Biochemical and fluorescence microscopic imaging approach has been adopted to investigate the accumulation of oil bodies at specific stages of seed development in Helianthus annuus L. cv. Morden. Seed filling in sunflower is marked with a rapid accumulation of proteins and lipids upto 30 DAA, after which protein accumulation declines whereas lipids continue to accumulate. Earliest signs of lipid accumulation are evident as early as during globular stage of embryo development. Spatially, a developing seed exhibits enhanced lipid deposition in peripheral cells. Oil body biogenesis is observed as early as 10 DAA, as is evident from the fluorescence microscopic detection of Nile red-positive entities in the protoplasts. To begin with, expression of one of the oleosin (the principal oil body membrane proteins) isoforms (16 kDa), is slower than the other two (17.5 and 20 kDa). Fatty acid composition of oil body lipids is quite similar to that of total seed lipids. An enhanced accumulation of linoleic acid is evident during later stages of seed filling. The proportion of major saturated fatty acids, palmitic (16:0) and stearic (18:0), however, do not alter much during the later phases of seed development. Present findings provide new information on oil body development, lipid accumulation and fatty acid composition, for a better understanding of the phasing of physiological and biochemical events associated with oilseed development.  相似文献

Mature canoia ( Brassica napus cv. Westar) seeds contain large quantities of the storage proteins cruciferin and napin and storage lipids rich in C18: 1 and C18:2 fatty acids. Both the quantity and quality of these products are altered by freezing during development. Further, the response to freezing changes during seed development. The effects include decreased fatty acid chain elongation, altered fatty acid unsaturation, higher lipid levels and lower protein levels. In addition, seeds in the pivotal moisture range (55%) may be predisposed to precocious germination, which is then inhibited by a lack of adequate seed moisture. The results indicate that freezing imparts its effect in two ways. Initially, there is a freezing (low temperature) component and this is followed by rapid desiccation of the seeds. Although most responses probably result from a combination of the stresses, it appears that inhibition of fatty acid chain elongation is caused by the freezing component and the gradual inhibition of storage protein accumulation is a result of accelerated seed desiccation.  相似文献

以甘蓝型油菜( Brassica napus L.)品种‘Westar’和‘Topas’为材料,通过超微结构观察和荧光定量PCR技术对油菜胚胎发育早期油体的发生、油体蛋白及脂肪酸合成转录因子基因的表达情况进行分析。结果显示:油体出现在油菜胚胎发育早期,在授粉9 ~ 11 d后(球形胚时期)的胚体和胚柄中均存在直径小于0. 5 μm的油体;荧光定量实验结果表明,除 BnCLO3 的表达量在整个胚胎发育阶段无明显变化外,其他油体蛋白基因 Oleosins 、 Steroleosins 和 BnCLO1 的表达量在心形胚时期就明显增多并持续增长;脂肪酸合成转录因子 BnLEC1 、 BnL1L 、 BnWRI1 和 BnFUS3 在胚胎发育阶段,基因表达规律均呈先上升再下降的趋势,但达到最高值的时间存在差异,其中 BnLEC1 最早, BnL1L 其次, BnWRI1 和 BnFUS3 较晚。研究结果表明甘蓝型油菜在球形胚时期出现油体,其结构蛋白和转录调控因子基因的表达自心形胚开始明显增多。  相似文献

MicroRNAs (miRNAs) and small interfering RNAs are important regulators of plant development and seed formation, yet their population and abundance in the oil crop Brassica napus are still not well understood, especially at different developmental stages and among cultivars with varied seed oil contents. Here, we systematically analyzed the small RNA expression profiles of Brassica napus seeds at early embryonic developmental stages in high-oil-content and low-oil-content B. napus cultivars, both cultured in two environments. A total of 50 conserved miRNAs and 9 new miRNAs were identified, together with some new miRNA targets. Expression analysis revealed some miRNAs with varied expression levels in different seed oil content cultivars or at different embryonic developmental stages. A large number of 23-nucleotide small RNAs with specific nucleotide composition preferences were also identified, which may present new classes of functional small RNAs.  相似文献

Cultured microspores of Brassica napus L. cvs Topas and Reston initiated cell divisions within 3 to 4 days, and globular, heart and torpedo shaped embryos were prevalent after approximately 6, 8, and 10 days, respectively. Embryos with rudimentary cotyledons were evident within 2 weeks, but those that reached this stage of development represented only 1–5% of the original microspore population. The fresh weight of microspore-derived embryos at all stages of development was significantly greater than that for zygotic embryos, but the pattern of change in fresh weight and fatty acid accumulation was similar in developing zygotic and microspore embryos. In freshly isolated microspores of both Topas (low erucic acid) and Reston (high erucic acid), the predominant fatty acid was 18:3, while 18:1 comprised less than 15% of total fatty acids. During development in both zygotic and microspore embryos, the level of 18:3 declined markedly while 18:1 rapidly increased. Erucic acid (22:1) was not detected in the early stages of embryogenesis in Reston. However, small amounts of 22:1 appeared by early cotyledonary stage and the level gradually increased in both zygotic and microspore embryos through the later stages of development. The fatty acid compositions of mature embryos was nearly identical to that of dry seed, except the level of 22:1 in Reston embryos was consistently less than in the seed. Triacylglycerols comprised only 15% of total lipids in freshly isolated microspores, but increased to more than 90% by 4 weeks. The fatty acid composition of the triacylglycerol fraction was generally similar to that of total lipids at all stages of development of microspore-derived embryos.  相似文献

The total lipid content, neutral and polar lipid composition, and fatty acid profiles were determined for the different developmental stages of the entomopathogenic fungus, Nomuraea rileyi. In general, the lipid composition of N. rileyi was found to be very similar to that of other Fungi Imperfecti. Triacylglycerides and sterols were the major neutral lipids and phosphatidylethanolamine, phosphatidylinositol, and phosphatidylserine were the major polar lipids. Phosphatidylcholine, resolved in conidial and mycelial samples, was not detected in hyphal body samples. The major fatty acids were palmitic, oleic, and linoleic. Stearic acid was detected as a minor component of hyphal body samples. During conidiogenesis the overall lipid composition was altered in (1) a reduction in total lipid content, (2) an increase in the polar/neutral lipid ratio, and (3) an increase in the degree of fatty acid unsaturation.  相似文献

Genomic fatty acid elongation 1 (FAE1) clones from high erucic acid (HEA) Brassica napus, Brassica rapa and Brassica oleracea, and low erucic acid (LEA) B. napus cv. Westar, were amplified by PCR and expressed in yeast cells under the control of the strong galactose-inducible promoter. As expected, yeast cells expressing the FAE1 genes from HEA Brassica spp. synthesized very long chain monounsaturated fatty acids that are not normally found in yeast, while fatty acid profiles of yeast cells expressing the FAE1 gene from LEA B. napus were identical to control yeast samples. In agreement with published findings regarding different HEA and LEA B. napus cultivars, comparison of FAE1 protein sequences from HEA and LEA Brassicaceae revealed one crucial amino acid difference: the serine residue at position 282 of the HEA FAE1 sequences is substituted by phenylalanine in LEA B. napus cv. Westar. Using site directed mutagenesis, the phenylalanine 282 residue was substituted with a serine residue in the FAE1 polypeptide from B. napus cv. Westar, the mutated gene was expressed in yeast and GC analysis revealed the presence of very long chain monounsaturated fatty acids (VLCMFAs), indicating that the elongase activity was restored in the LEA FAE1 enzyme by the single amino acid substitution. Thus, for the first time, the low erucic acid trait in canola B. napus can be attributed to a single amino acid substitution which prevents the biosynthesis of the eicosenoic and erucic acids.  相似文献

The lipid profile of Pleurotus sajor caju was studied in relation to mycelial and sporophore growth and different cultural factors. The growth was characterised by lipid synthesis during mycelial growth and utilisation during sporophore growth. The degree of instauration increased during mycelial growth and decreased during sporophore formation. The fatty acid composition of mycelium and sporophore was similar, linoleic acid (C18:2) being the most dominant acid in both. C:N ratio had a significant (P<0.05) positive effect on mycelial dry weight; however, per cent total lipids was similar. Non-polar lipids became more unsaturated as the temperature was raised from 10° to 25°C and pH from 3.0 to 6.0, but declined when the cultures were aerated. Mycelial dry weight increased significantly (P<0.05) when the liquid medium was supplemented with lipids. In general, fatty acids with carbon chain length C16 and C18 stimulated the growth of mycelium. Supplementation of solid substrate (cotton seed hulls) with safflower oil, soybean oil or rice bran significantly (P<0.05) increased the yield of sporophores. Total lipids and ratio of non-polar to polar lipids were not affected by lipid supplementation.  相似文献

The present study aimed to quantify the methyl esters of lenoleic acid (LA), γ-lenolenic acid (LNA) and oleic acid (OL) in the oil of Brassica napus mutants. Five stable mutants (ROO-75/1, ROO-100/6, ROO-125/12, ROO-125/14, and ROO-125/17)of B. napus cv. 'Rainbow' (P) and three mutants (W97-95116, W97-0.75/11 and W97-.075/13) of B. napus cv. 'Westar' (P) at M6 stage, exhibiting better yield and yield components, were analyzed for essential fatty acids. The highest seed yield was observed in the mutant (ROO-100/6) followed by ROO-125/14 of Rainbow, that is, 34% and 32% higher than their parent plants, respectively. Westar mutant W97-75/11 also showed 30% higher seed yield than its parent plant. High performance liquid chromatography analysis of the composition of fatty acids indicated that OL was the most dominant fatty acid, ranging from 39.1 to 66.3%; LA was second (15.3-41.6%) and LNA was third (18.1-28.9%). Mutant ROO-125/14 showed higher OL contents than parent (Rainbow). These results are expected to support the approval of ROO-125/14 in the National Uniform Varietal Yield Trials (NUVYT) as a new variety based on high oil quality.  相似文献

Westar)在机械损伤前后可溶性总蛋白的含量变化并试图通过蛋白质组学技术来检测可能发生变化的蛋白质。 蛋白质定量检测发现,同一植株同一叶片损伤前后可溶性总蛋白含量差异显著,损伤后蛋白表达量增高。 蛋白质双向凝胶电泳及其差异显示分 析损伤前后的蛋白质组,表明有8个蛋白质点发生明显的上调或下调。选择其中2个差异蛋白点经过MALDI-TOF质谱鉴定,它们分别是Rubisco小 亚基前体、果糖-1,6-二磷酸醛缩酶和粪卟啉-3-氧化酶,这些蛋白质可能在油菜叶片应答机械损伤过程中对维持植物的生理功能起到重要作用 。  相似文献

High-erucic acid (HEA) Brassica napus cultivars are regaining interest in industrial contexts. Erucic acid and its derivatives are important renewable raw materials utilized in the manufacture of plastic films, in the synthesis of Nylon 13,13, and in the lubricant and emollient industries. Theoretically, the highest level of erucic acid that can be achieved by means of classical breeding is 66 mol%; however, using new approaches on the basis of genetic engineering, it might be possible to develop a B. napus cultivar containing levels of erucic acid significantly above 66 mol% (>80 mol%). In an attempt to increase the amounts of very-long-chain fatty acids (VLCFAs), and erucic acid in particular, in Canadian HEA B. napus cultivars, we have focused on two targets using a transgenic approach. We examined both the role/function of the Arabidopsis thaliana FAE1 (fatty acid elongase) gene by expressing it under the control of the seed-specific napin promoter in B. napus germplasm with analysis of the changes in VLCFA content in the seed oil of transgenic lines, and the performance of the yeast SLC1-1 (sphingolipid compensation mutant) in B. napus cv. Hero transgenic progeny in the field. Here, we report analyses of the contents of 22:1, total VLCFAand oil in the seed oil, as well as seed yield of the field-grown FAE1 and SLC1-1 B. napus cv. Because of its high contents of protein, α-linolenic-rich oil, lignans, and fiber, demand is increasing for flax(Linum usitatissi-mum L.) and flax seed oil as a food source. In this comparative survey, we examined germination and the mobilization of seed storage products (lipids and soluble proteins) of 3-d-old seedlings from two flax cultivars (N 51 and H 52) challenged with moderate salinity (up to 200 mM NaCl). At the highest salt concentration, germination appeared to be cultivar-dependent, with that of ‘N 51’ being less impaired and delayed than in ’H 52’. Sodium chloride inhibited germination via osmotic and toxic effects, so that seed viability was altered, especially in ‘H 52’. At 200 mM NaCl, lipid mobilization was delayed in the earliest germination phases. This response was associated with increased proportions of linolenic acid contents in both cultivars and more linolenic acid-rich molecular species of TAGs. Irrespective of the salt level, soluble protein contents in both cultivars decreased over time, although a salt-related precocity of protein degradation occurred at 200 mM NaCl.  相似文献

Diets supplemented with relatively high levels of either saturated fatty acids derived from sheep kidney fat (sheep kidney fat diet) or unsaturated fatty acids derived from sunflower seed oil (sunflower seed oil diet) were fed to rats for a period of 16 weeks and changes in the thermotropic behaviour of liver and heart mitochondrial lipids were determined by differential scanning calorimetry (DSC). The diets induced similar changes in the fatty acid composition in both liver and heart mitochondrial lipids, the major change being the omega 6 to omega 3 unsaturated fatty acid ratio, which was elevated in mitochondria from animals on the sunflower seed oil diet and lowered with the mitochondria from the sheep kidney fat dietary animals. When examined by DSC, aqueous buffer dispersions of liver and heart mitochondrial lipids exhibited two independent, reversible phase transitions and in some instances a third highly unstable transition. The dietary lipid treatments had their major effect of the temperature at which the lower phase transition occurred, there being an inverse relationship between the transition temperature and the omega 6 to omega 3 unsaturated fatty acid ratio. No significant effect was observed for the temperature of the higher phase transition. These results indicate that certain domains of mitochondrial lipids, probably containing some relatively higher melting-point lipids, independently undergo formation of the solidus or gel phase and this phenomenon is not greatly influenced by the lipid composition of the mitochondrial membranes. Conversely, other domains, representing the bulk of the membrane lipids and which probably contain the relatively lower melting point lipids, undergo solidus phase formation at temperatures which reflect changes in the membrane lipid composition which are in turn, a reflection of the nature of the dietary lipid intake. These lipid phase transitions do not appear to correlate directly with those events considered responsible for the altered Arrhenius kinetics of various mitochondrial membrane-associated enzymes.  相似文献

利用荧光染料尼罗红染色和激光扫描共聚焦显微观察技术, 建立了油菜油体观察或生物体内中性脂类物质定性鉴定的研究体系。对高油品种宁油14号、宁油18号、ZH-088和低油品种ZL-366、NjY008、Westar共6个甘蓝型油菜品种子叶 贮藏细胞内的油体进行了观察。研究发现: 油菜种子成熟过程中, 油体从着色不明显的小颗粒, 逐渐发育形成着色清晰的球状大油体。种子成熟干燥后, 油体间很少发生聚合。在成熟干燥的种子中, 油体集中分布于子叶贮藏细胞中央, 呈椭圆形或不规则形状, 较少为圆形。通过研究种子内油体与含油量的关系, 发现高油品种组与低油品种组之间在单个子叶贮藏细胞内油体数量和截面积之和存在明显差异, 而在高油品种组内或低油品种组内的差异不明显。结果显示, 油菜种子细胞中油体的数量和总面积与含油量之间存在正相关, 可作为高油分材料的选择依据。  相似文献

Juvenile soft-shelled turtles (Pelodiscus sinensis) were fed 7 diets containing 8% of lard, soybean oil, olive oil, menhaden fish oil, or mixtures of 1 to 1 ratio of fish oil and lard, soybean oil, olive oil for 10 weeks. Growth and muscle proximate compositions of the turtles were not affected by different dietary treatments (p>0.05). Fatty acid profiles in muscle polar lipids, muscle non-polar lipids, and liver polar lipids reflected the fatty acid composition of dietary lipid source. Turtles fed diets containing fish oil generally contained significantly higher (p<0.05) proportion of highly unsaturated fatty acids (HUFA) in both polar and non-polar lipids of muscle and polar fraction of liver lipids than those fed other oils. Non-polar fraction of liver lipids from all groups of turtles contained less than 1% of HUFA. All turtles contained relatively high proportions of oleic acid in their lipids regardless of the dietary lipid source. Further, lipid peroxidation in both muscle tissue and liver microsomes of turtles fed fish oil as the sole lipid source was greater (p<0.05) than those fed fish oil-free diets. Turtles fed olive oil as the sole lipid source had the lowest lipid peroxidation rate among all dietary groups. The results indicate that dietary n-3 HUFA may not be crucial for optimal growth of soft-shelled turtles although they may be used for metabolic purpose. Further, high level of dietary HUFA not only increases the HUFA content in turtle tissues, but also enhances the susceptibility of these tissues to lipid peroxidation.  相似文献

Oat (Avena sativa) is unusual in comparison with other cereals since there are varieties with up to 18% oil content. The lipid content and fatty acid composition in different parts of the grain during seed development were characterized in cultivars Freja (6% oil) and Matilda (10% oil), using thin-layer and gas chromatography, and light and electron microscopy. The majority of lipids (86-90%) were found in the endosperm. Ninety-five per cent of the higher oil content of cv. Matilda compared with cv. Freja was due to increased oil content of the endosperm. Up to 84% of the lipids were deposited during the first half of seed development, when seeds where still green with a milky endosperm. Microscopy studies revealed that whereas oil bodies of the embryo and scutellum still contained a discrete shape upon grain maturation, oil bodies of the endosperms fused upon maturation and formed smears of oil.  相似文献

As a model system for studying possible oxidation changes insoybeans with ageing, whole soybean seeds, ground soybeans orsoybean oil were exposed to a heated oxygen atmosphere (105°C)for periods of up to 6 d. With the exception of polar lipidsof the embryonic axis, seed lipids were highly resistant tooxidative degradation provided seed structure was maintainedintact; however, the non-lipid fraction of the seed rapidlybecame discoloured. Polar lipids of ground seed material, andboth total and polar lipids in isolated oil, were less stableto oxidation than similar lipids within whole seeds. Seed filling is a dynamic, temporally regulated phase of seed development that determines the composition of storage reserves in mature seeds. Although the metabolic pathways responsible for storage reserve synthesis such as carbohydrates, oils, and proteins are known, little is known about their regulation. Protein phosphorylation is a ubiquitous form of regulation that influences many aspects of dynamic cellular behavior in plant biology. Here a systematic study has been conducted on five sequential stages (2, 3, 4, 5, and 6 weeks after flowering) of seed development in oilseed rape (Brassica napus L. Reston) to survey the presence and dynamics of phosphoproteins. High resolution two-dimensional gel electrophoresis in combination with a phosphoprotein-specific Pro-Q Diamond phosphoprotein fluorescence stain revealed approximately 300 phosphoprotein spots. Of these, quantitative expression profiles for 234 high quality spots were established, and hierarchical cluster analyses revealed the occurrence of six principal expression trends during seed filling. The identity of 103 spots was determined using LC-MS/MS. The identified spots represented 70 non-redundant phosphoproteins belonging to 10 major functional categories including energy, metabolism, protein destination, and signal transduction. Furthermore phosphorylation within 16 non-redundant phosphoproteins was verified by mapping the phosphorylation sites by LC-MS/MS. Although one of these sites was postulated previously, the remaining sites have not yet been reported in plants. Phosphoprotein data were assembled into a web database. Together this study provides evidence for the presence of a large number of functionally diverse phosphoproteins, including global regulatory factors like 14-3-3 proteins, within developing B. napus seed.  相似文献
