Summary

Large numbers of germinating seedlings of two rice cultivars exhibited d- type development consisting solely of completely white coleoptile growth after 15 days in the light in both static and periodically stirred water environments in completely sealed jars. Thus, whether germinated in static or agitated water in the sealed environments, coleoptile greening and subsequent normal seedling development were inhibited by hypoxic stress caused by the large numbers of germinating rice seedlings competing for the limited environmental oxygen supply. Consequently, the evidence presented points away from the formation of unfavourable oxygen diffusion gradients in static water environments in the sealed jars as having been responsible for d type seedling development observed in previous investigations. Continuous aeration of the 15-day-old static and periodically stirred aquatic environments, respectively, resulted in complete reversal of all d seedlings into normal green plants.  相似文献

Various factors affecting the yield and viability of Pinus pinaster Ait. cotyledon protoplasts and the mitotic activity of or regenerated cells are described. A study of the effect of sterilization procedures of the plant material showed that whereas the organs collected from disinfested seedlings allow for good yield and viability of isolated protoplasts, germination under non-sterile conditions favours a greater germinating capacity and stronger mitotic activity. Numerous clusters of from 10 to 15 cells were formed after 20 days of culture when a 5% aqueous solution of calcium hypochlorite was used as a sterilizing agent.
The effects of an additional purification of the enzymes showed that although yield and viability of the protoplasts are only slightly improved, the more highly purified enzymes on the other hand enhanced the mitotic activity markedly. Between the two total enzyme concentrations used (0.2 and 0.4%, and in which the relative ratio of each element was unchanged), only the lowest level supplied a debris-free protoplast suspension; mitotic activity occurred only in that case.
Comparison of the populations of cotyledon protoplasts collected from seedlings at two different growth stages (not fully-developed or fully-expanded cotyledons) did not reveal any appreciable difference in their size distribution. Neither was the extent of cellular viability affected by the degree of cell differentiation at the time of collecting. On the other hand, the yield of protoplasts and the mitotic activity of the regenerated cells were greater when partially-developed organs were used. Moreover a pretreatment of the elongating cotyledons with a mineral (half-strength MS macronutrients and full-strength micronutrients) and hormonal (15 μ M BAP, 0.5 μ M NAA) solution improved cell division frequency.  相似文献

Experimental investigations were carried out to evaluate the effect of continuous and delayed exposure of power frequency electromagnetic fields at 5, 50 and 100 μT on germinating Vicia faba seedlings as a model system. These studies included physical parameters (length and girth of primary roots, number as well as length of lateral roots and imbibition), major biochemical constituents (total sugar, protein, and fat) and activities of important housekeeping enzymes (amylases, proteases, and lipase) at 2, 4, and 8 days of growth. Also, mitotic index and rate of DNA synthesis were studied at day 8 of growth. There was no significant change in physical parameters and major biochemical constituents between control and experimental groups. Also, the comparison between the control and experimental group of seeds showed that α-amylase activity significantly decreased at 5, 50 and 100 μT on day 2 and 4 of growth. β-amylase and protease (37C & 50C) showed a significant decrease in activity on day 2 and 4 of growth at 100 μT, whereas activity of lipase significantly decreased only on day 2 of growth at 100 μT. At day 8 of growth, all enzyme activities reverted back to the same as control. Also, there was a significant increase in mitotic index as well as 3H-thymidine uptake at 100 μT delayed exposure on day 8. The present study suggests that exposure to power frequency electromagnetic fields up to 100 μT on germinating seedlings does not cause any permanent damage since the initial alteration under the magnetic fields in some important housekeeping enzymes involved in the onset of seed germination were returened to control values on day 8 of growth. Also, the growth of the germinated seedlings was found to be enhanced by the application of power frequency magnetic fields (100 μT) as evidenced by mitotic index and 3H-thymidine uptake.  相似文献

Summary

Three different oxygen-containing germination environments demonstrate the profound influence exerted by environmental oxygen (02) on growth and plastogenesis in coleoptiles of light-germinated rice seedlings. Coleoptile greening is extensive in low numbers of seedlings germinated in a sealed, initially air-saturated, static water environment and in large numbers of seedlings germinated under unagitated water underambient gaseous exchange conditions. In seedlings germinated in air (?21% 02), coleoptile greening is sparse and extension growth is much reduced compared with coleoptile extension growth of the submerged seedlings. Coleoptile greening and shoot and root growth are completely inhibited under hypoxia resulting from large numbers of germinating seedlings competing for the limited 02 supply in the sealed, initially air-saturated, static water environment. Coleoptile extension growth is highest under hypoxia and lowest under ?21% 02. The observations presented here demonstrate that 02 stress and non- stress conditions serve as environmental signals which influence growth behaviour and plastogenesis in coleoptiles of light-germinated rice seedlings.  相似文献

Dormant seeds of ponderosa pine (Pinus ponderosa) were stratified and then planted in the greenhouse. Changes occurring in the shoot apex during germination and growth of the seedlings were observed and are described. Cell divisions in germinating embryos are first noted on the flanks of the apex near the cotyledons. From these loci, mitotic reactivation proceeds up the flanks of the apex and inward toward cells of the rib meristem. Cells are dividing actively in all regions of the shoot apex of seedlings 12 days after planting, but no cytohistological zonation is evident due to differences in staining intensity and in cell and nuclear size. In 64-day-old seedlings the mitotic rate is reduced and the characteristic zonation of pine apices begins to appear. Zonation is even more evident in older seedlings. These observations are discussed in relation to the concept of initials and to theories concerning factors which regulate growth in the shoot apex.  相似文献

High population densities of germinating rice seedlings in initiallyair-saturated sealed aquatic environments exhibited dseedling growth consisting solely of coleoptile emergence inlight and dark environments. Residual oxygen tensions of 17–23%of the initially air-saturated water containing the dseedlings were evident after 15 d in both the light and dark.Coleoptiles of all d seedlings were stark white in appearance,lacked protochlorophyllide, and contained proplastids and amyloplasts,there being no evidence of normal etioplast development in thelight or dark and no chloroplast development in the light. Thus,complete environmental anoxia was observed to be unnecessaryfor inhibiting normal chloroplast photomorphogenesis in coleoptilesof light-germinated rice seedlings. Abstract

The phytotoxic action of Irpexil (Methylacethydroxamoilacetate) on higher plants. 1: action on growth and mitotic inhibition.—The fungal secondary metabolite Irpexil (Methylacethydroxamoilacetate) shows a rather marked inhibiting effect on the growth of mono-and dycotiledons seedlings. Both length and fresh weight of the axial parts are affected, particularly in wheat and pea seedlings. The inhibiting effect is at least partially due to the antimitotic activity of this compound; Irpexil does not seem to interfere at any stage of the mitotic cycle, at least at moderate concentrations (up to 70 ppm).  相似文献

Background and Aims

Flooding slows seed germination, imposes fatalities and delays seedling establishment in direct-seeded rice. This study describes responses of contrasting rice genotypes subjected to flooding or low oxygen stress during germination and discusses the basis of tolerance shown by certain cultivars.

Methods

In one set of experiments, dry seeds were sown in soil and either watered normally or flooded with 10 cm of water. Seedling survival and shoot and root growth were assessed and seed portions of germinating seedlings were assayed for soluble sugars and starch concentrations. The whole germinating seedlings were assayed for amylase and peroxidase activities and for ethylene production. Activities of enzymes associated with anaerobic respiration were examined and gene expression was analysed separately with seeds germinating under different amounts of dissolved oxygen in dilute agar.

Key Results

Flooding during germination reduced survival but to a lesser extent in tolerant genotypes. Starch concentration in germinating seeds decreased while sugar concentration increased under flooding, but more so in tolerant genotypes. Amylase activity correlated positively with elongation (r = 0·85 for shoot and 0·83 for root length) and with plant survival (r = 0·92). Tolerant genotypes had higher amylase activity and higher RAmy3D gene expression. Ethylene was not detected in seeds within 2 d after sowing, but increased thereafter, with a greater increase in tolerant genotypes starting 3 d after sowing. Peroxidase activity was higher in germinating seeds of sensitive genotypes and correlated negatively with survival.

Conclusions

Under low oxygen stress, tolerant genotypes germinate, grow faster and more seedlings survive. They maintain their ability to use stored starch reserves through higher amylase activity and anaerobic respiration, have higher rates of ethylene production and lower peroxidase activity as germinating seeds and as seedlings. Transgenic cotton (Gossypium hirsutum L.) lines expressing the tobacco glutathione S-transferase (GST) Nt107 were evaluated for tolerance to chilling, salinity, and herbicides, antioxidant enzyme activity, antioxidant compound levels, and lipid peroxidation. Although transgenic seedlings exhibited ten-fold and five-fold higher GST activity under normal and salt-stress conditions, respectively, germinating seedlings did not show improved tolerance to salinity, chilling conditions, or herbicides. Glutathione peroxidase (GPX) activity in transgenic seedlings was 30% to 60% higher under normal conditions, but was not different than GPX activity in wild-type seedlings under salt-stress conditions. Glutathione reductase, superoxide dismutase, ascorbate peroxidase, and monodehydroascorbate reductase activities were not increased in transgenic seedlings under salt-stress conditions, while dehydroascorbate reductase activity was decreased in transgenic seedlings under salt-stress conditions. Transgenic seedlings had 50% more oxidized glutathione when exposed to salt stress. Ascorbate levels were not increased in transgenic seedlings under salt-stress conditions. Malondialdehyde content in transgenic seedlings was nearly double that of wild-type seedlings under normal conditions and did not increase under salt-stress conditions. These results show that expression of Nt107 in cotton does not provide adequate protection against oxidative stress and suggests that the endogenous antioxidant system in cotton may be disrupted by the expression of the tobacco GST.  相似文献

When watermelon seeds were cultured in a Petri dish together with amaranth, barnyard grass, cockscomb, lettuce or tomato seeds, the shoot growth of amaranth and cockscomb was markedly promoted, whereas the shoot growth of lettuce and tomato was inhibited. The shoot growth of barnyard grass was not affected. These results suggest that plant-selective allelopathic substance(s) affecting the shoot growth of other plant seedlings were exuded from watermelon seeds. An allelopathic substance was isolated from the exudates of germinating watermelon seeds and identified as vanillic acid by its spectral analysis and Rf value on TLC. Vanillic acid promoted the shoot growth of cockscomb at the concentrations of 300 to 10 mg/l and that of amaranth at the concentrations of 30 to 3 mg/l, although the shoot growth of amaranth was inhibited by 300 mg/l of vanillic acid. The shoot growth of lettuce and tomato was inhibited at the concentrations higher than 30 mg/l by vanillic acid. However, the shoot growth of barnyard grass was not affected at the concentrations used. All these results suggest that vanillic acid may play as a major component of allelopathic substance(s), which shows plant-selective activity, in the exudates of germinating watermelon seeds.  相似文献

以莴苣幼苗为受体,用培养皿法检测从细果角茴香中分离得到的二氢血根碱对莴苣幼苗根生长和根毛发育的影响,并采用根尖细胞有丝分裂检测和单细胞凝胶电泳法对其可能的作用机制进行了初步研究.结果显示:较低浓度(25、50μmol/L)二氢血根碱能显著促进莴苣根的生长,较高浓度(200、300μmol/L)二氢血根碱显著抑制根的生长;二氢血根碱(10、20、30、40、50μmol/L)对莴苣幼苗根毛发育有极显著的抑制作用,且两者均表现了浓度依赖性.较低浓度(25、50μmol/L)二氢血根碱使根尖细胞有丝分裂指数显著增加,而对根尖细胞DNA没有显著影响;较高浓度(200、300μmol/L)二氢血根碱使根尖细胞有丝分裂指数显著下降,同时根尖细胞DNA受到显著性损伤.研究发现,低浓度的二氢血根碱对莴苣幼苗根生长的促进作用主要是由于根尖细胞有丝分裂活力增加所致;而高浓度二氢血根碱对莴苣幼苗根的抑制作用极可能是由于根尖细胞DNA受到损伤,使得细胞分裂活力降低,分裂期细胞数目减少,从而导致根生长受到抑制.  相似文献

To assess the evolutionary significance of persistent seed banks, phenotypes of naturally germinating seeds must be compared with those that remain dormant under the same environmental conditions. Dormant seeds can often be induced to germinate by application of gibberellic acid (GA). However, this method is valid only if there are no phenotypic “side effects” of GA that could confound comparisons between dormant and naturally germinating seeds. We examined this assumption in Lesquerella fendleri, a short-lived perennial mustard of the desert Southwest. We exposed 3840 seeds from 16 maternal sibships to two different GA treatments (0 or 1 g/L GA) in two different germination environments (greenhouse and growth chamber), and measured germination and postgermination traits. As expected, application of GA increased germination. GA also had strong and long-lasting effects on seedling morphology. Seeds that received GA developed into seedlings that were taller, with fewer but longer leaves, than seeds that did not receive GA. Effects of GA on both dormancy and postgermination traits varied among maternal sibships. Our results indicate that for this species and this concentration of GA, morphological effects can be substantial. Further study is required to determine whether such side effects are found for lower concentrations of GA, or under conditions that encourage faster seedling growth. Nonetheless, the present results illustrate the importance of testing potential confounding effects of GA in studies of the evolution of seed dormancy and its influence on postgermination traits.  相似文献

Changes in temperature and moisture as a result of climate forcing can impact performance of planted trees. Tree performance may also be sensitive to new soil conditions, for example, brought about by seeds germinating in soils different from those colonized by ancestral populations. Such “edaphic constraint” may occur with natural migration or human‐assisted movement. Pinus ponderosa seedlings, sourced from one location (“home” site), were grown across a field environmental gradient in either their original home soil or in soils from two different “away” sites. Seedlings were inoculated with site‐specific soil organisms by germinating seeds in living soil. After 6 months, the inoculated seedlings were transplanted into sterilized soils from the home or away sites. This experimental design allowed us to uncouple the importance of abiotic and biotic soil properties and test (1) how biotic and abiotic soil properties interact with climate to influence plant growth and stress tolerance, and (2) the role of soil biota in facilitating growth in novel environments. Seedlings grew least in hotter and drier away sites with away soil biota. Home soil biota ameliorated negative impacts on growth of hotter and drier away sites. Measurements of photosynthetic rate, stomatal conductance, and chlorophyll florescence (Fv/Fm) suggest that edaphic constraint reduced growth by increasing tree water stress. Results suggest that success of Ponderosa pine plantings into warming environments will be enhanced by pre‐inoculation with native soil biota of the seed source.  相似文献

Enantiomers of triaziflam and structurally related diaminotriazines were synthesized and their herbicidal mode of action was investigated. The compounds caused light and dark-dependent effects in multiple test systems including heterotrophic cleaver and photoautotrophic algal cell suspensions, the Hill reaction of isolated thylakoids and germinating cress seeds. Dose-response experiments revealed that the (S)-enantiomers of the compounds preferentially inhibited photosystem II electron transport (PET) and algae growth with efficacies similar to that of the herbicide atrazine. In contrast, the (R)-enantiomers of the diaminotriazines were up to 100 times more potent inhibitors of growth in cleaver cell suspensions and cress seedlings in the dark than the (S)-enantiomers. The most active compound, the (R)-enantiomer of triaziflam, inhibited shoot and root elongation of cress and maize seedlings at concentrations below 1 microM. The meristematic root tips swelled into a club shape which is typical for the action of mitotic disrupter herbicides and cellulose biosynthesis inhibitors. Microscopic examination using histochemical techniques revealed that triaziflam (R)-enantiomer blocks cell division in maize root tips 4 h after treatment. The chromosomes proceeded to a condensed state of prometaphase but were unable to progress further in the mitotic cycle. Disruption of mitosis was accompanied by a loss of spindle and phragmoplast micotubule arrays. Concomitantly, cortical microtubules decreased which could lead to isodiametric cell growth and consequently to root swelling. In addition, a decline in cellulose deposition in cell walls was found 24 h after treatment. Compared to the (R)-form, triaziflam (S)-enantiomer was clearly less active. The results suggest that triaziflam and related diaminotriazines affect enantioselectively multiple sites of action which include PET inhibitory activity, mitotic disruption by inhibiting microtubule formation and inhibition of cellulose synthesis.  相似文献

Summary Early and late germinating classes of seedlings of a population of Viola blanda were censused for three years. Seedlings germinating later had a significantly higher probability of death, especially during a period of relatively high mortality three years after germination. Later germinating seedlings were consistently smaller than earlier germinating seedlings. Mortality was size-dependent and concentrated among smaller individuals. It is hypothesized that the effect of germination time on mortality three years later is mediated through differential growth.  相似文献

人工调节土壤有效硅含量及盆栽试验,研究土壤有效硅对大豆生长发育和生理功能的影响。结果表明,土壤有效硅含量在55.1~202.8mg·kg^-1范围内,随着土壤有效硅含量的提高,大豆种子萌发过程中蛋白酶和脂肪酶活性升高,淀粉酶活性无显著变化,呼吸速率加快,种子活力升高,萌发速度加快,种子萌发率无显著变化;幼苗生长过程中叶片叶绿素含量无显著变化,光合速率加快,根系活力、硝酸还原酶活力升高,蒸腾强度减弱,水分利用效率和叶含水量升高,抗旱保水能力提高。大豆幼苗含硅量与土壤有效硅含量呈线性正相关趋势(r=0.994)。土壤有效硅含量大于2028mg·kg^-1,生理功能不再显著变化,说明土壤中的硅被大豆吸收后,改善了大豆萌发种子和幼苗的生理功能,使种子萌发和幼苗生长加快。  相似文献

The activities of trypsin inhibitors and Aspergillus-proteinase inhibitors have been assayed in barley embryos germinating in different conditions. The Aspergillus-proteinase inhibitor activity increased about 20-fold during 6 days at 20°C both in separated embryos germinating on nutrient agar and in the embryos of undissected grains germinating on agar with or without mineral salts. Significant inhibitor activity was also present in root tips at later stages of vegetative growth. Whether any changes occurred in the molecular weight distribution of the inhibitors present in the seedlings was studied by gel filtration on Sephadex G-75; only minor changes were observed. The trypsin inhibitor activity per seedling remained roughly constant in all cultures. In gel filtration experiments all the activity appeared to be due to the inhibitor species present in resting embryos.  相似文献
