length. In N.silvestris ethephon application did not reduce flowering, but most of the treated plants (62.5 %) did not attain the stage of inflorescence. Apical meristem (or inflorescence) and stem length were also reduced. The possible role of ethylene in regulation of transition to flowering is discussed.  相似文献

Changes in free IAA level were studied in the leaves of the central stem zone of short-day tobacco (Mcotianatabacum, cv. Maryland Mammoth) and long-day tobacco(Nicotiana silvestris) in inductive photoperiodic regime after 10, 20, 30 and 40 d, respectively. The leaves of SD tobacco Mammoth showed a high free IAA level in vegetative plants kept under long days but it significantly decreased (by ca. 50 %) after 10, 30 and 40 short days, respectively. After 20 short days the IAA level was as high as in the leaves of plants at the beginning of inductive treatment. The changes of freeIAA level in the leaves of LD tobacco N.silvestris were similar to those of SD Mammoth, but the IAA level in this species was significantly lower than that of Mammoth throughout the investigated period. Consequently, the changes observed in N. silvestris were much less pronounced. Plants of both tobacco species were fully induced to flowering by 30 inductive days and this was associated with differentiation of the flower organs. Application of 10 -4 M IAA during the last 10 d of the inductive treatment of 30 d significantly reduced flowering in SD tobacco Mammoth without changing the stem length and apex width. Apex length was slightly reduced. IAA application elicited almost no effect inN. silvestris. The results are discussed with respect to the possible role of IAA in flower induction in SD and LD plants.  相似文献

We analyze geographic variation in morphology for Sierra Nevadan populations of Brown-headed Cowbirds (Molothrus ater) in relation to two levels of geographic structure of populations: 1) subspecies and 2) vocal dialects. Two morphologically distinct subspecies occupy opposite slopes of the Sierra Nevada: M. a artemisiae on the east slope is larger than M. a. obscurus on the west slope, and its juveniles have predominantly white versus yellow rictal (bill) flanges. Populations of obscurus moved into California from the lower Colorado River around 1900 and invaded the western Sierra during the 1930's. A relatively constant type of flight whistle occurs in obscurus populations up to 700 km apart, but east-slope artemisiae are divided into many distinct dialects. The means of seven morphological measurements and of principal component 1 (i.e., body size) for 2,287 individuals show similar clinal patterns for adult males, yearling males, and females over a 60-km north–south transect along the eastern Sierra Nevada: large sizes occur at the southernmost study site at Bishop, decrease clinally to the north to Mammoth Lakes, and then increase clinally to the north from Mammoth Lakes to the northernmost site at Lee Vining. This reversal in cline slope is reiterated for variation in the frequency of white-flanged juveniles. Our data do not support adaptive explanations for the clines. Rather, the most parsimonious explanation is that there has been flow of obscurus genes into artemisiae from the west slope across the crest of the Sierra Nevada into the Mammoth Lakes area. Concordantly, the Sierran crest at Mammoth Lakes is considerably lower than any other point within about 80 km to the north or 180 km to the south. Two other findings strongly support this interpretation. First, wing lengths of our longest-winged samples (at Bishop and Lee Vining) are significantly shorter than those of cowbirds collected in the same region between 1912 and 1922. Second, the wing lengths of Mammoth Lakes adult males and females during 1978–1981 are significantly larger than those of the 1982–1985 period. This putative hybridization must have been rapid, as there have been fewer than 50 years since secondary contact could have occurred. Calculation of the gene-flow parameter Ι of Endler (1977) indicates that the gene-flow rate reported here is higher than for nearly all of the species he surveyed. This gene flow occurs between two subspecies with different flight whistles and across several cowbird flight-whistle dialects within artemisiae. Therefore, vocal differences among these dialects do not appear to be a strong deterrent to gene flow.  相似文献

Epigravettian dwellings from Eastern Europe give the Mammoth a privileged place: in the spectacular and elaborated architecture of their huts, like in the leading site Mezhyrich. However, the range of hunted and/or eaten game shows a variety of species including small ones such as the hare, testifying economical choices or even cultural practices where the Mammoth is not dominant. Magdalenian cave art, in Western Europe, gives the Mammoth a significant, sometimes exceptional place, like in Rouffignac or Combarelles, next to the even more important place of the Bison or the Horse. However, the dietary habits of the artists prove the prominent value of the Reindeer compared with the Horse. In Western Europe, wide and different landscapes and environments produced an economical-cultural mosaic. In the eastern part of Europe, the strong uniformity of climates and environments promoted an economical and environmental block leading to a sedentary lifestyle of the people, demonstrated by the concentration of huts. In the dreams of the hunters, as well as in their daily life, the incredible complexity of the relationship between men with animals revealed its creative and nourishing power.  相似文献

Agrobacterium tumefaciens binary vectors carrying kanamycin resistance gene and either C58 T-DNA gene 4 for cytokinin synthesis or genes 1 and 2 for auxin synthesis were constructed and used for transformation of a short-day tobacco Maryland Mammoth. Kanamycin resistant plants were regenerated from a small fraction of transformed tissue and the presence of T-DNA in their genome was verified by Southern blotting. The level of endogenous cytokinin in plants transgenic for gene 4 and the level of endogenous IAA in those transgenic for genes 1 and 2 increased by more than 100 %. A number of morphological characteristics distinguish them from untransformed controls.  相似文献

The stability of the florally determined state in terminal and axillary buds of two tobacco cultivars was studied. We used Hicks and Hicks Maryland Mammoth, near-isogenic cultivars of Nicotiana tabacum differing at the recessive maryland mammoth locus which confers short-day behavior. The experimental design consisted of growing plants in short-day conditions and subjecting them to three bioassays in long-day conditions: in vitro culture of apices consisting of meristems and three to four leaf primordia; rooting of buds consisting of meristems and 8 to 12 leaves, leaf primordia, and internodes; and release from apical dominance of axillary buds in situ. Cultured terminal and axillary apices expressed floral determination, indicating that meristems can be florally determined. Two lines of evidence indicate that rooting destabilizes an already acquired florally determined state: cultured apices from both axillary and terminal buds produced fewer nodes after excision than homologous buds which were rooted; and a lower percentage of rooted axillary buds from Hicks Maryland Mammoth plants expressed floral determination than did homologous axillary buds grown out in situ in noninductive conditions. Rooted buds from the two genotypes expressed floral determination at different frequencies, but produced abnormal inflorescences at similar frequencies, indicating that roots and the maryland mammoth allele influence common as well as unique processes associated with floral determination.  相似文献

Fluorescent antibodies (FA) prepared for Nitrobacter agilis and N. winogradskyi were highly reactive in homologous staining. Low-level cross-reactions between the two species were removed by adsorption. All 15 pure-culture isolates of Nitrobacter tested reacted strongly with either N. agilis FA or N. winogradskyi FA. All pure-culture isolates from soils were determined to be N. winogradskyi; those from Mammoth Cave sediments and a cattle waste oxidation ditch were N. agilis. No cross-reaction was found in extensive tests that included five isolates of Nitrosomonas europaea and 668 heterotrophic aerobic and anaerobic bacteria isolated from soil, sewage, and cave sites. The FA preparations were used to detect Nitrobacter species in Mammoth Cave sediments, in a cattle waste oxidation ditch, and in surface waters and sediments of a river and to observe that N. winogradskyi can outgrow N. agilis in enrichment culture.  相似文献

Summary The organization and distribution of microfilaments in freze substituted leaf tissues and root tips of tobacco plants (Nicotiana tabacum L. var. Maryland Mammoth) were investigated in detail. Three categories of microfilaments were recognized in interphase cells of all tissues including those in the root cap: (1) microfilament bundles; (2) single microfilaments; (3) cortical microtubuleassociated microfilaments. While the microfilament bundles appeared to be distributed throughout the cytoplasm, the single microfilaments were mainly confined to the cell periphery. All three categories of microfilaments were associated with various organelles. Our study indicates that the three categories of microfilaments are normal cytoskeletal components in higher plant cells. Microbial mats which lack cyanobacteria occur at 50° to 65° C in the sulfide-containing Mammoth Springs of Yellowstone National Park. The principal organisms within these mats are filamentous bacteria which resemble Chloroflexus aurantiacus. The incorporation of [14C]-HCO 3 - into mat material depended upon both light and sulfide, and was not inhibited when complete natural light was replaced with far-red and infra-red radiation. [14C]-acetate was incorporated in a light-dependent reaction which was stimulated by, but did not require, sulfide. In situ experiments with microelectrodes demonstrated net sulfide uptake by the mat in the light, and net sulfide production by the mat in the dark, suggesting the operation of a sulfur cycle.Filamentous phototrophic bacteria isolated from the mat were incapable of sustained growth in the presence of O2.Simultaneous exposure of cultures to light and O2 caused degradation of bacteriochlorophyll c. The stimulation of light-dependent [14C]-HCO 3 - -uptake by sulfide was more pronounced in these isolates than in strains of Chloroflexus aurantiacus.  相似文献

Summary The organization and distribution of microfilaments (MFs) in the preprophase bands (PPBs) of tobacco (Nicotiana tabacum L. var. Maryland Mammoth) root tip cells were studied with high pressure freezing and freeze-substitution methods. MFs were present predominantly as single filaments interspersed among microtubules (MTs) throughout the PPB. Some MFs appeared to be associated with MTs, whereas others were not. This is the first time that MFs have been demonstrated in the PPB at the electron microscope level.  相似文献

The ultrastructure of cells of the vegetative shoot apices is described for Chenopodium album, Kalanchoë blossfeldiana and K. laxiflora, Bryophyllum daigremontianum, Nicotiana rustica, and N. tabacum (Maryland Mammoth), and Ginkgo biloba. A less intensive study was made of the last three listed. The structures and organelles usually associated with meristematic cells were observed: dictyosomes, plastids (in various stages of development), mitochondria, endoplasmic reticulum (ER), vacuoles, lipid droplets, and plasmalemma. In addition, spherosome-like structures were observed in all zones of the shoot apices. Also, multivesicular bodies were observed in C. album and B. daigremontianum. Ribosome density is greater in cells of the flank meristem. Proplastids, plastids with prolamellar bodies, or grana have a differential distribution in the apex, characteristic for a particular species. Confirmation could not be given to the concept that vacuoles arise as a series of local dilations in long extensions of the so called "smooth ER." The tonoplast and ER are distinguishable at the time of inception of a vacuole, although the tonoplast may arise from the ER. Rapid growth of a vacuole and/or fusion with other vacuoles may result in irregularly shaped prevacuoles. No vacuoles were observed to originate from cisternae of dictyosomes in the species studied.  相似文献

Amount and products of photosynthesis during 10 minutes were measured at different 14CO2 concentrations in air. With tobacco (Nicotiana tabacum L. cv. Maryland Mammoth) leaves the percentage of 14C in glycine plus serine was highest (42%) at 0.005% CO2, and decreased with increasing CO2 concentration to 7% of the total at 1% CO2 in air. However, above 0.03% CO2 the total amount of 14C incorporated into the glycine and serine pool was about constant. At 0.005% or 0.03% CO2 the percentage and amount of 14C in sucrose was small but increased greatly at higher CO2 levels as sucrose accumulated as an end product. Relatively similar data were obtained with sugar beet (Beta vulgaris L. cv. US H20) leaves. The results suggest that photorespiration at high CO2 concentration is not inhibited but that CO2 loss from it becomes less significant.  相似文献
