Prochlorococcus is a ubiquitous marine oxyphotobacterium characterized by the presence of DV-chl a and b . In addition, the type strain Prochlorococcus marinus Chisholm et al. CCMP 1375 (or SS120), an isolate from the Sargasso Sea, contains low levels of an unusual phycoerythrin. Until now, it has been unclear if phycoerythrin occurs randomly within this systematic group and if the molecular characteristics of this phycoerythrin are restricted to this single strain. Here, we show that two additional Prochlorococcus strains from the Pacific Ocean also contain similar low levels of phycoerythrin. DNA sequence and phylogenetic analyses demonstrated that this phycoerythrin is very similar to the phycoerythrin of P. marinus SS120 and differs from the classic cyanobacterial phycoerythrins. In contrast, a third isolate from the Arabian Sea lacks phycoerythrin. Based on the DV-chl b:a ratio and 16S rRNA sequence data, we classify the two Pacific phycoerythrin-containing isolates as low-light-adapted strains and the Arabian Sea isolate as a high-light-adapted strain. Thus, we provide further evidence to link the physiology of an individual genotype and the presence or absence of functional phycoerythrin genes within the genus Prochlorococcus .  相似文献

Circadian periodicity in cell division and death was investigated in the cyanobacterium Anabaena flos-aquae (Lyngb.) Bréb in a phosphorus (P)-limited, N2-fixing chemostat culture. When entrained under 12:12 h LD cycles, not only cell division but also cell death showed a clear circadian rhythm in this filamentous cyanobacterium. The rhythm persisted under continuous light and was temperature compensated. Circadian rhythm was clearly observed in the steady-state cell number and instantaneous growth rate, μ(t), which reached a maximum at about 2 h before sunset and a minimum at about 2 h before sunrise. The number of dead cells and the instantaneous death rate γ(t) also showed a circadian periodicity; the peak of γ(t) occurred approximately 8 h before that of μ(t). Therefore, cell growth and death in A. flos-aquae appear to be under the control of circadian clocks, and thus it seems that their death is programmed cell death.  相似文献

Nine axenic microalgal (Chlorophyta) strains from three genera (Protococcus, Chlorella, and Scenedesmus) were analyzed for endogenous cytokinins. Cytokinin‐like activity was detected using the excised cucumber cotyledon bioassay. Five strains showed no cytokinin‐like activity and four strains, low cytokinin‐like activity. Ethanolic extracts of the microalgae containing a mixture of deuterium‐labeled standards were purified using a combined DEAE‐Sephadex octadecysilica column and immunoaffinity column based on wide‐range specific mon‐oclonal antibodies and analyzed by HPLC linked to a micromass single quadrupole mass spectrometer with an electrospray interface and a photodiode array detector. There were similar trends in cytokinin profiles for the nine microalgal strains investigated, although concentrations did vary. Both isopentenyladenine and isopentenyladenosine were detected in all nine strains. cis‐Zeatin and cis‐zeatin riboside occurred at higher concentrations than the trans isomers, whereas trans‐zeatin‐O‐glucoside and trans‐zeatin riboside‐O‐glucoside were dominant over the cis isomers. Dihydrozeatin and its conjugates were not detected in any significant amounts. The aromatic benzyladenine always occurred at higher concentrations than benzyladenosine. The topolins were well represented with all three isomers (ortho, meta, and para) being detected, with ortho‐topolin and ortho‐topolin riboside occurring at higher concentrations than the other isomers. However, for the O‐glucosides, the meta isomers (meta‐topolin‐O‐glucoside and meta‐topolin riboside‐O‐glucoside) occurred at higher concentrations than the other isomers. No N‐glucosides were detected (isopentenyladenine‐9‐glucoside, zeatin‐9‐glucoside, dihydrozeatin‐9‐glucoside, benzyladenine‐9‐glucoside, ortho‐topolin‐9‐glucoside, and meta‐topolin‐9‐glucoside). Generally, zeatin and topolin conjugates were the dominant forms of isoprenoid and aromatic cytokinins, respectively. There was no distinct trend in the proportions of isoprenoid to aromatic cytokinins.  相似文献

Raphidiopsis brookii P. J. Hill (cyanobacteria) was isolated from a small subtropical eutrophic pond (Biguá Pond) located in the grounds of Rio Grande University in the extreme south of Brazil, following a toxic bloom of this species. Growth, saxitoxin production, and release of carbohydrates and protein were monitored at three sodium nitrate concentrations (500, 1,000, and 1,500 μM), from inoculation up to the stationary growth phase. Growth was monitored by determining the biovolume, chl content, and trichome count. Growth was better described in terms of biovolume and chl measurements, because trichome fragmentation was observed to increase at the stationary growth phase. Carbohydrates and proteins were released in small amounts during most of the experiment, with a significant increase during the stationary phase. Extracellular polysaccharides were essentially composed of glucose, galactose, N‐acetyl‐glucosamine, mannose, xylose, rhamnose, arabinose, and fucose. The relative proportions of these units showed no significant variation during growth. Small quantities of extracellular free carbohydrates were also detected, and only fucose was released in significant amounts at the lowest nitrate concentration (500 μM). R. brookii produced both saxitoxin and dc‐saxitoxin, the former at four times the rate of the latter. This was the first study demonstrating saxitoxin production and the release of both carbohydrate and protein by R. brookii.  相似文献

The filamentous diazotrophic cyanobacterium Nodularia forms water blooms each year in the Baltic Sea. Filaments isolated from such water blooms vary in their trichome width, degree of coiling, and properties of their gas vesicles; previously, these characters have been used to classify individuals to species level. To test the validity of such a phenotypic classification, we determined the nucleotide sequences for a region of the phycocyanin locus that includes a noncoding intergenic spacer (PC-IGS), the IGS between two adjacent copies of the gvpA gene (which encodes the main structural gas vesicle protein) and the rDNA internal transcribed spacer (rDNA-ITS), for 13 clonal Nodularia isolates from the Baltic Sea during August 1994. The complete 16S-rDNA sequence was determined for three isolates and was found to be identical in each of them. Molecular sequences for noncoding regions of the genome were used to assign isolates to three groups on the basis of PC-IGS, two groups on the basis of gvpA -IGS, and three groups on the basis of rDNA-ITS. No consistent correlation was found between genotype and any of the phenotypic features examined, and no link was found between any of these features themselves, indicating that these characters are not useful for placing Nodularia isolates into meaningful taxonomic groups. The PC-IGS, gvpA -IGS, and rDNA-ITS genotypic groupings were not congruent. This might indicate that gene flow occurs between individuals in Nodularia populations.  相似文献

A simple, sensitive assay method for the isolation and enumeration of sparse populations of cyanobacteria in an estuarine system is described. The method, based on the standard membrane-filter plate count technique, differentiates between viable and nonviable cells. It was found that an estuarine water-based agar medium was the most suitable medium for isolation of cyanobacteria. Because of the restricted nature of colony development, isolation of individual species is easily accomplished.  相似文献

The taxonomy and toxicity of a single‐filament isolate from a filamentous cyanobacterial bloom in Lake Hakanoa (New Zealand) were examined by microscopy and liquid chromatography–mass spectrometry. Based on a morphological examination of environmental and cultured material, strain CAWBG02 was identified as Raphidiopsis mediterranea Skuja; however, subsequent phylogenetic analysis of the 16S rRNA gene sequence demonstrated that CAWBG02 was most likely to be a single culture of Aphanizomenon issatschenkoi (Usacev) Proshkina‐Lavrenko. Toxin testing confirmed that the original bloom and A. issatschenkoi isolate produced anatoxin‐a but did not produce homoanatoxin‐a or any cylindrospermopsins, saxitoxins, or microcystins. Despite the absence of cylindrospermopsin production, genes implicated in the biosynthesis of cylindrospermopsin were successfully amplified from A. issatschenkoi strain CAWBG02. To our knowledge, this is the first confirmation of an anatoxin‐a‐producing species in the Southern Hemisphere and the first report of anatoxin‐a production by A. issatschenkoi.  相似文献

2012年4月和10月在江西省鄱阳湖进行野外调查时, 发现一种浮游的丝状蓝藻。通过分离纯培养, 获得了6个纯化藻株。根据藻株主要形态学特征及其16S rRNA基因序列与比较, 鄱阳湖这些藻株与老挝的2株Aerosakkonema funiforme较为相近, 其中16S rRNA基因序列的相似度达到98%。鉴于这些藻株不同于颤藻目中其他属的藻株, 因此确定为我国一水华蓝藻新记录属气丝藻属Aerosakkonema Nanda Watanabe 2012, 模式种为索状气丝藻Aerosakkonema funiforme Nanda Watanabe 2012。    相似文献

Endogenous cytokinins were quantified in synchronized Chlorella minutissima Fott et Novákova (MACC 361) and Chlorella sp. (MACC 458) grown in a 14:10 light:dark (L:D) photoperiod. In 24 h experiments, cell division occurred during the dark period, and cells increased in size during the light period. Cytokinin profiles were similar in both strains, consisting of five cis‐zeatin (cZ) and three N6‐(2‐isopentenyl)adenine (iP) derivatives. Cytokinin concentrations were low during the dark period and increased during the light period. In 48 h experiments using synchronized C. minutissima (MACC 361), half the cultures were maintained in continuous dark conditions for the second photoperiod. Cell division occurred during both dark periods, and cells increased in size during the light periods. Cultures kept in continuous dark did not increase in size following cell division. DNA analysis confirmed these results, with cultures grown in light having increased DNA concentrations prior to cell division, while cultures maintained in continuous dark had less DNA. Cytokinins (cZ and iP derivatives) were detected in all samples with concentrations increasing over the first 24 h. This increase was followed by a large increase, especially during the second light period where cytokinin concentrations increased 4‐fold. Cytokinin concentrations did not increase in cultures maintained in continuous dark conditions. In vivo deuterium‐labeling technology was used to measure cytokinin biosynthetic rates during the dark and light periods in C. minutissima with highest biosynthetic rates measured during the light period. These results show that there is a relationship between light, cell division, and cytokinins.  相似文献

塔里木盆地塔北隆起轮南46井、英买1、2井奥陶纪石灰岩内含有大量的钙藻化石、蓝细菌以及疑难微体化石。这些钻井岩芯均在深达5000-6200m的地下深部取得。钙藻主要为绿藻类的Dasyporella,Ver‘miporella,Moniliporella以及?Plexa;红藻类的管孔藻类Solenoporaceans;钙化蓝细菌则有Girvanella,Botomaella,?Subtifloria等;疑难微体化石有Bevocastria,Nuia,Rothpletzella。这些钙藻生活于热带或亚热带正常盐度的浅海水内,其水深不到20m。世界各地的奥陶纪Vermiporella均位于古赤道的两侧,这表明它们是在气候炎热、温暖海水中生活的一类海洋藻类。Girvanella以藻灰结核和内碎屑最为常见,某些球粒可能代表Girvanella破碎后形成的单管或小棒。塔里木盆地钙藻植物群相似于哈萨克斯坦、波罗的海周围地区以及北美同时代植物群,这表明这些钙藻和蓝细菌化石具有遍布于全球的性质。塔北隆起早、中奥陶世沉积属于典型的碳酸盐岩台地相沉积。到晚奥陶世时,碳酸盐岩沉积被浅水陆棚沉积所取代,以陆源碎屑岩为主,夹少量的碳酸盐岩。  相似文献

Aphanizomenon Morren is an important member of the cyanobacterial community in the Baltic Sea, but studies of this genus have been hampered by the difficulty of growing it in laboratory culture. PCR amplification of DNA from colonies picked directly from water samples has circumvented this problem and made it possible to carry out an analysis of genetic diversity within the Baltic Sea and in two small North American lakes separated by just a few kilometers. The nucleotide sequence of the phycocyanin intergenic spacer and partial flanking coding regions of cpcB and cpcA was determined for 32 colonies of Aphanizomenon , 26 from the Baltic Sea, and 6 from the North American lakes. No variation was detected among the 26 Baltic Sea colonies, but two alleles, differing at 19 nucleotide positions (5.4%), were found in the North American lake colonies. Surprisingly, the two North American types were less closely related to each other than to the Baltic Sea genotype. The Baltic Sea Aphanizomenon is clearly distinct from A. flos-aquae at both the cpcB–cpcA and 16S rDNA loci, which lends phylogenetic support to their tentative separation based on ultrastructural analysis. We conclude that although there is significant genetic diversity in the genus Aphanizomenon , the population in the Baltic Sea is, in contrast to the Nodularia population, genetically homogeneous.  相似文献

A strain of Raphidiopsis (Cyanobacteria) isolated from a fish pond in Wuhan, P. R. China was examined for its taxonomy and production of the alkaloidal hepatotoxins cylindrospermopsin (CYN) and deoxy-cylindrospermopsin (deoxy-CYN). Strain HB1 was identified as R. curvata Fritsch et Rich based on morphological examination of the laboratory culture. HB1 produced mainly deoxy-CYN at a concentration of 1.3 mg·g 1 (dry wt cells) by HPLC and HPLC-MS/MS. CYN was also detected in trace amounts (0.56 μg·g 1). A mouse bioassay did not show lethal toxicity when tested at doses up to 1500 mg dry weight cells·kg 1 body weight within 96 h, demonstrating that production of primarily deoxy-CYN does not lead to significant mouse toxicity by strain HB1. The presence of deoxy-CYN and CYN in R. curvata suggests that Raphidiopsis belongs to the Nostocaceae, but this requires confirmation by molecular systematic studies. Production of these cyanotoxins by Raphidiopsis adds another genus, in addition to Cylindrospermopsis , Aphanizomenon , and Umezakia , now known to produce this group of hepatotoxic cyanotoxins. This is also the first report from China of a CYN and deoxy-CYN producing cyanobacterium.  相似文献

The evolutionary relationships of cultivated psychrophilic and psychrotolerant polar oscillatorians were examined, based on small subunit rDNA sequences. Psychrophilic oscillatorians from the Antarctic were affiliated in one well-supported clade, which also includes two Arctic strains. Two of the Antarctic psychrophiles contain an 11-nucleotide insertion that is identical to, and previously only described in, an Arctic oscillatorian. The psychrotolerant phenotype, conversely, has arisen multiple times in the cyanobacterial lineage, and psychrotolerant strains are sometimes most closely related to organisms of temperate latitudes. These findings support the hypothesis that oscillatorians in both polar regions originated from more temperate species. Furthermore, morphological designations of these filamentous cyanobacteria often do not have phylogenetic significance.  相似文献

In the North Atlantic over a wide geographic region that includes various oceanic regimes and a temperature range from 10 to 22° C, an increase in the number of nondividing Synechococcus cells (X) was generally accompanied by a greater-than-proportional increase in the number of dividing cells (Y). As a result, the fraction of dividing cells (FDC = Y · (Y + X)?1) was positively related to population size (Y + X). Recognizing that FDC is generally greater in a rapidly growing population than in a slowly growing one, our empirical finding implies a positive correlation between specific growth rate and standing stock for Synechococcus. One notable exception occurred during winter (T < 5°C) in a eutrophic coastal embayment when a decrease in cell abundance was not matched by a decrease in FDC.  相似文献

There is still a great deal of debate about whether cyanobacteria contain vacuoles. This might in part reflect our limited ability to isolate vacuoles. We found and isolated vacuoles from different cyanobacteria during spheroplast preparation. Lysozyme treatment induced two kinds of spheroplasts: vacuolated spheroplasts and nonvacuolated spheroplasts. Upon breakage in distilled water, vacuolated spheroplasts released transparent, spherical, and colorless vacuoles with diameters ranging from 2.3 to 16 μm. Large vacuoles could be generated by fusion of two or three small vacuoles. Additionally, large vacuoles also could engulf small ones or other cellular bodies. The isolated vacuoles could tolerate hypotonic condition, and some could be drawn into a thread. Nonvacuolated spheroplasts released few vacuoles after breaking apart. This successful confirmation and isolation of vacuoles will allow studies of the origin and function of cyanobacterial vacuoles.  相似文献

Oscillatorian cyanobacteria dominate benthic microbial mat communities in many polar freshwater ecosystems. Capable of growth at low temperatures, all benthic polar oscillatorians characterized to date are psychrotolerant (growth optima > 15° C) as opposed to psychrophilic (growth optima ≤ 15° C). Here, psychrophilic oscillatorians isolated from meltwater ponds on Antarctica's McMurdo Ice Shelf are described. Growth and photosynthetic rates were investigated at multiple temperatures, and compared with those of a psychrotolerant isolate from the same region. Two isolates showed a growth maximum at 8° C, with rates of 0.12 and 0.08 doublings·d ? 1, respectively. Neither displayed detectable growth at 24° C. The psychrotolerant isolate showed almost imperceptible growth at 4° C and a rate of 0.9 doublings·d ? 1 at its optimal temperature of ~23° C. In both photosynthesis versus irradiance and photosynthesis versus temperature experiments, exponentially growing cultures were acclimated for 14 days at 3, 8, 12, 20, and 24° C under saturating light intensity, and [14C] photoincorporation rates were measured. Psychrophilic isolates acclimated at 8° C showed greatest photosynthetic rates; those acclimated at 3° C were capable of active photosynthesis, but photoincorporation was not detected in cells acclimated at 20 and 24° C, because these isolates were not viable after 14 days at those temperatures. The psychrotolerant isolate, conversely, displayed maximum photosynthetic rates at 24° C, though photoincorporation was actively occurring at 3° C. Within acclimation temperature treatments, short‐term photosynthetic rates increased with increasing incubation temperature for both psychrophilic and psychrotolerant isolates. These results indicate the importance of temperature acclimation before assays when determining optimal physiological temperatures. All isolates displayed photosynthetic saturation at low light levels (<128 μmol·m ? 2·s ? 1) but were not photoinhibited at the highest light treatment (233 μmol·m ? 2·s ? 1). Field studies examining the impact of temperature on photosynthetic responses of intact benthic mats, under natural solar irradiance, showed the mat communities to be actively photosynthesizing from 2 to 20° C, with maximum photoincorporation at 20° C, as well as capable of a rapid response to an increase in temperature. The rarity of psychrophilic cyanobacteria, relative to psychrotolerant strains, may be due to their extremely slow growth rates and inability to take advantage of occasional excursions to higher temperatures. We suggest an evolutionary scenario in which psychrophilic strains, or their most recent common ancestor, lost the ability to grow at higher temperatures while maintaining a broad tolerance for fluctuations in other physical and chemical parameters that define shallow meltwater Antarctic ecosystems.  相似文献
