The rodent whisker system is a major model for understanding neural mechanisms for tactile sensation of surface texture (roughness). Rats discriminate surface texture using the whiskers, and several theories exist for how texture information is physically sensed by the long, moveable macrovibrissae and encoded in spiking of neurons in somatosensory cortex. However, evaluating these theories requires a psychometric curve for texture discrimination, which is lacking. Here we trained rats to discriminate rough vs. fine sandpapers and grooved vs. smooth surfaces. Rats intermixed trials at macrovibrissa contact distance (nose >2 mm from surface) with trials at shorter distance (nose <2 mm from surface). Macrovibrissae were required for distant contact trials, while microvibrissae and non-whisker tactile cues were used for short distance trials. A psychometric curve was measured for macrovibrissa-based sandpaper texture discrimination. Rats discriminated rough P150 from smoother P180, P280, and P400 sandpaper (100, 82, 52, and 35 μm mean grit size, respectively). Use of olfactory, visual, and auditory cues was ruled out. This is the highest reported resolution for rodent texture discrimination, and constrains models of neural coding of texture information.  相似文献

Metabolite gradients might guide mitochondrial localization in cells and angiogenesis in tissues. It is unclear whether they can exist in single cells, because the length scale of most cells is small compared to the expected diffusion times of metabolites. For investigation of metabolic gradients, we need experimental systems in which spatial patterns of metabolism can be systematically measured and manipulated. We used concentrated cytoplasmic extracts from Xenopus eggs as a model cytoplasm, and visualized metabolic gradients formed in response to spatial stimuli. Restriction of oxygen supply to the edge of a drop mimicked distance to the surface of a single cell, or distance from a blood vessel in tissue. We imaged a step-like increase of Nicotinamide adenine dinucleotide (NAD) reduction approximately 600 microm distant from the oxygen source. This oxic-anoxic switch was preceded on the oxic side by a gradual rise of mitochondrial transmembrane potential (Deltapsi) and reactive oxygen species (ROS) production, extending over approximately 600 microm and approximately 300 microm, respectively. Addition of Adenosine triphosphate (ATP)-consuming beads mimicked local energy sinks in the cell. We imaged Deltapsi gradients with a decay length of approximately 50-300 microm around these beads, in the first visualization of an energy demand signaling gradient. Our study demonstrates that mitochondria can pattern the cytoplasm over length scales that are suited to convey morphogenetic information in large cells and tissues and provides a versatile model system for probing of the formation and function of metabolic gradients.  相似文献

Research on the comparative morphology of pollen grains depends crucially on the application of appropriate microscopy techniques. Information on the performance of microscopy techniques can be used to inform that choice. We compared the ability of several microscopy techniques to provide information on the shape and surface texture of three pollen types with differing morphologies. These techniques are: widefield, apotome, confocal and two-photon microscopy (reflected light techniques), and brightfield and differential interference contrast microscopy (DIC) (transmitted light techniques). We also provide a first view of pollen using super-resolution microscopy. The three pollen types used to contrast the performance of each technique are: Croton hirtus (Euphorbiaceae), Mabea occidentalis (Euphorbiaceae) and Agropyron repens (Poaceae). No single microscopy technique provided an adequate picture of both the shape and surface texture of any of the three pollen types investigated here. The wavelength of incident light, photon-collection ability of the optical technique, signal-to-noise ratio, and the thickness and light absorption characteristics of the exine profoundly affect the recovery of morphological information by a given optical microscopy technique. Reflected light techniques, particularly confocal and two-photon microscopy, best capture pollen shape but provide limited information on very fine surface texture. In contrast, transmitted light techniques, particularly differential interference contrast microscopy, can resolve very fine surface texture but provide limited information on shape. Texture comprising sculptural elements that are spaced near the diffraction limit of light (~250 nm; NDL) presents an acute challenge to optical microscopy. Super-resolution structured illumination microscopy provides data on the NDL texture of A. repens that is more comparable to textural data from scanning electron microscopy than any other optical microscopy technique investigated here. Maximizing the recovery of morphological information from pollen grains should lead to more robust classifications, and an increase in the taxonomic precision with which ancient vegetation can be reconstructed.  相似文献

Questions: How should we evaluate the success of new distance measures combining community abundance and phylogenetic information? How do we interpret ordinations using these metrics? Methods: We generated synthetic data along a known environmental gradient with two hypothetical underlying phylogenetic structures: niche phylogenetically conserved or dispersed along a gradient. We also examined tree species composition associated with gradients in elevation and longitude in Oregon, USA. NMS ordinations of plots in species space from phylogenetic (PD) and Sørensen distance (SD) matrices allowed comparison of the use of PD in different scenarios. Results: PD caused plots to cluster based on the clades that they contained, reducing stress with the synthetic data but not with the real example. Phylogenetic distance highlighted clades related to gradients when these were associated. When phylogeny was not conserved along a gradient, that gradient was less strong. Regardless of phylogenetic conservation, NMS using SD consistently extracted the strongest gradients in species composition. Conclusions: The success of PD should be evaluated on how well it extracts gradients in species composition and allows community ecologists to determine which gradients are partially explained by phylogeny and not based on its ability to reduce ordination stress. PD ordinations can help community ecologists interpret niche conservation but may obscure gradients related to species composition when niches are not conserved along the gradient of interest at the scale of the study.  相似文献

Bone morphogenetic proteins (BMPs) are expressed broadly and regulate a diverse array of developmental events in vivo. Essential to many of these functions is the establishment of activity gradients of BMP, which provide positional information that influences cell fates. Secreted polypeptides, such as Noggin, bind BMPs and inhibit their function by preventing interaction with receptors on the cell surface. These BMP antagonists are assumed to be diffusible and therefore potentially important in the establishment of BMP activity gradients in vivo. Nothing is known, however, about the potential interactions between Noggin and components of the cell surface or extracellular matrix that might limit its diffusion. We have found that Noggin binds strongly to heparin in vitro, and to heparan sulfate proteoglycans on the surface of cultured cells. Noggin is detected only on the surface of cells that express heparan sulfate, can be specifically displaced from cells by heparin, and can be directly cross-linked to a cell surface proteoglycan in culture. Heparan sulfate-bound Noggin remains functional and can bind BMP4 at the plasma membrane. A Noggin mutant with a deletion in a putative heparin binding domain has reduced binding to heparin and does not bind to the cell surface but has preserved BMP binding and antagonist functions. Our results imply that interactions between Noggin and heparan sulfate proteoglycans in vivo regulate diffusion and therefore the formation of gradients of BMP activity.  相似文献

A simple technique has been developed for establishing stable gradients of a substance in agar. The technique involves the creation of a spherically symmetric concentration profile in which concentration varies inversely with the distance from the source and is independent of the diffusion coefficient of the substance. It has been shown that the gradients established with this technique are stable for at least 190 h. and, on a theoretical basis, they can be kept stable for more than 1000 h. Time-variant gradients can also be established, if desired, using the same system and limiting either the source or the agar sink. It must be emphasized that a stable gradient cannot be obtained by using a shallow agar layer as a sink. The use of such conditions (e.g. the agar in a standard petri dish) can result only in time-variant gradients. The solution to the diffusion equation in a spherically symmetric system establishes the expected concentration profile, the basis for adjusting it, and the parameters that control the behavior of the system. Some useful applications for examining chemotaxis on a solid surface as well as possible further developments are discussed.  相似文献

Endothelial cells in blood vessels are exposed to bloodflow and thus fluid shear stress. In arterial bifurcations and stenoses, disturbed flow causes zones of recirculation and stagnation, which are associated with both spatial and temporal gradients of shear stress. Such gradients have been linked to the generation of atherosclerotic plaques. For in-vitro studies of endothelial cell responses, the sudden-expansion flow chamber has been widely used and described. A two-dimensional numerical simulation of the onset phase of flow through the chamber was performed. The wall shear stress action on the bottom plate was computed as a function of time and distance from the sudden expansion. The results showed that depending on the time for the flow to be established, significant temporal gradients occurred close to the second stagnation point of flow. Slowly ramping the flow over 15 s instead of 200 ms reduces the temporal gradients by a factor of 300, while spatial gradients are reduced by 23 percent. Thus, the effects of spatial and temporal gradients can be observed separately. In experiments on endothelial cells, disturbed flow stimulated cell proliferation only when flow onset was sudden. The spatial patterns of proliferation rate match the exposure to temporal gradients. This study provides information on the dynamics of spatial and temporal gradients to which the cells are exposed in a sudden-expansion flow chamber and relates them to changes in the onset phase of flow.  相似文献

Studies of biodiversity along environmental gradients provide information on how ecological communities change in response to biotic and abiotic factors. For instance, distance to water is associated with several factors that shape the structure and the functioning of ecosystems at a range of spatial scales. We investigated the influence of distance to a perennial water source on ant communities in a semi‐arid savanna in northern Botswana. Ant abundance, taxonomic richness, and both alpha and beta diversity were generally higher during the wet than the dry season. However, there were strong seasonal influences on the effects of distance to water, with more pronounced effects during the wet season. While both abundance and beta diversity declined with increasing distances to water during the wet season, there was a contrasting increase in alpha diversity. There was no major effect of distance to water on taxonomic richness during either season. Beta diversity was as high across as along gradients, and we found support for modular rather than nested community structures along gradients. Our study demonstrated that small‐scale gradients in distance to water can influence several aspects of ant communities in semi‐arid savannas. However, our results also point to strong effects of small‐scale environmental variation, for instance associated with vegetation characteristics, soil properties, and plant community structure that are not directly linked to water access.  相似文献

It has been suggested that a measure of the gradients of vertical disparity over a surface may scale the mapping between horizontal disparity and perceived depth. We have investigated this possibility by obtaining estimates of the depth within stereograms that simulated two apposed fronto-parallel planes placed at different distances from an observer. The gradients of vertical disparity in a stereogram were set to simulate those appropriate to a viewing distance of 12.5 cm, 25 cm, 50 cm or 100 cm, whereas the distance specified by vergence and accommodative cues was always fixed at 50 cm. Judgements of the perceived depth between the two planes were uninfluenced by changes in the gradients of vertical disparity. It thus seems that the human visual system does not employ vertical disparity as a scaling parameter in stereoscopic depth judgements.  相似文献

Experiments have shown that the depletion of polymer in the region between two apposed (contacting or nearly contacting) bilayer membranes leads to fusion. In this paper we show theoretically that the addition of nonadsorbing polymer in solution can promote lateral contraction and phase separation of the lipids in the outer monolayers of the membranes exposed to the polymer solution, i.e., outside the contact zone. This initial phase coexistence of higher- and lower-density lipid domains in the outer monolayer results in surface tension gradients in the outer monolayer. Initially, the inner layer lipids are not exposed to the polymer solution and remain in their original "unstressed" state. The differential stresses on the bilayers give rise to a Marangoni flow of lipid from the outer monolayers in the "contact zone" (where there is little polymer and hence a uniform phase) to the outer monolayers in the "reservoir" (where initially the surface tension gradients are large due to the polymer-induced phase separation). As a result, the low-density domains of the outer monolayers in the contact zone expose their hydrophobic chains, and those of the inner monolayers, to the solvent and to each other across the narrow water gap, allowing fusion to occur via a hydrophobic interaction. More generally, this type of mechanism suggests that fusion and other intermembrane interactions may be triggered by Marangoni flows induced by surface tension gradients that provide "action at a distance" far from the fusion or interaction zone.  相似文献

Dynamic texture spreading is a filling-in phenomenon where a colored pattern perceptually spreads onto an area confined by virtual contours in a multi-aperture motion display. The spreading effect is qualitatively similar to static texture spreading but widely surpasses it in strength, making it particularly suited for quantitative studies of visual interpolation processes. We first carried out two experiments to establish with objective tasks that texture spreading is a genuine representation of surface qualities and thus goes beyond mere contour interpolation. Two subsequent experiments serve to relate the phenomenon to ongoing discussions about potentially responsible mechanisms for spatiotemporal integration. With a phenomenological method, we examined to what extent simple sensory persistence might be causally involved in the effect under consideration. Most of our findings are consistent with the idea of sensory persistence, and indicate that information fragments are integrated over a time window of about 100 to 180 ms to form a complete surface representation.  相似文献

Texture mapping is an increasingly popular technique in molecular modeling. It is particularly effective in representing high-resolution surface detail using a low-resolution polygonal model. We describe how texture mapping can be used with parametric molecular surfaces represented as expansions of spherical harmonic functions. We define analytically the texture image and its transformation to a parametric surface. Unlike most methods of texture mapping, this transformation defines a one-to-one correspondence between the surface and the texture; texture coordinates are derived from the location of the surface point and not from physical properties at the surface point. This has advantates for the interactive visualization of surface data. We control the interactive response time by lowering the resolution of the polygon mesh while retaining the high-resolution detail of the texture, or we can lower the resolution of the texture image with the same polygonal model. By using a well-defined convention for texture coordinates, we can use the same image for the original surface or its parametric representation, and we can rapidly switch between images that represent different surface properties without recomputing the texture coordinates. Parametric surfaces allow new flexibility for the visualization of molecular surface data.  相似文献

Biological similartiy typically decreases with geographical distance. Despite the recent attention to the distance decay relationship, there is no consensus on how the relationship varies across organism groups, geographic gradients and environments. We first conducted a quantitative meta-analysis of 401 distance decay relationships across a wide range of organisms, ecosystems and geographical gradients, and then united the effects of categorical and continuous variables on the rate of distance decay using a general linear model (GLM). As effect sizes we used the similarity at one km distance (initial similarity) and the distance that halves the similarity from its value at one km distance (halving distance). Both the initial similarity and halving distance were significantly affected by variables characterizing the spatial scale, organism properties, study region and ecosystem concerned. The patterns appear robust as the results of meta-analysis and GLM only differed in marginal details. According to GLM with Akaike's information criterion, the most parsimonious models explained 55.3 and 37.6% of variance in initial similarity and halving distance, respectively. Across large scales, similarity was decreasing slightly faster at high latitudes than at low latitudes, while small-scale turnover was higher at low latitudes. We also found significant differences in initial similarity among the realms, with terrestrial systems showing higher small-scale beta diversity. The decrease in community similarity at large scales was higher among organisms that are actively mobile than among passively dispersed organisms. We conclude that regression of similarity against distance unites several ecological phenomena such as dispersal propensity and environmental structuring, and provides an effective approach for gauging the spatial turnover across sites. We also found that the patterns in beta-diversity are highly scale-dependent.  相似文献

The luminance and colour gradients across an image are the result of complex interactions between object shape, material and illumination. Using such variations to infer object shape or surface colour is therefore a difficult problem for the visual system. We know that changes to the shape of an object can affect its perceived colour, and that shading gradients confer a sense of shape. Here we investigate if the visual system is able to effectively utilise these gradients as a cue to shape perception, even when additional cues are not available. We tested shape perception of a folded card object that contained illumination gradients in the form of shading and more subtle effects such as inter-reflections. Our results suggest that observers are able to use the gradients to make consistent shape judgements. In order to do this, observers must be given the opportunity to learn suitable assumptions about the lighting and scene. Using a variety of different training conditions, we demonstrate that learning can occur quickly and requires only coarse information. We also establish that learning does not deliver a trivial mapping between gradient and shape; rather learning leads to the acquisition of assumptions about lighting and scene parameters that subsequently allow for gradients to be used as a shape cue. The perceived shape is shown to be consistent for convex and concave versions of the object that exhibit very different shading, and also similar to that delivered by outline, a largely unrelated cue to shape. Overall our results indicate that, although gradients are less reliable than some other cues, the relationship between gradients and shape can be quickly assessed and the gradients therefore used effectively as a visual shape cue.  相似文献

Vision generally provides reliable predictions for touch and motor-control, but some classes of stimuli evoke visual illusions. Using haptic feedback on virtual 3-D surfaces, we tested the function of touch in such cases. Our experiments show that in the perception of 3-D shapes from texture cues, haptic information can dominate vision in some cases, changing percepts qualitatively from convex to concave and concave to slant. The effects take time to develop, do not outlive the cessation of the feedback, are attenuated by distance, and drastically reduced by gaps in the surface. These dynamic shifts in qualitative perceived shapes could be invaluable in neural investigations that test whether haptic feedback modifies selective activation of neurons or changes the shape-tuning of neurons responsible for percepts of 3-D shapes.  相似文献

Cell-cell interactions may often be characterized by a work of adhesion or equivalent surface tension between cells. This intercellular surface tension may vary from one part of an epithelium to another. Such gradients in surface tension may be important driving factors in morphogenesis. A computational method for the measurement of parameters of cell shape involved in surface tension has been developed. It permits detailed observational analysis of cell-cell interface curvature. The information thus obtained is utilized in an algorithm for estimating gradients of surface tension in epithelia. The use of these methods should permit cell biologists and embryologists to decide whether or not developing cells behave like bubbles, and if so, to what extent their adhesive forces contribute to various aspects of embryogenesis.  相似文献

In a series of experiments we have investigated the perception of Moiré patterns as a function of spatial density, rotation and temporal display parameters. Results indicate that the local correlation extraction process involved in the perception of these patterns is not feature specific, yet is driven by excitatory (correlated) and inhibitory (uncorrelated) information under a form of spatial summation. These results are comparable with recent results on texture discrimination where texture interpoint distance distributions (dipole statistics) have also been discovered to have excitatory and inhibitory components.  相似文献

Predictive models of tumor response based on heterogeneity metrics in medical images, such as textural features, are highly suggestive. However, the demonstrated sensitivity of these features to noise does affect the model being developed. An in-depth analysis of the noise influence on the extraction of texture features was performed based on the assumption that an improvement in information quality can also enhance the predictive model. A heuristic approach was used that recognizes from the beginning that the noise has its own texture and it was analysed how it affects the quantitative signal data. A simple procedure to obtain noise image estimation is shown; one which makes it possible to extract the noise-texture features at each observation. The distance measured between the textural features in signal and estimated noise images allows us to determine the features affected in each observation by the noise and, for example, to exclude some of them from the model. A demonstration was carried out using synthetic images applying realistic noise models found in medical images. Drawn conclusions were applied to a public cohort of clinical images obtained using FDG-PET to show how the predictive model could be improved. A gain in the area under the receiver operating characteristic curve between 10 and 20% when noise texture information is used was shown. An improvement between 20 and 30% can be appreciated in the estimated model quality.  相似文献
