The Tumor Necrosis Factor-alpha (TNF-α) is a pleiotropic cytokine that produces different stimuli in various physiological and pathological conditions. TNF-α contributes importantly to the development of T cells, B cells, and dendritic cells. However, TNF-α is also a potent inflammatory mediator and apoptosis inducer. The significance of the TNF-α involvement in the pathogenesis of systemic lupus erythematosus (SLE) remains controversial. From the genetic standpoint, a number of studies suggest that the TNF-α gene polymorphism is involved in the susceptibility of SLE. Moreover, there is a close association between the TNF-α gene expression and clinical manifestations. In addition, the increased serum level of TNF-α is observed in SLE patients and associated with disease activity and certain systemic manifestations. Treatment with anti-TNF agents is, however, controversial in SLE since induction of antinuclear antibodies, anti-dsDNA, anticardiolipin antibodies, and cases of drug-induced lupus have been observed in rheumatoid arthritis patients. In this context, this study reviewed the importance of TNF-α in the pathogenesis of SLE.  相似文献

Concanavalin A and anti-α-d-glucose antibodies form precipitin complexes with antigens havingα-d-glucose as terminal units. The sedimentation rates, molecular weights, gel electrophoretic mobilities, isoelectric points, and immunoglobulin type of Con A andα-Ab have been determined. The interactions of the compounds with antigens in the presence of potential inhibitors have been compared. The data show that the interaction of Con A with glucose units occurs with hydrogen bonding at hydroxyl groups at C1, 3,4, and 6 and van der Waals bonding at the pyranose ring oxygen. In theα-Ab complex with glucose units, in addition to the above bond types, a hydrogen bond at the hydroxyl at C2 occurs and this bond is essential for interaction.  相似文献

Transforming growth factor-β2 (TGF-β2) is found in increasing amounts in aqueous humor and reactive optic nerve astrocytes of patients with primary open-angle glaucoma (POAG), a major cause of blindness worldwide. The available data strongly indicate that TGF-β2 is a key player contributing to the structural changes in the extracellular matrix (ECM) of the trabecular meshwork and optic nerve head as characteristically seen in POAG. The changes involve an induction in the expression of various ECM molecules and are remarkably similar in trabecular meshwork cells and optic nerve head astrocytes. The ECM changes in the trabecular meshwork most probably play a role in the increase of aqueous humor outflow resistance causing higher intraocular pressure (IOP). In the optic nerve head, TGF-β2-induced changes might contribute to deformation of the optic nerve axons causing impairment of axonal transport and neurotrophic supply and leading to their continuous degeneration. The increase in IOP further adds mechanical stress and strain to optic nerve axons and accelerates degenerative changes. In addition, high IOP might induce the expression of activated TGF-β1 in trabecular meshwork cells and optic nerve head astrocytes; this again might significantly lead to the progress of axonal degeneration. The action of TGF-β2 in POAG is largely mediated through the connective tissue growth factor, whereas the activities of TGF-β1 and -β2 are modulated by the blocking effects of bone morphogenetic protein-4 (BMP-4) and BMP-7, by gremlin that inhibits BMP signaling and by several species of microRNAs.  相似文献

X-ray fluorescence microscopy was applied for two-dimensional elemental analysis of substantia nigra (SN) tissue. The samples representing Parkinson’s disease (PD) and control cases were examined at HASYLAB beamline L and at ESRF beamline ID22. Two-dimensional mapping of P, S, Cl, K, Ca, Fe, Cu, Zn, Se and Br was done with the spatial resolution of 15 and 5 μm. The masses per unit area of elements in neuromelanin reach nerve cells of SN were determined.The elemental data were processed using two multivariate techniques, namely cluster and discriminant analysis. The statistical methods were used for data reduction, both unsupervised and supervised classification as well as for the creation of a model that would simplify case identification based on the elemental analysis of SN tissue. The results of cluster analysis confirmed the statistical significance of the differences in elemental composition of PD and control SN nerve cells. Based on the results of discriminant analysis, the elements (P, Cl, Fe, Cu and Zn) that played the greatest role in the process of differentiation between neurons from examined groups were determined.  相似文献

Nonalcoholic fatty liver disease (NAFLD) is associated with hepatic steatosis, inflammation and liver fibrosis and has become one of the leading causes of hepatocellular carcinoma and liver failure. However, the underlying molecular mechanism of hepatic steatosis and the progression to nonalcoholic steatohepatitis (NASH) are not fully understood. Herein, we discovered that AMPKα2 catalytic subunit showed reduced expression in the liver following high fat diet (HFD) feeding to mice. Importantly, knockout of AMPKα2 in mice aggravated NAFLD, hepatic steatosis, inflammation and fibrosis. On the other hand, hepatocyte-targeted overexpression of AMPKα2 prevented or reversed NAFLD indications. In vivo mechanistic studies revealed that increased phosphorylation of IKKα/β and NF-κB in HFD-fed AMPKα2−/− mice compared to WT mice, and treatment of these mouse cohorts with an inhibitor of NF-κB signaling for 4 weeks, effectively attenuated the progression of steatohepatitis and metabolic disorder features. In summary, AMPKα2 provides a protective role in the process of hepatic steatosis to NASH progression through suppression of liver NF-κB signaling.  相似文献

The mechanism by which the fibroblast is able to trigger palmar fibromatosis is still not yet fully understood. It would appear certain that the “abnormal” fibroblasts continuously synthesise profibrotic cytokines which are able to determine the activation to myofibroblasts, to stimulate them to the further proliferation and synthesis of other cytokines, to modify the cells’ differentiation and ultrastructural characteristics, as well as the production of matrix and other proteins. Several fibroblast growth factors have been suggested to be responsible of an abnormal cell activation with an aberrantly elevated collagen synthesis and extracellular deposition in Dupuytren’s disease, as TGF-Beta, TNF-Alfa, PDGF, GM-CSF, free radicals, metalloproteinases, sex hormones, gene modified expression, mechanical stimulation. Although renal biopsy is the most accurate way of assessing renal inflammation in patients with lupus nephritis (LN), the technique is invasive and cannot be performed frequently. Currently used blood and urine biomarkers have limited utility in monitoring the activity of nephritis. In a previous issue of Arthritis Research and Therapy, Singh and colleagues showed that measuring urinary levels of vascular cell adhesion molecule 1 could be useful in both diagnosing and monitoring LN. These levels are higher in patients with lupus than controls, are higher in lupus patients who have active renal disease compared with those who do not, and correlate significantly with the histological activity index in renal biopsies of patients with LN.Lupus nephritis (LN) is one of the most severe forms of systemic lupus erythematosus (SLE). If not treated adequately, the disease can result in renal failure or death. In its early stages, LN may be almost asymptomatic and picked up only by carrying out routine blood and urine tests. Measurements of urine protein are especially helpful in this regard. In a previous issue of Arthritis Research and Therapy, Singh and colleagues described tests for three different urinary biomarkers in patients with SLE, investigating which is likely to be most helpful for monitoring renal disease activity [1].Treatment of LN was revolutionized by the introduction of combined corticosteroid/cyclophosphamide regimes and has advanced more recently with the replacement of cyclophosphamide by mycophenolate in many cases [2]. Early diagnosis and treatment has a major impact on clinical outcomes in patients with LN [3], but can also cause treatment-related adverse effects. Being able to identify exactly when treatment is needed during the course of the disease and when maintenance treatment needs to be increased is therefore important. The most accurate way to assess nephritis in patients with SLE is by carrying out a renal biopsy. The histological type of nephritis can be defined and the degree of inflammation can be quantified using an activity index [4]. A high activity index signifies active yet reversible disease, whereas the chronicity index shows irreversible damage.Renal biopsy is an invasive procedure, however, which cannot be repeated whenever a flare of renal lupus is suspected. We would therefore like to have biomarkers, measurable in blood or urine, which rise and fall with renal disease activity and are closely associated with the degree of inflammation in the kidney. Blood markers such as anti-double-stranded DNA, anti-nucleosome and anti-α-actinin antibodies have been studied. There is some evidence that increases in these markers are associated with renal disease activity [5], as measured by indices such as blood albumin and urine protein, but very little evidence for their association with renal biopsy scores.Given that the kidney is the main site of inflammation in LN, however, biomarkers in urine may reflect this inflammation more closely than those in the blood. There has been some interest in using urinary neutrophil gelatinase-associated lipocalin as a marker of renal inflammation in SLE. Studies have shown that urinary (but not serum) neutrophil gelatinase-associated lipocalin levels correlate with measures of renal disease activity and that a rise in urinary neutrophil gelatinase-associated lipocalin at one visit predicts flare of nephritis at the next visit [6,7]. These studies were cross-sectional and did not look at the association between urinary neutrophil gelatinase-associated lipocalin and renal histology.The current paper by Singh and colleagues is also a cross-sectional study but includes data on renal histology and compares three different urinary assays; CXCL16, monocyte chemoattractant protein 1 (MCP-1) and vascular cell adhesion molecule 1 (VCAM-1) [1]. Since all three of these molecules can play a role in the recruitment of inflammatory cells to the nephritic kidney, it would be entirely reasonable to predict that their levels would rise in the urine when active renal inflammation is at its height. The authors review both clinical and murine evidence supporting this prediction for each of these markers [8]. In this study they report creatinine-normalized urinary CXCL16, MCP-1 and VCAM-1 levels in 73 patients with SLE, 13 healthy volunteers and 22 patients with other forms of glomerulonephritis.The study results showed some evidence of utility for all three of these urinary markers. All markers were elevated in patients with SLE compared to healthy controls and correlated with the level of proteinuria. However, the comparison with healthy controls is complicated by the fact that 92% of these controls were Asian whereas none of the patients were. Disease activity in the study was assessed using the Systemic Lupus Erythematosus Disease Activity Index and correlated with MCP-1 and VCAM-1 but not CXCL16. When patients were defined as having active or inactive renal disease, on the basis of the renal components of the Systemic Lupus Erythema tosus Disease Activity Index score alone, MCP-1 and VCAM-1 but not CXCL16 distinguished those two groups. However, this result was based on a relatively small number of patients with inactive renal disease (n = 14), probably because there was a low threshold for considering a patient to have active renal disease - being positive for any single criterion of hematuria, pyuria, proteinuria or casts was sufficient. This difficulty in defining active LN based on disease activity measures is common to many papers of this type and is a major reason why the availability of renal biopsy data from the date of the urine sample in 24 of these patients is so important. These histological data provide a direct objective measure of nephritis and particularly support the utility of urinary VCAM-1 as a biomarker, since only VCAM-1 was significantly correlated with the activity index and also differentiated class IV LN from the other histological types.This study thus provides compelling evidence that measuring urinary VCAM-1 could be an important new biomarker in patients with LN, and some evidence supporting urinary CXCL16 and MCP-1. Investigation into whether an index combining the values of all three markers, along the lines of the serum chemokine index reported by Bauer and colleagues [9], would have more potential to distinguish active LN from inactive LN would be interesting. As suggested by the authors, however, the most important thing is to study levels of these urinary markers longitudinally in patients with LN to see whether they predict changes in activity of nephritis in individual patients over time.  相似文献

TRPM2 is a Ca2+-permeable non-selective cation channel that can be activated by adenosine dinucleotides, hydrogen peroxide, or intracellular Ca2+. The protein is expressed in a wide variety of cells, including neurons in the brain, immune cells, endocrine cells, and endothelial cells. This channel is also well expressed in β-cells in the pancreas. Insulin secretion from pancreatic β-cells is the primary mechanism by which the concentration of blood glucose is reduced. Thus, impairment of insulin secretion leads to hyperglycemia and eventually causes diabetes. Glucose is the principal stimulator of insulin secretion. The primary pathway involved in glucose-stimulated insulin secretion is the ATP-sensitive K+ (KATP) channel to voltage-gated Ca2+ channel (VGCC)-mediated pathway. Increases in the intracellular Ca2+ concentration are necessary for insulin secretion, but VGCC is not sufficient to explain [Ca2+]i increases in pancreatic β-cells and the resultant secretion of insulin. In this review, we focus on TRPM2 as a candidate for a [Ca2+]i modulator in pancreatic β-cells and its involvement in insulin secretion and development of diabetes. Although further analyses are needed to clarify the mechanism underlying TRPM2-mediated insulin secretion, TRPM2 could be a key player in the regulation of insulin secretion and could represent a new target for diabetes therapy.  相似文献

Alcoholism has complex etiology and there is evidence for both genetic and environmental factors in its pathophysiology. Chronic, long-term alcohol abuse and alcohol dependence are associated with neuronal loss with the prefrontal cortex being particularly susceptible to neurotoxic damage. This brain region is involved in the development and persistence of alcohol addiction and neurotoxic damage is likely to exacerbate the reinforcing effects of alcohol and may hinder treatment. Understanding the mechanism of alcohol’s neurotoxic effects on the brain and the genetic risk factors associated with alcohol abuse are the focus of current research. Because of its well-established role in neurodegenerative and neuropsychological disorders, and its emerging role in the pathophysiology of addiction, here we review the genetic and epigenetic factors involved in regulating α-synuclein expression and its potential role in the pathophysiology of chronic alcohol abuse. Elucidation of the mechanisms of α-synuclein regulation may prove beneficial in understanding the role of this key synaptic protein in disease and its potential for therapeutic modulation in the treatment of substance use disorders as well as other neurodegenerative diseases.  相似文献

Our previous investigations disclosed that the heat-stable and heat-labile factors obtained from brain cytosol are required for -hydroxylation and oxidation of lignoceric acid by rat brain particulate fraction. The heat-stable factor was recently found to contain glucose-6-phosphate, N-acetylaspartate, glutamate, aspartate, glutamine, inorganic phosphate and low levels of adenosine nucleotide as active components. A combination of these compounds was as effective as the crude heat-stable factor for enzymic activity. Using these compounds, we reinvestigated the requirement for the heat-labile factor. With crude heat-stable factor there was an absolute requirement for the heat-labile factor; however, with various combinations of the individual components of the heat-stable factor, some degree of activity was obtained without the heat-labile factor. When aspartate or one of its derivatives, N-acetylaspartate or oxaloacetate, was used in place of the heat-stable factor, the activity was relatively low but highly stimulated by the addition of heat-labile factor. On the other hand, higher activity was obtained when glutamate or one of its derivatives, glutamine or -ketoglutarate, was used without heat-labile factor. The addition of heat-labile factor to this system did not stimulate the activity. When studying the aspartate family, we discovered that the requirement for the heat-labile factor varied in a descending order: N-acetylaspartate > aspartate > oxaloacetate. Lignoceric acid oxidation was further characterized with rat brain particulate fraction, NADPH, Mg2+, glutamate, inorganic phosphate, and AMP without heat-stable and heat-labile factors. It was found that the requirement for NADPH was also partially eliminated with glutamate but not aspartate. The effects of various inhibitors, such as inhibitors of the electron transfer system, oxidative phosphorylation, the enzymes involved in citric acid cycle, and glycolysis, suggest that the heat-stable factor is involved in producing ATP or other high energy compounds to be used for the activation of lignoceric acid. ATP added to the system in place of heat-stable factor resulted in less than one-half of the lignoceric acid oxidation.  相似文献

The subcellular site of -amylase (EC 1.6.2.1) synthesis and transport was studied in barley aleurone layers incubated in the presence or absence of gibberellic acid (GA3). Using [35S]methionine as a marker, the site of amino-acid incorporation into organelles isolated from aleurone layers incubated with and without GA3 was determined following purification by isopycnic sucrose-density-gradient centrifugation. Incorporation of radioactivity into trichloroacetic-acid-insoluble proteins was greatest in those fractions exhibiting activity of an endoplasmic reticulum (ER) marker enzyme. Further fractionation of densitygradient fractions by sodium-dodecyl-sulfate polyacrylamide-gel electrophoresis showed that a major portion of the radioactivity in the ER fractions was present in a protein co-migrating with marker -amylase. This protein was identified as authentic -amylase by immunoadsorbent chromatography and affinity chromatography. The newly synthesized -amylase associated with the ER was shown to be sequenstered within the lumen of the ER by experiments which showed that the enzyme was resistant to proteolytic degradation. The labelled -amylase sequestered in the ER can be chased from this organelle when tissue is incubated in unlabelled methionine following a 1-h pulse of labelled methionine. The isoenzymic forms of -amylase found in tissue homogenates and incubation media of aleurone layers incubated with and without GA3 were characterized after chromatography on diethylaminoethyl cellulose. In homogenates of GA3-treated aleurone layers, five peaks of -amylase activity were detected, while in homogenates of aleurone layers incubated with-out GA3 only three peaks of activity were found. In incubation media, four isoenzymes were found after GA3 treatment and two were found after incubation without GA3. The 8^th International Workshop on Human Leucocyte Differentiation Antigens (chaired by HZ and managed by BS) was run over a 4-year period and culminated in a conference in December 2004. Here we review the achievements of the HLDA Workshops and provide links to information on CD molecules and antibodies against them, including the 93 new CDs assigned in the 8^th Workshop. We consider what remains to be achieved (including an estimate of the number of leucocyte surface molecules still to be discovered), and how the field can best move forward.  相似文献

Apart from differences in circadian phase position, individuals with different morningness–eveningness levels vary in many more characteristics. Particularly consistent relationships have been observed between morningness–eveningness and mood. Eveningness has been associated with disadvantageous mood, e.g. depressiveness in healthy individuals, and mood disorders. A concept of social jetlag suggests that evening subjects function in less advantageous environments due to discrepancies between internal and social time (societies promote morning-oriented functioning), which results in their lowered mood. Individual temperament, as defined by the Regulative Theory of Temperament (RTT), refers to the capacity of the human organism to meet environmental requirements – the greater the capacity, the less negative impact of external conditions. Thus, the aim of this study is to determine which RTT traits are linked to both morningness–eveningness and mood dimensions and to test whether they account for the relationship between morningness–eveningness and mood. A sample of 386 university students (267 female) aged between 19 and 47 (M?=?21.15, SD?=?4.23) years completed the University of Wales Institute of Science and Technology (UWIST) Mood Adjective Check List, Morningness–Eveningness Questionnaire and Formal Characteristics of Behaviour – Temperament Inventory. Analyses revealed lower endurance (EN) and higher emotional reactivity (ER) related to eveningness as well as to lower hedonic tone (HT), energetic arousal (EA) and to higher tense arousal (TA). Moreover, eveningness was associated with lower HT, EA and higher TA. Among RTT traits, EN was most strongly related to eveningness, and mediation analyses revealed that this temperamental trait fully mediated the relationship between eveningness and the three mood dimensions. The remaining RTT traits did not provide more explanation of the association between morningness–eveningness and mood than EN itself. If subjects did not differ in EN, the association between morningness–eveningness and mood was absent. EN is discussed as a protective factor against negative consequences of social jetlag and particularly lowered mood in evening individuals.  相似文献

Extraction of Triton Photosystem II chloroplast fragments with 0.2% methanol in hexane for 3 h results in the removal of 90 to 95% of the plastoquinone in the original preparation. The extracted fragments (chlorophyll : plastoquinone ratio, 900 : 1) showed no P-680 photooxidation at 15 K after a single laser flash. The extracted fragments also showed no light-induced C-550 absorbance change at 77 K. Reconstitution of the primary reaction of Photosystem II, as evidenced by restoration of low-temperature photooxidation of P-680, could be obtained by the addition of plastoquinone A but not by the addition of β-carotene. The addition of β-carotene plus plastoquinone A restored the C-550 absorbance change. These results indicate that plastoquinone functions as the primary electron acceptor of Photosystem II and that β-carotene does not play a direct role in the primary photochemistry but is required for the C-550 absorbance change.  相似文献
