The post-copulatory sexually refractory stage in the male cricket Gryllus bimaculatus consists of the two substages: the first refractory stage (RS1, time-variable) between copulation and spermatophore preparation, and the second refractory stage (RS2, time-constant) between spermatophore preparation and the recommencement of courtship. To understand the mechanism of the timer for RS2, subcuticular or intraganglionic injection of biogenic amines (10(-2) mol l(-1)) was performed immediately after spermatophore preparation. RS2 was shortened by octopamine, 5-HT, 5-HTP and NA-5-HT. Among these, 5-HTP was most potent. It shortened RS2 to maximally about 38% of the control. The shortening effect continued for 4.5 h after subcuticular injection even when the hemolymph was washed out with saline at 1 hour after injection. Simultaneous injection of 5-HTP with the inhibitor (NSD-1015) of 5-HT synthesis enzyme nullified the effect of 5-HTP, indicating that the shortening effect was caused by 5-HT synthesized from extrinsic 5-HTP. Injection of the inhibitor (CHX) of protein synthesis had no effect of on RS2. These results suggest that the reproductive timer in the TAG may be controlled by 5-HT or a second messenger mediated by 5-HT.  相似文献

This stage was defined as the interval between spermatophore protrusion and recommencement of copulation or a calling song. To inactivate the central nervous system locally and reversibly, different body regions were cooled to 10 degrees C for 20-30 min after spermatophore protrusion. A behavioural test then measured the duration of the refractory stage after males recovered from cooling. Males with the head, thorax and anterior abdomen cooled did not show a lengthening of that stage. In contrast, males with the entire abdomen or even the posterior abdominal segments containing only the 6th and terminal (7th-11th) abdominal ganglia showed a lengthening of the refractory stage up to, but not exceeding, the cooling duration. When 20-min cooling was interposed twice after spermatophore protrusion, the refractory stage was lengthened by about 40 min, indicating that interposed cooling did not reset the timer. These results are in agreement with our previous hypothesis that the reproductive timer for the refractory stage in the male cricket is located in the posterior abdominal ganglia, possibly within the terminal abdominal ganglion.  相似文献

【目的】揭示绿盲蝽Apolygus lucorum腹神经节的组成结构。【方法】采用免疫组织化学染色方法,利用突触蛋白抗体对绿盲蝽成虫的腹神经节进行免疫标记,激光共聚焦扫描显微镜扫描照相获得原始数据,用图像分析软件进行标记,构建三维结构模型。【结果】绿盲蝽成虫腹神经节位于腹神经索的末端,与其前方的后胸神经节和中胸神经节紧密融合,形成后部神经节。与脑和胸神经节类似,腹神经节由周围的细胞体和内部的神经髓构成。腹神经节的神经纤维束主要包括位于腹侧的两条纵向神经连索和向两侧发出的9束神经纤维。9束神经纤维连接着9个神经原节,即富含突触联系的神经髓。这些神经原节紧密融合,无明显的边界,最后两节形成膨大的末端腹神经节。两侧的神经原节由横向的神经连锁连接起来。腹神经节外周的细胞体数量较多,排列紧密,大小一致,仅在前端背侧中间和后端腹侧中间位置分别有2个和5个体积较大的细胞体。【结论】本研究结果明确了绿盲蝽腹神经节的结构,为进一步研究昆虫的行为调控及神经系统发育和演化奠定一定的形态学基础。  相似文献

The reproduction of pseudoscorpions involves indirect sperm transfer by means of spermatophores. The spermatophores are the product of the male genital atrium. A functional interpretation of spermatophore formation in Chthonius ischnocheles is based on evidence from (a) a morphological study of the genital atrium, the associated accessory glands and the musculature (b) males sectioned during spermatophore production (c) histochemical tests on the glands and their secretions (d) biochemical analyses of one gland (posterior dorsal) and its secretion (e) the behaviour of males during this process and (f) the structure of the spermatophore.
The anterior region of the genital atrium is concerned with the production of the sperm packet. The encysted sperm and the seminal fluid from the prostatic reservoir are encapsulated in a sperm packet by a secretion from two pairs of anterior glands. The posterior region of the genital atrium is responsible for the formation of the spermatophore stalk and its distal elaboration, the two lateral collars. These parts of the spermatophore arise from the fibroin secretion of the posterior dorsal gland; the shape of the spermatophore collars is correlated with their mould, the medial diverticula. In addition the lateral glands secrete a light oil which accumulates on a thickening of the spermatophore stalk proximal to the collars. This suggested that this secretion acts as a pheromone to attract females to the spermatophore since in this species males produce their spermatophores in the absence of females.  相似文献

Serotonin immunoreactivity of neurons in the gastropod Aplysia californica   总被引:2,自引:0,他引:2  
Serotonergic neurons and axons were mapped in the central ganglia of Aplysia californica using antiserotonin antibody on intact ganglia and on serial sections. Immunoreactive axons and processes were present in all ganglia and nerves, and distinct somata were detected in all ganglia except the buccal and pleural ganglia. The cells stained included known serotonergic neurons: the giant cerebral neurons and the RB cells of the abdominal ganglion. The area of the abdominal ganglion where interneurons are located which produce facilitation during the gill withdrawal reflex was carefully examined for antiserotonin immunoreactive neurons. None were found, but two bilaterally symmetric pairs of immunoreactive axons were identified which descend from the contralateral cerebral or pedal ganglion to abdominal ganglion. Because of the continuous proximity of this pair of axons, they could be recognized and traced into the abdominal ganglion neuropil in each preparation. If serotonin is a facilitating transmitter in the abdominal ganglion, these and other antiserotonin immunoreactive axons in the pleuroabdominal connectives may be implicated in this facilitation.  相似文献

This study addresses a potentially general basis of measuring time in a biological timer. Here, we examined the effects of biogenic amines on the time-fixed post-copulatory sexually refractory stage (ca. 1 h) which is defined as the time interval between spermatophore protrusion and the onset of a calling or a mating response in the reproductive cycle of the male cricket. For subcuticular injection of amines (0.15 ml, 10(-2) mol l(-1)), the interval of the refractory stage was shortened by octopamine, serotonin, 5-hydoxytryptophan and N-acetyl-serotonin but was unchanged by tryptophan, melatonin or 5-hydroxyindol-3-acetic acid. The effect of 5-hydoxytryptophan was most potent (maximum shortening, 38%) and long lasting (ca. 4.5 h) while other amines effected only the injected cycle. Injection of 5-hydoxytryptophan (180 nl, 10(-2) mol l(-1)) into the terminal abdominal ganglion also decreased the interval to a similar extent. Simultaneous injection of 5-hydoxytryptophan with the inhibitor of the serotonin synthesis enzyme reduced the 5-hydoxytryptophan effect suggesting that this effect results from synthesis of serotonin from 5-hydoxytryptophan. The protein synthesis inhibitor cycloheximide had no effect on the interval. These results suggest that the reproductive timer is regulated by serotonergic neurons in the terminal abdominal ganglion without protein synthesis during the interval of the time-fixed sexually refractory stage.  相似文献

We localized brain neurons responsible for the inhibition of copulation actions in the male cricket, Gryllus bimaculatus. Males with one connective-cut between the brain and the terminal abdominal ganglion (TAG) were able to perform normal reproductive behaviors such as courtship, copulation and spermatophore protrusion. However, copulation response tests using a female dummy showed that they failed to exhibit a transient inhibitory state after application of noxious stimulation in the mating stage and to exhibit a tonic inhibitory state in the post-copulatory sexual refractory stage. A partial cut of a single connective (hemilateral connective-cut) combined with the copulation response test and an axonal backfilling demonstrated that approximately 40 brain neurons (per one connective), whose somata were located in the posterior region of the protocerebrum and mainly contralaterally to their descending axons running through the dorso-medial part of the connective, were candidates for the inhibition of copulation actions, at least in the mating stage. In addition, we report here for the first time, that copulation actions can be elicited shortly after copulation even in intact males. This unusual behavior was frequently observed in fledglings after the final molt. The results are discussed from the perspective of the operation of the brain inhibition system on the central pattern generator (CPG) for copulation.  相似文献

The frontal ganglion contains approximately 20 cells and rests on the two posterior elevator muscles of the roof of the pharynx, thus locating the ganglion ventral and anterior to the brain. Two frontal nerves, a pair of lateral connectives, and the single recurrent nerve connect with the ganglion. There is a centrally located neuropile which is surrounded by the perineurium which in turn is covered by the neural lamella. The perineruium contains numerous glial cells and neurons with two large neurosecretory cells located in a dorsal lateral position of the ganglion.The neurosecretory cells were examined on five occasions during the year, and no significant changes occurred in the fine structure of the organelles or cellular products. The cells appear to be engaged in the synthesis of elementary neurosecretory granules throughout the year. This observation differs from previous studies on diapausing lepidopterous larvae and pupae. Axons from these two cells enter the lateral connectives and extend toward the protocerebrum.  相似文献

Effects of arginine vasotocin (AVT) on reproductive events such as courtship behavior, pheromone release, and spermatophore discharge were investigated in the male newt Cynops pyrrhogaster. AVT enhanced the incidence and frequency of androgen-induced courtship behavior. In this case, AVT was likely to act centrally because the behavior was evoked with a much smaller amount of AVT when the hormone was administered intracerebroventricularly than when given intraperitoneally. Involvement of endogenous AVT in spontaneously occurring courtship behavior was also evidenced by the fact that administration of a V1 (vasopressor) receptor antagonist, [d(CH2)5(1), Tyr(Me)2, Arg8-vasopressin] suppressed the expression of the courtship behavior. The water in which AVT-treated males had been kept showed considerable female-attracting activity as compared with the water in which saline-injected males had been kept. Moreover, the content of sodefrin, a female-attracting pheromone in the abdominal gland, was decreased by the intraperitoneal injection of AVT, suggesting that the neurohypophyseal hormone stimulated the release of sodefrin from the abdominal gland into the water. AVT induced contraction of the excised abdominal gland concentration-dependently, and, again, the V1 receptor antagonist suppressed the AVT-induced contraction. Thus, we concluded that AVT induces the pheromone discharge, acting peripherally on a contractile structure of the abdominal gland. AVT was also found to induce spermatophore deposition in the male kept in the absence of the female. Administration of the V1 receptor blocker to the sexually developed males suppressed the spermatophore deposition. All these results indicate the involvement of AVT in reproductive events acting centrally and peripherally.  相似文献

ABSTRACT. Rhythmic respiratory nerve activity was recorded in the dragonfly larvae, Anax parthenope Julius Brauer (Anisoptera). Alternating expiratory and inspiratory bursts of spikes occurred in abdominal nerve cords isolated from all peripheral connections. These bursts are similar to the activity recorded in semi-intact preparations, suggesting that the respiratory rhythm can be generated without peripheral sensory feedback. Expiratory bursts started and ended at the same time in different segments of semi-intact preparations. When connectives were severed, the nerve cord separated from the last abdominal ganglion did not normally show rhythmic bursts; the last ganglion alone and the nerve cord connected to the last ganglion exhibited the rhythmic bursts. However, in a few cases the nerve cord separated from the last ganglion exhibited the rhythm. The results suggest that the last ganglion contains the main oscillator, but that other weak oscillators occur elsewhere.  相似文献

The influence of the intermating interval on sperm number and spermatophore mass was examined in the bushcricket Poecilimon veluchianus. Males that remated after 1 d transferred about 50% of the sperm, but significantly more than 50% of the weight of spermatophores than males that remated after 2 d. Assuming a constant rate of replenishment of sperm and spermatophore material we concluded that available spermatophore material but not available sperm number influences remating interval in P. veluchianus males.  相似文献

The impulse responses of dorsal unpaired median neurons (DUMN) of the prothoracic ganglion of the cricketGryllus bimaculatus to acoustic and tactile sensory stimuli were studied. It has been established that among these cells there are mono- and bimodal neurons, some of them with the background impulse activity. To study effects of the cerebral ganglia on the DUMN activity, section of the anterior connectives of the prothoracic ganglion was performed. The recording of the DUMN responses to the auditory stimuli before and after disconnection of the links between the prothoracic and cerebral ganglia showed that in several experiments the section of the connectives resulted in suppression of the responses. At the same time, in some experiments, no statistically significant changes in the character of the DUMN responses after the section of the connectives were observed. The existence of some modulating descending influences of the subpharyngeal ganglion on the activity of the neurons investigated is discussed.  相似文献

ABSTRACT. The influence of hypoxia and hypercapnia on the ventilatory rhythm of the hellgrammite Corydalus cornutus L. (Megaloptera) was studied. In intact animals the frequency of rhythmic retractions and protractions of abdominal gills is increased by hypoxia (10% O2, 90% N2) but no ventilatory response is elicited by hypercapnia (1–5% CO2, 20% O2, 75–79% N2).
The ventilatory motor pattern was examined by recording extracellularly from the gill retractor muscle or its efferent nerve. In response to hypoxia (8% 02, 92% N2), there are decreases in the cycle-time, the interspike interval, and the burst length of the gill retractor motorneuron. In addition, previously quiescent motorneurons associated with gill protraction are recruited.
Individual ganglia or small groups of abdominal ganglia can be isolated both from the central ganglionic chain and from the periphery by selective cutting of roots and connectives. When exposed to hypoxia, preparations that include the first abdominal ganglion show characteristic changes in the ventilatory motor pattern similar to those in intact animals. Thus sensitivity to oxygen appears to be located centrally and not peripherally. In small animals (head width < 7 mm), abdominal ganglia 2–3 and 2–7 respond characteristically to hypoxia, but in larger animals (head width > 9 mm), chains of ganglia lacking abdominal ganglion 1 fail to respond. In larger animals oxygen sensitivity may thus be concentrated in abdominal ganglion 1, whereas in smaller animals the ability to initiate a ventilatory response to hypoxia is distributed among the abdominal ganglia.  相似文献

Summary The roles of the mechanosensory afferents from the wings, cerci, tergites and genitalia in copulation behavior were examined by ablation and stimulation in the male cricketGryllus bimaculatus DeGeer.The sexually excited male cricket exhibited an intense posture (IP) upon contact stimulation of the elytra and the 4th to 9th abdominal tergites. This posture allowed the backward slipping (BWS) or hooking to take place subsequently.Backward slipping (BWS), which is the movement to get under the female, was elicited during IP by contact stimulation to the middle and distal regions of the dorsal surface of the cercus.Hooking, the coordinated movements for hanging the epiphallus onto the female's subgenital plate could be induced during IP by contact either on the dorsum, periproct or proximal 2 mm regions of the cercus. The latter two regions played a role in performing hooking accurately.Among four types of mechano-sensilla on the cercus the trichoid type was crucial for the initiation of BWS and hooking. Calculations revealed that about 60 trichoid hairs (4% of all the trichoid hairs in one cercus) were sufficient for the male to carry out hooking normally.The input from the bristle hairs on the epiphallus initiated the spermatophore extrusion (SPE) by swelling the endophallus.These results demonstrated that copulation behavior in the male cricket consisted of several motor acts and each act is triggered by specific input from the contact-sensitive sensilla on the elytra, tergites, cerci and genitalia. The sequential execution of each motor act is achieved because one motor act results in a positional change in contact with the female which in turn gives rise to another act. The abdominal cerci of the wood cricket, Nemobius sylvestris, are covered by a variety of hair‐like sensilla that differ in length, thickness, and articulation. Fillings from the cercal nerves with cobalt chloride and fluorescent dyes revealed the projection of sensory axons into the terminal abdominal ganglion of the ventral nerve chain. Two projection areas on each side of the terminal abdominal ganglion midline could be identified: a posterior cercal glomerulus and an anterior bristle neuropil. Axons from some cercal sensilla ascend through the connectives to reach the metathoracic ganglionic mass. As their axons pass through each segmental abdominal ganglion, they project medial arborization. Cross‐sections of the terminal abdominal ganglion and retrograde fills with cobalt chloride and fluorescent dyes from connectives revealed several small cells and seven pairs of giant ascending interneurons organized symmetrically. Giant somata are located contralateral to their axons (diameters between 20 and 45 μm). The cercal projections overlap extensively with the dendritic fields of the giant interneurons. In the terminal abdominal ganglion, we identified nine longitudinal tracts, two major tracts, and seven smaller ones. ABSTRACT Central nervous system (CNS) of arachnids is still mysterious and has a rich unexplored field compare to what is known in insects or crustaceans. The CNS of the spider, Achaearanea tepidariorum, consists of a dorsal brain or supraesophageal ganglion and circumesophageal connectives joining it to the subesophageal mass. As the segmentation of the arachnid brain is still under discussion, we classify the brain as a protocerebral and tritocerebral ganglion depending on the evidences which generally accepted. The subesophageal nerve mass underneath the brain is the foremost part of the ventral nerve cord. All of this nerve mass is totally fused together, and forming subesophageal ganglia in this spider. In the brain, the nerve cells are packed in the frontal, dorsal and lateral areas, but are not absent from the posterior and ventral regions. In addition, the nerve cells of the subesophageal and abdominal ganglia are only restricted to the ventral and ventolateral regions. The CNS of the spider, Achaearanea tepidariorum is similar in feature to the Family Araneidae.  相似文献

A morpho-functional investigation of the sex pheromone-producing area was correlated with the pheromone release mechanism in the female gypsy moth Lymantria dispar. As assessed by male electroantennograms (EAG) and morphological observations, the pheromone gland consists of a single-layered epithelium both in the dorsal and ventral halves of the intersegmental membrane between the 8th and 9th abdominal segments. By using the male EAG as a biosensor of real-time release of sex pheromone from whole calling females, we found this process time coupled with extension movements of the ovipositor. Nevertheless, in females in which normal calling behavior was prevented, pheromone release was detected neither in absence nor in presence of electrical stimulation of the ventral nerve cord/terminal abdominal ganglion (TAG) complex. Tetramethylrhodamine-conjugated dextran amine stainings also confirm the lack of any innervation of the gland from nerves IV to VI emerging from the TAG. These findings indicate that the release of sex pheromone from the glands in female gypsy moths is independent of any neural control exerted by the TAG on the glands, at least by way of its three most caudally located pairs of nerves, and appears as a consequence of a squeezing mechanism in the pheromone-producing area.  相似文献

The detailed developmental morphology of the spermatophore of a cricket (Gryllus bimaculatus, De Geer) is described for the first time. A specific behavioural reference point is identified from which time developmental processes proceed in a more or less robotic manner. The structures previously identified as forming part of the fully formed spermatophore, that is, the ampulla, the sperm tube and the attachment plate are all identified in detail during their development. Interestingly the attachment plate material is extruded as an amorphous mass from the ejaculatory duct prior to its insertion into the dorsal pouch, the sperm do not enter the sperm sac until the end of spermatophore formation, and the sperm tube itself is formed from a flattened layer of material which rolls up into a tube and involves the use of a die mechanism.  相似文献

Nitric oxide (NO) has an important modulatory role on the processing of sensory signals in vertebrates and invertebrates. In this investigation we studied the potential sources of NO in the terminal abdominal ganglion of the crayfish, Pacifastacus leniusculus, using NADPH-diaphorase (NADPHd) histochemistry, with NADPHd acting as a marker for NO synthase (NOS). In the terminal ganglion a mean of 27 strongly labelled NADPHd-positive cell bodies were found, and of these 80% [of stained cell bodies] [corrected] occurred in three regions located in antero-lateral, central and posterior parts of the ganglion. Ventral and antero-ventral commissures as well as specific dorsal and ventral areas of the dendritic neuropil showed positive staining. Intense labelling was seen in the ventro-medial tract, and in the connective between the terminal ganglion and the 5th abdominal ganglion. In addition, some motor neurones and neurones with branches in the sensory commissures were NADPHd positive. Our finding that NADPHd-positive cells occur in consistent patterns in the terminal abdominal ganglion implies that NO may have a role in mechanosensory processing in the crayfish.  相似文献
