Rice seedlings germinated for up to 96 hr from the imbibition under strictly anaerobic conditions had a certain level of respiratory activity, though less than that of aerobic controls. Nitro-BT positive particles were recognized in coleoptile cells of these seedlings. Respiratory activity in shoots from these seedlings showed a marked rise after exposure to air.  相似文献

Activity of nitrate reductase in roots and cotyledons of cotton seedings (Gossypium hirsutum L. cv. Deltapine 16) increased rapidly on germination, reaching a maximum after 1 day of imbibition. Thereafter, activity declined until emergence and greening of the cotyledons, when it again began to increase steadily. Germinating soybean (Glycine max (L.) Merrill cv. Merit) and sunflower (Helianthus annuus L. cv. Peredovic) seedlings did not show the early peak of activity. The early peak depended on nitrate and was sensitive to cycloheximide, but not to actinomycin D or other inhibitors of RNA synthesis. The second, light-dependent increase was sensitive to actinomycin D. In roots, the early peak of activity occurred before any growth. After emergence of the root tip from the seed coat, activity was localized in the terminal 2 millimeters, whether expressed on a fresh weight, protein, or root basis. The difference in activity between the apical (0-2 millimeter) and subapical (2-4 millimeter) segments did not result from differences in nitrate availability, energy supply, or turnover rates of nitrate reductase. Root activity was similar to that of the cotyledons after emergence, in that both were sensitive to actinomycin D.  相似文献

Dehydroquinate synthase, an enzyme catalyzing the conversion of 3-deoxy-D-arabino-heptulosonate 7-phosphate (DAHP) to 3-dehydroquinate, was detected in cell-free extracts of etiolated Phaseolus mungo seedlings. The reaction product, 3-dehydroquinate, formed from [1-14C]DAHP was identified by paper-radiochromatography. The enzyme required NAD+ and Co2+ for activity.  相似文献

Occurrence, synthesis and localization of lectins in coleoptiles of 3-day old seedlings of wheat, rye, barley and rice were studied by a combination of high resolution ion-exchange chromatography, in vivo labelling with 35S-cysteine and immunocytochemistry. Whereas no lectin can be isolated or localized in barley coleoptile, 1.9 and 40 ng of lectin per coleoptile was obtained from wheat and rye respectively. Wheat germ agglutinin was localized in the outer layer of the wheat coleoptile and both inner and outer layers of rye coleoptile displayed a specific reaction. In rice, 250 ng of lectin is present in the coleoptile and is distributed throughout this organ. Nitrate reduotase is induced by nitrate in excised embryos and germinating intact seedlings of rice (Oryza sativa L.). The enzyme is induced 24 hr after imbibition. The rate of enzyme formation increases with the age of seedlings. There is a lag period of 30 to 40 min between the addition of substrate and the formation of nitrate reductase. Formation of the enzyme is promoted by the presence of ammonium. Chloramphenicol, actinomycin D and cycloheximide effectively inhibit the formation of nitrate reductase.  相似文献

Seed germination is a complex physiological process in plants that can be affected severely by heavy metals. The interference of germination by cadmium stress has not been well documented at the proteomic level. In the present study, in order to investigate the protein profile alternations during the germination stage following exposure to cadmium, a proteomic approach has been adopted in combination with morphological and physiological parameters. Seeds were exposed with a wide range of cadmium between 0.2 and 1.0 mM. Increases of cadmium concentration in the medium resulted in increased cadmium accumulation in seeds and TBARS content, whereas germination rate, shoot elongation, biomass, and water content were decreased significantly. Temporal changes of the total proteins were investigated by two-dimensional electrophoresis (2-DE). Twenty-one proteins were identified using MALDI-TOF mass spectrometry, which were upregulated at least 1.5-fold in response to cadmium stress. The identified proteins are involved in several processes, including defense and detoxification, antioxidant, protein biosynthesis, and germination processes. The identification of these proteins in the cadmium stress response provides new insight that can lead to a better understanding of the molecular basis of heavy metal responses of seeds at the germination stage.  相似文献

Carbohydrate metabolism in growing rice seedlings under arsenic toxicity   总被引:7,自引:0,他引:7  
We studied in the seedlings of two rice cultivars (Malviya-36 and Pant-12) the effect of increasing levels of arsenic in situ on the content of sugars and the activity of several enzymes of starch and sucrose metabolism: alpha-amylase (EC 3.2.1.1), beta-amylase (EC 3.2.1.2), starch phosphorylase (EC 2.4.1.1), acid invertase (EC 3.2.1.26), sucrose synthase (EC 2.4.1.13) and sucrose phosphate synthase (EC 2.4.1.14). During a growth period of 10-20 d As2O3 at 25 and 50 microM in the growth medium caused an increase in reducing, non-reducing and total soluble sugars. An increased conversion of non-reducing to reducing sugars was observed concomitant with As toxicity. The activities of alpha-amylase, beta-amylase and sucrose phosphate synthase declined, whereas starch phosphorylase, acid invertase and sucrose synthase were found to be elevated. Results indicate that in rice seedlings arsenic toxicity causes perturbations in carbohydrate metabolism leading to the accumulation of soluble sugars by altering enzyme activity. Sucrose synthase possibly plays a positive role in synthesis of sucrose under As-toxicity.  相似文献

Summary Tamarind, a multipurpose tropical tree species, is economically important for sustainable development of wasteland due to its hardy nature and adaptability to various agroclimatic ocnditions. Reports on in vitro morphogenesis in this species are limited, due to its recalcitrant and callogenic nature. To overcome these limitations, an attempt was made to induce meristematic activity in seedling explants. Seedlings were germinated in medium with or without thidiazuron (4.54, 9.08, 13.12, 18.16 μM). This growth regulator restricted the differentiation of the apical meristem to form shoots. It triggered proliferation of the meristematic tissue at the cotyledonary node and a large number of meristematic buds appeared in a ridial pattern around the node. The meristematic activity extended to the junction of the epicotyl and hypocotyl, giving rise to buds in the form of protuberances in all sides of the junction. These buds differentiated to form shoot primordia and subsequently to shoots in medium devoid of growth regulators. Plants developed by micrografting of these shoots on seedling-derived rootstocks survived in soil.  相似文献

Lipoxygenase (EC 1.13.1.13) was found in seedlings of Citrullus lanatus (Thunb.) Matsum. and Nakai (watermelon). The enzyme has pH optima of 4.4 and 5.5 and is inhibited by 0.2 mM nordihydroguaiaretic acid. During germination and early growth of the castor bean (Ricinus communis) nitrogenous constituents from the endosperm are transferred via the cotyledons to the growing embryo. Exudate collected from the cut hypocotyl of 4-day seedlings contained 120 millimolar soluble amino nitrogen and glutamine was the predominant amino acid present, comprising 35 to 40% of the total amino nitrogen. To determine the nature of nitrogen transfer, the endosperm and hypocotyl were removed and glutamine uptake by the excised cotyledons was investigated. Uptake was linear for at least 2 hours and the cotyledons actively accumulated glutamine against a concentration gradient. The uptake was sensitive to respiratory inhibitors and uncouplers and efflux of glutamine from the excised cotyledons was negligible. Transport was specific for the l-isomer. Other neutral amino acids were transported at similar rates to glutamine. Except for histidine, the acidic and basic amino acids were transported at lower rates than the neutral amino acids. For glutamine transport, the K(m) was 11 to 12 millimolar and the V(max) was 60 to 70 micromoles per gram fresh weight per hour. Glutamine uptake was diminished in the presence of other amino acids and the extent of inhibition was greatest for those amino acids which were themselves rapidly transported into the cotyledons. The transport of amino acids, on a per seedling basis, was greatest for cotyledons from 4-to 6-day seedlings, when transfer of nitrogen from the endosperm is also maximal. It is concluded that the castor bean cotyledons are highly active absorptive organs transporting both sucrose and amino acids from the surrounding endosperm at high rates.  相似文献

Effect of nitrogen (N) deficiency on antioxidant status and Cd toxicity in rice seedlings was investigated. N deficiency resulted in a reduction of shoot growth but not root growth. The contents of N-containing compounds such as nitrate, chlorophyll, and protein decreased in leaves of rice seedlings grown under N deficiency. Accumulation of abscisic acid and H2O2 in leaves was induced by N deficiency. The content of ascorbate and the activities of ascorbate peroxidase, glutathione reductase, and catalase in N-deficient leaves were lower than their respective control leaves. However, glutathione content was not affected and superoxide dismutase activity was increased by N deficiency. Cd toxicity in N-deficient seedlings was more pronounced than that in N-sufficient ones. Pretreatment with ascorbate or L-galactono-1,4-lactone, a biosynthetic precursor of ascorbate resulted in a reduction of Cd toxicity enhanced by N deficiency. N deficiency also resulted in an enhancement of Cd uptake in rice seedlings. The possible mechanism of Cd toxicity enhanced by N deficiency is discussed.  相似文献

Increased antioxidant activity in Cassia seedlings under UV-B radiation   总被引:2,自引:0,他引:2  
Cassia auriculata L. seedlings were irradiated with ultraviolet B (UV-B) in an environment-control chamber. The two doses assayed (7.5 and 15.0 kJ m−2) induced oxidative damage with an increase in lipid peroxidation and hydrogen peroxide and a decrease in chlorophyll and total phenol contents. The ascorbate and dehydroascorbate content as well as the reduced glutathione/oxidized glutathione content and ratio were significantly increased. The UV-B stress led to significant increases of the activity of superoxide dismutase, catalase, peroxidase and polyphenol oxidase. It is suggested that Cassia seedlings try to counteract high concentrations of oxygen species produced under UV-B stress through a co-ordinated increase in the contents and activities of antioxidants involved in their detoxification.  相似文献

Leaves and leaf sheaths of rice (Oryza sativa L.) seedlings responded to 100 μmol/L jasmonic acid with necrotic lesions and browning, respectively. By SDS-PAGE and immunoblot analysis, light-, dose- and time-dependent drastic reductions in ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBisCO) were found in jasmonate-treated leaves and leaf sheaths. Cycloheximide, free radical scavengers, a metal chelator, and protease and photosystem II inhibitors significantly reversed decrease in RuBisCO protein. Polypeptides were induced in light-incubated jasmonate-treated leaf sheaths. Surprisingly in dark incubated leaves only, induction was blocked by cycloheximide, indicatingde novo synthesis. Photosystem II inhibitors, the metal chelators 1,10-phenanthroline and EDTA, and a protease inhibitor, tosyl-lysine chloromethyl ketone, also blocked polypaptide induction, whereas free radical scavengers were only partially effective. Results indicate that jasmonate action in rice may be mediated through inhibition or damage of photosynthesis via production of reactive oxygen species and leading to necrotic lesion formation.  相似文献
