The IP4 highway and the new projected road network have been changing the critical Iberian wolf (Canis lupus signatus) habitat in Trás-os-Montes e Alto Douro region (north-eastern Portugal). A holistic stochastic dynamic methodology (SDM) was developed for predicting changes in Iberian wolf population due to the new highways construction, increasing road traffic density and urbanization. The ecological indicators for wolf presence and abundance were estimated indirectly from the density of attacks on livestock. The low density of wild ungulates in the area makes livestock the main source of food for wolves. These indirect ecological indicators and state variables are the underlying database of our SDM model. This model focus on the interactions between conceptually isolated key-components, such as the density of attacks on livestock and changes in habitat conditions, providing some basis to analyse the responses of wolf populations to the environmental scenarios that will characterize the region in the future. The model proposed was preceded by a conventional multivariate statistical procedure (stepwise multiple regression analysis) performed to discriminate the significant relationships between livestock attacks and socio-environmental variables. The holistic parameters of the model were estimated from the results of the statistical treatment and from regional data regarding tendencies within the use of land. The simulation results demonstrate the SDM capability in capturing the trends of this changing region, namely by showing a dramatically decrease of wolf attacks in the region for the next decade.  相似文献

The main goal of ex situ conservation programs is to improve the chances of long term survival of natural populations by founding and managing captive colonies that can serve as a source of individuals for future reintroductions or to reinforce existing populations. The degree in which a captive breeding program has captured the genetic diversity existing in the source wild population has seldom been evaluated. In this study we evaluate the genetic diversity in wild and captive populations of the Iberian wolf, Canis lupus signatus, in order to assess how much genetic diversity is being preserved in the ongoing ex situ conservation program for this subspecies. A sample of domestic dogs was also included in the analysis for comparison. Seventy-four wolves and 135 dogs were genotyped at 13 unlinked microsatellite loci. The results show that genetic diversity in Iberian wolves is comparable in magnitude to that of other wild populations of gray wolf. Both the wild and the captive Iberian wolf populations have a similarly high genetic diversity indicating that no substantial loss of diversity has occurred in the captive-breeding program. The effective number of founders of the program was estimated as ∼ ∼16, suggesting that all founders in the studbook pedigree were genetically independent. Our results emphasize also the genetic divergence between wolves and domestic dogs and indicate that our set of 13 microsatellite loci provide a powerful diagnostic test to distinguish wolves, dogs and their hybrids.  相似文献

The ability to coordinate with others' head and eye orientation to look in the same direction is considered a key step towards an understanding of others mental states like attention and intention. Here, we investigated the ontogeny and habituation patterns of gaze following into distant space and behind barriers in nine hand-raised wolves. We found that these wolves could use conspecific as well as human gaze cues even in the barrier task, which is thought to be more cognitively advanced than gazing into distant space. Moreover, while gaze following into distant space was already present at the age of 14 weeks and subjects did not habituate to repeated cues, gazing around a barrier developed considerably later and animals quickly habituated, supporting the hypothesis that different cognitive mechanisms may underlie the two gaze following modalities. More importantly, this study demonstrated that following another individuals' gaze around a barrier is not restricted to primates and corvids but is also present in canines, with remarkable between-group similarities in the ontogeny of this behaviour. This sheds new light on the evolutionary origins of and selective pressures on gaze following abilities as well as on the sensitivity of domestic dogs towards human communicative cues.  相似文献

Careful genetic management, including cryopreservation of genetic material, is central to conservation of the endangered Mexican gray wolf. We tested a technique, previously used to vitrify human and domestic animal oocytes, on oocytes from domestic dogs as a model and from the endangered Mexican wolf. This method provided a way to conserve oocytes from genetically valuable older female Mexican wolves as an alternative to embryos for preserving female genes. Oocytes were aspirated from ovaries of 36 female dogs in December and March (0 to 65 oocytes per female) and from six female wolves (4 to 73 per female) during their physiologic breeding season, or following stimulation with the GnRH agonist deslorelin. Oocytes from dogs were pooled; half were immediately tested for viability and the remainder vitrified, then warmed and tested for viability. All oocytes were vitrified by being moved through media of increasing cryoprotectant concentration, placed on Cryotops, and plunged into liquid nitrogen. There was no difference in viability (propidium iodide staining) between fresh and vitrified, warmed dog oocytes (65.7 and 61.0%, respectively, P = 0.27). Oocyte viability after warming was similarly assessed in a subset of wolves (4 to 15 oocytes from each of three females; total 29 oocytes). Of these, 57.1% of the post-thaw intact oocytes were viable, which was 41.4% of all oocytes warmed. These were the first oocytes from a canid or an endangered species demonstrated to have maintained viability after vitrification and warming. Furthermore, our results demonstrated that vitrification of oocytes with the Cryotop technique was an option for preserving female gametes from Mexican wolves for future use in captive breeding programs, although in vitro embryo production techniques must first be developed in canids for this technique to be used.  相似文献

In this study, 200 Escherichia coli isolates from 22 rainwater tank samples in Southeast Queensland, Australia, were tested for the presence of 20 virulence genes (VGs) associated with intestinal and extraintestinal pathotypes. In addition, E. coli isolates were also classified into phylogenetic groups based on the detection of the chuA, yjaA, and TSPE4.C2 genes. Of the 22 rainwater tanks, 8 (36%) and 5 (23%) were positive for the eaeA (belonging to enteropathogenic E. coli [EPEC] and Shiga-toxigenic E. coli [STEC]) and ST1 (belonging to enterotoxigenic E. coli [ETEC]) genes, respectively. VGs (cdtB, cvaC, ibeA, kpsMT allele III, PAI, papAH, and traT) belonging to extraintestinal pathogenic E. coli (ExPEC) were detected in 15 (68%) of the 22 rainwater tanks. Of the 22 samples, 17 (77%) and 11 (50%) contained E. coli belonging to phylogenetic groups A and B1, respectively. Similarly, 10 (45%) and 16 (72%) contained E. coli belonging to phylogenetic groups B2 and D, respectively. Of the 96 of the 200 strains from 22 tanks that were VG positive, 40 (42%) were carrying a single VG, 36 (37.5%) were carrying two VGs, 17 (18%) were carrying three VGs, and 3 (3%) had four or more VGs. This study reports the presence of multiple VGs in E. coli strains belonging to the STEC, EPEC, ETEC, and ExPEC pathotypes in rainwater tanks. The public health risks associated with potentially clinically significant E. coli in rainwater tanks should be assessed, as the water is used for drinking and other, nonpotable purposes. It is recommended that rainwater be disinfected using effective treatment procedures such as filtration, UV disinfection, or simply boiling prior to drinking.  相似文献

Gray wolves (Canis lupus) were immobilized with 0.5 mg/kg xylazine plus 7.5 micrograms/kg of either sufentanil (n = 8), etorphine (n = 8), or carfentanil (n = 2). Drug doses used in this study were selected to provide consistency for comparison and are not recommended doses for effective immobilization of wolves. Induction times were similar among groups (11.9 +/- 1.0 min). Thirty min after induction, wolves were given either 0.5 mg/kg naloxone hydrochloride plus 0.15 mg/kg yohimbine hydrochloride or saline only intravenously. Arousal times for wolves given naloxone and yohimbine (1.2 +/- 0.1 min) were shorter than wolves given saline (35.5 +/- 6.4 min). Respiratory rates were similar among the three drug groups (6.9 +/- 1.0 breaths/min). One animal given sufentanil then saline was found dead 108 min after induction. Presumptive diagnosis was renarcotization and hypothermia. Results indicated that sufentanil is an effective opioid immobilizing agent for gray wolves.  相似文献

The objective of the present study was to investigate whether nuclear transfer of postmortem wolf somatic cells into enucleated dog oocytes, is a feasible method to produce a cloned wolf. In vivo-matured oocytes (from domestic dogs) were enucleated and fused with somatic cells derived from culture of tissue obtained from a male gray wolf 6h after death. The reconstructed embryos were activated and transferred into the oviducts of naturally synchronous domestic bitches. Overall, 372 reconstructed embryos were transferred to 17 recipient dogs; four recipients (23.5%) were confirmed pregnant (ultrasonographically) 23-25 d after embryo transfer. One recipient spontaneously delivered two dead pups and three recipients delivered, by cesarean section, four cloned wolf pups, weighing 450, 190, 300, and 490g, respectively. The pup that weighed 190g died within 12h after birth. The six cloned wolf pups were genetically identical to the donor wolf, and their mitochondrial DNA originated from the oocyte donors. The three live wolf pups had a normal wolf karyotype (78, XY), and the amount of telomeric DNA, assessed by quantitative fluorescence in situ hybridization, was similar to, or lower than, that of the nuclear donor. In conclusion, the present study demonstrated the successful cloning of an endangered male gray wolf via interspecies transfer of somatic cells, isolated postmortem from a wolf, and transferred into enucleated dog oocytes. Therefore, somatic cell nuclear transfer has potential for preservation of canine species in extreme situations, including sudden death.  相似文献

A total of 318 Escherichia coli isolates obtained from diarrheic and healthy pigs in Ontario from 2001 to 2003 were examined for their susceptibility to 19 antimicrobial agents. They were tested by PCR for the presence of resistance genes for tetracycline, streptomycin, sulfonamides, and apramycin and of 12 common virulence genes of porcine E. coli. Antimicrobial resistance frequency among E. coli isolates from swine in Ontario was moderate in comparison with other countries and was higher in isolates from pigs with diarrhea than in isolates from healthy finisher pigs. Resistance profiles suggest that cephamycinases may be produced by > or = 8% of enterotoxigenic E. coli (ETEC). Resistance to quinolones was detected only in enterotoxigenic E. coli (< or = 3%). The presence of sul3 was demonstrated for the first time in Canada in porcine E. coli isolates. Associations were observed among tetA, sul1, aadA, and aac(3)IV and among tetB, sul2, and strA/strB, with a strong negative association between tetA and tetB. The paa and sepA genes were detected in 92% of porcine ETEC, and strong statistical associations due to colocation on a large plasmid were observed between tetA, estA, paa, and sepA. Due at least in part to gene linkages, the distribution of resistance genes was very different between ETEC isolates and other porcine E. coli isolates. This demonstrates that antimicrobial resistance epidemiology differs significantly between pathogenic and commensal E. coli isolates. These results may have important implications with regards to the spread and persistence of resistance and virulence genes in bacterial populations and to the prudent use of antimicrobial agents.  相似文献

We investigated fluctuating asymmetry in 13 traits of the skulls and jaws of historical and contemporary populations of Scandinavian gray wolves (Canis lupus). We hypothesized that there is a higher level of fluctuating asymmetry in the inbred contemporary population than in the historical population. Our analyses did not detect any difference in the level of fluctuating asymmetry as predicted. We propose different explanation for this lack of change in fluctuating asymmetry. It is assumed that a large number of studies have failed to find a positive correlation between the level of genetic stress and developmental instability and have therefore never been published, which hampers a good understanding of fluctuating asymmetry as an indicator of developmental instability. The current study is thus important in this context. The gray wolf population in Scandinavia is characterized by an extreme bottleneck followed by two and a half decades of strong inbreeding, but no associated change in fluctuating asymmetry is detected.  相似文献

2005 年10 月至2006 年4 月,采用焦点动物取样法和全事件扫描取样法,对黑龙江省哈尔滨北方森林动园4 对圈养狼交配活动进行了观察,以期了解无人干扰下圈养狼的交配过程及其交配模式。观察时间共计25 d, 225 h,实际录像时间为126 h,记录到爬跨741 次,成功交配46 次,成功爬跨交配占总爬跨次数的6. 2% 。狼在交配过程中有锁结现象,雄狼通常在一次爬跨、多次抽动后出现射精。交配行为一般发生在8∶ 00 ~10∶ 00和14∶00 ~ 16∶ 00。雌性具有明显的邀配模式,一旦邀配成功,雌狼站立不动,尾巴偏向一侧,腰部微下躬,配合雄狼爬跨。对交配参数进行单因素方差分析,4 只雄狼的抽插时间没有差异(P = 0. 827),而其锁结行为的时间差异极其显著(F = 71.43,P <0.001),交配期持续5 ~ 14 d,交配平均持续时间为534 ± 402 s,最长达1 588 s,

Wolf (Canis lupus) diet is determined by several ecological factors which can differ with latitude and human impact on the environment. Here we aim to compare Northern and Southern Europe with respect to wolf feeding habits. Scats were collected and analysed for nine years in South-central Scandinavia and four years in Tuscany, Italy, where prey density, predator-prey size relation and habitat heterogeneity, were compared in different ecological perspectives. Consumption followed prey density in Scandinavia but not in Tuscany and the main prey species, moose and wild boar respectively, were more seasonally age diversified in Scandinavia than in Tuscany. Most likely, the risk of injury was an important factor in prey age selection, especially in Tuscany. Diet composition in Scandinavia showed a negligible variance while in Tuscany, temporal and spatial variation were clearly recognised. The underlying mechanism is most likely related to the limited ecological diversity of landscape in Scandinavia contrasted with the higher variability of South European landscapes resulting in higher variation in prey abundance and consequently prey choice.  相似文献

Domestic dogs have been recognized for their social sensitivity and aptitude in human-guided tasks. For example, prior studies have demonstrated that dogs look to humans when confronted with an unsolvable task; an action often interpreted as soliciting necessary help. Conversely, wolves persist on such tasks. While dogs'' ‘looking back’ behaviour has been used as an example of socio-cognitive advancement, an alternative explanation is that pet dogs show less persistence on independent tasks more generally. In this study, pet dogs, shelter dogs and wolves were given up to three opportunities to open a solvable puzzle box: when subjects were with a neutral human caretaker, alone and when encouraged by the human. Wolves were more persistent and more successful on this task than dogs, with 80% average success rate for wolves versus a 5% average success rate for dogs in both the human-in and alone conditions. Dogs showed increased contact with the puzzle box during the encouragement condition, but only a moderate increase in problem-solving success. Social sensitivity appears to play an important role in pet and shelter dogs'' willingness to engage in problem-solving behaviour, which could suggest generalized dependence on, or deference to, human action.  相似文献

  • 1 The wolf Canis lupus, the most widespread of the four species of large carnivores in Europe, after centuries of population decline and eradication, is now recovering in many countries. Wolves contribute to regulating prey–predator dynamics and interact with human activities, mainly livestock farming and ungulate hunting. Although wolves are protected in most European countries, illegal or incidental killing is widespread.
  • 2 Wolf populations do not show any apparent phylogeographic structuring worldwide. Molecular and morphological studies of historical samples showed evidence of wolf ecomorph extinctions, coinciding with the great Pleistocene faunal turnover.
  • 3 Extant populations show recurrent long‐range dispersal during cycles of expansion and recolonization. Demographically stable populations, in contrast, seem to be characterized by very limited gene flow.
  • 4 Despite the potential for dispersal and ecological flexibility, landscape genetic approaches have demonstrated the existence of genetically distinct wolf populations, which originated through habitat and prey specializations.
  • 5 Small isolated wolf populations may suffer from inbreeding depression, although selection for heterozygotes and the rescue effect can foster rapid population recovery. Population structure and dynamics is efficiently monitored by non‐invasive genetic methods, which are also useful to identify wolf × dogCanis lupus familiaris hybridization.
  • 6 Despite technical advances and a better knowledge of wolf biology, wolf conservation is largely dependent on humans, and on the solution of conflicts with stakeholders.

Municipal sewage influent was screened for the presence of the virulence genes encoding Shiga-like toxins SLT-I and SLT-II (slt-I and slt-II) and intimin (eaeA) and those involved in biosynthesis of O157 (rfbE) and H7 (fliC) antigens by multiplex PCR to simultaneously identify the enterohemorrhagic Escherichia coli (EHEC) O157:H7 and its virulence factors in a single reaction. The screening was carried out monthly from October 2004 to September 2005. Direct PCR analysis using total DNA from sewage concentrate showed the presence of at least one virulence gene in 100% samples (n = 12). Sixty six percent of these samples were also positive for rfbE (O157) gene and fliC (H7) gene. The PCR amplification of these genes was possible when the concentration was above 20 cells ml−1. From the multiplex PCR of the isolates following plating on Cefixime-Tellurite Sorbitol MacConkey (CT-SMAC) agar to detect non-sorbitol fermenting (NSF) colonies (n = 600), one E. coli strain carrying slt-II gene and two strains of E. coli O157:H7 carrying slt-I were detected. The results show that municipal sewage represents a potential reservoir of EHEC. CT-SMAC agar was proved to have limited E. coli O157:H7 selectivity and only 0.005% (3/600) sensitivity for sewage samples due to the high frequency (43%) of NSF strains in sewage. The enrichment of sewage sample in modified E. coli broth (mEC) increased the sensitivity of PCR resulting in the clearer amplification of five genes. Amplification of target cell type in mEC broth implied that EHEC were present in sewage in a culturable and hence potentially infectious state. However, pre-enrichment did not affect the selectivity of CT-SMAC because frequency of NSF colonies remained the same as that obtained without enrichment. The study, therefore, underscores the need for more sensitive screening techniques that can be routinely employed for the regular monitoring of sewage influent.  相似文献

The analysis of mitochondrial DNA sequences has for a long time been the most extensively used genetic tool for phylogenetic, phylogeographic and population genetic studies. Since this approach only considers female lineages, it tends to give a biased picture of the population history. The use of protein polymorphisms and microsatellites has helped to obtain a more unbiased view, but complementing population genetic studies with Y chromosome markers could clarify the role of each sex in natural processes. In this study we analysed genetic variability at four microsatellite loci on the canid Y chromosome. With these four microsatellites we constructed haplotypes and used them to study the genetic status of the Scandinavian wolf population, a population that now contains 60-70 animals but was thought to have been extinct in the 1970s. In a sample of 100 male wolves from northern Europe we found 17 different Y chromosome haplotypes. Only two of these were found in the current Scandinavian population. This indicates that there should have been at least two males involved in the founding of the Scandinavian wolf population after the bottleneck in the 1970s. The two Scandinavian Y chromosome haplotypes were not found elsewhere in northern Europe, which indicates low male gene flow between Scandinavia and the neighbouring countries.  相似文献
