Biomass and eicosapentaenoic acid (EPA) productivities were investigated in a flat panel airlift loop reactor ideally mixed by static mixers. Growth with ammonium, urea and nitrate as nitrogen source were performed at different aeration rates. Cultures grew on ammonium but the decay of pH strongly inhibited biomass increase. On urea biomass productivity reached 2.35 g L–1d–1at an aeration rate of 0.66 vvm (24 h light per day, 1000 mol photon m–2s–1). Aeration rates between 0.33 vvm and 0.66 vvm and maximal productivities on urea were linearly dependent. Productivity on nitrate never exceeded 1.37 g L–1d–1. In the range of maximum productivity photosynthesis efficiency of 10.6% was reached at low irradiance (250 mol photon m–2s–1). Photosynthesis efficiency decreased to 4.8% at 1000 mol photon m–2s–1. At these high irradiances the flat panel airlift reactor showed a 35% higher volume productivity than the bubble column. At continuous culture conditions the influence of CO2concentration in the supply air was tested. Highest productivities were reached at 1.25% (v/v) CO2where the continuous culture yielded 1.04 g L–1d–1(16 h light per day, 1000 mol photon m–2s–1). The average EPA content amounted to 5.0% of cell dry weight, that resulted in EPA productivities of 52 mg L–1d–1(continuous culture, 16 h light per day) or 118 mg L–1d–1(batch culture, 24 h light per day).  相似文献

Laurencia brongniartii is usually found at depths below 4 m, but can be found in shallow subtidal areas in crevices and on the walls of a coral reef in Amami Oshima Island, Kagoshima Prefecture, Japan, where irradiances were significantly lower than those at similar depths in open water. In preparation for the possible cultivation of this species for its antibiotic compounds, the effects of temperature and irradiance on photosynthesis and growth were measured. Photosynthesis and growth rates of L. brongniartii explants were highest at 26 and 28 °C, which closely corresponded to temperatures found during August to late December when it was most abundant. The estimated maximum photosynthesis rate (P max) was 4.41 mol photon m–2 s–1 at 26 °C and 4.07 mol photon m–2 s–1 at 28 °C. Saturating irradiance occurred at 95 mol photon m–2 s–1 at 26 °C and 65 mol photon m–2 s–1 at 28 °C. In contrast, growth experiments at 41.7 mol photon m–2 s–1 caused bleaching of explants and the maximum growth rate observed during the study was 3.02 ± 0.75% day–1 at 28 °C and 25 mol photon m–2 s–1. The difference in the saturating irradiance for photosynthesis and the irradiance that caused bleaching in growth experiments suggests that long-term exposure to high irradiance was detrimental and should be addressed before the initiation of large scale cultivation.  相似文献

The effects of light intensity, oxygen concentration, and pH on the rates of photosynthesis and net excretion by metalimnetic phytoplankton populations of Little Crooked Lake, Indiana, were studied. Photosynthetic rates increased from 1.42 to 3.14 mg C·mg–1 chlorophylla·hour–1 within a range of light intensities from 65 to 150E·m–2·sec–1, whereas net excretion remained constant at 0.05 mg C·mg–1 chlorophylla·hour–1. Bacteria assimilated approximately 50% of the carbon released by the phytoplankton under these conditions. Excreted carbon (organic compounds either assimilated by bacteria or dissolved in the lake water) was produced by phytoplankton at rates of 0.02–0.15 mg C·mg–1 chlorophylla·hour–1. These rates were 6%–13% of the photosynthetic rates of the phytoplankton. Both total excretion of carbon and bacterial assimilation of excreted carbon increased at high light intensities whereas net excretion remained fairly constant. Elevated oxygen concentrations in samples incubated at 150E· m–2·sec–1 decreased rates of both photosynthesis and net excretion. The photosynthetic rate increased from 3.0 to 5.0 mg C·mg–1 chlorophylla· hour–1 as the pH was raised from 7.5 to 8.8. Net excretion within this range decreased slightly. Calculation of total primary production using a numerical model showed that whereas 225.8 g C·m–2 was photosynthetically fixed between 12 May and 24 August 1982, a maximum of about 9.3 g C·m–2 was released extracellularly.  相似文献

This study examines the influence of current velocity in the toxiceffect of copper in diatom-dominated biofilms grown in artificial channels.Effects on community structure, algal biomass and photosynthesis (carbonincorporation) caused by 15 g L–1 of copperwere tested at contrasting (1 and 15 cm s–1)velocities. Moreover, a possible threshold on the effect of copper on algalbiomass and photosynthesis related to current velocity was examined by usingprogressively increasing current velocity (1 to 50 cms–1) at 15 g L–1 Cu.Chlorophyll-a decreased ca. 50% as a result of addition of15 g L–1 Cu. Chlorophyll decrease occurredearlier at 15 cm s–1 than at 1 cms–1 when adding 15 g L–1Cu. Copper also caused a remarkable decrease in carbon incorporation(from 30 to ca. 50%), which was produced earlier at 15 cms–1 (three days) than at 1 cms–1 (seven days). Some taxa were affected by thecombination of copper and current velocity. Both Achnanthesminutissima and Stigeoclonium tenue becomedominant at 15 cm s–1 in the presence of copper.Significant inhibition of algal growth in 15 g L–1Cu occurred at low (1 cm s–1) and highvelocities (50 cm s–1), but not at intermediatevelocity (20 cm s–1). The experiments indicatethat current velocity triggers the effect that copper has on diatom-dominatedbiofilms, and that the effect is more remarkable at low and high than atintermediate current velocities.  相似文献

Summary The kinetics ofBordetella pertussis growth was studied in a glutamate-limited continuous culture. Growth kinetics corresponded to Monod's model. The saturation constant and maximum specific growth rate were estimated as well as the energetic parameters, theoretical yield of cells and maintenance coefficient. Release of pertussis toxin (PT) and lipopolysaccharide (LPS) were growth-associated. In addition, they showed a linear relationship between them. A comparison has been made of cyclopropene (CP), 1-methylcyclopropene (1-MCP), and 3,3-dimethyl-cyclopropene (3,3-DMCP) in their ability to protect plants against ethylene. In bananas, both CP and 1-MCP are effective around 0.5 nL L–1, and 3,3-DMCP was effective at 1 L L–1. Bananas treated with CP and 1-MCP again become sensitive to ethylene at 12 days and those treated with 3,3-DMCP at 7 days. Mature green tomatoes are protected by 5–7 nL L–1 of 1-MPC for 8 days at 25°C and tomatoes treated with 3,3-DMCP at 5–10 L L–1 are protected for 5 days. Carnation flowers are protected with CP or 1-MCP after exposure to 0.5 nL L–1 for 24 hours and by 1 L L–1 of 3,3-DMCP. The display life of Campanula flowers is increased from 3.3 to 5.4 days by 10 L L–1 of 3,3-DMCP and to 9 days by 20 nL L–1 of 1-MCP. Ethylene inhibition of pea seedlings is reduced by treatment with 1-MCP at 10 L L–1 of ethylene but as ethylene is increased to 3000 L L–1 growth inhibition increases. 3,3-DMCP treatment causes very little reduction of the ethylene effect even at very low concentrations.  相似文献

Jasmonic acid altered the accumulation of major anthocyanins in Vitis vinifera cell culture. Peonidin 3-glucoside content at day three was increased from 0.3 to 1.7 mg g–1 dry cell wt while other major anthocyanins were increased by smaller increments. By day 14, the content of methylated and acylated anthocyanins (peonidin 3-p-coumaroylglucoside and malvidin 3-p-coumaroylglucoside) was 6.3 mg g–1 DCW, in response to treatment with jasmonic acid, and comprising 45% (w/w) of total anthocyanins. In comparison, the untreated control culture contained 1.2 mg g–1 DCW which made up 32% (w/w) of total anthocyanins. Light further enhanced anthocyanin accumulation induced by jasmonic acid elicitation. The content of peonidin 3-glucoside at day 3 was 6.6 mg g–1 DCW, 22-fold higher than control cultures while the content in response to light irradiation alone was 0.6 mg g–1 DCW. When a highly pigmented cell line was elicited with jasmonic acid total anthocyanins increased from 9.2 to 20.7 mg g–1 DCW, but there was no change in the anthocyanin composition.  相似文献

Synopsis Arsenic persists in Chautauqua Lake, New York waters 13 years after cessation of herbicide (sodium arsenite) application and continues to cycle within the lake. Arsenic concentrations in lake water ranged from 22.4–114.81 g l–1, = 49.0 ag l–1. Well water samples generally contained less than 10 g l–1 arsenic. Arsenic concentrations in lake water exceeded U.S. Public Health Service recommended maximum concentrations (10 g l–1) and many samples exceeded the maximum permissible limit (50 g l–1). Fish accumulated arsenic from water but did not magnify it. Fish to water arsenic ratios ranged from 0.4–41.6. Black crappie (Pomoxis nigromaculatus) contained the highest arsenic concentrations (0.14–2.04 g g–1 ), X = 0.7 g g–1) while perch (Perca flavescens), muskellunge (Esox masquinongy) and largemouth bass (Micropterus salmoides) contained the lowest concentrations (0.02–0.13 g g–1). Arsenic concentrations in fish do not appear to pose a health hazard for human consumers.  相似文献

The light utilization efficiency and relative photon requirement of photosynthesis in pulsed and continuous light from light emitting diodes (LEDs) has been measured. First, we chacterized the photon requirement of photosynthesis from light of LEDs that differ in spectral quality. A photon requirement of 10.3±0.4 was measured using light from a 658 nm peak wavelength (22 nm half band width) LED over the range of 0–50 mol photons m–2 s–1 in 2 kPa O2 in leaves of tomato (Lycopersicon esculentum Mill., cv. VF36). Because the conversion of electrical power to photons increased with wavelength, LED lamps with peak photon output of 668 nm were most efficient for converting electricity to photosynthetically fixed carbon. The effect of pulsed irradiation on photosynthesis was then measured. When all of the light to make the equivalent of 50 mol photons m–2 s–1 was provided during 1.5 s pulses of 5000 mol photons m–2 s–1 followed by 148.5 s dark periods, photosynthesis was the same as in continuous 50 mol photons m–2 s–1. When the pulse light and dark periods were lengthened to 200 s and 19.8 ms, respectively, photosynthesis was reduced, although the averaged photon flux density was unchanged. Under these conditions, the light pulses delivered 1017 photons m–2, which we calculate to be equivalent to the capacitance of PS I or PS II. Data support the theory that photons in pulses of 100 s or shorter are absorbed and stored in the reaction centers to be used in electron transport during the dark period. When light/dark pulses were lengthened to 2 ms light and 198 ms dark, net photosynthesis was reduced to half of that measured in continuous light. This two-part study compared the efficacy of different plant species to extract Se from soils irrigated with Se-laden effluent. The species used were: Brassica napus L. (canola), Brassica juncea Czern L. and Coss (Indian mustard), and Hordeum vulgare L. (barley). In Study 1 we irrigated the plants with a saline effluent containing 0.150 mg Se L–1, while in Study 2, the same species were planted in a saline soil selenized with 2 mg Se L–1. Plants were simultaneously harvested 120 days after planting. In Study 1, there were only slight effects of treatment on dry matter (DM) yield. Plant Se concentrations averaged 21 g Se g–1DM for the Brassica species, and 4.0 g Se g–1 DM for barley. Total Se added to soils via effluent decreased by 40% for Brassica species and by 20% for barley. In Study 2, total DM decreased for all species grown in saline soils containing Se. Plant Se concentrations averaged 75 g g–1 DM for Brassica species and 12 g Se g–1 DM for barley. Total Se added to soils prior to planting decreased by 40% for Brassica species and up to 12% for barley. In both studies, plant accumulation of Se accounted for at least 50% of the Se removed in soils planted to Brassica and up to 20% in soils planted to barley. Results show that although the tested Brassica species led to a significant reduction in Se added to soil via use of Se-laden effluent, additional plantings are necessary to further decrease Se content in the soil.  相似文献

We determined for Vicia faba L the influence of nitrogen uptake and accumulation on the values of photon saturated net photosynthetic rate (P Nmax), quantum yield efficiency (), intercellular CO2 concentration (C i), and carboxylation efficiency (C e). As leaf nitrogen content (NL) increased, the converged onto a maximum asymptotic value of 0.0664±0.0049 mol(CO2) mol(quantum)–1. Also, as NL increased the C i value fell to an asymptotic minimum of 115.80±1.59 mol mol–1, and C e converged onto a maximum asymptotic value of 1.645±0.054 mol(CO2) m–2 s–1 Pa–1 and declined to zero at a NL-intercept equal to 0.596±0.096 g(N) m–2. fell to zero for an NL-intercept of 0.660±0.052 g(N) m–2. As NL increased, the value of P Nmax converged onto a maximum asymptotic value of 33.400±2.563 mol(CO2) m–2 s–1. P N fell to zero for an NL-intercept of 0.710±0.035 g(N) m–2. Under variable daily meteorological conditions the values for NL, specific leaf area (L), root mass fraction (Rf), P Nmax, and remained constant for a given N supply. A monotonic decline in the steady-state value of Rf occurred with increasing N supply. L increased with increasing N supply or with increasing NL.  相似文献

The effect of adding UV-A radiation (320–400 nm) to photosynthetically active radiation (PAR, 400–700 nm) during growth of the photosynthetic marine microalga Dunaliella bardawil was investigated in this work in terms of cell growth and carotenoid production. Although signs of slow cell growth (slight reduction of chlorophyll and protein content) were observed after 24 h of cell exposure to UV-A (40 mol photons m–2 s–1 and 70 mol photons m–2 s–1) plus 140 mol photons m–2 s–1 PAR , 84 h exposure to these UV-A conditions slightly stimulated cell growth and increased the photosynthetic efficiency of the exposed cultures. The enhanced cell growth was coupled with an increase in total carotenoid content. Besides -carotene as the major pigment, increases in the well-known antioxidants lutein and zeaxanthin of about 3-fold and 5-fold, respectively, were determined in cultures exposed to UV-A radiation of 70 mol photons m–2 s–1for 84 h. As a consequence, far from being negative to cell growth, low and medium UV-A radiation are stress factors that could be successfully applied to long-term processes for large scale carotenoid production using D. bardawil cultures with retention of cell viability. UV-A exposure has the advantage of being a factor either easily applied or removed as required, in contrast to other nutrient stresses, which require medium replacement for their application.  相似文献

The effects of eight microelements (I, BO3 3–, MoO4 2–, Co2+, Cu2+, Mn2+, Fe2+, Zn2+) on the biosynthesis of camptothecin and the growth of suspension cultures of Camptotheca acuminata were studied. The increase of I to 25 M l–1, Cu2+ to 1 M l–1, Co2+ to 2 M l–1 and MoO4 2– to 10 M l–1 in Murashige and Skoog (MS) medium resulted in 1.66, 2.84, 2.53 and 2.04 times higher of camptothecin yield than that in standard MS medium respectively. Combined treatment of I (25 M l–1), Cu2+ (1 M l–1), Co2+ (2 M l–1) and MoO4 2– (10 M l–1) lead to improve cell dry weight, camptothecin content, and camptothecin yield to 30.56 g l–1, 0.0299%, and 9.15 mg l–1, respectively, which were 20.2, 208.9 and 273.8% increment respectively when compared with those of control.  相似文献

Summary Observations of aperture changes as sucrose is added to the solution bathing epidermal strips ofCommelina communis L. allow calculation of the osmotic changes required to open or close the stomatal pore, for comparison with changes in potassium content. With isolated guard cells, in strips in which all cells other than guard cells have been killed, the internal osmotic changes required are 83 mosmol kg–1 m–1 below 10m aperture, 129 mosmol kg–1 m–1 in the range 10–15 m, and 180 mosmol kg–1 m–1 above 15 m. For opening against subsidiary cell turgor in addition to guard cell turgor, in intact strips with live subsidiary and epidermal cells, these figures should each be increased by about 33 mosmol kg–1 m–1. A change in subsidiary cell turgor is magnified in its effects on the water relations of the guard cell by a factor greater than 3.7 for equal changes in the water potential of the two cells, or greater than 4.7 at constant volume of the guard cell.  相似文献

Hydrodictyon reticulatum (L.) Lagerh. is a recent addition to the New Zealand flora and is expanding its distribution rapidly. Proliferations of the alga now constitute an economic nuisance in waters which have not previously suffered filamentous algal blooms. To better understand the current and likely future spread of the alga and to identify possible management options the alga's growth requirements have been investigated. A strain isolated from New Zealand tolerated temperatures between 5 and 40 °C and salinities from 0 to 5. Optimal growth was at 25 °C, 150 mol photon m–2s–1 and in freshwater. Nett photosynthesis was saturated at photon flux densities of 100 and 160 mol m–2s–1 at 12 and 20 °C, respectively. Growth rate was linearly related to internal N concentration and hyperbolically to internal P concentration. Minimum cellular nutrient contents, by weight, were 1% N and 0.2% P. Growth was saturated at contents of 5% N and 0.5% P under the conditions of culture (20 °C, 150 mol photon m–2s–1). The alga maintained optimal cellular N content at low ambient nitrate concentrations (100 mg m–3) half optimum content at 18 mg m–3. Affinity for filtrable reactive phosphorus was not unusually high compared to other filamentous algae. We suggest that this alga is occupying a niche in New Zealand which has been precluded from other filamentous nuisance algae by low N concentration and N:P ratio. The significance of these findings in setting environmental targets for management of this nuisance alga is discussed.Author for correspondence  相似文献

Summary We examined the transition from ectothermy to endothermy in nestling bank swallows (Riparia riparia) by measuring the peak metabolic response to cold (PMR) in groups of nestlings. Additionally aerobic capacity, as assessed by citrate synthase activity (CS), and contractile function, as assessed by myofibrillar ATPase activity (mATPase) were measured in the pectoralis and mixed leg muscles during development. During the first 65% of their growth (from 2–12 g) bank swallows do not increase their metabolic rate in response to cold (Fig. 1). Between 12 and 16 g the PMR increased from 4 to more than 10 ml O2 (g·h)–1. Citrate synthase activity increased throughout development, starting at 20 moles (min·g fresh mass)–1 in both tissues and increasing to 150 and 50 moles (min·g)–1 in the pectoralis and leg muscles, respectively (Fig. 5). The augmented aerobic capacity combined with large increases in muscle mass undoubtedly contributes to the improved thermoregulatory abilities of older nestlings. However, muscle mass and aerobic capacity increase continuously and do not show the sharp transition noted in PMR. In the leg muscle mATPase activity is constant throughout growth, but in the pectoralis muscle it undergoes an abrupt increase from 0.5 moles (min·mg myofibrillar protein)–1 in animals weighing less than 12 g to 0.9 moles (min·mg)–1 in nestlings weighing more than 15 g (Fig. 6). Excised root cultures of Gloriosa superba reached 7.5 g dry wt l–1 and accumulated 240±40 g colchicine g–1 cell dry wt after 4 weeks growth. While all precursors (except trans-cinnamic acid) enhanced colchicine content of root cultures without adversely affecting root growth, treatment with p-coumaric acid + tyramine (each at 20 mg l–1) increased colchicine content to 1.9 mg g–1 cell dry wt.  相似文献

Plant phenotype stability during ex vitro growth, one of the main requirements of plant micropropagation, was tested on tobacco. Plants cultivated in vitro in the presence of 3 % sucrose under photon flux density (PFD) of 200 mol m–2 s–1 (3 % HL plants) showed the best growth and photosynthetic parameters in the course of 7-day acclimation. However, significant change in phenotype of these plants appeared under a decrease in PFD to 50 mol m–2 s–1 during further ex vitro growth (in the period of 7th – 17th day). Much higher internodia elongation was found in 3 % HL plants in comparison with plants grown in vitro on sucrose media under PFD of 50 mol m–2 s–1 (3 % LL) or without sucrose either under PFD of 50 mol m–2 s–1 or 200 mol m–2 s–1 (0 % LL, 0 % HL). It can be presumed that 3 % HL plants show permanent demand for high PFD. Neither ABA or chlorophyll contents nor de novo thylakoid membrane synthesis were related to the morphogenic effect of low PFD. Changeable contents of hexoses in leaves of 3 % HL and 3 % LL plants were in no direct correlation to the elongated growth.  相似文献

The aim of the study was to determine the influence of total dissolvedsolids/salinity (mg L-1 TDS) on photosynthetic activity of Nodularia spumigena strain 001E isolated from Lake Alexandrina, SouthAustralia, using photosynthesis-irradiance (PI) curves. N. spumigena001E cultures were grown in ASM medium at a range of TDSconcentrations (360, 6,600, 13,200, 19,800, 26,400 mg L-1)at an irradiance of 30 mol m-2 s-1 (PAR, 400–700 nm) at 25 °C. The PI relationship was determined at 25 °Cfor irradiances between 0 and 500 mol photon m-2s-1 (PAR). The initial slope of PI curve, , a function of lightharvesting efficiency and photosynthetic energy conversion, decreasedproportionally with an increase in salinity from 360 to 26,400 mgL-1 TDS. The maximum rate of photosynthesis (Pmax),occurred at 6,600 mg L-1 TDS. No influence of salinity onIk, the irradiance at which Pmax was measured, or on Rd, the dark respiration rate, was identified.  相似文献
