Preliminary studies were conducted on wheat straw to test the technical viability of an integrated system of composting, with bioinoculants and subsequent vermicomposting, to overcome the problem of lignocellulosic waste degradation, especially during the winter season. Wheat straw was pre-decomposed for 40 days by inoculating it with Pleurotus sajor-caju, Trichoderma harzianum, Aspergillus niger and Azotobacter chroococcum in different combinations. This was followed by vermicomposting for 30 days. Chemical analysis of the samples showed a significant decrease in cellulose, hemicellulose and lignin contents during pre-decomposition and vermicomposting. The N, P, K content increased significantly during pre-decomposition with bioinoculants. The best quality compost, based on chemical analysis, was prepared where the substrate was treated with all the four bioinoculants together followed by vermicomposting. Results indicated that the combination of both the systems reduced the overall time required for composting and accelerated the composting of ligno-cellulosic waste during the winter season besides producing a nutrient-enriched compost product.  相似文献

A series of 1,3,5-triazine derivatives that contain 4-amino-2-trifluoromethyl-benzonitrile, 8-hydroxyquinoline, and different piperazines as substituents at the carbon atoms of the triazine ring have been synthesized by a simple and efficient synthetic protocol. The chemical structures of the compounds were elucidated with the aid of IR, 1H NMR and 13C NMR spectroscopy, and elemental analysis. The antimicrobial activity of the compounds was tested against seven bacteria (Staphylococcus aureus MTCC 96, Bacillus cereus MTCC 619, Escherichia coli MTCC 739, Pseudomonas aeruginosa MTCC 741, Klebsiella pneumoniae MTCC 109, Salmonella typhi MTCC 733, Proteus vulgaris MTCC 1771) and four fungi (Aspergillus niger MTCC 282, Aspergillus fumigatus MTCC 343, Aspergillus clavatus MTCC 1323, Candida albicans MTCC 183). The results indicate that some of the novel s-triazines have noteworthy activity in minimum inhibitory concentration as well as agar diffusion tests.  相似文献

研究了利用里氏木霉和黑曲霉混合培养产纤维素酶,以黑曲霉孢子悬浮液的不同活化浓度及不同的活化时间来寻找2个菌种发挥最大协同作用的结合点以及所产纤维素酶的水解特性。以里氏木霉单一培养和黑曲霉单一培养为参照进行对比研究。底物为农林废弃物之一的玉米秸秆,经过蒸气爆破预处理后,用作产酶C源。结果表明:黑曲霉孢子悬浮液活化浓度为10个/mL,活化时间为12 h时,滤纸酶比酶活最高,达3.32 U/mL,高于里氏木霉单一培养的2.25 U/mL,β-葡萄糖苷酶比酶活达1.32 U/mL,高于里氏木霉单一培养的0.57 U/mL。为进一步验证混合菌产纤维素酶的水解效果,利用混合菌产纤维酶的酶液及里氏木霉产纤维素酶的酶液进行酶水解实验,当酶用量为20 U/g绝干纤维素,底物质量浓度为100 g/L条件下水解48 h,混合菌所产酶液酶解得率达70.00%,高于里氏木霉所产酶液的酶解得率63.05%。实验表明里氏木霉与黑曲霉混合培养产酶是可行的,并优于单一菌种培养。  相似文献

We describe a general, simple and inexpensive method for the isolation of DNA from filamentous fungi. Starting from freeze-dried mycelium 01–015% by weight can be isolated as high molecular weight DNA suitable for restriction and ligation in 2 h. The preparation can be done in Eppendorf tubes and allows the processing of many samples in parallel. We have used the method with the basidiomycetes Phanerochaete chrysosporium, Coprinus cinereus and the ascomycete Aspergillus nidulans and others have used it with Trichoderma reesei, Aspergillus niger and for the isolation of DNA from tomato plants.  相似文献

Production of cellulase by Trichoderma reesei from dairy manure   总被引:6,自引:0,他引:6  
Cellulase production by the fungi Trichoderma reesei was studied using dairy manure as a substrate. Data showed that T. reesei RUT-C30 had higher cellulase production than T. reesei QM 9414 and that a homogenized manure, treated by a blender to reduce fiber size, led to higher cellulase production. The cellulase production was further optimized by growing T. reesei RUT-C30 on homogenized manure. The effects of manure concentration, pH, and temperature on cellulase production were investigated with optimal parameter values determined to be 10 g/l manure (dry basis), 25.5 degrees C, and pH 5.7, respectively. Elimination of CaCl2, MgSO4, nitrogen sources (NH4+ and urea) and trace elements (Fe2+, Zn2+, Co2+ and Mn2+) from the original salt solution had no negative influence on the cellulase production, while phosphate elimination did reduce cellulase production. Based on above results, the final medium composition was simplified with manure additives being KH2PO4, tween-80 and CoCl2 only. Using this medium composition and a reaction time of 6-8 days, a maximum cellulase production activity of 1.74 IU/ml of filter paper activity, 12.22 IU/ml of CMCase activity, and 0.0978 IU/ml of beta-glucosidase was obtained. This filter paper activity is the highest ever reported in cellulase production from agricultural wastes.  相似文献

Potato chips industry wastewater was collected and analyzed for biochemical oxygen demand (BOD), chemical oxygen demand (COD), total suspended solids (TSS) and total carbohydrates. Two Aspergillus species, A. foetidus and A. niger, were evaluated for their ability to grow and produce biomass and reduce the organic load of the wastewater. A. foetidus MTCC 508 and A. niger ITCC 2012 were able to reduce COD by about 60% and produce biomass 2.4 and 2.85 gl(-1), respectively. Co-inoculation of both Aspergillus strains resulted in increased fungal biomass production and higher COD reduction than in individual culture at different culture pH. pH 6 was optimum for biomass production and COD reduction. Amendment of the wastewater with different N and P sources, increased the biomass production and COD reduction substantially. Under standardized conditions of pH 6 and amendment of wastewater with 0.1% KH2PO4 and 0.1% (NH4)2 SO4, a mixed culture gave 90% reduction in COD within 60 h of incubation.  相似文献

This experiment was designed to characterize the physical, chemical and microbial properties of a standard commercial horticultural, greenhouse container, bedding plant medium (Metro-Mix 360), that had been substituted with a range of increasing concentrations (0%, 5%, 10%, 25%, 50% and 100% by volume) of pig manure vermicompost and to relate these properties to plant growth responses. The growth trials used tomatoes (Lycopersicon esculentum Mill.), grown in the substituted media for 31 days under glasshouse conditions, with seedling growth recorded in 20 pots for each treatment. Half of the tomato seedlings (10 pots per treatment) were watered daily with liquid inorganic fertilizer while the other half received water only. The percentage total porosity, percentage air space, pH and ammonium concentrations of the container medium all decreased significantly, after substitution of Metro-Mix 360 with equivalent amounts of pig manure vermicompost; whereas bulk density, container capacity, electrical conductivity, overall microbial activity and nitrate concentrations, all increased with increasing substitutions of vermicompost. The growth of tomato seedlings in the potting mixtures containing 100% pig manure vermicompost was reduced, possibly as a result of high soluble salt concentrations in the vermicompost and poorer porosity and aeration. The growth of tomato seedlings was greatest after substitution of Metro-Mix 360 with between 25% and 50% pig manure vermicompost, with more growth occurring in combinations of pig manure vermicompost treated regularly with a liquid fertilizer solution than in those with no fertilizer applied. Some of the growth enhancement in these mixtures seemed to be related to the combined effects of improved porosity, aeration and water retention in the medium and the high nitrate content of the substrate, which produced an increased uptake of nitrogen by the plant tissues, resulting in increased plant growth. When the tomato seedlings were watered daily with liquid inorganic fertilizer, substitution of Metro-Mix 360 with a very small amount (5%) of pig manure vermicompost resulted in a significant increase in the growth of tomato seedlings. Such effects could not be attributed solely to the nutritional or physical properties of the pig manure vermicompost. Therefore, it seems likely that the pig manure vermicompost provided other biological inputs, such as plant growth regulators into the container medium, that still need to be identified fully.  相似文献

Mycelia from the mushroom Marasmiellus troyanus were grown in the presence of radiolabeled benzo[a]pyrene in liquid culture. After 15 days, 8.1% of the label from M. troyanus cultures was recovered in CO2 as compared to 1.1% for Phanerochaete chrysosporium and 0.2% for Aspergillus niger. M. troyanus efficiently transformed B[a]P into water soluble metabolites with 64% of the label recovered in the water soluble fraction as compared to 11.7% for P. chrysosporium and 4.1% for A. niger. Glucuronic acid and sulfate conjugates of B[a]P were identified from the aqueous fraction of cultures of M. troyanus, after 17 days.  相似文献

The decomposition efficiency of Perionyx sansibaricus (Perrier) for vermicomposting was evaluated by using a variety of wastes such as agriculture waste, farm yard manure and urban solid waste. Vermicomposting resulted in significant increase in total N (80.8-142.3%), phosphorous (33.1-114.6%) and potassium (26.3-125.2%), whereas decrease in organic C (14.0-37.0%) as well as C:N ratio (52.4-69.8%) in different experimental beddings. P. sansibaricus showed maximum biomass production, growth rate (mg day(-1)), mean cocoon numbers, and reproduction rate (cocoon worm(-1)) in VLL (vegetable waste+leaf litter) as compared to other substrate materials. There was a consistent trend for earthworms' growth and reproduction rate, related to initial N-content of the substrate (P<0.05), but there was no clear effect of C:N ratio of the composted material on earthworm cocoon numbers and weight gain. Earthworm showed minimum total population mortality in VLL and maximum in HHCD (household waste+cow dung), after 150 days of experimentation. The increased level of plant metabolites in end product (vermicompost) and growth patterns of P. sansibaricus in different organic waste resources demonstrated the candidature of this species for wastes recycle operations at low-input basis.  相似文献

Vermitechnology was investigated as a means of reducing organic waste materials. Vermicomposting conditions were optimized to convert the biowastes to nutritious composts for amending agricultural soil. Studies were undertaken to select the most suitable earthworm species for vermicomposting, to enrich vermicompost by inoculation with beneficial microbes, to standardize an economically feasible method of vermicomposting, to achieve nutrient economy through vermicompost application in acid soils (pH 4.5), and to assess the performance of vermicompost as a bioinoculant in cow-pea, banana, and cassava. Earthworm species Eudrillus eugineae, Eisenia foetida, Perionyx sansibaricus, Pontoscolex corethrurus and Megascolex chinensis were compared for their efficiencies in biodegrading organic wastes. E. eugineae was found to be a superb agent. As a bioinoculant, vermicompost increased nitrogen and phosphorous availability by enhancing biological nitrogen fixation and phosphorous solubilisation. Vermicompost-amended acid-agriculture-soil significantly improved the yield, biometric character and quality of banana, cassava and cow-pea. Vermicompost application stimulated root growth, facilitating nutrient absorption and thereby favouring higher yield.  相似文献

设施蔬菜化肥过量施用造成土壤劣化、土传病虫害发生严重,从而导致蔬菜产量和品质下降。蚯蚓粪在改善土壤环境和促进蔬菜生长、提高产量和品质方面具有良好的效果。本试验在日光温室采用等量撒施和沟施两种方式研究了增施蚯蚓粪对土壤养分、酶活性、微生物数量及番茄生长、产量和品质的影响。结果表明: 与不施蚯蚓粪的对照相比,撒施蚯蚓粪和沟施蚯蚓粪处理均改善了土壤环境,显著增加了土壤有机质和氮、磷、钾等养分含量;显著提高了蔗糖酶和过氧化氢酶活性,增加了土壤中细菌、放线菌数量,降低了真菌数量。沟施蚯蚓粪处理明显促进了植株地上部的生长,撒施效果不显著。蚯蚓粪处理明显增强了植株根系活力,提高了叶片的光合作用和叶绿素含量,促进了植株氮和钾含量的积累。撒施和沟施蚯蚓粪处理的番茄产量比对照分别提高了22.7%和32.6%。沟施处理果实中可溶性蛋白、可溶性糖、维生素C和可滴定酸含量分别提高了66.1%、11.0%、122.6%和29.9%,硝酸盐含量降低了65.7%,而撒施处理对番茄果实品质提升效果不明显。  相似文献

A physico-chemical and structural characterization of three 1,4-beta-D-glucan cellobiohydrolases (EC. 3.2.1.91), isolated from a culture filtrate of the white-rot fungus Phanerochaete chrysosporium, reveals that the cellulolytic enzyme secretion pattern and thus the general degradation strategy for P. chrysosporium is similar to that of Trichoderma reesei. Partial sequence data show that two of the isolated enzymes, i.e., CBHI, pI 3.82 and CBH62, pI 4.85, are homologous with CBHI and EGI from T. reesei; while, the third, i.e., CBH50, pI 4.87, is homologous to T. reesei CBHII. Limited proteolysis with papain cleaved each of the three enzymes into two domains: a core protein which retained full catalytic activity against low molecular weight substrates and a peptide fragment corresponding to the cellulose binding domain, in striking similarity to the structural organization of T. reesei. CBHI and CBH62 have their binding domain located at the C-terminus, whereas in CBH50 it is located at the N-terminus. It is evident that synergistically acting cellobiohydrolases is a general requirement for efficient hydrolysis of crystalline cellulose by cellulolytic fungi.  相似文献

研究长期小麦连作施肥条件下土壤微生物量碳、氮,土壤呼吸的变化及其与土壤养分的相关性。以陕西长武长期定位试验为平台,应用氯仿熏蒸-K2SO4提取法、碱液吸收法和化学分析法分析了长达26a不同施肥处理农田土壤微生物量碳、微生物量氮和土壤呼吸之间的差异及其调控土壤肥力的作用。长期施肥及种植作物,均能提高土壤微生物量碳、氮含量,尤其是施用有机肥,土壤微生物量碳、氮含量高于单施无机肥的处理,土壤呼吸量也提高15.91%—75.73%,而施用无机肥对于土壤呼吸无促进作用。土壤微生物生物量碳氮、土壤呼吸与土壤有机质、全氮呈极显著相关。长期有机无机肥配施可以提高土壤微生物量碳氮、土壤呼吸,氮磷肥与厩肥配施对提高土壤肥力效果最好。微生物量碳氮及土壤呼吸可以反映土壤质量的变化,作为评价土壤肥力的生物学指标。  相似文献

The post-harvest residues of some local crops, e.g., wheat (Triticum aestivum), millets (Penniseum typhoides and Sorghum vulgare), and a pulse (Vigna radiata) were subjected to recycle through vermicomposting. The crop residues were amended with animal dung, and three types of vermibeds were prepared: (i) millet straw (S. vulgare + P. typhoides in equal quantity) + sheep manure (1: 2 ratio) (MS); (ii) pulse bran (V. radiata) + wheat straw (T. aestivum) + cow dung (1: 1: 2 ratio) (PWC); and (iii) mixed crop residues (mixing of all types crop residues, used in this study) + cow dung in 1:1 ratio (MCR + CD). The fourth treatment was cattle shed manure (CSM). Vermicomposting resulted in a significant increase in total N (97.3% to 155%), available P (67.5% to 123.5%), exchangeable K (38.3% to 112.9%), and exchangeable Ca (23.3% to 53.2%), and decrease in organic C content (20.4% to 29.0%) in the different vermibeds. The earthworm showed the higher biomass gain, growth rate (mg wt. worm?1 day?1) and cocoon numbers in the CSM vermibed. The quality of crop residues was directly related to the rate of organic matter mineralization during the vermicomposting. This study suggests that agriculture wastes could be converted into some value-added products, e.g., vermicompost and worm biomass through vermicomposting technology. The higher concentrations of plant nutrients in end products indicate a potential for using agriculture wastes in sustainable crop production.  相似文献

The relationships between vermicompost chemical features, enzyme activities, community-level physiological profiles (CLPPs), fungal community structures, and its microbial respiratory response to pesticides were investigated. Fungal community structure of vermicomposts produced from damaged tomato fruits (DT), winery wastes (WW), olive-mill waste and biosolids (OB), and cattle manure (CM) were determined by denaturing gradient gel electrophoresis of 18S rDNA. MicroResp? was used for assessing vermicompost CLPPs and testing the microbial response to metalaxyl, imidacloprid, and diuron. Vermicompost enzyme activities and CLPPs indicated that WW, OB, and DT had higher microbial functional diversity than CM. The microbiota of the former tolerated all three pesticides whereas microbial respiration in CM was negatively affected by metalaxyl and imidacloprid. The response of vermicompost microbiota to the fungicide metalaxyl was correlated to its fungal community structure. The results suggest that vermicomposts with higher microbial functional diversity can be useful for the management of pesticide pollution in agriculture.  相似文献

Hydrolysis of gallotannin to gallic acid by immobilized mycelia of Aspergillus niger MTCC 282, Aspergillus fischerii MTCC 150, Fusarium solani MTCC 350 and Trichoderma viride MTCC 167 in a packed bed bioreactor was studied. Fungal mycelia preinduced with 5 g L-1 gallotannin were immobilized in calcium alginate gel (1.5%) and the resultant beads were packed in a column to a bed volume of 175 mm3. Gallotannin dissolved in distilled water was passed through the column and the eluate was recycled after adjusting pH to 6 with ammonium hydroxide (10%). Maximum hydrolysis of gallotannin was recorded by immobilized mycelia of F. solani and T. viride at 35 degrees and 45 degrees C after 175 and 60 min of residency period respectively. Optimum substrate concentration required for maximum hydrolysis was 10 g L-1 at pH 5 for both the fungi. Immobilized mycelia of A. niger and A. fischerii revealed maximum operational stability. Loss of activity after eighth run was in the order of-A. niger (no loss), A. fischerii (7.5%), F. solani (18%) and T. viride (18%). Stability in terms of retention of enzyme activity after 150 days of storage at 4 degrees C was A. niger (58%), A. fischerii (26.8%), F. solani (83%) and T. viride (85.1%).  相似文献

A novel microbial phospholipase C (PLC) from Bacillus licheniformis MTCC 7445 was purified to homogeneity by ammonium sulphate fractionation, dialysis, anion exchange chromatography and gel exclusion chromatography. The bacteria growing on vegetable oils secreted significantly high amount of PLC. The enzyme was purified to 23.4-fold with 46% recovery and specific activity 398 U/mg. It exhibited optimum activity at 70°C and pH 10.0. Using diphosphatidylglycerol as substrate the PLC of B. licheniformis MTCC 7445 had a V max and K m of 0.68 mM/min and 32 mM, respectively. It hydrolyzed phosphatidylinositol and phosphatidylserine as well as phosphatidylcholine but not other glycerophospholipids. Its activity was enhanced by 113% with Mn2+ and 110% with Mg2+. During degumming of vegetable oils with this enzyme preparation, the phosphorus content of the oil became lower than 4 mg/kg after 5 h of enzyme treatment at 40°C. The novel PLC from B. licheniformis MTCC 7445 is potentially useful for the refining of high quality oils with 95% removal of phospholipids with attractive yield.  相似文献

Commercial cellulase enzymes have been used in the food, detergent, and textile industries, and are potentially effective for processing biomass feedstocks. A survey was undertaken to identify major manufacturers/distributors of cellulases in the USA and to evaluate 13 representative commercial preparations for enzyme activity, protein concentration, and chemical composition. Samples were subjected to activity measurements using filter paper, carboxymethylcellulose, cellobiose, and p-nitrophenyl-β-d-glucopyranoside as substrates. To ascertain the microbial origin of the commercial preparations, Western blots utilizing monoclonal antibodies specific for Trichoderma reesei CBH I and Aspergillus niger β-d-glucosidase were developed. Eleven of the cellulases tested were of T. reesei or T. viride origin and two were from A. niger.  相似文献
